Notizen: Abstract Feeding behaviour of five species of cereal aphids in wheat seedlings differing in hydroxamic acid (Hx) levels, was monitored via electrical penetration graphs (EPG). Aphid species could be grouped as sensitive to the feeding deterrent effect of Hx in the seedlings (Rhopalosiphum padi, Schizaphis graminum, Sitobion avenae, andMetopolophium dirhodum) or insensitive to them (Rhopalosiphum maidis). However, when feeding behaviour was studied in artificial diets containing Hx, all species were equally sensitive to Hx. The behavour ofR. maidis was further compared with that ofR. padi through detailed EPG analysis. It was found that the insensitivity ofR. maidis to Hx in seedlings may be due to a feeding strategy avoiding contact with the compounds by decreasing the number of cellular punctures in live tissues other than sieve elements during its way to the phloem.

Notizen: Abstract Hessian flyMayetiola destructor (Say) larvae are able to obtain food from their host plant without inflicting mechanical damage to the plant surface, apparently by secreting substances which elicit release of nutrients from plant cells surrounding the feeding site. Cells of fully susceptible plants retain their normal appearances, while in resistant plants extensive areas of cellular collapse occur. These responses indicate that hypersensitivity is the basis of wheat's resistance to the Hessian fly. The fly's feeding mechanism more closely resembles that of a pathogen than of a phytophagous insect; correspondingly, both the genetic relationship and resistance mechanism of the host plant to the parasite are of the sorts commonly associated with bacterial and fungal pathogens.

Notizen: Abstract The Russian wheat aphid, Diuraphis noxia (Mordvilko), is one of the most important aphid pests of wheat, Triticum aestivum L., worldwide. Among the various pest management options, plant resistance is an economical management tactic to control D. noxia in cereal crops such as wheat. Researchers have identified D. noxia resistant germplasm and it has been incorporated into wheat. This study compared D. noxia resistance between the ‘Betta’ wheat isolines Betta-Dn1, Betta-Dn2, and Betta-Dn5 and their corresponding donor gene plant introduction (PI) lines PI 137739 (Dn1), PI 262660 (Dn2), and PI 294994 (Dn5). Although the Betta isolines and PI lines showed D. noxia resistance when compared with Betta wheat, the degree of resistance in the isolines to D. noxia was different from their corresponding PI donors. Aphid number, aphid fecundity, and biomass per aphid were not different between Betta-Dn1 and PI 137739 or Betta-Dn2 and PI 262660; however, the same parameters were significantly lower on PI 294994 compared with Betta-Dn5. This indicated that aphid resistance in PI 137739 and PI 262660 was probably governed by a single dominant gene, while the resistance in PI 294994 was controlled by more than one gene. Additionally, plant biomass reduction was aphid density dependent, which suggested that use of appropriate aphid infestation level is important when using plant biomass reduction as an indicator of resistance. Plant resistance categorization showed that there was no detectable difference in antixenosis among the seven lines evaluated. However, the higher aphid fecundity observed on PI 262660 compared with PI 137739 and PI 294994, in addition to no significant differences among the three PIs in plant biomass reduction, suggested PI 262660 was a tolerant line, while PI 137739 and PI 294994 were antibiotic lines. Plant tolerance could not be elucidated among the three Betta isolines using aphid fecundity and plant biomass reduction as indicators.

Notizen: Abstract The effect of cereal leaf surface wax on Diuraphis noxia (Mordvilko), the Russian wheat aphid, probing behavior and nymphoposition was evaluated. Ultrastructure of leaf epicuticular wax from wheat (Triticum aestivum L.) c.v. ‘Arapahoe’ and ‘Halt’ was different from barley (Hordeum vulgare L.) c.v. ‘Morex’, and oat (Avena sativa L.) c.v. ‘Border’. Both wheat cultivars had similar rod-shaped epicuticular wax, while barley and oat plants had flakes. The chemical composition comparison of gas chromatograms also indicated that the extract of the two wheat cultivars had similar pattern of peaks, while the barley and oat leaves had similar peaks. Cereal variety significantly affected aphid probing behavior (P 〈 0.05), but wax removal using ethyl ether swab did not (P 〈 0.05). Aphids initiated significantly more probes on Border oat leaves than on Morex barley irrespective of wax removal, although total probing duration per aphid was not significantly different among the four cereals examined. Accumulative salivation duration per aphid on oat leaves with wax was significantly longer than other cereal leaves with wax, while accumulative ingestion duration per aphid on Arapahoe wheat and Morex barley was significantly longer than on oat. Nymphoposition of D. noxia on cereal leaves maintained on the benzimidazole-agar medium showed that aphids produced a greater number of nymphs on Morex barley and less on Border oat leaves, although wax removal did not affect aphid nymphoposition. Removal of leaf epicuticular waxes from the 4 cereal genotypes using ethyl ether swab indicated that the influence of wax on plant resistance to D. noxia probing and reproduction was limited. Morex barley was the most favorable, while Border oat was the least favorable cereal host of D. noxia.

Notizen: Abstract Extended sieve element salivation (E1 waveform in the electrical penetration graph) is a characteristic activity during early sieve element punctures, particularly in resistant plants. In order to explore a chemically-mediated mechanism of resistance associated with sieve element salivation, we compared the pattern of feeding behaviour of the aphid, Sitobion fragariae (Walker), on two cultivars of the wheat Triticum aestivum L., with different concentrations of hydroxamic acids (Hx). During 24 h of electronic monitoring, aphids dedicated over 50% of the total time to phloem ingestion from the sieve elements. Total time allocated to E1 in the experiment, time to first E1 within the experiment, time allocated to E1 before a sustained phloem ingestion (E2) and the contribution of sieve element salivation to the phloem phase (E1/[E1+E2]) were significantly higher in the high-Hx cultivar. The increased salivation in plants with higher contents of Hx suggests the existence, at least in this system, of a chemically-mediated sieve element constraint.

Notizen: Abstract The loss of biodiversity — the tendency of the world's fauna to become more and more homogeneous —is widely acknowledged as a problem. Biodiversity is threatened by neozoism and acculturation as much as the extinction of species, but these dangers are less well known. Neozoism and acculturation have an effect on two levels; they lead 1) to an equalisation of the faunal regions and their originally different, specially adapted biocenoses, and 2) to a reduction of genetic diversity below the species level. These processes develop very rapidly in the European waters, particularly in the lowland river systems. The present contribution gives a brief review on neozoans in European inland waters.

Notizen: Abstract Wheat (cv Chinese Spring) tissues were transformed using Agrobacterium tumefasciens and a new plasmid modular vector, pMVTBP. We constructed pMVTBP with unique restriction sites connecting (1) the CaMV 35S promoter, (2) a Kozak sequence, (3) the FLAG epitope, (4) the (His)6 epitope, (5) a coding region (for wheat TATA Binding Protein, wTBP) and (6) the CaMV 35S 3′UTR. This vector thus allows easy exchange of different regulatory or coding sequences. Explants of either germinating mature seeds, or immature embryos, were induced to callus for up to two weeks, treated with virulence-induced bacteria for one hour, then regenerated into plantlets. Transient expression of a GUS reporter gene, assayed at about one week, occurred in 10–12% of calluses. Expression of the FLAG-tagged wTBP was also detected, by immunostaining. Stable expression, by selective growth on geneticin, and by GUS expression at about six weeks, occurred in 1–2% of calluses, quite comparable to that achieved by other methods.

Notizen: Abstract A modern diatom-pH calibration data-set consisting of surface-sediment diatom assemblages from 118 lakes and 530 taxa is presented. The AL:PE data-set is from high-altitude or high-latitude lakes in the Alps, Norway, Svalbard, Kola Peninsula, UK, Slovenia, Slovakia, Poland, Portugal, and Spain (pH range = 4.5-8.0; DOC range = 0.2-3.2 mg l-1). In addition, 92 epilithon samples from 22 high-altitude or high-latitude lakes comprise an AL:PE epilithon diatom-pH data-set. Weighted averaging partial least squares regression is used to develop pH-inference models. The AL:PE data-set has a root-mean-square-error of prediction (RMSEP) of 0.33 and a maximum bias of 0.36 pH units and r2 of 0.82, as assessed by leave-one-out cross-validation. The epilithon data-set has, after data-screening and the deletion of one very obvious outlier, a RMSEP of 0.23 and a maximum bias of 0.18 pH units and r2 of 0.88. The 167 sample SWAP diatom-pH data-set from lowland or upland lakes in the UK, Norway, and Sweden has a RMSEP of 0.29 and a maximum bias of 0.23 pH units and r2 of 0.86. The pH optima, as estimated by weighted averaging and Gaussian regression, are compared for the three data-sets (AL:PE, SWAP, AL:PE epilithon). There is a good correspondence between the AL:PE and the AL:PE epilithon optima, but a consistent bias between the AL:PE and SWAP optima, with the SWAP optima being lower than the AL:PE estimates. The predictive performances of the AL:PE and SWAP calibration data-sets are compared using independent test samples and six core sequences, all from high-altitude lakes, one in south-east Siberia and five in eastern Scotland. The results show the importance of using the AL:PE data-set for inferring lake-water pH from diatom assemblages in high-altitude or high latitude lakes with low DOC concentrations.

Notizen: Abstract Ostracods have only recently been recognized as a valuable group for paleolimnology and as an essential instrument for investigations concerning climatic change. With the increasing knowledge of temperature and oxygen demands of species, their use in the study of shallow lakes with respect to climate and lake level changes and the onset of meromictic conditions has become increasingly important. Central European examples are described. They include a former extension of Neusiedlersee to the SE, most likely a precursor of the present lake, and several Carinthian lakes, including Längsee, and Lobsigensee in Switzerland, NW of its capital Bern.

Notizen: Abstract Samples of wheat (n = 25) and maize (n = 30) for animal consumption, collected in 1997 after harvest from western Romania, were analyzed by enzyme immunoassays for mycotoxin contamination. Toxins analyses included deoxynivalenol (DON), 3-acetylDON, 15- acetylDON, fusarenone X (FX), T-2 Toxin (T-2), diacetoxyscirpenol (DAS), zearalenone (ZEA), fumonisin B1 (FB1), aflatoxin B1 (AFB1), ochratoxin A (OA), and citrinin (CT). DON and acetylDONs were the major contaminants in wheat (100%) and maize (46%). Median values for DON, 3-acetylDON, and 15-acetylDON were 880 μg kg-1, 66 μg kg- 1, and 150 μg kg-1 in wheat, and 890 μg kg-1, 180 μg kg-1, and 620 μg kg- 1 in maize, respectively. Additionally, 3,15-diacetylDON was detected in some samples by HPLC-EIA analysis. All samples were negative for FX (〈150 μg kg-1). T-2 was found in wheat (n = 6) and maize (n = 1) at levels between 13 and 63 μg kg- 1. DAS (2.6 μg kg-1) was found in one maize sample. ZEA occurred in all wheat and in four maize samples, median values were 10 μg kg-1 and 250 μg kg-1, respectively. One maize sample contained FB1 (140 μg kg-1). All samples were AFB1-negative (〈4 μg kg-1). OA was found in one wheat sample (37 μg kg- 1), CT was found in one maize sample (580 μg kg- 1). This first reported natural occurrence of a range of mycotoxins in Romanian feeding stuff shows that DON and acetyl DONs may be present at levels which may affect animal production.

Notizen: Abstract Wheat for human consumption (140 samples) was collected after harvest from all regions of Bulgaria. The 1995 crop year was characterized by heavy rainfall in the spring and summer months. The internal mycoflora of wheat samples was dominated by Fusarium spp. and Alternaria spp., and storage fungi were rarely present. The samples were analysed for contamination with Fusarium mycotoxins deoxynivalenol (DON), 3-acetyldeoxynivalenol (3-AcDON), 15-acetyldeoxynivalenol (15-AcDON), T-2 Toxin (T-2), diacetoxyscirpenol (DAS), and zearalenone (ZEA), using enzyme immunoassay methods. DON and ZEA were the predominant toxins, with a contamination frequency of 67% and 69%, respectively. The average levels of these toxins in positive samples were 180 μg/kg (DON) and 17 μg/kg (ZEA), maximum concentrations were 1800 μg kg−1 and 120 μg kg−1, respectively. Acetyl derivatives of DON, namely 3-AcDON and 15-AcDON, were found in 2.1 % and 0.7% of the samples, at at maximum level of about 100 μg kg−1. Only one sample was positive for T-2 (55 μg/kg), DAS was not detected. This is the first report about the natural occurrence of a range of Fusarium mycotoxins in wheat for human consumption in Bulgaria.

