    Description: The goal of this book is simple: We would like to show how mortality dynamics can be visualized in the so-called Lexis diagram. To appeal to as many potential readers as possible, we do not require any specialist knowledge. This approach may be disappointing: Demographers may have liked more information about the mathematical underpinnings of population dynamics on the Lexis surface as demonstrated, for instance, by Arthur and Vaupel in 1984. Statisticians would have probably preferred more information about the underlying smoothing methods that were used. Epidemiologists likewise might miss discussions about the etiology of diseases. Sociologists would have probably expected that our results were more embedded into theoretical frameworks....

In the current crisis of the capitalist world system, elites in the United States, along with other central countries, promote fear of crime and terrorism. They shaped these fears so that people looked to authorities for security, which permitted extension of apparatuses of coercion like police and military forces. In the face of growing oppression, rebellion against elite hegemony remains possible. This book offers an analysis of the crisis and strategies for rebellion.

    Description: The fortieth anniversary of the Strategic and Defence Studies Centre’s founding provided the opportunity to assemble many of Australia’s leading analysts and commentators to review some of the more significant issues that should define Australian defence policy. In the first 20 years after its establishment, SDSC scholars played a prominent role in shaping the ideas and aspirations that eventually found official expression in the 1987 Defence of Australia White Paper. This policy sustained a coherent balance between strategy, force structure and budgets for well over a decade. In recent years, however, the cumulative effects of the end of the Cold War and watershed events like the East Timor experience; the attacks on New York and Washington, D.C., in September 2001; the Bali bombings in October 2002; and the invasion of Iraq in March 2003 have fractured the former consensus on defence policy. These developments have eroded acceptance of the core judgements underpinning defence policy. This has led to a more tenuous connection between some recent major equipment acquisitions and declared policy. The unravelling of the consensus on the ‘defence of Australia’ policy means that we must again undertake a balanced, long-term assessment of the nature of Australia’s strategic interests. Only by doing so can we determine the kinds of armed forces that would contribute most effectively to protecting those interests. The papers collected in this volume are not informed by a common view of where Australia should focus its defence policy, but all address themes that should figure prominently in this difficult but essential task.

    Description: This open access book explores the intersection of property law, relocation, and resettlement processes in the United States and among communities that grapple with migration as an adaptation strategy. As communities face the prospect of relocating because of rising seas, policy makers, disaster specialists, and community leaders are scrambling to understand what adaptation pathways are legally possible. While in its ideal application, law functions blindly and without variation, the authors find that legal contradictions come to bear on resettlement processes and place certain communities further in harm’s way. This book will unearth these contradictions in order to understand why successful community-based resettlement has presented such a challenge to communities that are experiencing increasing land deterioration as a result of climate change.

    Description: We cannot expect in East Asia over the foreseeable future to see the sort of conflation of sovereign states that has occurred in Europe. We must anticipate that, for the foreseeable future, the requirement will be for the sensible management and containment of competitive instincts. The establishment of a multilateral security body in East Asia that includes all the key players, and which the major powers invest with the authority to tackle the shaping of the regional security order, remains a critical piece of unfinished business.

    Description: The Hmong are among Australia’s newest immigrant populations. They came as refugees from Laos after the communist revolution of 1975 ended their life there as highland shifting cultivators. The Hmong originate from southern China where many still remain, and others live in Vietnam, Thailand and Burma. Hmong refugees are now also settled in the USA, Canada, France, Germany and French Guyana. Already the beauty and richness of traditional Hmong culture, in particular their shamanism and embroidered costume, has attracted the attention of the Australian public, but little is known about these people, their background or the struggles they have faced to adjust to a new life in Australia.This interdisciplinary collection of articles deals with their music and textiles, gender and language, their social adaptation and their global diaspora. The book aims to bring knowledge of the Hmong to a wider public and contribute to the understanding of these people.

    Description: The Representation of Science and Scientists on Postage Stamps examines how the postal authorities of the world have developed unique techniques to portray science and scientists in order to convey a message behind the stamp issue. It is a multi-disciplinary examination that investigates visual representation, semiotics, science, science history and politics, amongst other issues. The author introduces and describes a technique for looking at how and why images may have been chosen. Is the image a mirror reflecting a known reality or is it a lens to challenge, to prompt further thought or action? He also hypothesises that the Internet created a real change to science communication, influencing the way images of science were used. The stamp message changed from that of pursuing a public understanding approach to an awareness of science with its increasing contextual content.

