    Notizen: Abstract Rate constants and activation parameters for the formation and the dissociation of the monocomplex of nickel(II) with thiocyanate ion in dimethylsulfoxide have been determined by stopped-flow measurements. The results for thiocyanate ion, and also those for several other unidentate ligands, are consistent with a dissociative interchange mechanism in which solvent exchange constitutes the rate-determining step. On the other hand, previous results have shown that for multidentate ligands the ring-closure step also may be rate limiting.

    Notizen: Abstract The rate of the reaction Fe(CN) 6 4− +W(CN) 8 3− →Fe(CN) 6 3− +W(CN) 8 4− in aqueous solution has been studied at various temperatures and ionic strengths, in the presence of Group IA cations. The results, combined with earlier work on the ion pairing of these compounds, give an indication of the number of cations present in the activated complex, a number which increases steadily in going from Li+ to Cs+ as the associated cations. Activation parameters for the reaction at various total [M+] are also evaluated.

    Notizen: Abstract Kinetic studies of the hexacyanoferrate(III) oxidation of 1-propanol and 2-propanol have been carried out in aqueous alkaline medium. The reaction velocity is of first order with respect to alcohols, alkali and hexacyanoferrate(III). The kinetic data suggest that the oxidation involves the formation of an anion of the substrate undergoes oxidation with hexacyanoferrate(III) via charge transfer process. The free radical thus produced is further oxidised to form the final products.

    Notizen: Abstract The kinetics of the bromination of acetanilide has been studied with the diffusion layer titration method. The results have been obtained using the apparatus which consists of the ring-disc electrode and an amperostat-potentiostat system. Dependence of the ring current on the disc current has been determined as a function of rotation speeds of the electrode, of the solution concentration and temperature. It has been shown the bromination reaction of acetanilide exhibits by first order kinetics. In that case Br2 and Br3 − are the brominating species. The rate of bromination changes with the concentration of the Br− ions. This reaction rate depends on reactions of molecular bromine with acetanilide. For 0.033〈[Br−]〈0.173M the rate constant changes in the following range: $$17530M^{ - 1} s^{ - 1}〈 k〈 6400M^{ - 1} s^{ - 1} ([H + ] = 1,34;T = 298K)$$

Die Kinetik der Nitrosierung von Piperazin (PIP) in wäßriger Perchlorsäurelösung wurde mittels einer differentiellen spektrophotometrischen Methode verfolgt. Es ergab sich für denpH-Bereich 0,85–4,36 folgendes Zeitgesetz: $$v_0 = \left[ {nitrite} \right]_0 2 \left[ {PIP} \right]_0 /\left( {1 + f/\left[ {H^ + } \right]} \right)^2 \left( {g \left[ {PIP} \right]_0 + h + j\left[ {H^ \div } \right]} \right)$$ wobei [Nitrit]0 und [PIP]0 die anfänglichen stöchiometrischen Konzentrationen bedeuten. Bei 298,2 K und μ=1,0M,f=(1,17±0,11) 10−3 M,g=(3,5±0,7)·10−2 M s,h=2,6·10−6 M 2 s andj=(0,95±0,04)M s. Bei Erhöhung der Acidität ([HClO4]≥1M) tritt ein neuer kinetischer Term auf: $$v_0 ' = p\left[ {Nitrit} \right]_0 \left[ {PIP} \right]_0 $$ Bei 298,2 K und μ=3,0M,p=(1,9±0,2) 10−3 M −1 s−1. Es wird ein genereller Mechanismus für die Nitrosierung jedesN-nitrosierbaren Substrates in wäßriger Perchloratlösung vorgeschlagen, wobei als nitrosierende Agentien ausschließlich N2O3 und H2NO2 +/NO+ auftreten. Es werden die Besonderheiten dieses Mechanismus bezüglich derpK-Werte derN-nitrosierbaren Substrate diskutiert.
    Notizen: Abstract The mechanism of formation ofN-nitroso compounds, which are considered as potential chemical carcinogens was studied. The kinetics of nitrosation of piperazine (PIP) in aqueous solution of perchloric acid have been investigated using a differential spectrophotometric technique. A general mechanism for the nitrosation of anyN-nitrosable substrate in aqueous perchloric solution in which the only nitrosating agents are N2O3 and H2NO2 +/NO+ is proposed. Also, the various particularities of this mechanism, according to thepK of theN-nitrosable substrate, are discussed.

Die Oxidation ist erster Ordnung bei niedrigen Konzentrationen von Xylit und Galaktit und geht bei Erhöhung der Konzentration gegen null. In gleicher Weise wurde eine Kinetik erster Ordnung bezüglich Hydroxyl bei niedrigen Konzentrationen und eine erniedrigte Ordnung bei höheren Konzentrationen für die Oxidation von Xylit beobachtet; bei Galaktit bleibt die Oxidation auch bei höheren Hydroxyl-Konzentrationen erster Ordnung. Es wird angenommen, daß die Reaktion über einen aktivierten Komplex zwischen [KFe(CN)6]2− und dem Substrat-Anion verläuft; dieser Komplex zerfällt in [KFe(CN)6]3− und ein Substrat-Radikal. Ein möglicher Reaktionsmechanismus wird vorgeschlagen.
    Notizen: Abstract Kinetics of oxidation of xylitol and galactitol by hexacyanoferrate(III) ion in aqueous alkaline medium is reported. The reaction rate is of first order with respect to hexacyanoferrate(III) in each substrate. The reaction is first order at lower concentrations of xylitol and galactitol and tends towards zero order as the concentration increases. Similarly first order kinetics was obtained with respect to hydroxide ion at lower concentrations and tends to lower order at higher concentration in the oxidation of xylitol; in the oxidation of galactitol the reaction is first order with respect to hydroxide ion even up to manyfold variation. The course of reaction has been considered to proceed through the formation of an activated complex between [K Fe(CN)6]2− and substrate anion which decomposes slowly into radical and [K Fe(CN)6]3−. A probable reaction mechanism is proposed.

Im Temperaturbereich von 40–90° erfolgt die Auflösungsreaktion kinetisch kontrolliert, wobei eine Aktivierungsenergie von 14±1 kcal mol−1 (59±5 kJ mol−1) gefunden wurde.
    Notizen: Abstract Using the rotating disc method, the rates of dissolution of natural monoclinic pyrrhotite, FeS1.14, in oxygen-free aqueous solutionsS([H+]=0.1, [Na+]=0.9, [ClO 4 − ]=1.0 mol kg−1) were determined. In the temperature range 40–90 °C the dissolution reaction occurs under kinetic control; the activation energy being 14±1 kcal mol−1 (50±5 kJ mol−1).

Die Reaktion zeigt erste Ordnung gegenüberNBS und Allylalkohol. Bei relativ hoher Säurekonzentration zeigt sich keine Änderung der Reaktionsgeschwindigkeit, bei niedriger Säurekonzentration wird die Reaktionsgeschwindigkeit beträchtlich erhöht. Es wurde kein primärer Salzeffekt festgestellt. Bei varriierender Quecksilberacetatkonzentration bleibt die Reaktionsgeschwindigkeit gleich, bei Abwesenheit von Quecksilberacetat wird jedoch die Geschwindigkeitskonstante erhöht. Die kinetischen Parameter,E a, derArrheniusfaktorA, ΔH ≠, ΔG ≠ und ΔS ≠ wurden bestimmt. Ein Geschwindigkeitsgesetz in Übereinstimmung mit den experimentellen Befunden wurde abgeleitet und ein Mechanismus vorgeschlagen.
    Notizen: Abstract The kinetics of oxidation of allyl alcohol byN-bromosuccinimide (NBS) has been studied at 35 °C in aqueous medium. The reaction shows first order dependence on bothNBS and allyl alcohol. In fairly high acid concentration, there is no change in the rate of the reaction but at low acid concentration, the rate is considerably enhanced. There is no primary salt effect. At varying mercuric acetate concentrations, the rate constant remains the same. But in the absence of mercuric acetate, the rate is enhanced. The kinetic parameters,E a,Arrhenius factorA, ΔH≠, ΔG≠ and ΔS≠ have been calculated. A rate law in agreement with experimental results has been derived. A mechanism is proposed.

In allen Fällen ist die Reaktion von zweiter Ordnung hinsichtlich ICl und von erster Ordnung hinsichtlich des Substrats, d. h. mit einer Gesamtordnung von drei. Geschwindigkeitskonstanten wurden für jedes Substrat bei 20°, 30°, 40° und 50°C gemessen. AusArrhenius-Diagrammen wurden Aktivierungsenergien ermittelt und auch andere kinetische und thermodynamische Parameter wurden bestimmt. Ein Mechanismus wird vorgeschlagen, wobei die verschiedenen Parameter diskutiert werden. Lösungsmitteleffekte werden ebenfalls diskutiert und die Daten mit Nitrobenzol und Essigsäure als Lösungsmittel miteinander verglichen.
    Notizen: Abstract The kinetics of the addition of iodine monochloride (ICl) to some vinyl compounds in nitrobenzene solvent was investigated. In all cases the reaction follows second order dependence on ICl and first order on the substrate, making the total order three. Rate constants were measured for each substrate at 20°, 30°, 40° and 50°C.Arrhenius plots were made from which activation energies were evaluated. Other kinetic and thermodynamic parameters are reported. A suitable mechanism is proposed for the reaction and based on this, the various parameters are discussed. Solvent effects are discussed and the data with nitrobenzene and acetic acid solvents are compared.

