    Notes: Summary The kinetics of seeded crystal growth of calcium apatites were studied in dilute supersaturated solutions at various levels of fluoride concentrations. Initial precipitation rates were enhanced by fluoride concentrations higher than 0.05 ppm. The analytical results are consistent with the precipitation of fluoridated hydroxyapatites, Ca5Fx-(OH)1−x(PO4)3, FHA. The degree of fluoridation, X, appears to be determined by the activity of HF in solution, which varies for the various initial fluoride levels but remains fairly constant during precipitation. Thus the composition of the precipitating phase was the same for a given solution whether 25 or 10 mg of hydroxyapatite was added as seeds. All the experimental results are consistent with the BCF theory, which relates the mean linear rate of growth, RL, to the supersaturation, DS, by the expression RL=C1T(DS-1)1n(DS)tanh(C2/T 1n DS), in which DS is the supersaturation defined by mean molar activities with respect to the precipitating FHA, T the absolute temperature, and C1 and C2 are constants calculated from the experimental results. Consequently, the crystal growth appears to take place in surface kinks and to be controlled by surface diffusion. Since crystal growth in most biological systems takes place at fluoride concentrations within the experimental range used, it seems probable that it occurs along the model advanced in the present investigation.

    Notes: Summary To test the importance of 24-hydroxylation of vitamin D3 on bone mineralization, rat pups born to vitamin D-deficient females were given either 25-hydroxyvitamin D3 or 24,24-difluoro-25-hydroxyvitamin D3 for 16 days beginning at the time of weaning. Following such treatment analysis of blood samples revealed no detectable 24R,25-(OH)2D3 and 1,25-(OH)2D3 in the rats given the difluoro compound while revealing the expected 24,24-difluoro-25-hydroxyvitamin D3 and 24,24-difluoro-1,25-dihydroxyvitamin D3. The rats given 25-hydroxyvitamin D3 had the expected levels of 25-hydroxyvitamin D3, 24,25-dihydroxyvitamin D3, and 1,25-dihydroxyvitamin D3. Following sacrifice at day 17, postweaning bone mineralization and modeling were studied in long bones using histological methods. Bones taken from vitamin D-deficient rats at the beginning and end of the experimental period had lesions typical of rickets. These included wide growth plates, excessive amounts of osteoid, and metaphyseal fibrosis. Following treatment with either 25-hydroxyvitamin D3 or 24,24-difluoro-25-hydroxyvitamin D3, bone mineralization returned to normal. Growth plate widths and the amount of osteoid on bone surfaces were both substantially reduced and to a similar degree in both treatment groups. Normal cartilage core formation and trabecularization of the metaphyseal primary spongiosa were also restored to a similar degree in both groups. In effect, no difference was observed in any bone parameter studied between the 25-hydroxyvitamin D3- and the 24,24-difluoro-25-hydroxyvitamin D3-treated animals. These results provide strong evidence that 24-hydroxylation of the vitamin D molecule plays little or no role in the modeling and mineralization of bone.

    Notes: Summary Using the technique of short interval sequential tetracycline labeling, it was documented that the apposition of mineralized bone matrix in adult male Sprague-Dawley rats was inhibited by hydrocortisone. The inhibition occurred as early as six days after the onset of the treatment and was dose dependent over a dose range of 0.62 to 20 mg per kg body weight per day. Vitamin D2 supplements by injection protected bone from this hydrocortisone action. 64 I. U. of vitamin D2 injected daily was able to prevent the inhibition of bone apposition by 20 mg per kg body weight per day of hydrocortisone. The results imply that vitamin D or its metabolites may compete with hydrocortisone in some cellular mechanisms and support the usefulness of vitamin D supplements in the treatment and the prevention of steroid-induced osteoporosis.

    Notes: Summary The therapeutic effects of vitamin D analogs, 1,24(R)-dihydroxycholecalciferol [1,24(R)(OH)2D3], 1,24(S)-dihydroxycholecalciferol [1,24(S)(OH)2D3], and 1,25-dihydroxycholecalciferol [1,25(OH)2D3] on immobilization osteoporosis were studied in rats. The right hind limb was immobilized through application of a plaster cast following the section of the sciatic nerve. The left hind limb was intact. Vitamin D analogs were orally administered for 6 weeks at dose levels of 0.02 and 0.10µg/kg/day, respectively. The mean lengths of the immobilized femurs were not significantly different from those of the intact femurs in all the experimental groups. In the immobilized femur of animals treated with 1,24(R)(OH)2D3, 0.10µg/kg, dry and ash weights were heavier and calcium and phosphorus contents greater than those in the nontreated group. Furthermore, the amount of calcified bone mass and the cortical thickness of the femurs of the immobilized limb in 1,24(R)(OH)2D3-treated animals were greater than those in the nontreated animals. Treatment with 1,25(OH)2D3 at 0.10µg/kg caused an increase of the bone mass in both immobilized and intact femurs when compared with those of the control group. It was concluded that the administration of 1,24(R)(OH)2D3 diminished the effect of immobilization in the development of osteoporosis without any side effects.

Dans cet ordre d'idée, les arêtes de truites et de carpes ont été analysés pour établir si la composition du minéral osseux traduit la faible concentration en bicarbonate sérique. Le contenu en CO2 d'arêtes de poissons n'est pas significativement différent de celui des os normaux de mammifères. Des crîstaux d'apatite synthétique, préparés dans des conditions identiques de T, μ et (HCO 3 − ), ne contiennent que 1/7ème à 1/8ème du CO2 d'arêtes de poissons. Ces résultats indiquent que la composition des liquides dans l'os ne réfléchit pas simplement la composition du sérum circulant, ainsi qu'on l'admet généralement.
    Abstract: Zusammenfassung Es ist in der Literatur berichtet worden, daß Fische, um genügend Sauerstoff zu erhalten, einen Überschuß von Kohlensäure einatmen müssen und folglich einen niedrigen Gehalt an Bicarbonaten im Kreislauf aufweisen. Knochen von Forellen und Karpfen wurden analysiert, um festzustellen, ob ihre Mineralsalzzusammensetzungen die niedrigen Serum-bicarbonate-konzentrationen wiederspiegeln. Die Resultate zeigten keinen wesentlichen Unterschied zwischen dem Carbonatgehalt der Knochen von Fischen und Säugetieren. Synthetische Apatitkristalle, die unter ähnlichen Bedingungen von T, μ und HCO3 bereitet wurden, wiesen nur 1/7–1/8 des Carbonatgehaltes der Fischknochen auf. Diese Ergebnisse deuten zweifellos darauf hin, daß zwischen der Zusammensetzung der Gewebeflüssigkeit der Knochen und des Serums kein einfacher Zusammenhang besteht.
    Notes: Abstract It has been reported in the literature that fish, to get sufficient oxygen, must overventilate with respect to CO2 and therefore exhibit a low circulating level of bicarbonate. Accordingly, the bones of trout and carp were analyzed to learn if the composition of their bone mineral reflects the low serum level of bicarbonate. It was found that the CO2 content of fish bone is not significantly different from that of normal mammalian bone. Synthetic apatite crystals, made under comparable conditions of T, μ, and (HCO 3 − ) were found to contain only 1/7th to 1/8th the CO2 of fish bone. These data strongly suggest that the composition of the fluids in bone does not reflect, in a simple way, the composition of the circulating serum as is generally assumed.

