    Notes: Summary In the course of studying the plasma adrenocorticotropic hormone (ACTH) and corticosterone responses to synthetic corticotropin releasing factor (CRF), we noted some disparity in the responses. A higher dose (20 μg compared with 5 μg per rat i.a.) produced an equal plasma ACTH but greater plasma corticosterone response in adult male rats. Thus, we examined the possibility that CRF increases adrenocortical responsiveness to ACTH. CRF significantly (p〈0.0005) increased the plasma corticosterone response to ACTH in rats pretreated with dexamethasone. Thus, synthetic CRF increases corticosterone secretion in rats not only by stimulating ACTH secretion, but also by increasing the adrenocortical responsiveness to ACTH.

    Notes: Summary Surface carbohydrates on the Merkel cell of the outer root sheath (ORS) were investigated in 1to 4-day-old rat vibrissae by use of rhodamine isothiocyanate (RITC)-conjugated lectins. The red fluorescence of RITC provided a convenient assay for lectin binding to the Merkel cell, which is itself identified by its green fluorescence following selective uptake of the dye quinacrine. In monolayers or suspensions of freshly dissociated ORS cells, the Merkel cell showed high affinity for the α-fucose-specific lectin, Ulex europeus agglutinin I (UEA-I), thus revealing a novel feature for a basally located cell. Other high-affinity lectins included concanavalin A (Con A), wheat germ agglutinin (WGA), soybean agglutinin (SBA), and Ricinus communis agglutinin I (RCA-I). In contrast, Dolichos biflorus (DBA), Bandeiraea simplicifolia I and II (BS-I and BS-II), and peanut agglutinin (PNA) virtually excluded the Merkel cell, though PNA-binding sites were unmasked after neuraminidase treatment. Other dispersed ORS cells had varying lectin affinities, and generally binding was inhibited by a competing haptenic sugar. The pattern of lectin binding seen in cryostat and paraffin sections of the vibrissa suggested that the Merkel cells share surface properties with their neighboring basal and/or spinous cells; however, unshared properties are likely to exist since ingrowing mechanosensory nerves recognize the Merkel cells, and not other epidermal cells, as their targets.

    Notes: Summary The growth-promoting effect of caerulein on antral gastric mucosa was explored using Wistar rats. Implanted osmotic minipumps were used to administer submaximal doses of either caerulein or saline to normal rats for up to 4 days. In one group, reflux of bile and pancreatic juice into the stomach was avoided by previous surgical diversion of the distal common bile duct to the jejunum. DNA synthetic and mitotic activity in the antrum epithelium were estimated by 3H-thymidine pulse labelling and autoradiography during the administration of the peptide. The rate of cell migration was determined in animals killed 1, 2 and 3 days after the 3H-thymidine pulse. Administration of caerulein to normal rats provoked significant increases in both labelling and mitotic indices, and a significant acceleration of the upward cell migration in the glandular tubes. In the animals with distal diversion of bile and pancreatic secretions both labelling and mitotic indices were also increased over control values under the effect of the peptide. These data indicate that administration of caerulein stimulates cell proliferation in the antral gastric mucosa. This effect cannot be explained through increased reflux of pancreaticobiliary secretions in the stomach.

    Notes: Summary The volume and surface area of lipid inclusions often present in the cytoplasm of rat Sertoli cells was measured directly from semi-thin sections of perfusion-fixed testicular tissues using an image analyser linked to a light microscope. Sertoli cell nuclei were used as a reference for comparing any variations in the measured parameters of lipid inclusions during the rat spermatogenic cycle. Volume density of Sertoli cell lipid inclusions was assessed by morphometric analysis of Sertoli cells photographically reconstructed from electron micrographs. Maximum lipid content in Sertoli cells occurred during stages IX–XIV of the spermatogenic cycle, then declined at stages I–III and remained low from stages IV–VIII. The persistence and increase in number of many large Sertoli cell lipid inclusions beyond the stage where spermatid residual bodies are phagocytosed within the Sertoli cells (stage IX) suggests that the synthesis and lipolysis of Sertoli cell lipid inclusions represents an intrinsic functional cycle of the Sertoli cells. Stage-dependent variations in the lipid content of rat Sertoli cells offers morphological evidence that the metabolic duties of the Sertoli cells are synchronised with the spermatogenic cycle to provide local coordination of the proliferation and maturation of the germ cells.

    Notes: Summary Thy-1+ cells, producing Thy-1+ material, have been demonstrated by the indirect immunoperoxidase technique in the theca of growing ovarian follicles of the rat. OX-2 antigen, known as the minor glycoprotein of rat thymocytes, was detected in granulosa cells of non-growing follicles. Ia+ cells of dendritic type and/or activated macrophages were identified in the granulosa of advanced degenerating follicles, and remnants of the zona pellucida exhibited immunoglobulins. In some ovaries immunoglobulins were also bound to the zona pellucida of oocytes of early degenerating antral follicles. Medium-sized antral follicles with degenerating granulosa were occasionally invaded by cells carrying antigens of cytotoxic T lymphocytes or other T lymphocyte subsets, while degenerating large antral follicles were sometimes invaded by cells exhibiting antigen of cells with natural killer function (but not antigens of T lymphocytes). Granulosa cells of some degenerating antral follicles exhibited class-I antigens derived from the major histocompatibility complex. We suggest that cell-mediated control mechanisms of antigen expression and metabolism of tissue cells during their differentiation and degeneration should be considered in addition to the well-documented hormonal dependence of some tissues.

    Notes: Summary Antigenic markers characteristic of astrocytes and their differentiative states (i.e., glial fibrillary acidic protein (GFAP), vimentin, and M1 and C1 antigens) were investigated in the pineal gland of mouse and rat using double immunolabeling techniques. In both species the socalled interstitial cells as characterized by TEM were shown to be astrocytes, since they expressed vimentin, but neither fibronectin (a marker for fibroblasts and endothelial cells) nor the neuron-specific L1 antigen or tetanus toxin receptors. Subpopulations of vimentin-positive pineal astrocytes were also GFAP- and C1- antigen-positive. M1- antigenpositive cells were not detected. It is concluded that a considerable proportion of interstitial cells in the pineal gland of rat and mouse are immature astrocytes which, in contrast to other parts of the central nervous system, persist into adulthood.

    Notes: Summary Thyrotropic hormone (TSH) or cAMP accelerate the formation of follicular cavities in the explanted thyroid gland of the 15-day-old rat fetus. Cytochalasin B or vinblastine and nocodazole or colchicine, which disorganize microfilamental and microtubular structures respectively, inhibit or completely block in vitro-induced folliculogenesis. Exposure of the thyroid tissue to lumicolchicine, a structural isomer of colchicine deprived of antimicrotubular activity, does not inhibit the activation of folliculogenesis induced by TSH. These results are strong evidence for the supposition that microfilaments and microtubules are involved in the TSH-stimulated mechanisms resulting in thyroid folliculogenesis. Folliculogenesis requires the integrity of both microfilaments and microtubules.

    Notes: Summary Appearance of immunoreactive corticotropin-releasing factor (CRF)-containing neurons was studied in developing hypothalamus of the rat by use of antisera against rat- and ovine CRF. These neurons were first recognized in the lateral and paraventricular nuclei on days 15.5 and 16.5 of gestation, respectively, when antiserum against rat CRF was employed. Antiserum against ovine CRF revealed the cells two days later exclusively in the latter nucleus. In both nuclei, the neurons increased in number with development. The neurons in the paraventricular nucleus appeared to project their immunoreactive processes to the median eminence via the periventricular and lateral pathways. In the median eminence, the immunoreaction with antiserum to rat CRF was first recognized in its anterior portion in the form of dots on day 16.5 of gestation but as beaded fibers in the external layer on day 17.5; these structures increased in amount with development in rostro-caudal direction. Although antiserum to ovine CRF was less potent in immunostainability than antiserum to rat CRF, it also revealed the beaded fibers in the median eminence on day 17.5 of gestation. Since evidence is available that the paraventricular nucleus is involved in corticotropin release, it is concluded that, in rats, the hypothalamic regulatory mechanism controlling the release of corticotropin initially appears on days 16.5–17.5 of gestation.