    Notizen: Abstract The alcoholysis of various esters of aromatic carboxylic esters with octadecanol in the presence of lead stearate was investigated by chromatographic analysis of the reaction mixtures. The reactivity of the esters was found to be strongly affected by the substitution pattern of the aromatic nucleus as well as by the structure of the alkoxy group. Electron donating substituents in a suitable position lead to a remarkable increase in reactivity compared to the unsubstituted alkyl esters.

    Notizen: Abstract The influence of NaClO4, NaCl and Na2SO4 on the oxidation of Fe(phen) 3 2+ by Ce(IV) was investigated by means of the stopped-flow method. At the concentrations range of NaClO4 and NaCl 0.1–1.0M the rate constant values decrease from 1.03·105 to 0.56·105M−1s−1 and from 1.08·105 to 0.81·105M−1s−1 respectively. In varying concentrations of Na2SO4 solutions (0.05–0.35M) the rate constant values decrease from 1.05·105M−1s−1 to 0.45·105M−1s−1. Taking into account the negative salt effect the mechanism of the reaction progress is proposed.

Bis zu einer Cu(II)-Konzentration von≤2×10−5 M ist die Geschwindigkeitskonstante der Katalysatorkonzentration proportional; darüber wird eine konstantbleibende Geschwindigkeit beobachtet, die nun von der Cu(II)-Konzentration unabhängig ist. Neutralsalze haben keinen Effekt auf die Geschwindigkeitskonstante. Es wird für den katalysierten und unkatalysierten Reaktionsablauf ein Mechanismus vorgeschlagen und ein mathematischer Ansatz präsentiert.
    Notizen: Abstract The kinetics of uncatalysed and Cu(II) catalysed oxidation of arginine monohydrochloride was investigated. Both reactions follow a singular order dependence each in oxidant and substrate. An inverse order dependence is reported with the alkali concentration. A plot of observed rate constant versus Cu(II) concentrations Cu(II)≤2.0×10−5 M is linear; from the intercept the rate constant for the uncatalysed pathway was calculated. However, at high copper ion concentrations i.e. Cu(II)〉2.0×10−5 M a fixed value of rate constant was found for all catalyst concentrations. Added neutral salts show an insignificant effect on the reaction rate. Mechanisms were proposed for both cases and rate expressions were derived by applying steady state assumptions.

    Notizen: Abstract Rate constants of the cleavage reaction of silicon aryl linkages with HBr were estimated for various aryl derivatives by means of NMR. A row of substituents was obtained, dependent on the cleavage rates. Correlation with theHammett function shows the electrophilic character of the reaction. The different cleavage rates give the possibility for specific cleavages of aryl groups from silanes.

    Notizen: Summary The kinetics of seeded crystal growth of calcium apatites were studied in dilute supersaturated solutions at various levels of fluoride concentrations. Initial precipitation rates were enhanced by fluoride concentrations higher than 0.05 ppm. The analytical results are consistent with the precipitation of fluoridated hydroxyapatites, Ca5Fx-(OH)1−x(PO4)3, FHA. The degree of fluoridation, X, appears to be determined by the activity of HF in solution, which varies for the various initial fluoride levels but remains fairly constant during precipitation. Thus the composition of the precipitating phase was the same for a given solution whether 25 or 10 mg of hydroxyapatite was added as seeds. All the experimental results are consistent with the BCF theory, which relates the mean linear rate of growth, RL, to the supersaturation, DS, by the expression RL=C1T(DS-1)1n(DS)tanh(C2/T 1n DS), in which DS is the supersaturation defined by mean molar activities with respect to the precipitating FHA, T the absolute temperature, and C1 and C2 are constants calculated from the experimental results. Consequently, the crystal growth appears to take place in surface kinks and to be controlled by surface diffusion. Since crystal growth in most biological systems takes place at fluoride concentrations within the experimental range used, it seems probable that it occurs along the model advanced in the present investigation.

    Notizen: Summary A thermodynamic analysis has been made of the secondary transition stage in the spontaneous precipitation of calcium phosphate following the amorphous-crystalline transformation. The first formed crystalline material has a solubility similar to that of octacalcium phosphate (OCP) and the computed thermodynamic solubility product remains invariant in the pH range 7.00–8.60. The duration of the secondary stage is sensitive to pH and the transition appears to occur by hydrolysis of the first formed OCP-like phase to a more basic apatitic phase with a tricalcium phosphate (TCP) stoichiometry. The crystalline material at the end of this transition has an invariant solubility product, in the pH range 7.00 to 8.60, when the TCP-like molecular formula is assumed. Changes in the solution chemistry which accompany the solid-tosolid transitions are consistent with the above conclusions. The results of this study are also consistent with those of a previous study which suggest that the stability of the amorphous calcium phosphate phase is dependent upon the instability of the solution phase with respect to OCP formation.

    Notizen: Summary A method is described for the isolation and purification of a sulphated glycopeptide from human supragingival calculus. The compound was isolated after using EDTA treatment, 2 M CaCl2 extraction, proteolytic digestion, ethanol precipitation, and finally purified by DEAE cellulose chromatography. It migrated as a single component on cellulose acetate electrophoresis, and chemical and infrared spectral analysis showed the presence of covalently attached sulphate groups. The sulphated glycopeptide was distinguished from being a sulphated glycosaminoglycan.

    Notizen: Summary The study of interstellar molecules broadly includes two areas of interest. One area uses the unique ability of molecules to act as probes of the physical conditions in the cold, dense, visually opaque component of the interstellar medium. The physical properties of this and other components of the interstellar medium are summarized. The other area deals with the chemistry of interstellar molecules, recent aspects of which are emphasized in this review. Gas-phase chemistry, shock chemistry, and grain surface chemistry are discussed in the context of recent observations. No present observations suggest that surface reactions are relevant, but neither can they be ruled out. Ion-molecule reactions are clearly operative, at least for the simpler species. Chemical isotope fractionation is reviewed, andd it is concluded that the complexities of the chemistry allow no cosmological conclusions to be drawn from observations of deuterium in interstellar molecules, while the presence of13C in interstellar molecules permits an estimate of the12C/13C ratio which is consistent with the current concepts of the nucleosynthesis history of the Galaxy. Possible connections between interstellar molecules and the early molecular history of the solar system are discussed.

    Notizen: Abstract The kinetics of the diazotization of α-naphthylamine1 in water HCl solution from 0,2N to 2.0N at 0 °C were investigated. It was found that the nitrosation reaction $$\alpha --C_{10} H_7 NH_2 + NOCl\mathop \rightleftharpoons \limits^{k_v } \alpha --C_{10} H_7 NH_2 NO^ + + Cl^ - $$ is a preceeding advance-back-reaction (velocity coefficient of the nitrosation is 1.92·1010l mol−1 s−1). The decomposition of I by splitting off a proton is the rate determining reaction. The free enthalpy of activation for the nitrosation reaction equals 12.94 kJ/mol.

Es wurden die thermodynamischen Parameter der Reaktion bestimmt und auch einHammet'scher ϖ-Wert (+0,6) für die Oxidation der Methylarylketone ermittelt.
    Notizen: Abstract The kinetics of the reaction of three substituted acetophenones withN-Bromosuccinimide was studied in perchloric acid media in presence of mercuric acetate. The reactions were found to be zero order with respect toNBS while the order with respect to ketones and [H+] was found to be unity. The addition of succinimide mercuric acetate and sodium perchlorate has no effect on the rate of oxidation and the rate increases with the decrease in dielectric constant of the medium. Kinetic investigations have revealed that the order of reactivity ism-nitroacetophenone 〉 p-chloroacetophenone 〉 p-methylacetophenone. TheArrhenius equation has been found to be valid in the temperature range 35–55°. Thermodynamic parameters have been calculated. Mechanistic pathways of the reactions are discussed and a rate equation is derived.Hammett's plot gives a ϖ value of +0.60 for methyl-aryl-ketones.