    Description: Since September 11, 2001, our newspapers have been filled with the ‘war on terror’; our governments have mobilised their resources for ‘homeland security’; and people everywhere are braced for more terrorist attacks. Yet while the new threat is genuine, we must not lose sight of the continuing security concerns in the Asia-Pacific. Tensions persist on the Korean peninsula, in the Taiwan Straits and the South China Sea, and in Kashmir. The region is well supplied with weapons of mass destruction and may face an arms race, and there are a range of pressing human security issues. Likewise, the strategic realities of the region remain linked with US power, and with the emergence of China as a key regional player. The book examines the developing strategic relationships in the region, and clarifies the dilemmas for Australian policy-makers as they try to balance genuine engagement with the region against a long-standing and valued alliance with the United States. Emerging from discussions between the Shanghai Institute for International Studies and the University of New South Wales at ADFA, Asia-Pacific Security has a particular relevance for foreign-policy professionals and scholars of the region.

    Description: In the years between the two world wars of the twentieth century leaders in Western countries worried about a food surplus. The hardships of the Great Depression were intensified by a glut of wheat and consequent low prices on the world market. Yet at the same time nutrition scientists protested that significant proportions of populations, even in affluent countries, were unable to afford a diet ‘adequate for health’. Fresh fruits, vegetables, dairy products and meat were out of reach for the poor. This book traces the work of three men who sought to bring together the interests of farmers and the needs of the hungry: scientist and passionate campaigner for better nutrition, John Boyd Orr; Australian politician and international statesman, Stanley Melbourne Bruce; and Economic Adviser to Bruce at the Australian High Commission in London, Frank Lidgett McDougall. Bruce once said ‘McDougall brings me a new idea every morning’. One of those ideas became the genesis of their work, which helped bring about the formation of the Food and Agriculture Organization of the United Nations in 1945. All three undertook significant roles in the formative years of the organisation. The story of this contribution to the international world order is little known. The cooperation, diplomacy and persistence of these men provides inspiration for tackling the alarming prospect of food shortages in the present century.

    Description: In this book, we discuss the changing medical and public profile of fungal infections in the period 1850–2000. We consider four sets of diseases: ringworm and athlete’s foot (dermatophytosis); thrush or candidiasis (infection with Candida albicans); endemic, geographically specific infections in North America (coccidioidomycosis, blastomycosis and histoplasmosis) and mycotoxins; and aspergillosis (infection with Aspergillus fumigatus). We discuss each disease in relation to developing medical knowledge and practices, and to social changes associated with ‘modernity’. Thus, mass schooling provided ideal conditions for the spread of ringworm of the scalp in children, and the rise of college sports and improvement of personal hygiene led to the spread of athlete’s foot. Antibiotics seemed to open the body to more serious Candida infections, as did new methods to treat cancers and the development of transplantation. Regional fungal infections in North America came to the fore due to the economic development of certain regions, where population movement brought in non-immune groups who were vulnerable to endemic mycoses. Fungal toxins or mycotoxins were discovered as by-products of modern food storage and distribution technologies. Lastly, the rapid development and deployment of new medical technologies, such as intensive care and immunosuppression in the last quarter of the twentieth century, increased the incidence of aspergillosis and other systemic mycoses.

