    Beschreibung / Inhaltsverzeichnis: Zusammenfassung Ceutorhynchus constrictus Marsh. (Coleoptera: Chrysomelidae: Ceutorhynchinae) ist ein monophager Rüsselkäfer, der an Knoblauchhederich frisst. Das Wirtswahl-Verhalten dieses Käfers ist im Labor untersucht worden. Die meisten Crucifiren waren im Wahlversuche nicht akzeptiert, wenn Knoblauchhederich als Vergleichspflanze vorhanden war. Von Brassica nigra, Sinapis alba, und Thlaspi arvense wurden im Vergleich gleiche Mengen verzehrt wie von der Wirtspflanze. Blühende Descurainia sophia Pflanzen wurden, im Gegensatz zu Jungpflanzen der gleichen Art, angenommen. Die wichtichsten Phagostimulanten in Extrakten von Knoblauchhederich-Blättern waren ungeladene, wasserlösliche Substanzen. Das häufigste Glukosinolat im Knoblauchhederich, Sinigrin, war auch ein Phagostimulant. Doch war die phagostimulierende Wirkung von Sinigrin nur in Kombinationen mit noch nicht identifizierten, ungeladenen Substanzen aus Knoblauchhederich-Blätter nachweisbar. Wirtspfanzen-Beziehungen von monophagen Insekten werden diskutiert im Zusammenhang mit der Eigenart des Glukosinolat-Inhaltes ihrer Wirtspflanzen.
    Notizen: Abstract Host plant relations of the monophagous weevil Ceutorhynchus constrictus Marsh. (Coleoptera: Curculionidae: Ceutorhynchinae) feeding on garlic mustard, Alliaria petiolata (Bieb.) Cavara & Grande (Cruciferae) were studied in the laboratory. Most other crucifers were rejected in choice tests using garlic mustard as a reference plant, but Brassica nigra, Sinapis alba and Thlaspi arvense were as acceptable as the host plant. Flowering plants of Descurainia sophia were acceptable while young plants of this species were not. The most important feeding stimulants in extracts of garlic mustard were uncharged, water soluble compounds. The most abundant glucosinolate in garlic mustard, sinigrin, was a feeding stimulant, too. However, the feeding stimulatory activity of sinigrin was only expressed in the presence of still unidentified uncharged compounds from garlic mustard leaves. Host plant relations in monophagous crucifer-feeding insects is discussed in relation to the distinctness of glucosinolate patterns found in their host plants.

La répulsion a été associée à l'oeuf lui-même et non à une autre activité de l'adulte sur la graine. Des stimuli tant chimiques que physiques (tactiles) ont semblé impliqués dans la découverte des oeufs; la substance répulsive était plus soluble dans le méthanol que dans l'éther ou l'eau. Contrairement à des études antérieures sur C. chinensis, nous avons observé que les femelles n'étaient pas dissuadées de pondre sur des graines exposées préalablement à de fortes densités de mâles.
    Notizen: Abstract Several experiments were conducted to determine the mechanism by which ovipositing cowpea weevils, Callosobruchus maculatus (F.), avoid egg-laden seeds. Deterrence was associated with the egg itself and not with other adult activities on the host. Both chemical and physical (tactile) stimuli appear to be involved in egg recognition; the chemical deterrent was more soluble in methanol than in ether or water. Contrary to a previous study using C. chinensis (L.), we found that females were not deterred from ovipositing on seeds that had been exposed to high densities of males.

    Notizen: Abstract A field experiment with Drosophila melanogasterrevealed that when flies encounter a particular food type soon after emergence, the probability of their subsequently being attracted to such a resource is increased. In this experiment, the length of time flies experienced their postemergence environments was under the control of the flies themselves. The experiment thus realistically mimicked one form of experiential effect that may be important in nature. A theoretical model is developed which shows that enhanced adult preferences for the types of resources fed on as larvae can substantially increase the degree of host-based genetic subdivision within a polyphagous population.

    Beschreibung / Inhaltsverzeichnis: Résumé Les choix alimentaires d'un groupe de chrysoméles spécialistes ou généralistes a été étudié en utilisant 3 méthodes: plantes entières, feuilles coupées, rondelles de feuilles. Les méthodes utilisées out eu un effet très significatif sur les préférences relatives des chrysomèles du maïs, du haricot et de la courge. Dans quelques cas le seuil de signification statistique pour un choix particulier a été modifié par la méthode utilisée et dans d'autres cas l'ordre de préférence a été totalement inversé. Les choix alimentaires des spécialistes ont été moins affectés par la méthode utilisée que pour les espèces les plus généralistes. Il y a eu aussi un effet plus important des différences entre les méthodes rondelle de feuilles et feuille entière qu'entre feuille entière et plante entière. Les changements chimiques induits dans les plantes par les dégâts sont probablement très répandus et les chercheurs devraient en tenir compte quand ils programment et interprètent des expériences sur les préférences alimentaires.
    Notizen: Abstract Feeding preferences of a group of specialist and generalist chrysomelid beetles were examined using three test methods: whole plants, excised leaves and leaf discs. Method of testing had a very significant effect on relative preferences of the beetles for corn, bean and squash. In some cases the level of statistical significance for a particular preference was affected by test methodology and in other cases the direction of preference was reversed altogether. The feeding preferences of the specialist beetle species were less affected by test method than were the more generalist species. There was also a much greater effect of the difference in test method between the disc test and whole leaf test than between the whole leaf test and whole plant test. Chemical changes in plants induced by damage are probably quite widespread in plants and investigators should take this into account when designing and interpreting tests of insect feeding preference.