Notizen: Abstract A technique for the application of the15N isotope dilution technique for the quantification of plant associated biological nitrogen fixation (BNF) was tested and applied to quantify the BNF contribution to two genotypes ofPhaseolus vulgaris. The technique makes use of sequential measurements of the15N enrichment of soil mineral N, and the uptake of labelled N by the “N2-fixing” plant, to simulate its uptake of soil N (the “soil to plant simulation” technique). The test was made with two non-N2-fixing crops (non-nodulating beans and wheat) and two bean genotypes (PR 923450 and Puebla 152), at two levels of N fertilizer addition (10 and 40 kg N ha−1), to compare the actual N uptake with that simulated from the soil and crop15N data. The simulation of the soil N uptake by the non-nod bean crop using this “soil to plant simulation” technique underestimated by 20 to 30% the true N uptake, suggesting that the mineral N extracted from soil samples taken from the 0–15cm layer had a higher15N enrichment than that N sampled by the roots of this crop. In the case of the wheat crop the simulation resulted in a much greater underestimation of actual N uptake. In general the results using this technique suggested that BNF inputs to the bean cultivars was higher than would be expected from the nodulation and acetylene reduction data, except for the early PR beans in the 40 kg N ha−1 treatment. In this case the total N and simulated soil N accumulation were well matched suggesting no BNF inputs. An allied technique (the “plant to plant simulation technique”) was proposed where the15N enrichrnent of soil mineral N was simulated from the data for total N and labelled N accumulation taken from sequential harvests of either of the non-N2 -fixing control crops. This was then utilized in combination with the labelled N uptake data of the other crop to simulate its soil N uptake. However, the results using either technique indicated that the wheat and non-nod or nodulating beans exploited pools of N in the soil with completely different15N enrichments probably due to differences in exploitation of the soil N with depth.

Notizen: Abstract Data was assembled from experiments on the fate of15N-labelled fertilizer applied to wheat (Triticum spp.) grown in different parts of the world. These data were then ranked according to the annual precipitation-evaporation quotient for each experimental location calculated from the average long-term values of precipitation and potential evaporation. Percentage recovery of15N fertilizer in crop and soil varied with location in accordance with the precipitation-evaporation quotient. In humid environments more15N fertilizer was recovered in the crop than in the soil, while in dry environments more15N fertilizer was recovered in the soil than in the crop. Irrespective of climatic differences between locations 20% (on average) of the15N fertilizer applied to wheat crops was unaccounted for at harvest. Most of the15N fertilizer remaining in the soil was found in the 0–30 cm layer. The most likely explanation of these differences is that wheat grown in dry environments has a greater root:shoot ratio than wheat grown in humid environments and, further, that the residue of dryland crops have higher C/N ratios. Both factors could contribute to the greater recovery of15N fertilizer in the soil in dry environments than in humid ones.

Notizen: Abstract Long-term field experiments play an important role in understanding the complex interactions of plants, soils, climate and management and their effects on sustainable crop production. A long-term fertilizer experiment with maize-wheat-cowpea (fodder) is in progress since 1971 at Punjab Agricultural University farm Ludhiana, India. The experimental result for the first 21 years showed that application of N alone or in combination with P did not produce as much maize and wheat grains as the application of N, P and K together. Eight years after the start of the experiment, the optimal levels of N, P and K application (100% NPK) were unable to sustain the similar (maize) yield level as before because of Zn deficiency. Whereas in FYM amended plots the Zn deficiency did not appear and the higher crop yields could be sustained. The chemical control of weeds could not sustain the maize productivity at the same level as the manual removal of weeds. It was concluded that the high level of crop production can be sustained with the application of N, P and K under intensive cropping system provided deficiency of any of the micronutrient does not crop up. The deficiency of Zn is most likely to occur in semi-arid light textured alluvial soils under intensive cropping without the addition of farm yard manure/organic manures. In maize based cropping systems, manual control of weeds may be preferred to the chemical one. Addition of FYM in conjunction with 100% NPK is most beneficial both from bio-physical and economic point of view.

Notizen: Abstract In land use plans, fertilizer recommendations are indispensable to avoid soil nutrient depletion or soil water pollution. Nutrient relations of five cereals have been evaluated on the basis of a literature review with the aim of arriving at such fertilizer recommendations at regional level. Nutrients considered were nitrogen, phosphorus and potassium for millet, sorghum, maize, rice and wheat. The relevant nutrient relations are fertilizer nutrient application to nutrient uptake, and nutrient uptake to crop yield. In addition, post-anthesis nutrient uptake is considered. Subsequently, obtained results are used in simulation modelling exercises to calculate the time required to attain an equilibrium nutrient balance and to investigate the effect of erosion control and straw recycling. Although fertilizer requirements could be assessed for each of the five cereals, monitoring of nutrient supply from natural sources remains necessary. Moreover, research on fertilizer use should focus on improvement of fertilizer recoveries and multiperiod models for both N and P uptakes by crops to allow quantitative land use planning where the time scale is included.

Notizen: Abstract Winter wheat crops were grown with ostensibly adequate supplies of all soil nutrients in 1990 and 1991 with the aim of testing if late foliar supplements of K and N, applied at key development stages, could improve grain yield and grain N content. Foliar sprays of KNO3 solution, supplying up to 40 kg K ha−1 in total, at flag leaf unfolded, inflorescence completed and the watery-ripe stage of grain filling, had no effect on yield, yield components or grain N. Urea, supplying 40 kg N ha−1 at flag leaf unfolded, had no effects on grain yield and grain N in 1990, but in 1991 grain N was increased by 0.14% whilst yield was reduced by up to 0.6 t ha−1. Urea scorched flag leaf tips in both years. In 1990, the spring was very dry and foliar supplements might have been expected to have had an effect, but on this highly fertile soil all crop K and N requirements were met from the soil.

Five out of the seven extractants were found to be promising for the estimation of critical level of available Mn in these soils, as the amount of Mn extracted by these extractants was positively and significantly correlated with relative grain yield as well as Mn uptake. The critical deficiency level of soil available Mn with 0.005 M DTPA, 0.02% hydroquinone, 0.02 N sodium pyrophosphate, 0.1N H3PO4 and 0.05N HCl+0.025N H2SO4 was 3.1, 13.8, 23.5, 5.3 and 17.8 mg kg-1 soil, respectively. The 1N ammonium acetate and 0.01M CaCl2 were found to be unsuitable extractants for these soils. Further field trials at eight locations with varying levels of Mn deficiency showed successive increase in the grain yield of wheat with foliar Mn application, emphasizing the need for Mn fertilization when wheat is grown on Mn deficient soils.

Notizen: Abstract A multisite field experiment was conducted to study the effect of topdressed Se-enriched Ca(NO3)2 (CN) and basal applied NPK on the selenium (Se) concentration in spring wheat (Triticum aestivum L.). Selenium was applied either through CN (at the rates of 0, 6.45, and 12.91 g Se ha−1) or NPK (5.83 g Se ha−1). Selenium concentration in wheat grains increased consistently with increasing rate of Se-enriched CN or NPK. However, the superiority of Se-enriched CN over NPK in raising the Se concentration in wheat grain depended on location and growth conditions. At the same rate both methods of Se-application were found to be equally effective in raising the Se concentration of wheat grains. The Se concentration of grain was generally higher in the light textured soils than in the medium to heavy textured soils. Without Se application, the Se-concentration in wheat grain was about 16µg kg−1 which is regarded insufficient to meet the Se requirement for Se in animal and human. Calcium nitrate enriched with 25 mg Se kg−1 (6.45 g Se ha−1) increased the Se concentration in wheat grain to a desired level.

Notizen: Abstract The bioluminescently marked Pseudomonas fluorescens strain 5RL, has been used previously to follow colonisation of soy bean roots (De Weger et al. [1991] Appl. Environ. Microbiol. 57:36-41). In the present paper the method has been further developed and optimized for wheat roots and it is used to get a quick overview of the colonisation patterns of many different root systems at the same time. Colonisation was followed on wheat plants grown in our gnotobiotic sand system (Simons et al., 1996. Mol Plant Microbe Interact 9: 600–607) and the following results were obtained. (i) A spatio-temporal analysis of the colonisation of wheat roots showed that 4 days after planting the highest bacterial activity was observed at the upper part of the root. After 6 days the high bacterial activity at the upper part was further increased, whereas spot-like activities were observed on the lower root parts, possibly due to micro-colonies. (ii) Bacterial mutations causing lack of motility or auxotrophy for amino acids resulted in impaired colonisation of the lower root parts, indicating that motility and prototrophy for the involved amino acid(s) are important factors for wheat root colonisation by strain 5RL. (iii) Coinoculation of strain 5RL with other wild type Pseudomonas strains on the root influenced the colonisation pattern observed for strain 5RL. Colonisation was not visually affected when the competing strain was a poor root coloniser, but was severely reduced when the competing strain was a good root coloniser. The results show that the spatio-temporal colonisation of wheat root by P. fluorescens strain 5RL and derivatives is similar to that of strain WCS365 on tomato. The advantage of the use of lux-marked strains is that the results are obtained much quicker than when conventional methods are used and that the result is supplied as an image of the colonisation pattern of many different roots.

Notizen: Abstract The influence of prey density, within-field vegetation, and the composition and patchiness of the surrounding landscape on the abundance of insect predators of cereal aphids was studied in wheat fields in eastern South Dakota, USA. Cereal aphids, aphid predators, and within-field vegetation were sampled in 104 fields over a three year period (1988–1990). The composition and patchiness of the landscape surrounding each field were determined from high altitude aerial photographs. Five landscape variables, aggregated at three spatial scales ranging from 2.6 km2 to 581 km2, were measured from aerial photographs. Regression models incorporating within-field and landscape variables accounted for 27–49% of the variance in aphid predator abundance in wheat fields. Aphid predator species richness and species diversity were also related to within-field and landscape variables. Some predators were strongly influenced by variability in the composition and patchiness of the landscape surrounding a field at a particular spatial scale while others responded to variability at all scales. Overall, predator abundance, species richness, and species diversity increased with increasing vegetational diversity in wheat fields and with increasing amounts of non-cultivated lands and increasing patchiness in the surrounding landscape.

Notizen: Abstract Management of stored-grain insect pests by farmers or elevator managers should be based upon a knowledge of the grain storage environment and the ecology of insect pests. Grain storage facilities and practices, geographical location, government policies, and marketing demands for grain quality are discussed as factors influencing stored-grain insect pest management decisions in the United States. Typical practices include a small number of grain samples designed to provide grain quality information for segregation, blending and marketing. This low sampling rate results in subjective evaluation and inconsistent penalties for insect-related quality factors. Information on the efficacy of insect pest management practices in the United States, mainly for farm-stored wheat, is discussed, and stored-grain integrated pest management (IPM) is compared to field-crop IPM. The transition from traditional stored-grain insect pest control to IPM will require greater emphasis on sampling to estimate insect densities, the development of sound economic thresholds and decision-making strategies, more selective use of pesticides, and greater use of nonchemical methods such as aeration. New developments in insect monitoring, predictive computer models, grain cooling by aeration, biological control, and fumigation are reviewed, their potential for improving insect pest management is discussed, and future research needs are examined.

Notizen: Abstract Diaeretiella rapae (M'Intosh) (Hymenoptera: Aphidiidae) is a parasitoid of several aphid species, including the Russian wheat aphid (RWA),Diuraphis noxia (Mordvilko), and the cabbage aphid (CA).Brevicoryne brassicae (L.). The response of matedD. rapae females to odors from wheat, cabbage, and plant-host complexes was investigated using a four-choice olfactometer. Experienced parasitoids, but not inexperienced females, responded positively to odors of the wheat-RWA complex in a no-choice test. In choice tests, experienced parasitoids did not respond to odors of uninfested cabbage and wheat leaves, but did respond positively to aphid-infested plants and to aphids alone. The response ofD. rapae to the cabbage-CA complex and to CA alone was significantly greater than to the wheat-RWA complex and RWA alone, suggesting an innate odor preference for crucifer-feeding aphids.

Notizen: Abstract Hydroxamic acids (Hx) are natural products of Gramineae that are associated with cereal resistance to pests. We aimed at characterizing the induction of Hx accumulation in seedlings of wheat,Triticum aestivum, by short-term infestation of the cereal aphid,Rhopalosiphum padi. A load of 25 aphids increased significantly the Hx levels in the infested primary leaf in comparison with control levels. Lower loads did not increase Hx concentration. Aphid infestation lasting 16 hr did not elicit induction of Hx, even after a time-lag of 32 hr to allow the expression of any induced response. Forty-eight hours was the minimum duration of aphid infestation required to trigger Hx induction. The age of the infested tissue (the primary leaf) did not affect induction. Similar increases of Hx were found in unfolding, expanding, and totally expanded primary leaves. It was determined that the regime of nutrient supply (N-intensive nutritive solutions at low and high concentration) to wheat seedlings had no effect on the magnitude of the aphid-induced Hx (N-based secondary metabolites). Results obtained are discussed in the framework of general theories of plant defense allocation.