Ab ThO2:C=1:50 war die Reaktionsgeschwindigkeit vom Mischungsverhältnis unabhängig; das Endprodukt war immer ThC2. Der logarithmische Gewichtsverlust war direkt proportional der Zeit, und dieArrheniusgerade zeigte einen Knickpunkt bei 1710 K: unterhalb dieser Temperatur ergab sich eine Aktivierungsenergie von 440 kJ, oberhalb eine solche von 260 kJ. Die Temperatur des Knickpunkts entspricht der Umwandlungstemperatur von monoklinem in tetragonal raumzentriertes ThC2.
    Notizen: Abstract The reduction of thoria with excess graphite was studied with a thermo-balance in vacuum between 1620 and 1920 K. From Th02:C=1:50 the rate of reaction was independent of the ratio of the reactants; the endproduct was always ThC2. The logarithmic weight loss was directly proportional to the time, and theArrhenius plot showed a break at 1710 K: below this temperature the activation energy was found to be 440 kJ, above 260 kJ. The temperature corresponding to the break coincides with the transition temperature of monoclinic to body-centered tetragonal ThC2.

    Notizen: Abstract In the decomposition of α,α-dimethylbenzyl hydroperoxide (1) by use of copper(II) chlorocomplexes, the novel fact was found that the product ratio of acetophenone (2) to α,α-dimethylbenzyl alcohol (3) is 2 to 1. The kinetics of this result was investigated and a mechanism consistent with the experimental result is proposed.

    Notizen: Abstract The bromination of 15 dinuclear phenolic compounds (dihydroxydiphenylmethanes, methylene bisphenols) by molecular bromine in acetic acid was studied kinetically at 22°C. In all compounds the electrophilic substitution occurred inortho-position to the phenolic hydroxy group of the methyl phenol unit while the non reacting neighboring unit was differently substituted by H, CH3,t-Bu and NO2. A decrease in the reaction rate was observed in 2,2′-dihydroxydiphenylmethanes, where the +M-effect of the hydroxy group is diminished by an intramolecular hydrogen bond. The strength of this hydrogen bond may be influenced mainly by steric factors. Strong electron withdrawing substituents like NO2 show a rate decreasing influence on the reactivity of the neighboring unit also in 2,4′- and 4,4′-dihydroxydiphenylmethanes.

    Notizen: Abstract The kinetics of the oxidation of Mo(CN) 8 4− by IO 4 − has been studied in ethanol—water solvent mixtures over a temperature range of 15–35 °C. The effect of solvent composition on the reaction rate and the mechanism has been investigated. Activation parameters are given. An inner-sphere mechanism, consistent with the kinetic results, is proposed.

Die Reaktionsordnung ist sowohl bezüglich des Chinols als auch des Hg(II)-Ions erster Ordnung. Die Reaktionsgeschwindigkeit erhöht sich leicht mit der HNO3-Konzentration und auch mit abfallender Dielektrizitätskonstante des Mediums. Die Reaktionsgeschwindigkeit sinkt mit dem Zusatz von KNO3. Es ist keinerlei Hinweis auf eine Komplexbildung zwischen Chinol und Hg(II) festzustellen. Die Resultate der Untersuchungen legen HgNO + 3 als aktive Spezies nahe. Es wird ein möglicher Mechanismus mit einem Zweielektronen-Transfer im geschwindigkeitsbestimmenden Schritt vorgeschlagen. Das dabei produziertep-Benzochinon existiert nicht in freier Form, sondern es bildet einen stabilen 1 : 1-Komplex mit Quecksilbernitrat; dieser Komplex wurde mittels TLC und IR charakterisiert.
    Notizen: Abstract The kinetics of oxidation of quinol by mercuric nitrate in presence ofAcOH-H2O-HNO3 mixture has been investigated in order to find the active species of mercuric nitrate involved in the oxidation in this medium. The order of reaction both with respect to quinol and Hg(II) is found to be one. The reaction rate slightly increases with the increase in [HNO3] and the decrease of the dielectric constant of the medium. The reaction rate retards on addition of KNO3. There is no evidence for complex formation between quinol and Hg(II). These results suggest that HgNO + 3 might be the active species in this medium. A probable mechanism involving a two electron transfer in the rate determining step has been suggested. The producedp-benzoquinone does not exist in free state but forms a stable (1 : 1) complex with mercuric nitrate which has been characterized by TLC and IR studies.

    Notizen: Abstract Rate constants and activation parameters for the reductive dimerization of substituted benzylidenemalononitriles were obtained from derivate cyclic voltammetry measurements in dimethylformamide as solvent. Benzylidene-,p-methyl-andp-methoxybenzylidenemalononitrile react exclusively via dimerization of their anion radicals (radical-radical coupling) while forp-fluorobenzylidenemalononitrile 25% of the dimerization proceeds by coupling of the anion radical with the substrate.

    Notizen: Abstract An assessment is made of the relative contribution of certain classes of energetic particle precipitation to the chemical composition of the middle atmosphere with emphasis placed on the production of odd nitrogen and odd hydrogen species and their subsequent role in the catalytic removal of ozone. Galactic cosmic radiation is an important source of odd nitrogen in the lower stratosphere but since the peak energy deposition occurs below the region where catalytic removal of O3 is most effective, it is questionable whether this mechanism is important in the overall terrestrial ozone budget. The precipitation of energetic solar protons can periodically produce dramatic enhancement in upper stratospheric NO. The long residence time of NO in this region of the atmosphere, where catalytic interaction with O3 is also most effective, mandates that this mechanism be included in future modelling of the global distribution of O3. Throughout the mesosphere the precipitation of energetic electrons from the outer radiation belt (60°≲Λ≲70°) can sporadically act as a major local source of odd hydrogen and odd nitrogen leading to observable O3 depletion. Future satellite studies should be directed at simultaneously measuring the precipitation flux and the concomitant atmosphere modification, and these results should be employed to develop more sophisticated models of this important coupling.

    Notizen: Summary The kinetics of precipitation of hydroxyapatite, HA, was studied by seeding dilute supersaturated solutions with well characterized HA crystals. In solutions having initial degrees of supersaturation comparable to those present in human serum, the precipitation rates were related to the thermodynamic driving force (degree of supersaturation with respect to HA) and not to the solution composition. The following relationshipR 0=KA(DS) n , whereR 0=initial precipitation rate, A=amount of seeds, DS=degree of supersaturation, and K andn are parameters obtained from the experimental data, was found to apply over a DS range of 6.6×1010 to 3.3×106. These observations are consistent with the occurrence of a simple growth process on the HA seeds. No evidence for the formation of discrete calcium phosphates other than HA was detected. The Ca/P molar ratio of the precipitating phase, calculated from solution compositions, was invariably higher than that expected for HA; this result is shown to be consistent with an initial adsorption phenomenon. Anomalous kinetic behavior was observed at low seeding levels and may relate to the surface phenomenon described. It is concluded that, most probably, under physiological conditions, formation and remineralization of hard tissues occur through the reported crystal growth process.

    Notizen: Summary Cyanogen bromide (CNBr) peptides were prepared of the insoluble collagen of bovine dental cementum. Following chromatographic separation, the peptides were identified by their amino-acid composition. Type I collagen ([α1(I)]2α2) accounted for more than 90% of the organic matrix, while Type III collagen ([α1(III)]3) was present at a level of approximately 5%. Amino-acid analyses revealed that the CNBr peptides from α1(I) and α2 chains of cementum closely resembled the corresponding peptides from calf skin. The only systematic difference was a higher level of hydroxylation of prolyl and lysyl residues of the cementum peptides.

    Notizen: Summary The calcium homeostatic mechanism was challenged in adult female rats by feeding them a calcium-deficient diet containing oxalate, and by subjecting them to pregnancy and lactation. The regimen caused a substantial weight loss, especially in those animals which reared their young well. Severe hypocalcaemia was observed in the lactating rats. Serum-P was slightly elevated. The content of hydroxyproline in serum was considerably elevated, reflecting the degree of calcium deprivation. Serum proteins were least influenced. The calcium depriving regimen reduced the growth of long bones, but did not stop it. The ash content of the bones was considerably reduced, the degree of reduction depended on the degree of calcium deprivation. Ash as percentage of total bone organ was reduced, but not to the same extent as total ash. Analyses of different parts of femur showed that the proximal and distal parts had lost more bone mineral than the diaphyseal shaft. The ash content of cortical bone tissue from the femur was estimated by a volumetric method. No differences were observed between test groups and controls, indicating that no measurable amounts of bone mineral had been removed from the walls of the vascular canals or by osteocytic osteolysis. Planimetric determinations on cross sections from femora disclosed that a great amount of bone had been removed from the endosteal surface of the diaphysis, while the periosteal surface demonstrated reduced bone apposition.

    Notizen: Summary The bone resorptive factor and amphipathic antigen (AcA) previously identified by us in preparations fromActinomyces viscosus have been partially purified, characterized chemically, and compared. They elute at the same location on chromatography with Ac 22. The fatty acid composition of AcA and the bone resorptive factor is the same. Some differences in carbohydrate composition are observed. TheActinomyces factor does not affect calcium influx or cyclic AMP in isolated bone cells. Therefore it is concluded that AcA stimulates resorption either by gaining entrance into bone cells or by way of a yet undetermined second messenger.