Le (R*,S*)-1-éthylpropyl 2-méthyl-3-hydroxypentanoate a été identifié en 1987 comme le principal composé du sitophilate, la phéromone mâle d'agrégation de S. granarius. La composition énantiométrique du sitophilate a été déterminée par 3 méthodes: 1) tests biologiques des énantiomères synthétiques (2S,3R) et (2R,3S) du diastéréomère actif (R*,S*); 2) spectrométrie RMN 1H des esters Mosher dérivés de la phéromone naturelle et des sitophilates de synthèse (2S*,3R*)-et (2R*,3S*); 3) comparaison en capillarité GLC des temps de rétention des dérivés naturels de la phéromone et des 2 éniantiomères de synthèse. La combinaison des 3 méthodes confirme que le (2S,3R) énantiomère est la forme active du sitophilate. Le mâle produit 〉96% de l'énantiomère (2S,3R). Il n'y a pas eu attraction de S. granarius par le (2R,3S) sitophilate. S. oryzae L. et S. zeamais Motsch n'ont pas été attirés par le (2S,3R)-sitophilate. L'utilisation du (2S,3R)-1-éthylpropyl 2-méthyl-3-hydroxypentanoate dans les pièges devrait permettre une détection précoce de la présence de S. granarius dans des stocks de grains.
    Notizen: Abstract The enantiomeric composition of sitophilate, the granary weevil [Sitophilus granarius (L.)] male-produced aggregation pheromone [(R*,S*)-1-ethylpropyl 2-methyl-3-hydroxypentanoate)], was determined by three methods: (1) bioassaying the synthetic (2S,3R) and (2R,3S) enantiomers of the active (R*,S*) diastereomer; (2) 1H NMR spectroscopy of Mosher ester derivatives of the natural pheromone and synthetic (2S,3R)-and (2R,3S)-sitophilate; and (3) capillary GLC comparisons of the retention times of derivatized natural pheromone and the two synthetic enantiomers. The combined methods confirmed the (2S,3R) enantiomer as the active form of sitophilate. Male granary weevils were shown to produce 〉96% (2S,3R)-sitophilate. No significant attraction of S. granarius by the (2R,3S) enantiomer was observed. Rice and maize weevils [S. oryzae (L.) and S. zeamais Motschulsky] were not attracted by (2S,3R)-sitophilate.

    Beschreibung / Inhaltsverzeichnis: Résumé Les mécanismes de défense de la pomme de terre sauvage, S. berthaultii Hawkes, aux larves de Leptinotarsa decemlineata Say, ont été étudiés par ablation sélective des trichomes glandulaires et par l'élimination de leur exsudat des folioles, et par comparaison avec S. tuberosum L. qui a perdu les trichomes glandulaires défensifs A et B. L'ablation des trichomes A a augmenté la proportion de larves ayant consommé S. berthaultii. L'élimination de l'exsudat des trichomes B a augmenté la proportion de consommatrices et réduit la mortalité. Les principaux composés actifs de l'exsudat B, c'est-à-dire des esters d'acides gras de sucrose, n'étaient actifs qu'en présence de trichomes A. Les esters de sucrose n'ont pas modifié la consommation larvaire sur folioles de S. tuberosum, ou sur disques de feuilles de S. berthaultii dont les trichomes A avaient été enlevés. La croissance des larves survivantes n'a pas été modifiée significativement par l'ablation des trichomes A ou l'élimination de l'exsudat de B. La croissance des larves a été significativement augmentée quand les folioles de S. berthaultii ont été incorporés dans l'aliment artificiel après élimination de la barrière physique due aux pédoncules B. La croissance a été de même importance sur aliments artificiels contenant des feuilles (fraiches ou en poudre lyophylisée) de S. berthaultii ou de S. tuberosum, mais plus faible que sur folioles de S. tuberosum. La présence de trichomes A est indispensable à la résistance de S. berthaultii aux L, de L. decemlineata. Les gouttelettes de type B contenant des esters de sucrose augmentent l'expression de la résistance en présence d'une défense active par trichomes A.
    Notizen: Abstract The defensive mechanisms of the wild potato, solanum berthaultii Hawkes, to larvae of the Colorado potato beetle, Leptinotarsa decemlineata (Say), were studied by selective removal of glandular trichomes and trichome exudates from leaflets, and by comparing performance on S. berthaultii and on the cultivated potato, S. tuberosum L., which lacks defensively active type A and B glandular trichomes. Removal of type A trichomes increased the proportion of larvae that fed on S. berthaultii. Removal of the exudate from type B trichomes increased the proportion of larvae that fed and led to a decrease in mortality. The predominant active compounds in type B exudate, i.e. fatty acid esters of sucrose, were only effective in the presence of type A trichomes. Sucrose esters did not affect larval feeding on S. tuberosum leaflets or on S. berthaultii leaf discs from which the type A trichomes had been removed. Growth of surviving larvae was not significantly affected by removing type A trichomes or type B exudate. Growth of larvae was significantly increased when S. berthaultii leaflets were presented in artificial diet which eliminated the physical barrier of the type B stalks. Growth was no different on artificial diet containing either S. berthaultii or S. tuberosum leaf material (fresh or lyophilized powder) but was poorer on these diets than on S. tuberosum leaflets. The presence of type A trichomes is a fundamental requirement for expression of S. berthaultii resistance to L1 L. decemlineata. Type B droplets containing sucrose esters increase the expression of resistance in the presence of defensively-active type A trichomes.