    Notes: Summary The distribution of caldesmon (a calmodulin-binding, F-actin-interacting protein) (Sobue et al. 1982) and of actin was studied in the rat's small intestine by means of light-microscopic immunocytochemistry. Positive immunostaining for caldesmon was seen in smooth muscle cells of the intestinal wall, and of blood vessels, and in the apical portion of the absorptive epithelial cells. The immunoreactivity in goblet cells was difficult to recognize. The positive reaction to immunostaining for actin showed almost the same pattern as that for caldesmon. These results suggest that this calmodulin-binding protein may play an important role in the control of actin-myosin interaction in smooth muscle cells and in non-muscle cells.

    Notes: Summary Ferritin-concanavalin A (Fer-Con A) was used to label the apical plasma membrane of the lactating cell to determine whether membrane internalization takes place. Rat glands were infused in vivo via the teat with 0.2 mg of Fer-Con A in 0.2 ml tris buffer (pH 7.0) containing 0.1% trypan blue, the latter acting as a marker of the infusate. Tissues were obtained from separate animals 5, 10 and 60 min postinfusion. Fer-Con A was seen in alveolar lumina bound to the outer surfaces of apical plasma membrane, microvilli and milk fat globules. It was observed within lactating cells on the inner membrane surfaces of endocytotic vesicles, Golgi cisternae, and secretory vesicles containing casein micelles, and in multivesicular bodies and lysosomes. Internalization of the ferritin-lectin conjugate into casein-containing secretory vesicles was detectable in the 5-min postinfusion tissue. Lysosomes were the only structures in control tissue that contained particles bearing some resemblance to Fer-Con A. The data provide evidence that apical plasma membrane is internalized and distributed to a number of intracellular compartments.

    Notes: Summary Systemic administration of monosodium-1-gluta-mate by single injections of 4 mg/g body weight in infant rats (2–10 days of age) results in acute swelling of cytoplasm and nuclear pyknosis of neurons in the stratum zonale and stratum griseum superficiale of the superior colliculus. Multiple daily doses of 4 mg/g body weight monosodium-1-glutamate result in an almost complete loss of neurons in these two superficial layers. The deeper layers appear not to be affected. No pathological effects were observed in the lateral geniculate body or pretectal complex. Light-and electron-microscopic studies reveal that the optic nerves are remarkably shrunken and many myelinated as well as unmyelinated axons are lost. Injection of 3Hproline into the vitreous body of one eye results in limited transport to the suprachiasmatic nucleus, lateral geniculate body and to lateral portions of the superior colliculus. The small percentage of intact axons in the optic nerve, as well as the limited proline transport from the eye, suggest that administration of monosodium-1-glutamate leaves intact some optic fibers, a portion of which belongs to the retinohypothalamic tract.

    Notes: Summary In an attempt to identify a cellular Ca2+-pool, from which calcium is released when secretagogues are applied, tissue fragments of the rat exocrine pancreas were incubated and fixed with glutaraldehyde in the presence of calcium. By means of this procedure electron-dense deposits were found on plasma membranes. X-ray microanalysis showed that these deposits contain calcium. Stimulation of tissue fragments with the use of the secretagogues carbachol or cholecystokinin reduced the number of deposits by about 80%. When the antagonist atropine was applied after carbachol stimulation, deposits reappeared on cell membranes, which then disappeared again after a second stimulation with cholecystokinin. In the presence of procaine, carbachol was inhibited and only slightly reduced the Ca2+-deposits on the plasma membranes. These results suggest that a calcium pool, from which calcium is released to induce enzyme secretion on stimulation, is located in the cell membrane

    Notes: Summary The development of immunoreactive (ir) somatostatin-containing nerve terminals in the rat median eminence (ME) has been examined electron-microscopically. Nerve fibers containing ir particles scattered throughout the axoplasm are first seen in the external layer of the ME on day 18.5 of gestation, and, on day 21.5 appear to terminate on the basement membrane of the perivascular space of the portal vessels. After birth, the fiber terminals contain several membrane-limited granules, which are labeled with ir PAP particles. Ultrathin, Epon-embedded sections of ME, treated by the protein A gold-labeling method for somatostatin, demonstrate positively labeled granules in the nerve fibers in the postnatal ME, but in the prenatal tissue, no specific gold-labeling is found. These findings show that, in the external layer of the ME, somatostatin storing occurs in the granules in the axonal terminals after birth.

    Notes: Summary Vibratome sections obtained from perfusion-fixed rat brains were stained by means of silver impregnation and physical development according to Gailyas (1970). Small pieces of the cerebral cortex were postfixed with buffered osmium tetroxide solution and processed for electron microscopy to examine the localization of the silver deposit at the cellular level. The cell surfaces of pericytes and smooth muscle cells were completely outlined by silver grains. Endothelial cells and perivascular astrocytes, however, showed an asymmetric distribution of the silver deposit, i.e., the deposit was restricted to the abluminal endothelial surface and to the astrocytic membrane adjacent to the vessel wall, respectively. The method allowed a clear-cut distinction between perikarya of endothelial cells and pericytes as well as glial cells in perivascular position, even at the light-microscopic level.

    Notes: Summary The nucleoli of dictyate-stage growing oocytes in rat ovaries were examined both with routine electron microscopy and electron microscopy after silver nitrate and ammoniacal silver nitrate (Ag-AS) staining. The nucleoli of the unilaminar follicular oocytes consist of twisted strands of dense fibrillar components, aggregates of granular components, and small fibrillar centers. After Ag-AS staining, silver grains are numerous on the dense fibrillar strands, fewer on the fibrillar centers, and very sporadic on the granular aggregates. The same stainability of three nucleolar components with the Ag-AS method was also confirmed in the nucleoli segregated by actinomycin D. During the transition of growing oocytes from bilaminar to plurilaminar follicle stage, the nucleolar dense fibrillar strands gradually conglomerate and are transformed into large and compact spherules. The stainability of dense fibrillar components with the Ag-AS method was lost along with this nucleolar transformation. These results may provide some new clues on the functional significance of AgAS-positive proteins in the nucleoli.

    Notes: Summary Electron microscopy was used to study synaptogenesis in prepyriform cortex of fetal rat pups during early stages of synapse formation. Of special interest is the frequent occurrence of unapposed, developing synaptic specializations in axon and growth cone profiles. The location and morphology of the unapposed specializations suggests that thay are presynaptic in nature. These presumably immature presynaptic specializations are found in the lateral olfactory tract and subjacent cortex. Intermediate forms between uncontacted presynaptic specializations and definitive synapses suggest a synaptogenic sequence in which initial development of an immature presynaptic specialization begins without apposition of a postsynaptic element at that location. This implies that initiation of presynaptic development is not dependent upon postsynaptic contact and also raises the question of whether synaptic contacts could be established via presynaptic induction of postsynaptic formation.

    Notes: Summary The distribution of caldesmon (a calmodulin-binding, F-actin interacting protein; Sobue et al. 1982) and actin was studied in the rat thyroid gland by means of light-microscopic immunocytochemistry, and the fine-structural distribution of actin filaments was examined by use of heavy meromyosin (HMM). Caldesmon and actin were demonstrated in the apical cytoplasm of almost all the follicle epithelial cells in normal as well as TSH-treated animals. Immunoreactivities for both caldesmon and actin showed almost the same pattern in localization. The smooth muscle cells of the blood vessels were also positive for caldesmon and actin. By electron microscopy, numerous actin filaments decorated by HMM and running perpendicularly or randomly to the apical surface were recognized in the apical cytoplasm of the follicle epithelial cell. These results suggest that caldesmon and actin, in conjugation with calmodulin, play a role in the regulation of cellular activity such as exocytosis and endocytosis in the apical portion of the follicle epithelial cell.

    Notes: Summary The effect of short-time treatment with the ionophore monensin, administered intraluminally at concentrations of 5 and 10 μM, was studied on the Golgi apparatus of absorptive cells in the small intestine of the rat. At 2–3 min after treatment most of the Golgi stacks exhibited dilated cisternae. At 4–5 min stacked cisternae were absent; they were replaced by groups of smooth-surfaced vacuoles. Dilatation and vacuolization occurred in the entire stacks without preferential effect on any particular Golgi subcompartment. Monensin did not influence the cytochemical Golgi reaction of thiamine pyrophosphatase and acid phosphatase. The characteristic staining pattern of these two enzymes in all Golgi cisternae of absorptive cells in the proximal small intestine, and the reactivity restricted to trans cisternae in distal segments of the small intestine, were unchanged after treatment with monensin. In the distal small intestine, the cytochemical pattern allowed the monensin-induced vacuoles to be attributed to the former cisor trans-Golgi face. Further, the cytochemical results demonstrate that vacuolization is not restricted to the stacked cisternae, but includes the trans-most cisterna. The latter, usually located at some distance from the Golgi stacks, has been defined as belonging to the GERL system in several types of cells. The clear response to monensin, an agent that selectively affects the Golgi apparatus, indicates common properties between trans-most and stacked Golgi cisternae.

