    Beschreibung / Inhaltsverzeichnis: Zusammenfassung In einem Untersuchungsgebiet am westlichen Bodensee (Halbinsel Mettnau) zeichnet sich zwischen 1976 und 1997 eine deutliche Zunahme früh ausgeflogener Teichrohrsänger ab, was auf ein früheres Brüten schließen läßt. Weiterhin hat sich die Erstankunft des Teichrohrsängers am Bodensee in den letzten 16 Jahren (1982–1997) im Vergleich zum Zeitraum 1961 bis 1980 um genau eine Woche verfrüht (im Mittel vom 26.4. auf den 19.4.). Während sich zwischen der jährlichen Erstankunft des Teichrohrsängers am Bodensee und der Zahl früh ausgeflogener Jungvögel kein statistisch gesicherter Zusammenhang finden läßt, ist die Temperatur in der ersten Maihälfte ein guter (statistisch signifikanter) Indikator für die Zahl früh ausgeflogener Teichrohrsänger. Die vorliegende Untersuchung zeigt, daß sich offenbar auch Langstreckenzieher an die sich abzeichnende Klimaveränderung anzupassen beginnen.
    Notizen: Summary Data from a standardized mist-netting programme (the so-called "MRI-Programm") were used to assess the timing of breeding in Reed Warblers (Acrocephalus scirpaceus) at Lake Constance (South Germany). The number of Reed Warblers fledged early in the season increased significantly between 1976 and 1997 (rs=0.614; p〈0.01). This is most probably due to a long-term increase in spring temperature as suggested by the high correlation between the number of early-fledged Reed Warblers and mean temperature in the first half of May (rs=0.416; p〈0.01). While in the period from 1961 to 1980 the mean date of spring arrival was on the 26th of April, it shifted towards the 19th April from 1981 to 1997. However, the number of early-fledged Reed Warblers and the date of spring arrival are not significantly correlated. The lack of a significant correlation may be due to a number of reasons, for instance early arriving birds do not belong to the local population or there could be an observational bias among years. Yet, the most likely explanation is that the timing of breeding in Reed Warblers is triggered by vegetation growth and food supply rather than by the timing of spring arrival. There is growing body of evidence from this and other studies that the recent climate change is responsible for the shift in both the timing of migration and breeding in birds.

    Notizen: Abstract Megalurothrips sjostedti is an economic pest of cowpea in tropical Africa. There are no reports of the effect of temperature on the development of the pest and no diapause has been detected. The developmental rate and reproduction of M. sjostedti was studied under six constant temperatures (14, 17, 20, 23, 26 and 29 °C ) and a photoperiod of L12:D12 in the laboratory. Total developmental time ranged from 33.1 days at 14 °C to 19.2 days at 26 °C. At 29 °C, all larvae died during hatching. Egg, larval and pupal stages required 94.3, 97.1 and 105.3 deg-days (DD) above a threshold of 8.2, 9.1 and 10.4 °C, respectively, to complete development. Total developmental cycle was completed at 163.9 DD above a threshold of 12.6 °C. Adult longevity was lowest (11.1 days) at 29 °C and highest (28.7 days) at 14 °C. At constant temperatures, the highest pre-oviposition period was observed at 29 °C under a photoperiod of L16:D8. Egg production also ceased at this temperature/photoperiod combination. Total fecundity was highest at 26 °C under L10:D14. At cyclical temperatures, pre-oviposition period was longest (12.4 days) at 20/29 °C under L16:D8. Total fecundity was highest (168.4 eggs) at 14/26 °C under L10:D14, and lowest (8.2 eggs) at 20/29 °C under L16:D8. The significance of these findings in the management of M. sjostedti is discussed.

    Notizen: Abstract Life table parameters of Aphis gossypii Glover (Homoptera: Aphididae) on Gossypium hirsutum L. were determined at six temperatures (10, 15, 20, 25, 30, and 35 ± 0.5 °C) in the laboratory. Relationships of life table parameters with temperature were described with mathematical equations. Development was fastest at 30 °C, with a pre-larviposition period of 4.6 d. Survival to adult was greatest at 25 °C (81%). Fecundity was highest at 25 °C, with a total fecundity of 28.3 nymphs per female and a mean reproductive rate of 3.1 nymphs per female per day. Threshold temperatures for development in the first through fourth instar and the adult were 8.2, 8.0, 7.2, 6.2 and 7.9 °C, respectively. The durations of these stages, expressed as temperature sums above these thresholds, were 24.2, 23.7, 23.0, 25.5 and 168.8 degree-days (D°), respectively. A. gossypii achieved its maximum net reproductive number (24.4 nymphs per female) and greatest intrinsic rate of increase (0.386 d−1) at 25 °C. The high relative rate of population increase at 25 °C results in a daily population increase of 47% and a doubling time of only 1.8 d, illustrating the tremendous growth capacity of A. gossypii populations under favourable conditions. Compared to literature sources, our source of A. gossypii, fed on cotton, showed a comparatively great heat tolerance.

    Notizen: Abstract The Russian wheat aphid, Diuraphis noxia (Mordvilko), is one of the most important aphid pests of wheat, Triticum aestivum L., worldwide. Among the various pest management options, plant resistance is an economical management tactic to control D. noxia in cereal crops such as wheat. Researchers have identified D. noxia resistant germplasm and it has been incorporated into wheat. This study compared D. noxia resistance between the ‘Betta’ wheat isolines Betta-Dn1, Betta-Dn2, and Betta-Dn5 and their corresponding donor gene plant introduction (PI) lines PI 137739 (Dn1), PI 262660 (Dn2), and PI 294994 (Dn5). Although the Betta isolines and PI lines showed D. noxia resistance when compared with Betta wheat, the degree of resistance in the isolines to D. noxia was different from their corresponding PI donors. Aphid number, aphid fecundity, and biomass per aphid were not different between Betta-Dn1 and PI 137739 or Betta-Dn2 and PI 262660; however, the same parameters were significantly lower on PI 294994 compared with Betta-Dn5. This indicated that aphid resistance in PI 137739 and PI 262660 was probably governed by a single dominant gene, while the resistance in PI 294994 was controlled by more than one gene. Additionally, plant biomass reduction was aphid density dependent, which suggested that use of appropriate aphid infestation level is important when using plant biomass reduction as an indicator of resistance. Plant resistance categorization showed that there was no detectable difference in antixenosis among the seven lines evaluated. However, the higher aphid fecundity observed on PI 262660 compared with PI 137739 and PI 294994, in addition to no significant differences among the three PIs in plant biomass reduction, suggested PI 262660 was a tolerant line, while PI 137739 and PI 294994 were antibiotic lines. Plant tolerance could not be elucidated among the three Betta isolines using aphid fecundity and plant biomass reduction as indicators.

    Notizen: Abstract The smaller fruit tortrix, Grapholita lobarzewskii Nowicki, has become a major pest in the recent past in apple orchards north of the Alps. Nevertheless little is known about the biology and the behaviour of this species. The effect of temperature on post-diapause development and survival was studied under controlled conditions in order to establish a basis for the forecasting of adult emergence. Survival was found to be highest at 17 °C and lowest at 11.8 °C. The sex ratio did not differ significantly from 1:1. Based on a linear relationship between temperature and developmental rates, thermal thresholds of 9.6 °C and 9.8 °C were determined for females and males respectively. The thermal constants, i.e., the mean developmental times in physiological time units, were found to be 342 day-degrees for females and 317 day-degrees for males, suggesting a slight protandry. Based on the mean and the variance of the developmental times, a simple temperature-driven phenology model was built using a time-varying distributed delay. The model was validated by visually comparing the predictions with independent observations on adult emergence, and by calculating the temporal deviations of the predictions. In 4 out of 5 years the mean error was less than 3 days. The model was therefore found to give reliable forecasts of the emergence of G. lobarzewskii and can be used to determine the optimal time for the exposure of pheromone traps, the application of pheromone dispensers for mating disruption, and for the timing of insecticide applications.

    Notizen: Abstract Extended sieve element salivation (E1 waveform in the electrical penetration graph) is a characteristic activity during early sieve element punctures, particularly in resistant plants. In order to explore a chemically-mediated mechanism of resistance associated with sieve element salivation, we compared the pattern of feeding behaviour of the aphid, Sitobion fragariae (Walker), on two cultivars of the wheat Triticum aestivum L., with different concentrations of hydroxamic acids (Hx). During 24 h of electronic monitoring, aphids dedicated over 50% of the total time to phloem ingestion from the sieve elements. Total time allocated to E1 in the experiment, time to first E1 within the experiment, time allocated to E1 before a sustained phloem ingestion (E2) and the contribution of sieve element salivation to the phloem phase (E1/[E1+E2]) were significantly higher in the high-Hx cultivar. The increased salivation in plants with higher contents of Hx suggests the existence, at least in this system, of a chemically-mediated sieve element constraint.

    Notizen: Abstract A respirometric system was used to analyze the biodegradation of high molecular weight (120,000 to 200,000 g mol−1) polylactic acid (PLA) plastic films in soil under laboratory conditions. The respirometric system consisted of air-conditioning pretraps, a soil reactor, and a carbon dioxide (CO2) posttrap. A 200-g homogeneous soil mixture of all-purpose potting soil : manure soil : sand [1 : 1 : 1 (w/w)] and 1.5 g of PLA plastic films in 1 × 1-cm2 squares was added to each bottle. The respirometers were placed in a 28, 40, or 55°C water bath for 182 days. Treatments (three replicates) included native corn starch (positive control), polyethylene (Glad Cling Wrap; negative control), and three PLA films: Ca-I (Cargill Dow Polymers LLC, monolayer), GII (Cargill Dow Polymers LLC, Generation II), and Ch-I (Chronopol; monolayer). The degree of polymer mineralization was indicated by the cumulative CO2 liberated from each respirometer. The initial average mineralization rate and total percentage mineralized of the PLA plastic films at 28, 40, and 55°C was 24.3, 41.5, and 76.9 mg/day with a 27, 45, and 70% carbon loss, respectively. No decrease in soil pH was observed after 182 days of mineralization. Hence, increase in soil temperature drastically enhanced the biodegradation of PLA plastic films in soil under laboratory conditions (P 〈 0.0001).

    Notizen: Abstract Three high molecular weight (120,000 to 200,000 g mol−1) polylactic acid (PLA) plastic films from Chronopol (Ch-I) and Cargill Dow Polymers (GII and Ca-I) were analyzed for their degradation under various temperature and relative humidity (RH) conditions. Two sets of plastic films, each containing 11 samples, were randomly hung in a temperature/humidity-controlled chamber by means of plastic-coated paper clips. The tested conditions were 28, 40, and 55°C at 50 and 100% RH, respectively, and 55°C at 10% RH. The three tested PLA films started to lose their tensile properties when their weight-average molecular weight (M w) was in the range of 50,000 to 75,000 g mol−1. The average degradation rate of Ch-I, GII, and Ca-I was 28,931, 27,361, and 63,025 M w/week, respectively. Hence, GII had a faster degradation rate than Ch-I and Ca-I under all tested conditions. The degradation rate of PLA plastics was enhanced by the increase in temperature and relative humidity. This trend was observed in all three PLA plastics (Ca-I, GII, and Ch-I). Of the three tested films, Ch-I was the first to lose its mechanical properties, whereas Ca-I demonstrated the slowest loss, with mechanical properties under all tested conditions.

