    Notes: Abstract Polyribosomes (polysomes), active in an amino acid incorporation system in vitro, were isolated from tobacco leaf protoplasts. A comparison of polysome profiles indicated that the polysome/monosome ratio is greatly decreased in isolated protoplasts as compared to the intact leaf. In isolated protoplasts, a marked accumulation of ribosomal subunits was also found. The division of protoplasts, as investigated in the 8-cell and callus stages, was associated with a(n) (at least) partial regeneration of polysome profiles characteristic for leaves. Plasmolysis of leaves attached to the plant had no great effect on the polysome profile. However, leaf excision per se resulted in a dramatic loss of polysomes, even when the leaf tissue was floated on water. It is concluded that the isolation of the cell from its normal environment, and not the osmotic stress and associated increase in RNase activity, is the most important factor responsible for the loss of polysomes in isolated protoplasts.

    Notes: Abstract Forty-eight samples of 9 mm diameter articular cartilage and associated subchondral bone from, the tibial plateau of human knee joints were mounted in methylmethacrylate cement and subjected to controlled impact velocities. The controlled velocities provided for the testing of 21 samples at a strain rate of 500 s−1 and 27 samples at a strain rate of 1000 s−1. Data are presented on the energy absorption per unit volume of cartilage in relation to the associated stresses and strains. The data in each of the resultant nine graphs are subjected to a least squares fit to the polynomial $$Y = a_1 X + a_2 X^2 $$ The relevant statistical data for each of the strain rates and for the combined data are presented. While the total energy absorption of the articular cartilage of the knee joint at ambulatory stresses of 0·8 to 6·3 N/mm2 is calculated to be no more than 0·13 to 3·65 J, it is suggested that the compliance of the cartilage of the knee joint together with the incongruity of the tibia and femur are essential features that should be considered in the design of total knee-replacement prostheses.

    Notes: Abstract This paper reviews many of the mechanical properties of faulted and jointed rock under pressure and temperature and in the presence of water. At low effective confining pressures (below about 1 kilobar), the friction strength is quite variable and depends on the frictional resistance between gouge particles or asperities and on the dilatancy of the fault. At higher pressures the friction strength is nearly independent of mineralogy, temperature, and rate, at least for rocks whose friction strength is less than the failure strength. Water tends to slightly weaken the fault. The type of sliding motion, whether stick-slip or stable sliding, is much more affected by environmental and mineralogical factors. In general, stick-slip is dominant at high pressures and low temperatures, in the presence of strong minerals such as quartz and feldspar, in the absence of gouge, for lower surface roughness, and perhaps in the presence of water. The microscopic deformation mechanisms are poorly understood. At low temperatures, cataclasis dominates in rocks containing mostly quartz or feldspar, and plastic deformation in rocks containing mostly calcite or platy silicates. At high temperature most minerals deform plastically, producing a greater temperature-and rate-dependence of the friction strength. Glass has been found in some sliding surfaces in sandstone.

    Notes: Synopsis The relationship between standing in a dominance hierarchy and physiological stress was studied in rainbow trout. Individual fish were assigned relative dominance ranks, based on behavioral observations in a large, simulated stream tank. These ranks were compared to histometric measures of interrenal cell activity. Fish, isolated individually in the stream tank had significantly lower levels of interrenal activity than fish from the crowded holding tank. Groups of fish in the stream tank formed stable, linear dominance hierarchies. Interrenal activity correlated inversely with dominance rank, with the exception that top ranking fish had higher activity than expected. Possible cause and effect relationships are discussed.

    Notes: Summary Neurohypophyses from sexually mature female sturgeons, Acipenser güldenstädti Brandt, killed before, soon after spawning, and during down-stream migration were studied light and electron microscopically. Ovaries were examined only under a light microscope. A large amount of neurosecretory material (3.5–4.5 arbitrary units) is found in the neurohypophysial “roots” during up-stream migration to spawning grounds. Neurosecretory fibres and their terminals are replete with elementary neurosecretory granules. Side by side with the latter some terminals contain single residual granules and few “synaptic” vesicles. Soon after spawning the amount of neurosecretory material decreases markedly in most individuals (content up to 1–3 units), and elementary granules are few in the fibres and their terminals. The number of residual granules increases, and “synaptic” vesicles are especially numerous. Granulated, disintegrating neurosecretory granules and granule-shadows occur in the preterminal parts of the fibres. The number of pituicytes increases because some “light” tanycytes seem to migrate from the ependymal into the subependymal layer. The restoration of neurosecretory material (reaching again up to 3.5–4.5 units) occurs within a month after spawning. The number of elementary granules increases in fibres and terminals while that of residual granules and “synaptic” vesicles decreases. These changes in the neurohypophysis of females after spawning are due to discharge of great amounts of peptide neurohormones into the general circulation after spawning. Spawning is regarded as a “physiological” stress for these females.