    Notes: Summary Estrogen is accumulated from the blood by nerve cells in the ventromedial nucleus of the hypothalamus and can facilitate female reproductive behavior by acting on this region of the brain. This cell group was examined in ovariectomized female rats, given estrogen or control treatment, by use of light and electron microscopy. A significantly greater portion of the nerve cells in the estrogen-treated animals had protuberances on their nucleolar surfaces, apparent under the light microscope. The fine structure of such protuberances included dense, aggregated material, which is shown to contain DNA by the sodium tungstate staining technique. Because increased numbers of such protuberances were found in nuclei of cells of the experimental group where previous studies demonstrated a significant increase in ultrastructural signs of biosynthetic activity, they may be associated with increased RNA synthesis. Thus, they could indicate, ultrastructurally, increased synthetic rates for RNA in nerve cells through which estrogen promotes reproductive behavior.

    Notes: Summary In untreated adult male albino rats nuclear volume and the percentage of binucleate cells were determined in the first layer of hepatocytes adjacent to hepatic venous branches of varying diameters (〈40 μm, 40 μm–80 μm, 80 μm–120μm, 120 μm–160 μm, 〉160 μm), and in the third and fourth layer of hepatocytes in the remainder of the perivenous parenchyma. In the first layer of hepatocytes adjacent to the vascular structures means of nuclear volume are significantly lower and percentage of binucleate cells significantly higher than in the cells of the remainder of the perivenous parenchyma. Within each area measured distribution curves of nuclear volume classes were homogeneous but showed heterogeneity in comparison with each other. The morphometric data presented in this study strongly support the opinion of the heterogeneity of liver cells in the perivenous zone, as previously postulated on the basis of histochemical investigations.

    Notes: Summary Subcellular structures of hepatocytes in periportal and perivenous zones were examined during 24 h. The volume, surface and numerical profile densities of cytoplasmic organelles were analysed morphometrically. Most subcellular structures in periportal and perivenous hepatocytes were subject to strong circadian variations. In hepatocytes from both zones, the volume densities of smooth endoplasmic reticulum (sER), mitochondria, lysosomes, peroxisomes, polysomes and lipid droplets demonstrated peak values at 16.00 h, 20.00 h or 00.00 h; trough values were at 04.00 h, 08.00 h, or 12.00 h, except for peroxisomes (16.00 h). However, the volume densities of glycogen granules and rough endoplasmic reticulum (rER) in periportal and perivenous hepatocytes exhibited maximal values at 04.00 h, 08.00 h or 12.00 h and minimal values at 20.00 h. The surface densities of sER, mitochondria, lysosomes and peroxisomes, and the numerical profile densities of mitochondria, lysosomes and peroxisomes in periportal and perivenous hepatocytes showed similar trends. These events suggest that membranes of the rER show a partial correlation with the sER, mitochondrial and lysosomal membranes during the 24-h span. This may involve the interaction between ribosomes and rER. Almost all cytoplasmic organelles examined displayed significant differences between periportal and perivenous hepatocytes, morphometrically and in fine structure, indicating that the morphofunctional variability of hepatocytes differs depending on the location in the liver acinus.

    Notes: Summary To assess the distribution of the thyroid hormone triiodothyronine (T3) within intact living cells, freshly prepared dispersed rat hepatocytes were incubated with [125I]-T3 for periods of 5 min and 30 min. Lightand electron-microscopic (EM) radioautography was carried out to determine the distribution of grains over the isolated cells. Both procedures showed the grains distributed almost entirely over the cytoplasmic matrix rather than the nucleus. Grain counts under the EM were compared with expectation based on established quantitative methods. Only the mitochondria showed obvious and statistically significant grain counts, whereas the nucleus failed to accumulate grains in excess of expectations by chance alone based on area. The findings support the existence of mitochondrial binding of T3, presumably a prerequisite for its action in direct stimulation of the mitochondria.

    Notes: Summary Intercellular junctions were often found between macrophages in sinuses of regional lymph nodes of the rat after injection of large doses of cholesterol, cortisone acetate, and estrone at the footpad. They were identified by subplasmalemmal densities, 20–50 nm in width, beneath the plasma membranes of apposed macrophages. No distinct filamentous structures were visible in those dense regions. Electron-dense amorphous materials are lined up at the center of the intercellular space in the junctional regions. Some macrophages form clusters with intercellular junctions. No significant difference in the effect of cholesterol, cortisone acetate, and estrone on the number of intercellular junctions betwene macrophages was found.

    Notes: Summary The neuropil located ventral to the SON was investigated by the use of immunoperoxidase staining for neurophysins, oxytocin and vasopressin, and electron miroscopy. The study was performed in six groups of rats: 1) control; 2) infusion of isotonic saline into the CSF; 3) infusion of hypertonic saline into the CSF; 4) drinking hypertonic saline for 4 days; 5) same as group 4 but injection of colchicine into the CSF on second day of dehydration; 6) salt loading for 3 months. In the control rats the ventral neuropil contained a few immunoreactive processes, the general morphology of which was completely different from that of the neurosecretory axons emerging from the SON at its dorsal aspect. In rats of groups 3 to 6 the ventral processes (VP) became loaded with neurosecretory granules, whereas the perikarya and axons were depleted. Based on their general morphology and reactivity pattern it is suggested that the VP are dendrites. Most of these “dendrites” were embedded in a glial cushion formed by the processes of a particular type of marginal glia. Some of these “dendrites” enveloped an arteriole penetrating the optic tract. All VP were rich in synaptic contacts. The possibility that the VP of neurosecretory cells may be functionally related to the subarachnoid CSF and the arteriolar blood flow is discussed.

    Notes: Summary When rats were injected with 6-hydroxydopamine the catecholaminergic nerve terminals in their intermediate lobes exhibited distinct signs of degeneration. Morphometric examination of the Golgi apparatus in cells of the intermediate lobe of these rats showed significant enlargement of Golgi cisternae. The release of adrenocorticotropin, β-endorphin/lipotropin and α-melanotropin from intermediate-lobe cells in vitro was measured by radioimmunoassay. The high basal peptide release was inhibited by dopamine and stimulated by methyl-isobutyl-xanthine. In contrast, γ-aminobutyric acid, serotonin, histamine and noradrenaline, or corticotropin-releasing hormone, rat hypothalamic extract and vasopressin had no or only very weak effects. These observations indicate that the synthetic apparatus of intermediate-lobe cells is constantly depressed by dopaminergic nerves. We were not able to stimulate peptide release from intermediate-lobe cells by use of the abovementioned endogenous agents.

    Notes: Summary After maternal fasting for 72 h the pancreatic β cells of 18-day-old foetal rats show a conspicuous enrichment in secretory material, with an increase of pancreatic insulin concentration and a marked development of the rough endoplasmic reticulum and the Golgi apparatus. The morphometric analysis shows that the intracytoplasmic migration of the secretory granules is inhibited, principally inside the cell web. Consequently the number of secretory granules fused with plasma membrane decreases and this is associated with a decreased foetal plasma insulin. The difference in the ultrastructural aspect of the β cells of foetuses from fasting mothers and of foetuses from fed mothers is less conspicuous at 19 days of gestation and progressively disappears at 20 and 21 days. The modifications in ultrastructural aspect and in functional state are discussed.

    Notes: Summary After 2 h intraocular injections of 19 and 190 nmoles ibotenic acid in the rat retina produced an intensive vacuolization of the inner plexiform layer and cellular alterations, in the inner nuclear layer and ganglion cell layer. These alterations consisted of either cytoplasmic swelling accompanied by clumping of the nuclear chromatin or darkening of the cytoplasm along with nuclear condensation. A week later the retinas were thinner than the controls due to the disappearance of the affected cells. Pre-treatment with diazepam prevents the morphological alterations induced by 19 nmoles ibotenic acid; mainly the swelling, which was completely prevented, while the darkening was reduced drastically, although some vacuolization of the inner plexiform layer is still present.

