    Notizen: We present a theoretical model which describes a cooperative helix-coil liquid-crystal phase transition. We show that this model predicts a first-order phase transition where certain types of chainlike macromolecules in solution make a transition from a nearly coiled to a nearly rigid conformation accompanied by a simultaneous development of long-range nematic-type liquid crystalline orientational order. From this model, the phase boundaries between nematic and isotropic phases are obtained as functions of concentration of macromolecules and of other physical parameters.

    Notizen: As a study preliminary to attempts at photoaffinity and cooperative affinity labelling of cells and cell membrane vesicles bearing opiate receptors, the title compounds were prepared and tested by rat brain membrane binding assays and by a modified guinea-pig ileum bio-assay. The potency of the two compounds in both systems was considerably greater than that of the standard peptide, [Leu5]enkephalin, justifying further work with these and similar compounds.

    Notizen: Because of its overall steric resemblance to the phenylalanine analogue, carboranylalanine, the title compound was prepared by the modified Strecker synthesis of Patel & Worsley. The use of (S)-(-)-α-methylbenzylamine in the synthesis, the positive trend of [α]D with increasing protonation, and the thin-layer chromatographic behaviour of synthetic diastereomeric dipeptides are strong indications that the configuration at the asymmetric carbon atom is S (natural L). Its optical purity was ascertained by purification via the quinine and ephedrine salts of t-butoxy-carbonyl-adamantylalanine. The new amino acid shall be used for studies of structure-activity relationships of phenylalanine and leucine residues in biologically active peptides. In addition, a convenient synthesis in excellent yield of the starting material, 2-(1-adamantyl)-ethanal, by Pfitzner-Moffat oxidation of 2-(1-adamantyl)-ethanol is described.

    Notizen: The heterogeneous polymerization of L-proline N-carboxy anhydride was carried out in acetonitrile and benzene, using butylamine as the initiator. Polyproline I was formed in acetonitrile and polyproline II in benzene in the course of the polymerization. The conversion attained 99% and 66% after 1 week-polymerization in acetonitrile and benzene, resp. Extended chain crystals were obtained by the polymerization in acetonitrile. On the other hand, poly(L-proline) obtained by the polymerization in benzene gave aggregates into which many growing chain ends were occluded. The differences in the crystallization behaviour during the polymerization between the two different liquids may be due to the conformation of the resultant polymers. Polyproline I exhibits a cross-section per chain of 0.709 nm2, whereas polyproline II shows only one of 0,386 nm2. The wider the cross-section per chain the more the growing, active chain ends can react with the monomer, giving rise to a higher conversion. In contrast with the case of crystallization of such α-helical chains as poly(L-alanine) during polymerization, hexagonal lamellar crystals were formed.

    Notizen: Using immobilized glucose isomerase, the effects of superficial velocity of the reaction solution flowing through a packed-bed reactor on the apparent kinetic constants of reversible reaction system were studied. The results showed that the apparent kinetic constants, both Vm″ and Km″, of the forward reaction varied significantly as the superficial velocity is changed, whereas those of the reverse reaction varied only slightly. Using the kinetic data determined experimentally, computer simulation of the enzyme reactor performance was carried out, and the importance of the external mass transfer in the proximity of immobilized-enzyme particles was recognized. The reactor performance, expressed in terms of productivity, was examined as a function of the reactor height-to-diameter ratio, H/D. The productivity of the reactor system goes through a maximum value at a H/D ratio of about 1.6. and decreases as the H/D ratio increases. Theoretical analysis of the reaction kinetics of immobilized-enzyme system that has reversible reaction kinetics is also presented. The experimental results showed good agreement with the results found from the theoretical analysis and the computer simulation studies. Based on the principles of the methods and the results presented in this paper, it is anticipated that one can predict the optimal design and operating conditions for the glucose isomerase reactor system and that application of the results could be extended to other enzyme systems with reversible reaction kinetics.

    Notizen: It is well established that both molecular weight (M) and its distribution (MD) affect many polymer properties such as mechanical behavior. Thus studies have shown that fatigue life is enhanced by increases in M. Research here has shown that with notched specimens fatigue crack propagation (FCP) rates are dramatically decreased by increasing M, even when the M is high enough that the static fraeture energy has essentially reached its asymptotic limit. In this study, specimens of poly(methyl methacrylate) containing either high- or low-M tails were prepared and characterized. The earlier finding that FCP rates are inversely related to average M was confirmed, but specific effects of M distribution were observed. At constant Mn, a low-M tail had little effect on FCP resistance, while a high-M tail improved FCP resistance of polymers whose average M was too low for effective entanglements. Thus with high-M tails, it was possible to test specimens whose average M's were too low to permit machining. It is proposed that the effects noted are due to relative stabilization or destabilization of crazes ahead of the crack.