The beetles ‘reflex bleed’ when disturbed, thereby deploying the alkaloid, which is provenly deterrent to spiders and ants. Newly emerged adults lack the alkaloid, but the compound builds up to deterrent levels in their blood within days. Eggs and larvae ofEpilachna are devoid of the compound.

    Beschreibung / Inhaltsverzeichnis: Résumé Les allozymes d'Hypera de l'est des USA (Beltsville, Maryland; Washington Co., Illinois), de l'ouest des USA (Logan et St Georges, Utah) et ‘égyptiens’ (Yuma, Arizona; Westmorland, Californie) ont été analysés par électrophorèse sur gel d'acrylamide. L'étude a porté sur 22 loci de 12 enzymes (ACPH, ADH, AMY, AO, EST, GOT, G-6PDH, MDH, ME, SOD, TYR, XDH). L'hétérozygotie moyenne de la population était 0.231, avec une moyenne de loci polymorphes de 0.536. La distance génétique moyenne de l'ensemble des populations était de 0.033 et l'indice de fixation de 0.024. Des loci caractéristiques ont été trouvés qui pourraient permettre de distinguer les Hypera occidentaux des orientaux et des ‘égyptiens’. La faible distance génétique entre les Hypera orientaux et ‘égyptiens’ suggère qu'ils appartiennent à la même souche et sont certainement différents des occidentaux. A partir de cela et d'autres éléments, nous concluons que tous les Hypera des USA sont H. postica Gyllenhal et que l'utilisation d'H. brunneipennis Boheman pour désigner les Hypera ‘égyptiens’ d'Amérique du Nord doit être abandonnée.
    Notizen: Abstract Allozyme profiles of eastern weevils (Beltsville, Maryland; Washington Co., Illinois), western weevils (Logan and St. George, Utah), and Egyptian weevils (Yuma, Arizona; Westmorland, California) were compiled by acrylamide gel electrophoresis. Twenty-two gene loci from 12 enzymes (ACPH, ADH, AMY, AO, EST, GOT, G-6PDH, MDH, ME, SOD, TYR, XDH) were analyzed. Mean heterozygosity of these populations was 0.231, with an average proportion of polymorphic loci of 0.536. The mean genetic distance of all weevil populations was 0.033 and the fixation index was 0.024. Diagnostic loci were found which could distinguish western weevils from eastern and Egyptian weevils. The small genetic distance between the eastern and Egyptian weevils suggests that they may be the same strain and are certainly different from the western weevil strain. Based on this and other evidence, we conclude that all weevil strains in the United States are Hypera postica (Gyllenhal), and that the use of H. brunneipennis (Boheman) for the Egyptian alfalfa weevil of North America should be discontinued.

    Notizen: Abstract Alfalfa weevils (Hypera postica (Gyllenhal)) with vestigial hind wings were discovered in a population from Wageningen, the Netherlands, and two populations from the United States—an eastern weevil strain from Beltsville, Maryland and an Egyptian weevil strain from Atascadero, California. Such a mutant was absent from 23 other populations surveyed in the United States—three from eastern, seven from western, and 13 from Egyptian weevil strains. This mutation is due to a dominant autosomal gene with normal-wing individuals as recessive. The mutant gene can be transferred from eastern weevil to the western weevil strain. The short-wing trait may be useful for genetic manipulation to control the alfalfa weevil.