    Description: "The Australian National University’s Strategic & Defence Studies Centre (SDSC) is Australia’s premier university-based strategic studies think tank. Fifty years after the Centre was founded in 1966, SDSC celebrated its continued research, publications, teaching and government advisory role with a two-day conference entitled ‘New Directions in Strategic Thinking 2.0’. The event saw the podium graced by many of the world’s premier thinkers in the strategic studies field. An evening between those tours to the lectern brought together academics, practitioners and other honoured guests at a commemorative dinner held beneath the widespread wings of the ‘G for George’ bomber in the Australian War Memorial—an event that included SDSC’s own Professor Desmond Ball AO making his last public appearance. Since SDSC’s 25th anniversary, the world has seen the collapse of the Soviet Union and the end of the Cold War. Bipolarity gave way to the emergence of the United States as the world’s sole superpower, a status many now see as under threat.     Description: Paul Dibb AM has had an extraordinary career. He enjoys an international scholarly reputation of the highest order, while at the same time he has done much distinguished public service. He was a pioneer in moving back and forth between posts in government departments, notably the Department of Defence, and academia. He began as a student of Soviet economic geography, and then spent nearly two decades in Australian Defence intelligence, including service as Head of the National Assessments Staff (NAS) in the Joint Intelligence Organisation (JIO) from 1974 to 1978, Deputy Director of JIO in 1978–80, Director of JIO in 1986–88, and Deputy Secretary of Defence (Strategy and Intelligence) in 1988–91, before becoming a Professor in the Strategic and Defence Studies Centre (SDSC) at The Australian National University (where he is now an Emeritus Professor). He has been quite happy to engage in vigorous public debate about important and controversial strategic and defence issues, giving him a high public profile. The contributors include two former Chancellors of ANU, one a former Minister of Defence, and the other a former Secretary of the Department of Defence, a former Chief of the Defence Force (CDF), and other former senior officials, as well as academic specialists in geography, international relations, and strategic and defence studies. ‘This would be a high-quality set of essays for any edited volume, but for a festschrift – a genre that sometimes generates uneven collections – this is an exceptional assembly.     Description: Where the last three decades of the 20th century witnessed a China rising on to the global economic stage, the first three decades of the 21st century are almost certain to bring with them the completion of that rise, not only in economic, but also political and geopolitical terms. China’s integration into the global economy has brought one-fifth of the global population into the world trading system, which has increased global market potential and integration to an unprecedented level. The increased scale and depth of international specialisation propelled by an enlarged world market has offered new opportunities to boost world production, trade and consumption; with the potential for increasing the welfare of all the countries involved. However, China’s integration into the global economy has forced a worldwide reallocation of economic activities. This has increased various kinds of friction in China’s trading and political relations with others, as well as generating several globally significant externalities. Finding ways to accommodate China’s rise in a way that ensures the future stability and prosperity of the world economy and polity is probably the most important task facing the world community in the first half of the 21st century. The book delves into these issues to reflect upon the wide range of opportunities and challenges that have emerged in the context of a rising China.

    Description: Asia looks and feels very different now compared to the days of the Cold War. The sense that Asia now works differently can be traced to a single source – the re-emergence of China. China was the dominant power in greater Asia for most of recorded history. This historical norm was interrupted from the early 19th century, too far into the past to be recognisable and readily accommodated by the actors in today’s international arena. A powerful China feels new and unfamiliar. Arriving peacefully at mutually acceptable relationships of power and influence that are very different from those that have prevailed for the past half century will be a demanding process. The world’s track record on challenges of this kind is not terrific. It will call for statesmanship of a consistently high order from all the major players, and building the strongest possible confidence among these players that there are no hidden agendas.

