    Notes: Summary Flux gate and resonance type of magnetometers (free precession, optically pumped), which can in some cases replace the SQUID magnetometer, are discussed. The principles of operation, sensitivity, dynamical range and frequency response are described. The application of these magnetometers for biomagnetic measurements is mentioned.

    Notes: Summary We have exploited the spatial resolution of the magnetic sensor to provide numerical estimates of the atrial contribution to magnetic fields recorded just prior to ventricular depolarization. The contribution is found to be small, strenghening our original interpretation that such fields originate mostly in the conduction system of the heart.

La procedura è stata applicata per esaminare i MCG di 16 pazienti affetti da pregresso infarto del miocardio. Sulla base di questa elaborazione è stato possibile dividere i pazienti in due classi, una delle quali contiene tutti i pazienti con depolarizzazioni tardive.
    Notes: Summary A possible clinical application of a high-resolution magnetic isofield contour mapping technique in cardiography is discussed. Preliminary results are presented in applying this technique to patients with recent myocardial infarction. The patients could be divided into two classes; one of them contains patients with electrical late potentials as a subclass.

    Description / Table of Contents: Riassunto Finora la magnetoencefalografia spontanea (MEG) in soggetti normali e patologici non è stata investigata a fondo. Si conoscono pochissimi contributi in questo campo. A parte il lavoro del gruppo di biomagnetismo di Roma, il solo contributo clinico finora è stato quello di Hughes e collaboratori che era abbastanza limitato nel numero e nella scelta dei soggetti studiati. La nostra esperienza condotta fin dal 1980 su un gran numero di casi (principalmente epilessie focali) mostra che il MEG può evidenziare attività significative anche se non sono presenti anormalità EEG; esso ha inoltre una risoluzione spaziale maggiore rispetto all’EEG. I foci epilettici mostrano un’area limitata di attività massima. Finora non è stata osservata nessuna evidenza sperimentale di una differente distribuzione delle ampiezze del campo sullo scalpo. La risoluzione spaziale del MEG riguarda anche la possibilità di risolvere la morfologia dei segnali EEG (per esempio onde aguzze EEG risolte in gruppi di spikes dal MEG) o di separare topograficamente le anormalità EEG mescolate e non ben localizzate (per esempio, attività delta ed epilettiche mescolate nell’EEG e discriminate nel MEG). I migliori risultati si osservano quando la localizzazione è nella corteccia esterna. Dopo una descrizione del materiale studiato, si sottolinea l’importanza dei risultati ottenuti per la prima volta in casi di epilessie focali.
    Notes: Summary So far spontaneous magnetoencephalography (MEG) in normal and pathological subjects has not been deeply investigated. Very few contributions in this field are available. Aside from the work of the Rome biomagnetic group in this field, the only clinical contribution up to now has been that of Hughes and co-workers which was rather limited in the number and in the selection of subjects investigated. Our experience, carried out since 1980 on a large number of cases—mainly focal epilepsies— shows that the MEG may evidence significant activities even if EEG abnormalities are not present and has a higher spatial resolution with respect to the EEG. Epileptic foci show a limited area of maximal activity. So far no experimental evidence of a different distribution of field amplitude over the scalp has been observed. The spatial resolution of the MEG also regards the possibility of resolving the morphology of EEG signals (for instance, EEG sharp waves resolved into groups of spikes by MEG) or topographically separate EEG abnormalities mixed up and not well localized (for instance, delta and epileptic activities mixed up in the EEG, but differently in the MEG). The best results are observed when the localization is in the outer cortex. After a description of the material studied, the importance of the results obtained for the first time in cases of focal epilepsies is outlined.

    Description / Table of Contents: Riassunto Durante l’ultimo anno gli autori hanno condotto una ricerca sistematica su un gran numero di soggetti patologici registrando simultaneamente l’EEG e il MEG. La presente versione del nostro apparato sperimentale permette di effettuare misure MEG con un solo sensore magnetico, impedendo la rilevazione simultanea di attività MEG su punti differenti della testa. Ciò ha condotto gli autori a concentrarsi sulle epilessie focali. Fin dalle prime misure il sensore magnetico ha dimostrato un notevole potere localizzante, rafforzando l’interesse per la localizzazione in casi patologici. In questo lavoro si riportano risultati che sembrano confermare la straordìnaria capacità delle misure MEG nel localizzare sorgenti di attività cerebrali patologiche in casi di epilessie focali.
    Notes: Summary During the last year the authors have been carrying out a systematic investigation on a large number of pathological subjects simultaneously recording the EEG and the MEG. The present version of our experimental apparatus allows MEG measurements with only one magnetic detector, thus preventing the detection of simultaneous MEG activities over different points of the head. This has led the authors to concentrate upon focal epilepsies. Since the first measurements a remarkable localizing power was shown by the magnetic detector, thus strengthening the interest in pathological localization. In this contribution we report results which seem to confirm the extraordinary capability of MEG measurements to localize sources of pathological brain activities in cases of focal epilepsies.

Si è trovato che la componente antero-posterioreX vettore magnetico del cuore è la piú facile da scoprire. Si suggeriscono conduttori ideali per le altre due componenti. Comunque questi hanno una sensibilità inferiore. Le differenze rispetto ai risultati di precedenti ricercatori sono state spiegate con le differenze tra i modelli.
    Notes: Summary The ideality and sensitivity of several vector magnetocardiographic leads were tested by using an inhomogeneous, physical torso model. The postero-anterior component of the heart’s magnetic vector was found easiest to detect. New, more ideal leads for the other two components are suggested. However, these have lower sensitivity. The differences from the results of former investigators can be explained by the differences of the models.

    Description / Table of Contents: Riassunto Si è studiato lo spettro del rumore di un gradiometro superconduttore a derivata secon da in un agglomerato urbano. I dati sono stati ottenuti dal gradiometro con tre accelerometri ortogonali e un magnetometro a flussometro elettronico triassiale attaccato al dewar. Questi dati sono stati analizzati usando tecniche di elaborazione dei segnali, soprattutto un cancellatore adattativo di rumori, per ridurre i rumori nei dati del gradiometro. I risultati mostrano qui che il rumore dell’esterno e/o il rumore del movimento del dewar possono essere ridotti di 40 dB in potenza di rumore. Si è anche mostrato che, nel caso di basso rapporto segnale-rumore (S/N∼1), può essere estratta un’onda sunusoidale di 10 Hz. Questa tecnica non solo promette riduzione dei rumori, ma ha anche lo scopo d’identificare i segnali di rumore che potrebbero essere veramente considerati parte del responso evocato. L’analisi dei dati che contengono i responsi evocati è ora in corso.
    Notes: Summary The noise spectrum of a superconducting second-derivative gradiometer has been investigated in an urban environment. Data have been acquired from the gradiometer with three orthogonal accelerometers and a triaxial fluxgate magnetometer attached to the dewar. These data have been analyzed by using signal processing techniques, primarily an adaptive noise canceller, to reduce noise in the gradiometer data. Results shown here indicate that the environmental noise and/or the dewar motion noise can be reduced as much as 40 dB in noise power. It is also shown, in the case of poor signal-to-noise ratio (S/N∼1), that a 10 Hz sine wave can be extracted. This technique not only shows promise for noise reduction, but also aids in the identification of noise signals which might be misconstrued as part of the evoked response. Analysis of data containing evoked response is now underway.

    Notes: Summary The instrumentation for biomagnetic applications is described. The small size and weight of the SQUID gradiometer permit easy transport and positioning of the instrument, thus making biomagnetic measurements possible in any noise-free location. The equipment is provided with a battery power supply, and, when filled with 60 cm3 of liquid helium, will operate continuously for 6 h. The device is suitable for magnetocardiography.

