    Notizen: Abstract A model is proposed describing the mechanical evolution of a shear zone along compressional and extensional plate boundaries, subject to constant strain rate. The shear zones are assumed as viscoelastic with Maxwell rheology and with elastic and rheological parameters depending on temperature and petrology. Stress and strain are computed as functions of time and depth. For both kinds of boundaries the model reproduces the existence of a shallow seismogenic zone, characterized by a stress concentration. The thickness of the seismogenic layer is evaluated considering the variations of shear stress and frictional strength on faults embedded in the shear zone. Assuming that a fault dislocation takes place, the brittle-ductile transition is assumed to occur at the depth at which the time derivative of total shear stress changes from positive to negative values. The effects of different strain rates and geothermal gradients on the depth of the brittle-ductile transition are studied. The model predictions are consistent with values inferred from seismicity data of different boundary zones.

    Beschreibung: The Zagros thrust belt is a zone of deformed crustal rocks well exposed along the southwest region of Iran. To obtain a better knowledge of this mountain chain, we elaborated a 2D model reproducing the thermal structure of the “Mountain Front Fault”. This study, which is focused on the Lurestan region, is based on a model made by merging published sections and available information on the depth of the Moho. We present the isotherms and the geotherms calculated using an analytical methodology. The calculation procedure includes the temperature variation due to the re-equilibrated conductive state after thrusting, frictional heating, heat flow density data, and a series of geologically derived constraints. In order to perform the temperature calculations, the crustal structure in the Lurestan region is simplified as composed of two domains: A lower unit made by crystalline basement and an upper unit including all the lithostratigraphic units forming the sedimentary cover. The resulting model is compared with the numerical results obtained by previous studies to improve the description of the thermal structure of this geologically important area.

    Beschreibung: In this study, we elaborated a 2D model that reproduces the thermal structure of the central-northern Adriatic offshore and adjacent onshore area of the Italian peninsula. Based on the crustal structure along the trace of the CROP-03 deep section, the geotherms offshore Gabicce (northern Marche region) were obtained by an analytical procedure taking into account the role of thrusting within the sedimentary cover. Basement involvement at depth beneath the neighbouring Mondaino area to the SW, where a crustal thrust ramp dips towards the hinterland, required the use of a different analytical procedure. The results obtained in this study allowed us to define a satisfactory description of the thermal state of the northern Marche coastal area and adjacent Adriatic offshore. These results, integrated with those obtained by previous studies, confirm that the isotherms of 250°C and 400°C are placed in the stable Adriatic lithosphere at depths of about 11 km and 22 km, respectively. Furthermore, the 400°C isotherm is deeper in the onshore area, reaching a depth of about 30 km in the zone comprised between Gabicce and Mondaino, whereas the 250°C isotherm deepens towards the SW along the Adriatic Sea sector, to reach a maximum depth of 13 km in coastal area, rising again at a depth of 11 km in the innermost sector of the studied section.

    Beschreibung: The data here reported introduce the wound-healing assay as a tool for testing probiotics aimed at protecting gastrointestinal mucosal surfaces and to verify the consistency of their manufacturing. At the scope, we compared the in vitro effects of two multi-strain high concentration formulations both commercialized under the same brand VSL#3 but sourced from different production sites (USA and Italy) on a non-transformed small-intestinal epithelial cell line, IEC-6. The effects on cellular morphology, viability, migration, and H 2 O 2 -induced damage, were assessed before and after the treatment with both VSL#3 formulations. While the USA-sourced product (“USA-made”) VSL#3 did not affect monolayer morphology and cellular density, the addition of bacteria from the Italy-derived product (“Italy-made”) VSL#3 caused clear morphological cell damage and strongly reduced cellularity. The treatment with “USA-made” lysate led to a higher rate of wounded monolayer healing, while the addition of “Italy-made” bacterial lysate did not influence the closure rate as compared to untreated cells. While lysates from “USA-made” VSL#3 clearly enhanced the formation of elongated and aligned stress fibers, “Italy-made” lysates had not similar effect. “USA-made” lysate was able to cause a total inhibition of H 2 O 2 -induced cytotoxic effect whereas “Italy-made” VSL#3 lysate was unable to protect IEC-6 cells from H 2 O 2 -induced damage. ROS generation was also differently influenced, thus supporting the hypotesis of a protective action of “USA-made” VSL#3 lysates, as well as the idea that “Italy-made” formulation was unable to prevent significantly the H 2 O 2 -induced oxidative stress.     Beschreibung: The Zagros thrust belt is a large orogenic zone located along the southwest region of Iran. To obtain a better knowledge of this important mountain chain, we elaborated the first 3-D model reproducing the thermal structure of its northwestern part, i.e., the Lurestan arc. This study is based on a 3-D structural model obtained using published geological sections and available information on the depth of the Moho discontinuity. The analytical calculation procedure took into account the temperature variation due to: (1) The re-equilibrated conductive state after thrusting, (2) frictional heating, (3) heat flow density data, and (4) a series of geologically derived constraints. Both geotherms and isotherms were obtained using this analytical methodology. The results pointed out the fundamental control exerted by the main basement fault of the region, i.e., the Main Frontal Thrust (MFT), in governing the thermal structure of the crust, the main parameter being represented by the amount of basement thickening produced by thrusting. This is manifested by more densely spaced isotherms moving from the southwestern foreland toward the inner parts of orogen, as well as in a lateral variation related with an along-strike change from a moderately dipping crustal ramp of the MFT to the NW to a gently dipping crustal ramp to the SE. The complex structural architecture, largely associated with late-stage (Pliocene) thick-skinned thrusting, results in a zone of relatively high geothermal gradient in the easternmost part of the study area. Our thermal model of a large crustal volume, besides providing new insights into the geodynamic processes affecting a major salient of the Zagros thrust belt, may have important implications for seismotectonic analysis in an area recently affected by a Mw = 7.3 earthquake, as well as for geothermal/hydrocarbon exploration in the highly perspective Lurestan region.