    Description: Large amounts (estimates range from 70 Tg per year to 300 Tg per year) of the potent greenhouse gas methane are oxidized to carbon dioxide in marine sediments by communities of methanotrophic archaea and sulphate-reducing bacteria, and thus are prevented from escaping into the atmosphere. Indirect evidence indicates that the anaerobic oxidation of methane might proceed as the reverse of archaeal methanogenesis from carbon dioxide with the nickel-containing methyl-coenzyme M reductase (MCR) as the methane-activating enzyme. However, experiments showing that MCR can catalyse the endergonic back reaction have been lacking. Here we report that purified MCR from Methanothermobacter marburgensis converts methane into methyl-coenzyme M under equilibrium conditions with apparent V(max) (maximum rate) and K(m) (Michaelis constant) values consistent with the observed in vivo kinetics of the anaerobic oxidation of methane with sulphate. This result supports the hypothesis of 'reverse methanogenesis' and is paramount to understanding the still-unknown mechanism of the last step of methanogenesis.     Description: Humans have engaged in endurance running for millions of years, but the modern running shoe was not invented until the 1970s. For most of human evolutionary history, runners were either barefoot or wore minimal footwear such as sandals or moccasins with smaller heels and little cushioning relative to modern running shoes. We wondered how runners coped with the impact caused by the foot colliding with the ground before the invention of the modern shoe. Here we show that habitually barefoot endurance runners often land on the fore-foot (fore-foot strike) before bringing down the heel, but they sometimes land with a flat foot (mid-foot strike) or, less often, on the heel (rear-foot strike). In contrast, habitually shod runners mostly rear-foot strike, facilitated by the elevated and cushioned heel of the modern running shoe. Kinematic and kinetic analyses show that even on hard surfaces, barefoot runners who fore-foot strike generate smaller collision forces than shod rear-foot strikers. This difference results primarily from a more plantarflexed foot at landing and more ankle compliance during impact, decreasing the effective mass of the body that collides with the ground.     Description: Malaria is a devastating infection caused by protozoa of the genus Plasmodium. Drug resistance is widespread, no new chemical class of antimalarials has been introduced into clinical practice since 1996 and there is a recent rise of parasite strains with reduced sensitivity to the newest drugs. We screened nearly 2 million compounds in GlaxoSmithKline's chemical library for inhibitors of P. falciparum, of which 13,533 were confirmed to inhibit parasite growth by at least 80% at 2 microM concentration. More than 8,000 also showed potent activity against the multidrug resistant strain Dd2. Most (82%) compounds originate from internal company projects and are new to the malaria community. Analyses using historic assay data suggest several novel mechanisms of antimalarial action, such as inhibition of protein kinases and host-pathogen interaction related targets.     Description: Jasmonoyl-isoleucine (JA-Ile) is a plant hormone that regulates a broad array of plant defence and developmental processes. JA-Ile-responsive gene expression is regulated by the transcriptional activator MYC2 that interacts physically with the jasmonate ZIM-domain (JAZ) repressor proteins. On perception of JA-Ile, JAZ proteins are degraded and JA-Ile-dependent gene expression is activated. The molecular mechanisms by which JAZ proteins repress gene expression remain unknown. Here we show that the Arabidopsis JAZ proteins recruit the Groucho/Tup1-type co-repressor TOPLESS (TPL) and TPL-related proteins (TPRs) through a previously uncharacterized adaptor protein, designated Novel Interactor of JAZ (NINJA). NINJA acts as a transcriptional repressor whose activity is mediated by a functional TPL-binding EAR repression motif. Accordingly, both NINJA and TPL proteins function as negative regulators of jasmonate responses. Our results point to TPL proteins as general co-repressors that affect multiple signalling pathways through the interaction with specific adaptor proteins.     Description: Tumours with mutant BRAF are dependent on the RAF-MEK-ERK signalling pathway for their growth. We found that ATP-competitive RAF inhibitors inhibit ERK signalling in cells with mutant BRAF, but unexpectedly enhance signalling in cells with wild-type BRAF. Here we demonstrate the mechanistic basis for these findings. We used chemical genetic methods to show that drug-mediated transactivation of RAF dimers is responsible for paradoxical activation of the enzyme by inhibitors. Induction of ERK signalling requires direct binding of the drug to the ATP-binding site of one kinase of the dimer and is dependent on RAS activity. Drug binding to one member of RAF homodimers (CRAF-CRAF) or heterodimers (CRAF-BRAF) inhibits one protomer, but results in transactivation of the drug-free protomer. In BRAF(V600E) tumours, RAS is not activated, thus transactivation is minimal and ERK signalling is inhibited in cells exposed to RAF inhibitors. These results indicate that RAF inhibitors will be effective in tumours in which BRAF is mutated. Furthermore, because RAF inhibitors do not inhibit ERK signalling in other cells, the model predicts that they would have a higher therapeutic index and greater antitumour activity than mitogen-activated protein kinase (MEK) inhibitors, but could also cause toxicity due to MEK/ERK activation. These predictions have been borne out in a recent clinical trial of the RAF inhibitor PLX4032 (refs 4, 5). The model indicates that promotion of RAF dimerization by elevation of wild-type RAF expression or RAS activity could lead to drug resistance in mutant BRAF tumours.     