Si riesaminano il principio di misurazione e i lavori recenti di altri ricercatori. Si studia l’effetto del movimento legato al cuore della cassa toracica anteriore sul segnale MSPG.I risultati suggeriscono che il movimento del confine cassa toracica-aria può contribuire in modo significativo (∼75%) al segnale MSPG quando è registrato vicino al cuore.
    Notes: Summary Magnetic-susceptibility plethysmography (MSPG) is a new method proposed for monitoring cardiac-volume changes. The measurement principle and recent work by other investigators are reviewed. The effect of the cardiac-related motion of the anterior chest wall on the MSPG signal is studied. The results suggest that the motion of the chest-wall-air boundary may contribute significantly (∼75%) to the MSPG signal when recorded near the heart.

The fields are affected by muscle activity, decaying with the time of relaxation of the leg.

    Description / Table of Contents: Riassunto In questo lavoro si tratta il problema dell’ottimizzazione di sistemi di sensori gradiometrici metrici superconduttori di secondo ordine che sono stati progettati per la misurazione di campi magnetici deboli disomogenei in un ambiente disturbato magneticamente. Si considerano le condizioni di massimo rapporto segnale-rumore dell’uscita del quantomagnetometro superconduttote (SQM). Si analizza l’influenza dei parametri misurati della sorgente del segnale, del rumore di una sorgente di disturbo esterna e di quello proprio del magnetometro sulla selezione della lunghezza di base del gradiometro.
    Notes: Summary In this paper we deal with an optimization problem of the second-order superconducting gradiometric sensor systems that have been designed for measurement of the weak inhomogeneous magnetic fields in a magnetically disturbed environment. The conditions of maximum signalto-noise ratio of the output of superconducting quantum magnetometers (SQM) are considered. The influence of measured signal source parameters, of an outside disturbing source and of the own noise of the magnetometer on the gradiometer base length selection is analysed.

    Notes: Summary We show that substantial improvements can be obtained in the shielding properties of an eddy-current-shielded room by covering the doorway with conductive plates. Results suggest that it is unnecessary to install a conductive door and show that it is worthwhile covering a small part of the doorway permanently.

Sono state osservate rampe maggiori dopo aver indotto con farmaci il blocco del nodo atrioventricolare di primo grado, ed è stata osservata una completa ripolarizzazione atriale a seguito d’isolate contrazioni atriali in due animali con blocco di secondo grado. Queste rampe, che variano fino a 6.7 pT in ampiezza e provengono dagli atri, possono complicare l’individuazione magnetica dell’attività di His-Purkinje.
    Notes: Summary Sloping signals were observed in thePR segment of normal magnetocardiograms recorded from five dogs. Larger ramps were observed after first-degree AV nodal blockade was induced pharmacologically, and complete atrial repolarization was observed following isolated atrial contractions in two animals with second-degree block. These ramps, ranging up to 6.7 pT in size and arising from the atria, may complicate magnetic detection of His-Purkinje activity.

    Description / Table of Contents: Riassunto La motivazione fondamentale per lo studio dei fenomeni elettrofisiologici cardiaci tramite il metodo biomagnetico è dovuta al fatto che questa tecnica offre un’alta risoluzione spaziale e sembra fornire informazioni complementari rispetto alle sole registrazioni elettriche. Fino ad oggi l’interesse degli studiosi si è concentrato su due differenti fenomeni cardiaci: 1) lo studio dell’attività del sistema di conduzione atrioventricolare e 2) lo studio della depolarizzazione ventricolare anormalmente ritardata (late potentials). L’interesse clinico per lo studio non invasivo del sistema di His-Purkinje (HPS) è ben noto. Registrazioni cliniche non invasive di late potentials sono particolarmente desiderabili in quanto possono essere preziosi indicatori di rischio di morte improvvisa. In questo lavoro si presenta una rassegna sullo stato delle ricerche in entrambi i campi d’indagine.
    Notes: Summary The fundamental motivation for investigating cardiac electrophysiological phenomena by means of the biomagnetic method has been that this technique should have added spatial resolution and provided complementary information as opposed to electric recordings only. So far the interest of experimentalists has been concentrated on two different kinds of cardiac phenomena: 1) the study of the activity of the cardiac conduction system and 2) of abnormally delayed ventricular depolarization (late potentials). The clinical interest for the noninvasive investigation of the His-Purkinje system (HPS) is well known. Noninvasive clinical recording of late potentials is particularly desirable as they seem to be precious markers for risk of sudden death. A review of the state of research in both fields is presented in this paper.

    Description / Table of Contents: Riassunto Ein dal 1980 è iniziato uno studio sistematico dell’attività del sistema di His-Purkjnje (HPS) su soggetti normali e patologici con il metodo biomagnetico. Allo scopo di risolvere il problema di quanta parte dei segnali misurati sia attribuibile al sistema di His-Purkinje e quanta piuttosto alla ripolarizzazione atriale, è stata recentemente sviluppata una strumentazione appositamente disegnata per misure sul HPS e con una sensibilità di $$20fT/\sqrt {Hz}$$ nella banda di frequenze interessanti ((0.5÷300) Hz). Al contempo si è altresí sviluppato un nuovo modello teorico in stretta corrispondenza con le caratteristiche anatomiche del HPS che fornisce una distribuzione di campo magnetico sul torace in buon accordo qualitativo con i risultati sperimentali.
    Notes: Summary Since 1980, systematic investigations of the activity of the His-Purkinje system (HPS) by the biomagnetic method have been reported on both normal subjects and patients affected by different kinds of conduction disturbances. A first attempt to interpret the experimental results by means of a theoretical model satisfactorily accounted for the measured «ramp» patterns. Moreover, a good temporal coincidence was observed between magnetic signals and invasively recorded His bundle electrograms (HBE). An alternative explanation of the «ramp» pattern, in terms of atrial repolarization, has been recently proposed on the basis of animal experimental investigation. In order to try to solve this problem, we have carried out a twofold approach: a new second-order gradiometer, specifically designed for HPS activity investigation has been successfully set up and tested; it provides a sensitivity of $$20fT/\sqrt {Hz}$$ in the band width of interest ((0.5÷300) Hz). Second, a new model based on a closer correspondence to anatomical features of the HPS has been developed. This model provides well-defined patterns which are in quite good agreement with experimental results.

I risultati mostrano che entrambi i componenti magnetici sono localizzati nella corteccia occipitale superficiale tangenziale al cuoio capelluto. La distribuzione diM120 suggerisce che i generatori dei componenti elettrici e magnetici sono correlati, ma il campo magnetico elettrico registrato è una modificazione elaborata di quello prodotto dalla sorgente.
    Notes: Summary Corresponding electrical and magnetic components of the brain’s visual evoked response peaking at 120 and 180 ms poststimulus are examined. Results show that both magnetic components are localized in the superficial occipital cortex tangential to the scalp. The distribution ofM120 suggests that the generators for the electrical and magnetic components are related, but the electrical field recorded is an elaborate modification of that produced by the source.

