    Notizen: The mixed culture of Lactobacillus plantarum and Propionibacterium shermanii grown anaerobically in glucose minimal medium exhibits features typical of a commensal interaction even though a number of complicating factors, such as a large maintenance requirement of L. plantarum and inhibition of growth of P. shermanii at low pH, are present. A simple mathematical model of the system is presented and is shown to reproduce rather well some of the features of the continuous mixed culture system in both steady-state and transient situations.

    Notizen: The potential of sand as a support for immobilized enzymes was investigated by preparing alkylamine sand and devising methods to measure the total number of amine groups present and the fraction available for immobilization of enzymes. Alcohol dehydrogenase (alcohol:NAD oxidoreductase, EC 1.1.1.1) and lactate dehydrogenase (L-lactate: NAD oxidoreductase, EC 1.1.1.27) were immobilized on alkylamine sand, and the stability of the immobilized protein and dehydrogenase activity was measured. Urease (urea amidohyrdrolase, EC 3.5.1.5) was also immobilized on sand to test the applicability of these methods to larger scale immobilizations. Results suggest that sand shows promise as a support for immobilized enzymes.

    Notizen: Protein solubility can be adequately represented by the classical Cohn equation for the salting-out of alcohol dehydrogenase and fumarase from clarified yeast homogenate with ammonium sulfate. However, the constant β in this equation is a function of the contacting procedure employed. The kinetics of continuous salting-out were similar for alcohol dehydrogenase and fumarase. The overall rate equation for precipitation had a variable order which was high initially, up to 3.1, but approached unity on completion of precipitation. This was followed by a partial resolution stage which was first order with respect to the concentration driving force. Precipitate particle size was estimated as 0.5 to 5 μm with continuous flow precipitation producing the largest particles.

    Notizen: The production of cholesterol oxidase by 3 liter batch cultures of Nocardia rhodocrous growing on a glycerol/yeast extract medium was investigated. Cholesterol was shown to be a good inducer of the enzyme. The optimum time for cholesterol addition and the quantity to be added were determined, resulting in a 15-fold yield increase. Cholesterol oxidase synthesis was influenced by the dissolved oxygen tension. Maximum cholesterol oxidase production was obtained at 30-40% air saturation. The effect of growth conditions on the extraction of cholesterol oxidase by Triton X-100 was investigated. The scale-up of the fermentation to 800 liters in a pilot-plant fermenter is described.

    Notizen: Using whole cells containing glucose isomerase, mathematical models for the enzymatic conversion of D-glucose to D-fructose and for the inactivation of the enzyme catalyst have been postulated and verified experimentally. The heat of reaction, the equilibrium constant, and the individual rate constants and their activation energies have been estimated. The model can be used to predict the time course for the enzymatic production of fructose in a batch reactor within the tested experimental range of 40-80°C.

    Notizen: The authors studied the influence of periodic variations (one cycle per 24 hr) of the feeding mash concentration of a continuous anaerobic culture of Saccharomyces cerevisiae in sugarcane molasses media at constant dilution rate. The yield of cells and their susceptibility to infection by FMD virus are equivalent to conventional Roux monolayer systems. The potential for the commercial application of the DEAE Sephadex A50 system is discussed in relation to other unit process monolayer systems and in particular to the system in which cells are cultured in a deep bed of small glass spheres.

    Notizen: Urease from Jack bean was immobilized on nonporous glass beads by covalent bonding and its kinetics were studied in a packed-column differential reactor. To facilitate comparison, the urease was immobilized by both diazo and glutaraldehyde coupling. The kinetic properties of immobilized urease were similar to those of the soluble enzyme and different immobilization methods did not appreciably alter the kinetic properties. The affects of three different amino acid activators appear to follow predictions obtained from a relatively simple competitive model, except at very low substrate levels.