Description: The evolution and maintenance of sexual reproduction has puzzled biologists for decades. Although this field is rich in hypotheses, experimental evidence is scarce. Some important experiments have demonstrated differences in evolutionary rates between sexual and asexual populations; other experiments have documented evolutionary changes in phenomena related to genetic mixing, such as recombination and selfing. However, direct experiments of the evolution of sex within populations are extremely rare (but see ref. 12). Here we use the rotifer, Brachionus calyciflorus, which is capable of both sexual and asexual reproduction, to test recent theory predicting that there is more opportunity for sex to evolve in spatially heterogeneous environments. Replicated experimental populations of rotifers were maintained in homogeneous environments, composed of either high- or low-quality food habitats, or in heterogeneous environments that consisted of a mix of the two habitats. For populations maintained in either type of homogeneous environment, the rate of sex evolves rapidly towards zero. In contrast, higher rates of sex evolve in populations experiencing spatially heterogeneous environments. The data indicate that the higher level of sex observed under heterogeneity is not due to sex being less costly or selection against sex being less efficient; rather sex is sufficiently advantageous in heterogeneous environments to overwhelm its inherent costs.     Description: Biodiversity indicators provide a vital window on the state of the planet, guiding policy development and management. The most widely adopted marine indicator is mean trophic level (MTL) from catches, intended to detect shifts from high-trophic-level predators to low-trophic-level invertebrates and plankton-feeders. This indicator underpins reported trends in human impacts, declining when predators collapse ("fishing down marine food webs") and when low-trophic-level fisheries expand ("fishing through marine food webs"). The assumption is that catch MTL measures changes in ecosystem MTL and biodiversity. Here we combine model predictions with global assessments of MTL from catches, trawl surveys and fisheries stock assessments and find that catch MTL does not reliably predict changes in marine ecosystems. Instead, catch MTL trends often diverge from ecosystem MTL trends obtained from surveys and assessments. In contrast to previous findings of rapid declines in catch MTL, we observe recent increases in catch, survey and assessment MTL. However, catches from most trophic levels are rising, which can intensify fishery collapses even when MTL trends are stable or increasing.     Description: Reprogramming of somatic cell nuclei to yield induced pluripotent stem (iPS) cells makes possible derivation of patient-specific stem cells for regenerative medicine. However, iPS cell generation is asynchronous and slow (2-3 weeks), the frequency is low (〈0.1%), and DNA demethylation constitutes a bottleneck. To determine regulatory mechanisms involved in reprogramming, we generated interspecies heterokaryons (fused mouse embryonic stem (ES) cells and human fibroblasts) that induce reprogramming synchronously, frequently and fast. Here we show that reprogramming towards pluripotency in single heterokaryons is initiated without cell division or DNA replication, rapidly (1 day) and efficiently (70%). Short interfering RNA (siRNA)-mediated knockdown showed that activation-induced cytidine deaminase (AID, also known as AICDA) is required for promoter demethylation and induction of OCT4 (also known as POU5F1) and NANOG gene expression. AID protein bound silent methylated OCT4 and NANOG promoters in fibroblasts, but not active demethylated promoters in ES cells.     Description: Eusociality, in which some individuals reduce their own lifetime reproductive potential to raise the offspring of others, underlies the most advanced forms of social organization and the ecologically dominant role of social insects and humans. For the past four decades kin selection theory, based on the concept of inclusive fitness, has been the major theoretical attempt to explain the evolution of eusociality. Here we show the limitations of this approach.     