    Notes: Summary The effect of an LH pulse on the rate at which 3H-thymidine is incorporated into cultured ovaries of metestrous rats was studied. In comparison to ovaries cultured with tonic LH, an LH pulse (1) “rescued” follicles from atresia, (2) induced thecal cell proliferation, and (3) increased the rate at which granulosa cells enter mitosis. It is concluded that LH pulses increase follicular growth by first triggering thecal cell proliferation and then inducing mitotic divisions within the granulosa cells of both atretic and non-atretic follicles.

    Notes: Summary The localization of autologous antiperoxidase immunoglobulin G (IgG) was studied in the choroid plexus of Lewis rats immunized against horseradish peroxidase (HRP). This experiment was performed to study the permeability of the choroid plexus to intravascular IgG. It was shown that autologous IgG was present in the extravascular spaces. The transendothelial transfer appeared to occur mainly via the fenestrations and some interendothelial junctions. No transfer of IgG at the level of epithelial cells toward the cerebrospinal fluid was demonstrated. Interstitial spaces in contact with the connective-tissue cells of the choroid stroma were strongly labeled. The significance of these spaces remains hypothetical and raises the question of the fate of IgG from the interstitial space.

    Notes: Summary The appearance and intracellular localisation of glial fibrillary acidic protein (GFAP) in pituicytes in neural lobe cultures of newborn rats aged 7 to 30 days were investigated by use of the indirect immunofluorescence method. GFAP-immunoreactive cells were observed mostly in the outgrowth zone. GFAP was localised in the perikaryal cytoplasm as well as in pituicyte processes. GFAP-positive pituicytes showed considerable morphological polymorphism. The presence of GFAP — astrocytic marker — in pituicytes in vitro and the evident morphological similarity to cultured astrocytes suggest the astroglial character of these cells.

    Notes: Summary Cyclic guanosine monophosphate-phosphodiesterase (cGMP-PDEase) activity was studied histo- and cytochemically in the retinal rods of the rat with the use of a newly developed technique. Intense activity of cGMP-PDEase was evenly distributed over the outer segments of the rods. Reaction product was observed on the plasmalemma and on the disk membranes of the outer segments. A weak reaction product occurred also on the plasmalemma of the inner segments; however, no precipitate was found in the perinuclear and synaptic portions of the rod cells. The enzyme activity was strongly inhibited by 2 mM theophilline and by 2 mM 3-isobutyl-1-methylxanthine (IBMX). To confirm the specificity of this new cGMP-PDEase method, the localization of 5′nucleotidase (5′GMPase) was also studied. In contrast to the activity of cGMP-PDEase, the activity of 5′GMPase was distributed on the plasma membrane of the photoreceptor cells extending over a wide range from the synaptic endings in the outer plexiform layer to the tip of the outer segments.

    Notes: Summary The rat retina was studied by immunohistochemistry with antibody to S-100 protein during the first three postnatal weeks. Immunoreactive astrocytes are first detected subjacent to the inner limiting membrane close to the optic disc. They gradually increase in number and spread toward the ora serrata along the inner surface of the retina as the development proceeds. S-100-immunostained Müller cells are first identified on the 12th postnatal day although their immunoreactivity is much weaker than that of astrocytes at the same stage. This differential intensity of the immunoreactivity of the two cell types facilitates observation of the entire shape of the astrocyte. This characteristic reveals that cellular investments of blood vessels in the inner retina are formed by astrocytic processes whereas those in the outer plexiform layer are derived from processes of Müller cells. The cellular investment becomes complete by the 18th postnatal day.

    Notes: Summary Macrophages of the adrenal cortex were studied in normal male and female, ovariectomized and estradiol-injected rats. In normal male rats few macrophages with numerous granules were observed in the zona fasciculatazona reticularis border, and in the zona reticularis. Granules, identified as lysosomes, were limited by a single membrane with a heterogeneous matrix; they exhibited acid phosphatase- and aminotriazole-resistant peroxidatic activities. A larger number of macrophages had identical distributions in normal female rats. In ovariectomized and estradiol-injected rats the number and distribution of adrenal macrophages were similar to those in normal females; however, in spayed animals the number of these cells in the zona reticularis was higher than in the other experimental groups. Lysosomes in macrophages of treated animals were more numerous and their contents more complex than in normal male animals. These results indicate that the adrenal macrophage system is stimulated in experimental conditions involving high levels of circulating estrogens.

These cells, corresponding to the small, intensely fluorescent cells, showed an extensive rough endoplasmic reticulum, a large Golgi apparatus and a very large number of granular vesicles. In addition to the granular vesicles, 70–160 nm in diameter, in which the dense core filled most of the vesicle, most cells of the hydrocortisone-injected rats contained also larger granular vesicles, up to 350 nm in diameter, in which the dense core was eccentrically located. A minority of the cells contained only granular vesicles 70–100 nm in diameter, which was the only type seen in the saline-treated control rats. Thirty days after discontinuation of the hydrocortisone treatment, most of the cells with large granular vesicles had disappeared, and only two profiles of such cells were seen on day 40. The other small cells contained only granular vesicles 70–160 nm in diameter, and these cells could not be distinguished from the small granule-containing cells of 40-day-old control rats treated early postnatally with saline. Hydrocortisone treatment, first on days 3–9 and subsequently on days 40–46, caused reappearance of the small granule-containing cells with large granular vesicles up to 350 nm in diameter, the dense core of which was eccentrically located. Hydrocortisone treatment on days 40–46 only was not followed by appearance of such cells in rats treated with saline on days 3–9.

    Notes: Summary The intercellular clefts of the brain and the leptomeninges, and the perivascular spaces were studied with reference to the results obtained in a previous study (Krisch et al. 1983). The spatial relationships of these compartments were analyzed at the electron-microscopic level. Horse-radish peroxidase (HRP) was injected into the brain or into the contralateral ventricle. The pattern of distribution of HRP depends on the boundary situation in the individual compartments. The inner and outer pial layers accompany the vessels intruding into the brain. In the Virchow-Robin space the pial funnel obliterates within a short distance. The inner arachnoid layer is continuous with the outer arachnoid layer when it covers the vessels traversing the meningeal space. The perivascular compartment is not in communication with the arachnoid space; moreover, the pial funnel within the Virchow-Robin space is sealed off against the arachnoid space. Thus, blood vessels traversing the meningeal spaces and subsequently penetrating the brain surface are exposed to the common intercellular compartment represented by the intercellular clefts of the brain and the leptomeninges; this compartment does not communicate with the other compartments. The cerebrospinal fluid located in this intercellular compartment is preferentially drained into the upper cervical lymph nodes.

    Notes: Summary Rat liver fixed by perfusion with low glutaraldehyde concentrations was incubated in diaminobenzidine-containing medium to stain for peroxidase. Endogenous peroxidatic activity was found not only in Kupffer cells but also in the endothelial cells lining the sinusoids and central veins. The reaction product was localized in the nuclear envelope and endoplasmic reticulum. The peroxidatic activity in endothelial cells showed a concentration-dependent sensitivity to glutaraldehyde: in liver samples fixed with 0.25% glutaraldehyde, approx. 23% of the sinusoidal endothelial cells and 65% of central vein endothelium were peroxidase-positive; with 0.5% glutaraldehyde, only approx. 8% of the sinusoidal endothelial cells contained detectable amounts of the reaction product; with 1.5% glutaraldehyde all endothelial cells were consistently peroxidase-negative. No peroxidatic activity could be found in liver endothelial cells following isolation by centrifugal elutriation. Endothelial cell peroxidase may possibly be involved in defense responses of liver and/or, as a part of prostaglandin synthase system, in prostanoid production.

    Notes: Abstract The stress for 12 sulfur-containing synthetized volatiles was evaluated in male Wistar rats and compared to that for fox-dropping extract concentrate. Stress behavior was analyzed by quantifying various stress responses in a standard open field and measuring the increase in plasma corticosterone concentration. Nine compounds induced stress—a dihydrothiazole, two cyclic polysulfides, five mercaptoketones, and a mercaptan. For the mercaptoketones, the following structure-activity relationships were observed. Size can vary considerably; the mercapto group can be either alpha or beta and either secondary or tertiary. The keto group is not essential, since a structurally related mercaptan remains active. The mercapto group is essential for activity in mercaptoketones, since conversion to a methyl sulfide resulted in a neutral response. This type of odorant could function as an allomone and may have potential in rat control as an area repellent.