    Description / Table of Contents: Riassunto In questo lavoro si presentano alcuni tentativi di considerazioni modello, basati sul concetto di dipolo di corrente equivalente per tener conto delle risposte sonore stimolate osservate, con lo scopo di tentare un confronto delle variazioni in ampiezza e latenza con calcoli basati sul modello di nervo altamente semplificato di Rall. Si presenta un’analisi fenomenologica dei dati sperimentali sul campo magnetico sonoro tardo evocato dalla corteccia uditiva umana. Qui si pone l’accento sulle relazioni tra le proprietà del segnale (per esempio, ampiezza e latenza) e tra le proprietà del segnale e le proprietà dello stimolo (per esempio, ampiezza/latenza e frequenza in discesa relativa Δf/f per uno stimolo con frequenza in discesa) piuttosto che su problemi correlati con la posizione del dipolo di corrente equivalente, per esempio l’organizzazione tonotopica discussa da Elberlinget al. e Romaniet al. Si presentano alcuni risultati di un calcolo numerico basato su un modello a cavo lineare e li si confronta con i risultati sperimentali.
    Notes: Summary In this paper we present some tentative model considerations, based on the concept of an equivalent current dipole to account for observed auditory evoked responses, with the aim of attempting a comparison of variations in amplitude and latency with calculations based on Rall’s greatly simplified nerve model. A phenomenological analysis of experimental data on the late, auditory evoked magnetic field from the human auditory cortex is presented. The emphasis is here placed on relations between signal properties (e.g. amplitude and latency) and between signal properties and stimulus properties (e.g. amplitude/latency and relative frequency glide Δf/f for a frequency glide stimulus) rather than on questions related to the location of the equivalent current dipole,e.g. the tonotopic organization discussed by Elberlinget al. and by Romaniet al.. Some results of a numerical calculation based on a linear-cable model are presented and compared with experimental results.

L’attività a livello delle cortecce uditive, 2.6 cm al di sotto della calotta cranica, spiega la distribuzione di campo osservata $$\overline {N100m}$$ . Un’ulteriore sorgente di corrente vicino al vertice è necessaria per spiegare le corrispondenti distribuzioni di potenziale.     Notes: Summary The sources of auditory evoked electric and magnetic responses were modelled with multiple current dipoles. The activity at the auditory cortices, 2.6 cm beneath the skull, explains the observed $$\overline {N100m}$$ field distribution. An additional current source near the vertex is needed to explain the corresponding potential distributions. The locations of the current sources of $$\overline {N100m}$$ did not show any systematic dependence on the tonal frequency.

Si è trovato che l’ampiezza AEF è correlata all’intensità dello stimolo in modo quadratico, l’ampiezza AEP in modo, lineare. Gli AEF erano di ampiezza maggiore in risposta alla stimolazione contralaterale che in risposta alla stimolazione ipsilaterale. Gli AEP non presentano questa relazione. In un secondo esperimento gli AEF e AEP dell’emisfero destro in risposta al tono di stimolazione nell’orecchio controlaterale in questi 12 soggetti sono stati combinati con dati simili precedenti su 24 soggetti, ottenendo cosí un totale di 36 soggetti, per esaminare la comparabilità della forma dell’onadaP50 di AEP e dell’analogaP50 di AEF. Si è trovato che la latenza dellaP50 decrese in funzione dell’intensità crescente dello stimolo sia per AEF che per AEP, e che la latenza diP50 è decisamente inferiore nelle registrazioni magnetiche rispetto alle registrazioni del potenziale.
    Notes: Summary Auditory evoked magnetic fields (AEFs) and EEG auditory evoked potentials (AEPs) were recorded from left and right auditory cortical regions of 12 normal adult subjects. The magnetic sensor was a figure-eight SQUID gradiometer with a 4 cm baseline oriented so as to be maximally sensitive to a current dipole oriented normal to the Sylvian fissure. Stimuli were 100 ms long 1 kHz tone pips with a modal interstimulus interval of 700 ms delivered at sound pressure levels of 40, 60, 80 and 100 dB. AEF amplitude was found to be related to stimulus intensity in a quadratic fashion, AEP amplitude in a linear fashion. AEFs were of larger amplitude in response to contralateral as compared to ipsilateral stimulation. AEPs did not exhibit such a relationship. In a second experiment right-hemisphere AEFs and AEPs in response to contralateral ear tone stimulation in these 12 subjects were combined with similar previous data from 24 subjects, providing a total of 36 subjects, to examine the comparability of the AEPP50 wave form and the AEFP50 analog. The latency of theP50 was found to decrease as a function of increasing stimulus intensity for both AEFs and AEPs, and theP50 latency was consistently shorter in magnetic compared to potential recordings.

In alcuni soggetti sono state effettuate mappe di campi magnetici generati dall’attività spontanea del cervello. Le mappe dell’ampiezza media dei segnali del ritmo α hanno mostrato 2 massimi dell’attività su 2 regioni simmetriche dello scalpo. Comunque, malgrado una generale simmetria fra gli emisferi, la complessità delle mappe misurate suggerisce una corrispondente complessità dei generatori responsabili dell’attività α.
    Notes: Summary An investigation of the spontaneous brain activity from normal subjects has been carried out by means of the neuromagnetic method. Two different systems were used for magnetic measurements. Both used 2nd-order gradiometers specifically designed for brain studies: 2.9 cm diameter, 5 cm baseline. A r.f. SQUID and a d.c. SQUID were coupled to the gradiometers. The measured noise levels are $$40fT/\sqrt {Hz}$$ and $$25fT/\sqrt {Hz}$$ , respectively. Thorough magnetic mapping of the spontaneous activity was performed on a few subjects. By mapping the average amplitude of the α-rhythm signals two maxima of activity over two symmetric regions of the scalp were clearly observed. Despite a general symmetry between hemispheres, however, the complexity of the measured maps suggests a corresponding complexity of the generators responsible for α-activity.

    Notes: Summary The concurrent administration of tricyclic antidepressants has been shown in man to result in a clinically significant impairment of the antihypertensive effect of clonidine. This interaction is thought to be related to competition for central α2 receptors where clonidine acts as an agonist and the tricyclics act as antagonists. Although it seems to cause less cardiovascular effects than tricyclic antidepressants, the tetracyclic antidepressant, mianserin also has been reported to be an α receptor antagonist and may, therefore, also interfere with the antihypertensive activity of centrally-acting drugs. This study investigates the effects of acute and chronic mianserin administration in patients with essential hypertension established on long term treatment with either clonidine or methyldopa. The first dose of mianserin was not associated with an increase in blood pressure and during a further two weeks of mianserin therapy there were no significant alterations in blood pressure, supine or erect. Similarly, mianserin did not alter heart rate either after acute or after chronic administration. Mianserin itself had a sedative effect but there was no interference with the sedation attributable to clonidine or methyldopa. Mianserin caused no reduction in salivary flow and did not influence the reduced saliva production caused by clonidine. Both clonidine and methyldopa are associated with a reduction in sympathetic outflow but there was no evidence in this study of any further change in plasma noradrenaline or 24 h urinary catecholamine excretion. This study demonstrates that if mianserin is given acutely or chronically, it does not interfere with the effects of the centrally acting antihypertensive drugs, clonidine and methyldopa. Mianserin may therefore be a suitable antidepressant for patients receiving these antihypertensive agents if drug treatment for depression is indicated.

    Notes: Summary Cadralazine (ISF 2469) was administered to 24 hypertensive patients in single oral doses of 7.5, 10, 15, 20 and 30 mg, according to a single-blind, placebo-controlled, within-patient change-over design. The study was done in 2 stages: in the first a range including the upper and lower doses was studied (7.5, 15, 30 mg and placebo), and in the second the range of doses was restricted (10, 15, 20 mg and placebo). The drug produced a significant decrease in blood pressure in the supine and standing positions. The decrease became clinically important starting from the 15 mg dose. Its action was still significant 12 h after administration. A significant increase in heart rate was also observed. All the effects were correlated with the dose. Side effects occurred mainly after the 30 mg dose. Thus, cadralazine, in a single oral dose in man, showed good antihypertensive activity starting from the 15 mg dose, and its effect was dose-related, slow in onset and long-lasting.

    Notes: Summary The disposition of oral metoprolol was studied in 5 women during the last trimester of pregnancy and 3 to 5 months after delivery. After a single oral dose of 100 mg the individual peak plasma concentration in the pregnant state was only 20–40% of that after pregnancy. The plasma half-lives of metoprolol were about the same during (average 1.3 h) and after pregnancy (average 1.7 h). By contrast, the area under the plasma concentration versus time curve was much smallerduring (mean 262 nmol/l×h) thanafter (mean 1298 nmol/l×h) pregnancy, resulting in an average apparent oral clearance (Clo) of metoprolol that was 4.4times higher during (362 ml×kg−1 body-weight×min−1) than after pregnancy. The increased Clo in pregnancy is assumed to be due to enhanced hepatic metabolism of the drug. The possible clinical consequence of the difference in the disposition of metoprolol is discussed.