    Notizen: Abstract Electrosynthesis of cupric oxide powder was carried out on a laboratory scale in an electrochemical cell under various experimental conditions. The electrolysis was appraised in terms of the particle size of the cupric oxide product and the anodic dissolution of the copper electrode. Using a previously determined pH value of 7.50, the other optimum electrolysis operating conditions established were a current density of 4000Am−2, temperature of 353K, and Na2SO4 concentration of 0.5m. The optimum values of current efficiency, cell voltage and specific energy consumption for the electrochemical synthesis of cupric oxide powder were determined.

A classification scheme is introduced and presented in a tabular form called TEA (Table of Evolutionary Algorithms). It distinguishes between different classes of evolutionary algorithms (e.g., genetic algorithms, ant systems) by enumerating the fundamental ingredients of each of these algorithms.     Notizen: Abstract The ADP-dependent phosphofructokinase (PFK) from Thermococcus zilligii has been purified 950 fold; it had a specific activity of 190 U mg−1. The enzyme required Mg2+ ions for optimal activity and was specific for ADP. The forward reaction kinetics were hyperbolic for both cosubstrates (pH optimum of 6.4), and the apparent K m values for ADP and fructose-6-phosphate were 0.6 mM (apparent V max of 243 U mg−1) and 1.47 mM (apparent V max of 197 U mg−1), respectively. Significantly, the enzyme is indicated to be nonallosteric but was slightly activated by some monovalent cations including Na+ and K+. The protein had a subunit size of 42.2 kDa and an estimated native molecular weight of 66 kDa (gel filtration). Maximal reaction rates for the reverse reaction were attained at pH 7.5–8.0, and the apparent K m values for fructose-1,6-bisphosphate and AMP were 0.56 mM (apparent V max of 2.9 U mg−1) and 12.5 mM, respectively. The biochemical characteristics of this unique ADP-dependent enzymatic activity are compared to ATP and pyrophosphate-dependent phosphofructokinases.

    Notizen: Abstract A protease of a molecular mass of approximately 30 kDa was isolated and purified from the haloalkaliphilic archaeon Natronomonas (formerly Natronobacterium) pharaonis. The enzyme hydrolyzed synthetic peptides, preferentially at the carboxyl terminus of phenylalanine or leucine, as well as large proteins. Hydrolysis occurred over the range of pH from 6 to 12, with an optimum at pH 10. The temperature optimum was 61°C. The enzyme was nearly equally active over the range of salt concentration from 0.5 to 4 M (NaCl or KCl). A strong cross-reaction with a polyclonal antiserum against human chymotrypsin was observed. Enzymatic activity was inhibited by typical serine protease inhibitors. There was significant homology between N-terminal and internal sequences from autolytic fragments and the sequence of bovine chymotrypsinogen B; the overall amino acid composition was similar to that of vertebrate chymotrypsinogens. Evidence for a zymogen-like processing of the protease was obtained. Cell extracts from other halobacteria exhibited similar proteolytic activity and immunoreactivity. The data suggested a widespread distribution of a chymotrypsinogen B-like protease among halo- and haloalkaliphilic Archaea.

    Notizen: Abstract  The mouse Lsh/Ity/Bcg locus regulates natural resistance to intracellular pathogens, and the Nramp1 gene was isolated as its candidate. Nramp is part of a small family of at least two genes, Nramp1 and Nramp2. In the present study, a full-length cDNA for carp NRAMP has been isolated and characterized. Nucleotide and predicted amino acid sequence analysis indicate that the carp NRAMP encodes a 548 amino acid membrane protein with 12 putative transmembrane domains, two N-linked glycosylation sites, and an evolutionarily conserved consensus transport motif. The peptide sequence identity among carp and human NRAMP2 is 78%, and 65% with human NRAMP1. Reverse transcription-polymerase chain reaction revealed that carp NRAMP is ubiquitously expressed. Phylogenetic analysis, using neigbor-joining, showed that the carp NRAMP protein clustered together with mammalian NRAMP2 proteins.

    Notizen: Abstract  The receptor of a T lymphocyte (TCR) recognizes nonself antigens in the company of major histocompatibility complex (MHC) molecules presented to it by the antigen-presenting cell. The variable region of TCR is encoded by either a concatenation of variable region (TCR-V), diversity region (TCR-D), and joining region (TCR-J) genes, or a concatenation of TCR-V and TCR-J genes. The TCR-V genes exist as a multigene family in vertebrate species. Here we study the evolutionary relationships of TCR-V genes from humans, sheep, cattle, rabbits, mice, and chicken. These six species can be classified into two groups according to the frequency of γδ T-cells in their peripheral T-cell populations. The "γδ low" group of species includes humans and mice, in which γδ T-cells constitute very limited portion of the T-cell population. The "γδ high" group includes sheep, cattle, rabbits, and chicken, in which γδ T-cells comprise up to 60% of the T-cell population. Here, we compiled TCR-V sequences from the six species and conducted a phylogenetic analysis. We identified various TCR-V gene subgroups based on the analysis. We found that humans and mice have representatives from nearly all of the subgroups identified, while other species have lost subgroups to different extent. Therefore, the γδ low species have a high degree of diversity of TCR-V genes, while γδ high species all have limited diversity of TCR-V genes. This pattern is similar to that found for immunoglobulin variable region (IGV) genes.

    Notizen: Abstract  A full-length and several partial cDNAs encoding IGK light chains from the marsupial South American opossum, Monodelphis domestica, were isolated and characterized. Using these clones as a starting point, the expressed IGKV repertoire was sampled by anchored polymerase chain reaction using an IGKC-specific primer. Based on nucleotide sequences of twenty unique, expressed IGKV-J combinations, there are at least four IGKV families and two J segments. Southern blot analysis revealed each IGK-V family contains multiple gene segments totaling at least thirty-five IGKV in the opossum genome. No evidence for particular, recurrent IGKV-J combinations in the opossum IGK repertoire was seen, rather the V-J combinations appeared random and diverse. Each of the four IGKV families appear more closely related to V segments from placental mammals than to each other, suggesting the duplication of the IGKV families prior to the separation of marsupials and placental mammals more than one-hundred-million years ago. Overall, the complexity of opossum light chain V segments appears greater than that found in the heavy chain, and light chains are likely to contribute significantly to Ig diversity in this species.With this report, the homologues encoding all three classes of eutherian Ig chains, IGH, IGL, and IGK, have been described in a non-placental mammal.

    Notizen: Abstract  Proper peptide presentation by major histocompatibility complex (MHC)-encoded class II antigens is dependent on the products of the MHC DM loci. We identified the rabbit orthologues (RLA-DMA and -DMB) of human HLA-DMA and -DMB and found that they have 76.9% and 78.8% identity with HLA-DMA and -DMB, respectively. Like classical class II MHC genes, RLA-DM genes are more closely related to human HLA-DM genes than to mouse H2-DM. Among the DM family, there is a high degree of variability at the amino terminus of the DMa chains, and length variability in the cytoplasmic tails of both DMα and DMβ. The rabbit DM genes are coexpressed with class II genes in lymphoid tissues, as are the DM genes of other mammals. The RLA-DM locus maps to the class II region of the rabbit MHC, and is flanked by the DP and DOB loci. Despite having some similarities to class II genes of bony fishes, the DM family represents a separate branch of the MHC class II family.

    Notizen: Abstract The star-nosed mole (Condylura cristata) has a snout surrounded by 22 fleshy and mobile appendages. This unusual structure is not an olfactory organ, as might be assumed from its location, nor is it used to manipulate objects as might be guessed from its appearance. Rather, the star is devoted to the sense of touch, and for this purpose the appendages are covered with thousands of small mechanoreceptive Eimer's organs. Recent behavioral studies find that the star acts much like a tactile eye, having a small behavioral focus, or “fovea” at the center – used for detailed explorations of objects of interest. The peripheral and central nervous systems of the mole reflect these behavioral specializations, such that the small behavioral focus on the nose is more densely innervated in the periphery, and has a greatly enlarged representation in the somatosensory cortex. This somatosensory representation of the tactile fovea is not correlated with anatomical parameters (innervation density) as found in other species, but rather is highly correlated with patterns of behavior. The many surprising parallels between the somatosensory system of the mole, and the visual systems of other mammals, suggest a convergent and perhaps common organization for highly developed sensory systems.

    Notizen: Abstract Peripheral auditory frequency tuning in the ensiferan insect Cyphoderris monstrosa (Orthoptera: Haglidae) was examined by comparing tympanal vibrations and primary auditory receptor responses. In this species there is a mis-match between the frequency of maximal auditory sensitivity and the frequency content of the species' acoustic signals. The mis-match is not a function of the mechanical properties of the tympanum, but is evident at the level of primary receptors. There are two classes of primary receptors: low-tuned and broadly tuned. Differences in the absolute sensitivity of the two receptor types at the male song frequency would allow the auditory system to discriminate intraspecific signals from sounds containing lower frequencies. Comparisons of tympanal and receptor tuning indicated that the sensitivity of the broadly tuned receptors did not differ from that of the tympanum, while low-tuned receptors had significantly narrower frequency tuning. The results suggest that the limited specialization for the encoding of intraspecific signals in the auditory system of C. monstrosa is a primitive rather than a degenerate condition. The limited specialization of C. monstrosa may reflect the evolutionary origin of communication-related hearing from a generalized precursor through the addition of peripheral adaptations (tympana, additional receptors) to enhance frequency sensitivity and discrimination.

    Notizen: Abstract The unusual morphology of a sound-activated plurisegmental ascending interneurone (AN5-AG7) in an insect (Ancistrura nigrovittata, Ensifera, Phaneropteridae) is described. This neurone's soma is located in the penultimate abdominal ganglion. The most prominent arborisations with smooth endings are found in the prothoracic ganglion. The neurone terminates with numerous beaded endings in the brain (protocerebrum). All abdominal ganglia including the penultimate contain only tiny side branches of beaded appearance. The neurone's morphology is compared to the morphology of a `typical' sound-activated plurisegmental neurone of bushcrickets with its soma in the prothorax. In the prothoracic ganglion and in the brain the arborisations of the two cells are very similar. Graded potentials and action potentials are generated in the prothoracic portion of both neurones. Both receive excitation mainly by ultrasound, and inhibition by soma-ipsilateral stimuli. Neither wind, substrate vibration nor touch of the abdomen evoke responses in AN5-AG7. It is assumed that early in evolution this neurone had its dendrites in the ganglion which houses the cell body (like cercal interneurones of this neuromere). Profound evolutionary changes probably have taken place to bring about this neuron's modern morphology.

    Notizen: Abstract Male moths responding to their species-specific sex pheromone, may cease their upwind flight when pheromone components of sympatric species are added to the mixture. The interspecific interaction between the pheromone response of the tortricid moths Cydia pomonella and Adoxophyes orana was investigated in field-trapping and wind-tunnel studies. Addition of the A. orana pheromone [(Z9)-tetradecenylacetate and (Z11)-tetradecenylacetate] to a source containing the C. pomonella pheromone [(E8, E10)-dodecadienol] resulted in a significant inhibition of attraction by male C. pomonella. It is demonstrated that this behavioural antagonist for C. pomonella must be emitted from the same point source to induce this inhibitory effect. A spatial separation of the two interspecific pheromones (at 14 cm, 5 cm and 0.5 cm crosswind) restored the attraction of the conspecific pheromone for male C. pomonella. In contrast to C. pomonella, male A. orana were not inhibited by point sources releasing both the C. pomonella and A. orana pheromone. We suggest that the discrepancy in the interspecific pheromone interaction between these two tortricids can be explained if we consider the evolutionary ecology of interspecific pheromone communication in C. pomonella and A. orana.