La précipitation spontanée ou catalysée sont approximativement aussi sensibles. L'inhibition par des combinaisons de Mg avec des ions de Sr ou de F est additive. Ces résultats montrent que ces inhibiteurs ont acucune action spécifique sur la catalysation par la collagène de l'os de mouton qui soit différente de celle sur la nucléation de l'apatite en soi.
    Abstract: Zusammenfassung Mg, Sr und F Ionen hemmten die Fällung von Apatit wenn sie zu einem Calciumphosphatpuffer einzeln oder zusammen hinzugefügt wurden. Die Fällung von Apatit durch Apatitkeime war weniger empfindlich hinsichtlich der Hemmung als die spontane Nucleation und als die Fällung von Apatit, welche von Schafsknochenkollagen katalysiert werden kann. Die spontane und katalysierte Fällung sind ungefähr gleich empfindlich. Die Hemmung von Mg in Kombination mit Sr oder F Ionen war additiv. Diese Resultate weisen darauf hin, daß keine spezifischen Effekte bei der Katalyse von Knochenkollagen vorliegen, welche von den Effekten bei der Nucleation von Apatit an sich verschieden sind.
    Notes: Abstract Mg, Sr and F ions acted as inhibitors of apatite formation when added to a calcification buffer separately or in combinations. The precipitation of apatite under the influence of apatite seeds was less susceptible to inhibition than were spontaneous nucleation and apatite formation catalyzed by sheep bone collagen, while the latter two were about equally susceptible. Combinations of Mg with Sr or F ions showed additive effects. These results suggest that no specific effects of these inhibitors occurred on the catalytic action of the bone collagen different from their effects on the nucleation of apatite as such.

    Notes: Summary A simplified method is described for the measurement of calcium absorption in the dog. This method uses45Ca and takes less than 8 h to perform. Calcium absorption was measured in normal and uremic dogs on different intakes of calcium and phosphorus and compared to the results obtained in the same animal maintained on the same diet by another method using47Ca and a double dilution procedure. The correlation coefficient between the two methods was 0.99 (P〈0.001). The advantages of the simplified method are discussed.

    Notes: Summary Thaw-mount autoradiographic studies after injection of3H-1,25-D3 were conducted on 18-and 20-day-old rat fetuses. In maxillary bones, ribs, and tibia, nuclear concentration of radioactivity was found in osteoprogenitor cells and osteoblasts. Osteocytes and chondrocytes in epiphyseal plates were either unlabeled or weakly labeled. In competition experiments, nuclear concentration of radioactivity was blocked by the injection of a high dose of nonradioactive 1,25-D3 prior to the administration of the labeled hormone, but not by a similar dose of nonradioactive 25-D3. The results are interpreted as indicating that osteoprogenitor cells and osteoblasts are target cells for the direct action of 1,25-D3 on fetal bone.

    Notes: Summary The role of vitamin D in early skeletal development was studied by measuring serum calcium and phosphorus, osseous tissue quantity and mineralization, and endochondral bone elongation in rat fetuses and pups from vitamin D-replete and vitamin D-deficient mothers. At the 20th day of pregnancy there was a slight, yet significant, increase in the amount of osteoid on trabecular bone surfaces in fetuses from vitamin D-deficient mothers. The fetal bones otherwise appeared normal in spite of severe skeletal changes in the vitamin D-deficient mothers. After parturition, the importance of vitamin D in skeletal development becomes progressively more obvious. Serum calcium levels were slightly, yet significantly, lower in vitamin D-deficient than in vitamin D-replete pups and these levels continued to fall in the vitamin D-deficient pups through lactation and after weaning. At 3 days postpartum, there was a small, yet significant, increase in the amount of osteoid on bone surfaces of the vitamin D-deficient pups. The relative amounts of osteoid in the vitamin D-deficient pups continued to increase through lactation and after weaning when compared with vitamin D-replete pups. By the 14th day of lactation and at later periods, there were significant reductions in metaphyseal mineralized tissues in the vitamin D-deficient pups when compared with the vitamin D-replete pups. At weaning and after weaning, there were substantial increases in growth plate thickness and decreases in longitudinal bone growth in the vitamin D-deficient pups when compared with the vitamin D-replete pups. The results from this study indicate that vitamin D does not appear to play a major role in fetal skeletal development. However, after birth, vitamin D becomes progressively more important with age for normal bone development, mineralization, and endochondral growth.

    Notes: Summary Recent evidence of the occurrence of brushite in newly formed bone mineral prompted a study of the solubility properties of brushite:apatite mixtures under physiological conditions and the influence on them of pH, lactate, pyruvate, and, particularly, noncollagenous bone proteins (NCBPs). Brushite alone was surprisingly stable in solution at pH 7.4, 37°C. In the presence of increasing amounts of apatite, hydrolysis of brushite to an insoluble phase occurred. A decrease of 0.1 pH unit or the addition of 1.5 mM pyruvate or 10 mM lactate increased the ion activity product (Ca2+×HPO 4 2− ) 3 or more times. However, within the loose envelope of bone such conditions so different from those in the circulation might be only local or temporary. NCBPs, on the other hand, stabilized brushite in solution alone as well as in the presence of apatite for days. They probably act by adsorbing strongly to the crystal surface and preventing nucleation by apatite. This brushite-apatite-bone protein system exhibits solubility characteristics that can resolve the old problems presented by the participation of the skeleton in extracellular calcium homeostasis on the one hand, and by the apparent insolubility of the apatite mineral of bone on the other.

    Notes: Summary The present study examined calcium phosphate precipitation in aqueous suspensions of multilamellar liposomes as a possiblein vitro model for matrix vesicle mineralization. Liposomes were prepared by dispersing CHCl3-evaporated thin films of 7:2:1 and 7:1:1 molar mixtures of phosphatidylcholine, dicetyl phosphate, and cholesterol in aqueous solutions containing 0, 25, or 50 mM PO4 and 0 or 0.8 mM Mg. After removal of unencapsulated PO4 by gel filtration, the liposomes were suspended in 1.33 mM Ca/0.8 mM Mg solutions and made permeable to these cations by the addition of the ionophore X-537A. All experiments were carried out at pH 7.4, 22°C, and 240 mOsm. In the absence of entrapped PO4, Ca2+ taken up by the liposomes was largely bound to inner membrane surfaces. With PO4 present, Ca2+ uptake increased as much as sixfold with maximum accumulations well above values sufficient for solid formation. Precipitated solids appeared to be located predominantly in the aqueous intermembranous spaces of the liposomes. Amorphous calcium phosphate (ACP) precipitated initially in the presence of entrapped Mg2+, then subsequently converted to apatite intermixed with some octacalcium phosphate. The stability of the liposomal ACP was somewhat greater than that observed in bulk solutions under comparable conditions of pH, temperature, and electrolyte makeup. In time, the mineral deposits caused entrapped PO4 to leak from the liposomes. These findings suggest that the precipitation within liposomes is similar to that which occurs in macro-volume synthetic systems but that the precipitated solid eventually impairs the integrity of the surrounding intermembranous space. These observations may offer a partial explanation for the ability of matrix vesiclesin vivo to act as nidi for extracellular mineralization.