    Notizen: Summary Equilibrium and kinetic experiments were conducted to investigate the factors determining the adsorption of salivary macromolecules onto hydroxyapatite. Using amino acids and other small adsorbates, it was determined that the carboxyl attached to the α carbon does not appear to adsorb onto HA and the affinities of side-chain carboxyls are much smaller than that of the phosphate group (phosphoserine). Hydroxyl (serine) displays an extremely high affinity, but its adsorption site on HA is different and the number of such sites is much smaller than found for the rest of the functional groups investigated. It is shown that the information obtained from small molecules cannot be readily applied to prediction of the adsorption behavior of salivary macromolecules and polypeptides. The kinetics of adsorption of the salivary phosphopeptide statherin, a polyaspartate, and the salivary prolinerich phosphoprotein PRP3 are consistent with the reversibility of the adsorption process; no conclusion was possible in the case of the protein PRP1. Apparent irreversibility cannot be explained on the basis of multipoint binding or the properties of the carboxyl versus phosphate group; it appears that secondary structure determines to a significant extent the adsorption properties of the macromolecules. Calculation of the thermodynamic molar quantities of adsorption of PRP1, PRP3, andl-ASP onto HA showed that the process is entropically driven. The functional relationship between partial molar entropy and adsorption coverage is similar for the two proteins, but quite different from that for aspartate. Explanations for these results are advanced on the bases of changes in structure configurations and displacement of water from the adsorbate and the adsorbent surface, the second factor being the dominant one in the adsorption of a small molecule such asl-ASP.

    Notizen: Abstract Kinetic parameters of the glyoxysomal and mitochondrial malate dehydrogenase (EC 1.1.1.37) of watermelon (Citrullus vulgaris Schrad.) cotyledons were compared. The data were obtained by initial rate experiments at pH 8.5 in both directions of the reaction using homogeneous enzyme preparations. Substrate inhibition at physiologically significant concentrations was observed with reduced nicotinamide adenine dinucleotide (NADH) (50% inhibition at 0.65 mmol·l-1 NADH), but not with oxaloacetate, L-malate or oxidized nicotinamide adenine dinucleotide. The inhibition of both isoenzymes by 5′adenosine monophosphate was studied. Inhibition was found to be competitive with respect to NADH and non-competitive with respect to oxaloacetate. The apparent inhibitor constants at 200 μmol·l-1 of the fixed substrates were 3.2 and 1.6 mmol·1-1 for NADH, and 3.2 and 5.2 mmol·l-1 for oxaloacetate with the glyoxysomal and mitochondrial isoenzymes, respectively. The energy of activation was determined for oxaloacetate reduction by glyoxysomal (E a =3.14×104J×mol-1) and mitochondrial (E a =4.10×104 J x mol-1) MDH from Arrhenius plots, which exhibited constant slopes throughout the range of thermal stability. Despite considerable structural differences, the results indicate very similar kinetic behaviour of the glyoxysomal and mitochondrial isoenzymes. The physiological significance of the data are discussed in relation to the gluconeogenic processes occuring in cotyledons during germination.

    Notizen: Abstract The denaturation of aldolase from rabbit muscle in various solvents leads to significant qualitative and quantitative differences with respect to the structural disintegration of the enzyme. The differences refer to the quaternary structure and to the conformation which is changed only slightly in MgCl2 while in guanidine · HCl or urea at pH ∼ 2 the molecule is close to the state of the random coil. Using the enzymic activity as a quantitative measure for the refolding process, the reaction order and the rate constants of the processes of structure formation (θ i → N*) are found to be identical. This observation suggests a common intermediate D in the process of renaturation after denaturation and dissociation in the different solvent media. D may be considered an intermediate state with a defined number of nucleation centers whose rapid formation is predetermined by the aminoacid sequence. As taken from the first order kinetics in the given range of enzyme concentration, transconformation reactions are rate limiting in the obligatory pathway of refolding. At low enzyme concentrations second order steps gain importance which indicates that the enzymic activity is significantly modified by the formation of the native quaternary structure.

Essential points are: 1) In the scattering experiment, amplitude resolution of 2 · 10−5 (single flash) by application of a differential detector, stability of the same magnitude due to optical compensation, high intensity at the detector due to special optics for the scattered light and semiconductor sources. 2) In the absorption measurement, elimination of scattering contributions by the dual wavelength-method and by high aperture optics. 3) Simultaneous measurement of absorption and scattering. The application of the method is described in using signals from isolated bovine rod outer segments. A reliable procedure is described by the use of which the originally measured light-scattering effects can be split up into single signals. The method allows comparative kinetic analysis of absorption and scattering signals. The possible causal connections between pigment and membrane structure processes can be selected.

    Notizen: Abstract Viroids are an independent class of plant pathogens which are distinguished from viruses by the absence of a protein coat and by their unusually small size. They are single-stranded circular RNAs composed of about 360 nucleotide residues. Sequence analysis and physicochemical studies of the potato spindle tuber viroid (PSTV) have shown that, as a result of intra-molecular base pairing, viroids form a unique rod-like secondary structure which is characterized by a serial arrangement of double-helical sections and internal loops. There is no indication for an additional tertiary structure because all parts of the molecule are freely accessible to ligand interaction. During the denaturation all of the native base pairs of viroids are dissociated in one highly cooperative transition, and in the same process very stable hairpins which are not present in the native structure are newly formed. Most of the properties of the structure and structural transitions of PSTV have been found also in citrus exocortis viroid, chrysanthemum stunt viroid and four different viroid-like RNAs associated with the cadang-cadang disease. The close similarity between these viroids is more expressed in the overall structure and in thermodynamic and functional domains than in the primary sequence. The stiffness of all viroids can be described by an unique persistence length of 300 å. Characteristically, regions of premelting, regions of stable hairpins, and the sequence UACUACCCGGUGG which is opposite to one of the stable hairpins, are the most conservative sequences in the molecules. Current hypotheses about the function of viroids are discussed on the basis of their structural and thermodynamic features. The suggestion that viroid RNA has features similar to DNA has been supported by the finding that they are replicated in vitro by the DNA-dependent RNA polymerase II of the host plant. The highly conserved sequence in viroids mentioned above corresponds very closely to a segment at the 5′-end of the small nuclear RNA U1 of eukaryotes. Because this segment is discussed in recent models, to be involved in the splicing process, a hypothesis is proposed in which viroids interfere with the splicing process leading to a pathogenic misregulation of mRNA processing.

    Notizen: Abstract The kinetics of reduction of horse heart cytochrome c have been investigated using the reductants sodium dithionite and potassium ferrocyanide. Sodium dithionite reduction at pH 7.0 yields rate constants of 2.8×108M−1sec−1 for SO 2 − and 6×105M−1sec−1 for S2O 4 = at infinite dilution. Moreover,the data presented demonstrates the participation of positively charged amino acid side chains at the site of electron transfer. The effect of pH on the reduction of ferricytochrome c requires a minimum of two pK Values for description (pK1=7.0±0.4,pK2=9.3±0.3). Based on the pK values determined, one or more lysines and a residue(s) with a low pK are implicated as the positively charged residues participating in electron transfer. From a comparison of the rates of reduction of various denatured forms of cytochrome c we feel that the most viable conclusion is that electron transfer takes place at the exposed heme edge in the vicinity of the amino acid side chains indicated above. Ferrocyanide reduction of ferri-horse heart cytochrome c takes place in a kinetically complex manner. A mechanism is described which includes complexes of ferrocyanide and ferricytochrome c and ferricyanide and ferrocytochrome c. As was found for dithionite reduction a positively charged region of the cytochrome c participates in electron transfer. Combining our results with ferrocyanide and dithionite we conclude that available data is compatible with a single mechanism of electron transfer. It is suggested that the kinetic distinction between different reductants lies in the lifetime of the transient complex formed, with the order ferrocyanide≫ S2O 4 = 〉SO 2 − .

    Notizen: Abstract The kinetic and molecular properties of cyanobacterial glucose-6-phosphate dehydrogenase, partly purified from Anabaena sp. ATCC 27893, show that it undergoes relatively slow, reversible transitions between different aggregation states which differ in catalytic activity. Sucrose gradient centrifugation and polyacrylamide gel electrophoresis reveal three principal forms, with approximate molecular weights of 120 000 (M 1), 240 000 (M 2) and 345 000 (M 3). The relative catalytic activities are: M 1≪M 2〈M 3. In concentrated solutions of the enzyme, the equilibrium favors the more active, oligomeric forms. Dilution in the absence of effectors shifts the equilibrium in favor of the M 1 form, with a marked diminution of catalytic activity. This transition is prevented by a substrate, glucose-6-phosphate, and also by glutamine. The other substrate, nicotinamide adenine dinucleotide phosphate (NADP+), and (in crude cell-free extracts) ribulose-1,5-diphosphate are negative effectors, which tend to maintain the enzyme in the M 1 form. The equilibrium state between different forms of the enzyme is also strongly dependent on hydrogen ion concentration. Although the optimal pH for catalytic activity is 7.4, dissociation to the hypoactive M 1 form is favored at pH values above 7; a pH of 6.5 is optimal for maintenace of the enzyme in the active state. Reduced nicotamide adenine dinucleotide phosphate (NADPH) and adenosine 5′-triphosphate (ATP), inhibit catalytic activity, but do not significantly affect the equilibrium state. The relevance of these findings to the regulation of enzyme activity in vivo is discussed.