    Beschreibung: The migration capability of dendritic cells (DCs) is regulated by their response to factors, namely chemokines, that characterize maturation stage and shape their functional activities. This study examines the morphology, expression of chemokines/chemokine receptors, and migration properties of DCs generated after treatment of monocytes with type I interferon (IFN) and granulocyte-macrophage colony-stimulating factor (GM-CSF) (IFN-DCs). IFN-DCs showed phenotypical and morphologic features undetectable in DCs generated in the presence of interleukin 4 (IL-4) and GM-CSF, such as expression of CD83 and CD25 and the presence of CD44+, highly polarized, thin, and long dendrites. IFN-DCs markedly migrated in response to β-chemokines (especially MIP-1β) and expressed the Th-1 chemokine IP-10. Notably, IFN-DCs showed an up-regulation of CCR7 as well as of its natural ligand MIP-3β, characteristics typical of mature DCs. Of interest, IFN-DCs exhibited a marked chemotactic response to MIP-3β in vitro and strong migratory behavior in severe combined immunodeficient (SCID) mice. In SCID mice reconstituted with human peripheral blood leukocytes, IFN-DCs induced a potent primary human antibody response and IFN-γ production, indicative of a Th-1 immune response. These results define the highly specialized maturation state of IFN-DCs and point out the existence of a “natural alliance” between type I IFN and monocyte/DC development, instrumental for ensuring an efficient connection between innate and adaptive immunity.

    Beschreibung: Purpose: We have developed a novel DC population differentiated from human monocytes in the presence of GM-CSF and IFN-alpha (IFN-DC), which is highly efficient in internalizing tumour cell antigens and in the cross-priming of CD8+T cells, promoting efficient anti-lymphoma response. Preclinical results prompted us to start a clinical trial of IFN-DC combined with low-dose Rituximab(RTX) in patients with Follicular lymphoma (FL), which is an indolent, immune-responsive, but still incurable cancer. Patients and Methods: In October 2014 we started a phase I clinical trial (EudraCT: 2013-003158-25) testing safety, immunogenicity and systemic clinical responses of low-dose RTX combined with autologous IFN-DC, administered by intra-nodal injections. Patients features: age 18-75y, indolent FL, relapsed or refractory (R/R), stage III-IV low tumour burden, with at least 1 superficial pathologic lymph node. One leukapheresis was scheduled at study entry for harvesting mononuclear cells. Eight cycles of RTX (5-20mg) and IFN-DC (30±10x106) were planned: the first 4 every two weeks, followed by other 4 cycles administered monthly. All injections were delivered with the aid of an ultrasound guide. Blood test for immune-responses and autoimmunity markers were scheduled at 0,+72d, +132 days ; +9; +18;+24 months. Results: as of the 31st July 2018, 8 patients have completed treatment and have been evaluated for response (median follow-up=24 months; range=8-45 mo). Median age of the study population was 60 years (range 27-76). All subjects had disseminated disease and enlarged lymph nodes or lesions greater than 1.5 cm at multiple sites available for local injections and monitoring for distant response. All patients were R/R after previous systemic therapy (median lines=2, range1-4) and 3/8 (37.5%) after auto-transplantation. The treatment was very well tolerated and manageable. No relevant adverse events were observed. Four out of 8 (50%) patients had objective response (OR) with 3/8(37.5%) attaining complete remission (CR); 2/8 (25%) are in stable disease at +8 and +25 months; 2/8 (25%), who did not achieve OR, progressed at +8 and +14 respectively. One patient progressed 4 months after achieving CR (Figure1). In all patients who achieved OR or who are in stable disease, induction of a tumour specific immune response was observed. Conclusions: local RTX and IFN-DC combined therapy was effective in eliciting in 50% of R/R FL with low tumour burden systemic and durable responses, which were associated with induction of specific anti-FL T cell responses.     Beschreibung: Cross-presentation allows antigen-presenting cells to present exogenous antigens to CD8+ T cells, playing an essential role in controlling infections and tumor development. IFN-α induces the rapid differentiation of human mono-cytes into dendritic cells, known as IFN-DCs, highly efficient in mediating cross-presentation, as well as the cross-priming of CD8+ T cells. Here, we have investigated the mechanisms underlying the cross-presentation ability of IFN-DCs by studying the intracellular sorting of soluble ovalbumin and nonstructural-3 protein of hepatitis C virus. Our results demonstrate that, independently from the route and mechanism of antigen entry, IFN-DCs are extraordinarily competent in preserving internalized proteins from early degradation and in routing antigens toward the MHC class-I processing pathway, allowing long-lasting, cross-priming capacity. In IFN-DCs, both early and recycling endosomes function as key compartments for the storage of both antigens and MHC-class I molecules and for proteasome- and transporter-associated with Ag processing–dependent auxiliary cross-presentation pathways. Because IFN-DCs closely resemble human DCs naturally occurring in vivo in response to infections and other danger signals, these findings may have important implications for the design of vaccination strategies in neoplastic or chronic infectious diseases.