    Notizen: Glucose oxidase from Aspergillus niger was immobilized on nonporous glass beads by covalent bonding and its kinetics were studied in a packed-column recycle reactor. The optimum pH of the immobilized enzyme was the same as that of soluble enzyme; however, immobilized glucose oxidase showed a sharper pH-activity profile than that of the soluble enzyme. The kinetic behavior of immobilized glucose oxidase at optimum pH and 25°C was similar to that of the soluble enzyme, but the immobilized material showed increased temperature sensitivity. Immobilized glucose oxidase showed no loss in activity on storage at 4°C for nearly ten weeks. On continuous use for 60 hr, the immobilized enzyme showed about a 40% loss in activity but no change in the kinetic constant.

    Notizen: In previous reports from this laboratory it has been shown that the extended aeration process for biological treatment of organically laden municipal and/or industrial waste could be successfully employed for concurrent purification and sludge disposal. Also results using a modified process in which autodigestion was aided and controlled by periodic partial hydrolysis of small portions of the recycle sludge showed that operational control was feasible. There was some question regarding the success of such a process if the original waste contained a large portion of inorganic solids. Accordingly, a 1½ year pilot plant study was made using a waste (hydrolyzed trickling filter sludge) of exceptionally high ash content (50-60%). It was found that the ash content of activated sludge grown on this substrate did not continually increase nor did the high ash content of the waste interfere in any way with the efficiency of removal of organic matter. In general it exceeded 90 percent. Also a highly nitrified effluent was produced. A variety of analyses were performed: COD, BOD, TOC, suspended solids, NH3-N, organic-N, NO3-N, etc. Interrelationships between these important monitoring parameters for assessing plant performance offered useful insight into operational control for hydrolytically assisted extended aeration processes.

    Notizen: A model is proposed that accounts for the decreases in yield which occur in chemostat cultures of mesophilic yeasts at superoptimal growth temperatures. Two yield depressing effects were identified, one due to increased maintenance requirements by the viable fraction of the population, the other due to energy substrate dissipation by the nonviable fraction. The two effects are functions of the dilution rate, as is the fraction of nonviable cells. Experimental results were obtained on the yield, maintenance, and dissipation of energy substrate in a glucose-limited chemostat culture of a respiration-deficient mutant of Saccharomyces cerevisiae at 39°C. The rates of glucose utilization for maintenance and for dissipation constituted, respectively, 33-28% and 15-9% of the total glucose utilization rate over the range of dilution rates tested (0.038-0.064 hr-1), while the yield varied over this range from 0.066-0.085 g of biomass (dry wt) per gram of glucose.

    Notizen: The models of Monod and Williams, for the growth of unicellular organisms in chemostats, give strongly damped transients in the biomass and cell number when the flow rate of the chemostat is changed. A simple trick is used to incorporate time delay in these models while still allowing a conventional stability analysis. For long enough time delays the equilibrium point is unstable and limit cycles can be computed. Results obtained using Williams' model, with weakly damped transients as a result of using moderately long time delay, are compared with his data in which cell numbers show weak damping but biomass shows strong damping.

    Notizen: An efficient method to grow Escherichia coli W to high cell concentrations on the pilot scale is described and discussed. The method involves growth linked introduction of glucose; and ammonia to the culture, sparing with oxygen, and maintenance of aerobic conditions by gradually decreasing the temperature in the culture in order to keep the oxygen demand within the limits of the capacity of supply. Under these conditions the linear rate of cell mass production is actually the result of exponential growth with a gradually decreasing growth-rate constant.About 10 kg packed cells were produced in a 50 liter working-volume fermentor in one run of 13 hr. The concentration of the cells at the end of the growth was about 47 g dry cells/liter. The expenditure for nutrients was minimal and the controls were of simple automatic nature. From the determined yield constants for glucose, nitrogen, phosphorus, and oxygen it may be inferred that the cells grown by this method are similar to those grown exponentially at constant temperature.