    Notizen: Abstract Acetate uptake by strains of Synechococcus and Aphanocapsa in short experiments required light, and was strongly inhibited by m-dichlorocarbonyl cyanide phenylhydrazone and dichlorophenyl dimethyl urea. Acetate carbon was distributed in amino acids and in the acyl portion of lipids in the same way as during growth experiments when CO2 was available, but the reduced incorporation in the absence of CO2 was primarily into the lipid fraction. An apparent K m for uptake by Synechococcus and for Aphanocapsa 6308 of 20 and 180 μM at pH 7.4 was obtained; corresponding V max values were 6 and 11 nmol x min-1 x mg protein-1. Uptake with Synechococcus was affected by pH, with affinity decreased and maximal rate increase with rising pH. Acetate uptake was not affected by propionate or butyrate when both were added at the same time, but a light and concentration dependent inhibition developed if suspensions were preincubated with propionate. Acetate carbon moved rapidly into acid insoluble material, but after 10–15 s 75% or more of the recovered intracellular counts were in acetyl CoA. Counts in this compound were reduced by preincubation with propionate. Kinetic measurements of acetyl CoA synthetase in fractionated cell extracts gave values for K m of about 50 μM for acetate, 5 mM for propionate, 100 μM for CoA and 0.38 mM for ATP. The internal pool of free CoA was measured to be about 20 μM, and was reduced by preincubation with propionate. This suggests that the activity of CoA-mediated reactions may be regulated by the availability of this cofactor.

    Notizen: Abstract The stability, pH-dependence and kinetic properties of the Mn2+ and FDP-activated NAD-dependent lactic acid dehydrogenases from Lactobacillus casei ssp. casei (ATCC 393) and L. curvatus (DSM) 20010) were studied after the enzymes were purified to homogeneity by affinity chromatography. Both enzymes are virtually unidirectional, catalysing efficiently only the reduction of pyruvate. They are similar with respect to the effector requirement and pH-optimum. They differ, however, in their electrophoretic mobility, heat stability, pH-dependence of the Mn2+ requirement and several kinetic properties. It is suggested that most of these differences are caused by differences of the negative charges in the vicinity of the FDP-binding site or the site responsible for the interaction of the subunits of the enzymatically active oligomeres.

    Notizen: Abstract In order to study phosphate uptake kinetics the cyanobacteriumOscillatoria agardhii was grown in continuous culture under a phosphorus limitation. The affinity of the uptake system reflected in the initial slope of the uptake rate versus external substrate concentration curve (dV/ds) was found to be unaffected by the growth wate. The maximum phosphate uptake rate (V m ) decreased as the growth rate was increased. Attempts were made to relate the decrease ofV m to the increase in phosphorus content of the cells that occurred a higher growth rates. Accumulation of phosphate during pulse experiments indeed resulted in a decrease ofV m . However feedback regulation ofV m by accumulated phosphorus was found to occur only to a small extent in steady state growing cells. The main part of the regulation of the activity of the phosphate uptake system seemingly is determined by a long term process that is, at least longer than 2 h. The presence of short term feedback inhibition by accumulated phosphorus on the activity of the uptake system provides an explanation of the phenomenon thatOscillatoria agardhii is not able to grow at nearμ max growth rates under a phosphorus limitation.

    Notizen: Summary The fat globules of the cream fractions of human and rat milk were stained with ruthenium red. Under the electron microscope, discrete granules and an amorphous coat of lesser density are seen at the surface of the milk fat globules. Since ruthenium red binds anionic groups selectively, it is probable that the granules containe the greatest concentration of these groups. The cream fraction of rat milk contains hexoses, hexosamines, methylpentoses and sialic acid. Methylpentoses and hexosamines are significantly enriched in the cream fraction. It is concluded that the finding of a surface coat in milk fat globules is in keeping with the Bargmann-Knoop model and suggests a distinct mechanism for carrying certain complex carbohydrates in milk. The role of the negative charges at the outer surface of the membrane coat in maintaining fat globules in suspension and in binding certain cations such as calcium is suggested.

    Notizen: Summary The effect of autonomic denervation upon the kinetics of the ileal epithelium of rats was studied by tracing 3H-thymidine labelled nuclei and counting mitotic figures and goblet cells. Counts of labelled nuclei and goblet cells provided information about cell migration along the intestinal epithelium. The mitotic index and turnover time of this population were calculated from the mitotic figures counts. Comparing denervated animals with sham operated controls, it was possible to conclude that autonomic denervation, either sympathectomy or parasympathectomy results in a decrease in mitotic activity. As a result of lower mitotic activity in the crypts, the turnover time increases and there is a delay in the migration of cells from the crypts towards the villi. Low addition levels of high enrichment isotope (〈1% of the total nitrogen pool with 95 to 99 atom percent excess15N) and isotope dilution analysis, were used to establish the kinetics of nitrogen flow. The pools examined were NH4, NO3+NO2, soluble organic-N and forest floor organic matter. This approach allows a more realistic assessment of rates of N movement at the levels of nitrogen concentration encountered in natural systems.

    Notizen: Summary The sensitivity of the mineralization of nitrogen by a range of soils contaminated with heavy metals (up to 340 μg Cd g−1, 7500 μg Pb g−1 and 34000 μg Zn g−1) to the addition of heavy metals in solution were studied using pot incubations (ammonification) and a soil perfusion technique (nitrification). The ammonification of peptone showed little correlation between treatments with Cd, Zn (1000 and 5000 μg g−1) and Pb (10000 and 20000 μg g−1) and origin of the soil. Nitrification was considerably more sensitive to heavy metals than ammonification. All the soils had active, often large, populations of ammonifying and nitrifying organisms which showed substantial similarities between the soils. The rate of nitrifying activity (NO3−N production) was logrithmic in most cases. The presence of tolerant populations of nitrifying organisms in the contaminated soils was demonstrated. Tolerance was also eventually acquired after a longer lag phase, by the non-contaminated soil populations although the rate of activity was often reduced. Metals added in solution were adsorbed by the soil within 4 hours. Differences in toxicity between metal salts (chlorides, sulphates and acetate) were attributed to the amount left in solution. However, in many instances, acetate was found to stimulate all the stages in the mineralisation of nitrogen.

    Notizen: Abstract A method is described for calculating from first principles the coefficients in the Becker-Döring equations for the rate of change of the distribution of cluster sizes in a low-density lattice gas with Kawasaki dynamics. The method depends on solving a diffusion problem for the concentration of particles near a given cluster. The coefficients are calculated for cluster sizes up to 6, on a simple cubic lattice at a temperature 0.59 times the critical temperatures, and extrapolated to larger sizes. The resulting version of the Becker-Doring equations is then solved numerically. Comparison with the results of a computer simulation (at overall concentration 0.075) carried out by Kalos and others indicates that the method gives quite good predictions of the dependence of the cluster distribution on the critical cluster size (usually denoted by l*) but that the predicted rate of change of critical cluster size with time is too small, at this overall concentration, by a factor of about 0.3.

    Notizen: Abstract We make a quantitative comparison between the predictions of the Becker-Döring equations and computer simulations on a model of a quenched binary A-B alloy. The atoms are confined to the vertices of a simple cubic lattice, interact through attractive nearest neighbor interactions, and move by interchanges of nearest neighbor pairs (Kawasaki dynamics). We study in particular the time evolution of the number of clusters of A atoms of each size, at four different concentrations: ρA=0.035, 0.05, 0.075, and 0.1 atoms per lattice site. The temperature is 0.59 times the critical temperature. At this temperature the equilibrium concentration of A atoms in the B-rich phase is ρ A eq =0.0145 atoms/lattice site. The coefficients entering the Becker-Döring equations are obtained by extrapolation from previously published low-density calculations, leaving the time scale as the only adjustable parameter. We find good agreement at the three lower densities. At 10% density the agreement is, as might be expected, less satisfactory but still fairly good-indicating a quite wide range of utility for the Becker-Döring equations.