    Notizen: Abstract Wheat (cv Chinese Spring) tissues were transformed using Agrobacterium tumefasciens and a new plasmid modular vector, pMVTBP. We constructed pMVTBP with unique restriction sites connecting (1) the CaMV 35S promoter, (2) a Kozak sequence, (3) the FLAG epitope, (4) the (His)6 epitope, (5) a coding region (for wheat TATA Binding Protein, wTBP) and (6) the CaMV 35S 3′UTR. This vector thus allows easy exchange of different regulatory or coding sequences. Explants of either germinating mature seeds, or immature embryos, were induced to callus for up to two weeks, treated with virulence-induced bacteria for one hour, then regenerated into plantlets. Transient expression of a GUS reporter gene, assayed at about one week, occurred in 10–12% of calluses. Expression of the FLAG-tagged wTBP was also detected, by immunostaining. Stable expression, by selective growth on geneticin, and by GUS expression at about six weeks, occurred in 1–2% of calluses, quite comparable to that achieved by other methods.

    Notizen: Abstract The seasonal sedimentation pattern of diatom valves in Lake Holzmaar was investigated during 1995 by deploying sediment traps at three different lake depths. According to the sedimentation pattern, the major reproduction zone of diatoms was restricted to the upper 6 m of the water body. The population growth started late in April and blooms of Cyclotella cf. comensis Grun., which dominates the plankton diatoms, and Fragilaria crotonensis Kitton were collected in traps during June and September, and July, respectively. During summer, the seasonal sedimentation pattern of each taxon, as collected in the upper traps, was reflected in the concentrations in the lowest trap. However, in May and from September onwards, the community composition in the lowest trap and augmented trapping rates suggest both sediment focusing and resuspension of bottom sediments. The temperature signals as recorded by δ18O values of diatom valves should, therefore, reflect integrated temperatures between 0 and 6 m depth. However, temperatures during summer and autumn are expected to be accentuated in the sedimentary record since the isotopic signal is weighted by both the number and the weight-mass of the valves. During summer, the transfer of these signals by the sedimenting diatoms retains the information pattern recorded, while in spring and autumn/winter additional influxes caused by resuspension may somewhat alter those temperature informations. The proxy signals finally stored in the sediments, may, therefore, not precisely represent the successive temperatures currently recorded during 1995 within mid-lake.

    Notizen: Abstract The effect of water availability (water activity,aw; 0.995–0.90) and temperature (18–30 °) on in vitro interactions between an ochratoxin producing strain of Aspergillus ochraceus and six other spoilage fungi was assessed in dual culture experiments on a maize meal-based agar medium. Inprimary resource capture of nutrient substrate, A. ochraceus was dominant against many of the interacting species, being able to overgrow and replace A. candidus, and sometimes A. flavus and the Eurotium spp. regardless of aw or temperature. However, with freely available water (0.995 aw) A. alternata and A. niger were dominant, with mutual antagonism between A. ochraceus and A. flavus at 25–30 °C. In the driest conditions tested (0.90 aw) there was also mutual antagonism between A. ochraceus and the two Eurotium spp. Overall, under allconditions tested the Index of Dominance for A. ochraceus was much higher than for other competing species combined suggesting that A. ochraceus wasa good competitive colonist able to replace a numberof other species. However, the growth rate ofA. ochraceus was modified and decreased by the interaction with competitors. Interaction between A. ochraceus and species such as A. alternata (18°C/0.995) and Eurotium spp. (0.995–0.95 and 25–30 °C) resulted in a significant stimulation of ochratoxin production. Theresults are discussed in relation to the effect that environmental factors have on the possible competitiveness of A. ochraceus in the maizegrain ecosystem and the role of ochratoxin in nicheexclusion of competitors.

    Notizen: Abstract Seven chemical extractants were tested for their relative performance to predict the response of wheat to Mn application in coarse textured alkaline soils of semi-arid region. Five out of the seven extractants were found to be promising for the estimation of critical level of available Mn in these soils, as the amount of Mn extracted by these extractants was positively and significantly correlated with relative grain yield as well as Mn uptake. The critical deficiency level of soil available Mn with 0.005 M DTPA, 0.02% hydroquinone, 0.02 N sodium pyrophosphate, 0.1N H3PO4 and 0.05N HCl+0.025N H2SO4 was 3.1, 13.8, 23.5, 5.3 and 17.8 mg kg-1 soil, respectively. The 1N ammonium acetate and 0.01M CaCl2 were found to be unsuitable extractants for these soils. Further field trials at eight locations with varying levels of Mn deficiency showed successive increase in the grain yield of wheat with foliar Mn application, emphasizing the need for Mn fertilization when wheat is grown on Mn deficient soils.

    Notizen: Abstract In apple orchards in northern Greece, females of Panonychus ulmi Koch were found to lay diapause eggs from late August to the beginning of October. The course of diapause termination in the field was determined by transferring diapause eggs during winter and early spring from apple orchards with the varieties ‘Starkinson’ and ‘Firiki’ to short days (LD 8:16) (1992–1996), and long days (LD 16:8) (1994–1995), both at 20 °C. Percentages of diapause termination were very low to zero from October to the beginning of January, then progressively increased throughout January and February. Diapause termination in 50% of the eggs occurred in the first half of February in lowland mite populations irrespective of the year and location from which the eggs originated, and about one month earlier in a population originating from an altitude of 300 m. For each sampling date throughout the winter, the mean number of days required for 50% egg hatch at 20 °C (T50%) was similar under either a long (LD 16:8) or a short (LD 8:16) photoperiod. Diapause eggs collected in October 1995 from two orchards and maintained at 0, 5, 10 and 15 °C for various periods were subsequently transferred to 20 °C and LD 8:16, where TP50% was determined. It was shown that temperature, duration of maintenance at the different temperatures and the orchard from which the eggs originated had a significant effect on T50% and therefore on diapause development. Additionally, in our strains diapause intensity was much weaker than in strains from more northern latitudes and was terminated even without any cold exposure. The variation in diapause intensity in different strains of P. ulmi may have an adaptive significance for this widespread species.

    Notizen: Abstract The influence of prey density, within-field vegetation, and the composition and patchiness of the surrounding landscape on the abundance of insect predators of cereal aphids was studied in wheat fields in eastern South Dakota, USA. Cereal aphids, aphid predators, and within-field vegetation were sampled in 104 fields over a three year period (1988–1990). The composition and patchiness of the landscape surrounding each field were determined from high altitude aerial photographs. Five landscape variables, aggregated at three spatial scales ranging from 2.6 km2 to 581 km2, were measured from aerial photographs. Regression models incorporating within-field and landscape variables accounted for 27–49% of the variance in aphid predator abundance in wheat fields. Aphid predator species richness and species diversity were also related to within-field and landscape variables. Some predators were strongly influenced by variability in the composition and patchiness of the landscape surrounding a field at a particular spatial scale while others responded to variability at all scales. Overall, predator abundance, species richness, and species diversity increased with increasing vegetational diversity in wheat fields and with increasing amounts of non-cultivated lands and increasing patchiness in the surrounding landscape.

    Notizen: Abstract Management of stored-grain insect pests by farmers or elevator managers should be based upon a knowledge of the grain storage environment and the ecology of insect pests. Grain storage facilities and practices, geographical location, government policies, and marketing demands for grain quality are discussed as factors influencing stored-grain insect pest management decisions in the United States. Typical practices include a small number of grain samples designed to provide grain quality information for segregation, blending and marketing. This low sampling rate results in subjective evaluation and inconsistent penalties for insect-related quality factors. Information on the efficacy of insect pest management practices in the United States, mainly for farm-stored wheat, is discussed, and stored-grain integrated pest management (IPM) is compared to field-crop IPM. The transition from traditional stored-grain insect pest control to IPM will require greater emphasis on sampling to estimate insect densities, the development of sound economic thresholds and decision-making strategies, more selective use of pesticides, and greater use of nonchemical methods such as aeration. New developments in insect monitoring, predictive computer models, grain cooling by aeration, biological control, and fumigation are reviewed, their potential for improving insect pest management is discussed, and future research needs are examined.

The historical lore of most fields of scientific endeavor preserve these reputations, often through the identification of founders, innovators, and prolific workers whose contributions are considered fundamental to progress in the field. Historians frequently rely on the historical lore of scientists to guide their studies of the development of ideas, exhibiting justifiable caution in reassessing reputations in the light of current knowledge. However, the transmission of historical lore can obscure the relative importance of accomplishment, influence and personality in shaping contemporary reputations, leaving the historian to either accept reputations at face value or attempt to reconstruct the context in which they were created. The science of taxonomy, because of its rules of priority, leaves a relatively accurate record of historical accomplishment through the persistence of taxa in catalogues and faunal guides. These records allow the modern historian an unbiased means to assess the relative accomplishments of historical figures and therefore a means to critically reassess reputations independent of personality and influence. In the historical lore of North American ichthyology, Louis Agassiz at Harvard and Spencer Baird at the Smithsonian emerge as central figures in the early development of the field during the mid-1800s, contributing not only through the quality and quantity of their science, but also through their roles as institutional leaders and mentors to workers who followed. Charles Girard, originally a student of Agassiz's and later a coworker with Baird, receives little notice in the history of ichthyology, and his reputation is that of a minor player in the initial description of the North American fish fauna, and one whose work appears to have been flawed or even careless when compared to his contemporaries. However, a review of both contemporary and modern taxonomic works reveals that Girard's productivity far exceeded that of either Agassiz or Baird. Furthermore, an examination of the tendency of Girard and his contemporaries to introduce synonymous names into the literature, which might reflect careless or uncritical work, suggests that Girard was among the more accomplished workers of hisera, including Agassiz and Baird. Girard's low ranking in the folklore of North American ichthyology, therefore, can not be attributed to discernible shortcomings in his scientific work, but rather to a public and private campaign of criticism waged by Agassiz after Girard's departure from Harvard. While Agassiz's dispute with Girard stemmed from their personal interactions, he expressed them as criticisms of Girard's work, and thus helped shape Girard's scientific reputation as it has been transmitted through the lore of ichthyology. This case study reveals how scientific reputation may not always rest on accomplishment, but can be influenced by personal interactions obscured by time but nonetheless important to history.

    Notizen: Abstract  Analysis and hydrogeochemical modeling of hydrocarbonate groundwater, including its buffering geochemical properties, have demonstrated that this water has undergone a geochemical transformation almost throughout the whole of one industrial region. It is known that hydrocarbonate groundwater possesses a high protective natural geochemical potential, supporting neutralization of acid atmospheric precipitation. Natural hydrocarbonate water undergoes three stages of anthropogenic transformation caused by acid atmospheric precipitation over more than 50 years. The first stage is transformation of neutral hydrocarbonate water into alkaline water, accompanied by calcite precipitation and sodium carbonate dissolution from the unsaturated zone. The second stage is transformation of alkaline water into neutral hydrocarbonate water; the hydrocarbonate content, being less than the lower limit for background concentrations, showed reduced water buffering properties or protective potential relative to acid precipitation. The third stage is transformation of neutral hydrocarbonate water into acidic water, with a complete loss of protective geochemical potential. This unfavorable ecological situation with natural geochemically hydrocarbonate groundwater shows that natural groundwater less protected from the impact of acid atmospheric precipitation is in a worse ecological condition, which is confirmed by observations in northern and Western Europe and other regions of the world.

They are characterized by six invariant amino acid residues in the homeodomain, while the residue at position 50 can be a serine, glutamine or lysine as in the Pax-type, Q50 Paired-like or the K50 Paired-like homeodomains respectively. Genes in this class emerged early in animal evolution: three distinct Pax genes and two Q50 Paired-like genes have recently been characterised from cnidarians. Phylogenetic molecular reconstructions taking into account homeodomain and paired-domain sequences provide some new perspectives on the evolution of the Paired-class genes. Analysis of 146 Paired-class homeodomains from a wide range of metazoan taxa allowed us to identify 18 families among the three sub-classes from which the aristaless family displays the least diverged position. Both Pax-type and K50 families branch within the Q50 Paired-like sequences implying that these are the most ancestral. Consequently, most Pax genes arose from a Paired-like ancestor, via fusion of a Paired-like homebox gene with a gene encoding only a paired domain; the Cnidaria appear to contain genes representing the ’before’ and ’after’ fusion events.