Description: Gram-negative bacteria, such as Escherichia coli, frequently use tripartite efflux complexes in the resistance-nodulation-cell division (RND) family to expel various toxic compounds from the cell. The efflux system CusCBA is responsible for extruding biocidal Cu(I) and Ag(I) ions. No previous structural information was available for the heavy-metal efflux (HME) subfamily of the RND efflux pumps. Here we describe the crystal structures of the inner-membrane transporter CusA in the absence and presence of bound Cu(I) or Ag(I). These CusA structures provide new structural information about the HME subfamily of RND efflux pumps. The structures suggest that the metal-binding sites, formed by a three-methionine cluster, are located within the cleft region of the periplasmic domain. This cleft is closed in the apo-CusA form but open in the CusA-Cu(I) and CusA-Ag(I) structures, which directly suggests a plausible pathway for ion export. Binding of Cu(I) and Ag(I) triggers significant conformational changes in both the periplasmic and transmembrane domains. The crystal structure indicates that CusA has, in addition to the three-methionine metal-binding site, four methionine pairs-three located in the transmembrane region and one in the periplasmic domain. Genetic analysis and transport assays suggest that CusA is capable of actively picking up metal ions from the cytosol, using these methionine pairs or clusters to bind and export metal ions.     Description: Sugar efflux transporters are essential for the maintenance of animal blood glucose levels, plant nectar production, and plant seed and pollen development. Despite broad biological importance, the identity of sugar efflux transporters has remained elusive. Using optical glucose sensors, we identified a new class of sugar transporters, named SWEETs, and show that at least six out of seventeen Arabidopsis, two out of over twenty rice and two out of seven homologues in Caenorhabditis elegans, and the single copy human protein, mediate glucose transport. Arabidopsis SWEET8 is essential for pollen viability, and the rice homologues SWEET11 and SWEET14 are specifically exploited by bacterial pathogens for virulence by means of direct binding of a bacterial effector to the SWEET promoter. Bacterial symbionts and fungal and bacterial pathogens induce the expression of different SWEET genes, indicating that the sugar efflux function of SWEET transporters is probably targeted by pathogens and symbionts for nutritional gain.     Description: Messenger RNA lacking stop codons ('non-stop mRNA') can arise from errors in gene expression, and encode aberrant proteins whose accumulation could be deleterious to cellular function. In bacteria, these 'non-stop proteins' become co-translationally tagged with a peptide encoded by ssrA/tmRNA (transfer-messenger RNA), which signals their degradation by energy-dependent proteases. How eukaryotic cells eliminate non-stop proteins has remained unknown. Here we show that the Saccharomyces cerevisiae Ltn1 RING-domain-type E3 ubiquitin ligase acts in the quality control of non-stop proteins, in a process that is mechanistically distinct but conceptually analogous to that performed by ssrA: Ltn1 is predominantly associated with ribosomes, and it marks nascent non-stop proteins with ubiquitin to signal their proteasomal degradation. Ltn1-mediated ubiquitylation of non-stop proteins seems to be triggered by their stalling in ribosomes on translation through the poly(A) tail. The biological relevance of this process is underscored by the finding that loss of Ltn1 function confers sensitivity to stress caused by increased non-stop protein production.     Description: Interconversion between conductive and non-conductive forms of the K(+) channel selectivity filter underlies a variety of gating events, from flicker transitions (at the microsecond timescale) to C-type inactivation (millisecond to second timescale). Here we report the crystal structure of the Streptomyces lividans K(+) channel KcsA in its open-inactivated conformation and investigate the mechanism of C-type inactivation gating at the selectivity filter from channels 'trapped' in a series of partially open conformations. Five conformer classes were identified with openings ranging from 12 A in closed KcsA (Calpha-Calpha distances at Thr 112) to 32 A when fully open. They revealed a remarkable correlation between the degree of gate opening and the conformation and ion occupancy of the selectivity filter. We show that a gradual filter backbone reorientation leads first to a loss of the S2 ion binding site and a subsequent loss of the S3 binding site, presumably abrogating ion conduction.     Description: Sex determination is a fundamental biological process, yet its mechanisms are remarkably diverse. In vertebrates, sex can be determined by inherited genetic factors or by the temperature experienced during embryonic development. However, the evolutionary causes of this diversity remain unknown. Here we show that live-bearing lizards at different climatic extremes of the species' distribution differ in their sex-determining mechanisms, with temperature-dependent sex determination in lowlands and genotypic sex determination in highlands. A theoretical model parameterized with field data accurately predicts this divergence in sex-determining systems and the consequence thereof for variation in cohort sex ratios among years. Furthermore, we show that divergent natural selection on sex determination across altitudes is caused by climatic effects on lizard life history and variation in the magnitude of between-year temperature fluctuations.