(Carabidae) a été effectuée dans deux parcelles de 4 ha en utilisant des pièges distribués sur une grille de 10 sur 10. Pendant la première année d'échantillonnage, Lymantria dispar L. (Lymantriidae) était abondant et les calosomes se sont reproduits, tandis que la seconde année les papillons étaient beaucoup moins nombreux. La distribution des larves de carabes a été examinée aussi pendant la première année sur l'une des stations, et celle des chenilles pendant les deux années avec des toiles d'emballage sur l'autre station. Les dispersions des adultes et larves de calosomes et des chenilles de Lymantria ont été calculées en utilisant la régression du logarithme naturel de la variance de l'échantillon sur le logarithme naturel de la moyenne (méthode de Taylor) et la régression de l'indice de Lloyd d'agrégation moyenne sur la moyenne (méthode d'Iwao). La tendance à l'agrégation est plus forte chez les mâles (d'après la pente du logarithme de la moyenne par rapport au logarithm e de la variance de l'échantillon). La migration et le degré d'agrégation paraissent varier ensemble dans une station, tandis que dans l'autre une telle relation n'apparaît pas clairement. Les femelles malgré des taux de migration variés mais généralement faibles, n'ont jamais pr'esenté de tendance significative à l'agrégation. A l'opposé, les larves de calosomes et de Lymantria ont presenté des distributions agrégatives.
    Notizen: Abstract Mark-release studies of Calosoma sycophanta L. (Carabidae) in two 40000 m2 areas were done using traps deployed in 10 by 10 grids. In both areas, gypsy moths, Lymantria dispar L. (Lymantriidae), were abundant the first but not the second year of sampling. The distribution of carabid larvae was also investigated in the first year at one of the sites and the distribution of gypsy moth larvae found under burlap bands determined for 2 years at the other site. Dispersions of beetles and prey were evaluated by Taylor's logarithmic meanvariance method and Iwao's mean-mean crowding method. Taylor's method gave the most consistent results, and the slopes of the regression lines were used to determine degree of clumping. To evaluate dispersal of adult beetles, recapture data were analysed using the Fisher-Ford and Jolly-Seber methods in conjunction with Jackson's method for separating survival and emigration. A new method for estimating emigration was found to be most consistent with the Fisher-Ford-Jackson procedure. The tendency of the beetles to aggregate (measured via Taylor's method) was generally highest for males. In one site, emigration and degree of aggregation for males appeared to vary together, while at the other site no relationship was evident. Females showed few tendencies to aggregate. Both beetle larvae and gypsy moth larvae had clumped distributions.

No significant utilization of ethanol or acetaldehyde occurred at low concentrations (〈 1.5% and 0.1% respectively), however both female and male longevity was increased at concentrations of 1.0 and 1.5% acetic acid.     Notizen: Abstract The role of light intensity and temperature in determining the onset of flight in the crepuscular dung beetle Onitis alexis Klug (Coleoptera: Scarabaeidae) was examined. Flight under natural light was highly synchronized, with two-thirds of the beetles that flew flying over a period of 10–12 min. In six flights on different days, the mean time of onset varied by up to 13 min, but mean onset occurred at fixed light intensity. Absolute light intensity therefore appears to be a vital cue in determining flight onset. Mean onset remained at this intensity when dusk was advanced artificially by up to about 8 min. However, when dusk was brought further forward, mean flight occurred at lower intensities and onset of flight took place over a longer period. This is interpreted as an overlapping of the period of light intensities suitable for flight with the circadian rhythm that brings the beetles to the surface. No beetles flew when kept under constant bright light or in the dark during the dusk period. Under conditions of constant dusk, the beetles appeared uncoordinated and, although the median time of onset of flight was only 3 min later than in the control flight, onset of flight was spread out over a much longer period. At soil temperatures of 20–22 °C, over 90% of the beetles flew. This percentage decreased with decreasing temperature, and less than 10% flew at temperatures of 16–17 °C.

    Notizen: Abstract A model is presented that describes the reemergence of parent spruce bark beetles, Ips typographus, based on breeding density and temperature conditions. Laboratory data obtained at one constant temperature provide the distribution of reemergence time and different combinations of threshold temperatures and degreedays (°D) at mean reemergence. Of these combinations, 167.71 °D above 7.5 °C gave the best correspondence between model prediction and field data obtained from densely infested standing trees. The model was in good agreement with the reemergence pattern found for two groups of infested logs under different temperature conditions when adjusted for the lower breeding density. The applicability of the model in the study of bark beetle reproductive biology and in forest management is discussed. The method used to construct the model should be useful in developing other models for poikilothermic processes.

Le comportement des deux espèces a été observé en filmant en vidéo des carabes à jeun, maintenus à différentes températures constantes, dans des enceintes semées en orge de printemps. Des éléments du comportement, communs aux deux espèces, ont été définis: 1) immabilité, 2) marche et course, 3) recherche, 4) affrontement. P. cupreus a été plus actif à toutes les températures, B. lamprosa été inactif au-dessous de 10°C. La part de temps consacrée à la recherche, le nombre de plantes prospectées, et la vitesse ont augmenté avec la température chez les deux espèces. Dans des enceintes similaires colonisées par R. padi, P. cupreus a significativement augmenté le temps consacré à la recherche dans les enceintes, parallèlement à l'augmentation de la densité des pucerons. Après la découverte d'une colonie de pucerons, P. cupreus escalade et prospecte la plante et ses voisines immédiates; tandis que les plantes des enceintes sans pucerons sont rarement escaladées.     Notizen: Abstract Maximum consumption rates were determined for two carabids, Bembidion lampros Herbst. and Pterostichus cupreus L., feeding on the cereal aphid Rhopalosiphum padi L. at different temperatures in the laboratory. Mean daily consumption increased with increasing temperature for both species, B. lampros consuming a maximum of 16 1–3 instar nymphs and 9 apterous adult aphids at 25°C. P. cupreus was particularly voracious and consumed 125 apterous adult R. padi per day at 20°C. The behaviour of both species was analysed by video filming starved beetles, maintained at different constant temperatures, in arenas sown with spring barley. The behavioural components (1) still; (2) run/walk; (3) search and (4) confrontation were identified and were common to both species. P. cupreus was more active over the temperature range tested; B. lampros was inactive under 10°C. The proportion of time spent searching, number of plants searched, and velocity increased with increasing temperature for both species. When observed in similar arenas seeded with R. padi colonies, individuals of P. cupreus significantly increased their time spent searching in arenas with increasing aphid density. Following discovery of an aphid colony, individuals climbed and searched the host plant and its nearest neighbours. Plants in aphid free arenas were rarely climbed. B. lampros was not observed climbing in either aphid free arenas or in arenas with increasing aphid densities, and did not significantly increase its time spent searching in response to increased prey density. The few B. lampros that found aphids caught them walking on the soil surface. The relative efficiences of these two carabids as predators of R. padi are discussed, and the results are compared with similar studies elsewhere with predators of Sitobion avenae on winter wheat.