    Notes: Small-angle neutron scattering, SANS, stands forth as one of the most important of the new tools for evaluating polymer chain conformation and morphology. This paper reviews the SANS literature through 1982, with a few early 1983 references added. The theory of SANS is outlined and compared to light scattering. SANS values of polymer molecular weights and radii of gyration obtained in the bulk state were found to be in agreement with values obtained from dilute solutions by light scattering. In each case, deuterated fractions of polymer were inserted into the hydrogeneous matrix, or vice versa, to provide contrast. Several new research areas are then discussed, including unidirectional stretching of elastomers, stress-relaxation, polymer-polymer miscibility, crystallization from the melt compared with crystallization from dilute solutions, nonclassical aggregation during bulk polymerization of thermoset systems, morphology of polymer blends, block copolymers and ionomers, the core-shell structure of latexes and polymer blocks, and grafts as surfactants in emulsions and latexes. Much new and sometimes unexpected information is being provided by the SANS research now in progress.

    Notes: Effects of pressure and temperature on time-dependent changes in physical/mechanical properties of graphite/epoxy composites were investigated. Samples were cut from the eight-ply-thick laminates of commercially used composites, post-cured, and then quenched to environments of various temperature and pressure. Time-dependent changes in their properties were analyzed by thermal and thermomechanical (dynamic mechanical) measurements. An increase in the glass-transition temperature was found to occur as a function of time. The rate of this process was enhanced by an increase in temperature and/or a decrease in pressure. An explanation was offered in terms of types and mechanisms of molecular events that occur in the glassy state. Time dependent decrease in free volume (and enthalpy) takes place but is not the sole mechanism responsible for the observed increase in Tg. After a certain period of time (which depends on T and P of the environment), additional crosslinking appears to take place.

    Notes: Water-ground Phlogopite micas were classified into narrow particle-size distributions containing flakes with well-defined diameters and thicknesses in order to evaluate the influence of particle size and flake aspect ratio on the mechanical properties of mica-filled polypropylenes, For the purposes of comparison, most of the injection-molded specimens contained 40 percent (by weight) mica. As expected, the flexural and tensile modulus values increased in proportion to the aspect ratio over the range from 30 to 60 to a maximum of 8 GPa. The measured tensile strengths of the mica-filled polypropylenes increased substantially as the flake diameter became smaller, but did not correlate with the flake aspect ratio. The attainable properties were frequently dependent upon the method of mixing, and considerable care was necessary to ensure proper dispersion and adequate coupling. Intensive mixing, as in a Gelimat Mixer, may cause in situ delamination and particle-size reduction of the mica filler particles, leading to a marked increase in tensile strength of the resulting composite. The mica-filled compounds could be reprocessed many times without significant loss of properties, particularly compounds having mica particles less than 40 μm in diameter. The fracture energies (notched Izod) and the heat-distortion temperatures were not appreciably influenced by the size or aspect ratios of the mica within this range. Increased fracture toughness could be achieved by reducing the mica concentration or employing a polypropylene copolymer. Guidelines are presented to indicate the preferred characteristics of mica fillers and the influence of mixing conditions on performance.

    Notes: Three blown-film-grade low-density-polyethylene (LDPE) resins were studied using different rheological techniques. Eccentric rotating disks (ERD), cone-plate viscometry, capillary rheometry, annular die extrusion, and non-isothermal stretching of a filament were used. The viscoelasticity of the melts was found to play a dominant role in the observed behavior. Extrudate appearance in annular flow, melt strength, and extensibility are affected by melt elasticity. A correlation was found between the maximum draw ratio of a filament stretched under non-isothermal conditions and minimum film thickness.

    Notes: This paper investigates three aspects of linear-low-density polyethylene (LLDPE) rheological properties: shear viscosity variations with shear rate and temperature, tensile behavior determined with an extensiometer, and extrusion defects. The differences in shear viscosity variation with shear rate and temperature between LLDPE and LDPE (low-density polyethylene) are shown. These differences, attributed to wider molecular weight distribution and to long chain branching (LCB) in LDPE, involve different extrusion behaviors. The lack of LCB in LLDPE can be demonstrated by comparison of the measured Newtonian viscosity with the value of the same parameter calculated from molecular weight distribution and composition law of Newtonian viscosities. The lack of LCB leads to good melt extensibility, which is shown by tensile properties of polyethylene melts determined with a non-isothermal extensiometer. The melt fracture phenomenon is studied because it promotes surface defects on bubbles in film application. Extrudate distortions are examined at the laboratory extruder outlet. This test shows differences between LLDPE and LDPE, but also between some LLDPE samples.

    Notes: Inverse gas chromatography (IGC) has been used to investigate thermodynamic miscibility of a molten poly(ethylene oxide) (PEO)/poly(methyl methacrylate) (PMMA) blend. Toluene, benzene, and chloroform have been employed as probes in pure and mixed stationary phases of these polymers. Experimental measurements have been taken over a narrow range of temperatures because of the high PMMA glass transition temperature as well as the degradation of the PEO. The interaction parameter χ23 determined at 150°C is slightly negative and dependent on the interacting probe, as has been also noted in previous chromatographic studies on polymer-polymer miscibility. The last section is devoted to a model calculation, using Flory's equation of state theory. Different χ23-concentration curves have been simulated, with the interaction energy parameter X23 as an adjustable parameter.

    Notes: A quantitative model which described the microscopic and macroscopic refractive index properties of uniaxially oriented crystalline polymers has been extended in relation to molecular bond polarizabilities in this work. Application of this extended modeling methodology in analyzing measured refractive index data for a series of unoriented and oriented samples of linear polyethylene provided Δco = 0.0585 and Δao = 0.194 as the most probable crystalline and noncrystalline intrinsic birefringences for samples exhibiting spherulitic morphology. With these intrinsic birefringences, noncrystalline orientation functions were determined from the optical measurements coupled to the model and the results compared to values obtained from infrared measurements. This comparison of noncrystalline orientation functions, as well as from low density polyethylene reported by other investigators, provided experimental justification for our modeling methodology to examine the possibility of changing intrinsic birefringences for polyethylene as a function of orientation and morphology. The results of this examination demonstrated that values for Δco = 0.0585 and Δao = 0.12 should be used for both low and high density polyethylene samples oriented above the spherulitic to fibrillar transition region.

    Notes: “Living” anionic polymerizations carried out in a continuous stirred-tank reactor (CSTR) and operated under forced-feed oscillations are theoretically studied to investigate the effects on time average values of average chain lengths, polydispersity, and reagent conversions. The instantaneous mean residence time is maintained constant. The period of the oscillations is varied between 60 and 0.05 mean residence times, and various amplitudes are considered. Some advantages are observed with the periodic forcing as compared to the steady operation; polymers with average polydispersities above 10 may be produced at low frequencies, with polydispersities below the steady-state value of 2 at intermediate frequencies, and with polydispersities of 2 but with increased monomer conversion at fast cycling.

    Notes: The high-temperature electrical conductivity and thermal decomposition characteristics of Sylgard® 184 with and without hollow microspheres of glass, silica, and ceramic have been determined to 600 to 700°C in air and nitrogen environments. The materials are silicone-based dielectrics and are used as electronic encapsulants. Results show that a peak in the conductivity temperature dependence at ∼300°C results principally from volatilization of [Si(CH3)2O]n with some evolution of water, that oxygen accelerates decomposition, and that the microspheres may help form a network of interconnected conductive pathways in the residual material. There is a good correlation between thermal stability and temperature-dependent electrical properties.

    Notes: An abbreviated review of the current state of knowledge of polymer interfacial phenomena is given. Classical thermodynamics treats the interfacial zone (the interphase) as a “black box” and yields rigorous relationships among interfacial quantities. A recent reformulation of interphase thermodynamics, which eliminates the use of a Gibbs dividing surface, is shown to be an invaluable tool for investigating interfacial properties. Microscopic theories, such as the gradient theory, yield more details about what is in the black box, but the information is only approximate. The gradient theory has been used to: (1) relate the surface tension of a polymer liquid to its isothermal compressibility, (2) develop a quantitative theory of polymer liquid surface tension, and (3) determine the interfacial tension between two immiscible polymer liquids. The gradient theory will be shown to be in harmony with the microscopic theory of Helfand and co-workers although the latter treats polymer interfaces from a completely different point of view.