    Notizen: Abstract  Hairless is a member of the Notch signalling pathway, where it acts as antagonist by binding to Suppressor of Hairless [Su(H)], thereby inhibiting Notch target gene activation. The pathway and its members are highly conserved in metazoans from worms to humans. However, a Hairless orthologue from another species has not yet been identified. The identification of Hairless in largely diverged species by cross-hybridization has failed so far probably due to a low degree of conservation. Therefore, we turned to D. hydei where a Hairless mutation has been described before. The D. hydei Hairless orthologue is reasonably well conserved with regard to gene structure and expression. The prospective Hairless protein orthologues share several highly conserved regions which are separated by quite diverged stretches. As to be expected, the largest region of high conservation corresponds to the Su(H) binding domain. This region is also functionally conserved, since this D. hydei protein domain binds very strongly to the D. melanogaster Su(H) protein. The other conserved regions support our earlier structure-function analysis since they nicely correspond to previously defined, functionally important protein domains. Most notably, the very C-terminal domain which is very sensitive to structural alterations, is nearly identical between the two species. In summary, this evolutionary study improves the knowledge on functionally significant domains of the Hairless protein, and may be helpful for the future identification of homologues in other animals, especially in vertebrates.

    Notizen: Abstract  Evolutionary change in developmental mode in sea urchins is closely tied to an increase in maternal provisioning. We examined the oogenic modifications involved in production of a large egg by comparison of oogenesis in congeneric sea urchins with markedly different sized oocytes and divergent modes of development. Heliocidaris tuberculata has small eggs (95 µm diameter) and the ancestral mode of development through feeding larvae, whereas H. erythrogramma has large eggs (430 µm diameter) and highly modified non-feeding lecithotrophic larvae. Production of a large egg in H. erythrogramma involved both conserved and divergent mechanisms. The pattern and level of vitellogenin gene expression is similar in the two species. Vitellogenin processing is also similar with the gonads of both species incorporating yolk protein from coelomic and hemal stores into nutritive cells with subsequent transfer of this protein into yolk granules in the developing vitellogenic oocyte. Immunocytology of the eggs of both Heliocidaris species indicates they incorporate similar levels of yolk protein. However, H. erythrogramma has evolved a highly divergent second phase of oogenesis characterised by massive deposition of non-vitellogenic material including additional maternal protein and lipid. Maternal provisioning in H. erythrogramma exhibits recapitulation of the ancestral vitellogenic program followed by a novel oogenic phase with hypertrophy of the lipogenic program being a major contributor to the increase in egg size.

    Notizen: Abstract  The amphibian Xenopus laevis is one non-mammalian vertebrate in which the major histocompatibility complex (MHC) has been analyzed extensively. Class IIβ, class Ia, LMP2, LMP7, HSP70, C4, Factor B, and Ring3 genes have been identified and mapped to the MHC. Here, we report the isolation of a transporter associated with antigen processing (TAP) gene, TAP2, and demonstrate its linkage to the MHC. While the ATP-binding region of Xenopus TAP2 is highly conserved in evolution, amino acid identity to other vertebrate TAP proteins was not detected in the N-terminal region. Segregation analysis of 34 individuals from two families showed exact restriction fragment length polymorphism matching between the MHC class Ia gene and the one TAP2 gene demonstrating linkage conservation since the mammalian/amphibian divergence ∼350 million years ago. In addition, one non-MHC-linked TAP2–hybridizing fragment was detected in approximately half of the individuals tested. Interestingly, TAP2 allelic lineages appear to match those of LMP7 and classical class I, which previously were categorized into two highly divergent groups that emerged at least 60 million years ago. Similar to LMP7 and class Ia,TAP2 is expressed ubiquitously with highest levels in intestine and spleen.

The human genes encoding these activators (PSME1 and PSME2, respectively) were analyzed by sequencing. Each gene comprised 11 exons, consistent with gene duplication during vertebrate evolution. The intron/exon organization of both genes was highly conserved, the major difference being the absence of the exon encoding the lysine and glutamic acid-rich 'KEKE' motif in PA28β. Two other genes of relevance to the immune system were located close to those for PA28 at 14q11.2 including ISGF3G, a protein involved in transcription after IFNα signalling. These sequences were also characterized.

    Notizen: Abstract  Analysis of cattle major histocompatibility complex (MHC) (BoLA) class I gene expression using serological and biochemical methods has demonstrated a high level of polymorphism. However, analysis of class I cDNA sequences has failed to produce conclusive evidence concerning the number and nature of expressed genes. Such information is essential for detailed studies of cattle immune responses, and to increase our understanding of the mechanisms of MHC evolution. In this study a selective breeding programme has been used to generate a number of MHC homozygous cattle expressing common serologically defined class I specificities. Detailed analysis of five class I haplotypes was carried out, with transcribed class I genes identified and characterized by cDNA cloning, sequence analysis, and transfection/expression studies. Surface expression of the gene products (on lymphocytes) was confirmed using monoclonal antibodies of defined BoLA specificity. Phylogenetic analysis of available transcribed cattle MHC class I sequences revealed complex evolutionary relationships including possible evidence for recombination. The study of individual haplotypes suggests that certain groupings of related sequences may correlate with loci, but overall it was not possible to define the origin of individual alleles using this approach. The most striking finding of this study is that none of the cattle class I genes is consistently expressed, and that in contrast to human, haplotypes differ from one another in both the number and composition of expressed classical class I genes.

MhcOnmy-UBA*01 is similar to Onmy-UA-C32 and the classical major histocompatibility complex class I genes of other fish species, whereas Onmy-UAA*01 is divergent from all class I genes so far characterized. Onmy-UAA*01 is expressed at lower levels than Onmy-UBA*01. Although Onmy-UAA*01 exhibits restriction fragment length polymorphism on Southern blotting, the encoded protein is highly conserved. Two allotypes, which differ only by substitution at amino acid position 223 of the α3 domain, have been defined. Onmy-UAA*01 has an exon-intron organization like other class I genes and contains a Tc1-like transposon element in intron III. Orthologues of Onmy-UAA*01 have been characterized in four other species of salmonid. Between four species of Oncorhynchus, UAA*01 proteins differ by only 2–6 amino acids, whereas comparison of Oncorhynchus with Salmo trutta (brown trout) reveals 14–16 amino acid differences. The Onmy-UAA*01 gene has properties indicative of a particularly divergent non-classical class I gene.

    Notizen: Abstract  A comparison of the major histocompatibility complex (Mhc) region between human and mouse highlights both stability and differences. The class II and class III regions are orthologous; they probably existed in the ancestor in a similar organization and were not subjected to major rearrangement. The class I genes, by contrast, are definitely paralogous, having been reorganized several times. As long as only class I genes were identified, the class I regions of human and mouse were difficult to compare directly. The identification of non-class I genes has allowed a comparative map to be drawn, which shows that the class I region is orthologous between human and mouse as well. The lack of orthology specifically applies to the class I sequences. However, the comparative map shows that the non-orthologous class I sequences occupy homologous locations with regard to the conserved genes. I propose a model to explain this paradox. The conserved genes may represent samples of a dense "framework" of genes whose alterations are deleterious. The homologous positions occupied by class I genes would thus represent the few permissive places allowing major perturbations. The evolution of the class I sequences, by duplication and deletion, independently in the two species, has taken place within the scope defined by the framework: insertion at the permissive places, and expansion by creation of class I-related DNA by duplication, thus pushing back the boundaries of the framework.

    Notizen: Abstract  Class I genomic clones of the quail (Coturnix japonica) major histocompatibility complex (MhcCoja) were isolated and characterized. Two clusters spanning the 90.8 kilobase (kb) and 78.2 kb class I gene regions were defined by overlapping cosmid clones and found to contain at least twelve class I loci. However, unlike in the chicken Mhc, no evidence for the existence of any Coja class II gene was obtained in these two clusters. Based on comparative analysis of the genomic sequences with those of the cDNA clones, Coja-A, Coja-B, Coja-C, and Coja-D (Shiina et al. 1999), these twelve loci were assigned to represent one Coja-A gene, two Coja-B genes (Coja-B1 and -B2), four Coja-C genes (Coja-C1-C4), four Coja-D genes (Coja-D1-D4), and one new Coja-E gene. A class I gene-rich segment of 24.6 kb in which five of these genes (Coja-B1, -B2, -D1, -D2 and -E) are densely packed were sequenced by the shotgun strategy. All of these five class I genes are very compact in size [2089 base pairs (bp)–2732 bp] and contain no apparent genetic defect for functional expression. A transporter associated with the antigen processing (TAP) gene was identified in this class I gene-rich segment. These results suggest that the quail class I region is physically separated from the class II region and characterized by a large number of the expressible class I loci (at least seven) in contrast to the chicken Mhc, where the class I and class II regions are not clearly differentiated and only at most three expressed class I loci so far have been recognized.

    Notizen: Abstract  Lymphocytes of a New World primate, the cotton-top tamarin (Saguinus oedipus), express classical G–related major histocompatibility complex (MHC) class I molecules with unusually limited polymorphism and variability. Three G-related loci, an F locus, an E locus, and two pseudogenes (So-N1 and So-N3) have been identified by cDNA library screening and extensive PCR analysis of both cDNA and genomic DNA from the cotton-top tamarin. Furthermore, each genus of the subfamily Callitrichinae (tamarins and marmosets) appears to express its own unique set of MHC class I genes, likely due to a rapid turnover of loci. The rapid emergence of unique MHC class I genes in the Callitrichinae genera, resulting from an active process of duplication and inactivation of loci, may account for the limited diversity of the MHC class I genes in the cotton-top tamarin. To determine the nature of the entire complement of MHC class I genes in the cotton-top tamarin, we synthesized a genomic DNA library and screened it with MHC class I-specific probes. We isolated nine new MHC class I pseudogenes from this library. These newly isolated tamarin G–related MHC class I pseudogenes are not closely related to any of their functional counterparts in the tamarin, suggesting that they do not share a recent common ancestral gene with the tamarin's currently expressed MHC class I loci. In addition, these tamarin sequences display a high rate of nonsynonymous substitutions in their putative peptide binding region. This indicates that the genes from which they have derived were likely subject to positive selection and, therefore, were once functional. Our data support the notion that an extremely high rate of loci turnover is largely responsible for the limited diversity of the MHC class I genes in the cotton-top tamarin.

    Notizen: Abstract  HLA-G is a nonclassical major histocompatibility complex (MHC) class I molecule that is expressed only in the human placenta, suggesting that it plays an important role at the fetal-maternal interface. In rhesus monkeys, which have similar placentation to humans, the HLA-G orthologue is a pseudogene. However, rhesus monkeys express a novel placental MHC class I molecule, Mamu-AG, which has HLA-G-like characteristics. Phylogenetic analysis of AG alleles in two Old World primate species, the baboon and the rhesus macaque, revealed limited diversity characteristic of a nonclassical MHC class I locus. Gene trees constructed using classical and nonclassical primate MHC class I alleles demonstrated that the AG locus was most closely related to the classical A locus. Interestingly, gene tree analyses suggested that the AG alleles were most closely related to a subset of A alleles which are the products of an ancestral interlocus recombination event between the A and B loci. Calculation of the rates of synonymous and nonsynonymous substitution at the AG locus revealed that positive selection was not acting on the codons encoding the peptide binding region. In exon 4, however, the rate of nonsynonymous substitution was significantly lower than the rate of synonymous substitution, suggesting that negative selection was acting on these codons.