Les aliments durs seuls usent moins le bord incisif (extrémité de la mandibule) et la surface coupante, que leur association avec le fouissage. En présence exclusive d'aliments durs, les femelles avaient érodé plus vite leurs mandibules que les mâles, tandis que chez ceux-ci le fouissage seul les avaient usé plus vite que chez les femelles. Des femelles récoltées dans un champ de céréales contenaient plus d'aliments durs que les mâles. Les différences de comportement avaient provoqué apparemment des types d'usure différents suivant les sexes. Les adultes usent leurs mandibules au cours de leur première année de vie imaginale. Cependant, la taille des mandibules des femelles échantillonnées dans la nature correspondait mieux à celle des femelles (de même âge) provenant des expériences ayant provoqué une forte usure (aliment dur et fouissage du sol). Les mandibules des mâles échantillonnés présentaient seulement une usure modérée et ressemblaient à celles des mâles ayant consommé soit des aliments durs, soit foui le sol. La discussion a traité des relations entre l'usure des mandibules, la fécondité et la longévité de P. melanarius et d'autres espèces de carabes.
    Notizen: Abstract The effects of food texture and burrowing on mandible wear in the predatory carabid beetle Pterostichus melanarius Illiger were investigated by feeding adults soft or hard food in jars with or without soil. Both the incisor cusp (mandible tip) and cutting surface of the mandibles erode more in response to the combination of hard food and burrowing than to hard food alone. Females were found to erode their mandibles more than males in the hard-food-only treatment, whereas males wore down their mandibles more than females in the soil-only treatment. Female P. melanarius collected in a cereal field contained signficantly more solid food items compared with males. Differences in behaviour apparently resulted in differential wear patterns between the sexes. It is shown that the process of mandible wear in P. melanarius will result in worn mandibles in their first year as adults. However, mandible size of field-sampled females were found to be best correlated with females (of similar age) in the experiment exhibiting extensive wear (foraged hard food and burrowed in soil). The mandibles of field-sampled males were similar to those of males in the hard-food-only and the soil-only treatments, which showed only slight to moderate wear. The influence of mandible wear on fecundity and survival in P. melanarius as well as in other carabid species is discussed.

La température de 23 °C a été défavorable aux larves récoltées en septembre dans des souches de chênes et élevées sur aliment B: les larves étaient mortes en 30 jours. Avec aliment B+C, l'effet défavorable a été neutralisé et le poids des larves augmenté de 543%, 897% et 1179% en 1, 2 et 3 mois. Des larves néonates d'été ou d'hiver se sont bien développées sur B+C, mais toutes les larves d'hiver étaient mortes sur B en 40 j. Des larves de mêmes parents, écloses à différentes époques de l'année, ont présenté des différences saisonnières du taux de survie et de la vitesse de développement lors de leur élevage à 23 °C sur B+C.     Notizen: Abstract The effects of constant temperature (23 °C), food quality (B, natural diet i.e. powdered oak bark; B+C, natural diet enriched with powdered sweet crackers) and season on larval development of the cerambycid Morimus funereus L. were studied. The temperature of 23 °C exerted an unfavourable effect on larvae collected from oak stumps in September and bred on B; i.e. the larvae died within 30 days. When the larvae consumed B+C the unfavourable effect of temperature was abolished and the larvae increased their weight by 543%, 897% and 1179% in 1, 2 and 3 months, respectively. Newly hatched summerand winter-larvae developed successfully on B+C, while all the winter-larvae died within 40 days on B. Larvae of an identical parentage hatched in different phases of the annual cycle and showed seasonal differences in the rate of development and survival when reared on B+C at 23 °C. The fastest development and the highest survival rate were observed in larvae which emerged in early- and midsummer, whereas the slowest development and the lowest survival rate were in those hatched in late summer. The synchronized seasonal changes of adults and their offspring, as expressed at different levels of biological organization, suggest the existence of an endogenous annual rhythm which is dependent upon the expression of different genes in the course of the annual cycle.