    Notizen: An analytical expression for the rate efficiency factor of planar localized enzyme systems is derived. The derivation takes into account the isothermal kinetic effect under the externally imposed perturbation of combined electrostatic and high frequency time-varying fields. The contribution of each individual field to the enzyme reaction is examined through the basic mechanism in which charged substrates interact, with the specific perturbing field. The interaction mechanisms for the electrostatic and for the time-varying fields are found to be different. This difference regulates the different manners in which enzymatic reaction rates are altered. Enzymatic reactions under electrostatic perturbation can be retarded or enhanced depending on the field polarization. At sufficiently high field intensities the reaction rate may approach zero or approach a maximum value equal to the turnover number of the enzyme. Time-varying field perturbations, on the other hand, always enhance the enzymatic reactions if bunching effects are negligible. At sufficiently high field intensities, the reaction may approach a value equal to that of the free enzyme system. Several typical numerical examples on pure electrostatic field perturbations, pure time-varying field perturbations, and combined field perturbations are also presented.

    Notizen: The pressure required for initiation of flow when freeze-pressing with the X-press is related to the phase boundaries of water, particularly those between ice I and liquid even at temperatures around -25°C and lower. Widening the orifice of the pressure chamber to diameters larger than 2.5 mm leads to lower pressures and less extensive cell disintegration.Pressing Saccharomyces cerevisiae slowly with the aid of a manual hydraulic jack at -25°C produces a disintegration of 60-75% irrespective of cell concentration. Pressing at -35°C shows no clear differences.Pressing more rapidly with the aid of a motor-driven hydraulic press produces a similar extent of disruption of diluted cell suspensions (5.4 mg/g) as slow pressing. However, freeze-pressing a paste of baker's yeast (270 mg/g) increases the degree of disintegration. Under these conditions the disintegration is further enhanced by a lower temperature, -35°C, and by a high velocity of flow through the orifice, such that more than 95% of the S. cerevisiae is disrupted by one pressing at less than 2 × 108 Pa.Mechanisms for flow through the X-press are suggested and discussed in relation to the phase diagram of water.

    Notizen: The characteristic flavor of hard Italian cheeses is associated with the presence of fatty acids, particularly butyric acid, liberated from milk fat during the ripening process. To ensure proper development and control of flavor, animal pregastric esterases or lipases are routinely added to the milk before coagulation of the curd. Such esterases are also used to generate flavor in enzyme modified cheese and other dairy products.Esterases from microbial sources have been investigated as agents to enhance flavor in cheese. We have found that an esterase from Mucor miehei exhibits the type of lipolytic activity needed for this application. Romano and fontina cheeses of excellent quality have been prepared by the use of this esterase. It has also been used successfully in the preparation of enzyme modified cheese, and, in turn, processed American cheese.

    Notizen: This study illustrates approaches using botanical proteinases and a pancreatic lipase to improve the palatability of two types of food. A third example is presented to illustrate the use of a chelating agent in growing corn to increase the sugar content by over 50%, presumably by influencing enzyme activity in the corncob itself. The purpose is to show that one is not limited to using only the microbial approach for producing flavors but to point out the broader concept of food palatability which includes texture and flavor.

    Notizen: There are four main sources of enzymes in foods - these being the inherent enzymes, enzymes from microbial contaminants, enzymes elaborated by microorganisms added to foods, and specific enzymes added to foods. This study primarily deals with the latter two sources of enzymes in food. Although both plants and animals serve as sources of enzymes, they are not as economical or versatile sources as are enzymes obtained from microorganisms. In the meat industry, proteases are used to tenderize muscle and to obtain flavor precursors. In the preparation of cured meat products such as sausages, lipases, and proteases from bacterial cultures are utilized. Similarly, proteases and lipases are used in the dairy industry to develop flavor compounds. Proteases and amylases also have applications in the baking and milling industries where they are used to produce precursors for the nonenzymatic browning reactions. Carbohydrases such as amylase, amyloglucosidase, and glucose isomerase have found usage in the starch and syrup industry for the production of high dextrose and high fructose syrups. Other enzymes such as glucose oxidase, pectinase, and naringinase are of value to the wine and fruit juice industries. A better understanding of the mode of action of enzymes as well as the mechanisms of development of flavor compounds will further enhance the use of microbial enzymes to develop specific and desired flavors in foods.