    Notes: Summary Several β-blockers increase VLDL-TG and decrease HDL-cholesterol concentrations. The underlying mechanism ist not yet clear. Some studies have suggested that the effect is less pronounced during treatment with selective β-blockers. The effects of 2 such drugs, metoprolol 200 mg/day and atenolol 50 mg/day, have been compared in 50 hypertensive patients (WHO Stage I–II), mean age 47 years. Serum lipoproteins were determined in 20 patients before treatment and after treatment with either drug for 3 months. Both drugs were equally effective in reducing blood pressure. After atenolol the initial VLDL-cholesterol concentration of 1.04 mmol/l had not changed, but it rose to 1.29 mmol/l after metoprolol (p〈0.05). The HDL-cholesterol concentration 1.42 mmol/l did not fall during atenolol treatment, but during metoprolol there was a small reduction to 1.31 mmol/l (p〈0.05). Hyperlipoproteinaemia is common in hypertensive patients, 40% of the present group had hypertriglyceridaemia and 25% had hypercholesterolaemia. Thus, atenolol 50 mg was found not to affect lipoproteins, whereas metoprolol 200 mg increased the VLDL concentration in 75% of the patients.

    Notes: Summary The unusual observation of a withdrawal syndrome due to guanfacine in a hypertensive patient with chronic renal failure led to a study of the kinetics of the drug in this patient. The principal pharmacokinetic parameters of guanfacine were greatly altered, with extended biotransformation and a decrease in the half-life compared to the values observed in other cases of severe renal insufficiency. Associated treatment with phenobarbital had had a considerable effect, as shown by the results of a further kinetic study 2 months after withdrawal of the phenobarbital. The findings then were in good agreement with reference values which strongly suggests a consequence of the enzyme inducing effect of phenobarbital.     Notes: Summary The effects of captopril and propranolol on blood pressure, heart rate and plasma noradrenaline, renin and aldosterone, and on the responses to changes in posture and to isometric exercise were measured in patients with essential hypertension. During placebo administration blood pressure, heart rate and plasma noradrenaline rose on standing and during isometric exercise. The rise in diastolic blood pressure during isometric exercise correlated significantly with the rise in plasma noradrenaline. During captopril treatment blood pressure was significantly lower than during placebo administration when the patients were lying, standing or sitting, but the reduction during isometric exercise was not significant. Plasma renin increased, but heart rate, plasma noradrenaline and plasma aldosterone remained unchanged. The acute changes in blood pressure, heart rate and plasma noradrenaline produced by standing and isometric exercise during captopril treatment were similar to those during placebo administration. During propranolol treatment diastolic blood pressure was significantly lower than during placebo administration when the patients were lying, standing or sitting and during isometric exercise. Heart rate also fell. Plasma noradrenaline during standing, sitting and isometric exercise was significantly greater than during placebo administration. The changes in plasma noradrenaline measured during propranolol treatment with the patients supine were negatively correlated with noradrenaline values obtained during placebo administration: plasma noradrenaline fell in patients with higher, and increased in those with lower, initial concentrations. The expected acute increase in heart rate on standing and during isometric exercise was blunted by propranolol, but the changes in blood pressure and plasma noradrenaline were unaffected. We conclude that in essential hypertension noradrenaline is involved in the pressor response to isometric exercise. Angiotensin converting enzyme inhibition by captopril did not interfere with the responses of the sympathetic nervous system to postural changes and isometric exercise. During propranolol treatment there was no evidence that reduced sympathetic activity was involved in the hypotensive response.

    Notes: Summary The haemodynamic effects of a new vasodilating drug, felodipine, were studied in eight, healthy, male subjects, aged 22–31 years. The drug was given as an oral solution in the dose of 0.15 mg/kg. Thirty-five minutes later further dose of 0.15 mg/kg was administered. Felodipine induced a pronounced decrease in diastolic blood pressure (maximal effect 15±4 mm Hg) and in the systemic vascular resistance. Cardiac output increased (maximum by 4.2±0.3 l/min), due to an increase both in the stroke volume and the heart rate. The maximal increase in the stroke volume (measured from echo cardiograms) and the heart rate were 33±5 ml and 23±3 beats/min, respectively. Felodipine caused a significant decrease in the pre-ejection period (23±3 ms) and an increase in the left ventricular ejection time (29±3 ms). The quotient PEP/LVET fell from 0.36±0.01 to 0.28±0.01. Significant activity of felodipine could be recorded at a plasma level of about 15 nmol/l. When the maximal haemodynamic effects were recorded the plasma level was about 40 nmol/l. After a cumulative dose of 0.30 mg/kg, there was a twofold variation in the maximal plasma level (from 31 to 61 nmol/l). The results of the present investigation are in agreement with previous haemodynamic studies in animals. It would appear that felodipine is a potent arteriolar vasodilator and it might well be of considerable value in the management of patients with hypertension or congestive cardiac failure.

After an initial placebo test, nifedipine 20, 40 or 60 mg was given in random order at 72-h intervals, in a single administration crossover study. The placebo and the active drug were given at 8 a.m. Blood pressure and heart rate were measured twice by the same observer, every 20 min from 7 to 8 a.m., and then hourly until 8 p.m., first in recumbency and again after 1 min of standing upright. Plasma nifedipine was assayed in samples taken hourly from 8 a.m. to noon, every 2 h from noon to 8 p.m., and 24 and 48 h after drug administration. All 3 doses significantly lowered blood pressure; the fall during recumbency was significantly larger (−18%) and lasted longer (12 h) after 60 mg than after 20 mg (−11% and 7 h). All 3 doses caused a similar increase in heart rate (+29 to +38%), which reached its maximum after 2 h and lasted for 5 h. The maximum plasma concentration and the area under the plasma concentration — time curve were dose-dependent despite large inter-subject variation. Absorption, bioavailability and elimination were linear between the 20 and 60 mg doses. Plasma nifedipine levels were strongly correlated with the concomitant decrease in mean arterial blood pressure (r=0.61,p〈0.001). Four patients experienced mild side effects (headaches, flushes, drowsiness or weakness). This tablet form of nifedipine has a potent antihypertensive action which lasts longer than that of the capsule presentation.

    Notes: Summary A naturalistic study was set up to screen, identify and treat hypertensive patients aged 20–60 years in a rural general practice. 3,222 patients (92%) of a stable population of 3,489 were screened by 2 nurse research assistants and of these 455 patients (14%) were found to be hypertensive or borderline hypertensive. After careful assessment, 192 of these patients were found suitable for treatment and subsequently 138 entered the study. Two well recognised treatment regimes were used and no significant difference between patient response resulted. 84 patients (60.9%) completed the 2 year duration of the study discussed here. The cost of the study is not feasible in an average general practice, but day to day running of such a project, run along clearly defined treatment regimes was managed easily by 2 research assistants: this reduced, therefore, the work load on individual general practitoners.