    Notizen: Abstract The concept of encapsulating semiochemicals into a starch matrix is being studied for potential use in corn rootworm (CRW) management programs. During 1987, experiments were conducted to determine: 1) If volatile plant-derived Diabrotica spp. attractants could be encapsulated in a starch borate matrix (SBM), and 2) If various SBM-semiochemical formulations would attract Diabrotica species over time in field corn. Chemical analyses of fresh SBM formulations indicated that indole, estragole, veratrole, phenylacetaldehyde, and trans-anethole were not retained during formulation but trans-cinnamaldehyde, Beta-ionone, 1,2,4,-trimethoxybenzene, eugenol and isœugenol were successfully encapsulated. Encapsulated semiochemical formulations were made into 20 mesh granules, placed in Pherocon ® 1C traps that were tied to corn plants, and sampled for CRW adults every 4 days from 11 July to 8 September. Field data indicated that encapsulated semiochemicals were retained in the SBM for varying lengths of time and were released at rates attractive to CRW adults. A two-component mixture of trans-cinnamaldehyde and 1,2,4-trimethoxybenzene was the most effective formulation tested; however, no formulation was effective during corn silking and pollination. Although seasonal variation in CRW response could limit the usefulness of some plant-derived semiochemicals, the starch matrix concept may be useful as a delivery system for semiochemicals and may have potential as a tool that could be used in the development of new more biorational CRW management programs.

    Notizen: Abstract Flight activity of corn rootworm beetles, the northern (NCR), Diabrotica barberi (Coleoptera: Chrysomelidae), and western (WCR), D. virgifera virgifera, was monitored using malaise traps placed at the edges of two small cornfields. Populations of beetles in the fields peaked while corn was flowering, but capture in malaise traps remained low until silks had dried. Capture of NCR and (to a lesser extent) WCR then increased and remained high through the season. Malaise traps were constructed to permit separate collections of beetles that entered on each of two opposite sides. Female NCR that were captured in sides that faced toward corn tended to contain fewer mature eggs than those in sides facing away from corn. These findings support the hypothesis that female NCR emigrate from cornfields to feed when fresh silk and corn pollen (favored foods) become unavailable, but that egg-laden NCR actively seek corn when searching for oviposition sites.

    Beschreibung / Inhaltsverzeichnis: Zusammenfassung Mit eine quantitativen enzymabhängigen Immunoabsorptionstest wurde immunoreaktives Material aus Beutetieren als Eiweissgehalt (IRM/P) in Larven eines polyphagen räuberischen Carabiden, Poecilus cupreus, bestimmt. Wir bestimmten den Beuteverzehr und die Geschwindigkeit des IRM/P Abbaus bei Larven, die während verschiedener Zeit gehungert hatten. Der Beuteverzehr nahm exponentiell zu über 7 Tage Hunger. Die Geschwindigkeit des Beute- IRM/P Abbaus schien logarithmisch. Die Beuteaufnahme nahm fast zwanzigfach zu über 7 Tage Hunger von 2.7 ng auf 50.2 ng IRM/P/10 Mikroliter Larvenextrakt. Der grösste Teil der Verdauung erfolgte am zweiten bis dritten Tag nach der Nahrungsaufnahme, unabhängig von der verdauten Menge. Das Niveau 4 Tage nach der Fütterung zeigte, dass Larven, die vorgängig 7 Tage gehungert hatten, 5.0 ng IRM/P/10 Microliter Extrakt zurückhielten; dies war mehr als die Aufnahmemenge nichthungernder Larven. Die Verdauungsgeschwindigleit nahm zu mit der Dauer des Hungerns, doch wurde auch mehr Beute im Nahrungskanal zurückgehalten.
    Notizen: Abstract Prey antigen levels were determined by a quantitative enzyme-linked immunosorbent assay in larvae of a carabid predator, Poecilus cupreus L. under laboratory conditions. We determined prey consumption and rate of prey protein decay in larvae having starved for different periods. Prey consumption increased exponentially over 7 days of starvation. The rate of prey immunoreactive material decay seemed to be logarithmic, similar in larvae which had starved for different periods. Most of the digestion took place in the first 3 days period after feeding, irrespective of the amounts ingested. The rate of decay increased with increasing period of starvation. There were also signs of food retention in the alimentary canal in larvae which starved longer.