Poly(L-lysine)-sodium dodecyl sulfate complexes undergo a β-helix transition upon raising the pH of the solution. The transition pH tends to shift downward at elevated temperatures. No helix-β transition can be detected for poly(L-lysine) in sodium dodecyl sulfate solution (pH 〉 11) even after 1-hr heating at 70°C. This is in marked contrast with uncharged poly(L-lysine) solution without sodium dodecyl sulfate, which is converted into the β-form upon mild heating of the solution above 50°C.

    Notizen: A normal coordinate analysis of V-amylose has been performed for an isolated 61 helical chain. Negligible splitting from interactions of vibrations of successive residues is expected between A and E vibrational species due to the large size of the monomer unit. As a result, calculation of only the totally symmetric A modes represents an adequate approximation to the vibrational spectrum of helical polysaccharides. Using this method together with a valence force field we have obtained good agreement between the observed and calculated frequencies. In addition, the computed potential energy distribution and Cartesian displacement coordinates match previous experimental assignments, based on deuterium exchange. The analysis also supports the proposed mechanism for conversion of V-amylose to the more extended B-form. This conversion results in an observed frequency shift for the Raman line at 946 cm-1 which is predicted by the calculations.

    Notizen: The circular dichroism of Ac-(Ala)x-OMe and H-Lys-(Lys)x-OH with x = 1, 2, 3, and 4 has been measured in aqueous solutions. The oligomers with x = 4 show similar circular dichroism spectra in water when the lysyl amino groups are protonated, and they respond in similar fashion to heating and to sodium perchlorate. Both oligomers at 15°C exhibit a positive circular dichroism band at 217-218 nm, which is eliminated by the isothermal addition of 4 M sodium perchlorate or by heating. The positive circular dichroism of the lysine oligomer is also eliminated when the pH is elevated to deprotonate the amino groups. Positive circular dichroism is still observed for Ac-(Ala)4-OMe at elevated pH. Circular dichroism spectra have been estimated for poly(L-alanine) and poly(L-lysine) as statistical coils under the above conditions, based on the trends established with the oligomers. Poly(L-lysine) and poly(L-alanine) are predicted to exhibit similar circular dichroism behavior in aqueous solution so long as the lysyl amino groups are protonated. The circular dichroism of the statistical coil of poly(L-lysine), but not poly(L-alanine), is predicted to change when the pH is elevated sufficiently to deprotonate the lysyl amino groups. These results suggest that the unionized lysyl side chains participate in interactions that are not available to poly(L-alanine). Hydrophobic interactions may occur between the unionized lysyl side chains. Protonation of the lysyl amino groups is proposed to disrupt these interactions, causing poly(L-alanine) and protonated poly(L-lysine) to have similar circular dichroism properties.

    Notizen: Poly-β-benzyl-L-aspartate (poly[Asp(OBzl)]) forms either a lefthanded α-helix, β-sheet, ω-helix, or random coil under appropriate conditions. In this paper the Raman spectra of the above poly[Asp(OBzl)] conformations are compared. The Raman active amide I line shifts from 1663 cm-1 to 1679 cm-1 upon thermal conversion of poly[Asp(OBzl)] from the α-helical to β-sheet conformation while an intense line appearing at 890 cm-1 in the spectrum of the α-helix decreases in intensity. The 890 cm-1 line also displays weak intensity when the polymer is dissolved in chloroform-dichloroacetic acid solution and therefore is converted to the random coil. This line probably arises from a skeletal vibration and is expected to be conformationally sensitive. Similar behavior in the intensity of skeletal vibrations is discussed for other polypeptides undergoing conformational transitions.The Raman spectra of two cross-β-sheet copolypeptides, poly(Ala-Gly) and poly(Ser-Gly), are examined. These sequential polypeptides are model compounds for the crystalline regions of Bombyx mori silk fibroin which forms an extensive β-sheet structure. The amide I, III, and skeletal vibrations appeared in the Raman spectra of these polypeptides at the frequencies and intensities associated with β-sheet homopolypeptides. Since the sequential copolypeptides are intermediate in complexity between the homopolypeptides and the proteins, these results indicate that Raman structure-frequency correlations obtained from homopolypeptide studies can now be applied to protein spectra with greater confidence.The perturbation scheme developed by Krimm and Abe for explaining the frequency splitting of the amide I vibrations in β-sheet polyglycine is applied to poly(L-valine), poly-(Ala-Gly), poly(Ser-Gly), and poly[Asp(OBzl)]. The value of the “unperturbed” frequency, V0, for poly[Asp(OBzl)] was significantly greater than the corresponding values for the other polypeptides. A structural origin for this difference may be displacement of adjacent hydrogen-bonded chains relative to the standard β-sheet conformation.

    Notizen: Mixtures of a weak polybase (polyethylenimine) and a weak polyacid acrylamide-acrylic acid copolymer in aqueous solutions at several ionic strengths and polymer concentrations are studied potentiometrically. When the concentrations of the polyethylenimine and acrylamide-acrylic acid copolymer charges are not too different, phase separation into two liquid phases (“complex coacervation”) is observed. In the pH region where no phase separation occurs, potentiometric titrations are performed on mixtures of both polymers. From the titrations of polyethylenimine solutions, acrylamide-acrylic acid copolymer solutions, and the mixtures, the free energy of interaction has been evaluated according to the theory of Litan. The dependence of the free energy of interaction on pH, polymer concentrations, and ionic strength is explained quantitatively with a model of cooperative electrostatic physical association.

    Notizen: The synthesis and characterization of a series of sequential polypeptides with the repeating sequences Aze-Pro-Aze, Pro-Aze-Pro, Pro-Aze-Gly, Aze-Pro-Gly, Ala-Aze-Gly, Aze-Ala-Gly, and Pro-Pro-Gly are reported. The polymers were prepared by the active ester method, using the p-nitrophenyl, pentachlorophenyl, and N-hydroxysuccinimide esters as the polymerizable tripeptide derivatives. Except for poly(Ala-Aze-Gly) obtained via the N-hydroxysuccinimide ester, all polymers were isolated in good yields and have weight-average molecular weights in the range 10,000-30,000. The molecular weights have been determined by applying the calibrated gel chromatography system described by Fairweather et al. [J. Chromatogr. (1972) 67, 157] and by viscometry. All di- and tripeptide intermediates were chemically and optically pure.

    Notizen: Synthetic regular polytripeptides of the type (Gly-R2-R3) where R2, R3, or both, are imino acids have been widely studied as model compounds for collagen. One such polytripeptide is poly(Gly-Pro-Hyp), since triplets with this sequence constitute about 10% of collagen. Recently, a new model has been proposed for this polytripeptide in which one of the three peptide bonds in the tripeptide unit is in the cis conformation, and the γ-hydroxyl group of hydroxyproline forms a direct interchain hydrogen bond within the triple helix. We have confirmed this structure by model building using computer techniques, and the helical parameters obtained by us are close to the experimentally observed values. The model is also found to be comparable in stability with other models from energy considerations.

    Notizen: Several globular proteins have values of the Scheraga-Mandelkern β parameter significantly below the theoretical minimum value, β0 = 2.112 × 106, for an impermeable sphere. Using the Felderhof-Deutch generalization of the Debye-Bueche-Brinkman theory of hydrodynamics of porous spheres, we have shown that values of β slightly below this supposed minimum are theoretically expected. A porous sphere of uniform density has a minimum β of 2.084 × 106 at a Debye shielding ratio of 6.5, corresponding, for example, to a sphere radius of 11 Å and an inverse hydrodynamic shielding length of 0.6 Å-1, values not far from those of small proteins. A two-layer porous sphere model gives similar results. Although this is the first theoretical explanation of values of β below β0, the theory is incomplete since β values as low as 2.03 × 106 are observed.

    Notizen: NMR measurements of poly(γ-benzyl-L-glutamate) are reported in several different strengths of magnetic field to determine the relaxation time of the helix-coil transition. Nmr spectra of various samples had line shapes varying from the double to single, depending on the extent of the polydispersity of the sample. This result indicated that the correct line shape of a polypeptide is obscured in the overlapping of multipeaks, which are due to the heterogeneity of the molecular weight in the sample. Thus, the conventional line-shape analysis could not be applied to the kinetic study of the helix-coil transition of polypeptides without consideration of this polydispersity effect on the line shape.To overcome this difficulty, we measured linewidths of nmr spectra for fairly monodisperse samples, using various nmr spectrometers, having field strengths from 60 to 220 MHz. The results were analyzed by a quadratic equation, which involves an additional term proportional to the frequency difference of two sites. The equation differs from the conventional quadratic equation, usually utilized in the case of the fast-exchange limit, only in this additional term. This modification is required to evaluate correctly the unusual broadening of the linewidth resulting from the polydispersity effect and to determine the relaxation time reflected in nmr.Nmr spectra of three samples (DP-35, 85, and 250) were measured by 220-, 100-, and 60-MHz spectrometers in trifluoroacetic acid/chloroform at 28°C and linewidths were analyzed. Relaxation times of the helix-coil transition obtained at the transition midpoint are 2.5 × 10-4, 7 × 10-4, and 1.1 × 10-3 sec, for DP-35, 85, and 250, respectively.