    Notizen: The development of the unique flavor of blue type cheese depends on the concerted action of numerous enzymes of Penicillium roqueforti involved in protein and lipid metabolism. Protease(s) by degrading casein modify the texture and background flavor of the ripening cheese. Lipase by hydrolyzing milk triglycerides provides flavorful fatty acids and precursors of methyl ketones. The enzyme complex involved in the partial oxidation of free fatty acids and the properties of β-ketoacyl decarboxylase which generates the major flavor components of blue cheese are discussed. Fermentation of P. roqueforti for the rapid production of methyl ketones is briefly reviewed.

    Notizen: Trypsin was coupled on an agarose gel which was modified with a spiropyran compound. The trypsin-spiropyran (agarose) gel showed reverse photochromism. The activity of the trypsin-spiropyran gel in the dark was 12% of that of native trypsin, and it was higher than that under visible light. The apparent Michaelis constant of the trypsin-spiropyran gel in the dark was larger than that under visible light. On the other hand, the maximum velocity in the dark was higher than that under visible light. The optimum pH of the trypsin-spiropyran gel in the dark was the same as that under visible light. Immobilized trypsin was stable in the pH range from 3 to 9. The trypsin-spiropyran gel was more stable against heat than the native trypsin.

    Notizen: Cell recycle and vacuum fermentation systems were developed for continuous ethanol production. Cell recycle was employed in both atmospheric pressure and vacuum fermentations to achieve high cell densities and rapid ethanol fermentation rates. Studies were conducted with Saccharomyces cerevisiae (ATCC No. 4126) at a fermentation temperature of 35°C. Employing a 10% glucose feed, a cell density of 50 g dry wt/liter was obtained in atmospheric-cell recycle fermentations which produced a fermentor ethanol productivity of 29.0 g/liter-hr. The vacuum fermentor eliminated ethanol inhibition by boiling away ethanol from the fermenting beer as it was formed. This permitted the rapid and complete fermentation of concentrated sugar solutions. At a total pressure of 50 mmHg and using a 33.4% glucose feed, ethanol productivities of 82 and 40 g/liter-hr were achieved with the vacuum system with and without cell recycle, respectively. Fermentor ethanol productivities were thus increased as much as twelvefold over conventional continuous fermentations. In order to maintain a viable yeast culture in the vacuum fermentor, a bleed of fermented broth had to be continuously withdrawn to remove nonvolatile compounds. It was also necessary to sparge the vacuum fermentor with pure oxygen to satisfy the trace oxygen requirement of the fermenting yeast.

The immobilized cellulase was more stable than the native cellulase. The substrate cellulose was hydrolyzed quantitatively with immobilized cellulase. The final reaction product was identified as glucose. Immobilized cellulase was used in a fluidized bed reactor where the pressure drop of the fluidized bed reactor was low and constant. Cellulose was hydrolyzed to glucose by the cellulase-bead fluidized bed reactor. The minimum flow velocity (Umf) was 0.5 cm/sec and the optimum flow velocity of the cellulose hydrolysis was 1 cm/sec.

    Notizen: A means to avoid the glucose effect in the production of baker's yeast from glucose and/or molasses in a fed batch culture by controlling the feed rate of fresh medium with an ad hoc measurement of the respiratory quotient, RQ, is presented. The feed rate is changed stepwise here such that the value of RQ ranges from 1.0 to 1.2 throughout the cultivation. Thus far, the specific growth rate based on the total cell mass and the growth yield obtained throughout are 0.24 hr-1 and 0.55 g cell/g glucose.Prior to the experimental run mentioned above, equations to predetermine the feed rate and concentration of glucose in the feed are derived from the mass balance of limiting substrates (glucose). Since values of either RQ or IO2 (QO2 x, oxygen consumption rate with respect to the total cell mass in the fermenter) can be measured quite easily and reliably, computer control of the fermentation in light of this information is discussed.