Notizen: Abstract ICP Forests was launched in 1985 under the Working Group on Effects of the Convention on Long-range Transboundary Air Pollution of UN/ECE. As part of the Programme, forest condition is surveyed annually on up to 33,600 sample plots with about 622,000 sample trees in 35 countries, representing about 225 million hectares of forest. Results of the national surveys are reported as country related mean values, split into tree species and age groups. A special transnational survey is performed in cooperation with the European Commission, based on a uniform 16 × 16 km grid of 4,800 plots with 103,000 trees in 27 countries. Results show that large scale forest decline has been less dramatic than was suggested in the early 1980s. Nevertheless, a general worsening of forest condition can be shown in many parts of Europe. In some areas, particularly in Central and Eastern Europe, several thousand hectares of forest have died. Whilst the most important probable causes reported for the observed forest damage are adverse weather conditions, insects and fungi, most countries consider air pollution as a threatening to forest health.

Notizen: Abstract Open-top chambers ventilated with ambient or chiarcoal-filtered air were used to assess the impact of air pollution on the yield of local cultivars of wheat and rice, at a site on the outskirts of Lahore. At this location, 6-h mean O3 concentrations reach 60 ppb in certain months, and annual mean NO2 concentrations are 20–25 ppb. The experiments showed significant yield reduction in two successive seasons which ranged from 33% to 46% in wheat and from 37% to 51% in rice. The major yield parameter affected was the number of ears or panicles per plant, although there was also evidence of small effects on 1000 grain weight and on the number of grains per ear/panicle. These results have significance in terms of the maintenance of agricultural yields as pollution emissions rise in south and south-east Asia.

Notizen: Abstract In closed-chamber fumigation experiments dry matter partitioning and chlorophyll fluorescence of wheat were studied, analysing the effects of ozone during different stages of plant development. Ozone causes enhanced leaf senescence, leading to a loss of green leaf area and, consequently to a decreased supply of assimilates, affecting (in increasing order of severeness) stem, ear and grain productivity because of reduced storage pools for translocation. Leaves of plants before shooting stage were most sensitive but the lack of green leaf area after ear emergence had the most pronounced effects on grain yield. Measurements of photochemical capacity showed that evidence for negative ozone effects could be found in changes of chlorophyll fluorescence parameters in leaf sections not yet showing visible ozone injury. Negative effects on photosynthesis were more distinct with increasing accumulated ozone dose, with increasing age of leaf tissue and with increasing ozone sensitivity of the cultivar. The changes in chlorophyll fluorescence are most likely to be explained by a decreased pool size of plastoquinones caused by ozone.

Notizen: Abstract In Britain wheat is an important crop accounting for 41% of the total cereal production. In this study ozone concentrations for 1989 estimated as described in Part 1 of the paper are integrated with the estimated wheat distribution to derive a detailed estimate of the impact of ozone on wheat yields at a fine spatial scale (1km × 1km). These data provide estimates for calculating regional and national yield losses. The methodology can be applied to other crop species. Recent research on a range of crops has established relationships between the economic yield loss for certain crops, including wheat, and ozone exposure. Exposure is described as the accumulated exposure above a threshold experienced during the daylight hours (AOT). Critical AOT values are derived from yield exposure relationships which show linear reductions of yield loss with increasing ozone concentrations. This study has made use of land cover data from remotely sensed imagery at 25m resolution and nationally collected agricultural statistics for counties. These data were combined using an areal interpolation technique to provide more spatially articulate estimates of the location and intensity of wheat production. The results demonstrate the economic importance of ozone as a pollutant. Wheat yield losses attributed to ozone vary between different parts of the country but, for years when ozone levels are high, yield losses are likely to be significant in some areas.

Notizen: Abstract A wheat cDNA encoding a glycine-rich RNA-binding protein, whGRP-1, was isolated. WhGRP-1 contains two conserved domains, the RNA-binding motif (RNP motif) combined with a series of glycine-rich imperfect repeats, characteristic of a conserved family of plant RNA-binding proteins. Northern analysis revealed that whGRP-1 mRNA accumulates to high levels in roots and to lower levels in leaves of wheat seedlings. whGRP-1 mRNA accumulation is not enhanced by exogenous abscisic acid in seedlings and accumulates to very high levels during wheat embryo development, showing a pattern different from that of the ABA-inducible wheat Em gene.

Notizen: Abstract Isolation of cDNAs encoding individual members of a gene family is essential for assessing their role in a biological phenomenon. However, this process is often laborious and slow due to highly conserved protein-coding region that interferes with the isolation of the individual members. Identification of gene-specific probes from 3′ non-coding regions of different members can assist in the fast retrieval and characterization of individual members of a multigene family. We used the recent technique of differential display for the same purpose. As an example of a multigene family in plants, we selected a heat shock protein gene family, HSP16.9 from wheat, with estimated 12 members. We modified the original differential display technique for selective amplification of the 3′ non-coding regions of different wheat HSP16.9 genes by replacing the random 10-mer in the original method with a conserved HSP16.9 gene family-specific primer. Sixteen cDNA fragments from these experiments were sequenced and they represent 8 different members of a 12 member gene family. Our succes can be attributed to shorter 3′ non-coding regions that are typical of higher-plant genes and use of highly conserved gene family-specific primer in these experiments. This modified differential display technique can be of general application to other plant systems where cloning of the different members of a gene family is desired.

Notizen: Abstract To isolate genes specifically expressed at the initiation of plant embryo development we have applied a sensitive subtractive hybridization technique for three isogenic wheat lines of the so-called ‘Salmon system’ with either zygotic or autonomous embryo development. Here we present a gene sequence showing a high homology to grass pollen allergens of type II/III thought to be expressed in pollen tissue only. Surprisingly, the pollen allergen-like sequence, designated Tri a III, is also expressed in gynoecia of the sexual, male fertile wheat line ‘(aestivum)-Salmon’, whereas the two parthenogenetic and male sterile wheat lines ‘(caudata)-Salmon’ and ‘(kotschyi)-Salmon’ completely lack any Tri a III transcript. Our data suggest a positive correlation between the expression of this clone and the manifestation of male fertility. Northern and in situ hybridization analysis revealed that, in addition to its presence in pollen, Tri a III is expressed in the parenchymatous tissue of ‘(aestivum)-Salmon’ ovaries exclusively at the day of anthesis. This precise temporal and spatial expression pattern suggests a more general function of the pollen allergen-like sequence Tri a III not limited to the exhibition of allergens in pollen grains.

Notizen: Abstract In wheat (Triticum aestivum L.), water deficit during meiosis in the microspore mother cells (MMCs) induces pollen abortion, resulting in the failure of fertilization and a reduction in grain set. In stressed plants, meiosis in MMCs proceeds normally but subsequent pollen development is arrested. Unlike normal pollen grains, which accumulate starch during the late maturation phase, stress-affected anthers contain pollen grains with little or no starch. Stress also alters the normal distribution of starch in the anther wall and connective tissue. To determine how starch biosynthesis is regulated within the developing anthers of stressed plants, we studied the expression of ADP-glucose pyrophosphorylase (AGP), which catalyzes the rate limiting step of starch biosynthesis. Two partial-length cDNAs corresponding to the large subunit of AGP were amplified by RT-PCR from anther RNA, and used as probes to monitor AGP expression in developing anthers of normal and water-stressed plants. These clones, WAL1 and WAL2, had identical deduced amino acid sequences and shared 96% sequence identity at the nucleic acid level. In normal anthers, AGP expression was biphasic, indicating that AGP expression is required for starch biosynthesis both during meiosis and later during pollen maturation. AGP expression in stressed anthers was not affected during the first phase of starch accumulation, but was strongly inhibited during the second phase. We conclude from these results that the reduced starch deposition later in the development of stressed pollen could be the result of a lower expression of AGP. However, this inhibition of AGP expression is unlikely to be the primary cause of male sterility because anatomical symptoms of pollen abortion are observed prior to the time when AGP expression is inhibited.

Notizen: Abstract A genomic clone encoding the precursor of wheat leaf ferredoxin has been isolated and characterised. The uninterrupted PetF gene encodes a polypeptide of 143 amino acid residues, consisting of an N-terminal presequence of 46 amino acid residues and a mature polypeptide of 97 amino acid residues. Southern blot analysis suggests that six copies of the PetF gene are present in the wheat haploid genome. Northern blot analysis has shown that the genes are both developmentally and light regulated in wheat seedlings and provides evidence that a circadian rhythm regulates the steady-state levels of ferredoxin transcripts. The intact wheat gene and several chimeric constructs, containing portions of the 5′-upstream region fused to the β-glucuronidase reporter gene, have been introduced into tobacco plants, but levels of β-glucuronidase activity above background were not detected, suggesting that the 5′-upstream region is unable to function as a promoter in tobacco plants.

Notizen: Abstract The mitochondrial outer membrane of eukaryotic cells contains voltage-dependent anion channels (VDAC) also termed porins. Three cDNAs from wheat (Triticum aestivum) were isolated and sequenced (Tavdac 1–3). They share 65% similarity of their amino acid sequences, and therefore they probably represent isoforms. The deduced amino acid sequence of one of the cDNAs was found to be identical to the purified VDAC protein from wheat mitochondria [8]. Secondary structure analysis of the deduced amino acid sequences of the three vdac cDNAs revealed a characteristic α helix at their N-terminal and β-barrel cylinders characteristic of VDAC channels. The Tavdac cDNAs are differentially expressed in meristematic tissues. The transcript levels of Tavdac 1 in all wheat tissues is at least 2.5-fold higher than Tavdac 2 and Tavdac 3. Tavdac 2 has a low level of expression in all floral tissues whereas Tavdac 3 is highly expressed in anthers. This is the first report on differential expression of vdac genes in plants. The Tavdac genes have been mapped on the wheat genome. Tavdac 1 is located on the long arm of chromosome 5, Tavdac 2 on the long arm of chromosome 1 and Tavdac 3 on the long arm of chromosome 3. A phylogenetic reconstruction indicates that vdac genes underwent numerous duplication events throughout their evolution. All duplications occurred after the separation of plants from animals and fungi, and no orthologous genes are shared among phyla. Within plants, some of the vdac gene duplications probably occurred before the monocotydelon-dicotydelon split.

Notizen: Abstract A novel cDNA encoding for a peptidyl-prolyl-cis-trans-isomerase (PPIase) belonging to the FK506-binding protein (FKBP) family was isolated from wheat. It contains an open reading frame of 559 amino acids and it represents the first plant FKBP-PPIase to be cloned. It possesses a unique sequence which is composed of three FKPB-like domains, in addition to a putative tetratricopeptide repeat (TPR) motif and a calmodulin-binding site. The recombinant FKBP-PPIase expressed in and purified from Escherichia coli exhibits PPIase activity that is efficiently inhibited by the immunosuppressive drugs FK506 and rapamycin. Northern blot analysis showed that wheat FKBP was found mainly in young tissues. Polyclonal antibodies revealed the presence of cross-reacting proteins in embryos, roots and shoots. The unique structural features, the enzymatic activity and the presence of putative isoforms in wheat tissues indicate the possibility of the involvement of wheat PPIase in essential biological functions, similar to other members of the FKBP gene family.

Notizen: Abstract Dynamical aspects of three chloroplast promoters responding to change in light condition were examined in mature chloroplasts of wheat (Triticum aestivum) by in vitro transcription. The wheat psbD/C operon has four distinct promoters, two of which named as D/C-3 and D/C-4 promoters dominantly function in mature chloroplasts to produce the mRNAs encoding D2/CP43 and CP43 alone, respectively. Activity of the D/C-3 promoter in mature chloroplasts was reduced to less than 30% by 24 h dark adaptation and recovered by re-illumination to the original level within 30 to 60 min. The activation of the D/C-3 promoter which requires de novo cytoplasmic protein synthesis was induced by low fluence of light (e.g. 16 µE m-2 s-1), but the extent of activation increased with increasing light fluence. The accumulation of mRNAs from the D/C-3 promoter saturated at 2- to 3-fold higher level within 2 h when the dark-adapted seedlings were transferred to the lig at 72 µE m-2 s-1, concomitant with the increase in rate of D2 synthesis, suggesting that synthesis of D2 in mature chloroplasts is controlled via the D/C-3 promoter activity in a light-dependent way. Activity of the D/C-4 promoter slightly increased in the dark and decreased in the light. Effect of light on the psbA promoter activity was not observed at all in mature chloroplasts. In vitro transcriptional analysis of the D/C-3 promoter with 5′ deletion mutations revealed that at least two cis elements which are located within the sequences of -78 to -47 and -46 to -29 of the transcription initiation site, respectively, act as enhancing elements in the D/C-3 promoter. The light-switching element of the transcription, however, was suggested to be located in the core promoter sequence downstream of the -35 element.

Notizen: Abstract We have identified a new wheat PKABA1/-like protein kinase gene, TaPK3/, that is expressed in greening wheat seedlings. TaPK3 has high sequence homology (97% similarity with some sequence diversity at the 3' end) to the wheat PKABA1 protein kinase mRNA, which is upregulated by cold-temperature treatment, dehydration and abscisic acid (ABA). Use of a TaPK3 gene-specific probe has revealed that TaPK3 is differentially expressed with respect to PKABA1. TaPK3 mRNA accumulates in greening shoot tissue of wheat, but is not affected by dehydration, cold-temperature treatment or ABA. Based on sequence and expression differences, we conclude that expression of the PKABA1/-like protein kinases is not limited to stress responses.