    Notes: Summary To evaluate the relationships of a 25-hydroxy vitamin D absorption test to intestinal fat absorption and to the absorption of radioactively labeled vitamin D and 25-hydroxy vitamin D, an investigation was undertaken in 19 patients with gastrointestinal disorders. A correlation was noted between the results of the 25-hydroxy vitamin D absorption test and baseline 25-hydroxy vitamin D levels. No correlation was found between results of the 25-hydroxy vitamin D absorption test and quantitative fecal fat excretion. Peak values of the 25-hydroxy vitamin D absorption test correlated with net absorption of14C vitamin D. No correlation existed between the values obtained in the 25-hydroxy vitamin D absorption test and3H-25-OH D absorption. These studies indicate that the 25-hydroxy vitamin D absorption test probably does not serve as an effective screening test for intestinal fat malabsorption. The results of the test probably most accurately reflect the body stores of vitamin D at the time of testing, but there appears to be little advantage to performing a 25-hydroxy vitamin D absorption test in lieu of a single determination of the serum level of 25-hydroxy vitamin D as a method of evaluating vitamin D nutritional status.

    Notes: Summary The ESR spectra of oxovanadium(IV) ions, (VO2+), adsorbed on hydroxyapatite(OHAp), fluorhydroxyapatite(FHAp), Mg-containing tricalcium phosphate(Mg-TCP), octacalcium phosphate (OCP), dicalcium phosphate dihydrate (DCPD), and amorphous calcium phosphate(ACP) were measured at room temperature. The ESR parameters of VO2+ adsorbed on these compounds were slightly different from one another and accordingly, the ESR technique by use of VO2+ was useful for an analysis of the calcium phosphates precipitated from supersaturated solutions. The ESR parameters of VO2+ adsorbed on ACP and Mg-TCP were found to be very similar to each other, suggesting that ACP and TCP resemble each other in the structure of their crystal surfaces.

    Notes: Summary The infrared (IR) spectra of KBr pellet preparations of tetracycline (TC) complexes with hydroxyapatite (HA) indicated that the binding of the antibiotic to bone mineral can occur independently at each of two ligands. The shifts in the IR spectrum occur for the keto-enol ligand of rings B and C and for the tricarbonylmethane grouping of ring A. Analogs that lack one or the other of these groups are able to form complexes with HA in which the common portion resembles that of the parent molecule.

    Description / Table of Contents: Résumé Les effects d'une dose de 1 mg de vitamine D3 (40 000 Unités,) administrée par voie intra-veineuse, ont été étudiés sur l'histologie osseuse et le métabolisme du phosphore chez 19 sujets contrôles dont l'histologie était normale et 28 malades présentant les caractères histologiques de l'ostéomalacie par carence vitaminique D. L'administration de la vitamine D n'a entraîné aucune modification histologique ou biologique significative chez les sujets contrôles. Mais chez les ostéomalaciques, il est apparu en moins de sept jours une augmentation très significative du front de calcification à l'interface tissue-ostéoïde-tissue-calcifié. Cette modification s'accompagnait d'une ascension progressive de la phosphatémie et de la réabsorption tubulaire du phosphore atteignant dans le même délai des valeurs normales.
    Abstract: Zusammenfassung Bei 19 Patienten mit normaler Knochenhistologie und bei 28 Patienten mit histologisch gesicherter Osteomalacie wurde die Wirkung von 1 mg Vitamin D3 (40000 Iv) i.v. auf die Knochenhistologie und den Phosphatmetabolismus untersucht. Bei den Kontrollpatienten konnten keine signifikanten Änderungen nach Vitamin D festgestellt werden, wogegen die Osteomalacie-patienten innerhalb von 7 Tagen eine deutliche Zunahme der Verkalkungszone an der Grenze zwischen Osteoid- und Knochengewebe zeigten. Diese Änderung war von einer fortschreitenden Zunahme des Serum-Phosphates, verbunden mit einer gesteigerten renalen tubulären Rückabsorption des Phosphates begleitet; beide kehrten anschließend gleichzeitig wieder zur Norm zurück.
    Notes: Abstract The effect of a 1 mg dose of intravenous Vitamin D3 (40,000 i.u.) on bone histology and phosphate metabolism was investigated in 19 patients with normal bone histology and 28 patients with histological evidence of osteomalacia. No significant changes occurred in the control patients after Vitamin D but the patients with osteomalacia showed a marked increase, within seven days, in the proportion of osteoid having a calcification front. This was accompanied by a progressive rise in the serum phosphate, which was associated with an increase in the renal tubular reabsorption of phosphate to normal.

    Notes: Summary It has previously been observed that after the third decade of life the tensile strength of bone decreases faster than its density [1], indicating that some microstructural change occurs in bone with advancing age. In order to try to elucidate this observation, the orientation and particle size of the mineral phase of bone have been studied with respect to age. Samples were taken from femora varying in age from 13 to 97 years. The results show that the degree of preferred orientation does not vary with age after the first decade of life. However, the percentage of small particles increased up to the fourth decade of life and thereafter decreased, a pattern of change similar to that observed for bone strength. It would seem to indicate that an increase in mineral particle size is a factor that contributes to the loss of bone strength and that an increased percentage of large particles in the later years of life could, along with osteoporosis, help to explain the increased fragility in the bones of the elderly.

Sans apport de vitamine D la croissance était lente, la cendre de la métaphyse diminuait, le serum calcium abaissait le serum phosphore augmentait. L'activité des encymes dans la métaphyse et la diaphyse de la tibia et du femur était la suivante: alkaline phosphatase, acide β-glycerophosphatase, activité protéolytique a pH 8, ta leucin-aminopéptidase et la glutamatdehydrogenase était augmenté dans la métaphyse; la lactatdehydrogenase et l'acide phénylphosphatase était diminué. Des changements comparables étaient decouvertes dans la diaphyse a l'éxception d'une activité de lactatdehydrogenase et de phenylphosphatase non alterée; en outre l'activité proteolytique a pH 5 était augmentée. Le manque de la vitamine D restait sans influence sur l'aspartattrans-aminase et alanintransaminase. L'activité proteolytique a pH 3,7 n'était pas changée. Ces altération encymatiques ne peuvent pas etre provoquées par une malnutrition parce que'elles n'etaient pas observées chez des animales avec une nuriture identique mais a, laquelle on avait ajouté de la vitamin D. Les encymes cataboliques ne manquait pas, la calcémie basse n'est donc pas éxpliquable par une réduction de la résorption osseuse. Les résultats sont plustôt en faveur d'une résorption osseuse très active.
    Abstract: Zusammenfassung Der Effekt von Vitamin D-Mangel auf Wachstum, Serumionen und Enzymaktivität im Knochen wurde bei der Ratte untersucht. Die Diät enthielt 0,6% Calcium und 0,6% Phosphor. Vitamin D-freie Diät hatte verzögertes Wachstum, Abnahme der Knochenasche in der Metaphyse, erniedrigtes Serumcalcium und erhöhtes Serumphosphat zur Folge. Die enzymatische Aktivität in der Metaphyse und Diaphyse von Tibia und Femur veränderte sich: alkalische Phosphatase, saure β-Glycerophosphatase, proteolytische Aktivität bei pH 8, Leucin-Aminopeptidase und Glutamatdehydrogenase waren in Knochenproben der Metaphyse vermehrt, Lactatdehydrogenase und saure Phenylphosphatase vermindert. Ähnliche Veränderungen der Enzymaktivität im Knochen fanden sich in der Diaphyse, mit Ausnahme unveränderter Aktivität von Lactatdehydrogenase und saurer Phenyphosphatase sowie erhöhter proteolytischer Aktivität bei pH 5. Der Vitamin D-Mangel ließ die Aspartat-und Alanin-Transaminasen sowie die proteolytische Aktivität bei pH 3,7 unverändert. Die Enzymveränderungen sind nicht durch geringere Nahrungsaufnahme bedingt, weil sie bei Tieren mit identischer Ernährung, aber ohne Vitamin D-Entzug, nicht beobachtet wurden. Ein Mangel an Abbaufermenten, welcher Hemmung der Knochenresorption bewirken und für den Abfall des Serumcalciums verantwortlich sein könnte, ließ sich nicht finden. Die Resultate sprechen eher für vermehrte Knochenresorption.
    Notes: Abstract The effect of vitamin D deficiency on growth, serum ions and bone enzymatic activity has been studied in rats. The diet contained 0.6% calcium and 0.6% phosphorus. Omission of vitamin D from the diet resulted in decreased growth, decreased metaphyseal bone ash, decreased serum calcium and increased serum phosphorus. Changes in enzymatic activity in metaphyseal and diaphyseal bone from the tibia and femur also occurred. Alkaline phosphatase, acid β-glycerophosphatase, proteolytic activity at pH 8.0, leucine aminopeptidase, and glutamate dehydrogenase were increased in metaphyseal bone samples. Lactate dehydrogenase and acid phenylphosphatase were decreased. Similar changes in bone enzymatic activity occurred in the diaphysis except that the lactate dehydrogenase and acid phenylphosphatase activities were not decreased and proteolytic activity at pH 5.6 was increased. The vitamin D deficiency did not affect aspartate or alanine transaminase or proteolytic activity at pH 3.7. These enzyme changes were not due to decreased food consumption because they did not occur in pair-fed animals. There was no lack of degradative enzymes that would impair bone resorption and lead to a fall in serum calcium. Rather the results were more consistent with an increase in bone resorption.