The break in the symmetry relation HV = VH is related to the tilt angle between \documentclass{article}\pagestyle{empty}\begin{document}$ \vec \mu _a $\end{document} and \documentclass{article}\pagestyle{empty}\begin{document}$ \vec \mu _e $\end{document}. The theory is applied to a sonicated DNA-2-hydroxy-4,4′-diamidinostilbene complex, in the blue and red emission bands of this peculiar dye. Simultaneous measurements of linear dichroism and fluorescence lead to the determination of an angle of 47° between a fluorescent bound dye and the DNA axis, with no difference for the blue- and red-emitting species, but confirm the presence of nonfluorescent bound dye in a more perpendicular arrangement.

    Notizen: The translational drag, rotational drag, and intrinsic viscosity of spherical multisubunit structures have been calculated analytically using the Felderhof-Deutch theory of polymer frictional properties. The structures considered were hollow shells, spheres with uniform subunit density, and spheres covered with a subunit layer of different density. Changes in the transport coefficients resulting from the random removal of subunits and from the variation of subunit size are calculated. For the case of the shell, the results agree with the numerical computations of Bloomfield, Dalton, and Van Holde [Biopolymers 5, 135, 149 (1967)].

    Notizen: Interactions of glutaraldehyde with either n-butylamine, poly(α,L-lysine), or collagen resulted in a fast release of protons in dilute aqueous solutions at various pH values, followed by much slower changes. The latter reactions, which extended over hours and days, were followed spectrophotometrically and revealed the formation of distinct absorption bands in the visible and near-ultraviolet regions in all the above systems. The visible-range bands disappeared upon treatment with sodium borohydride. A qualitative relationship between oxygen uptake by the system n-butylamine-glutaraldehyde and the slow formation of colored products has been established, while the chemical nature of the reaction products has not been determined.Sedimentation velocity, viscosity, and optical rotation measurements on the products of interaction between poly(L-lysine) and glutaraldehyde in aqueous solution indicated large conformational changes in the polyamino acid present in excess (in residues) over the dialdehyde. In particular, the intrinsic viscosity dropped considerably after interaction, indicating intramolecular crosslinking. At molar ratios of 1:1 between polylsine residues and aldehyde groups, intermolecular crosslinking of polylysine was obtained at pH 8.6.Electron microscopic examinations of collagen samples treated by glutaraldehyde at various pH values indicated changes from unordered to more ordered structures upon treatment with glutaraldehyde, in particular at pH 10.The present structural and optical investigations are considered to be relevant to tanning processes of hides and to fixation procedures.

    Notizen: The template directed synthesis of poly[d(A-T)] from the nucleoside triphosphates in the presence of DNA polymerase I is carried out continuously in a stirred flow reactor for the first time. The initial objective is to test the kinetic stability of the established steady states at various flow rates. Graphical analysis predicts instable steady states for certain high flow rates. As a consequence of instabilities multiple steady states and steady-state hysteresis may occur. Steady-state hysteresis has now been found experimentally. For a different enzyme fraction of low exonuclease activity we found the steady-state absorbance at 260 nm to be almost invariant with flow rate at high enzyme concentrations even if the flow rate was increased by a large factor. We call this phenomenon kinetic buffering. Relaxation of a large flow perturbation approaches the steady state in a sigmoidal fashion. Concentration oscillations at 260 nm occurred in one experiment using an enzyme fraction of low exonuclease activity after perturbing the steady state by monomer (dATP). Advantages of the stirred flow reactor method over serial transfer are discussed.

    Notizen: The far-ultraviolet circular dichroic spectrum of the 39-residue peptide hormone porcine corticotropin and the biologically active fragment corticotropin 1-24 is negative from 250 nm to 195 nm in water, but in 6M guanidinium chloride a positive band appears at about 225 nm. The temperature and guanidinium chloride dependence of this spectral transition indicates the absence of any stable ordered secondary structure in corticotropin and the spectrum is seen to be in only partial agreement with results using the model peptide chromophore, Ala-Ala-Ala. Using oligopeptides containing aromatic amino acid residues sandwiched between glycyl residues, it is shown that the shape and intensity of the corticotropin 225 nm positive band which appears in 6M guanidinium chloride is in agreement with the far-ultraviolet transitions of the aromatic chromophores in the hormone. Curve resolution of the near-ultraviolet circular dichroic spectrum of corticotropin and comparison of the rotational strengths of the phenylalanyl and tyrosyl bands reveals no evidence for increased rotational freedom in 6M guanidinium hydrochloride. Spectral changes are observed, however, in the transitions arising from the single tryptophan. This study suggests that corticotropin in aqueous solution may serve as a better model for the circular dichroic spectrum of the aperiodic regions in globular proteins than either synthetic homopolypeptides or reference proteins for which spectral and X-ray diffraction data are available.

    Notizen: An all-order classical coupled oscillator theory in which monomer band shapes are explicity taken into account is applied to the calculations of polynucleotide circular dichroism. Calculations are shown for ApA and oligoadenylic acid of varying chain lengths, in both RNA and B-DNA geometry, and the advantages of this theory are assessed. By introducing an effective dielectric constant, good agreement with measured spectra is obtained. Variations in monomer parameters are tested in an attempt to eliminate remaining discrepancies between calculated and measured spectra.

    Notizen: New techniques in laser Raman spectroscopy are used to obtain spectra of aqueous solutions of lysozylme for frequency shifts as small as 5 cm-1. In addition, Raman measurements are made on two crystalline forms of hen egg white lysozyme. The spectra obtained from the solution and from the crystal are found to be similar for frequencies above 100 cm-1. However, a low-frequency band at 25 cm-1 observed in crystalline lysozyme is not found in the solution, indicating that this band cannot be attributed to an internal molecular vibration.

    Notizen: The adsorption of protein films on polystyrene latex spheres was studied by optical mixing spectroscopy. With this technique, we show that both the hydrodynamic thickness of protein films and their optical density can be measured. Thus, we found that films of the glycoproteins isolated from the human erythrocyte membrane were four times as thick as films of either human serum albumin or bovine serum albumin for about the same surface coverage. This result suggests an end-on orientation for the adsorbed glycoprotein molecules, which is consistent with the model proposed by others for the orientation of these molecules at the surface of the red blood cell itself.

    Notizen: Solvent binding to bovine serum albumin in 2-chloroethanol-water mixed solvents of different composition, measured previously by Inoue and Timasheff (Biopolymers (1972) 11, 737-43) is applied to a hydrodynamic study of the solvated protein.From sedimentation and diffusion data, the apparent molecular weight of the solvated protein particle can be calculated, provided an average partial specific volume, computed from the composition of the particle, is introduced in Svedberg's equation. The unsolvated molecular weight of the protein can than be calculated by subtraction of the bound solvent. Further data on the hydrodynamic particle (f/fmin and axial ratio of the equivalent ellipsoid) are readily calculated from these experiments, and reinforce the supposition that 2-chloroethanol is a strong helix-inducing solvent.

    Notizen: In order to obtain a molecular picture of the A and B forms of a DNA subunit, potential energy calculations have been made for dGpdC with C(3′)-endo and C(2′)-endo [or C(3′)-exo] sugar puckerings. These are compared with results for GpC. The global minima for dGpdC and GpC are almost identical. They are like A-form duplex DNA and RNA, respectively, with bases anti, the ω′, ω angle pair near 300°, 280°, and sugar pucker C(3′)-endo. For dGpdC, a B-form helical conformer, with sugar pucker C(2′)-endo and ω′ = 257°, ω = 298°, is found only 0.4 kcal/mol above the global minimum. A second low-energy conformation (2.3 kcal/mol) has ω′ = 263°, ω = 158° and ψ near 180°. This has dihedral angles like the original Watson-Crick model of the double helix. In contrast, for GpC, the C(2′)-endo B form is 6.9 kcal/mol above the global minimum. These theoretical results are consistent with experimental studies on DNA and RNA fibers. DNA fibers exist in both A and B forms, while RNA fibers generally assume only the A form. A low-energy conformation unlike the A or B forms was found for both dGpdC and GpC when the sugars were C(3′)-endo. This conformation - ω′,ω near 20°,80° - was not observed for C(2′)-endo dGpdC. Energy surface maps in the ω′,ω plane showed that C(2′)-endo dGpdC has one low-energy valley. It is in the B-form helical region (ω′ ∼ 260°, ω ∼ 300). When the sugar pucker is C(3′)-endo, dGpdC has two low-energy regions: the A-form helical region and the region with the minimum at ω′ = 16°, ω = 85°.

