    Notes: Direct introduction of a N-atom in one step at C (5) of 5-hydroxyuridine (4a) or 5-hydroxy-2′-deoxyuridine (4b) by certain primary amines led to the synthesis of two novel C(5)-N-spin-labeled nucleoside analogs and to several C(5)-N-aryl adducts. Substitution by 4-amino-2,2,6,6-tetramethylpiperidinooxyl (3) at C(5) of 4a or 4b led to the spin-labeled nucleosides 5-[(1-oxyl-2,2,6,6-tetramethyl-4-piperidinyl)amino]uridine and -2′-deoxyuridine (2a and 2b, respectively). The analogous C(5)-substituted aniline adducts 5-anilino uridine (5a) and 5-anilino-2′-deoxyuridine (5b) and the p-toluidine adducts 5-(p-toluidino)uridine (6a) and 5-(p-toluidino)-2-deoxyuridine (6b) were also prepared. In addition, results of the antiviral and antimetabolic activity of some of these analogs are reported.

    Notes: N-Aminoalkylbenzamides and N-aminoalkyl-o-methoxybenzamides have been prepared and examined for their pKa, log P and dopamine receptor affinity. The pKa values range from ca. 7.5 for the derivatives having a one-C-atom side-chain, to ca. 10.3 for the N-aminobutyl derivatives. These variations with chain length are satisfactoryly explained by a field model. The variations in (log P)-values as a function of chain length and substitution at the N-atom indicate the involvement of proximity and conformational effects. The complete inability of the compounds to displace 3H-spiperone and 3H-sulpiride from their specific rat striatal binding sites demonstrates the critical role of adequate aromatic substitution at positions 4 and 5.

    Notes: The modified nucleoside Πd (1) was used in the synthesis of the oligonucleotide d (TpTpΠpCpGpTpCpApApApApTpC). Diester methodology being unsatisfactory, the triester synthesis was investigated with the unnatural nucleoside. An improved method of nucleoside phosphorylation was developed for the synthesis of the fully-protected nucleotide 2e. This molecule could be cleanly and selectively deprotected, and allowed the efficient synthesis of the desired oligonucleotide.

    Notes: Photochemistry of 5,6-Epoxy-1,3-dienes: Influence of a 7-Hydroxy Substituent on the Carbene FormationOn singlet excitation (λ=254 nm) in MeCN the hydroxy-epoxydiene (E)-4 undergoes photocleavage to the carbene intermediates d and e as main processes. The carbene d, showing behaviour typical of vinyl carbenes, undergoes addition to the adjacent double bond furnishing the cyclopropenes 5A + B. The carbene e, however, undergoes an insertion reaction into the neighbouring carbinol C,H-bond leading to the enol intermediate 21, which gives rise to the compounds 6A + B and 7A + B. To a lesser extent the products 8A + B are formed via another enol intermediate (32). On photolysis of (E)-4 in MeOH instead of MeCN the enol intermediates 21 and 32 undergo rapid tautomerisation to the ketones 9A + B (main products) and 11.

    Notes: Bamboo carbohydrates were hydrolyzed with commercial amylases and a mixture of fungal culture broths containing cellulolytic and hemicellulolytic enzymes. The effects of cooking temperature and the size of fiber particles were also investigated. It was found that the higher the cooking temperature, the higher the rate of sugar formation and the lower the viscosity of the slurry. Additions of cellulose and hemicellulose digesting enzymes increased the sugar yield and decreased the viscosity of both the cooked and noncooked slurries. A smaller size of particle appeared to favor the average saccharification rate. Although glucose, xylose, and cellobiose were present in the hydrolysates, only 50% of the total carbohydrate was digested, and 78.9% of this was converted to reducing sugars. The alcohol efficiency for the fermentation of cooked and noncooked mashes by Saccharomyces was about 85%.

Durch Anwendung unterschiedlicher Methoden der 13C-Kernresonanz konnten alle Grundgerüst-C-Atome in [D6]DMSO ebenso wie in [D6]DMSO/NaOD zugeordnet werden. - Die Bildung des Phenolatanions von 1 bewirkte bei ortho- und ipso-C-Atomen auf der einen bzw. beim para-C-Atom auf der anderen Seite markante, entgegengesetzte Verschiebungen der betreffenden Signale. Diese Methodik erwies sich als entscheidend bei der Absicherung von C-3 als hydroxyliertes C-Atom, so daß die andere Möglichkeit, C-2, ausgeschlossen werden konnte.
    Notes: From the Lauraceae Ocotea bucherii has been isolated the 3-hydroxy-1,2,9,10-tetramethoxy-aporphine (1, 3-hydroxyglaucine) whose structure was determined using spectroscopic methods. Application of various techniques in 13C NMR led to the spectral assignment of each skeletal carbon in [D6]DMSO as well as [D6]DMSO/NaOD. - The phenolate anion corresponding to 1 gave rise to pronounced, but opposite variations of the chemical shifts of the ipso and ortho carbons on the one hand or the para carbon on the other. This method proved to be decisive in confirming C-3 as the carbon bearing the hydroxy group, whereas the alternative C-2 was excluded.

    Description: Selective pharmacological inhibition of thromboxane A2 synthesis did not prevent arachidonate-induced aggregation of human platelets in vitro. Prevention was instead achieved by a combination of thromboxane A2 inhibitors with low concentrations of aspirin. The latter partially reduced the proaggregatory cyclooxygenase products that accumulated when thromboxane A2 synthesis was blocked. The aspirin concentrations did not affect per se either platelet aggregation or prostacyclin synthesis in cultured human endothelial cells.     Description: The external and internal interosseous intercostal muscles were separately stimulated at end-expiratory lung volume in anesthetized dogs. These muscles were all found to elevate the ribs into which they insert.     Description: The involvement of plasma fibronectin in phagocytosis of bacteria was investigated by testing the binding of fibronectin to several species of bacteria and by evaluating the ability of fibronectin to promote binding and endocytosis of two species of these bacteria by phagocytic cells.