    Notizen: Abstract  The presence of Rh30-like polypeptides with an apparent relative molecular mass of 33 000 in the erythrocyte membranes from nonhuman primates and nonprimate mammals (mouse, rat, and dog) was demonstrated by immunoblotting. Nonhuman primates (orangutan, baboon, New World monkeys, lemur) and mouse Rh-like transcripts were amplified and sequenced. Analysis of the deduced amino acids sequences allowed us to determine the amino acid variability of Rh-like polypeptides which correlated with the hydrophylicity indexes. Hence, the putative transmembrane domains exhibited low indexes of variability, while the highest indexes were observed on extramembrane loops with a maximum on the sixth external loop. The cDNA sequences were compared with those previously reported in human, nonhuman primates, and cattle. The time of coalescence of mammalian Rh cDNA sequences was estimated by phylogenetic analysis to be 100 million years.

    Notizen: Abstract The sequences and predicted secondary structures of the four catalytic group-I introns in the psbA gene of Chlamydomonas reinhardtii, Cr.psbA-1–Cr.psbA-4, have been determined. Cr.psbA-1 and Cr.psbA-4 are subgroup-IA1 introns and have similar secondary structures, except at the 3′ end where Cr.psbA-1 contains a large inverted-repeat domain. Cr.psbA-4 is closely related to intron 1 of the Chlamydomonas moewusii psbA gene, with which it shares the same location, high nucleotide identity in the core, and an identically placed ORF that shows 58% amino-acid identity. Cr.psbA-2 is a subgroup-IA3 intron, and shows similarities to the Chlamydomonas eugametos rRNA intron, Ce.LSU-1. Cr.psbA-3 is a subgroup-IA2 intron, and is remarkably similar to the T4 phage intron, sunY. Interestingly, a degenerate version of Cr.psbA-3 is located in the intergenic region between the chloroplast petA and petD genes. All four introns contain ORFs, which potentially code for basic proteins of 11–38 kDa. The ORFs in introns 2 and 3 contain variants of the GIY-YIG motif; however, the Cr.psbA-2 ORF is free-standing, whereas the Cr.psbA-3 ORF is contiguous and in-frame with the upstream exon. The Cr.psbA-4 ORF contains an H-N-H motif, and possibly a GIY-YIG motif. These data indicate that the C. reinhardtiipsbA introns have multiple origins, and illustrate some of the evolutionary DNA dynamics associated with group-I introns in Chlamydomonas.

    Notizen: Abstract  The [2Fe-2S] protein from Azotobacter vinelandii that was previously known as iron-sulfur protein I, or Shethna protein I, has been shown to be encoded by a gene belonging to the major nif gene cluster. Overexpression of this gene in Escherichia coli yielded a dimeric protein of which each subunit comprises 106 residues and contains one [2Fe-2S] cluster. The sequence of this protein is very similar to that of the [2Fe-2S] ferredoxin from Clostridium pasteurianum (2FeCpFd), and the four cysteine ligands of the [2Fe-2S] cluster occur in the same positions. The A. vinelandii protein differs from the C. pasteurianum one by the absence of the N-terminal methionine, the presence of a five-residue C-terminal extension, and a lesser number of acidic and polar residues. The UV-visible absorption and EPR spectra, as well as the redox potentials of the two proteins, are nearly identical. These data show that the A. vinelandii FeS protein I, which is therefore proposed to be designated 2FeAvFdI, is the counterpart of the [2Fe-2S] ferredoxin from C. pasteurianum. The occurrence of the 2FeAvFdI-encoding gene in the nif gene cluster, together with the previous demonstration of a specific interaction between the 2FeCpFd and the nitrogenase MoFe protein, suggest that both proteins might be involved in nitrogen fixation, with possibly similar roles.

    Notizen: Abstract The paper describes an investigation of pressure barriers (plugs) for an underground gas storage facility which were to be constructed from fibre shotcrete. An extensive measurement program on a test plug was carried out to verify the suitability of fibre shotcrete for this purpose. The results of stress and temperature measurements carried out during the construction of the test plug and during cyclic loading of the plug to a water pressure up to 13 MPa are presented. The measurement program provided information on the stress and temperature development inside the plug body during hydration and hardening and also on the residual tensile stresses at the interface between the plug and the rock mass after the end of hydration. Another important output was information on the quality of the plug-rock mass interface, which was critical for a possible leakage. The results obtained during the cyclic loading of the plug revealed uneven stress distribution at the contact and in the adjacent rock mass. It also provided information on the rate of stress redistribution at the contact and inside the rock mass. The results were used to modify the technology of the shotcrete technique to be applied in the final plugs. The gas storage facility using the modified design has been in trial operation since the end of 1998.

This paper offers an econometric methodology for the detection of self-organisational change (defined in terms of the presence of time irreversibility, structural change and fundamental uncertainty) in economic processes that follow logistic diffusion growth paths in historical time. The approach we adopted is built upon recent developments in `moving window' spectral methods which are applied to the scaled residuals generated by estimated logistic diffusion models. We illustrate the use of such methods by examining the case of a financial instrument, namely, the Australian Building Society Deposit, which experienced logistic growth in its market share until bank deregulation was enacted in the 1980s. We show that there is clear evidence that self-organisational change is present over the historical period considered.

    Notizen: Abstract. It is shown that the equilibrium notion of an evolutionary stable strategy (ESS) does have predictive power for standard models of Bertrand competition. This is in contrast to a recent claim by Qin and Stuart (1997). The claim is based on the observation that the solution concept ESS behaves discontinuously when finite (discrete) action games approach an infinite (continuous) action game in the limit. Furthermore, it is argued that from a model-theoretic point of view evolutionary stability in prices (i.e. in the Bertrand model) is quite different from evolutionary stability in quantities (i.e. in the Cournot model).

    Notizen: Abstract  Homeobox genes encode a family of DNA-binding regulatory proteins which are crucial for development. The first plant homeobox gene identified was knotted1 which plays a major role in leaf development. The knotted1 gene has a homeobox which encodes a homeodomain (HD) and HD proteins have been shown to function as transcription factors. A phylogenetic classification of the KNOTTED1 HD is presented. Here, we report six kn1 HDs from the cereals oat, barley, wheat, rye and rice. The KN1 class-I and -II genes can be divided into two distinct clades. Further, we hypothesize that KN1 and BELL1/MEIS HDs, (the closest non-KN1 class HDs) evolved from a common ancestor after divergence from the common precursor of all the homeobox genes. Our analysis clearly shows the presence of an ancestral KN1 HD from which all the known plant kn1 class of genes evolved.

    Notizen: Abstract  Domesticated rice differs from the wild progenitor in large arrays of morphological and physiological traits. The present study was conducted to identify the genetic factors controlling the differences between cultivated rice and its wild progenitor, with the intention to assess the genetic basis of the changes associated with the processes of rice domestication. A total of 19 traits, including seven qualitative and 12 quantitative traits, that are related to domestication were scored in an F2 population from a cross between a variety of the Asian cultivated rice (Oryza sativa) and an accession of the common wild rice (O. rufipogon). Loci controlling the inheritance of these traits were determined by making use of a molecular linkage map consisting of 348 molecular-marker loci (313 RFLPs, 12 SSRs and 23 AFLPs) based on this F2 population. All seven qualitative traits were each controlled by a single Mendelian locus. Analysis of the 12 quantitative traits resolved a total of 44 putative QTLs with an average of 3.7 QTLs per trait. The amount of variation explained by individual QTLs ranged from a low of 6.9% to a high of 59.8%, and many of the QTLs accounted for more than 20% of the variation. Thus, genes of both major and minor effect were involved in the differences between wild and cultivated rice. The results also showed that most of the genetic factors (qualitative or QTLs) controlling the domestication-related traits were concentrated in a few chromosomal blocks. Such a clustered distribution of the genes may provide explanations for the genetic basis of the “domestication syndrome” observed in evolutionary studies and also for the “linkage drag” that occurs in many breeding programs. The information on the genetic basis of some desirable traits possessed by the wild parent may also be useful for facilitating the utilization of these traits in rice-breeding programs.

    Notizen: Abstract.  Chromosome pairing at metaphase-I was analyzed in F1 hybrids among T. turgidum (AABB), T. aestivum (AABBDD), and T. timopheevii (AtAtGG) to study the chromosome structure of T. timopheevii relative to durum (T. turgidum) and bread (T. aestivum) wheats. Individual chromosomes and their arms were identified by means of C-banding. Homologous pairing between the A-genome chromosomes was similar in the three hybrid types AAtBG, AAtBGD, and AABBD. However, associations of B-G were less frequent than B-B. Homoeologous associations were also observed, especially in the AAtBGD hybrids. T. timopheevii chromosomes 1At, 2At, 5At, 7At, 2G, 3G, 5G, and 6G do not differ structurally from their counterpart in the A and B genomes. Thus, these three polyploid species inherited translocation 5AL/4AL from the diploid A-genome donor. Chromosome rearrangements that occurred at the tetraploid level were different in T. turgidum and T. timopheevii. Translocation 4AL/7BS and a pericentric inversion of chromosome 4A originated only in the T. turgidum lineage. The two lines of T. timophevii studied carry four different translocations, 6AtS/1GS, 1GS/4GS, 4GS/4AtL, and 4AtL/3AtL, which most likely arose in that sequence. These structural differences support a diphyletic origin of polyploid wheats.

    Notizen: Abstract  Intron-2 of the Oryza sativa CatA catalase gene is similar in nucleotide sequence to p-SINE1, a retroposon, and seems to have been added to the ancestral genome of rice. To examine when the p-SINE1-like intron was inserted into CatA during the evolutionary divergence of Oryza species, and to elucidate the evolutionary relationships among Oryza species using the sequence of the intron as a marker, we performed polymerase chain reaction (PCR) analyses of 32 accessions of 17 Oryza species with various genome types. Agarose-gel electrophoresis of the PCR products revealed that all the Oryza species with an AA genome have the CatA homolog with the intron, whereas other Oryza species have the CatA homolog without the intron. These results indicate that intron-2 of CatA is a good marker for distinguishing species with an AA genome among Oryza species. Sequencing of the PCR products showed that all the introns are similar to p-SINE1, though with slight variations in length. We also performed PCR analyses using four accessions of three species in genera related to Oryza, and found that there is an intron in the CatA homolog of Leersia perrieri. On the other hand, the CatA homolog of Porteresia coarctata has no intron. Sequence data showed that the L. perrieri homolog has a p-SINE1-like intron similar to that in Oryza species with an AA genome. These results suggest that the p-SINE1-like intron was already present in the common ancestor of Oryza and L. perrieri and was then lost in the ancestors of P. coarctata and of the Oryza species other than those with an AA genome. The phylogenetic tree of Oryza species with an AA genome based on the nucleotide sequences of the introns leads us to propose that Oryza species with an AA genome evolved from an ancestor of Oryza longistaminata.