Notizen: Abstract The low-temperature (2 °C)-specific wheat cDNA, pTACR7, represents a gene designated tacr7 from hard red winter wheat (HRWW; Triticum aestivum L. cv. Winoka). The term low-temperature-specific (LTS) is used because tacr7 is not induced by ABA or stresses such as salt, dehydration, and heat. pTACR7 was isolated by RT-PCR with mRNA from wheat crown tissue, the oligonucleotide primers derived from the barley cognate pHVCR8 (GenBank accession number L28091). Based on the deduced amino acid sequence, TACR7 is highly hydrophobic, with a single transmembrane domain and an amino acid bias for leucine (19%). Thus, the encoded protein TACR7 is unique among low-temperature-regulated wheat proteins described in the literature. Analysis of steady-state levels of tacr7 transcripts (630 nt) showed accumulation in wheat seedlings, crown tissue, and callus cultures after transfer from control (25 °C) to low temperature (2 °C). No detectable transcripts were observed by northern blot hybridization with pTACR7 probe from seedling or callus treated with ABA, salt, dehydration, or heat stress. tacr7 transcripts accumulated during 2 °C exposure to a greater amount in a freeze-resistant HRWW (FR; SDmut 16029) than in a freeze-susceptible HRWW (FS; SDmut 16169) crown tissue, with the largest difference between genotypes being 30% ± 3% at 3 weeks.

Notizen: Abstract We have developed a method for the accelerated production of fertile transgenic wheat (Triticum aestivum L.) that yields rooted plants ready for transfer to soil in 8–9 weeks (56–66 days) after the initiation of cultures. This was made possible by improvements in the procedures used for culture, bombardment, and selection. Cultured immature embryos were given a 4–6 h pre-and 16 h post-bombardment osmotic treatment. The most consistent and satisfactory results were obtained with 30 μg of gold particles/bombardment. No clear correlation was found between the frequencies of transient expression and stable transformation. The highest rates of regeneration and transformation were obtained when callus formation after bombardment was limited to two weeks in the dark, with or without selection, followed by selection during regeneration under light. Selection with bialaphos, and not phosphinothricin, yielded more vigorously growing transformed plantlets. The elongation of dark green plantlets in the presence of 4–5 mg/l bialaphos was found to be reliable for identifying transformed plants. Eighty independent transgenic wheat lines were produced in this study. Under optimum conditions, 32 transformed wheat plants were obtained from 2100 immature embryos in 56–66 days, making it possible to obtain R3 homozygous plants in less than a year.

Notizen: Abstract Photosynthetic pathways are reported for 305 species ofChenopodiaceae from Africa, Asia and Europe. Ecological characteristics, phytogeography and life forms of all species are given, and their correlation with relevant CO2-fixation types are discussed. 205 species (67.2%) exhibit the C4-pathway and 100 species (32.8%) the C3-pathway of CO2-fixation. Most of the C4 species are of Irano-Turanian origin. The diversity of C4 species of the Irano-Turanian phytochoria, with very harsh winters, are interpreted by the active period of Chenopods in summer. There is a close relationship between some special morphotypes and respective photosynthetic type. Halophytes and xerophytes with articulated stems and stem succulents ofAnabasis-type are exclusively C4. Leaf succulent halophytes and xerohalophytes are predominantly C4. Hygrohalophytes with leaf or stem succulence are often C3. Probably many C4 Salsoleae have been evolved in the Afroasiatic arid zone after the climatic changes of the Miocene. Among them there is a high proportion of annuals that have a younger origin. There is some evidence that the present Chenopods of the subfamily “Salsoloideae” may have their ancient stock in NW Africa. We found close taxonomic relationships between photosynthetic pathways and infrageneric classification in genera with both types of C3 and C4, likeAtriplex andSuaeda.

Notizen: Abstract Several isolates ofPhoma sp., certain nonsporulating fungi, as well asPenicillium andTrichoderma, all isolated from zoysiagrass rhizosphere, promoted growth of wheat and soybean under greenhouse conditions. However, the ability of these rhizosphere fungi to enhance plant growth varied with the crop tested. For example, most of the fungi effectively promoted the growth of wheat, whereas only a few fungi were effective on soybean. In consecutive plantings of wheat and soybean grown in soil previously infested with these zoysiagrass rhizosphere fungi, the growth promotion ability of the fungi was lowered. However, addition of fresh potting medium appeared to restore their growth-promotive effects. It appears that the activation of plant growth-promoting fungi in soil might depend on the availability of organic substrates to colonize, as evidenced by the promotion of plant growth.

Notizen: Abstract A partial genetic linkage map constructed using 150 single seed descent (SSD) lines generated from a cross between the hexaploid wheat varieties ‘Schomburgk’ and ‘Yarralinka’ was used to identify loci controlling milling yield. Milling yield data were obtained using seed collected from field trials conducted at different sites over two seasons. The estimated broad-sense heritability of milling yield in this population was calculated as 0.48. In the preliminary analysis, two regions were identified on chromosomes 3A and 7D, which were significantly associated with milling yield and accounted for 22% and 19% of the genetic variation, respectively. Bulked segregant analysis in combination with AFLP identified other markers linked to these loci, as well as an additional region on chromosome 5A, which accounted for 19% of the genetic variation. The applicability of these markers as selection tools for breeding purposes is discussed.

Notizen: Abstract Transgenic wheat plants containing the gene encoding snowdrop lectin (Galanthus nivalis agglutinin; GNA) under the control of constitutive and phloem-specific promoters were generated through the particle bombardment method. Thirty-two independently derived plants were subjected to molecular and biochemical analyses. Transgene integration varied from one to twelve estimated copies per haploid genome, and levels of GNA expression from 0 to ca. 0.2% of total soluble protein were observed in different transgenic plants. Seven transgenic plants were selected for further study. Progeny plants from these parental transformants were selected for transgene expression, and tested for enhanced resistance to the grain aphid (Sitobion avenae) by exposing the plants to nymphal insects under glasshouse conditions. Bioassay results show that transgenic wheat plants from lines expressing GNA at levels greater than ca. 0.04% of total soluble protein decrease the fecundity, but not the survival, of grain aphids. We propose that transgenic approaches using insecticidal genes such as gna in combination with integrated pest management present promising opportunities for the control of damaging wheat pests.

Notizen: Abstract The Yr17 gene, which is present in many European wheat cultivars, displays yellow rust resistance at the seedling stage. The gene introduced into chromosome 2A from Aegilops ventricosa was previously found to be closely linked (0.5 cM) to leaf and stem rust resistance genes Lr37 and Sr38, respectively. The objective of this study was to identify molecular markers linked to the Yr17 gene. We screened with RAPD primers, for polymorphism, the DNAs of cv. Thatcher and the leaf rust-resistant near-isogenic line (NIL) RL 6081 of cv. Thatcher carrying the Lr37 gene. Using a F2 progeny of the cross between VPM1 (resistant) and Thésée (susceptible), the RAPD marker OP-Y15580 was found to be closely linked to the Yr17 gene. We converted the OP- Y15580 RAPD marker into a sequence characterized amplified region (SCAR). This SCAR marker (SC-Y15) was linked at 0.8 ± 0.7 cM to the Yr17 resistance gene. We tested the SC-Y15 marker over a survey of 37 wheat cultivars in order to verify its consistency in different genetic backgrounds and to explain the resistance of some cultivars against yellow rust. Moreover, we showed that the Xpsr150-2Mv locus marker of Lr gene described by Bonhomme et al. [6] which possesses A. ventricosa introgression on the 2A chromosome was also closely linked to the Yr17 gene. Both the SCAR SC-Y15 and Xpsr150-2Mv markers should be used in breeding programmes in order to detect the cluster of the three genes Yr17, Lr37 and Sr38 in cross progenies.

Notizen: Abstract We recently showed that the Lr10 wheat leaf rust resistance gene cosegregated with the candidate resistance gene Lrk10 which encodes a putative receptor-like kinase. The aim of this study was to develop Lrk10-derived molecular markers for the detection of the Lr10 gene in breeding material. Different subfragments of Lrk10 were tested as RFLP markers for the Lr10 resistance gene. The most specific fragment (Lrk10-6) was converted into the PCR-based STS marker STSLrk10-6. Both the RFLP and the STS marker did not give a signal with near isogenic lines containing a different Lr gene. The applicability of these markers for the detection of Lr10 in genetically diverse material was tested with 62 wheat and spelt breeding lines, mostly from European breeding programmes. Twelve varieties known to have Lr10 showed the same alleles as the originally characterized line ThatcherLr10. Most of the lines with unknown composition at the Lr10 locus had a null allele with both the RFLP marker Lrk10-6 and the marker STSLrk10-6 whereas 20% of the lines had a different allele. For six lines, including a traditional spelt variety derived from a landrace, both markers showed the same allele as Thatcher Lr10. Artificial infections of these lines with an isolate avirulent on Lr10 resulted in a hypersensitive reaction of all these lines, indicating also the presence of the Lr10 resistance gene. These data demonstrate that the markers derived from sequences of Lrk10 are highly specific for the Lr10 gene in breeding material of very diverse genetic origin. The markers will allow the defined deployment of Lr10 in wheat breeding programmes and will contribute to the elucidation of the role of Lr10 in polygenic resistances against leaf rust.

Notizen: Abstract Near-isogenic lines in conjunction with bulked segregant analysis were used to identify a DNA marker in wheat (Triticum aestivum L.) associated with the H21 gene conferring resistance to biotype L of Hessian fly [Mayetiola destructor (Say)] larvae. Near-isogenic lines were developed by backcross introgression BC3F3:4 (‘Coker 797’ * 4 / ‘Hamlet’) and differed by the presence or absence of H21 (on 2RL) derived from ‘Chaupon’ rye (Secale cereale L.). Bulked DNA samples were prepared from near-isogenic lines and BC3F2 population individuals segregating for reaction to Hessian fly biotype L and screened for random amplified polymorphic DNA markers using 46 10mer primers. Random-amplified polymorphic DNA markers from resistant and susceptible individuals and parental lines were scored and these data were used to identify a 3 kb DNA fragment that was related to the occurrence of H21. This fragment was amplified from DNA isolated from Hamlet, a near-isogenic line carrying 2RL, and bulked DNA from resistant BC3F2 individuals, but not from the recurrent parent Coker 797 or DNA bulks from susceptible BC3F2 plants. Analysis of 111 BC3F2 segregating individuals and BC3F2:3 segregants confirmed the co-segregation of the 3 kb DNA marker with the H21 resistance gene to Hessian fly. Use of this marker could facilitate more rapid screening of plant populations for Hessian fly resistance and monitoring the introgression of H21.

Notizen: Abstract In 1993–1996, the virulence of regional populations of the wheat powdery mildew pathogen (Erysiphe graminis DC f. sp. tritici Marchal) from the Czech Republic, Austria, Hungary and Slovakia against 13 resistance genes was investigated. The populations differed mainly at the regional level. Populations from the Czech Republic, mainly from the western regions, showed higher values of virulence against the Pm4b gene. Lower frequency of virulence against Pm4b was found in Austria, and the lowest value was observed in Hungary. The differences in frequencies of virulence against Pm4a and Pm4b showed a similar geographic pattern across the four countries: a continuous decline from west to east and from north to south. Virulence against Pm2 decreased in all countries considered; virulence to pm5, Pm6, Pm8 and Mli was high throughout. Genes and gene combinations that can ensure a relatively effective biological protection against this pathogen across Central Europe at present are Pm3b, Pm2+Mld and Pm1+2+9. Czech and Slovak populations were the most complex: virulence complexity reached a maximum in Slovakia in 1994. A similar evolution, though less significant, was observed in the Czech Republic. Data on complexity of isolates suggest that Central European populations of wheat powdery mildew tend to reach an intermediate level representing the optimal number of virulence genes. This process is probably a consequence of stabilizing selection.

Notizen: Abstract Field experiments with winter cereals grown on soil inoculated withC. gramineum showed that wheat and rye cultivars possess some resistance to the pathogen, while the triticale cultivars were the most susceptible. Higher tolerance of the tested wheat cultivars was connected mainly with slow development of disease symptoms; rye cultivars had, on average, lower percentages of plants infected byC. gramineum. The greatest variation in susceptibility toC. gramineum occurred among the selected cultivars of triticale.

Notizen: Abstract Apothecia of the eyespot fungus,Tapesia yallundae, were found on 0–18% of straws in plots of wheat stubble in February–March 1994. The fungicides carbendazim, prochloraz or carbendazim plus prochloraz had been applied repeatedly to the same plots in each of the previous 9 years in which successive wheat crops had been grown. The factors most strongly correlated with the incidence of apothecia were the incidence and severity of eyespot in the preceding wheat crop and the frequency of carbendazim-resistant W-type fungus in populations recovered from that wheat crop. Plots treated with carbendazim, which had previously had more disease and more resistance to carbendazim in the pathogen population relative to untreated plots, therefore yielded most apothecia. Plots treated with prochloraz, which had selected for predominantly R-type fungus and decreased eyespot, yielded few apothecia. Single-ascospore isolates were all of the W-type and were more frequently carbendazim-sensitive than expected, except those from plots treated only with carbendazim. None showed decreased sensitivity to prochloraz. The implications of applying fungicides regularly for controlling eyespot on the capability of the eyespot fungus for genetic variation through sexual reproduction are discussed.