Cette opération a produit une augmentation du dépôt de radiocalcium dans les fémurs des rats rachitiques et également des rats traités par la vitamine D comparés à leurs contrôles respectifs ayant eu une opération simulée. Cependant, une analyse kinétique des données sur l'assimilation squelettique du radiocalcium a démontré que les degrés de déposition du minéral de l'os étaient amoindris par la suppression des capsules surrénales du moins chez les animaux traités par la vitamine D. D'autre part, la vitamine D a augmenté le degré de déposition plus que celui des contrôles rachitiques à la fois chez les animaux ayant eu ou non leurs capsules surrénales supprimées. De plus on a démontré que la vitamine D a rehaussé l'absorption intestinale autant que la déposition squelettique du radiocalcium chez le rat rachitique à qui on a enlevé les capsules surrénales.
    Abstract: Zusammenfassung In dieser Arbeit wurde die Auswirkung der Nebennierenentfernjng auf die Verteilung radioaktivem Calciums in rachitischen und Vitamin D behandelten Ratten untersucht. Entfernung der Nebennieren hatte eine erhöhte Ablagerung von radioaktivem Calcium in den Obserschenkelknochen der rachitischen und mit Vitamin D behandelten Ratten gegenüber den jeweiligen scheinoperierten Kontrollen zur Folge. Jedoch konnte durch kinetische Analyse der Ablagerungswerte von radioaktivem Calcium im Skelet festgestellt werden, daß die Menge der abgelagerten Knochenmineralien durch Entfernung der Nebennieren zumindest bei den mit Vitamin D behandelten Ratten herabgesetzt wurde. Auf der anderen Seite hat die Verabreichung von Titamin D die Ablagerungsmenge bei Ratten, denen die Nebennieren entfernt wurden und in solchen, die nicht operiert wurden, den rachitischen Kontrollen gegenüber erhöht. Zusätzlich konnte gezeigt werden, daß bei rachitischen Ratten nach Nebennierenentfernung und nach Behandlung mit Vitamin D erhöhte Darmabsorption und Skeletablagerung radioaktiven Calciums stattfand.
    Notes: Abstract The effect of adrenalectomy on radiocalcium distribution in rachitic and vitamin D-treated rats was studied. Adrenalectomy resulted in an increased deposition of radiocalcium in the femurs of both rachitic and vitamin D-treated rats when compared with their respective shamoperated controls. However, kinetic analysis of the data on the skeletal uptake of radiocalcium demonstrated that accretion rates of bone mineral were decreased by adrenalectomy at least in vitamin D-treated animals. Vitamin D administration, on the other, hand increased the accretion rate over that of rachitic controls in both adrenalectomized and non-adrenalectomized animals. In addition vitamin D was shown to enhance intestinal absorption as well as skeletal deposition of radiocalcium in the adrenalectomized rachitic rat.

La collagène d'os de mouton se trouvait être un bon catalysateur, tandis que des expériences antérieures ont démontré que la collagène de tendons de queues de rats était un catalysateur très faible. Le phase rapide de la déposition de l'apatite dans la collagène se termine aussitôt que le contenu du minéral a attaint au plus 50 à 60 pour cent, bien que la supersaturation du tampon est encore bien èlevée. Les résultats montrent que dans un tel système modèle la quantité du depôt minéral est réglée par des facteurs semblables à ceux qui opèrent pendant la calcification biologique.
    Abstract: Zusammenfassung Kollagen aus Schafsknochen und Rattenschwanzsehnen und Apatitkeime wurden verwendet in einem Modell-System zur Untersuchung der katalytischen Nukleation und der Fällung von Mineral in einem metastabilen Calciumphosphatpuffer. Kollagen aus Schafsknochen war ein guter Katalysator für die Nukleation, während in früheren Versuchen sich herausstellte, daß Rattenschwanzkollagen ein ganz schlechter Katalysator ist. Die schnelle Phase der Apatitfällung im Kollagen war beendet, wenn der Mineralgehalt bis zu 50–60% angestiegen war, obwohl der Puffer noch stark übersättigt war. Die Resultate weisen daraufhin, daß die Menge des gefällten Minerals in einem solchen Modell-System von ähnlichen Faktoren reguliert wird wie die biologische Verkalkung.
    Notes: Abstract Sheep bone collagen, rat tail tendon collagen and apatite seeds were used in a model system to study nucleation catalysis and mineral deposition in a metastable calcification buffer. Sheep bone collagen was shown to be a good nucleation catalyst, while earlier experiments have shown that rat tail tendon collagen was a very poor catalyst. The rapid phase of apatite deposition in the collagen was terminated as soon as a mineral content of not more than 50–60 per cent was reached, although the buffer was still highly supersaturated. The results suggest that the amount of mineral deposited in such a model system is regulated by factors similar to those operating in biological calcification.

    Notes: Summary Apatite in some samples of common urinary calculi consisting mainly of apatite or of calcium oxalate and apatite was subjected to X-ray and thermal analysis and electron microscopic investigation. The X-ray analysis has determined the lattice parameters agreeing with hydroxyapatite, and the mean crystallite size normal to the basal plane in a range of 200–800 Å. The thermal analysis is given by differential thermal analysis (DTA) and thermogravimetric (TG) curves. From the TG curve of a mixed apatite-calcium oxalate calculus, an approach is given to estimate the proportions of the constituent materials; the result is as follows: whewellite, 51.7%; weddellite, 16.6%; apatite, 19.1%; and organic materials, 12.6%. Analysis with the scanning electron microscope with energy dispersive X-ray spectrometer has shown that apatite is found as tiny spherules (5–10 µm in diameter) with fluffy surfaces resulting from projecting fine fibers or needles (about 0.5 µm in length). The fibers are considered to be mosaic aggregates of crystallites. The spherules fill interspaces of well-formed oxalate crystals or are included sporadically in individual crystals. Occasionally well-formed crystal-shaped oxalates are bordered with narrow bands consisting of apatite spherules (often as hemispheres) linked in a row.

