A method for the quantification of phenformin, an oral hypoglycemic biguanide, from biological fluids based upon gas chromatography and chemical ionization mass spectrometry, is reported. Phenformin is quantitatively cyclized to the 2-substituted-4-trifluoremethyl-2,6-diamino-1,3,5-S-triazine when acylated with trifluoroacetic anhydride. This cyclized product was measured with respect to the deuterium substituted phenformin. The plasma levels achieved following the administration of a therapeutic dose of phenformin to a diabetic subject are reported.