Notizen: Abstract One could predict that the capacity for travelling great distances might predispose long-distance migratory birds to be good colonists and to exhibit wider geographic distributions than their non-migratory or short-distance migratory relatives or non-volant mammals. This prediction is not supported by the data on avian biogeography. The distributions of species, genera and families of North American and Eurasian birds and mammals are indeed related to migratory status, but long-distance migratory birds exhibit a great deal of biogeographic regionalism. In particular, at all taxonomic levels their distributions tend to be confined to either the Eastern or Western Hemisphere, suggesting that there has been little successful east–west dispersal between North America and Eurasia. Compared to non-migratory birds, short-distance migratory birds and non-volant mammals, long-distance migrants appear to be subject to severe constraints on their physiology, behaviour and ecology, w hich have prevented colonization of distant regions.

Notizen: Abstract The longitudinal distribution of Trichoptera at 37 sampling sites along 800 km of the Loire River (France) was analyzed in order to better understand large-scale changes in community structure occurring on a stream continuum. Different simple ordination methods — centered PCA and Correspondence Analysis — were performed to investigate these changes. Both analyses showed that a major change occurs in the community in the upper reaches of the Loire River between the Serre de la Fare and Vorey sampling sites with the appearance of Hydropsyche contubernalis and H. Exocellata. Even though other changes in community structure could exist, revealed either by Correspondence Analysis at the first sampling sites (crenal), or by PCA at sampling sites influenced by dams, this Trichoptera community appears to be stable enough in the latter part of the Loire River with the addition of a few species, Hydropsyche bulgaromanorum, Ecnomus tenellus and E. deceptor, rather than changes in species. The results are discussed in the light of different concepts in stream ecology, and the validity of such data analyses to describe some community patterns.

Notizen: Abstract Meiosis was examined in pollen mother cells of F1 hybrids made from crosses between wheat (Triticum aestivum) and lymegrass (Leymus arenarius and L. mollis). Fluorescence genomic in situ hybridization detected pairing between wheat and lymegrass chromosomes during prophase I and metaphase I. Such pairing, when resulting in bivalent formation, was likely to yield correct disjunction, and hence intergenomic recombination could be incorporated into the gametes. Bivalents in these hybrids, however, were more frequently formed between chromosomes of the same parental origin. Univalents were common, whereas multivalents were not clearly detected. Meiotic behaviour in some cells was not totally aberrant, and this may have accounted for the presence of normal pollen. The results are discussed in relation to intergenomic pairing, meiotic behaviour in wide-hybrids and genome relationships, including the Leymus genome origin.

Notizen: Abstract Fluorescencein situ hybridization (FISH) was used to determine the breakpoint of the translocation chromosome in two bread wheat (Triticum aestivum) germplasm lines withThinopyrum intermedium chromatin carrying resistance to either wheat streak mosaic virus (WSMV) or barley yellow dwarf virus (BYDV). In addition, genome-specific random amplified polymorphic DNA (RAPD) markers were used to ascertain the genomic sources of theTh. intermedium chromosomes carrying the WSMV or BYDV resistance. CI17766, a WSMV-resistant wheat germplasm line derived from induced homoeologous pairing by using theph1b mutant, had a translocation chromosome composed of the complete 4AL and about 45% of proximal 4AS from wheat, and the entire 4ES ofTh. intermedium. The BYDV-resistant translocation line, TC14, derived from tissue culture, had a very short distal segment of 7StL fromTh. intermedium terminally attached to 56% of the proximal 7DL. These observations indicate that translocations in these wheat germplasm lines did not involve centromeric breaks and fusion but were a result of homoeologous chromosome recombination.

Notizen: Abstract Analysis of genes controlling flowering time (heading date) contributes to our understanding of fundamental principles of plant development and is of practical importance because of the effects of flowering time on plant adaptation and crop yield. This review discusses the extent to which plants may share common genetic mechanisms for the control of flowering time and the implications of such conservation for gene isolation from the major cereal crops. Gene isolation may exploit the small genome of rice in map-based approaches, utilizing the conservation of gene order that is revealed when common DNA markers are mapped in different species. Alternatively, mechanisms may be conserved within plants as a whole, in which case genes cloned from the model dicot Arabidopsis thaliana provide an alternative route.

Notizen: Abstract In plants, several types of receptor-like kinases (RLK) have been isolated and characterized based on the sequence of their extracellular domains. Some of these RLKs have been demonstrated to be involved in plant development or in the reaction to environmental signals. Here, we describe a RLK gene family in wheat (wlrk, wheat leaf rust kinase) with a new type of extracellular domain. A member of this new gene family has previously been shown to cosegregate with the leaf rust resistance gene Lr10. The diversity of the wlrk gene family was studied by cloning the extracellular domain of different members of the family. Sequence comparisons demonstrated that the extracellular domain consists of three very conserved regions interrupted by three variable regions. Linkage analysis indicated that the wlrk genes are specifically located on chromosome group 1 in wheat and on the corresponding chromosomes of other members of the Triticeae family. The wlrk genes are constitutively expressed in the aerial parts of the plant whereas no expression was detected in roots. Protein immunoblots demonstrated that the WLRK protein coded by the Lrk10 gene is an intrinsic plasma membrane protein. This is consistent with the hypothesis that WLRK proteins are receptor protein kinases localized to the cell surface. In addition, we present preliminary evidence that other disease resistance loci in wheat contain genes which are related to wlrk.

Notizen: Abstract I compare the primatological literature in 4 Latin American (Brazil, Perú, Mexico and Argentina) 2 European (Great Britain and France) and one African (Kenya) countries with regard to: (1) total number of publications; (2) productivity of scientists; (3) quality of the publications; and (4) the range of subjects studied. Data are from Current Primate References (CPR; 1985–1994). Publications included in the sample show an address from the countries considered. The results showed that GB and France had a far larger number of publications than the other countries. They have respectively 〉7 and 4 times more authors than Brazil, the country with the highest number of authors among Latin American and African countries. But the mean number of publications per author shows little difference among the 7 countries. The quality of publications as measured by the percentage indexed by ISI shows that, GB and France fare much better than the Latin American countries. Kenya did not follow the pattern of the other underdeveloped countries and showed a high percentage of indexed publications. Not all subjects reviewed in CPR were studied by scientists from the 7 countries. GB and France presented the widest range of subjects. Scientists in countries with greater primate diversity concentrated their publications in behavior, ecology and conservation, colony management and general primatology with the exception of Kenya. These results suggest that scientific production in Latin American countries can be increased by increasing the number of scientists, either through educational programs or via incentives for the career. However, more specific action is needed to improve the quality of publications and the range of subjects studied.

Notizen: Abstract The effect of 24 treatment combinations of cultures of Streptomyces costaricanus sp. nov. (ATCC55274), Bacillus thuringiensis (ATCC55273) and a strain of Paecilomyces marquandii, nematicide (cadusaphos), and/or wheat mash on growth and response of potted banana plants (Musa AAA) and populations of Radopholus similis, Helicotylenchus multicinctus and free living nematodes were studied in Río Frío, Costa Rica. The best plant responses (height, leaf numbers, healthy root weight), lowest numbers of plant parasitic nematodes and highest numbers of free living nematodes were observed for treatments containing wheat as a component. Two treatments, viz. wheat + Streptomyces costaricanus (200-ml culture) and wheat + P. marquandii (200-ml culture), gave the overall best results. Numbers of free living nematodes increased up to 1500-fold only for treatments containing wheat. Significant positive correlations existed between numbers of free living nematodes and shoot weight, healthy root biomass, plant height, and leaf numbers. Non-wheat treatments, including nematicide only, gave the poorest responses in general. Observations of nematodes sampled 50 days following planting in wheat-containing treatments showed most of the free-living nematodes (≈ 90%) to be infected by nematophagous fungi (species not recorded). The results show that an organic amendment to soil, with or without a microbial component, can be an effective inducer of processes that regulate plant-parasitic nematode populations in soil.

Notizen: Abstract In 1944 a single mandibular specimen was found in Pyrgos, southern Greece.von Koenigswald (1972) described this specimen and allocated it to a new taxon,Graecopithecus freybergi.de Bonis and colleagues later described new hominid specimens from northern Greece and allocated them to the new taxon,Ouranopithecus macedoniensis.Martin andAndrews (1984), however, argued that there was no reason for maintaining a generic or specific distinction between the northern and southern Greek fossil populations as suggested byde Bonis andMelentis. This paper reexamines the taxonomic status of the southern and northern Greek hominids in light of recent fossil discoveries and concludes that there is no evidence for recognizing two hominine genera from the Miocene of Greece. There is evidence, however, to suggest that the Macedonian hominid is specifically distinct from the southern Greek hominid. Thus, becauseGraecopithecus has taxonomic priority overOuranopithecus, it is concluded that the Macedonian specimens should be allocated to a separate species ofGraecopithecus.

The results are clearly consistent with the existence of four species (H. italica, H. paivae, H. reverchonii andH. mauritanica). It is proposed that the latter be considered to comprise two subspecies,H. mauritanica subsp.mauritanica andH. mauritanica subsp.vincentina (comb. nov.). Notizen: Abstract Hay malting and hay meadows have long been of fundamental ecological, economic and social importance in temperate Europe. A variety of archaeological sources suggests that hay making may date back to the Iron Age, but direct archaeobotanical evidence for this practice is problematic. Past grassland communities are imperfectly represented and preserved in archaeobotanical assemblages, and ancient meadow and pasture communities may not resemble present-day communities in terms of management practices or botanical composition. This paper explores the potential of ‘FIBS’ (Functional Interpretation of Botanical Surveys) in the archaeobotanical investigation of ancient grassland management. The botanical composition of present-day grassland communities was analysed in terms of functional attributes (e.g. canopy height) relevant to cutting, grazing and habitat productivity. The utility of these attributes for distinguishing between present-day meadow, pasture and unmanaged grassland communities has been evaluated. Similar analyses were performed on archaeobotanical data from Neolithic to post-Medieval northwestern and central Europe. Functional shifts over time, interpreted in the light of the functional analysis of modern grassland, suggest that hay-making was well established by the Iron Age. Avenues are suggested for the refinement and further development of the FIBS methodology in the archaeobotanical investigation of grassland management.

Notizen: Abstract Although maize endosperm undergoes programmed cell death during its development, it is not known whether this developmental feature is common to cereals or whether it arose inadvertently from the selection process that resulted in the enlarged endosperm of modern maize. Examination of wheat endosperm during its development revealed that this tissue undergoes a programmed cell death that shares features with the maize program but differs in some aspects of its execution. Cell death initiated and progressed stochastically in wheat endosperm in contrast to maize where cell death initiates within the upper central endosperm and expands outward. After a peak of ethylene production during early development, wheat endosperm DNA underwent internucleosomal fragmentation that was detectable from mid to late development. The developmental onset and progression of DNA degradation was regulated by the level of ethylene production and perception. These observations suggest that programmed cell death of the endosperm and regulation of this program by ethylene is not unique to maize but that differences in the execution of the program appear to exist among cereals.

Notizen: Abstract A new screening technique for tolerance to high concentrations of boron, namely a filter paper technique, and a soil experiment were compared to investigate the response of wheat genotypes known to differ in tolerance to high concentrations of boron. Under high boron concentrations in filter papers, the more tolerant genotypes had significantly longer roots than those of the more sensitive genotypes. There was no significant correlation between the root lengths at the control treatment and the other three boron treatments (50, 100, 150 mg B L-1). Thus, the differences in root lengths at the high boron treatments could not be attributed to inherent differences in root growth but to the genetic variation in response to high boron concentrations among varieties. Root lengths at the three boron treatments in filter papers were highly significantly correlated with the three characters determined for plants grown in soil containing high levels of boron, namely the concentrations of boron in the shoots, plant dry weight and plant symptoms, indicating that root length could be used as a selection criterion in a genetic study or breeding program for boron tolerance.

Notizen: Abstract To obtain nutrients mineralised from organic matter in the soil, plants have to respond to its heterogeneous distribution. We measured the timing of nitrogen uptake by wheat from a localised, 15N labelled organic residue in soil, as well as the timing of changes in root length density. We calculated the rates of N uptake per unit root length (inflows) for roots growing through the residue and for the whole root system. A stimulated local inflow appeared to be the main mechanism of exploitation of the residue N during the first five days of exploitation. 8% of the N that the plants would ultimately obtain from the residue was captured in this period. Roots then proliferated in the residue. This, together with a rapidly declining N inflow, contributed to the capture, over the next seven days, of 63% of the N that the plants derived from the residue. After that time, massive root proliferation occurred in the residue, but relatively little further N was captured.