    Notes: Summary The relationship between the plasma concentrations of bufuralol and its major hydroxymetabolite (Ro 3-7410) and β-blocking activity was studied in 10 patients with uncomplicated essential hypertension. Blood samples and haemodynamic data were obtained during rest and after a single-level exercise test on a bicycle cycloergometer, prior to and up to 32 h after administration of a single oral dose of bufuralol 30 mg. Bufuralol was rapidly absorbed, following a first-order process with a lag time. The calculated maximal plasma concentration ranged from 44.6 to 200.3 ng/ml. The half-life of bufuralol was 2.75±1.15 h (mean±SD). Up to 50% of the parent drug was transformed into Ro 3-7410, which showed less interpatient variability in concentration and a fairly constant half-life, which was three times longer than that of the parent drug. In general, the heart rate (HR) was slightly decreased, although 2/10 patients showed an initial increase. The resting HR returned to its pre-treatment level within 6 h, the exercise HR took up to 32 h to return to the pre-treatment level. The drug reduced both resting and exercise blood pressure (BP). The former was reduced from 153.0±14.2/93.5±8.5 to 134.5±14.0/77.0±6.8 mmHg (systolic/diastolic BP; mean±SD) with 6 h after treatment. Similarly, the exercise BP was reduced from 199.0±15.2/98.5±8.8 to 171.0±9.9/88.5±8.5 mmHg at the 6th h post-dosing. The BP values had not returned to their pre-treatment levels even 32 h after treatment. Thus, bufuralol and its metabolite Ro 3-7410 induced a long-lasting antihypertensive effect and inhibited the cardio-acceleratory effect of exercise, and there was a good correlation between the pharmacokinetic and pharmacodynamic behaviour of the drug.

    Notes: Summary The effects of endralazine, a new antihypertensive hydrazinopyridazine derivative, on heart rate, mean blood pressure (mBP), glomerular filtration rate (GFR), effective renal plasma flow (CPAH), urine volume (V), the clearance of Na, K, urea (Ur) and uric acid (UA), plasma renin activity (PRA) and plasma aldosterone (PA) were studied in hypertensive patients after a single oral dose of 10–15 mg, and after 8–17 days of treatment with daily doses of 15–90 mg. In the acute experiments, heart rate increased by 27%, mBP decreased on average by 17% and GFR by 33% and CPAH fell by only 5%. Urine volume and electrolyte clearance were also depressed. There was a significant increase in PRA and PA. The fall in GFR correlated directly with mBP, CPAH and the product (mBP×CPAH). The logarithms of the Na clearance and V were correlated with GFR and mBP. The logarithms of the fractional excretion of Na and water also correlated with mBP, suggesting that tubular reabsorption of sodium and water may be affected by change in mBP. The fractional potassium excretion correlated directly with CPAH and ln PA. In contrast, on sustained daily treatment, mBP was less depressed (9%), but GFR increased strikingly by 27% and CPAH by 46%. The body weight increased by 4.5% as a consequence of salt and water retention. GFR was correlated with CPAH, the product (mBP×CPAH) and the increase in body weight. Thus, the improvement in GFR and effective renal plasma flow observed under these conditions may be due, in part, to volume expansion. However, a direct renal vasodilating effect of the drug appears to be the more important determinant.

No correlations were found between the mean arterial blood pressure (post-therapy) or the fall in blood pressure as a result of the P therapy (p〉 〉0.05) and P dose, peak P plasma concentrations, peak 4-hydroxypropranolol (4OHP) plasma concentrations or peak (P plus 4OHP) plasma concentrations. However, a positive nonlinear relationship was found between the daily P dose (independent variable) and peak P plasma concentrations over the daily dose range 30–160 mg/day. The elimination half-lives of NLA for patients in the third trimester of pregnancy were significantly shorter (p=0.072, df=13) than those when the patients were at least three months post-partum. Also, the areas under the plasma level-time curves of NLA were significantly less (p〈0.05, df=13) for patients in the third trimester of pregnancy than when these patients were at least three months post-partum. The results of this study indicate that the pharmacokinetics of P, PGLUC, 4OHP and 4OHPGLUC are not significantly altered by pregnancy. However, the kinetics of NLA do appear to be altered. The formation of NLA by N-dealkylation of P and further oxidation, appears to be competitively inhibited by unidentified substances, perhaps endogenous steroids, especially in the third trimester when compared to at least three months post-partum.

    Notes: Summary Ketanserin is a new, specific serotonin receptor blocking agent, which causes vasodilatation, presumably by an action on the vascular wall. The antihypertensive response to ketanserin 40 mg twice daily as monotherapy was assessed in 8 patients with essential hypertension. The investigation was an 8 week, double-blind, cross over study, which also included measurements during isometric (handgrip) and dynamic exercise (bicycle ergometry), as well as determination of plasma catecholamines and ketanserin. Ketanserin caused a reduction of supine and standing systolic and diastolic blood pressure (BP) during rest and a slight bradycardia. Although there was attenuation of the pressor response to handgrip, treatment with ketanserin did not really affect the changes in BP or heart rate during exercise, i.e. the base-line differences remained the same. There was no significant correlation between the effect on BP and the plasma level of ketanserin. The changes in BP produced by ketanserin showed little correlation with the initial levels of plasma catecholamines or with alterations in those levels. Although the results did not indicate direct interference by ketanserin with sympathetic tone, the lack of reflexogenic tachycardia, as well as the lack of increase in plasma noradrenaline during hand grip, indicates at least some modulation of autonomic function. It is concluded that ketanserin lowers BP in essential hypertension without interference with cardiovascular reflexes during standing or exercise, and that the compound may offer an alternative approach in the treatment of hypertension.

    Notes: Summary Fourteen men with moderately severe essential hypertension were treated with prizidilol hydrochloride 400–700 mg once daily (mean±S.D. 612±56 mg/day). The study was open and ambulatory, with an initial placebo period followed by dose titration of prizidilol. Prior to treatment and during optimal control of blood pressure cardiovascular adaptation was examined in a submaximal exercise test. Plethysomographic assessment of vascular flow, resistance and tone in the calf musculature during supine rest and during maximal vasodilatation was also performed. A highly significant reduction in systolic (from 164±4.5 to 141±2.7 mmHg; p〈0.001) and diastolic blood pressure (from 105±1.6 to 87±1.3 mmHg; p〈0.001) at supine rest was noted during therapy with prizidilol. There was no significant change in heart rate. Systolic pressure in the standing position was reduced (from 159±4.2 to 139±2.9 mmHg; p〈0.001) and so was the diastolic pressure (from 111±2.5 to 95±1.9 mmHg; p〈0.001). The heart rate in the standing position was significantly increased compared to supine rest in the placebo period and during optimal treatment with prizidilol. The β-adrenoceptor blocking properties of prizidilol were apparent as a reduction in the exercise-induced heart rate response at even the lowest work load. During prizidilol therapy an increase in resting calf muscle blood flow was found from 3.1±1.5 ml/min·100 ml to 4.3±2.1 ml/min·100 ml (p〈0.025). Vascular resistance and vascular tone were significantly reduced. No change regarding blood flow or resistance during maximal vasodilatation was noted. It is considered that prizidilol has a clear antihypertensive effect combining β-receptor blocking and vasodilator properties.

    Notes: Summary Nineteen out-patients with moderate to severe essential hypertension were treated daily for 3 years, with an average dose of 13 mg endralazine, a new peripheral vasodilator, in free combination with pindolol 3×5 mg. The blood pressure showed a statistically significant reduction from 172/110 mmHg to 154/92 mmHg after treatment for 3 weeks. Tachyphylaxis was not observed during the 3 year period. Oedema was the most frequent side-effect, but it disappeared spontaneously. No difference in efficacy and tolerance between slow and fast acetylators was found. Only 2 patients developed a weak positive antinuclear antibody titre, which disappeared spontaneously from one during continued treatment. No clinical evidence of a systemic lupus erythematosus-like syndrome was noted. It is concluded that the differences between endralazine and hydralazine in dosage and metabolism may explain the lower immunogenic activity of endralazine.