    Beschreibung / Inhaltsverzeichnis: Résumé Vingt-deux lignées deC. maculatus Fab., liées à 6 plantes-hôtes, ont été examinées quant à leurs préférences — sans choix—parmi les graines de 9 légumineuses. Ces préférences se sont révél'ees semblables à celles observées lors de choix binaires; 3 paramètres ont donné les mêmes résultats: nombre moyen d'oeufs pondus par l'ensemble des femelles, nombre moyen d'oeufs émis par l'ensemble de celles acceptant la plante, fréquence des femelles acceptant la plante. L'analyse en composantes principales des préférences pour les 9 plantes a révélé que la majorité des mesures se situe sur les 2 premiers axes, qui ont rendu compte de 89% de la variation. Les performances des lignées sur le premier axe suggèrent qu'il s'agit d'un critère général de préférence. La classification automatique des plantes a montré un aspect général semblable à celui des composantes principales. Par ces 2 méthodes, les plantes ne se sont pas classées nettement selon leurs affinités taxonomiques. Bien que la classification automatique n'ait pas mis en évidence une similarité de comportement chez les insectes liés au même hôte, ceci était apparemment le cas. Les lignées liées à des hôtes présentant de fortes — ou à l'opoosé — de faibles préférences, ont montré différents seuils d'acceptation pour les 9 substrats de ponte. A l'opposé, il n'y a aucune preuve d'altération évolutive de la hiérarchie de préférences. On en a déduit que ces insectes étaient motivés par un petit nombre de stimulus, tels que les allélochimiques et al texture de tégument, pour déceler l'adéquation des substrats potentiels. (La microscopie électronique a suggéré une corrélation entre la texture du tégument et la hiérarchie des préférences de ces populations). La confiance dans les stimulus recueillis contribue à limiter les stratégies évolutives disponibles pour la population. A partir de là, la sélection en faveur —ou contre — des plantes, à l'origine, faiblement préférées, peut conduire, dans la niche de ponte, à l'incorporation — ou à l'exclusion — d'un groupe d'espèces taxonomiquemment éloignées, mais phénétiquement semblables. La probabilité d'altérations importantes de la hiérarchie des préférences paraît extrémement faible.
    Notizen: Abstract A survey of 22 strains ofCallosobruchus maculatus associated with six different leguminous hosts indicates extensive genetic differentiation for oviposition preference. Female behavior suggests the existence of two distinct host preference groups among the nine oviposition substrates tested. Strains associated with plants in these two groups show distinct preference profiles. Genetic differentiation of oviposition preference is due to evolutionary modification of the threshold of acceptance of host plants, not to alteration of the preference hierarchy. From principal components analysis of these results, it is likely that very few independent plant traits govern oviposition preference.

    Beschreibung / Inhaltsverzeichnis: Summary In order to meet its sexual partner, Zabrotes subfasciatus must rely on the olfactory signals emitted either by the female or the male insect. The attractiveness of male and female odours were therefore studied and compared in an olfactometer. The behaviour of virgin and inseminated females and males, left in contact with the host plant's seeds or not, was studied in terms of the age of the imagos. In all cases examined, male odours did not attract female insects. Virgin females kept without seeds of the host plant did not attract males. On the other hand, the odour of virgin females left in contact with bean seeds significantly attracted virgin males. The females' attractive power varied according to their age: older females released a chemical cue that was more attractive for the males. Emission of the sex-pheromone stops after insemination of the females but begins again after a certain period of time. Results are discussed in terms of what is known on the reproductive regulation of this bruchid.

Le pouvoir attractif de l'odeur des mâles et des femelles sont donc étudiés et comparés par olfactométrie. Le comportement de mâles et de femelles vierges et inséminées, mis en contact des graines de la plante hôte ou non, est observé en fonction de l'âge des imagos. En aucun cas, les odeurs des mâles n'exercent une attraction sur les femelles. Les femelles vierges en absence de graines de haricot (Phaseolus vulgaris) n'exercent pas d'attraction sur les mâles vierges. Par contre les odeurs des femelles vierges mises en contact avec les graines de haricot sont très attractives pour les mâles vierges et varie en fonction de l'âge: il est plus affirmé chez les femelles plus âgées. L'émission de la phéromone sexuelle est interrompue après l'insémination des femelles mais reprend au bout d'un certain temps après celle-ci.

    Beschreibung / Inhaltsverzeichnis: Résumé Le concept de réponse optimale d'un parasite vis-à-vis de l'hôte le plus favorable pour son développement demeure surtout théorique et n'a pu être vérifié que dans les conditions de laboratoire. Nous avons montré que Drosophila melanogaster s'avère être, par rapport à D. simulans, l'hôte le plus favorable pour le développement du cynipide parasite Leptopilina boulardi. Une étude sur le terrain a démontré que ce parasite présente une réponse fonctionnelle densité dépendante vis-à-vis de D. melanogaster et non vis-à-vis de D. simulans, avec un effet de bascule. D'autre part, il s'avère que ce parasite exploite beaucoup mieux son hôte, en évitant le superparasitisme, ceci étant démontré au laboratoire et dans la nature. Enfin, il apparaît qu'il est capable d'allonger sa période de ponte lorsque cet hôte est rare, ce qui ne se produit pas avec D. simulans.
    Notizen: Abstract The hypothesis of optimal host species selection predicts that when a parasitoid has the choice between two host species, it will choose the species thay gives the best survival chances for its progeny. We confirmed this hypothesis by laboratory experiments with Leptopilina boulardi Barb. et al., a cynipid parasitoid which prefers Drosophila melanogaster Meigen (the host species most suitable for parasitoid survival) above D. simulans Sturt. As far as fitness parameters are concerned, the fertility of L. boulardi is higher with D. melanogaster; the egg laying can be spread out over a long period when this host is relatively scarce. This does not occur with D. simulans in which parasitic oviposition stops soon when this host is not abundant. Investigations of this foraging strategy were done under more complex natural conditions. We found that L. boulardi has a type III functional response with D. melanogaster only; furthermore, it seems that a switching effect may exist with this host. Parasitoid females appear to distribute their eggs more regularly on D. melanogaster, thus avoiding superparasitism. This seems to be independent of the relative frequency of this host. However, superparasitism of D. simulans did occur more frequently when this host was scarce.