    Notizen: Polyamides were prepared from C6 to C12 diamines with brassylic acid, a linear C13 dicarboxylic acid, derived from Crambe seed oil. One distinct characteristic of these polymers is their low moisture adsorption as compared to nylon 66 and nylon 6. To modify the properties of these nylons, multi-component copolyamides were prepared from hexamethylene diamine and mixtures of brassylic acid with adipic, terephthalic, or isophthalic acids. It was found that the melting points of the co-polyamides were changed by the choice and the levels of the diacids used. The melting point-composition curves all show a eutectic minimum. The glass transition temperature of nylon 6,13 is also changed by the incorporation of other diacids. Water adsorption of nylon 6,13 increases with increased substitution of brassylic acid by other diacids in the order of adipic 〉 isophthalic ≥ terephthalic. Mechanical properties of some copolyamides are in the same range as the commercial nylon 11 and nylon 12. The low moisture absorption, reduced fabrication temperature, and the wide range of properties obtainable through copolymerization make copolyamides derived from brassylic acid potentially suitable as specialty tubing, powder coatings, and molded machine parts. They will be commercially viable when brassylic acid becomes available on a large scale and is competitively priced.

    Notizen: A variant of murine L5178Y lymphoma resistant to procaine hydrochloride (PH) was selected by exposing the cells to gradual increments of PH in the growth medium until the cell grew exponentially in the presence of 1.5 mM PH. Using cinephotomicrography, it was observed that the majority of cells that initially succumbed to PH failed to undergo successful mitosis. With respect to chromosomal, cell size distribution and flow microfluorometric analyses, the PH-resistant cells are very similar to a spontaneous tetraploid cell line (R1T) previously cloned. The isolated cells, designated R1/P, were also found to be cross-resistant to analogues of PH, namely, lidocaine, tetracaine and dibucaine. The naturally-occurring tetraploid cell line (R1T) was also found to be more resistant to local anesthetics, although not to the same extent as R1/P cells. Since the enzyme that hydrolyzes procaine appears to be absent in all these lymphoid cell lines, the difference in resistance does not appear to depend on differences in the ability of these cells to remove the agent. It is suggested that an alteration in the structure and/or function of the plasma membrane in R1/P cells have rendered them either less sensitive to the membrane-perturbing effects of the local anesthetics or less permeable to local anesthetics molecules. The ability of local anesthetics to affect membranes and cytoskeleton structures may play a role in the genesis and/or selection of these cell variants.

    Notizen: The effect of Ca+2 on the transport and intracellular distribution of Na+ and K+ in Ehrlich ascites tumor cells was investigated in an effort to establish the mechanism of Ca+2-induced hyperpolarization of the cell membrane. Inclusion of Ca+2 (2mM) in the incubation medium leads to reduced cytoplasmic concentrations of Na+, K+ and Cl- in steady state cells. In cells inhibited by ouabain, Ca+2 causes a 41% decrease in the rate of net K+ loss, but is without effect on the rate of net Na+ accumulation. Net K+ flux is reduced by 50%, while net Na+ flux is unchanged in the transport-inhibited cells. The membrane potential of cells in Ca+2-free medium (-13.9 ± 0.8 mV) is unaffected by the addition of ouabain. However, the potential of cells in Ca+2-containing medium (-23.3 ± 1.2 mV) declines in one hour after the addition of ouabain to values comparable to those of control cells (-15.2 ± 0.7 mV).The results of these experiment are consistent with the postulation that Ca+2 exerts two effects on Na+ and K+ transport. First, Ca+2 reduces the membrane permeability to K+ by 25%. Second, Ca+2 alters the coupling of the Na/K active transport mechanism leding to an electrogenic hyperpolarization of the membrane.

    Notizen: Freshly explanted human myeloma cells formed colonies of monoclonal plasma cells in soft agar in the presence of medium conditioned by the adherent spleen cells of mineral oil-primed BALB/c mice. The medium showed peak activity at a dilution of 1:4. 2-mercaptoethanol or monothioglycerol was necessary for colony formation. Other thiols tested were ineffective in promoting colony growth. Colony-forming cells adhered to nylon wool, but not glass beads or plastic dishes. The presence of E-rosetting cells was not required for myeloma colony formation. Antibody prepared against a human myeloma cell line, RPMI 8226, reduced colony formation. These studies demonstrate the usefulness of this bioassay for determining functional properties of the myeloma colony-forming cell.