    Notizen: Abstract  The Endosperm Balance Number (EBN) hypothesis was developed in the early ’80s to explain the basis for normal seed development after intra- and inter-specific crosses, first in the potato and then in several other crop species. According to this hypothesis, each species has a genome-specific effective ploidy, the EBN, which must be in a 2 : 1 maternal to paternal ratio in the hybrid endosperm for normal development of the endosperm itself. This paper reviews how the EBN may act as a powerful isolating mechanism in sexual reproduction, maintaining the genome integrity of the species and playing an important role in the speciation of polyploids from diploids. We also provide further evidence that EBN is more important than chromosome ploidy in determining the success or failure of interspecific crosses. In fact, results from inter-ploidy and inter-EBN crosses to infuse 1EBN Solanum commersonii into 4EBN S. tuberosum demonstrated that the knowledge and manipulation of EBN is a useful tool in designing breeding schemes and in predicting the offspring ploidy and EBN. In this paper we also discuss the exceptions to the 2 : 1 EBN ratio, and report the evidence for endomitosis in the polar nuclei to explain exceptions to the EBN model in the potato.

    Notizen: Abstract A number of hypotheses have been proposed to explain the evolution of multiple mating in the honeybee queen. In particular, the consequences of reduced intracolonial relatedness provide plausible explanations for multiple mating with up to ten drones, but fail to account for the much higher mating frequencies observed in nature. In this paper, we propose an alternative mechanism which builds on non-linear relationships between intracolonial frequencies in genotypic worker specialization and colony fitness. If genes for any worker specialization confer an advantage on colony fitness only when they are rare, this would require a stable mix of sperm from a few drones which contribute that trait, and many which do not. To ensure both specific, low within-colony proportions of “rare specialist” genes, and to reduce random variation of these proportions would require mating with high numbers of drones. The quantitative implementation shows that moderate to very high numbers of matings are required to exploit colony advantages from genotypic allocation of workers to rare tasks. Extreme polyandry thus could result from colony selection dependent on the intracolonial frequency of rare genetic specialists.

    Notizen: Abstract Homeobox genes located in the 5’ part of the HoxA and HoxD complexes are required for proliferation of skeletal progenitor cells of the vertebrate limb. Specific combinations of gene products determine the length of the upper arm (genes belonging to groups 9 and 10), the lower arm (groups 10, 11 and 12) and the digits (groups 11, 12 and 13). In these different domains, individual gene products quantitatively contribute to an overall protein dose, with predominant roles for groups 11 and 13. Quantitative reduction in the gene dose in each set results in truncations of the corresponding anatomical regions. The physical order of the genes in the HoxA and HoxD complexes, as well as a unidirectional sequence in gene activation, allow for completion of the process in a precise order, which in turn makes possible the sequential outgrowth of the respective primordia. While the skeletal patterns of upper and lower limb are relatively stable throughout the tetrapods, more variation is seen in the digits. Molecular analysis of the underlying regulatory processes promises further exciting insights into the genetic control of development, pathology and the course of evolution.

    Notizen: Abstract To date, no conclusive evidence has been presented for the existence of neuronal-like elements in Porifera (sponges). In the present study, isolated cells from the marine sponge Geodia cydonium are shown to react to the excitatory amino acid glutamate with an increase in the concentration of intracellular calcium[Ca2+]i. This effect can also be observed when the compounds L-quisqualic acid (L-QA) or L-(+)-2-amino-4-phosphonobutyric acid (L-AP-4) are used. The effect of L-QA and L-AP-4, both agonists for metabotropic glutamate receptors (mGluRs), can be abolished by the antagonist of group I mGluRs, (RS)-α-methyl-4-carboxyphenylglycine. These data suggest that sponge cells contain an mGluR-like protein. A cDNA encoding rat mGluR subtype 1 has been used to identify the complete nucleotide sequence of G. cydonium cDNA coding for a 528-amino-acid-long protein (59 kDa) that displays marked overall similarity to mGluRs and to γ-amino-butyric acid B receptors. The deduced sponge polypeptide, termed putative mGlu/GABA-like receptor, displays the highest similarity to the two families of metabotropic receptors within the transmembrane segment. The N-terminal part of the sponge sequence shows similarity to mGluR4 and mGluR5. These findings suggest that the earliest evolutionary metazoan phylum, the Porifera, possesses a sophisticated intercellular communication and signaling system, as seen in the neuronal network of higher Metazoa.

    Notizen: Abstract The red heterobasidiomycetous yeast Xanthophyllomyces dendrorhous (perfect state of Phaffia rhodozyma) contains a novel type of carotenoid biosynthetic enzyme. Its structural gene, designated crtYB, was isolated by functional complementation in a genetically modified, carotenogenic Escherichia coli strain. Expression studies in different carotenogenic E. coli strains demonstrated that the crtYB gene encodes a bifunctional protein involved both in synthesis of phytoene from geranylgeranyl diphosphate and in cyclisation of lycopene to β-carotene. By sequence comparison with other phytoene synthases and complementation studies in E. coli with various deletion derivatives of the crtYB gene, the regions responsible for phytoene synthesis and lycopene cyclisation were localised within the protein.

    Notizen: Abstract In contrast to animals, the slowly evolving mitochondrial nucleotide sequences of plants appear well suited to investigate phylogenetic relations between old taxonomic groups. Analysis ofnad5 gene sequences in 47 bryophytes, the living representatives of very early land plants, confirm this assessment. Statistically reliable phylogenetic trees are obtained with different mathematical approaches. A group I intron sequence conserved in thenad5 gene of all 30 mosses and 15 liverworts investigated supports a sister group relationship of the two classes. The intron sequence adds phylogenetic information for fine resolution on top of the conserved exon sequences down to the level of classically defined orders or families, respectively. This intron is not present in the hornwortsAnthoceros husnotii andA. punctatus. The results allow statements on diverging taxonomic interpretations and support the monophyly of the liverworts, mosses, Jungermanniidae, Marchantiidae and Bryidae, and allow recognition of subclasses like Hypnanae and Dicrananae. Among the mosses, the derived orders (subclass Bryidae) are confidently set apart from the Sphagnales, Andreaeales, Polytrichales and Tetraphidales with Buxbaumiales occupying a mediating position. Among the liverworts, full support is found for the classic separation of simple (jungermanniid) and complex thalloid (marchantiid) species with a strikingly low mitochondrial sequence divergence among the latter.

    Notizen: Abstract To determine whether inbreeding depression accounts for the maintenance of outcrossing in populations of the self-compatible orchidEncyclia cochleata, the estimated selective advantage of selfing was compared to a measure of inbreeding depression. Individuals from three populations ofE. cochleata and some of their progeny were phenotyped using isozyme analysis. The electrophoretic data were used to estimate the outcrossing rate and the theoretical cost of outcrossing. Inbreeding depression was estimated by comparing the fitness of the progeny resulting from both types of pollinations. The seeds from outcrossed and selfed hand-pollinations and naturally pollinated seeds from a population of the triandrous form ofE. cochleata were grown aseptically on culture media, and their development over the next three years recorded. Inbreeding was common, particularly in one population (outcrossing rate 40%). However, the level of inbreeding depression was only 1–2%, considerably less inbreeding depression than expected.

    Notizen: Abstract This paper considers the evolution of cooperative behaviors as the interaction among agents using a genetic algorithm to improve the performance of the task in a group (group performance). Previous research often usedthe group fitness method, which evaluates group performance for the evolution of multiple groups in parallel. However, this entails large simulation costs and the evolution speed is slow.The individual fitness method that evaluates theindividual performance of the task entails a smaller simulation cost. However, it can not improve the group performance since each agent behaves selfishly. To optimize the group performance, it is important to include bothcompetition andsharing. Therefore, this paper presentsthe species fitness method, which shares the individual performances of agents belonging to the same species in a group that all have the same chromosomes. We show comparative experiments on these three methods on the evolutionary simulation of a foraging task in a group. To test the interaction among the agents, four kinds of species are evolved which show their communication ability by demonstrating whether the agent can send or receive the signal for food. Experimental results show that evaluating the species variance fitness leads the agents into reciprocative actions.

    Notizen: Abstract The acquisition process of bipedal walking in humans was simulated using a neuro-musculo-skeletal model and genetic algorithms, based on the assumption that the shape of the body has been adapted for locomotion. The model was constructed as 10 two-dimensional rigid links with 26 muscles and 18 neural oscillators. Bipedal walking was generated as a mutual entrainment between neural oscillations and the pendulous movement of body dynamics. Evolutionary strategies incorporated, for example, as fitness in the genetic algorithms were assumed to decrease energy consumption, muscular fatigue, and load on the skeletal system. An initial population of 50 individuals was created, and an evolutionary simulation of 5000 steps was conducted. As a result, the shape of the body changed from that of a chimpanzee to that of a modern human, and the body size nearly reached the size of a modern human. These simulation results show that improving locomotive efficiency and reducing the load on the musculo-skeletal system are important factors affecting the evolution of the human body shape and bipedal walking. Such computer simulations help us to understand the process of evolution and adaptation for locomotion in humans.

    Notizen: Abstract Since 1992, as part of a study to circumscribe the genus Porphyra in New Zealand, an extensive culture programme has been developed at the Museum of New Zealand Te Papa Tongarewa. This collection currently houses more than 800 cultures and 150strains. We currently recognise at least 20 species in New Zealand, most of which are undescribed. Close observation of reproduction and the development of conchocelis-phase filaments grown in the laboratory, under controlled conditions of temperature, irradiance and photoperiod, has enabled the recognition of characters that can assist in species separation. The comparative taxonomic value of a range of such characters is discussed, including reporting a novel reproductive mode.

    Notizen: Abstract Although Porphyra is commercially farmed in many countries, in South Africa only small harvests of wild populations for sale as nori have been carried out. The discovery that Porphyra improves growth of South African abalone (Haliotis midae) farmed inland-based tanks has led to increased pressure to harvest wild populations. This paper reports on a survey of the distribution and seasonality of Porphyra in the southern Western Cape. Porphyrawas present at all sites surveyed, and showed considerable temporal variation. A significant amount of the Porphyra present is in reserves and therefore protected from harvesting. Close rexamination of one site revealed seasonal populations of Porphyra that occupied different niches dependent on season. Recruitment peaked in spring and autumn, leading to dense summer and winter populations. Summer populations generally grew lower in the eulittoral than winter populations. No pattern in the mortality of larger thalli wasde tected, though sporeling mortality was high following recruitment peaks. Although it seems that most sites in the southern Western Cape are suitable for harvesting, the taxonomy of the genus in the region urgently needs revision if populations are to be appropriately managed.

    Notizen: Abstract A partial genetic linkage map constructed using 150 single seed descent (SSD) lines generated from a cross between the hexaploid wheat varieties ‘Schomburgk’ and ‘Yarralinka’ was used to identify loci controlling milling yield. Milling yield data were obtained using seed collected from field trials conducted at different sites over two seasons. The estimated broad-sense heritability of milling yield in this population was calculated as 0.48. In the preliminary analysis, two regions were identified on chromosomes 3A and 7D, which were significantly associated with milling yield and accounted for 22% and 19% of the genetic variation, respectively. Bulked segregant analysis in combination with AFLP identified other markers linked to these loci, as well as an additional region on chromosome 5A, which accounted for 19% of the genetic variation. The applicability of these markers as selection tools for breeding purposes is discussed.

    Notizen: Abstract Transgenic wheat plants containing the gene encoding snowdrop lectin (Galanthus nivalis agglutinin; GNA) under the control of constitutive and phloem-specific promoters were generated through the particle bombardment method. Thirty-two independently derived plants were subjected to molecular and biochemical analyses. Transgene integration varied from one to twelve estimated copies per haploid genome, and levels of GNA expression from 0 to ca. 0.2% of total soluble protein were observed in different transgenic plants. Seven transgenic plants were selected for further study. Progeny plants from these parental transformants were selected for transgene expression, and tested for enhanced resistance to the grain aphid (Sitobion avenae) by exposing the plants to nymphal insects under glasshouse conditions. Bioassay results show that transgenic wheat plants from lines expressing GNA at levels greater than ca. 0.04% of total soluble protein decrease the fecundity, but not the survival, of grain aphids. We propose that transgenic approaches using insecticidal genes such as gna in combination with integrated pest management present promising opportunities for the control of damaging wheat pests.