    Notes: The technique of Thermo Stimulated Creep (TSC) has been applied to the study of anelastic properties of polyethylene, polypropylene, their copolymers and blends. In the temperature range -200 to 100°C, complex TSC peaks were observed in all samples, namely around 0°C, about the same temperature as for the homopolypolymer polypropylene. By applying “fractional stresses”, with a convenient choice or the loading program, these peaks have been experimentally resolved. Two components can be distinguished: 1. The “low temperature” component is characterized by mechanical retardation times following a compensation law. It has been attributed to microbrownian motions of polypropylene sequences liberated at the glass transition of the “true” amorphous regions. 2. The “high temperature” component which is influenced by thermal treatment has been assigned to microbrownian motions of polypropylene sequences liberated at the glass transition of the “constrained” amorphous regions. In block polymers, an additional TSC peak is observed around -50°C: it has been associated with the glass transition of ethylene-propylene-rubber (EPR) interphase. The coupling of this interphase with polyethylene and polypropylene phases is insured by diffusion of some ethylene and propylene sequences in-EPR. At about -140°C, a TSC peak associated with the low temperature component of the glass transition of polyethylene can be distinguished in all the materials studied.

    Notes: The formation of polyethylene terephthalate (PET) has been modeled to have reactions with monofunctional compounds, redistribution, and cyclization reactions in addition to the usual polycondensation step. In the final stages, the overall polymerization is mass-transfer controlled and solution of the reactor performance equations have been determined through the orthogonal collocation technique. This technique is found to be considerably more efficient for PET reactors compared to the finite difference method; the use of ten collocation points gives results which are close to the exact solution.

The purpose of this paper is to summarize the authors' theoretical work on the structures, conformational energies, intermolecular interactions, and electronic properties of PBO and PBT chains, including the protonated forms known to exist in strong acids.     Notes: The evolution of poly(methyl methacrylate) (PMMA) during physical aging at 90°C is followed by torsional microcreep tests. On the aged specimen a longitudinal stress is applied which induces a strain of 5 percent after 30 days of creep. The torsional microcreep tests are performed during the longitudinal creep in order to compare the structure evolution of PMMA caused by straining with its evolution measured during aging. The microcreep, for the first 800s, follows a reversible logarithmic law. In this stage the mobile defects achieve their activated form which is perfectly reversible when unloaded. The physical aging reduces this logarithmic part of microcreep. This is due to the decrease of either the number or the volume of the mobile defects. Beyond a critical elongation ∊ = 1 percent, the longitudinal straining has just the opposite influence, i.e., the logarithmic part of microcreep increases. This critical elongation ∊ = 1 percent corresponds to the beginning of the steady state longitudinal creep. The transient that precedes this steady state has no detectable influence on the structure of the specimen.

    Notes: A method based on network theory is developed for characterizing molecular orientation in amorphous polymers. The proposed procedure gives not only the orientation distribution function for the chain segments in the polymer network (and hence the average orientation) but also a quantitative measure of how this orientation is distributed among the various types of entanglement junctions. The orientation of chain segments can be characterized by two parameters, one which gives the average orientation and another which reflects how much orientation is concentrated in long time entanglement junctions. The new method of characterizing orientation is used to interpret tensile strength data for both brittle and ductile failures.

    Notes: A study of the crystalline orientation, light transmission, and surface roughness of polyethylene tubular film prepared in our laboratories is presented. The present studies were primarily carried out on low-density (LDPE) and linear-low-density (LLDPE) polyethylene films. The optical properties of a few films of high-density polyethylene (HDPE) prepared for a previous study of morphology were characterized for comparison to the LDPE and LLDPE films. Wide angle X-ray diffraction and birefringence were used to characterize orientation. Both the LDPE and LLDPE films exhibited crystalline texture in which the b-axes tended to be perpendicular to the film surface and the a-axes had some tendency to align with the machine direction. The c-axes tended to be concentrated in the plane of the film with nearly equal biaxial orientation with respect to the machine and transverse directions. Little variation in the crystalline orientation was found with changes of process conditions in the range studied. Birefringence results indicate that the amorphous regions developed an orientation in which the chains tend to be normal to the film surface. The majority of light scattering from these films and a series of HDPE films was from the surface and not from the film interior. The transmission coefficient for the surface contribution was found to be a monotonic decreasing function of the standard deviation of the surface height obtained from surface profiles measured by profilometer. The surface asperites were largest for the HDPE and smallest for the LDPE samples. The intensity of both the surface and interior contributions to the scattering increased with increasing frostline height, i.e., a slower cooling rate. As draw-down ratio and blow-up ratio increase the scattering contribution from the film interior decreases but the contribution from the surface increases somewhat. These effects are discussed in terms of the changes in crystalline morphology and surface roughness produced by flow defects generated during extrusion.

    Notes: Cylindrical specimens of poly(methyl methacrylate) (PMMA) were quenched from above the glass transition and subsequently tested in torsion. Torque and normal-force relaxation responses were recorded simultaneously. The experiments were performed at 40, 60, and 80°C, at increasing aging times and at deformations ranging from γ = 0.0027 to γ = 0.074. It was found at 40 and 60°C, that, contrary to the classical picture of aging, the small-deformation torque-relaxation curves could not be superposed by any combination of vertical and horizontal shifts. On the other hand, at 80°C, and in the nonlinear deformation range even at lower temperatures, the relaxation curves are superimposable simply by horizontal shifts, but as γ increases they involve decreasing values of the double-logarithmic shift rate. At the same deformation level, however, the shift rate for the normal force is significantly higher than is that for the torque over most of the γ range investigated.

    Notes: Wood pulp fibers possess strength and modulus properties which compare favorably with glass fibers when the differences in fiber densities are considered. Softwood pulp fibers with fiber aspect ratios near 100 are readily dispersed into high-density polyethylene or isotactic polypropylene with the aid of carboxyic dispersing agents to form mixtures containing 50 weight-percent wood pulp which can be readily injection molded. The mechanical properties of the molded specimens were similar for all types of pulp including Kraft (bleached and unbleached), mechanical and chemical-mechanical pulps, waste pulps, and reclaim newspapers. Comparisons of the stiffness/weight efficiencies revealed that pulp composites equal or exceed the stiffness of most traditional materials of construction including steel, aluminum, glass-fiber composites, and talefilled polyolefins, while retaining a major material cost advantage. The measured strength values of the pulp composites were less than the theoretically predicted values due to the presence of voids created by the formation of volatiles during processing. Mechanical pulps which were available in dry form were preferred because of lower cost and ease of handling. Wood fibers are non-abrasive so that relatively large concentrations may be incorporated into polyolefins without causing serious machine wear during mixing and fabrication.

    Notes: It has been observed in dynamic mechanical measures that the range of temperatures in which relaxation properties are affected by the diminishing of the free volume has, as a lower limit the temperature at which the maximum of G″ or tgδ, corresponding to the β relaxation, occurs. In dielectric measures on polymers in which the β relaxation is not predominant over the α relaxation, this temperature range is scarcely affected by changes in frequency, whereas the peak shifts according to its activation energy. This fact leads to the conclusion that the decrease in free volume due to a slow cooling from a temperature above Tg mainly affects mobility of the main chains under Tg, without disturbing the groups that give rise to the secondary relaxations.

    Notes: Standard test specimens of mortar were cured under water for 15 days to achieve satisfactory strength. They were dried without allowing dehydration reactions to occur, evacuated to 40 millimeters (mm) Hg, and impregnated with a styreneacrylonitrile comonomer, which gives the highest strength after polymerization. Positive pressure of 0.6 N/mm2 was applied and thermal catalytic polymerization was carried out. Optimum initiator concentration was found to be 2 percent. Strength (compressive and tensile) increases with polymerization temperature up to 120°C, above which rapid deterioration occurs due to monomer loss, production of short chains with high polymerization rates, and the possibility of polymer degradation at high temperature. Optimum polymerization period of' 10 hours was selected. Inexpensive methods for field polymerization were tried at 95°C. Hot water yields a strength more than double that obtained with a, flow of hot air and about 80 percent of that obtained radiolytically. While hot air accelerates evaporation, hot water forms a hydraulic head above the capillaries and decreases the monomer losses tremendously.

    Notes: For free radical oligomerization of styrene, a scheme for calculating the molecular weight distribution and conversion in a continuous-stirred tank reactor (CSTR) train is developed, which also allows the calculation of molecular weight distribution (MWD) for batch reaction. Calculations show that under conventional or near dead-end condition: (1) increasing initial initiator concentration, reaction time and reaction temperature, and decreasing initial monomer concentration cause P̄n and P̄w to decrease and MWD to narrow; (2) increasing initial initiator concentration, reaction time and reaction temperature, and increasing monomer concentration cause monomer conversion to increase; (3) a single CSTR gives a lower rate of oligomer production, but a narrower MWD than does a batch reactor.