After 3 months of therapy with indapamide 2.5 mg/d the mean arterial pressure (MAP) had decreased from 135±6 to 112±2 mmHg (p〈0.001); the heart rate (HR) had not changed, VR had decreased from 6.1±1.2 to 4.8±1.8 (pg·min·kg)−1 (p〈0.05), and BRS had increased from 8.3±3.7 to 12.2±5.3 ms/mmHg (p〈0.005), with a leftshift of the relationship between BP and heart period. An inverse correlation was found between the pre-treatment systolic blood pressure and the change in baroreceptor sensitivity after indapamide (r=0.59; p〈0.05). In conclusion, chronic treatment with indapamide enhances BRS and resets the reflex. The resetting may account for the lack of tachycardia at rest observed after treatment with indapamide. The mechanism by which indapamide interferes with the baroreceptor reflex requires further investigations.

    Notes: Summary In a double-blind, cross-over trial, 10 men with primary hypertension, not adequately controlled with a β-blocker alone, were also given felodipine or placebo for periods of one week. Placebo was administered single-blind for 2 weeks and 1 week, respectively, before randomization and between treatments. The dose of felodipine ranged from 6.25 mg to 25 mg. The addition of felodipine resulted in a pronounced (20%), statistically significant reduction in blood pressure (BP) and a small but significant increase in heart rate (HR). The effects were seen within 1–2 h and were maximal after 3–4 h. During steady state treatment the duration of BP reduction was at least 12 h. No orthostatic reaction was seen. There was a significant correlation between the plasma concentration of felodipine and change in BP. The most frequently reported side-effects were headache and ankle oedema, the latter probably being due to pronounced pre-capillary vasodilatation. There was no weight increase and thus no indication of general water retention. No clinically significant change in laboratory variables and no influence on the P-Q time were seen. Thus, felodipine in combination with a β-blocker seems to be a useful addition to the treatment of hypertensive patients whose BP is not adequately controlled with a β-blocker alone.

    Notes: Summary The influence of various disease states, other than hypertension, on the pharmacokinetic behaviour of hydralazine is not completely known. In the present study the pharmacokinetics of oral hydralazine has been evaluated in 7 patients with severe, chronic heart failure, using 8 compensated hypertensives as controls. The pharmacokinetics was evaluated by measuring the plasma concentrations of hydralazine (“apparent” and “real” hydralazine) and hydralazine pyruvate hydrazone, and by assessing acetylator phenotype after a small dose of dapsone. The AUC (area under the plasma concentration curve) following a single, oral 50 mg dose was significantly larger in patients with chronic heart failure NYHA Class III–IV than in patients with essential hypertension without cardiac decompensation. A decreased rate of hepatic elimination of hydralazine is suggested as a major contributory factor to this finding.

A group of 20 patients whose blood pressure was unsatisfactory on a 3 drug regimen had their third drug stopped and after a 2 week period nifedipine was added to their beta-blocker plus diuretic therapy. Eleven became normotensive on 30 mg nifedipine daily and a further 6 on 60 mg daily; giving on overall success rate of 85%. This result was achieved with a reduction in side effects and an absence of any haemodynamic or metabolic complications.

    Notes: Summary To evaluate the role of adrenergic mechanisms in the acute response of renin to furosemide, plasma renin activity (PRA) and plasma catecholamine concentrations were measured for 3 h after i.v. administration of furosemide 1 mg/kg to 8 patients with mild essential hypertension. Furosemide induced a prompt and long-lasting increase in renin, with PRA more than doubled at all times. The increase in PRA within the first 30 min paralleled the peak increases in urinary water and sodium flow rates, and significant decreases in plasma volume and central venous pressure. There was no change in plasma catecholamine concentrations. Plasma noradrenaline was increased significantly at 60 min and adrenaline at 90 min, once furosemide had induced a marked loss of body-fluid and ∼65% decrease in central venous pressure. Both catecholamines remained elevated until the end of the study, whereas urinary water and sodium flow rates had returned to their pre-treatment values by 150 min. Mean blood pressure was essentially unchanged throughout the study, whereas heart rate increased significantly after 90 min. The findings suggest that in mildly hypertensive patients adrenergic mechanisms are not involved in the initial renin response to furosemide, but they come into play later, probably as a result of reflex sympathetic activation triggered by marked volume depletion.

    Notes: Summary In an open study increasing doses of a retarded tablet formulation of pinacidil were given twice daily for four weeks to 9 patients with untreated essential hypertension (WHO I–II). In all patients a decrease in diastolic blood pressure to below 100 mmHg, or a fall exceeding 15 mmHg, was obtained 2 h after tablet intake (p〈0.02), but in only two patients was the effect maintained after 10 hours (n.s.). At a mean serum concentration of 100 ng/ml 2 h after pinacidil 30 mg, the mean blood pressure had decreased by 14 and 12.7 mmHg in the supine and erect positions, respectively (p〈0.05). In contrast, mean blood pressure 10 h after the same dose was unchanged, when the mean serum concentration was 47.5 ng/ml. No change in heart rate was observed. Pharmacokinetic and pharmacodynamic investigations showed a tendency towards a more gradual and longer lasting antihypertensive effect and serum concentration-time curve after the retarded tablet than the previous tablet. Pinacidil 40 mg in the retarded tablet reduced mean blood pressure and increased heart rate for at least 8 h. There was a linear correlation between the serum concentration and the changes in mean blood pressure, and between the changes in mean blood pressure and in heart rate. There was no indication of tachyphylaxis. A serum level of 50 ng/ml of pinacidil appeared to be the minimal effective concentration. The side effect consisted of fluid retention, and the body weight increased by 1.0 kg (p〈0.05); four patients complained of oedema. Therapy was discontinued in one patient after a fainting episode following an increase in the dose. Thus, pinacidil was able to lower blood pressure during monotherapy for 4 weeks provided that an adequate serum concentration was achieved. The present retarded tablet formulation is not suitable for b. d. dosing. The tendency towards fluid-retention suggests that pinacidil should be used in combination with a diuretic.

    Notes: Summary Nifedipine, a calcium antagonist with a predominant vasodilator action, was evaluated for the treatment of hypertension. A 20 mg-tablet, with a slower absorption and a more sustained blood-level than provided by the 10 mg-capsule was administered to 20 patients. The duration of the trial was 20 weeks. All patients achieved a significant reduction in both systolic (p〈0.05) and diastolic (p〈0.001) blood-pressure (B.P.), but 10 patients were withdrawn before completion of the trial period. Two patients, although achieving a fall in B.P. which was significant, did not reach to target level (〈160/90) on maximal dosage, one patient suffered a stroke due to a cerebral infarct, and seven patients were withdrawn because of side-effects due mainly to vasodilatation. The remaining 10 patients obtained a satisfactory response. In nine patients, who had achieved a satisfactory result, there was no change in plasma renin activity (P.R.A.) during chronic nifedipine administration.

    Notes: Abstract Species composition and density of cladoceran populations changed in Lakes Zürich, St. Mortiz and Baldegg as human populations increased in these watersheds. Lake Zürich sediments became annually laminated in the 1890's as a result of increased organic input as the size of the cities surrounding the lake grew. At the same time, the Bosmina species changed from a oligotrophic form (longispina) to a eutrophic form (longirostris). An increase in Daphnia spp. populations also occurred at this time in the lake's history. Bosmina longispina reappeared in the lake in 1965 as the lake's trophic status changed from eutrophic to mesotrophic due to effective sewage treatment facilities. Annual laminations appear in the Lake St. Moritz sediments about 1910. Shortly thereafter, a shift from B. longispina to B. longirostris occurred. This change in trophic status is associated with increased tourism in the area. Lake Baldegg sediments also show annual laminations beginning in 1885 and a similar shift in the Bosmina species. Other cladoceran remains were too scarce to be useful in interpreting the histories of these lakes.