    Notizen: Abstract The Yr17 gene, which is present in many European wheat cultivars, displays yellow rust resistance at the seedling stage. The gene introduced into chromosome 2A from Aegilops ventricosa was previously found to be closely linked (0.5 cM) to leaf and stem rust resistance genes Lr37 and Sr38, respectively. The objective of this study was to identify molecular markers linked to the Yr17 gene. We screened with RAPD primers, for polymorphism, the DNAs of cv. Thatcher and the leaf rust-resistant near-isogenic line (NIL) RL 6081 of cv. Thatcher carrying the Lr37 gene. Using a F2 progeny of the cross between VPM1 (resistant) and Thésée (susceptible), the RAPD marker OP-Y15580 was found to be closely linked to the Yr17 gene. We converted the OP- Y15580 RAPD marker into a sequence characterized amplified region (SCAR). This SCAR marker (SC-Y15) was linked at 0.8 ± 0.7 cM to the Yr17 resistance gene. We tested the SC-Y15 marker over a survey of 37 wheat cultivars in order to verify its consistency in different genetic backgrounds and to explain the resistance of some cultivars against yellow rust. Moreover, we showed that the Xpsr150-2Mv locus marker of Lr gene described by Bonhomme et al. [6] which possesses A. ventricosa introgression on the 2A chromosome was also closely linked to the Yr17 gene. Both the SCAR SC-Y15 and Xpsr150-2Mv markers should be used in breeding programmes in order to detect the cluster of the three genes Yr17, Lr37 and Sr38 in cross progenies.

    Notizen: Abstract The effect of light, temperature and ammonium on inorganic nitrogen uptake by phytoplankton was investigated from June 1994 through December 1995 at three sites in the Tagus estuary (Portugal), during high tide of neap tides. Ammonium concentrations higher than 10 μM reduced nitrate uptake down to 24% but never prevented it. Below this threshold concentration, nitrate uptake was neither inhibited nor changed. Uptake of both nitrate and ammonium as a function of light intensity exhibited a saturation response. Uptake reduction occurred in the near bottom phytoplankton populations, particularly for nitrate. The ammonium uptake system was less limited by light than the nitrate uptake system, indicating the importance of ammonium as a nitrogen source for the phytoplankton which is likely to experience high changes in light in the well-mixed water column of this estuarine environment. Ammonium uptake was exponentially related to temperature in the upper estuary whereas in the mid and lower estuary this relationship was linear. The effect of temperature on nitrate uptake was linear but far less marked than for ammonium uptake.

    Notizen: Abstract Three embryonic stages of Atlantic salmon (Salmo salar L.) were subjected to eight constant incubation temperatures (4, 7, 10, 12, 14, 16, 19 and 22 °C) exceeding the range usually experienced in natural conditions. A change in thermal tolerance during the embryonic and larval development was registered: pre-hatching stages showed an upper thermal limit at about 16 °C, while hatched larvae survived until 22 °C. Temperature significantly affects developmental rate, resulting in a faster development and, consequently, lower yolk weight percentage at higher temperatures. We found positive relationships between incubation temperature and body size (length and weight) in the less developed stages, in which some yolk remained, but size decreased at increasing temperatures when yolk was completely exhausted.

    Notizen: Abstract The influence of temperature on growth and biomass composition of two species of Spirulina, S. maxima and S. platensis used for food was studied. A 4L fermenter with temperature and agitation control was used to cultivate both species. Under continuous light, maximum cell production of 2.4 g l−1 was verified for both cultures studied at temperatures above 25 °C: S. maxima (30 °C and 35 °C) and S. platensis (25 °C and 30 °C). An accentuated lag phase was observed for all cultures at lower temperatures (15–20 °C), and a maximum biomass production of 1.5 g l−1 was achieved. It was also observed that an increase of temperature caused a marked decrease in protein content, while carbohydrate synthesis was stimulated. The concentration of γ-linolenic acid varied from 11–16% for S. maxima and from 12–14% for S. platensis, at the optimum growth temperatures. Greater culture volumes were also studied in order to compare the performance of glass and plastic containers. At optimum growth temperature, S. maxima produced the same cell growth and similar final biomass composition.

    Notizen: Abstract Infection by two isolates of impatiens necrotic spot tospovirus (INSV) under temperature regimes of 25/18°C (day/night) or 33°C (continuous) was studied in Capsicum annuum (systemically susceptible to tomato spotted wilt tospovirus, TSWV), C. chinense PI 152225 and PI 159236 (reacting hypersensitively to TSWV) and Nicotiana benthamiana (systemically susceptible to both tospoviruses). At 25/18°C infection was systemic in all hosts tested. At 33°C infection in N. benthamiana was systemic whereas in C. annuum and C. chinense it was restricted to the inoculated leaves. The result differed from that reported for TSWV, where high temperature made plants more susceptible, or caused no difference. Exchanging temperature regimes 6 h to 4 days after inoculation did not affect the final results one month later, with plants being only locally infected at 33°C continuous regime, or systemically infected at 25/18°C alternate regime. The two INSV isolates were biologically and serologically stable for 5 passages in N. benthamiana held continuously at 33°C.

TheKnoxieae are monophyletic ifOtiophora, Otomeria, andPentas are included. The tribeAnthospermeae is supported as monophyletic, but its subtribes are not. ThePaederieae, together withTheligonum, form a paraphyletic grade basal to theRubieae. TheHedyotideae, includingSchismatoclada, form a grade at the base of theSpermacoceae. TheGaertnereae are monophyletic and distinct from thePsychotrieae. TheMorindeae are monophyletic and includeDamnacanthus andMitchella. Schradera is the sister group of theMorindeae. ThePsychotrieae are monophyletic when theGaertnereae, Lasianthus, andDeclieuxia are excluded. The recognition of a subtribeHydnophytineae leaves the rest of thePsychotrieae paraphyletic.Psychotria is paraphyletic with respect to all other genera of the tribe. Approximately 50 genera are here classified for the first time based on molecular data.

    Notizen: Abstract Previous molecular phylogenetic analyses have demonstrated that Saxifragaceae sensu lato are polyphyletic, with component lineages scattered throughout the eudicots. As part of our effort to elucidate the relationships of members of Engler and Prantl's Saxifragaceae s. l., we undertook a molecular systematic study of subfamily Brexioideae, which comprises three genera:Brexia, Ixerba, andRoussea. Not all taxonomic treatments have concurred, however, in placing these genera together. To elucidate relationships among these three genera as well as their relationships to other angiosperms we constructed large data sets ofrbcL, 18S rDNA, andrbcL + 18S rDNA sequences. Our phylogenetic analyses indicate clearly that Brexioideae are polyphyletic.Brexia is part of a celastroid clade that also includesParnassia, Lepuropetalon, and Celastraceae.Ixerba appears as sister to a large eurosid I clade;Roussea appears as part of Asterales. Molecular data, therefore, indicate that Brexioideae are a polyphyletic assemblage and component genera should ultimately be incorporated into other groups. Our studies continue to demonstrate the polyphyly not only Saxifragaceae s. l., but also of its constituent subfamilies.

    Notizen: Abstract Tourist-OsaCatA, a transposable element, was found in the 5′-flanking region of the rice gene CatA. The characteristics of this element are similar to those of the other Tourist elements so far found in Oryza sativa. PCR and sequence analyses of 37 accessions of 18 species revealed that all the Oryza species examined, except for one accession, have either a full-length or a partial Tourist element at this locus. Unlike the Tourist elements previously reported, this Tourist element is found in all four Oryza species complexes in the Oryzeae tribe. All AA genome Oryza species, except O. longistaminata, contain the full-length Tourist element. O. longistaminata and the species of the O. officinalis, O. meyeriana and O. ridleyi complexes contain the partial element. A phylogenetic tree of Oryza species based on the nucleotide sequences of these Tourist elements was constructed. The O. longistaminata accessions were placed near the neighboring cluster of the officinalis complex. We propose that the ancestor of O. longistaminata and that of other species with the AA genome diverged, and the ancestor(s) of the O. officinalis, O. ridleyi and O. meyeriana complexes then diverged from the ancestor of O. longistaminata in the course of the evolution of the Oryza species. The Tourist elements associated with CatA and its orthologs thus provide useful tools for examining evolutionary relationships among Oryza species.

    Notizen: Abstract Bordetella pertussis and B. bronchiseptica are genetically very closely related but differ significantly in their virulence properties. Using Representational Difference Analysis (RDA), 11 DNA fragments specific for B. pertussis Tohama I or B. bronchiseptica BB7865 were identified. All B. bronchiseptica BB7865-derived fragments also hybridized with chromosomal DNA from B. parapertussis but not from the B. pertussis strains Tohama I and W28, underlining the close phylogenetic relationship between B. bronchiseptica and B. parapertussis. The B. pertussis type strain BP18323 is a special case, as it contains DNA sequences characteristic for both B. pertussis and B. bronchiseptica. As demonstrated by pulsed-field gel electrophoresis, several of the BB7865-derived fragments are present on a single 30-kb XbaI fragment. Based on the sequences of putative coding regions, four of these fragments may code for proteins involved in carbohydrate metabolism or transport. In agreement with this notion, a mutant for one of these loci synthesizes a significantly altered lipopolysaccharide that lacks the O-specific side chains. The analysis of the corresponding genomic region in various Bordetella species showed that this locus is present in B. bronchiseptica and B. parapertussis but not in B. pertussis. This confirms that the RDA approach has identified a novel strain-specific LPS biosynthesis locus which accounts for the differences between the LPS structures elaborated by different Bordetella species.

    Notizen: Abstract Although maize endosperm undergoes programmed cell death during its development, it is not known whether this developmental feature is common to cereals or whether it arose inadvertently from the selection process that resulted in the enlarged endosperm of modern maize. Examination of wheat endosperm during its development revealed that this tissue undergoes a programmed cell death that shares features with the maize program but differs in some aspects of its execution. Cell death initiated and progressed stochastically in wheat endosperm in contrast to maize where cell death initiates within the upper central endosperm and expands outward. After a peak of ethylene production during early development, wheat endosperm DNA underwent internucleosomal fragmentation that was detectable from mid to late development. The developmental onset and progression of DNA degradation was regulated by the level of ethylene production and perception. These observations suggest that programmed cell death of the endosperm and regulation of this program by ethylene is not unique to maize but that differences in the execution of the program appear to exist among cereals.

    Notizen: Abstract Granule-bound starch synthase (GBSS), a product of the waxy gene in rice (Oryza sativa L.), is necessary for the synthesis of amylose in the endosperm. In an extended pedigree of 89 rice cultivars, we have previously shown that all cultivars with more than 18% amylose had the sequence AGGTATA at the leader intron 5′ splice site, while all cultivars with a lower proportion of amylose had the sequence AGTTATA. This single-nucleotide polymorphism reduces the efficiency of GBSS pre-mRNA processing. It also results in alternate splicing at multiple sites, some of which have non-consensus sequences. Here we demonstrate that this same G-to-T polymorphism is also associated with differential sensitivity to temperature during the period of grain development. Cultivars with the sequence AGTTATA have a substantial increase in accumulation of mature GBSS transcripts at 18 °C compared to 25 or 32 °C. The selection of leader intron 5′ splice sites is also affected by temperature in these cultivars. A 5′ splice site −93 upstream from that used in high-amylose varieties predominates at 18 °C. At higher temperatures there is increased utilization of a 5′ splice site at −1 and a non-consensus site at +1. Potential implications of differential 5′ splice site selection and associated differences in 3′ splice site selection on transcript stability and translational efficiency are discussed.