    Abstract: Résumé Influence de la résistance des particules sur la compression d'un agrégat broyé Les auteurs examinent les paramètres qui affectent la compressibilité des agrégats de roches et présentent des résultats expérimentaux obtenus avec des échantillons de calcaire, de dolérite et de grès, sur l'influence du diamètre des grains en-dessous de 19 mm. Les résultats suggèrent que l'indice des vides initial impose le taux de la déformation à la compression dont on dispose, cependant que le diamètre des grains (d m) et la résistance à la compression (S c) définissen les caractéristiques de la courbe de compressibilité.
    Notes: Summary The Effect of Particle Strength on the Compression of Crushed Aggregate The paper examines the variables controlling the compression of rock aggregates, and presents experimental data on the behaviour of limestone, dolerite and sandstone samples in size ranges up to 19.0 mm.     Description / Table of Contents: Résumé L'effet d'une alimentation, riche en calcium, par rapport à un régime sans calcium, est déterminé au niveau de la diaphyse tibiale chez le rat. Le régime sans calcium provoque une augmentation de la région médullaire et l'administration consécutive d'un régime riche en calcium provoque une diminution de cette région. Cependant, la proportion d'endoste perdue en 10 jours de régime sans calcium n'est pas reconstituée après 78 jours de régime riche en calcium. La diminution de zone médullaire est en rapport avec la diminution de résorption de l'endoste et surtout avec une apposition augmentée de l'endoste. La formation osseuse périostée et épiphysaire est inchangée: ce qui indique que le régime, riche en calcium, ne provoque pas une augmentation généralisée de l'ostéogenèse. L'augmentation de la formation d'endoste est limitée aux régions d'endoste les plus résorbées, au cours de la période de manque en calcium. Ce fait indique qu'un facteur local est partiellement responsable de la stimulation de l'apposition osseuse de l'endoste pendant cette période. Un stress mécanique, qui stimule la formation osseuse, est augmenté chez les animaux privés de calcium et revient à la normale après administration de calcium. De plus, le stress mécanique est probablement le plus important dans les régions d'apposition osseuse et pourrait constituer le facteur local, responsable de l'augmentation de l'endoste, pendant l'administration de calcium.
    Abstract: Zusammenfassung Bei Ratten, die nach einer Calcium-freien eine Calcium-reiche Diät erhielten, wurden die hieraus resultierenden Veränderungen der Tibia-Diaphyse bestimmt. Das Calcium-freie Futter verursachte eine Zunahme des Markraumes und die anschließende Verfütterung einer Calcium-reichen Diät hatte eine Reduktion des Markraumes zur Folge. Die Menge des endostalen Knochens, die während der 10 tägigen Verfütterung der Calcium-freien Diät verloren ging, war jedoch nach 78 Tagen Calcium-reicher Diät nicht voll ersetzt. Die Abnahme des Markraumes wurde durch eine geringere endostale Knochenresorption und besonders durch eine erhöhte endostale Knochenbildung hervorgerufen. Die Knochenbildung am Periost und an den Epiphysen war unverändert, ein Zeichen dafür, daß die Calcium-reiche Diät keine generalisierte Zunahme der Knochenbildung zur Folge hatte. Die Zunahme der endostalen Knochenbildung beschränkte sich auf jene Bereiche entlang dem Endost, wo während der Calcium-Mangelperiode am meisten Knochensubstanz verloren gegangen war. Dies weist darauf hin, daß ein lokaler Faktor mindestens teilweise verantwortlich ist für die Stimulierung von endostaler Knochenbildung während der Calcium-Sättigungsperiode. Die mechanische Belastung, welche die Knochenbildung anregt, war bei Calcium-armen Tieren erhöht und kehrte während der Calcium-Sättigung wieder zur Norm zurück. Zusätzlich war die mechanische Belastung wohl dort am größten, wo die stärkste Knochenbildung stattfand, und diese Belastung könnte somit als der lokale Faktor angesprochen werden, welcher während der Calcium-Sättigungsperiode zur gesteigerten endostalen Knochenbildung beitrug.
    Notes: Abstract The changes in the tibial diaphysis as a result of feeding a high calcium diet to rats previously fed a calcium free diet were determined. The calcium free diet resulted in an increase in the medullary area, and the subsequent feeding of a high calcium diet caused a reduction in medullary area. However, the amount of endosteal bone lost during ten days of feeding a calcium free diet was not completely restored after 78 days of feeding a high calcium diet. The decrease in medullary area was brought about by decreased endosteal bone resorption and particularly by increased endosteal bone formation. Bone formation at the periosteum and at the epiphyses were unchanged, indicating that the high calcium diet did not cause a generalized increase in bone formation. The increase in endosteal bone formation was limited to those sites along endosteum where greatest loss of bone had occurred during the calcium depletion period. This indicates that a local factor is at least partially responsible for the stimulation of endosteal bone formation during calcium repletion. Mechanical stress, which stimulates bone formation, was increased in calcium deficient animals and returned to normal during calcium repletion. In addition, mechanical stress was probably greatest in those sites where the greatest amount of bone repletion occurred and may have been the factor which contributed to the increase in endosteal bone formation during calcium repletion.