    Notes: The performance of liquid rubbers as low-temperature plastcizers for bitumen was evaluated by the torsional braid technique developed by Gillham. The low-temperature dynamic mechanical properties of bitumen-liquid rubber mixtures were measured from ambient to -100°C and compared with the corresponding fracture behavior using a modified Fraass test. Microscopic observation of the mixture revealed that the liquid rubber was partially soluble in the bitumen at 23°C and at concentrations greater than 10 percent tended to form colloidal dispersions of rubber particles having mean diameters of approximately 10 μm. The loss tangent maxima shifted to lower temperatures and became less pronounced as the concentration of liquid polybutadiene increased. The colloidal rubber dispersion further contributed to the ductility and reduced the brittle temperature of the bitumen at -30°C at 20 percent liquid rubber concentration. The reduction in the brittle temperature of the bitumen is related to the glass-transition temperature of the added rubber, those rubbers having, the lowest glass-transition temperatures being most efficient. Such modifications are necessary to reduce the tendency of asphaltic paving materials to crack in cold climates.

    Notes: The absorption of water vapor by propellants based on hydroxy-terminated polybutadiene and an isocyanate from atmospheres of increasing relative humidities, and the swelling index of the propellant which had absorbed various proportions of water, were measured in an attempt to establish the extent of hydrolytic damage. The method was found incapable of assessing such damage, but it was shown that there was no measurable absorption of vapor below 92 percent relative humidity (RH). A variety of linear and network vinylic components are studied including polymethacrylonitrile homopolymer and copolymer, and polyacenaphthalene homopolymer and copolymer. The polyurethane and polyvinyl components phase separate during the in situ polymerization resulting in a three-phase morphology. Detailed small-angle X-ray scattering analysis provides measurements of phase mixing, domain spacings, and diffuse phase boundary thicknesses. A comparison of these parameters for parent polymers and composites reveals only slight differences; indicating that the microdomain structures of the phase-separated polyurethane networks are preserved in the composite. Appreciable molecular interpenetration of components is not observed. Dynamic mechanical loss measurements for several of the composites demonstrate the presence of loss processes occurring over extremely broad ranges of temperature. The strong mechanical interaction between components observed suggest that the urethane and vinyl phases may be bicontinuous in these materials.

    Notes: A series of stretch-blow molded poly(ethylene terephthalate) bottles were investigated using measurements of birefringences (in plane and out of plane) and wide angle x-ray diffraction (WAXS) pole figures. Both bottles prepared in our laboratories and commercial bottles were investigated. Birefringences Δn13 and Δn23 of inside and outside surfaces of bottles were determined as a function of position along the length of the bottle (1-bottle axis, 2-hoop direction, 3-thickness direction). The Δn13, Δn23 for the inside surface of the bottles were found to be greater than those for the outside surface indicating heterogeneity through the thickness of the bottles. The WAXS studies indicate that the benzene rings in the polymer align parallel to the bottle surface. The chain axis in crystalline regions exhibit orientations close to equal biaxial orientation. Both Δn13 and Δn23 increased with inflation pressure.

    Notes: Small angle neutron scattering experiments were made on an ionomer synthesized from perdeuterostyrene and an ion pair comonomer. A Debye plot based on data from the sample with lower ion concentration gave a correlation length of 5.3A. Clustering is not significant in this sample. With a higher concentration of ionic groups a peak in the scattering curve is interpreted as due to clusters about 20A in diameter. These studies along with X-ray scattering and electron microscopy also indicate the presence of larger ion rich domains.

    Notes: An experimental study has been carried out on rubber I transfer molding. It reveals that the filling is frequently limited more by the resistance of flow across the transfer pot than by resistance of flow through the sprue holes into the cavities. A mathematical model has been derived, which predicts semi-quantitatively the molding behavior observed. The mode1 predicts that fill time is proportional to the ratio of compound viscosity divided by molding pressure raised to about the fourth power. For the common cases where most of the fill time is from the resistance to the transverse flow on the top of the sprue plate, the fill time is proportional to about the fifth power of the ratio of transverse distance divided by the charge thickness. Experimental results showed that preheating and mastication of the compound reduced transfer time substantially. The charge pattern did not seem to have a major influence on transfer time.

    Notes: Drying of hygroscopic plastics in preparation for molding or extrusion is characterized by very low moisture levels in the polymers and by very low humidity in the drying air. In these circumstances, diffusivity is essentially independent of moisture concentration but surface concentration will vary with time in the general case. An exponential surface concentration/time function is derived from fundamentals and applied to the solution of the diffusion equations for slabs and spheres. Under ideal drying conditions, the solutions reduce to the elementary case in which cs = cf for θ 〉 0. For non-ideal drying, the exponential factor b̃ in the surface concentration function appears in the solutions of the diffusion equations. Experiments on slabs were used to verify the analysis since ideal or non-ideal drying conditions could readily be set up with molded slab samples. Experimental drying curves are in agreement with theory. To complete the analysis for engineering applications, equations for counter flow heat transfer in the dryer hopper are derived and combined with the diffusion solutions to generate dryer performance data.

    Notes: The morphology of blends of high-density polyethylene (PE) and isotactic polypropylene (PP) was studied by mesans of optical and scanning-electron microscopy. In the range of 10 to 90 percent by weight PE, these blends are two-phase systems, the components of which crystallize separately into discrete phases. The presence of PE has a definite and pronounced effect on the crystalline structure of the PP, whose spherulitic structure becomes increasingly irregular and coarse with increasing PE content. The light transmission of these blends during melting and crystallization was also studied in an attempt to characterize them.

    Notes: An experimental study was made of the effects of die geometry and extrusion velocity on parison swell for three high-density-polyethylene blowmolding resins. Four annular dies were used: a straight, a diverging, and two converging dies. Diameter and thickness swells were measured as functions of time under isothermal conditions and in the absence of drawdown. This was accomplished by extruding into an oil having the same density and temperature as the extrudate. It was observed that 60 to 80 percent of the swell occurs in the first few seconds and that equilibrium swell is attained only after 5 to 8 minutes have elapsed. The diameter and thickness swells appear to be independent phenomena, as the relationship between them depends strongly on die design. The ranking of the resins in terms of the magnitude of the swell was found to be the same for all die geometries and extrusion rates used.

    Notes: Tensile, tensile impact, compact tension, and fatigue tests were carried out on ABS (acrylonitrile-butadiene-styrene), HIPS (high impact polystyrene), and toughened PMMA (poly(methyl methacrylate)). Dumbbell specimens machined from test pieces were then subjected to sinusoidal tension-compression cycling at low stress amplitudes. The HIPS specimens produced asymmetrical hysteresis loops characteristic of multiple crazing, whereas PMMA gave more symmetrical, regularly shaped, loops. The behavior of the ABS polymers varied with loading history. It was concluded that the hysteresis test provides a useful additional technique for studying deformation mechanisms, especially under conditions that do not readily permit volumetric measurements.

    Notes: This paper addresses the apparent controversy surrounding the role of the solid bed mechanics in the Maddock melting mechanism. It is demonstrated that the inability of the melting models based on the freely deformable solid bed concept to predict accurately the pressure gradients in the melting zone is not exclusively due to the highly simplified isothermal Newtonian treatment of the melt pool as presumed previously. This study has shown that when using a non isothermal non-Newtonian flow model for the melt pool, the freely deformable solid bed concept still results in unrealistically low pressure gradients while it may give good predictions of the melting rates. To the contrary, when a rigid solid bed is assumed, the pressure predictions tend to represent the experimental data more closely, whereas the theoretical melting rates seems to become less realistic. In view of the fact that both the freely deformable and the rigid solid bed concepts show such inconsistencies, it has been concluded that the mechanics governing the solids and melt transport in the melting zone require some additional examination, most notably, the influence of the constitutive behavior of the solid bed and of the cross-channel melt circulation around the solid bed, and possibly of the melting kinetics for semicrystalline polymers.