    Notes: Abstract Magnetic susceptibility measurements have been used to correlate synchronous depths in ca. 50 l m. cores of recent sediment from a small lake (area ca. 55 ha) in Scania, southern Sweden. The three-dimensional picture of sediment accumulation which emerges provides a basis for studying sediment deposition patterns through time. Using a palaeomagnetic chronology, the results show that the pattern and rate of accumulation have dramatically altered during the past 350 years, thus making semiquantitative studies of downcore sediment properties in a single core problematical. Possible reasons for these phenomena are briefly discussed.

    Notes: Abstract Traunsee is the deepest (189 m) lake in Austria and has been holomictic until the beginning of industrial discharges from the alkali works in Ebensee. The input of soluble compounds such as CaCl2 and NaCl have produced meromictic conditions. The relatively high oxygen content of the monimolimnion is due to water movements promoted by the large inflow and wind-action. The industrial discharges have caused both meromictic conditions and the deposition of highly alkaline sediment in the southern portion of the profundal zone. The effect of the discharges on the benthic fauna was evaluated by comparing the subfossil benthic fauna of four cores taken inside and outside the affected area. The ostracod fauna resembles that of other large lakes in the Salzkammergut, but also includes groundwater species such as Kovalevskiella and Cypridopsis subterranea. Leucocythere mirabilis has been found for the first time in an Austrian lake. Ostracods and other benthic organisms such as Testacea were not apparently influenced by the meromictic condition. They are, however, missing in all areas covered by the alkaline sludge.

    Notes: Abstract Microfossil analysis was carried out on 90 samples of a sediment core from Grossensee. The diversity of subfossil Cladocera, Chironomidae, Chaoborus, and Ostracoda reflects at least five different stages of development. Three extended cycles correspond to different climatic periods in the first 10 000 years of the lake's history, while during the last 2 000 years, two shorter cycles can be identified which correlate with proofs or even historical events for human activity in the catchment area. During this time the profundal fauna changed from stenoxibiontic to euryoxibiontic species. In the planktonic fauna a succession of three Eubosmina types, B. longispina, B. coregoni kessleri, and B. coregoni coregoni, was observed. The chydorid and ostracod assembly of the littoral showed no clear change.

    Notes: Abstract The distribution of 137Cs and 239,240Pu in sediment core samples of the Finnish lakes Laukunlampi, Lovojärvi and Pääjärvi were determined. The sediment samples were collected using dry ice and liquid nitrogen freezing methods. The sediments of these lakes are annually laminated. A clear maximum concentration of 137Cs and 239,240Pu was found in sediment layers formed during 1962–1964, the years of maximum fallout, and the middle of the 1950's can be estimated from the 137Cs and 239,240Pu profiles. The highest concentrations, 11 500 and 820 pCi kg−1 dry wt for 137Cs and 239,240Pu, respectively, were found in the sediment of Laukunlampi. The vertical distribution was similar for 137Cs and 239,240Pu in the lakes investigated. A slight migration of 239,240Pu and 137Cs was found and the migration of 137Cs seems to be higher than that of 239,240Pu. The advantages of 137Cs dating method are rapidity and simplicity. 239,240Pu is preferable when the sample size is small. The agreement found between 137Cs and 239,240Pu dates and the annual laminae show that these fallout radio isotopes can be used for dating sediments formed during the past 25 years.

Despite this, considerable doubt still surrounds the nature of the processes by which 210Pb is deposited in lake sediments, and this has lead to a situation where there is a choice of dating models offering different interpretations of 210Pb data. In assessing 210Pb data it is therefore essential to first of all determine whether data is consistent with the assumptions of the dating model, and to then compare the 210Pb chronology with independent dating evidence. We have tested 210Pb data from a wide variety of sites, and our calculations indicate that the crs (constant rate of 210Pb supply) model provides a reasonably accurate chronology when the total 210Pb contents of cores from neighbouring locations are comparable.

    Notes: Abstract Hidden Lake is a perched, brown-water lake located in the centre of Great Sandy Island (Fraser Is.), S.E. Queensland. It is highly acid (pH 4.0), oligotrophic and is thermally and chemically stratified for most of the year. The sediments revealed a 137Cs profile which departed from the temporal pattern of 137Cs fallout in Brisbane and was represented by an exponential increase of 137Cs towards the surface sediments from ca. 32 cm depth. The possible causes of the divergent profile are discussed, including physical and biological mixing, lag in the transport of catchment material to the sedimentary basin, diffusion, recycling and biological concentration. It is hypothesised that a combination of the last four processes, with diffusion facilitated by the highly acid conditions, are the major causes of the observed 137Cs profile. Possible recycling and bioconcentration of 137Cs raises questions as to the validity of this method of dating in similar environmental conditions, and as to the interpretation of other palaeochemical data. These hypotheses are to be tested against profiles obtained from 14C, 210Pb, 239/240Pu analyses of the sediment, and the measurement of 137Cs activity in the water and biota of the lake.

    Notes: Abstract Previous measurements of the 14C activity of lake sediments and Potamogeton plants from Säynäjälampi, collected in 1970, indicated a severe hard-water effect (Donner et al. 1971). Samples from new cores, collected in 1977, were sliced into thin sections down to 18 cm. The 14C concentration in samples down to 7 cm was similar to that obtained by the Helsinki Laboratory. The corresponding apparent 14C age is almost 2 000 years. Light-coloured layers, apparently due to catchment drainage, were used together with density diagrams to correlate the cores. The drainage history gives a time-scale showing that the lowest samples dated to correlate the cores. The drainage history gives a time-scale showing that the lowest samples dated were just old enough to be pre-bomb sediments. Three samples of submerged plants were also examined, indicating some exchange with the air and an appreciable reservoir age. The activity of the plants was significantly lower in 1978 than in 1970. One sample, well below the lowest drainage stratum, indicates a reservoir age of ca. 1 100 years. This sample yielded an apparent age of the INS fraction which was significantly different from that of the SOL fraction. The difference was determined to ca. 600 years. Total 210Pb was determined via 210Po using isotope dilution while 226Ra was measured with the help of radon emanation. The 226Ra was used to estimate the supported 210Pb. The unsupported 210Pb showed a higher sediment accumulation rate for the top nine cm of one core (the length investigated was 16.5 cm). The varying accumulation rate was explained by human activities, as could be seen from the heterogenous sediment composition of the cores. Recent vegetational change is described from direct observations since 1957. Pollen analysis does not give any reliable indication of agriculture.

    Notes: Abstract The recent sediments of Frains Lake, Michigan contain a rich and well preserved association of chrysophycean cysts. Forty one forms are revealed by scanning electron microscopy (SEM) and light microscopy (LM). Taxonomic descriptions. and SEM micrographs are provided for the dominant forms. The three dominant taxa throughout the sediments, Cysta minima, C. modica and C. subbavaricum, do not show significant shifts in proportional abundance associated with European settlement and the onset of cultural eutrophication. However, certain subdominant taxa do show clear trends. Density counts indicate a dramatic decline in cyst concentration (by volume and by dry mass) and a sharp increase in absolute accumulation (net annual influx) following settlement. The Frains Lake profile of chrysophycean cysts is compared to sequences of other North American and European temperate lakes. The utility of chrysophycean cysts as paleoenvironmental indicators is considered on the basis of these results.

    Notes: Abstract X-ray radiographs of unextruded sediment cores are used for the documentation and the interpretation of primary and secondary sedimentary structures, and for the correlation of synchronous laminations between different cores. The vertical variation in bulk density, water content and void ratio, and the increase of solids with increasing sediment depth are calculated from the recorded film density along the radiographs. The X-ray radiographic technique for the study of sedimentary properties is fast and non-destructive. The technique is especially valuable when studying the uppermost part of the sediment cover and the processes of sediment redistribution and sediment accumulation. Studies are being undertaken in several Swedish lakes and coastal bays, where mono- and stereoradiographs are being used to classify bottom and sediment types, to interpret changes in the depositional environment, and to calculate past and present rates of sediment accumulation.

