Syntheses of Bile Pigments, III1). - The Total Synthesis of Mesobilirhodin Dimethyl EsterMesobilirhodin (1) which is probably formed together with isomesobilirhodin (2) either by rearrangement of urobilin IXα or by oxidation of urobilinogen has been synthesised for the first time. The key reaction of the synthesis consists in the cyclisation of the δ-(pyrrol-2-yl)- laevulinic acid derivative 11 to give the corresponding 2,3-dihydro-5(1 H)-pyrromethenone 13. Compounds of this type have so far been inaccessible by means of the usual synthetic methods for bile pigments. Starting from the mixture of diastereomeric 11 two products are obtained (namely rac. erythro- and rac. threo- 13) which can be separated chromatographically. Their condensation reaction with the methyl ester of 5′-formylisoneobilirubinic acid, for which an improved synthesis is given, yields rac. erythro- and rac. threo-mesobilirhodin, respectively (rac. 1a and rac. 1b, resp.). Electronic and mass spectra of both diastereomers agree with the published data. The NMR data would indicate, however, that the product obtained from the oxidation of urobilinogen has the constitution 2 (i. e. isomesobilirhodin), or consists of a mixture of both 1 and 2.

Mesobilirhodin (1), das vermutlich neben Isomesobilirhodin (2) sowohl durch Umlagerung des Urobilins IXα als auch bei der Oxidation von Urobilinogen entsteht, ist erstmalig totalsynthetisch dargestellt worden. Schlüsselreaktion ist dabei die Cyclisierung des δ-(Pyrrol-2-yl)- lävulinsäure-Derivats 11 zum 2,3-Dihydro-5(1 H)-pyrromethenon 13. Verbindungen des Typs 13 waren bisher mit Hilfe der herkömmlichen Methoden zur Synthese von Gallenfarbstoffen nicht zugänglich. Aus dem Diastereomeren-Gemisch von 11 werden zwei Reaktionsprodukte erhalten (racem. erythro- und racem. threo-13), die sich chromatographisch voneinander trennen lassen. Ihre Kondensation mit 5′-Formylisoneobilirubinsäure-methylester (17), dessen Darstellung gegenüber der Literaturvorschrift verbessert wurde, liefert racem. erythro- bzw. racem. threo-Mesobilirhodin (racem. 1a bzw. racem. 1b), deren elektronen- und massenspektroskopische Daten mit den in der Literatur angegebenen Werten übereinstimmen. Das 1H-NMR-Spektrum deutet jedoch darauf hin, daß es sich zumindest beim Oxidationsprodukt des Urobilinogens um das Isomesobilirhodin 2 oder um ein Gemisch von 1 und 2 handeln konnte.