Notizen: Abstract Yield of eight wheat cultivars was evaluated under rainfed and irrigated conditions in a Mediterranean environment. Variation in grain yield resulted from variation in both aboveground biomass production and in harvest index. Under rainfed compared to irrigated conditions, grain yield, biomass and days to heading were decreased, whereas harvest index was increased. Grain yield of the different cultivars under rainfed conditions correlated with that under irrigated conditions in one of the two years. Among cultivars, harvest index under rainfed and irrigated conditions were correlated in both years. Water was used more efficiently for biomass production, and equally efficiently for grain production, under irrigated compared to rainfed conditions. Under rainfed conditions, crop water use efficiency was higher for cultivars developed for rainfed environments than for those developed for high-rainfall or irrigated environments. Cultivars with low-rainfall target environments had the lowest evapotranspiration under rainfed conditions. Under rainfed conditions, differences between the cultivar groups in crop water use efficiency corresponded with trends in water use efficiency of individual plants and with the ratio of photosynthesis to transpiration, measured on plants grown in a growth room. Early in the season, water was used more efficiently for biomass production at high sowing densities than at low sowing densities. Through faster biomass production and ground cover a smaller proportion of the evapotranspired water was lost in soil evaporation and a larger proportion was transpired. However, the net effect was a greater water use in the early phases of growth and consequently a lower water availability later in the season, leading to similar yields regardless of sowing density.

Notizen: Abstract Wheat plants differing in zinc efficiency (Excalibur; Zn-efficient, Gatcher and Durati; Zn-inefficient) were grown in HEDTA chelate-buffered nutrient solution in controlled conditions and supplied with 0 or 40 μg chlorsulfuron L-1 . Zinc uptake rates of 12-d-old plants were measured over 80 or 90 minutes using65 Zn added to nutrient solutions. Increasing the zinc concentration of the solution increased the rate of zinc uptake, while the percentage of zinc transported to shoots was decreased. Addition of chlorsulfuron to uptake solutions for 90 minutes did not influence rate of zinc uptake or transport of zinc to shoots. Pretreating plants with chlorsulfuron for 5 days decreased zinc uptake rates, but transport to shoots was proportionally increased. Three-day pretreatment with chlorsulfuron was the minimum required for significant differences in uptake and transport of zinc to occur. Plants exposed to chlorsulfuron for 3 days required a further 5 days of growth in chlorsulfuron-free solutions before uptake rates recovered to control plant rates. It is concluded that chlorsulfuron deleteriously but reversibly affects uptake of zinc across the plasma membrane after prolonged exposure.

Notizen: Abstract A crown rot disease in wheat caused by the fungusFusarium graminearum Schw. Group 1 is a widespread problem in chronically Zn-deficient Australian soils. A link between crown rot and Zn deficiency was established by Sparrow and Graham (1988). This paper reports a test of a further hypothesis, that wheat genotypes more efficient at extracting zinc from low-zinc soils are more resistant to infection by this pathogen. Three wheat cultivars (Excalibur, Songlen and Durati) of differential Zn efficiency were tested at three zinc levels (0.05, 0.5 and 2.0 mg Zn kg−1 of soil) and three levels ofF. graminearum S. Group 1 inoculum (0.1 g and 0.3 g kg−1 live chaff-inoculum and control having 0.1 g kg−1 dead chaff inoculum). Six weeks after sowing dry matter production of shoots and roots was decreased byFusarium inoculation at 0.05 mg and 0.5 mg kg−1 applied Zn.Fusarium inoculum at 0.1 g was as effective as 0.3 g kg−1 for infection and decreasing dry matter. The infection at the basal part of culm decreased significantly by increasing the rate of Zn application. Excalibur, a Zn-efficient cultivar (tolerant to Zn deficiency) produced significantly more shoot and root dry matter, and showed less disease infection compared with Zn-inefficient cultivars (Durati and Songlen) at low (0.05 mg Zn kg−1 soil) and medium (0.5 mg Zn kg−1 soil) Zn fertilization rates. Higher rate of Zn fertilization (2.0 mg Zn kg−1 soil) reduced the disease level in Durati to the level of Excalibur but the disease level of Songlen was still high, indicating its high Zn requirement and or sensitivity to crown rot. The data on Zn uptake show that Excalibur, being Zn-efficient, was able to scavenge enough Zn from Zn-deficient soil, we suggest that besides sustaining growth Excalibur was able to build and maintain resistance to the pathogen; inefficient cultivars needed extra Zn fertilization to achieve performance comparable to that of Excalibur. The present study indicates that growing Zn-efficient cultivars of wheat along with judicious use of Zn fertilizer in Zn-deficient areas where crown rot is a problem may sustain wheat production by reducing the severity of the disease as well as by increasing the plant vigour through improved Zn nutrition. ei]Section editor: R Rodriques-Kalana

Notizen: Abstract The influence of arbuscular mycorrhizal fungi on drought tolerance and recovery was studied in safflower (Carthamus tinctorius L.) and wheat (Triticum aestivum L.). Plants were grown with and without the mycorrhizal fungus, Glomus etunicatum Becker & Gerd., in nutrient-amended soil under environmentally-controlled conditions to yield mycorrhizal and nonmycorrhizal with similar leaf areas, root length densities, dry weights, and adequate tissue phosphorus. When drought stress was induced, mycorrhizal infection did not affect changes in leaf water, osmotic or pressure potentials, or osmotic potentials of leaf tissue rehydrated to full turgor in either safflower or wheat. Furthermore, in safflower, infection had little effect on drought tolerance as indicated by the level of leaf necrosis. Mycorrhizal wheat plants, however, had less necrotic leaf tissue than uninfected plants at moderate levels of drought stress (but not at severe levels) probably due to enhanced phosphorus nutrition. To determine the effects of infection on drought recovery, plants were rewatered at a range of soil water potentials from –1 to –4 MPa. We found that although safflower tended to recover more slowly from drought after rewatering than wheat, mycorrhizal infection did not directly affect drought recovery in either plant species. Daily water use after rewatering was reduced and was correlated to the extent that leaves were damaged by drought stress in both plant species, but was not directly influenced by the mycorrhizal status of the plants.

Notizen: Abstract The effects of Rht alleles on root growth and distribution in isogenic lines of spring wheat (Triticum aestivum L.) are described under different environmental conditions. Above-ground biomass, root length, root dry-weight and their distribution along the soil profile were measured by destructive sampling for growth of aerial biomass and extraction of soil cores containing roots. Field experiments were conducted under non-limiting water and nutritional conditions during two consecutive years, using an early and a late sowing date each year. Dwarfing genes significantly reduced plant height and above-ground biomass at anthesis. In addition, stem mass ratio also was reduced with increases in the allelic dosage. Conversely, total root length and root dry-weight per unit area at anthesis were increased with decreased plant height, therefore, root mass ratio tended to be negatively correlated with plant height. Differences in distribution of root length and root dry-weight through the soil profile among lines were largely confined to the upper soil layers (i.e. the top 30 cm). Differences in root dry-weight were more important than in root length, so that the dwarf line had the highest root mass per unit root length. Furthermore, a significant positive correlation between the root mass ratio and stem mass per unit stem length was found. It is suggested that increases in root mass per unit root length associated with Rht alleles are evidencing a surplus of photoassimilates during stem elongation which are used for thickening the roots due to the lack of alternative sinks. Agronomic implications of this effect are discussed.

Notizen: Abstract The residual effects of Zn supplied (+Zn: 1 mg kg−1 soil, -Zn: no Zn added) in subsoil to four oilseed rape genotypes (Zhongyou 821, Xinza 2 and Narendra: Brassica napus type, and CSIRO-1: Brassica juncea type) were studied on the following wheat in a glasshouse experiment in pots (100 cm long, 10.5 cm diameter). The topsoil (upper 20 cm soil in pots) was supplied with Zn (0.5 mg kg−1 soil) in all treatments whereas Zn (1 mg Zn kg−1 dry soil) was either supplied or omitted from the subsoil at time of sowing of oilseed rape genotypes. Oilseed rape plants were harvested at maturity and wheat (variety Songlen) was grown to evaluate the residual effects of subsoil Zn and oilseed rape genotypes. Subsoil residual Zn significantly improved the root growth, yield attributes, grain and straw yield of wheat. Wheat grain Zn concentration was 4 times higher in +Zn subsoil residual than under -Zn subsoil residual. Oilseed rape genotypes did not vary in their residual effect on grain and straw yield of wheat, but reflected a differential effect on Zn concentration and Zn content of wheat grain. Zinc uptake per wheat plant (grain + shoot + root Zn uptake) was about 4 times higher in +Zn subsoil residual than -Zn subsoil residual. Out of total Zn uptake per plant, 77%, 15% and 8% of Zn was loaded in wheat grains, shoots and roots respectively in +Zn subsoil residual. However in -Zn subsoil residual, distribution of Zn in grains, shoots and roots was 55%, 29% and 16% respectively. Zinc loading in grains of wheat was significantly higher in wheat grown after Narendra and Xinza 2 compared with wheat grown after Zhongyou 821 (Zn-inefficient genotype). These results indicate that subsoil residual Zn and Zn-efficient genotypes of oilseed rape have the potential to increase the Zn loading in wheat grains.

Notizen: Abstract The simulation of crop - soil systems with a model requires an appropriate description of the root dynamics. An empirical root growth model that simulates root-shoot relations, root distribution and a dynamic response to environmental conditions is presented. The root model extends an existing crop model and links it to a soil model to calculate dry matter accumulation, water and nitrogen dynamics of a wheat crop. Simulated roots are distributed over soil layers according to carbon supply from the shoots by using a 'top down principle'. This principle favours the top layers for root growth by first providing all available carbon to the first layer. Under unfavourable soil conditions in that layer, carbon is given to the next deeper soil layer. This procedure is repeated until a separately calculated rooting depth is reached. At that depth all available carbon is used for root growth regardless of current soil conditions. Under most simulated conditions the 'top down principle ' results in a negative exponential function of a monotone decrease of root distribution with soil depth. However, it can also account for larger root densities deeper in the profile when water or nitrogen deficiency occurs in soil. In addition to soil water and soil nitrogen supply the root model considers soil compaction, aeration and root distribution history for root growth simulation. The new model, consisting of an existing crop and soil model and linked through a new developed root model, was calibrated and tested using two independent field experiments. A sensitivity analysis was carried out by varying parameters, initial soil conditions and hypothetic weather patterns as part of the validation process. Root length density distribution (r2(1:1)=0.65), shoot, grain and total root biomass (r2(1:1)=0.87) were predicted satisfactorily, thus providing a useful tool for specific simulation studies on that site.

Notizen: Abstract Experiments were conducted under laboratory and greenhouse conditions to study the effect of 2,4-D on rooting and salinity tolerance of wheat. Seeds of one commercial wheat (Inqalab-91) and three salt-tolerant wheat lines (WL-41, WL-359, and WL-1073 developed through wide hybridization) were included in the study. Preliminary and short-term experiments were conducted to determine the level of 2,4-D (administered through seed soaking for 24.5 h. at 25 °C in the dark) at which the maximum number of roots emerged. Under hydroponic conditions, 2,4-D treatment of seeds caused an increase of 60 to 100% in the number of primary roots. The maximum increase in the number of roots was observed in one of the salt tolerant wheat lines (WL-41). The roots appeared in bunches but showed stunted growth at higher levels of 2,4-D. Dry matter accumulation decreased markedly; the effect was more pronounced in Inqalab-91 which is less tolerant to stress than other wheat lines. In all wheat types, allocation of dry matter to roots relative to shoot increased due to 2,4-D treatment. In soil, seeds treated with different levels of 2,4-D showed a germination delay of 1–3 days. Although the number of primary roots increased, 2,4-D treatment caused a decrease in total dry matter accumulation by plants grown for 40 days. In another experiment, conducted under greenhouse conditions, seed germination and growth of seedlings was significantly retarded in saline compared to that in non-saline (normal) soil. Initially, the pace of germination of treated seeds as well as seedling growth was slower in both soils, but after six weeks, the leaf area of seedlings raised from treated seeds was greater than those raised from untreated seeds. Towards maturity, plants arising from treated seeds developed wider and longer flag leaves leading to enhanced yield. Root biomass decreased in saline soil as compared to normal soil. However, 2,4-D treatment caused a substantial increase in root biomass in saline soil and the roots were harder in texture in wheats other than Inqalab-91. Seed treatment with 2,4-D led to a significant improvement in the number of productive tillers, yield of straw and grain, and grain protein content of all wheats grown in saline soil. Plants grown in normal soil did not show any marked effect of seed treatment on grain yield and other agronomic parameters. The four wheats showed substantial differences for different parameters but the salt tolerant wheat lines performed better compared to the commercial variety Inqalab-91.