    Notizen: Differential speed two roll milling is an effective pretreatment for increasing the susceptibility of cellulose to enzymatic hydrolysis. Using mills with three, six, and ten in. diam rolls and processing times of 10 min or less results in the following percent increases in susceptibility over untreated controls: cotton, 1100; maple chips, 1600; white pine chips, 600; newspaper, 125. In comparison, ball milling of newspaper for 24 hr gives only a 62% increase. A further advantage of the roll mill is the increased wet density of the product permitting higher slurry concentrations during hydrolysis. Important parameters of mill effectiveness are roll clearance and processing time.

    Notizen: The efficiency of conversion of the carbon-energy source to product is of primary importance in many fermentation processes. In order to assess the efficiency of a process, one must know how close the actual conversion yield is to the theoretical maximum. Theoretical conversion yields are useful, therefore, as guides in improving a process. This knowledge is particularly important today because the cost of raw materials is rapidly rising. In this study, the biochemical pathway of penicillin synthesis was used to estimate the theoretical yield of penicillin from glucose, ammonia, and sulfate. These values are compared with experimental data from the literature. An analysis of the role of glucose in the synthesis of cell mass and penicillin and in the maintenance of cells makes it possible to assess the efficiency of carbon-source utilization and to direct further advances in penicillin fermentations.

    Notizen: A mathematical model for simulation of oxygen transfer in airlift fermentors is presented. The airlift fermentor is represented by a number of interconnected compartments, each of which is assumed to be well mixed. In the annular region, the model includes both upflow and downflow for the gas phase. The model contains several adjustable parameters through which important hydrodynamic effects affecting oxygen transfer are incorporated. The effect of hydrostatic pressure is also included in the model. The model is simple enough to be used in design studies and it can be easily adapted to other airlift system configurations. The simulation results show good qualitative agreement with available experimental results.

    Notizen: The microbiological leaching of a chalcopyrite concentrate has been investigated using a pure strain of Thiobacillus ferrooxidans. The optimum leaching conditions regarding pH, temperature, and pulp density were found to be 2.3, 35°C, and 22%, respectively. The energy of activation was calculated to be 16.7 kcal/mol. During these experiments the maximum rate of copper dissolution was about 215 mg/liters/hr and the final copper concentration was as high as 55 g/liter. This latter value is in the range of copper concentrations which may be used for direct electrorecovery of copper. Jarosite formation was observed during the leaching of the chalcopyrite concentrate. When the leach residue was reground to expose new substrate surface, subsequent leaching resulted in copper extractions up to about 80%. On the basis of this experimental work, a flow sheet has been proposed for commercial scale biohydrometallurgical treatment of high-grade chalcopyrite materials.

    Notizen: Five marine phytoplankton species (Phaeodactylum tricornutum, Thalassiosira pseudonana, Skeletonema costatum, Monochrysis lutheri, and Dunaliella tertiolecta) were grown in enriched laboratory continuous cultures and natural populations were mass cultured outdoors for 16 months. Competition among the species was shown to be highly dependent on temperature, although the actual production of plant organic matter at the low growth rates used was relatively independent of this variable. Control of marine species in mass cultures does not appear economically feasible, but this drawback may be overcome by selecting herbivorous shellfish that are capable of assimilating those temperature-dependent phytoplankton species dominating in a particular locale.

    Notizen: Alcohol yields of 6.5% were obtained with Saccharomyces cerevisiae in lactasehydrolyzed acid whey permeate containing 30-35% total solids. Maximum alcohol yields obtained with Kluyveromyces fragilis were 4.5% in lactase-hydrolyzed acid whey permeate at a solids concentration of 20% and 3.7% in normal permeate at a solids concentration of 10%. Saccharomyces cerevisiae efficiently converted the glucose present in lactase-hydrolyzed whey permeates containing 5-30% total solids (2-13% glucose) to alcohol. However, the galactose, which comprised about half the available carbohydrate in lactase-hydrolyzed whey, was not utilized by S. cerevisiae, so that even though alcohol yields were higher when this organism was used, the process was wasteful in that a substantial proportion of the substrate was not fermented. For the process to become commercially feasible, an efficient means of rapidly converting both the galactose and glucose to alcohol must be found.

