I.1). - Phosphorus Analogs of α-Amino Acids and Peptides, VII2). - Synthesis of Methyl (Ethyl) Amino(diethoxyphosphoryl)acetate, 2-Phosphonoglycine, and its Derivatives from Diethyl (Isocyanomethyl)phosphonateAfter metalation with potassium tert-butoxide diethyl (isocyanomethyl)phosphonate (5a) yields with alkyl chloroformates or carbamoyl chlorides (type 6) or methyl isocyanate (10) the diethyl 4-oxazolylphosphonates 9 containing a masked carboxy group at C-5. These on hydrolysis yield the title compounds 2 and 1 as well as compounds 3. Compound 5b reacts with di-tert-butyl dicarbonate to the tert-butyl (diethoxyphosphoryl)isocyanoacetate 5c which can be hydrolyzed to the compounds 4. - On metalation of 9b (sodium hydride in dichloromethane) the Wittig-Horner reagent 13 which gives ethyl (Z)-3-aryl(heteroaryl)-2-isocyanoacrylates 15 with aromatic (heteroaromatic) aldehydes 14 is formed.
    Notes: (Isocyanmethyl)phosphonsäure-diethylester (5a) liefert nach Metallieren mit Kalium-tert-butylat mit Chlorameisensäureestern oder Carbamoylchloriden (Typ 6) bzw. mit Methylisocyanat (10) die 4-Oxazolylphosphonsäure-diethylester 9 mit einer maskierten Carboxylfunktion an C-5. Deren Hydrolyse liefert die Titelverbindungen 2 und 1 sowie die Phosphonsäurehalbester 3. Mit Ditert-butyldicarbonat reagiert 5b zum tert-Butylester 5c, der zu den Verbindungen 4 hydrolysierbar ist. - Durch Metallieren mit Natriumhydrid in Methylendichlorid erhält man aus 9b das Wittig-Horner-Reagens 13, das mit aromatischen (heteroaromatischen) Aldehyden 14 die (Z)-3-Aryl(Heteroaryl-)-2-isocyanacrylsäure-ethylester 15 ergibt.