The objectives of this work were to (i) determine whether tiller emergence under low phosphorus availability is a function of the availability of assimilates for growth or a direct result of low P availability, (ii) attempt to establish a quantitative relation between an index of the availability of P in the plant and the effects of P deficiency on tiller emergence, and (iii) to provide a better understanding of the mechanisms involved in tiller emergence in field-grown wheat. Wheat (Triticum aestivum L., cv. INTA Oasis), was grown in the field under drip irrigation on a typic Argiudol, low in P (5.5 μg P g-1 soil Bray & Kurtz I) in Balcarce, Argentina. Treatments consisted of the combination of three levels of P fertilization 0, 60 and 200 kg P2O5 ha-1, and two levels of assimilate availability, a control (non-shaded) and 65% of reduction in incident irradiance from seedling emergence until the end of tillering (shaded). Phosphorus treatments significantly modified the pattern of growth and development of the plants. Shading reduced the growth and concentration of water-soluble carbohydrates in leaves and stems. Leaf photosynthetic rate at saturating irradiance was reduced by P deficiency, but was not affected by shading. At shoot P concentrations less than 4.2 g P kg-1 the heterogeneity in the plant population increased with respect to the number of plants bearing a certain tiller. At a shoot P concentration of 1.7 g P kg-1 tillering ceased completely. Phosphorus deficiency directly altered the normal pattern of tiller emergence by slowing the emergence of leaves on the main stem (i.e. increasing the phyllochron), and by reducing the maximum rate of tiller emergence for each tiller.

Notizen: Abstract Wheat production is a major source of income for most people and forms the backbone of the economy in Central Anatolia, Turkey. Sunn pest, known in Turkey since 1927, is one of the most harmful pests of wheat in this region where the infested area has reached 0.3 million ha and the cost of chemical application has reached US$ 2 million per year. Two species of Eurygaster, E. maura and E. austriaca, are prevalent in the region. Observations of tests carried out on different genotypes under natural field conditions and controlled greenhouse tests showed variable response of wheat to sunn pest.

Notizen: Abstract A method for efficient genetic transformation of wheat has been developed using immature embryos as targets for microprojectile-mediated gene transfer and a helium driven particle delivery system. Screening and selection of transgenic cells, somatic embryos and regenerated plants are performed with the gus-gene and the phosphinothricin acetyl transferase (bar) gene coding for Basta-resistance as the selectable marker. On average, one fertile transgenic plant can be obtained from about 100 microprojectile treated, immature embryos. The number of integrated copies of the transferred gene ranges from 1 up to about 10. Stable integrated genes are inherited in most of the transgenic lines in a normal mendelian fashion segregating 3:1 in the F2. Homozygous, as well as heterozygous, lines have been followed and analysed genetically at the molecular level and up to F5. Apart from normal stable gene expression, examples have also been found which showed a loss of gene activity or unexpected segregation pattern. For applied aspects, different genes are transferred aiming for improved disease resistance, modification of quality, or other characteristics. First results from these transgenic lines are reported, and problems still existing with the production of stable transgenic wheat lines are discussed.

Notizen: Abstract Substitution lines with reciprocal substitutions of chromosomes containing recessive alleles of the homoeologous group 5 chromosomeVrn genes between varieties of winter wheat with high vernalisation requirement (‘Mironovskaya 808’) and low vernalisation requirements (‘Bezostaya 1’) have been created. On this basis the genetic determination of vernalisation requirement was established. Substitution lines Mironovskaya 808 (Bezostaya 1 5A), Mironovskaya 808 (Bezostaya 1 5B), Mironovskaya 808 (Bezostaya 1 5D) and reciprocal substitution lines Bezostaya 1 (Mironovskaya 808 5A), Bezostaya 1 (Mironovskaya 808 5B) and Bezostaya 1 (Mironovskaya 808 5D) were grown under different durations of vernalisation (3, 4, 5, 6, 7 and 8 weeks) and their response was evaluated. Photoperiodic sensitivity of the original parental genotypes was also determined. Reciprocal substitution lines of the same chromosome that carries the same vrn allele responded differently to vernalisation deficit. Differences have been shown between all group 5 reciprocal substitutions. Lines carrying chromosomes 5A and 5D of Mironovskaya 808 had a high vernalisation requirement whereas lines carrying chromosome 5B of Bezostaya 1 (vrn2B) had a low vernalisation requirement. The reciprocal lines had a reverse requirement. This explains the different vernalisation requirements of the original varieties: Mironovskaya 808 with a high vernalisation requirement carries two alleles (vrn1M and vrn3M) in its genotype that increase the vernalisation requirement, whereas Bezostaya 1 with a lower requirement for vernalisation contains only one such allele (vrn2B). By combination of the alleles in the lines with the substitution of chromosome 5B carrying vrn2 allele that in both original genotypes work inversely to the other alleles, transgressive genotypes have been formed: genotype vrn1M vrn2B vrn3M determines a higher vernalisation requirement than original variety Mironovskaya 808, and genotype vrn1B vrn2M vrn3B determines a lower vernalisation requirement than the original Bezostaya 1. An incomplete vernalisation requirement prolonged the time to heading, with exponential dependence on the vernalisation deficit, or prevented heading altogether. The original varieties further differed in photoperiodic sensitivity (Mironovskaya 808 sensitive, Bezostaya 1 less sensitive) that also influenced the background of substitution lines. The impact of the background on the heading time showed itself by about one week difference between Mironovskaya 808 and Bezostaya 1 grown under 8 weeks vernalisation and normal photoperiod. The difference between the lines with Mironovskaya 808 background and the lines with Bezostaya 1 background was approximately the same and was not significantly changed in different vernalisation variants of the lines. This difference may be caused by different photoperiodic sensitivity of the original varieties, but also by other genes, such as genes of earliness per se.

Notizen: Abstract The effect of plant breeding on yield and their physiological determinants has been widely studied in wheat. However, it is poorly understood how, and to what extent, yield stability has been modified. To attempt a direct analysis of changes in absolute and relative yield stability, data of yield of cultivars released in different eras in different environments were obtained from records from our lab and from the literature. Depending on the availability of data, effects of plant breeding on yield stability of cultivars released in Argentina, Australia, Italy and the United Kingdom were evaluated using a quantitative approach. In this paper it was assumed that the slope of yield vs. environmental index estimates the instability of the cultivars. In addition, a more qualitative approach for Mexico, and the former USSR complemented this analysis. There was a clear decrease in yield stability assessed in absolute terms as a consequence of wheat breeding. In Argentina, Australia, Italy and the UK this decrease was related to the magnitude of yield increases. However, the decrease in yield stability in Argentina and Australia was less than for Italy and the UK, particularly so during the last 30 years. Modern cultivars released in Argentina and Australia showed a trend to maintain yield stability as a percentage of their yield similar to that of their predecessors, while the two European countries analysed tended to a slight decrease in yield stability even in relative terms. The complementary, less quantitative evaluation of Mexico and the former USSR appeared to confirm the quantitative trends described for the other countries, i.e. a general decrease in yield stability (assessed in absolute terms) with genetic gains in yield potential.

Notizen: Abstract Chromosome arm 1RS of rye ( Secale cereale L.), when transferred to wheat ( Triticum sp.), significantly influences variety performance, because it carries genes for resistance to disease and insect pathogens. Inserted into wheat, 1RS also promotes haploid production, affects end-product quality, and sometimes affects yield. Therefore, its detection by breeders and geneticists is important. The entire 1RS arm is present in chromosome substitutions and in Robertsonian translocations involving chromosomes 1A, 1B, or 1D of wheat. In recombinant lines, a segment of 1RS has been exchanged with a segment of a group-1 wheat chromosome. Determining the wheat chromosome arm involved in a translocation, the source of rye chromatin, and the amount of 1RS chromatin introduced is necessary for a complete characterization of the introgressed segment. Biochemical, molecular, and cytogenetic technologies are described which enable such a characterization of 1RS in wheat. Examples of using gel electrophoresis, high-performance liquid chromatography, monoclonal antibodies, rye-specific molecular probes, RFLP and PCR assays, chromosome banding, in situ hybridization, and flow cytometry are provided. A comparison of these technologies is made and the advantages and disadvantages of each technology are discussed relative to modern wheat breeding efforts.

Notizen: Abstract Fifty-five spring bread wheat (Triticum aestivum L.) cultivars, mostly released between 1975 and 1991 in eight leaf rust-prone spring wheat growing regions of the former USSR, were tested in the seedling growth stage for reaction to 15 Mexican pathotypes of Puccinia recondita f. sp. tritici. In total, seven known and at least two unknown genes were identified, either singly or in combinations: Lr3 (7 cultivars), Lr10 (14), Lr13 (5), Lr14a (1), Lr16 (1), Lr23 (3); the unknown genes were identified in 14 cultivars. The first unknown gene could be either Lr9, Lr19, or Lr25; however, the second unknown gene in 9 cultivars was different from any named gene. Twelve of the 15 pathotypes are virulent for this gene, hence its use in breeding for resistance will be limited. The cultivars were also evaluated at two field locations in Mexico with two pathotypes in separate experiments. The area under the disease progress curve and the final disease rating of the cultivars indicated genetic diversity for genes conferring adult plant resistance. based on the symptoms of the leaf tip necrosis in adult plants, resistance gene Lr34 could be present in at least 20 cultivars. More than half of the cultivars carry high to moderate levels of adult plant resistance and were distributed in each region.

Notizen: Summary Studies were conducted to determine the inheritance and allelic relationships of genes controlling resistance to the Russian wheat aphid (RWA), Diuraphis noxia (Mordvilko), in seven wheat germplasm lines previously identified as resistant to RWA. The seven resistant lines were crossed to a susceptible wheat cultivar Carson, and three resistant wheats, CORWA1, PI294994 and PI243781, lines carrying the resistance genes Dn4, Dn5 and Dn6, respectively. Seedlings of the parents, F1 and F2 were screened for RWA resistance in the greenhouse by artificial infestation. Seedling reactions were evaluated 21 to 28 days after the infestation using a 1 to 9 scale. All the F1 hybrids had equal or near equal levels of resistance to the resistant parent indicating dominant gene control. Only two distinctive classes were present and no intermediate types were observed in the F2 segregation suggesting major gene actions. The resistance in PI225262 was controlled by two dominant genes. Resistance in all other lines was controlled by a single dominant gene. KS92WGRC24 appeared to have the same resistance gene as PI243781 and STARS-9302W-sib had a common allele with PI294994. The other lines had genes different from the three known genes.

Notizen: Summary Somatic embryogenesis was initiated from ‘immature embryos’ on Murashige-Skoog (MS) medium plus 2 mg.l-1 2,4-dichlorophenoxyacetic acid, 2% sucrose and 0.6% agarose. Somatic embryos were isolated and regenerated into whole green plants on MS medium devoid of 2,4-D. These regenerants were previously demonstrated to differ in their mitochondrial DNA organization. In order to estimate their characteristics three progenies of short-term culture regenerants and three progenies of long-term culture regenerants were analyzed and compared to the parental line. These somaclones obtained from the wheat variety Chinese Spring were evaluated for variation of 13 agronomic and morphological quantitative characters in comparison to the parental line. Significant variation was observed for plant height, spike length, main tiller diameter, between the somaclones regenerated from long-term culture and their parent. Differences were observed to increase with the duration of culture, leading to a significant modification of the structure of the plants. Several changes occurred during the somatic tissue cultures, but to a lesser extent than has previously been described in the literature.

Notizen: Summary Wide crosses in wheat are reviewed in relation to various factors that facilitate wide crossing to show that wide crosses can be as wide as one can make them. Included in this review is a particular reference to wheat-wheatgrass (Agropyron complex) crosses and an update on wide crosses of wheat with various genera of Agropyron complex. Hybrid seed set is too variable to predict whether a wide hybrid, where no seed was obtained in one attempt, will not be possible. High crossability genes seem to facilitate not only fertilization but also seed development, enabling embryo rescue. Variability for crossability occurs not only in wheat but also in alien species. Contrary to conventional thinking, several wide hybrids with wheat can be produced when species with lower chromosome numbers are used as female parents. Pre-and post-fertilization barriers to wide crosses do not appear to be equally strong, and can be overcome by the development and application of various technologies. Considering these aspects of wide hybridization, and based on recent successes in producing previously unsuccessful and very wide hybrids, it is concluded that how wide cross between plant species can be made is an open question and that many new and wider hybrids can be produced in future.

Notizen: Summary The end-use quality of wheat (Triticum aestivum L.) is determined in large part by the texture of the grain (soft or hard). Endosperm texture is currently determined by several empirical methods. These methods are limited because the use bulk grain lots, as opposed to individual kernels; assess phenotypic, as opposed to genotypic hardness; require a quantity of grain greater than that generally available in the early generations of wheat breeding programs, and are destructive. Recent approaches that use single kernels address the problems associated with bulk grain lots, but suffer the other limitations of providing only the phenotype and being destructive. An objective method for determining the texture genotype of single kernels of wheat was developed using starch granule-associated friabilin, a family of closely related 15 kDa proteins, as a biochemical marker. The occurrence of friabilin on water-washed wheat starch granules is apparently unaffected by the environment and is perfectly correlated (no exceptions) with grain softness. The technique presented here can detect friabilin on as little as 0.2 mg of starch and provides a 250-fold improvement in friabilin detection compared to previous methods. The method is capable of correctly assessing the genotype of F1 heterozygotes from hard x soft and soft x hard crosses. Further, the method uses only a portion of the endosperm from the kernel and therefore accommodates embryo propagation and high molecular weight glutenin subunit characterization. This single kernel method also facilitates the genetic characterization of mixed, bulk grain lots.