    Notizen: A heat and caustic pretreatment process has been tested to determine the increase in gas yield that can be obtained from fermentation of organic municipal refuse. A treatment temperature of 130°C and a NaOH concentration of 3 g/100 g of dry solids resulted in the highest gas yield for the conditions tested. The probable increase in gas yield was 20% for the best conditions tested. The treatment procedure also substantially increased the rates of gas production. A high conversion efficiency is possible at much shorter reactor retention times with pretreatment than without pretreatment.

    Notizen: A conceptual model of aerobic microbial growth in semisolid matrices was developed as a first step in the prediction of the rate of breakdown of semisolid cellulosic material. The conceptual model was described by a series of equations simplified by the assumption of steady-state microbial activity, and heat and mass transfer limitation. Temperature and oxygen distribution in compost piles were measured experimentally at the Butler County Mushroom Farm, Butler County, Pennsylvania, to test the validity of these assumptions. The compost piles consisted of ground corn husks, straw, and race horse manure. The data fit with the model was excellent with deviations between model predictions (as solved by an analog computer) and actual temperature measurements never exceeding 3°C. The effects of compost pile geometry, external temperature, compost density, external oxygen concentration, and insulation at the bottom of the pile were then predicted using a digital computer to solve the model. The predictions show that the maximum breakdown rate occurs for an optimum height (which depends upon the system), insulating the base increases the breakdown rate, increasing the external temperature increases the initial breakdown rate but decreases the pseudo-steady-state breakdown rate and the uniformity, and any increase in the external oxygen concentration increases the breakdown rate but decreases the uniformity.

    Notizen: Lysozyme has been immobilized on chitosan, a polyaminosaccharide, without using any intermediate reagent. The best pH conditions for operating the chitosan columns have been determined and the best eluting agent was found to be a 2% solution of propylamine. The lysozyme activity was determined after reacting lysozyme with the product of glycolchitin and Remazol Brilliant Blue R. The recovery of lysozyme from chicken egg white yields lysozyme with 55% activity.

    Notizen: High activity alkaline protease was obtained when the enzyme was immobilized on Dowex MWA-1 (mesh 20-50) with 10% glutaraldehyde in chilled phosphate buffer (M/15, pH 6.5). Activity yields of the protease and rennet were 27 and 29, respectively. The highest activities appeared at 60°C, pH 10 for alkaline protease and 50°C, pH 4.0 for rennet. The properties of both proteases were not essentially changed by the immobilization except that the Km values of both enzymes were increased about tenfold as a result of immobilization. Both proteases in the immobilized state were more stable than those in the free state at 60°C. Other peptide hydrolases, β-galactosidase, invertase, and glucoamylase, were successfully immobilized with high activities, but lipase, hexokinase, glucose-6-phosphate dehydrogenase, and xanthine oxidase became inactive.

    Notizen: Various aspects of process water recycle in a continuous flow fermentation process are analyzed. Simple mass balance equations in terms of product and feed components for a single-stage reactor producing biomass are developed. Constraints on the recycle ratio, imposed by the efficiency of the dewatering stage, are examined. The recycle analysis is extended using a kinetic growth model incorporating water soluble product formation and growth inhibition. The potential effect of recycle on substrate conversion and product accumulation is also examined and the concept of a critical recycle ratio in fermentation processes is developed.

The properties of DEAE-cellulose-Ti-PMGL and of porous glass-Ti-PMGL were compared with those of the native enzyme; despite the presence of the metal and the heterogeneity of the substrate, pectin, typical substrate-enzyme-support interactions were demonstrated by shifts in pH optima and KM values.     Notizen: The degree of emulsification, measured as surface area of oil generated, was studied. The effect of interfacial tension, volume fraction of oil, and power per unit volume on the Sauter mean diameter of the oil drops was determined in an airlift system with motionless mixers. A mathematical expression to predict the Sauter mean diameter was developed using regression techniques. From this equation another equation, which will predict the surface area of oil in terms of the same variables, was derived. The effects of water air surface tension and power per unit volume on the gas hold-up were obtained using similar techniques. The results show that the interfacial tension and the surface tension are important variables when hydrocarbon fermentations are carried out in airlift systems.