    Notes: Summary The elemental composition of trabecular bone was compared for: (a) rats made rachitic on a low phosphorus, vitamin D-deficient diet; (b) rats fed the same diet but supplemented with vitamin D; (c) normal rats fed a standard laboratory diet with normal phosphorus and vitamin D levels. Quantitative energy dispersive X-ray microanalysis was performed on mineralized bone matrix at four sites: (1) clusters of mineral crystals in osteoid; (2) bone matrix adjacent to osteoid containing mineralization clusters; (3) peri-lacunar bone matrix; and (4) deep bone matrix distant from osteocytes. Estimations were also made of serum calcium, phosphorus, and 25-hydroxy-vitamin D, and of calcium, phosphorus, and hydroxyproline in whole bone. At bone sites 2, 3, and 4, the mineral content was greater in the normal group than in the other two groups. At each site, the mineral content of the rachitic bone matrix was greater than that from the vitamin D-treated group. A normal pattern of increasing mineral content with distance into the bone from a recently mineralized border was found in the normal and vitamin D-treated groups but was notably absent in the rachitic bones. Microprobe measurements of Ca:P molar ratios in hydroxyapatite standards and in normal rat bone were approximately 1.7. In both rachitic and vitamin D-treated bones, the Ca:P molar ratio was significantly higher than that in normal bones and correlated with serum Ca:P ratios. It is suggested that the increased Ca:P ratios in the rachitic and vitamin D-treated bones may be explained by an increased carbonate deposition.

    Notes: Summary Using transmission and scanning electron microscopy, we have studied the ultrastructure of a number of urinary calculi, mainly composed of calcium phosphate. Three fundamental kinds of calcium phosphates were detected: nonstoichiometric carbonate apatite, nonhexagonal octacalcium phosphate, and calcium-magnesium whitlockite. The influence that the organic matter, substitutions in the phosphate lattice of CO3 and Mg, and apatitic stoichiometry have on the ultrastructure of the calcium phosphate calculi has been detailed. An originating apatitic unity named U2 is assumed to be the responsible for all the different structures of calcium apatites appearing in renal calculi. On the basis of our observations, a mechanism whereby apatites grow is postulated; magnesium functions as an inhibitor for the growing mechanism.

    Notes: Summary Several phosphoprotein preparations (phosvitin, rat incisor and fetal calf molar dentin phosphoproteins) all inhibit apatite growth/replication from pre-existing crystal seeds in metastable solutions. Two stages of the crystal growth process were inhibited by these phosphoproteins. First, an initial lag period was induced, probably associated with seed surface phenomena. This period was prolonged indefinitely when a combination of phosphoprotein precoated seeds was used together with soluble phosphoproteins in the crystal growth reaction. Second, the phosphoproteins prolonged that stage of the reaction where octacalcium phosphate is the predominant mineral phase present prior to its conversion to the final apatite product. Pretreatment of the phosphoproteins with calcium diminished their inhibitory activity to seeded crystal growth as well as towards de novo apatite formation in synthetic extracellular fluids. The presence of collagen diminished the inhibitory activity of the phosphoproteins towards de novo precipitation but had no effect on phosphoprotein-modulated apatite crystal growth in the seeded systems. These results suggest a potential regulatory role for phosphoproteins in dentin mineralization.

    Notes: Summary Octacalcium phosphate (Ca8H2(PO4)6·5H2O) is considered to be a precursor in the formation of apatite in bones and teeth; a crucial step for incorporation of impurities appears to occur during its hydrolysis. The present study examines the role that octacalcium phosphate plays in the process of incorporation of carbonate into apatite. Chemical, X-ray diffraction, and infrared techniques were used. When octacalcium phosphate is hydrolyzed in the presence of sodium and carbonate ions in aqueous media, approximately one sodium and one carbonate ion seem to substitute for a calcium and phosphate ion, respectively, in forming apatite, and thea axis is shortened. The infrared spectrum of the product indicates that the carbonate is in the type B site, which is presumed to be a phosphate site. This mechanism is of particular importance since the presence of carbonate in human enamel appears to be related to caries susceptibility. A structural mechanism for the incorporation of impurities during hydrolysis of octacalcium phosphate is presented.

    Notes: Summary Twelve hours after the diet of 3-week-old chicks was changed from a 1% to a 0.1% concentration of calcium (Ca), the growth rate and circulating levels of growth hormone had fallen while renal 25-hydroxecholecaiferol-24-hydroxylase had risen. The amount of47Ca incorporated into bone from an injection given 18 h previously was lower than in the control birds. Over the following 21/2 days on the low Ca diet, the renal 1-hydroxylase activity rose and the plasma prolactin level fell, but the other parameters moved back toward the control level. It was concluded that early adjustments in hormonal and mineral metabolism counteract the acute effects of a dietary Ca shortage until longer-term adaptive changes begin to compensate for a continuing Ca deficiency. The renal hydroxylase activities were not directly influenced by the level of circulating growth hormone or prolactin.

    Notes: Summary Everted gut-sac-technique was used to study diurnal periodicity of active calcium transport in the small intestine of the adult rat maintained under 12:12 hours light-dark cycle. Similar to intact rats, diurnal fluctuations of the transport capacity persisted in adrenalectomized animals and in rats treated with 100,000 units of vitamin D3. Bilateral nephrectomy resulted in significant dampening of diurnal transport fluctuations. In contrast to the differences in calcium transport, diurnal changes of the calcium binding protein did not appear to be statistically significant when investigated in the intact, vitamin D-treated and nephrectomized rats.

    Notes: Summary The hydrolysis of previously prepared amorphous calcium phosphate (ACP) was studied in a solution “saturated” with ACP; this eliminated the initial consumption of acid due to ACP dissolution. The procedure established that conversion of a high-concentration ACP slurry to an apatite involves two processes: the first process consumes acid and indicates the formation of a more acidic calcium phosphate intermediary with the solubility of octacalcium phosphate (OCP); the second process consumes base and indicates the conversion of the intermediary to apatite and, possibly, direct conversion of ACP to apatite. The thermodynamic analysis of the solution composition data suggests that ACP converts into a nonstoichiometric apatite when the OCP-like intermediary is formed, and a stoichiometric apatite is formed when no OCP-like intermediary is involved.

    Notes: Summary The effect of vitamin D on bone changes during the reproductive cycle in female rats has been investigated. One group of female rats was maintained on a vitamin D-deficient diet and another group on a vitamin D-replete diet from weaning. Both groups were mated with normal males and changes in their bones were determined histomorphometrically during pregnancy, lactation, and after weaning. All vitamin D-deficient rats had bone changes typical of rickets. Pregnancy caused significant reductions in mineralized tissue of trabecular and cortical bone in the vitamin D-deficient rats. Lactation caused further significant reductions in mineralized tissues of cortical and trabecular bone in both the vitamin D-deficient and vitamin D-replete animals, with the greatest changes seen at weaning. Some restoration of mineralized tissues occurred following weaning. There was an increase in tetracycline-labeled bone surface in the vitamin D-replete animals during lactation, likely due to an increase in bone formation rates. In the vitamin D-deficient animals during lactation, there was a decrease in tetracyclinelabeled bone surface, likely due to severely depressed bone mineralization. These results indicate that the mobilization of calcium from bone to maintain pregnancy and lactation occurs by a mechanism independent of vitamin D.

