    Description: Operating within safe and just Earth system boundaries requires mobilizing key actors across scale to set targets and take actions accordingly. Robust, transparent and fair cross-scale translation methods are essential to help navigate through the multiple steps of scientific and normative judgements in translation, with clear awareness of associated assumptions, bias and uncertainties. Here, through literature review and expert elicitation, we identify commonly used sharing approaches, illustrate ten principles of translation and present a protocol involving key building blocks and control steps in translation. We pay particular attention to businesses and cities, two understudied but critical actors to bring on board.

    Description: Superconducting links are an innovative solution for bulk power transmission, distinguished by their compact dimensions, high efficiency and small environmental footprint. As with any new technology field, there is a large amount of design possibilities for such links, each of them having a profound impact on the system configuration. For instance, changing the material can imply a change in the working temperature from 20 to 70 K and has consequences on the maximum link length. This article presents the dichotomic decision possibilities for the optimized design of a high-power superconducting link, focusing on some of the key components of the cable system. The complex design optimization process is exemplified using the European project Best Paths, in which the first 3-gigawatt-class superconducting cable system was designed, optimized, manufactured, and successfully tested.

    Description: The health of the ocean, central to human well-being, has now reached a critical point. Most fish stocks are overexploited, climate change and increased dissolved carbon dioxide are changing ocean chemistry and disrupting species throughout food webs, and the fundamental capacity of the ocean to regulate the climate has been altered. However, key technical, organizational, and conceptual scientific barriers have prevented the identification of policy levers for sustainability and transformative action. Here, we recommend key strategies to address these challenges, including (1) stronger integration of sciences and (2) ocean-observing systems, (3) improved science-policy interfaces, (4) new partnerships supported by (5) a new ocean-climate finance system, and (6) improved ocean literacy and education to modify social norms and behaviors. Adopting these strategies could help establish ocean science as a key foundation of broader sustainability transformations.

    Description: In Afghanistan, local communities have played a critical role in security, especially in rural areas. Afghan national security forces are important to the top-down strategy, but the Afghan government and NATO forces also need to leverage local communities to gain a complementary bottom-up strategy. This analysis discusses the viability of establishing local security forces in Afghanistan and addresses concerns about the wisdom of such policies.

    Description: Listeners with hearing impairments have difficulties understanding speech in the presence of background noise. Although prosthetic devices such as hearing aids and cochlear implants may improve the hearing capability, listeners with hearing impairments still complain about their speech perception in the presence of noise. The basic tonal audiometry only gives a cursory idea of the degree of difficulty in spoken communication caused by hearing loss because it does not assess the ability to understand speech. Therefore, the use of speech-in-noise tests to measure hearing loss in complex scenes is an integral part of a patient's audiological study. Most research has concentrated on studying only stationary sound sources, but in natural acoustic scenes, conversations may become very difficult to understand in the presence of moving sound sources such as a moving talker or a passing vehicle. Therefore, this thesis deals with quantifying speech perception in the presence of moving maskers through virtual sound sources presented binaurally via headphones. Significant differences in several conditions were found, revealing that the auditory system assesses differently the moving maskers than the stationary maskers. Therefore, the inclusion of moving conditions in clinical listening tests is recommended, in order to assess speech-in-noise perception in a more realistic environment.