    Notizen: This paper is concerned with optimization of the operating mode of a fermentor. Combining the various modes of operation - batch, semibatch, and continuous - the operating pattern which maximizes the desired metabolic product in a single fermentor is determined by using Kelley's transformation method with Pontryagin's maximum principle. Kelley's transformation method is a device which avoids the singular situation which occurs when the usual procedure of selecting the optimal control function by the maximum principle breaks down. This is the case in the problem considered in this paper. For lysine fermentation, the best operating mode depends on the fermentor capacity and operating time. The results of this study are summarized thus: (i) when the operating time is “long enough,” optimal conditions require that continuous operation follows either semibatch and/or batch operation, and (ii) when the fermentor capacity becomes “large enough,” semibatch operation becomes important.

    Notizen: This paper describes a mathematical model of the lag phases of Saccharomyces cerevisiae that incorporates the basic concepts previously presented in a two-stage deterministic model for the growth of this organism under conditions of oxygen excess with a sugar as the growth-limiting substrate. The model structure was suggested by an extensive investigation of the causes of the lag phases of S. cerevisiae which found that, in contrast to the traditionally accepted trends, the length of the lag phase was not inoculum-size dependent. This was consistent with other previously published work which suggested that a major factor in the length of the lag phases in S. cerevisiae was the need to synthesize adequate levels of glycolytic and respiratory enzymes. These suggestions were confirmed experimentally with lag-age data. Based on this conclusion a mathematical model was developed incorporating a description of the levels of glycolytic and respiratory enzymes and their effect on the growth rate and metabolism. This model was tested experimentally and the initial results indicate indicate that many aspects of the lag phase of this organism may be described mathematically. The experimental findings further support the concept of primary regulatory control proposed by Bijkerk and Hall.

    Notizen: In order to compare the process economics of making glucose from cellulose, a plant design is presented using acid hydrolysis which can be compared with a published design using enzyme hydrolysis. A common design basis is used; namely, an input capacity of 885 ton/day newsprint with a common technique of cost estimation. The cost of making glucose is in the range of 1.75 to 2.45 cents/lb, depending on the slurry concentration fed to the reactor for the acid hydrolysis. This cost range is less than the published estimate of 5.2 cents/lb for enzymatic hydrolysis.

    Notizen: Culture broths of cellulolytic fungi were used together with commercial amylases to enhance the saccharification of cassava starch slurry. It was found that the addition of appropriate concentration of the cellulases Trichoderma viride and a soil isolated Basidiomycete, increased both the rate of sugar formation and the degree of solubilization, and decreased the viscosity of the hydrolyzates. Owing to the improvement of the rheological properties of the must, and the additional sugar produced, an increased ethanol yield would be expected from the alcoholic fermentation of this hydrolyzate.

    Notizen: The effects of two grinding methods, hammer milling and defibrizing by disk refining, on the fermentability of ryegrass straw were investigated. Disk refined or defibrized straw produced more sugar than hammer milled straw. Release of sugar was especially pronounced when H2SO4 was added to the straw during the defibrizing process. In vitro rumen digestibility was significantly higher (P 〈 0.1) for defibrized than for hammer milled straw. With semisolid culture the level of yeast growth was about three times as high on the defibrized as on hammer milled straw. A scanning electron micrograph revealed that defibrizing removed the waxy surface of the straw as well as separating fiber bundles, so that the surface area of the exposed fiber structure was increased.

    Notizen: The feed value of annual ryegrass straw was improved by treatment with various concentrations of NaOH or NH3 followed by fermentation of the treated straw with a mixed culture of Cellulomonas sp. and Alcaligenes faecalis. Laboratory feeding trials with voles showed that NaOH or NH3 treatment considerably increased the feed efficiency of straw, but apparently gave a poorly palatable product. Fermentation tended to decrease the in vitro rumen digestibility (IVRD) of alkali-treated straw. The fermentations were carried out aerobically on a semisolid straw matrix having 11-86% moisture. Treatment by both NaOH and NH3 increased the IVRD of straw. NH3 also increased the nitrogen content in straw. The optimum condition for alkaline treatment of the straw was 4-6% NaOH for 1 hr or with 3% NH3 for four weeks at room temperature. A minimum of 63% moisture was needed for significant fermentation of the straw. The combined effects of NaOH treatment and fermentation more than doubled crude protein, doubled crude fat, and increased IVRD by 75%. The NH3 plus fermentation treatment tripled crude protein, doubled crude fat, and increased IVRD by 60%. Acetic acid was the main volatile fatty acid in the fermented straw.