    Notes: Summary Silicon has been reported to be involved in an early stage of bone formation as a result of earlier in vitro and in vivo studies in this laboratory. It is now possible to demonstrate that silicon exerts an effect on bone formation independent of the action of vitamin D. Day-old cockerels were fed Sideficient and Si-supplemented diets with adequate and no dietary vitamin D under trace element controlled conditions. At the end of 4 weeks chicks receiving dietary vitamin D3 (600 IU) exhibited optimal rates of growth whereas the growth rates of chicks receiving D2 (6000 IU) and no vitamin D were markedly depressed. There were no significant differences between growth of Si-deficient and Si-supplemented chicks on the same level of vitamin D. Skull and bone size was proportional to overall chick growth. However, all chicks on Sideficient diets irrespective of the level of dietary vitamin D had gross abnormalities of skull architecture, the overall skull appearance being narrower and shorter. The frontal area was narrower and the dorsal median line at the front parietal junction was depressed with a narrowing both posterior and laterally, stunting parietal and occipital areas. These abnormal areas showed fewer trabeculae and less calcification. Analyses of skull frontal bones for hexosamine, collagen, noncollagenous protein, and bone mineral demonstrated that the major difference was in collagen content, the Si-deficient skulls showing considerably less collagen at each level of vitamin D. These findings demonstrate that silicon has a significant effect on the bone matrix independent of vitamin D, and support the earlier postulate that silicon is involved in an early stage of bone formation.

    Notes: Summary Bone mineral content (BMC) was measured by125I photon absorptiometry every 3 months in 264 normal females (45–54 years) over a 2-year period together with serum samples for calcium, phosphate, magnesium, creatinine, alkaline phosphatases, potassium, and protein. A 48-h urinary calcium and creatinine measurement was obtained. The subjects were divided into 7 treatment groups and 3 placebo groups. Five of the treatments (thiazide, vitamin D3, fluoride + vitamin D3, fluoride, and 1αD3) were ineffective at the doses used; the annual loss of compact bone was 1.5–2.2% (-X=1.8%), similar to the loss seen with placebos. Estrogen and estrogen + thiazide, in contrast, produced a 1.34% annual increase of BMC. The subjects were divided into groups with low, medium, and high initial BMC. Also, individual regressions for bone change were calculated and the subjects were divided into groups of responders, maintainers, and losers (annual change of 〉0%, 0 to −1%, and 〉−1%, respectively). The initial BMC status did not consistently affect bone or biochemical responses to the therapeutic agents. Estrogen was effective even in subjects with high BMC, whereas the other agents did not inhibit bone loss even in subjects with low initial BMC. Virtually all subjects responded to estrogen positively; in contrast we could not identify a subset of “responders” with any of the other treatments. Time since menopause appeared to influence the bone changes, although it was not a significant effect given the sample size. Bone loss in groups not treated with estrogens was 2%/year at 20 months after menopause with a decline to 1.3%/year at 45 months post-menopause. There was no apparent decline in the bone response to estrogen during the first 4 years after menopause, and in fact bone response tended to increase with time.

    Notes: Summary The rates of diffusion of small ions and neutral molecules through isolated calvaria have been determined. Compared with data published on self-diffusion and diffusion through cartilage, H2O, 3-O-methylglucose, lactate, sulfate, and methylamine diffuse at approximately 2/3 the expected rate. Diffusion of H2O and sulfate was unaffected by the administration of the diphosphonate 1-hydroxyethane-1,1-diphosphonate (EHDP), although phosphate fluxes are markedly diminished. Diffusion of water was nearly doubled by vitamin D deficiency. A 1-week treatment with 1,25(OH)2D3 had no effect on H2O diffusion while returning calcium influx to normal. It is concluded that bone matrix permits a flow of small neutral and ionized molecules nearly comparable to that in cartilage. When changes in calcium and phosphate influx are observed, they can be ascribed to the exchange properties of bone mineral and not to changes in matrix permeability.

Ces faits semblent confirmer la possibilité non-apatitique de la phase non-cristalline du minéral osseux. Il peut s'agir d'un composé séparé et distinct.
    Abstract: Zusammenfassung Der erste feste Bestandteil, welcher beim Fällen von Calciumphosphat aus alkalischen Lösungen erscheint, ist strukturmäßig nicht-kristallin. Allerdings ist das Vorkommen eines nicht-kristallinen, aus einer Lösung hergestellten Orthophosphatsalzes nicht nur dem Calcium-phosphat-System eigen, können doch auch nicht-kristalline Magnesiumphosphate und Magnesiumammoniumphosphate unter ähnlichen Bedingungen gewonnen werden. Im Gegensatz zum Calciumsalz sind diese Verbindungen jedoch in Lösung anscheinend stabil. Alkalisch hergestellte Lösungen von Strontium- und Barium-Orthophosphaten ergeben keine nicht-kristalline Zwischenverbindungen. Für Strontium ist die intermediäre Phase Sr3(PO4)2·4H2O, während das Barium sogleich als Bariumhydroxyapatit ausfällt. Das Tristrontiumphosphat hingegen verwandelt sich autokatalytisch zu Strontiumhydroxyapatit. Die ähnliche stöchiometrische Form und das identische kinetische Verhalten der Strontium-Zwischenverbindung mit jener des amorphen Calciumphosphates läßt den Schluß zu, daß das Calciumphosphat eher ein Tricalciumphosphat als ein kryptokristallines, calciumarmes Hydroxyapatit sein könnte. Diese Annahme erhärtet die These, wonach die nichtkristalline Phase im Knochenmineral kein Apatit, sondern eine bestimmte, eigenständige Verbindung wäre.
    Notes: Abstract The first solid to appear in the precipitation of calcium phosphates from basic solutions is structurally non-crystalline. The existence, however, of a solution-prepared, non-crystalline orthophosphate salt is not unique only to the calcium phosphate system. Non-crystalline magnesium phosphates and magnesium ammonium phosphates can also be prepared under comparable conditions, but unlike the calcium salt, which converts into hydroxyapatite, these latter compounds are apparently solution stable. Preparations of strontium and barium orthophosphates from basic solution do not yield non-crystalline intermediates. For strontium, the intermediate phase is Sr3(PO4)2·4H2O, whereas the barium precipitates immediately as a stable barium hydroxyapatite. The tristrontium phosphate, however, converts autocatalytically into strontium hydroxyapatite. The analogous stoichiometry and identical kinetic behavior of the strontium intermediate with that of the amorphous calcium phosphate suggest that the latter could be a tricalcium phosphate rather than a cryptocrystalline Ca-deficient hydroxyapatite. This supports the possibility that the non-crystalline phase in bone mineral may not be an apatite but instead a distinct and separate compound.