    Description: In multicellular organisms, states with a high degree of tissue turnover like embryogenesis, development, and adult tissue homeostasis need an instantaneous, tightly regulated and immunologically silent clearance of these dying cells to ensure appropriate development of the embryo and adult tissue remodelling. The proper and swift clearance of apoptotic cells is essential to prevent cellular leakage of damage associated molecular patterns (DAMPs) which would lead to the stimulation of inflammatory cytokine responses. In addition to the clearance of apoptotic cells (efferocytosis), backup mechanisms are required to cope with DAMPs (HMGB-1, DNA, RNA, S100 molecules, ATP and adenosine) and other intracellular material (uric acid, intracellular proteins and their aggregates) released from cells, that were not properly cleared and have entered the stage of secondary necrosis. Furthermore, under certain pathologic conditions (e.g. gout, cancer, diabetes) non-apoptotic cell death may transiently occur (NETosis, necroptosis, pyroptosis) which generates material that also has to be cleared to avoid overloading tissues with non-functional cellular waste. Efficient efferocytosis is therefore indispensable for normal tissue turnover and homeostasis. The characterization of various signalling pathways that regulate this complex and evolutionary conserved process has shed light on new pathogenetic mechanisms of many diseases. Impaired clearance promotes initiation of autoimmunity as well as the perpetuation of chronic inflammation, but may also foster anti-tumor immunity under certain microenvironmental conditions. Immunological tolerance is continuously being challenged by the presence of post-apoptotic remnants in peripheral lymphoid tissues. Besides the autoimmune phenotype of chronic inflammatory rheumatoid disorders a plethora of pathologies have been associated with defects in genes involved in clearance, e.g. atherosclerosis, cancer, gout, diabetes, some forms of blindness, neuropathy, schizophrenia and Alzheimer’s disease. The main goal of this research topic is to collect contributions from various disciplines committed to studying pathogenetic mechanisms of the aforementioned disorders and dealing with alterations in the clearance of dying and dead cells, their remnants, and their constituents that leak out after membrane rupture. Integrating the combined collection of knowledge on efferocytosis and clearance of dead cells and their derived waste from different fields of research in physiology and pathophysiology could improve the molecular understanding of these increasingly prevalent diseases and may ultimately result in new therapeutic strategies.

    Description: Massive Open Online Courses (MOOCs) are changing the educational field, challenging traditional institutional strategies and recognition schemes and opening up new opportunities for learners and educators both from within and outside formal education. However, while the potential benefits and risks of the MOOCs have been discussed by scientists and policy makers, the corresponding empirical data is scarce. What’s more, the evidence that is available is usually restricted to a single course or single provider. MOOCKnowledge (http://moocknowledge.eu/), funded by the European Commission’s Institute for Prospective Technological Studies (IPTS), aims to facilitate a shared understanding of the value and efficacy of MOOCs by developing a set of analysis tools and applying them to a wide range of MOOCs. The most powerful outcome of the project would be the possibility to correlate different dimensions of MOOC production, execution, and learners experience. For example, identifying links between financial investment, learning design, and learner outcomes. To do this, we must first develop a conceptual model of the factors which determine or contribute to the value of a MOOC.

    Description: Adult stem cells are responsible for tissue regeneration and repair throughout life. Their quiescence or activation are tightly regulated by common signalling pathways that often recapitulate those happening during embryonic development, and thus it is important to understand their regulation not only in postnatal life, but also during foetal development. In this regard, skeletal muscle is an interesting tissue since it accounts for a large percentage of body mass (about 40%), it is highly amenable to intervention through exercise and it is also key in metabolic and physiological changes underlying frailty susceptibility in the elderly. While muscle-resident satellite cells are responsible for all myogenic activity in physiological conditions and become senescent in old age, other progenitor cells such as mesoangioblasts do seem to contribute to muscle regeneration and repair after tissue damage. Similarly, fibro-adipogenic precursor cells seem to be key in the aberrant response that fills up the space left from atrophied muscle mass and which ends up with a dysfunctional muscle having vast areas of fatty infiltration and fibrosis. The complex interplay between these stem/progenitor cell types and their niches in normal and pathological conditions throughout life are the subjects of intense investigation. This eBook highlights recent developments on the role of stem cells in skeletal muscle function, both in prenatal and postnatal life, and their regulation by transcriptional, post-transcriptional and epigenetic mechanisms. Additionally, it includes articles on interventions associated with exercise, pathological changes in neuromuscular diseases, and stem cell aging.