The fermentor was operated at a single stirrer speed of 400 rpm and air rate of 1 v/v/m. The effect of four inoculum sizes (0.5, 1.0, 3.0 and 5.0%) on the growth pattern and the aeration profiles was examined. Logarithmic growth of the fungus was observed. The aeration profile changed with inoculum size and at 5.0%, it was found that the oxygen uptake rate was controlled by the oxygen supply rate, during which the oxygen tension was zero.

    Notizen: Studies to examine the microbial fermentation of coal gasification products (CO2, H2 and CO) to methane have been done with a mixed culture of anaerobic bacteria selected from an anaerobic sewage digestor. The specific rate of methane production at 37°C reached 25 mmol/g cell hr. The stoichiometry for methane production was 4 mmol H2/mol CO2. Cell recycle was used to increase the cell concentration from 2.5 to 8.3 g/liter; the volumetric rate of methane production ran from 1.3 to 4 liter/liter hr. The biogasification was also examined at elevated pressure (450 psi) and temperature to facilitate interfacing with a coal gasifier. At 60°C, the specific rate of methane production reached 50 mmol/g cell hr. Carbon monoxide utilization by the mixed culture of anaerobes and by a Rhodopseudomonas species was examined. Both cultures are able to carry out the shift conversion of CO and water to CO2 and hydrogen.

    Notizen: As shown in a previous paper, threshold concentrations of lower and intermediate fatty acids inhibit the uptake of inorganic phosphate, growth, and cell division in yeast cells, This paper demonstrates that, apart from these effects, the acids cause an increase in the respiration quotient (RQ), inhibition of CO2 fixation, production of ethanol at the expense of anabolic processes, and inhibition of active amino acid transport in the yeast Candida utilis. On the other hand, the threshold concentrations have no effect on intracellular pH. The inhibition of the inorganic phosphate uptake cannot be the sole primary mode of action of fatty acids since the omission of inorganic phosphate in the incubation medium brings about an inhibition of anabolic processes that is lower than that brought about by fatty acids at concentrations still permitting some phosphate uptake, Although 2,4-dinitrophenol and caproic acid at low concentrations cause an analogous decrease in biomass yield, their combination does not bring about any marked increase in the effect. Considering the physicochemical properties of fatty acids and their preferential action on energy-requiring processes, one of the key sitesof action can be assumed to be the mitochondrial membrane. Fatty acids might inhibit the transport of anions, especially phosphate, across the membrane, and disturb the membrane potential by affecting the transport protons. The physicochemical properties of fatty acids may also give rise to their binding to other intracellular membranes and to a subsequent interference with the function of the corresponding organelles.

    Notizen: Glucose isomerase (D-xylose ketol-isomerase EC 5.3.1.5) from Bacillus Coagulans was partially purified and immobilized by adsorption to anion exchangers. The highest activities were obtained when the enzyme was adsorbed to DEAE-cellulose. On immobilization to DEAE-cellulose the measured optimum pH value for enzyme activity shifted from 7.2 to 6.8. There was no appreciable difference between the heat stabilities of soluble and immobilized enzyme. The Km app values for the immobilized enzyme were found to be 0.25M in the presence of 0.01M Mg2+ and 0.19M with 0.005M Mg2+, while those enzyme were 0.11 and 0.17M, re spectively. Under conditions of contimuous of D-glucose, a decrease of activity with time was observed, but this decrease was less at a low Mg2+ concentration and was affected by column geometry. There were no appreciable diffusional limitation effects in packed-bed columns.