    Notes: Summary The present study examined precipitate development induced in phosphate solutions by the ionophoric translocation of Ca2+ across a bulk organic solvent barrier. Experiments were conducted at pH 7.4 and 25 or 37° in a three-compartment Pressman cell. The aqueous reaction (0, 2.2 or 22 mM phosphate, 100 mM K+) and donor (1.3 or 13 mM Ca2+, 0.8 mM Mg2+, 100 mM Na+) compartments were separated by a CHCl3 compartment rendered permeable to cations by the addition of the carboxylic ionophore X-537A (2 or 20 mM). The resultant cation movements increased Ca2+, Mg2+, and Na+ concentrations in the reaction compartment at the expense of K+ loss to the donor compartment. The magnitude of the K+ counterflow and the efficiency of the ionophore-mediated Ca2+ ↔ 2K+ exchange reaction resulted in appreciable Ca2+ overshoots in the reaction compartment. In the absence of phosphate, Ca2+ increases exceeded donor levels by several-fold. With phosphate present, the Ca2+ flux was sufficient to induced precipitation. Generally, the first solid formed was amorphous. The amorphous precipitate, however, was unstable and converted to an apatite-like crystalline phase. Both carbonate (26 mM) and Mg2+ (0.8 mM) in the reaction solution delayed but did not prevent the conversion. The possible relevance of these findings to matrix vesicle calcification is discussed.

    Notes: Summary The present study examined crystal growth on enamel and synthetic apatite seed surfaces in dilute supersaturated solutions by means of transmission electron microscopy. At all supersaturations, new growth initially appeared on the ends of the seed crystal. In solutions undersaturated with respect to octacalcium phosphate (OCP), this growth was needlelike in appearance. Above the solubility point for OCP, the growth frequently took the form of thin, platelike crystals. The relevance of these findings to precursor phase formation is discussed.

    Notes: Summary To evaluate the effect of prednisone and triple treatment (sodium fluoride, calcium, and vitamin D) on trabecular and cortical bone serial bone mineral content (BMC) measurements were made at a metaphyseal (BMCD) and diaphyseal (BMCP) site on the forearm on 31 consecutive and previously bone-healthy patients scheduled for at least 24 weeks high-dose prednisone treatment. The patients were randomized into two further treatment groups: group I (n=16) received prednisone plus triple treatment and group II (n=15) received only prednisone. The two groups were similar with regard to age, sex, prednisone dose, and initial BMC. During 24 weeks treatment, BMCD (partially representing trabecular bone) and BMCP (mainly representing cortical bone) fell significantly and similarly, demonstrating that there is no preventive effect on bone mineral loss on the triple regimen. The BMC fall after 12 weeks was significantly more pronounced for metaphyseal (partially trabecular) than for diaphyseal (cortical) bone, whereas the values did not differ significantly after 24 weeks; this indicates a greater sensitivity to the hormone treatment of trabecular bone. In the entire group, the fall in BMC correlated positively with individual prednisone dose, significant at the diaphyseal site (r=0.39,P〈0.05), but not at the metaphyseal site (r=0.31, P=0.08). It is concluded that corticosteroid-induced osteopenia is a diffuse bone disease which affects trabecular as well as cortical bone, suggesting that BMC measured on the forearm reflects changes in bone mineral at other locations.

The purpose of this research is to examine the role of proteolipid in cartilage calcificationin vivo by comparing the proteolipid concentration of normal and vitamin D-deficient chick epiphyseal cartilage, the relationship of proteolipid to other tissue lipids, and its ability to supportin vitro apatite formation. Proteolipid was isolated from the upper growth centers (reserve cell zone, upper proliferative zone) and lower growth centers (lower proliferative, hypertrophic, and calcified cartilage zones) of long-bone epiphyses from 3-week-old normal and rachitic male white leghorn chicks by Sephadex LH-20 chromatography of the crude phospholipid component of the total lipid extract. In both normal and rachitic tissue the proteolipid/dry weight and proteolipid/total lipid ratios were greater in the lower growth center than in the upper zones. No statistically significant change in the proteolipid/total lipid ratio in rachitic tissues relative to comparable cell zones in normal cartilages was observed. However, there was an increase in the nonproteolipid phospholipid content of rachitic tissues, altering the relative proteolipid/phospholipid composition. Whereas proteolipids from normal tissue supportedin vitro calcification, proteolipids from rachitic tissues did not, indicating a direct effect of vitamin D on proteolipid structure. These data support the hypothesis that failure of rachitic cartilage to calcifyin vivo may be due in part to alterations in phospholipid and proteolipid metabolism.

    Notes: Summary Chick embryos were injected in the yolk sac at various ages with various doses of different vitamin D3 metabolites. Serum concentrations of total calcium and inorganic phosphate were determined 24 h after the injection and histological and electron microscopic studies of the tibiae were conducted 3–6 days after. Confirming previous results, the injection of 1,25(OH)2D3 was found to produce significant hypercalcemia and hypophosphatemia. The dose required to produce these effects decreased with age: 100 ng on the 9th day, 50 ng on the 11th, and 10 ng on the 15th. This finding is interpreted as resulting from the fact that the specialized cells in the chorionic epithelium which are considered to be involved in mineral resorption from the shell differentiate between the 11th and 13th days. Although no bone changes were observed in embryos injected before the 11th day, a rim of unmineralized trabeculae (osteoid) was observed at the periphery of the cortex of the tibial diaphysis in the embryos which had been injected after that age. Thus, in embryos injected on the 11th day with 100 ng 1,25(OH)2D3, the trabeculae formed between the 11th and 14th day remained unmineralized until the 15th or 16th day at which time they completed their mineralization. In the embryos injected on the 14th day, the alterations were more severe and could be produced with doses 10 times smaller than those required when the injections were made on the 11th day. At all ages, the doses that produced an osteoid rim also induced hypercalcemia and hypophosphatemia. The electron microscopical study of the osteoid trabeculae showed that osteoblasts and osteocytes had normal cytological characteristics and that the bone matrix did not present changes other than the reduction in mineral deposition. While the above findings do not exclude a direct action of 1,25(OH)2D3 on bone cells as the mechanism of osteoid formation, they do underline the importance of the humoral changes at least as partial determinants of this phenomenon. The activities of various vitamin D metabolites were compared using as parameter the threshold-dose required to produce a rim of unmineralized trabeculae in the tibia of 14–15 days embryos (T-D). The most active metabolite appeared to be 1,25(OH)2D3 (T-D: 10 ng); it was followed by 1,24,25(OH)3D3 (T-D: 100 ng) and 1,25,26(OH)3D3 (T-D: 100 ng). Vitamin D3 itself (T-D: 100 µg), 25(OH)D3 (T-D: 2.5µg) and 24,25(OH)2D3 (T-D: 5 µg) produced similar responses but only when administered in much larger doses.

    Notes: Summary An aqueous density fractionation for calcifying tissues was tested for its ability to prepare fractions corresponding to precise mineralization stages, suitable for further protein and crystal studies. Two fractions of immature enamel corresponding to different densities were prepared, using cooled cesium salt saturated solutions, and compared for crystal size and amelogenin molecular weight distribution. For the first time, a steep increase in crystal width was directly correlated to protein degradation in keeping with increasing mineralization. Thus, this paper describes a method for obtaining an accurate fractionation of a calcifying tissue according to its true density heterogeneity. The recovered fractions are shown to be suitable for both crystal and protein studies.

    Notes: Summary An original method for fractionating and preparing isolated crystals of homogeneous size was developed. It was demonstrated that enamel apatite crystals are at least 100 µm long. The flexibility of the very long crystallites was demonstrated. Crystal curvatures, accounting for the irregular course of the prisms through the enamel thickness, were visualized and measured. It was shown that in the deep forming enamel layer, lateral branches may grow out of the crystals and crystal fusing often occurs, inducing the crystallites to assume pyramidal shapes with their wide bases pointing toward the dentino-enamel junction and one or two tops toward Tomes' processes. During the maturation process, the two tops of the still immature crystals also fuse so that the mature crystals acquire a rodlike aspect, with parallel faces and steplike graduations along thec axis, allowing a close contact between the crystals. These results support the hypothesis that the crystallites would be continuous from the dentino-enamel junction to the surface.

