Die Methylierung von 4.5-Diamino-uracil mit Dimethylsulfat führt zu 4.5-Di-methylamino-1.3-dimethyl-uracil, welches mit Essigsäureanhydrid in 1.3.7.8-Tetramethyl-xanthin, mit Formamid in Coffein übergeht. Aus 4.5-Diamino-1.3-dimethyl-uracil bzw. seiner Monoacetyl-Verbindung entsteht mit verd. Schwefelsäure ein Gemisch von 1.3.1′3′-Tetramethyl-bis-alloxazin und 1.3.1′.3′-Tetramethyl-hydurilsäure. Mit 4.5-Diamino-3-methyl-uracil bzw. seiner Monoacetyl-Verbindung bilden sich entsprechend 3.3′-Dimethyl-bisalloxazin und 3.3′-Dimethyl-hydurilsäure.
