Note on an Improved Chromatographic Separation of Oligonucleotides**The separation of large amounts of oligonucleotides in a stepwise synthesis can be achieved by single-column chromatography by: appropriate choice of alcohols or mixtures of alcohols (methanol/isopropyl alcohol), use of ionic strength gradients of appropriate steepness, and maintaining the ionic strength constant as a substance elutes. It is only important to know the sequence-specific properties of an oligonucleotide chain, which, to a first approximation, can be taken to be the sum of the properties of the constituent monomers.

Die Trennung größerer Mengen von Oligonucleotiden bei schrittweiser Synthese ist säulen-chromatographisch in einem Durchlauf möglich durch Verwendung von geeigneten Alkoholen oder Alkoholgemischen (Methanol/Isopropylalkohol), durch Verwendung von Ionenstärkegradienten geeigneter Steilheit und durch die Technik, den Ionenstärkegradienten anzuhalten, wenn eine Verbindung eluiert wird. Erforderlich ist nur die Kenntnis der sequenzspezifischen Eigenschaften einer Oligonucleotidkette, die sich in erster Näherung als eine Summe der Eigenschaften der sie zusammensetzenden Monomeren verstehen lassen.