Notizen: Summary A glasshouse study was conducted to describe the dynamics of leaf and tiller appearance of four accessions of T. tauschii (Tt 04, Tt 17, Tt 65 and Tt 74) and to determine the influence of moisture stress (treatments were high and low moisture, imposed seven days after transplanting) on these and other aspects of development in this wild wheat. Under high moisture conditions, accessions differed greatly in flag leaf dimensions, culm length and seed number per spike, the values being lower in Tt 04 than in the other accessions. Low moisture strongly reduced values for these traits, with Tt 04 being least affected, but overall, there was no apparent association between the values obtained for these variables in the high moisture conditions and the effects of moisture stress. For three of the four accessions, final leaf number on the main culm was significantly lower in the low moisture treatment than in the respective control (P〈0.05), but the differences between treatments (ca. 0.5 leaves or less) were very small. Maximum tiller number, on the other hand, was strongly reduced by low moisture, and initiation of tillering was inhibited until water was reapplied. There were no apparent after-effects of the moisture regime on the rate of subsequent tiller appearance. The four accessions differed in their leaf appearance rates, giving phyllochron values (117–142° Cd leaf-1) within the range reported for hexaploid wheat. Low moisture tended to increase phyllochron, but in only one accession was this effect significant. Thus, depending on the accession, low moisture did not affect, or slightly decreased (by ca. 15–20%) the rate of leaf appearance. These effects were similar to those reported for cultivated wheat suggesting that there would be little scope for using these accessions of T. tauschii in breeding for stress tolerance.

Notizen: Summary Hordeum chilense is a wild barley extensively used in wide crosses in the Triticeae. It could be a valuable source of resistance to Fusarium culmorum and Septoria nodorum. Some H. chilense x Triticum spp. amphiploids, named tritordeums, were more resistant than the parental wheat line to these diseases, others were not. Average contents of ergosterol and deoxynivalenol (DON) suggested that resistance to colonization by Fusarium was the highest for Hordeum chilense, followed by tritordeum and wheat in decreasing order. In particular, the H. chilense genotypes H7 and H17 enhanced the wheat resistance to F. culmorum in its tritordeum offsprings. Resistance to S. nodorum in tritordeum was not associated with tall plant height. There is sufficient genetic variation for resistance to F. culmorum and S. nodorum among tritordeum to allow the breeding of lines combining short straw and resistance to both diseases.

Notizen: Summary The difficulties in developing inter-varietal chromosome substitution lines in wheat are reviewed. The use of genetical, cytological and molecular markers is suggested as a way of overcoming them. These difficulties and the use of markers, as well as the need to develop duplicate lines to detect background variation, are described using the development and analysis of the Cappelle-Desprez (Bezostaya 1) chromosome substitution set as an example. The effects of substituting Bezostaya 1 chromosomes on final plant height and adult-plant resistance to yellow rust are reported. The large number of aneuploids and substitution lines available in wheat provides a tremendous, international resource, which should be exploited in the future.

Notizen: Summary The group 1 and 6 inter-varietal chromosome substitution lines of Cappelle-Desprez (Bezostaya 1) were intercrossed along with the donor and recipient varieties, Cappelle-Desprez and Bezostaya 1, to give 36 genetically different families. The analysis of the means of these families showed that variation in SDS-sedimentation volume fitted a predominantly additive model. There were no significant within or between chromosome interactions among the group 1 and 6 chromosomes. Nor was there any evidence for interactions between these chromosomes and those of the background. Significant dominance/within chromosome interactions amongst the background chromosomes were however detected. Some of the positive effects on SDS-sedimentation were associated with increased grain hardness. Chromosome effects on % grain protein were not correlated with SDS-sedimentation.

Notizen: Summary The advent of molecular marker systems has made it possible to develop comparative genetic maps of the genomes of related species in the Triticeae. These maps are being applied to locate and evaluate allelic and homoeoallelic variation for major genes and quantitative trait loci within wheat, and to establish the pleiotropic effects of genes. Additionally, the known locations of genes in related species can direct searches for homoeologous variation in wheat and thus facilitate the identification of new genes. Examples of such analyses include the validation of the effects of Vrn1 on chromosome 5A on flowering time in different crosses within wheat; the indication of pleiotropic effects for stress responses by the Fr1 locus on chromosome 5A; the detection of homoeologous variation for protein content on the homoeologous Group 5 chromosomes; and the detection of a new photoperiod response gene Ppd-H1 in barley from homoeology with Ppd2 of wheat.

Notizen: Summary The chromosomal assignment of 64 PCR-amplified microsatellite loci and 29 additional fragments amplified by the same primer pairs is described for bread wheat (Triticum aestivum). The distribution over the different chromosomes and chromosome arms appears to be random. The highest proportion of microsatellite loci is found on the B genome, followed by the A and D genome. About half of the primer pairs amplified unique fragments, while the other half amplified additional fragments. 25% of the primer pairs, mostly designed to clones of a PstI-library, amplify fragments on homoeologous chromosomes. In some cases, more than one fragment on a single chromosome or fragments on non-homoeologous chromosomes occurred. The use of an automated DNA sequencer accounts for the accurate resolution of multiple fragments and enables to differentiate between fragments, amplified by a single primer pair, with size differences as small as two base pairs.

Notizen: Summary In order to obtain high levels of environmental adaptability in wheat varieties it is essential they flower at times appropriate to particular environmental conditions. The influence of three distinct genetic systems that together determine time of flowering is reviewed here. Vernalization genes are seen to be particularly important to winter wheats for their direct or indirect effects on winter hardiness. Vernalization genes play a minor role in determining flowering time in autumn sown winter wheats but insensitivity is essential if spring sown wheats are to flower. Day length sensitive photoperiod genes play a major role in determining flowering time and adaptability of autumn sown wheats. Insensitivity can promote yield advantages of over 35% in Southern European environments. 15% in Central Europe and offers benefits even in the UK. At present only a single allele of Ppd1 appears to have been introduced into commercial European wheat varieties. The merits of alternative Ppd1 alleles or different loci are discussed. The influence of earliness per se genes that determine flowering time independently of environmental stimuli is less well documented than the effect of photoperiod and vernalization genes. It is likely that genes on chromosomes belonging to groups 2, 3, 4, 6 and 7 may act to modify flowering time independently of environmental stimuli probably by determining numbers of vegetative and floral primordia being initiated or the rate of initiation of the primordia. Earliness per se genes appear to be widespread in European wheats and play a significant role in determining the exact time plants flower.

Notizen: Summary The improvement of lodging resistance by introducing major dwarfing genes, classified either as GA insensitive or GA sensitive, is one of the main strategies chosen by cereal breeders. In the present paper the current knowledge about the genetics, chromosomal localisation and the homoeoallelic relationships of the dwarfing genes in wheat and rye is reviewed. The confusing system of the symbolisation of the GA insensitive dwarfing genes/alleles in wheat is discussed and a nomenclature based on rules for gene symbolisation in wheat is proposed.

Notizen: Summary Grain size in wheat is the most stable yield component and has a favorable effect on flour yield. To identify the chromosomes associated with the large grains of line G603-86, (grain weight over 60 mg and grain length of about 9 mm), F3 lines, extracted from F2 populations obtained from F1 monosomics of crosses between G603-86 (P1) and the monosomic set of Favorit (P2) were tested in the field. ANOVA showed significant differences among parents for grain weight and grain length, but not for grain width or the factor expressing the difference in grain form and density. Homoeologous groups had significant effects on grain weight and on all components of grain weight, while genomes were not significantly different for any of these characters. Grain weight was significantly increased by chromosomes 6D and 4A of G603-86. Grain length was significantly increased by chromosomes 4A, 4B, 2B, 3A and 1B, grain width by chromosomes 1A and 1B, and the factor form-density by chromosomes 6D and 6A. The high grain size in G603-86 results from the effects of genes located on many chromosomes which affect grain dimensions, form and density.

Notizen: Summary The wheat varieties Chinese Spring, Marquis and Thatcher and five intervarietal ‘Chinese Spring’ substitution lines for chromosomes 2B (2 lines), 2D (2 lines) and 4A*, differing from the recipient variety in alleles for hybrid dwarfing genes and/or the photoperiodic response genes were analysed for tissue culture response (TCR). It could be demonstrated that only chromosome 2B has a major effect on TCR. Allelic variation at the hybrid dwarfing loci seems to have no effect on tissue culture performance, even in the combination D1D2D3 which gives the grass dwarf phenotype. Also the allelic constitution at the Ppd loci, gave no indication for a direct major effect of those alleles, however there seems to exist genetical factors for TCR on the homoeologous group 2 chromosomes which may be closely linked to the Ppd loci.

Notizen: Summary Triticum miguschovae, a genome addition synthetic, was used as a source for transfer of leaf rust (Puccinia recondita tritici) resistance to common wheat. This synthetic, developed from two wild species Triticum militinae and Aegilops squarrosa, proves a valuable donor of the genes for leaf rust resistance. Leaf rust resistance was transferred from T. miguschovae by both dominant and recessive genes. Stable lines phenotypically similar to their recurrent parents Kavkaz and Bezostaya 1 but differing from them in a high level of leaf rust resistance were obtained. The genes for resistance in 3 selected lines differed from each other and from the known effective genes Lr9, Lr19, and Lr24. The resistance of one of them (line 1229) is controlled by two complementary interacting genes located on chromosome 7B and 1D was revealed by monosomic analysis.

Notizen: Summary Wheat varieties tend to be chromosomally unstable producing on average 2–3% of plants with abnormal chromosome numbers. A number of semi dwarf wheat varieties, carrying the gibberellic acid insensitive dwarfing genes Rht1 or Rht2, have been seen to produce distinct tall off types due to reduction in dosage of the chromosome carrying the dwarfing gene. The UK variety ‘Brigand’, carrying Rht2 on chromosome 4D, produced very distinct tall off types when this chromosome was reduced in dosage. The frequency of tall off types was sufficiently high to cause the variety to fail United Kingdom statutory uniformity tests. An attempt to prevent the loss of chromosome 4D was made by constructing translocation chromosomes involving the short arm of chromosome 4D, which carries Rht2, and the long arm of chromosome 4S l from Aegilops sharonensis, which carries a gene(s) conferring preferential transmission. The work in this paper describes the field evaluation of two lines carrying 4DS.4DL-4S l L translocations, and demonstrates their success in preventing spontaneously occurring monosomy of chromosome 4D in semi-dwarf wheats.

Notizen: Summary The sequential combination of C-banding and in situ hybridization techniques applied in this or in a reverse order, are used to recognize targeted chromosomal regions in cereals. Both methods are described whereby standard chromosome squash preparations are followed by: i) C-banding technique using Leishman stain and a slightly modified in situ hybridization technique using biotin-labeled DNA probes, or ii) fluorescence in situ hybridization technique and C-banding. Both approaches have been successfully used onto mitotic chromosomes of rye and wheat resulting suitable for both their identification and detection of targeted sites.

Notizen: Summary Fluorescent in situ hybridization (FISH) of DNA to plant chromosomes has proved to be a powerful cytogenetic tool. The value of fluorescent in situ hybridization of total genomic DNA (GISH) of related species is demonstrated in the determination of wheat/alien chromosome pairing in hybrids. Its use for assessing the relative merits of the various genes that affect chromosome pairing is also shown. The ability of GISH to identify the presence in wheat of whole alien chromosomes or alien chromosome segments is illustrated. The potential of FISH for detecting repeated DNA sequences, low copy sequences and single copy genes is discussed.

Notizen: Summary Wheat breeding efforts for a half century in Japan were investigated by using of 129 varieties registered in MAFF from 1929 to 1984 in the view point of case history for three years. Varieties released in each breeding station are classified apparently by growth habit associated closely with duration of the cold requirement. Heading date is earlier in varieties released in southwest breeding stations (southern varieties) than in varieties released in northern breeding stations (northern varieties). Culm length is higher and the pre-harvest sprouting is more sensitive in northern varieties. In quality characters, northern varieties has the higher milling rate, lower apparent amylose content and larger particle size of flour. In varieties released in the Tohoku district, trend of breeding direction with time is smaller ear numbers and larger 1,000-grain weight and greater resistance to powdery mildew. In varieties released in the Kyushu district, a significant correlation with released year was clearly observed with regard to early maturity, short culm length, less grain crude protein content, less apparent amylose content and higher milling rate on breeding advancement.