To determine whether an abnormality of vitamin D metabolism exists in this disease, the profiles of the metabolites of vitamin D were determined in normal andHyp mouse plasma.Hyp and normal mice were fed a vitamin D-deficient diet and received 1,23H-vitamin D3 at 16 Ci/mmol by stomach tube at 5 ng/g body weight (0.21 µCi/g b.w.) on alternate days for 14 days. The dose of vitamin D given maintained near normal plasma 25-OH-vitamin D. Thus the mice were in a vitamin D-replete state with all metabolite pools labeled with3H. Plasma was collected from 4 normal and 4Hyp mice. The plasma was extracted, and the extracts were chromatographed separately for each mouse on an LH-20 column. Each major peak of radioactivity was rechromatographed using high performance liquid chromatography on a Zorbax-Sil column using solvent systems known to resolve several vitamin D metabolites. Twenty-one radioactive peaks were identified. The disintegrations per minute of3H in each peak were quantified and converted to plasma concentration using the known specific activity of the administered vitamin D. The 25-OH-vitamin D accounted for 55% of the circulating radioactivity, and 24,25-(OH)2-vitamin D accounted for 22%. The plasma levels of 24,25-(OH)2-vitamin D were similar to levels previously reported by us using protein binding assays. No peaks of radioactivity were missing in the plasma extracts of theHyp mice. Also there was no evidence that plasma 24,25-(OH)2-vitamin D was elevated in theHyp mice.

    Notes: Summary Eighty-eight adult male Sprague-Dawley rats were given a diet with either (a) 0.5% Ca and 0.6% P or (b) 0.01% Ca and 0.6% P. Osteopenia was created by adding prednisolone to the diet. The prophylactic effect of oral 1,25(OH)2D3 on the osteopenia was studied. It was found that prednisolone osteopenia in the rat was associated with defective Ca absorption. By giving an oral dose of 1,25(OH)2D3, it was possible to maintain normal Ca absorption during prednisolone treatment and to prevent the bone loss. No significant hypercalcemia or any kidney calcifications were seen. These results are in contrast to earlier findings, in which subcutaneous administration of 1,25(OH)2D3 failed to prevent prednisolone osteopenia because of its tendency to increase bone resorption.

    Notes: Summary Tritium-labeled 1,25 (OH2) vitamin D3, when injected into vitamin D-deficient adult and pregnant rats is concentrated and retained strongest in nuclei of cells in the outer root sheath of the hair, followed by the stratum granulosum, spinosum, and basale of the epidermis. In the hair follicle, in addition to the most heavily labeled outer root sheath, nuclear labeling exists also in cells of the hair bulb and of the inner root sheath, as well as in basal cells of the sebaceous gland. In contrast, cells of the dermal papilla and the connective tissue of the dermis are generally unlabeled, except for labeled cells in the outer connective tissue sheath at the infundibulum of vibrissae of 20-day fetal rats and a few scattered labeled cells in the dermis, probably macrophages. In the developing hair, in 18- and 20-day fetal rats, a distinct topographic pattern of labeled cells can be seen, which is characteristic of the different stages of hair follicle development. In the hair germ, heavily labeled cells appear first in the stratum spinosum. In the hair peg, they remain in this position in its juxtaepidermal portion; however, when a dermal papilla develops, heavily labeled cells assume a marginal position. This suggests a sequential epidermal-epidermal and mesenchymal-epidermal receptor induction. Injection of tritium labeled 25 (OH) vitamin D3 did not show nuclear concentration in these tissues and excess unlabeled 25 (OH) vitamin D3 — unlike excess 1,25 (OH2) vitamin D3 — did not prevent nuclear uptake of tritium labeled 1,25 (OH2) vitamin D3. The results indicate differential effects of 1,25 (OH2) vitamin D3 on different structures in the epidermis and dermis.

    Notes: Summary Specific antibodies raised against human cerebellar calciumbinding protein (CaBP) intensely labelled the cochlear hair cells of the rat. The vestibular hair cells also stained weakly. In both inner and outer cochlear hair cells, the cuticular plate was the most stained area. These results suggest that CaBP may prevent excessive concentrations of intracellular calcium and thus modulate some Ca2+-mediated biochemical processes, especially at the level of the cuticular plate and stereocilia; CaBP could be involved in the mechanochemical coupling of hearing or vestibular function.

    Notes: Synopsis Diets deficient in vitamin D effected a significant increase in plasma triiodothyronine (T3) concentration in raibow trout (Salmo gairdneri); different levels of dietary calcium exerted no effect on plasma T3 levels. These effects of vitamin D deficiency on plasma T3 levels appeared to be reversible, vitamin D supplementation after a period of vitamin D deficiency lowered T3 levels. Vitamin D3, vitamin D2 and the metabolites 25(OH)-D3 and 1, 25(OH)2D3 were all effective in lowering plasma T3 levels; vitamin D3 appeared to be more effective than vitamin D2. There appeared to be a correlation between weight gain and plasma T3 concentration in the groups fed different types and levels of vitamin D supplementation suggesting that the increased T3 levels may be a compensatory increase to the reduced weight gain of the vitamin D deficient fish. Plasma T4 levels were not affected by dietary vitamin D deficiency.

Both light and electron microscopy showed (1) that labelling was entirely restricted to the Purkinje cells, (2) that it appeared very early in Purkinje cell development, (3) that the entire cell was labelled from the tip of the smallest dendrites to the axonal terminals, and (4) that with increasing age, the immunoreaction appeared to be progressively restricted to the cell and organelle membranes.

    Notes: Abstract The dielectric constant of bone and its two components (collagen and apatite) were determined at 9.2 GHz in a room temperature environment by the cavity perturbation technique. After samples were exposed to ultraviolet light, repeat measurements under identical conditions showed decreases in both the real and the imaginary parts of the dielectric constant in all cases. The present work describes the technique and a possible mechanism of ultraviolet interaction with bone.

    Notes: Summary The vitamin D-dependent calcium-binding protein (CaBP) was studied in relation to the age of the cell, in isolated epithelial cell populations removed from rat duodenum. Alkaline phosphatase and thymidine kinase activities were used as markers to characterize differentiated villus cells and undifferentiated (mitotically active) crypt cells, respectively. CaBP distribution along the length of the villus, as established by radioimmunoassay, appears as a gradient increasing from the crypt to the tip of the villus. CaBP concentration in cells is shown to be (i) negatively correlated with the thymidine kinase activity of cells, and (ii) positively correlated with the alkaline phosphatase activity of cells. This indicates that CaBP is absent in crypt cells and appears in differentiated cells with the development of the brush border. Thus CaBP, like alkaline phosphatase, can be considered as an indicator of enterocyte maturation. These data were also confirmed by studying the cellular localization of the protein. In addition both indirect immunofluorescence and immunoperoxidase staining methods reveal that antibody against CaBP decorates the terminal web, but not the microvilli of the brush border of mature absorptive cells. The results suggest that CaBP may act as a modulator of some Ca2+-mediated biochemical processes at the level of the enterocyte brush border.