    Notes: Dynamic and static mechanical properties of a soluble aromatic polyimide (PI 2080) were investigated. It, unlike poly(pyromellitimide) (PI), was essentially amorphous in X-ray diffraction and did not exhibit any heterogeneous two-phase structure having ordered phases after annealing at temperatures around its glass transition temperature. The mechanical properties including thermal-distortion temperature and elastic recovery were not much affected by the annealing. The difference in molecular aggregation and mechanical properties between PI 2080 and PI may be due to their different chemical structures. The flexible carbonyl group and the statistical distribution of copolymeric repeating units in the main chain of PI 2080 seem to prevent the formation of molecular order.

    Notes: A method of determining fiber orientation in composites using wide-angle X-ray diffraction (WAXS) is described. Oriented crystalline fibers are suspended in an amorphous polymer matrix. The WAXS reflects characteristics of the fiber are used to determine the mean orientation and orientation distribution of the crystallographic axes representing the polymer chain relative to preferred axes located in the test specimen. The chain direction crystallographic axis is taken as representing the fiber axis, and the orientation of this axis to represent the orientation of the fibers. Experimental studies were carried out using Kevlar (poly(p-phenylene terephthalamide)) fibers suspended at a 20 volume percent loading in a polymethyl methacrylate matrix. The Kevlar fibers had Hermans orientation factors of 0.92. Specific attention is given to how through circular dies. We have examined both extrudates and the material frozen-in when the composite in the reservoir and die is cooled to room temperature. Fiber orientation factors, corresponding to Hermans orientation factors, 0.3 to 0.38 were obtained for the extrudates. Orientation factors for fibers within the die is about 0.45. Specially prepared completely oriented samples had orientation factors of 0.93, which closely corresponds to the orientation of the fiber.

    Notes: A general-purpose finite element program has been used to simulate the flow of Newtonian, power-law, and viscoelastic fluids in the entry and exit regions of a slit die. It was found that shear-thinning increases the entrance correction while it decreases the exit correction. Shear-thinning reduces the size of the small corner vortex that forms in the entry flow of a Newtonian fluid. The swelling ratio had a value of 1.196 for Newtonian fluids and decreased as the value of the power-law index decreased. Viscoelastic calculations were performed using the Criminale-Ericksen-Filbey (CEF) constitutive equation. Convergence of the iterative scheme was unattainable for Deborah numbers above 1.0. The results showed a decrease of the entrance correction and an increase of the exit correction with elasticity. Extrudate swell first decreased slightly and further increased with the Deborah number.

    Notes: As crazes grow in areal extent they also increase in width. The areal growth involves craze tip advance which has been shown to occur by the Taylor meniscus instability. Craze widening, at least for air crazes, occurs by drawing more fibrillar material from the craze-bulk polymer interfaces at essentially constant extension ratio. Simple arguments will be given to predict the scale of the fibrillation in terms of the stress S at the craze tip and interfaces and an effective polymer surface energy (Γ) where:\documentclass{article}\pagestyle{empty}\begin{document}$$ {\Gamma = \gamma + {\raise0.5ex\hbox{$\scriptstyle 1$}\kern-0.1em/\kern-0.15em\lower0.25ex\hbox{$\scriptstyle 4$}}d \cdot v_E U_b} $$\end{document} which assumes that all entangled chain crossing the surface are broken [γ represents the van-der-Waals (intermolecular) surface energy, d is the entanglement mesh size, vE is the entanglement density, and Ub is the energy required to break a single backbone bond]. These arguments also give the rate of fibrillation as a function of S, a nominal plastic resistance σy and Γ and can explain the fact that the stress for crazing increases relative to that for shear deformation as the entanglement density of the polymer is increased. The geometrically necessary entanglement loss (either by scission as assumed above or by disentanglement- at temperatures just below Tg) that accompanies fibril formation has important consequences for fibril stability. The probability p that a given entangled chain is lost can be computed from simple geometrical considerations knowing the fibril diameter D, its extension ratio λ and the mesh size d; p increases rapidly as Dλ½ becomes comparable to or less than d. These concepts can be tested in blends of high molecular weight polymer with chains of the same polymer that are too short to entangle.

    Notes: Enthalpy relaxations in glassy poly(vinyl chloride) following varied pre-aging treatments and under varied aging conditions have been compared through observations of sub-Tg endothermal DSC (differential scanning calorimetry) aging peaks. The extent of enthalpy relaxation for a fixed time and temperature of aging is progressively enhanced by the imposition and release of increasing mechanical stress before aging. The same effect is produced by sorption and desorption of increasing amounts of CO2 or CH3Cl vapor before aging. In contrast, the continued application of mechanical stress, or the presence of vapor, during the aging period suppresses enthalpy relaxation. The extent of suppression increases with increasing vapor pressure and solubility or increasing stress. These effects are interpreted as consequences of an increase in the enthalpy of the polymer under mechanical or sorptive stress and an enthalpy relaxation following the release of this stress. In addition to these effects on the DSC endotherm, a pronounced exotherm between the aging peak and Tg is observed for samples which have undergone shear yielding or orientation either before or during aging. This exotherm may be the result of release of stored strain energy during the DSC scan.

    Notes: Effects of structural relaxation and the glass-transition process are clearly manifested in the electrical behavior of glassy-chalcogenide films. Analysis of the transport and trapping of photoinjected carriers and of the thermal generation of free carriers in the bulk reveal that in chalcogenides each of these processes is mediated by a specific manifold of localized electronic states residing in the mobility (forbidden) gap. It is demonstrated that in the vicinity of Tg the population of these states varies systematically with the structural state of the glassy film always tending toward a temperature-dependent quasi-equilibrium. The present-observations thus clearly establish the structural origin of key electronic-gap states and simultaneously reveal the mechanism by which relaxation impacts electrical behavior.

    Notes: The kinetics of isothermal crystallization of polymer blends in which the matrix is a crystallizable polymer is considered. It is shown that depending on the difference in interfacial energies the inclusions are rejected or engulfed by the growing spherulite. Other factors influencing rejection, engulfing, and/or deformation of dispersed particles of the second polymer are the viscosity of the melt, the spherulite growth rate, and the size of dispersed particles. If the difference in interfacial energies is positive, then rejection or engulfing requires additional work to be done by the crystallization front. This dissipation of energy decreases the spherulite growth rate. It is estimated that the rejection of the second component is the most important phenomenon in the crystallization of blends. The spherulite growth rate of isotactic polypropylene in blends with low-density polyethylene and several elastomers was studied as a function of crystallization temperature and concentration. The comparison of growth rate data with morphological changes occuring during crystallization of blends studied shows very good agreement with the theoretical predictions based on energetics considerations.

    Notes: Critical strains causing environmental stress cracking of injection-molded poly(acrylonitrile-butadiene-styrene) (ABS) and poly(styrene-acrylonitrile) (SAN) plaques were determined upon exposure to methanol. Measurements were obtained for samples strained either parallel or perpendicular to the melt flow direction and for samples located at various distances from the mold gate. Critical strains were significantly higher in the direction parallel to the melt flow compared to the transverse direction. The degree of anisotropy increased with increasing rubber content. For ABS containing 46 percent rubber, the critical strain at one point was determined to be 2.99 percent in the direction of melt flow, but only 0.47 percent in the orthogonal direction. For this material, critical strains determined parallel to the melt flow decreased with distance from the gate; whereas, critical strains for SAN and ABS containing 30 percent rubber remained essentially constant. Orientation of the plaques was assessed using shrinkage determinations and a thermal conductivity technique. Though a straightforward correlation of orientation with critical strain is observed for ABS, a similar relationship is not observed for SAN. These results suggest that although stress cracking occurs in the glassy matrix of ABS, it is the dispersed rubbery phase which controls the magnitude of strain required to initiate cracking.

    Notes: A basic study of the kinematics, dynamics, and heat transfer occuring during tubular film extrusion of polyethylene is outlined. Three rheologically characterized polyethylenes, a low-density polyethylene (LDPE), a linear-low-density polyethylene (L-LDPE), and a high-density polyethylene (HDPE) were used in this study. The kinematics and stability of the tubular film process were investigated over a wide range of blow-up ratios, drawdown ratios, and frost-line heights. Local deformation rates along the bubble have been determined. Regions of stability and instability are described. Tensions and inflation pressures have been measured and expressed in terms of locol elongational viscosities. Temperature profiles along the bubble were determined and interpreted in terms of local heat transfer coefficients. Positions of crystallization and temperature profiles have been noted and used to estimate rates of crystallization. The characteristics of the LDPE, LLDPE, and HDPE are contrasted.