    Notizen: Abstract Experiments were conducted under laboratory and greenhouse conditions to study the effect of 2,4-D on rooting and salinity tolerance of wheat. Seeds of one commercial wheat (Inqalab-91) and three salt-tolerant wheat lines (WL-41, WL-359, and WL-1073 developed through wide hybridization) were included in the study. Preliminary and short-term experiments were conducted to determine the level of 2,4-D (administered through seed soaking for 24.5 h. at 25 °C in the dark) at which the maximum number of roots emerged. Under hydroponic conditions, 2,4-D treatment of seeds caused an increase of 60 to 100% in the number of primary roots. The maximum increase in the number of roots was observed in one of the salt tolerant wheat lines (WL-41). The roots appeared in bunches but showed stunted growth at higher levels of 2,4-D. Dry matter accumulation decreased markedly; the effect was more pronounced in Inqalab-91 which is less tolerant to stress than other wheat lines. In all wheat types, allocation of dry matter to roots relative to shoot increased due to 2,4-D treatment. In soil, seeds treated with different levels of 2,4-D showed a germination delay of 1–3 days. Although the number of primary roots increased, 2,4-D treatment caused a decrease in total dry matter accumulation by plants grown for 40 days. In another experiment, conducted under greenhouse conditions, seed germination and growth of seedlings was significantly retarded in saline compared to that in non-saline (normal) soil. Initially, the pace of germination of treated seeds as well as seedling growth was slower in both soils, but after six weeks, the leaf area of seedlings raised from treated seeds was greater than those raised from untreated seeds. Towards maturity, plants arising from treated seeds developed wider and longer flag leaves leading to enhanced yield. Root biomass decreased in saline soil as compared to normal soil. However, 2,4-D treatment caused a substantial increase in root biomass in saline soil and the roots were harder in texture in wheats other than Inqalab-91. Seed treatment with 2,4-D led to a significant improvement in the number of productive tillers, yield of straw and grain, and grain protein content of all wheats grown in saline soil. Plants grown in normal soil did not show any marked effect of seed treatment on grain yield and other agronomic parameters. The four wheats showed substantial differences for different parameters but the salt tolerant wheat lines performed better compared to the commercial variety Inqalab-91.

    Notizen: Abstract Phosphorus (P) deficiency limits the yield of wheat, particularly by reducing the number of ears per unit of area because of a poor tiller emergence. The objectives of this work were to (i) determine whether tiller emergence under low phosphorus availability is a function of the availability of assimilates for growth or a direct result of low P availability, (ii) attempt to establish a quantitative relation between an index of the availability of P in the plant and the effects of P deficiency on tiller emergence, and (iii) to provide a better understanding of the mechanisms involved in tiller emergence in field-grown wheat. Wheat (Triticum aestivum L., cv. INTA Oasis), was grown in the field under drip irrigation on a typic Argiudol, low in P (5.5 μg P g-1 soil Bray & Kurtz I) in Balcarce, Argentina. Treatments consisted of the combination of three levels of P fertilization 0, 60 and 200 kg P2O5 ha-1, and two levels of assimilate availability, a control (non-shaded) and 65% of reduction in incident irradiance from seedling emergence until the end of tillering (shaded). Phosphorus treatments significantly modified the pattern of growth and development of the plants. Shading reduced the growth and concentration of water-soluble carbohydrates in leaves and stems. Leaf photosynthetic rate at saturating irradiance was reduced by P deficiency, but was not affected by shading. At shoot P concentrations less than 4.2 g P kg-1 the heterogeneity in the plant population increased with respect to the number of plants bearing a certain tiller. At a shoot P concentration of 1.7 g P kg-1 tillering ceased completely. Phosphorus deficiency directly altered the normal pattern of tiller emergence by slowing the emergence of leaves on the main stem (i.e. increasing the phyllochron), and by reducing the maximum rate of tiller emergence for each tiller.

    Notizen: Abstract The effect of plant breeding on yield and their physiological determinants has been widely studied in wheat. However, it is poorly understood how, and to what extent, yield stability has been modified. To attempt a direct analysis of changes in absolute and relative yield stability, data of yield of cultivars released in different eras in different environments were obtained from records from our lab and from the literature. Depending on the availability of data, effects of plant breeding on yield stability of cultivars released in Argentina, Australia, Italy and the United Kingdom were evaluated using a quantitative approach. In this paper it was assumed that the slope of yield vs. environmental index estimates the instability of the cultivars. In addition, a more qualitative approach for Mexico, and the former USSR complemented this analysis. There was a clear decrease in yield stability assessed in absolute terms as a consequence of wheat breeding. In Argentina, Australia, Italy and the UK this decrease was related to the magnitude of yield increases. However, the decrease in yield stability in Argentina and Australia was less than for Italy and the UK, particularly so during the last 30 years. Modern cultivars released in Argentina and Australia showed a trend to maintain yield stability as a percentage of their yield similar to that of their predecessors, while the two European countries analysed tended to a slight decrease in yield stability even in relative terms. The complementary, less quantitative evaluation of Mexico and the former USSR appeared to confirm the quantitative trends described for the other countries, i.e. a general decrease in yield stability (assessed in absolute terms) with genetic gains in yield potential.

    Notizen: Abstract Chromosome arm 1RS of rye ( Secale cereale L.), when transferred to wheat ( Triticum sp.), significantly influences variety performance, because it carries genes for resistance to disease and insect pathogens. Inserted into wheat, 1RS also promotes haploid production, affects end-product quality, and sometimes affects yield. Therefore, its detection by breeders and geneticists is important. The entire 1RS arm is present in chromosome substitutions and in Robertsonian translocations involving chromosomes 1A, 1B, or 1D of wheat. In recombinant lines, a segment of 1RS has been exchanged with a segment of a group-1 wheat chromosome. Determining the wheat chromosome arm involved in a translocation, the source of rye chromatin, and the amount of 1RS chromatin introduced is necessary for a complete characterization of the introgressed segment. Biochemical, molecular, and cytogenetic technologies are described which enable such a characterization of 1RS in wheat. Examples of using gel electrophoresis, high-performance liquid chromatography, monoclonal antibodies, rye-specific molecular probes, RFLP and PCR assays, chromosome banding, in situ hybridization, and flow cytometry are provided. A comparison of these technologies is made and the advantages and disadvantages of each technology are discussed relative to modern wheat breeding efforts.

    Notizen: Abstract Frost is one of the main climatic stresses which has to be overcome by a winter pea crop. Some forage lines show a delayed floral initiation, which helps them to escape the main winter freezing periods, as a higher susceptibility to frost is observed after floral initiation commences. Frost tolerant forage peas have been used in all winter pea breeding programs in France and our main purpose was to evaluate to what extent those lines and the current winter varieties are variable for the date of floral initiation in field conditions. A field experiment was carried out during two years at the INRA experimental station of Mons (northern France). Different genotypes (9 in 1995–1996 and 12 in 1996–1997) were sown at approximately monthly intervals between September and June to provide a range of photothermal conditions. The date of floral initiation was determined by destructive sampling. We observed variability for the date of floral initiation among the different genotypes and sowing dates. Our data particularly highlighted the different reactions to photoperiod. Three varieties had no detectable reaction. Six varieties exhibited a quantitative response, with varied intensities. Lastly, the three forage varieties showed a qualitative, high response to photoperiod, which indicated the presence of the Hr allele, already described in Pisum.

    Notizen: Abstract Wheat cultured cells were used to study the role of Ca2+ in regulating protein kinases during the induction of defense-related genes by fungal elicitor treatments. Manipulation of intracellular Ca2+ concentrations by treatment with calcium ionophore A23187 in the presence of high extracellular Ca2+ resulted in the induction of mRNA expression of WCK-1, a gene encoding mitogen-activated protein (MAP) kinase. The induction of WCK-1 mRNA by A23187 did not occur when extracellular Ca2+ was chelated by 1,2-bis(2-aminophenoxy)ethane-N,N,N′,N′-tetraacetic acid (BAPTA). The WCK-1 mRNA was also induced by Typhula ishikariensis-derived elicitors, suggesting a possible involvement of WCK-1 in the plant defense response against pathogens. BAPTA and a calcium channel blocker, La3+, inhibited the elicitor-induced expression of the WCK-1 mRNA. A recombinant fusion protein of WCK-1 (GST-WCK-1) autophosphorylated at the Tyr residue and exhibited an autophosphorylation-dependent protein kinase activity towards myelin basic protein. Alteration of Tyr-196 in the conserved ‘TEY’ motif in GST-WCK-1 to Phe by site-directed mutagenesis abolished the autophosphorylation. The GST-WCK-1 protein was activated by elicitor-treated wheat cell extracts but not by the control extract. These results suggest that fungal elicitors activate WCK-1, a specific MAP kinase in wheat. Furthermore, the results suggest a possible involvement of Ca2+ in enhancing the MAP kinase signaling cascade in plants by controlling the levels of the MAP kinase transcripts.

    Notizen: Abstract A wheat gene, denoted Sbe1, encoding a type I starch-branching enzyme (SBEI) was isolated from a genomic library and shown to comprise 14 exons distributed over a 5.7 kb DNA region. Analyses of kernel RNA by 5′ rapid amplification of cDNA ends (5′-RACE) and reverse transcription-polymerase chain reaction (RT-PCR) demonstrated a considerable sequence variation at the 5′ ends of SBEI gene transcripts. DNA sequence alignments between the 5′-RACE products and the Sbe1 genomic DNA indicated that the first two exons and first intron were differentially processed to generate three classes of the mature transcript. One form of the SBEI gene transcript in 12-day old kernels contained the exon I+II+III combination at the 5′ end, whereas other forms differed by inclusion of intron 1 or exclusion of exon II sequences. RT-PCR analysis of Sbe1-uidA::nptII chimeric mRNA produced in transgenic wheat cultured cells confirmed that the isolated Sbe1 was able to produce all three forms of SBEI gene transcripts by alternative splicing of the primary mRNA. The variants of processed Sbe1 mRNA were potentially translated into N-terminal variants of the SBEI precursor with different transit peptide sequences.

    Notizen: Abstract In order to investigate the effects of soil texture on possible non-hydraulic signals under field conditions, spring wheat plants (Triticum aestivum L. cv. Cadensa) grown in sand and loam soils and with a well developed root system were exposed to slow soil drying in the late vegetative stage of growth. Soil water potential and content were measured daily at different depths and plant responses were measured in flag leaves. When the average soil water potential in the top soil layers (0–25 cm depth in sand and 0–45 cm depth in loam) dropped to –60 or –70 kPa and the lower soil layers were still at field capacity, morning xylem [ABA] (0.03–0.04 vs. 0.06–0.08 mmol m-3) and midday leaf ABA concentration increased (250–300 vs. 400–450 ng/g DW) and leaf conductance decreased relatively to well-watered (control) plants (0.75–0.88 vs. 0.64–0.70 mol m-2 s-1). These responses took place before any decrease in leaf water potential occurred as compared with control plants, indicating that they were triggered by root-borne signals due to reduced root water status in the top soil layers. At this stage the soil water content was as low as 6% by volume, the fraction of roots in ‘wet’ soil was 0.12 and relative available soil water was 45% in sand and still high 20%, 0.48 and 70%, respectively, in loam of the whole soil profile indicating that roots were responding to soil water availability and not soil water content at a certain evaporative demand. In addition, similar responses occurred at high and low evaporative demands (3.4–5.2 vs. 0.6–4.0 mm/day of potential evapotranspiration).