    Notes: Abstract The salinity of lakes is subject to variations imposed by climatic changes. These variations are recorded in the salinity profile of pore waters. Meromictic lakes, such as the Dead Sea, are a special case where waters which underlie the mixolimnion reflect salinity variations. In a sediment core from Dead Sea shallow waters, the salinity profile exhibited a minimum at about 1.9 m depth. It is shown by a diffusion model that this minimum can be attributed to lower salinities which prevailed at the sediment water interface for several decades around the turn of this century. No such minimum was observed in a sediment core from the deepest part of the lake where, during the last two centuries, the overlying brines had a constant salinity.

    Notes: Abstract The recent history of Little Round Lake, a small meromictic lake in southeastern Ontario, is considered. Pollen analyses were used to identify past changes in terrestrial vegetation, whilst limnological conditions were interpreted on the basis of diatom and chrysophyte microfossils. Contemporaneous with the arrival of European settlers (ca. A. D. 1850), the predisturbance assemblage of oligotrophic Cyclotella diatoms was replaced by Synedra spp., which then succeeded to a eutrophic flora dominated by Stephanodiscus hantzschii. Meanwhile, synuracean algae were almost completely excluded. Over the last 30 years, the algal microfossils indicate that the lake underwent a marked return to oligotrophy. Evidence is presented which suggests that this shift was related to the cultural enhancement of meromixis by the seepage of road salt.

    Notes: Abstract Lake Balaton, the largest shallow lake in Central Europe, is about 20 000 years old. An enormous increase in tourism and the disproportionate building development of the last few decades has resulted in the acceleration of eutrophication in the lake. Widespread research to reveal the causes of water-quality deterioration and possible ways of protection against it have recently started. The investigation of the larvae of non-biting midges (Diptera: Chironomidae) in the sediment of the open-water zone has also begun. The contemporary faunal composition strongly correlates with the trophic gradient along the longitudinal axis of the lake. We therefore supposed that the eutrophication process should be identifiable from the analysis of subfossil chironomid head capsules from the upper (15 cm thick) layer of the sediment. We found that quantitative results could only be obtained when fragments as well as relatively intact head capsules are considered. Our data verify that the originally oligo-mesotrophic community has been gradually replaced by eutrophic species in a west to east direction. Large-bodied larvae belonging to the Chironomus plumosus group mix the sediment down to 15 cm as they build their tubes and consequently alter the original proportions of head capsules at the different levels. So the sequence of communities through the sediment-layers is not quite reliable.

    Notes: Abstract The horizontal and vertical distribution of the gram-positive, non-motile, spore-forming and rod-shaped bacterium Clostridium perfringens Holland was studied. The aim of the study was to estimate the quantity of C. perfringens at different depths of the sediment and evaluate the effect of human effluent which the lake received between 1940 and 1956. C. perfringens lives in the colon of man. Because it is spore forming and cannot multiply under a temperature of 20 °C and, according to the studies of Seppänen et al. (1979) it can be at least 300 years old, it may be a suitable paleolimnological indicator of pollution by human effluent. The results showed that the amounts of Clostridium increased at the same level where redox potential decreased in the sediment due to the beginning of effluent disposal at a depth of 40 mm. The maximum number of Clostridium colonies occurred between 0–30 mm depth.

    Notes: Abstract The transition from an arid, glacial Late Pleistocene to an early Holocene (Gamblian ‘pluvial’) moist period has not been documented in Central America. Finding accessible volcanic lakes too youthful, and knowing that most Florida and Yucatan lakes were dry during glacial ages, we sought appropriate deposits lying deeper than 40 m in the deeper karst lakes of the Peten, in northern Guatemala. The bottom half of a 19.7 m core from Lake Quexil, and the bottom third of a 15 m core from Lake Salpeten, appear to be of Pleistocene age. The sediments contain lacustrine shells, sponge spicules, and Pinus pollen, and include several bands of humified gyttja with fragments of wood, but are dominantly montmorillonitic and mixed-layer clays and may be in part colluvial, like the later Holocene Maya clay. Calcite, gypsum, and (in presently saline Lake Salpeten only) dolomite indicate shallow, closed, moderately saline lakes 30–40 m lower than at present. In both cores a layer of inorganic sediment with gypsum dominant, perhaps recording the most arid phase of the glacial Late Pleistocene, overlies a similar clay layer with calcite dominant. As calcite and dolomite occur throughout the section(s), both minerals are believed to be detrital, but one source of calcite is algal crusts, formed and exposed today in the littoral and supralittoral zones. The early Holocene rise of lake levels formed several meters of fossiliferous gyttja with pollen of mesic tropical forest, now assigned to the pan-tropical Gamblian moist episode. Where deposited in oligomictic or meromictic lakes 〉 30 m deep, Gamblian gyttja of pollen zone Pl is finely laminated, the dark layers being richer in Ptot and Stot and poorer in Fe, Mn, Mg, and K than the light (clay) layers, but we cannot yet say that the laminae are annual.

    Notes: Abstract Pollen analysis was carried out on the sediments of 10 lakes in eastern Holstein. Sediment accumulation rates varied considerably both from lake to lake and also between respective periods. Increases in sediment accumulation rate often coincide with the Neolithic settlement periods. Faunal remains have been examined from two of these lakes (cf. Günther 1983; Hofmann 1983).

    Notes: Abstract The following stages in the formation of sediments in Lake Baikal have been distinguished. In the Middle Miocene lacustrine deposits accumulated in the southern depression. In Early and Middle Pliocene, as well as in Early-Middle Pleistocene the bottom of the southern and middle Baikal depressions was occupied by large lakes, but the northern basin was mainly dry with a system of small lakes, streams and rivers. This accounts for the great facies variety and the genetic heterogeneity of sediments in the northern depression. At the end of Middle and in the Late Pleistocene an abrupt warping of the depression took place, and a shore-line similar to the present one was formed. Sands in the upper part of the sediments are considered to be the product of intensive erosion which probably occurred in the pluvial epoch in the Late Pleistocene period.

    Notes: Abstract Laminated sediment is deposited in the deep areas (〉 30 m) of Lake Pääjärvi. In the diatom microstratigraphy a regular alteration of planktonic and littoral floras, which goes in pace with the visual structure, reflects the yearly cycle of sedimentation. Considerable variation in sedimentation was found to have occurred during the past two decades. A sequence, dated to the early 1960's, in which the organic content was exceptionally high, could be identified in the sediment at 6–14 cm depth even in shallower areas where laminations are not formed. This level was used as a marker horizon when determining the recent distribution of sediment by means of 19 cores taken from different parts of the lake. The average annual sediment accumulation in the profundal zone of the lake is 340 g m−2a−1 dry weight, of which 25 g m−2 a−1 (1100 kJ m−2 a−1) is organic carbon.

    Notes: Abstract The Early and Middle Flandrian geological development and paleolimnology of Lake Spitaalijärvi was studied using pollen, diatom and cladoceran analysis and 14C dating. Spitaalijärvi was isolated from the Ancylus Lake about 9000 B.P., at which time birch and pine and plants typical of open habitat communities grew on the solitary island. The rational limits for Alnus and Picea were ca. 8300 B.P. and 3700 B.P., respectively. During the first few hundred years after isolation Spitaalijärvi was probably fairly eutrophic, with a low water level. Water level began to rise before the Alnus rise (A°) and the lake became oligotrophic. After another transgression, which started before the Picea rise (P°), Tabellaria binalis and Semiorbis hemicyclus appear in the diatom stratigraphy indicating ultra-oligotrophic conditions. The main reason for the ultra-oligotrophy of Lake Spitaalijärvi is the character of the parent material in the catchment, consisting of glacial and littoral deposits derived from the local quartz sandstone of highly siliceous nature.