    Description / Table of Contents: Résumé Un mécanisme nouveau tente d'expliquer comment des forces mécaniques augmentées ou diminuées, appliquées à l'os, sont transformées en activité ostéoblastique et/ou ostéoclastique. Une hypothèse mécano-chimique du remaniement osseux, induit par stress mécanique, est émise pour expliquer ce phénomène. L'os répond á un stress mécanique par une croissance différentielle de façon á résister au stress appliqué: par suite le remaniement osseux induit mécaniquement est probablement sous le controle d'un système “feedback” négatif. L'hypothèse consiste dans le fait qu'un changement de charge au niveau de l'os modifie la tension au niveau des cristaux d'apatite de l'os. Ceci modifie la solubilité des cristaux qui fournit ainsi le message de “feedback” négatif nécessaire aux cellules osseuses, sous la forme d'un changement chimique, indiut par un phénomène mécanique. Les cellules aggissent alors pour compenser les modifications de l'activé calcique localisée soit en construisant de l'os pour redistribuer le stress augmenté, ou en résorbant de l'os, qui est en surplus des besoins structuraux, liés à un stress réduit. Pour tester cette hypothèse, des cristaux d'hydroxyleapatite sont soumis à un stress et des changements d'activité ionique du calcium sont enregistrés à l'aide d'une électrode à cation divalent. Les résultats démontrent qu'un effet mécano-chimique peut etre détecté dans les cristaux d'apatite qui, soumis à un stress, engendrent une activité en calcium de 9×10−5 moles/litres et de 7×10−5 moles/litre, dans des conditions sans stress. Les résultats expérimentaux de cette étude et les données de la physiologie cellulaire s'accordent avec l'hypothèse mécano-chimique proposée.
    Abstract: Zusammenfassung Um zu erklären, in welcher Weise auf Knochen applizierte, erhöhte oder verminderte mechanische Stresse zu einer Osteoblasten- und/oder Osteoklastenaktivität führen, wird ein neuer Mechanismus vorgeschlagen. Eine mechanochemische Hypothese der Knochenneubildung, verursacht durch mechanische Stresse, wird besprochen, um dieses Phänomen zu erklären. Knochen reagiert auf mechanische Stresse mit differenziertem Wachstum, um damit dem angewandten Stress zu widerstehen; daraus folgt, daß mechanisch induzierte Knochenneubildung vermutlich durch ein negatives Feedback-System reguliert wird. Die Hypothese besteht darin, daß eine Veränderung der Ladung des Knochens eine Veränderung der Spannung der Hydroxyapatitkristalle im Knochen zur Folge hat. Dies wiederum verändert die Löslichkeit der Kristalle und gibt damit die benötigte negative Feedback-Botschaft an die Knochenzellen in Form einer mechanischinduzierten chemischen Umgestaltung weiter. Die Zellen verhalten sich nun dementsprechend, um der Änderung der örtlichen Calciumaktivität entgegenzuwirken; entweder wird Knochengewebe gebildet, um einen erhöhten Stress neu zu verteilen, oder jener Knochenanteil wird beseitigt, der bei reduziertem Stress einen Überschuß der Strukturbedürfnisse darstellt. Um diese Annahme zu überprüfen, wurden synthetische Hydroxyapatitkristalle einem Stress unterworfen, und die Änderung in der Aktivität der Calciumionen wurde mittels einer divalenten Elektrode zur Messung der Kationenaktivität registriert. Die Resultate zeigen, daß ein mechanochemischer Effekt in den Hydroxyapatitkristallen nachgewiesen werden kann, die unter Stress eine Calciumaktivität von 9·10−5 Mol/Liter, dagegen ohne Stress eine solche von 7·10−5 Mol/Liter erzeugen. Die Ergebnisse dieser Untersuchungen einerseits und die Aussagen der Zellenphysiologie andererseits stimmen mit der hier vorgeschlagenen mechanochemischen Hypothese überein.
    Notes: Abstract A new mechanism is presented to explain how increased or decreased mechanical stresses applied to bone are translated into osteoblastic and/or osteoclastic activity. A mechano-chemical hypothesis for bone remodeling induced by mechanical stress is presented in an attempt to explain this phenomenon. Bone responds to mechanical stress by differential growth so as to resist the applied stress; therefore mechanically induced bone remodeling is probably regulated by a negative feedback system. The hypothesis is that a change in the loading of bone results in an altered straining of the hydroxyapatite crystals in bone. This in turn alters the solubility of the crystals, providing the required negative feedback message to the bone cells in the form of a mechanically induced chemical change. The cells then take appropriate action to compensate for the alteration in the localized calcium activity either by building up bone to redistribute an increased stress, or by removing bone which is surplus to the structural needs imposed by a reduced stress. In order to test the hypothesis, synthetic hydroxyapatite crystals were stressed and changes in calcium ion activity were recorded from a divalent cation activity electrode. The results show that a mechanochemical effect can be detected in hydroxyapatite crystals which, when stressed, generate a calcium activity of 9×10−5 moles/l compared to 7×10−5 moles/l when unstressed. The experimental results in this study and evidence from cellular physiology are consistent with the mechanochemical hypothesis proposed here.