    Notizen: The nmr titration curves of chemical shifts versus pH were observed for the protons of various histidine-containing di- and tripeptides. With these results, the macroscopic pKa values and the chemical shifts intrinsic to each ionic species were determined by a computer curve-fitting based on a simple acid dissociation sequence. The pKa value of the imidazole ring in N-acetyl-L-histidine methylamide was assumed to represent the intrinsic (or unperturbed) pKa of the imidazole rings of histidine having peptide linkages at both the CO and NH sides. The pKa values of the imidazole rings observed for most di- and tripeptides were reasonably reproduced by simple calculations using the intrinsic value and the perturbations due to the CO2- and NH3+ groups located at various positions. Some other factors affecting the pKa value of the imidazole ring are also discussed.

    Notizen: The hyperchromic and hypochromic changes in the intensity of the amide-I and amide-III lines of polypeptides and certain ring vibrations of the bases of polynucleotides are shown to be related to similar changes in the lower energy uv absorption bands. The selection rules strictly limit the pairs of excited electronic states that can contribute to the elements of the polarizability matrix. An energy-dependent term in this equation weights the contribution of the pairs of electronic transitions in favor of those involving the lower energy transitions. For both polypeptides and polynucleotides, there is a large hypochromic inensity change in the first π → π* exciton band upon the coil-to-helix transition. Through the selection rules, certain conformationally sensitive Raman lines are shown to derive their intensity predominantly from this band and hence also display hypochromism. Again, through an application of the selection rules, certain Raman lines can be demonstrated to depend predominantly for their intensity upon the n → π* transition, and consequently have the opposite hyperchromic intensity change upon the same conformational transition.

    Notizen: Transient electric birefringence experiments have been performed on well-characterized highly monodisperse samples of tobacco mosaic virus (TMV). Experiments and calculations are presented that show that the sensitivity and accuracy of the traditional transient electric birefringence technique can be enhanced by several orders of magnitude through the use of a low-powered laser and crystal polarizing optics, signal averaging, and digital data processing. Measurements of the field-free decay of the birefringence yield for the rotational diffusion coefficient, DR = 318 ± 3 sec-1. The length of the rodlike virion, calculated from the Broersma equation with this value of DR and a rod diameter of 15 nm, is 292.5 ± 1.0 nm. Measurements of the growth of the birefringence when an electric field is applied across the sample yield μ2/kT(α∥ - α⊥) = 0.47 ± 0.07, where μ is the permanent dipole moment (assumed parallel to the symmetry axis of the virus) and α∥ - α⊥ is the electrical polarizability anistropy of the virus. Using the value of α∥ - α⊥ measured by O'Konski et al., we then obtain μ = 2.5 × 10-14 esu cm.

    Notizen: The high-resolution nmr spectrum of baker's yeast tRNACUALeu, a recently sequenced non-denaturable tRNA, has been compared with the spectra of the native and denatured conformers of the closely related species tRNAUUGLeu. Because of the presence of many common base pairs in the different tRNA's, it is possible to assign most of the low-field resonances to specific secondary-structure base pairs. A comparison of the observed positions of the various resonances with those predicted by a semiempirical ring-current shift theory shows a root-mean-square deviation of 0.14, 0.11, and 0.12 ppm for tRNAUUGLeu (native), and tRNAUUGLeu (denatured), respectively. These results support the ring-current shift theory currently used to interpret the low-field nmr spectra of the tRNA molecules. Differences between the predicted and observed positions of some resonances provide new evidence for higher order effects such as shifts from second nearest neighbors, anomalous shifts exerted by G·U base pairs, and tertiary-structure effects. A model that was previously proposed for the denatured conformer of tRNAUUGLeu is also supported.

    Notizen: The properties of apomyoglobin were examined in aqueous solutions and various helix- and random-coil-forming solvents by solvent perturbation, optical rotation, circular dichroism, and viscosity measurements. The solvent perturbation data obtained in neutral aqueous solutions suggest 25-40% exposure of the two tryptophyl residues and 50-60% exposure of the three tyrosyls. The estimates of burial of these groups are in the ranges expected for myoglobin based on its X-ray structure. In the helicogenic alcohols, methanol, ethanol, 2-chloroethanol, trifluoroethanol, and 1-propyl alcohol, as well as in acidic solutions, 8 M urea and 6M guanidine hydrochloride, essentially all the tryptophyl and tyrosyl residues are found to be exposed to solvent based on this method. Analysis of the ORD and CD data indicates that in the alcohols the α-helix content of apomyoglobin has in most cases changed from 58-59% to about 80-95%. Analysis of the intrinsic viscosity data based on the equations of Simha and Kirkwood and Auer indicates that the polypeptide chain in these solvents has the dimensions of fully extended α-helical rods, with lengths of 221-251 Å and mean diameters of 12.8-13.6 Å. It is concluded that apomyoglobin in the various alcohols must have an extended but somewhat irregular rodlike structure, having a few bend or irregular sequences between the α-helical segments due largely to the presence of the four proline residues, 37, 88, 100, and 120 in the amino acid sequence.

    Notizen: The fluorescence parameters, lifetime, relative quantum yield, maximum and mean wavelength, half-width, and polarization, of bovine serum albumin (BSA) were measured at 15°C in aqueous solutions containing varying concentrations of different chemical perturbants, glycerol, Cu2+ ions, guanidine hydrochloride, and urea. By considering a quenching mechanism as being either dynamic or static, depending upon whether the quenching is or is not accompanied by a change in the fluorescence lifetime, we were able to correlate the changes produced in the various fluorescence parameters by the different chemical perturbants with changes in macromolecular structure as the concentration of perturbant was gradually increased. The addition of glycerol and of Cu2+ ions indicated that in aqueous BSA both tryptophan residues are below the surface of the macromolecule, out of contact with solvent water, and, as a consequence, they are statically quenched. “Ultra-Pure” guanidine hydrochloride at 2.4 M or more caused a drastic conformation change, which resulted in the emergence of a visible tyrosine peak at 304 nm in the BSA fluorescence spectrum when either 260- or 270-nm excitation was employed. With the same excitation, the enhancement of BSA tyrosine fluorescence by 6-8 M ultra-pure urea produced only a shoulder near 304 nm in the BSA fluorescence spectrum. We have introduced the use of a new relative quantum yield for protein fluorescence, q′, referenced to the quantum yield of unquenched free tryptophan, which eliminates the quenching action of water from the reference.

    Notizen: The molecular theory of the previous paper in this series is extended to determine the effect of divalent metal ions on helix stability relative to coil at fixed ionic strength and nucleotide phosphate concentration. Specification of the state of condensed counterions, as well as their concentration, is essential for the solution of this problem, and it is assumed that they translate freely within a thin cylindrical shell close to the polynucleotide. As a function of divalent counterion concentration mB2+ the relative stability of the helix is highly nonlinear. Although the overall trend is that the helix stability increases with addition of divalent metal ion, there is a narrow concentration range for which it decreases slightly. The behavior of the relative stability as a function of mB2+ is determined by the translational degrees of freedom of the counterions, both univalent and divalent, both condensed and uncondensed. Detailed comparison of the theory with data is not given here, but it is pointed out that the calculated values of the relative stability are consistent with the order of magnitude of the observed effect Mg2+ on the melting temperature.

    Notizen: The stability constant of the complex of tRNA with 50S subunits of ribosomes was compared in ordinary and heavy water. A considerable effect (about fourfold) was observed, showing the importance of hydrogen bonds in this interaction. In addition, the isotope effect of complementary polynucleotide interaction was measured for two examples. In the case of the binary complex of heptainosinic acid oligomers with poly(C) in the presence of 10-3 M MgCl2, the transfer from ordinary to heavy water gave an increase of the stability constant of about 5%. But in the case of a ternary complex of hexaadenylic acid with poly(U) under the same conditions, the stability constant in D2O increased threefold. The isotope effect depends strongly on magnesium ion concentration and is possibly due to some specific mechanism of magnesium ion complexing involving water molecules.

    Notizen: The storage and loss shear moduli, G′ and G″, have been measured for dilute solutions of unaggregated and aggregated tobacco mosaic virus samples in glycerol-water mixtures, by the Birnboim-Schrag multiple-lumped resonator modified for use with aqueous solvents. The frequency range was 100-5800 Hz, the concentration range 0.6-2.1 × 10-3 g/ml, and the temperatures 25.0° and 37.8°C. The number-average and weight-average molecular weights of the aggregated sample were estimated as 1.4 and 2.0 × 108, respectively, from electron microscopy. The extrapolated intrinsic moduli [G′] and [G″] were compared with the predictions of the Kirkwood-Auer theory for rigid rodlike molecules. For the unaggregated sample, the frequency dependence of [G′] and [G″] agreed well with the theory assuming the intrinsic viscosity to be 27 ml/g, though the asymptotic limit of [G′]M/RT at higher frequencies was slightly larger than the theoretical value of 3/5. For the aggregated sample, the data agreed with theory for rigid rods as modified to account for molecular-weight distribution.