    Beschreibung / Inhaltsverzeichnis: Résumé Le comportement de vol d'Ips calligraphus GERMAR (Scolytidae) a été examiné au laboratoire par une technique de vol captif. La génération parentale avait été artificiellement introduite, suivant la taille des adultes (1,6 à 2,1 mm) dans des morceaux de Pinus elliottii Engelm. variété elliottii au phloeme épais (TK=2,5 à 3,5 mm) ou fin (TN=0,5 à 1,5 mm) les larves se sont développées dans ces morceaux de pin. Le poids et la teneur en lipide des adultes obtenus à partir de TN étaient significativement inférieure à ceux de TK. Le poids des mâles les plus gros a été plus réduit que celui des femelles; les mâles les plus petits semblaient être moins efficaces dans l'initiation des attaques sur les arbres, et les femelles les plus petites ont eu une fécondité réduite. Les durées de vol quotidiennes pendant la vie des mâles TK vierges étaient en moyenne deux fois plus longues que celles des mâles TN. Les durées de vol quotidiennes étaient très variables: en moyenne moins de 20 minutes, mais la plus longue a été de 173 mn. En moyennes les adultes TN et TK ont commencé à voler, et volaient en plus grand nombre, le premier et le second jour après l'émergence. Ensuite la fréquence d'adultes volants et le pourcentage de surviveants, ont graduellement diminué jusqu'à tomber à zéro le neuvième jour. Les mâles TN et TK avaient des vols quotidiens plus longs et volaient environ 1 jour de plus que les femelles. L'activité de vol accrue des mâles reflète leur besoin de trouver dans la nature des arbres convenables; les vols des femelles peuvent généralement être plus brefs, étant plus directement orientés comme une réponse aux phéromones mâles. Les voiliers ont vécu environ 2 fois plus longtemps que les non-voiliers. Parmi les voiliers TK et TN il y avait une corrélation positive étroite entre le poids du corps et la durée totale des vols. Il n'y en a qu'une faible corrélation entre cette durée totale des vols et la longévité, suggérant un faible coût physiologique du vol. L'influence de cette durée des vols et du prélèvement consécutif de lipides sur la fécondité n'a pas été examinée. L'initiation au vol chez les individus fécondés a été sérieusement retardée (maximum le sixième jour) par rapport aux vierges, ce qui suggère un dégénérescence des muscles du vol associée à la reproduction, suivie d'une régénération importante, comme cela a été signalé chez d'autres Scolytes. Ces résultats contribuent à notre compréhension des facteurs conditionnant le comportement de vol et d'autres aspects fondamentaux de la biologie des insectes des pins. Une telle connaissance est nécessaire au développement d'une politique efficace de protection.
    Notizen: Abstract Rearing larvae of Ips calligraphus (Germar) (Coleoptera: Scolytidae) in bolts of typical slash pine, Pinus elliottii Engelm. var. elliottii, with ‘thin’ (TN; 0.5–1.5 mm) phloem (inner bark) relative to adult beetle body width significantly reduced body weight and lipid content of emerging adults compared with ‘thick’ (TK; 2.5–3.5 mm) phloem-reared adults. Unmated (pre-nuptial) TK beetles averaged 2-fold longer daily and life-time flight duration than TN beetles. Daily flights were highly variable, averaging 〈20 min (longest daily flight=173 min). TK and TN beetles initiated flight, and the greatest number flew, on the first or second day after emergence form their rearing bolt; % of beetles flying and survival gradually declined to zero by day 9. Males (both TK and TN) averaged 2.5 days of flight, ca. 1 day longer than females. Fliers lived ca. 2-fold longer than non-fliers. Among TK and TN fliers, significant but weak positive correlations occurred between body weight and lifetime flight duration. Poor correlation between lifetime flight duration and longevity suggests little physiological cost to flight, but effects on fecundity were not assessed. Flight by mated, egg-laying (post-nuptial) beetles was substantially delayed (peaking on day 6), suggesting degeneration and subsequent regeneration of flight muscles.

    Notizen: Abstract Laboratory tests with eggs of Diabrotica virgifera virgifera LeConte showed that during a 10-day hatching period, hatch of male eggs predominated on the first and second days, eggs of mixed sex, with ca. 1:1 ratio, hatched on the third and fourth days, and eggs hatching from the fifth to the tenth days were nearly all female. Overall, female eggs hatched a mean of 2.9 days later than male eggs. Not only did female eggs hatch later, but the time for posthatch development to the adult stage was 1.8 days longer for females. The later egg hatch and longer posthatch development for females resulted in female adults emerging a mean of 4.7 days later than male adults. Total adult emergence lasted 14 days; of this, males predominated during the first 5 days, and females predominated during the last 9 days. Males of D. v. virgifera appear to have evolved protandry (the tendency for males to emerge before females) by developing both a postdiapause embryonic stage and a combined larval and pupal stage of shorter duration.

L'une est inhibée, tandis que l'autre se comporte comme si elle était seule. Le signal provoquant ces réactions, qui peut traverser du papier métallique, est dû vraisemblablement aux vibrations de mastication des larves; ces dernières perçoivent ces signaux tout au long de leur vie active. La larve la plus âgée ne l'emporte pas obligatoirement, ce qui signifie que le signal indique plus que l'âge. En ayant eu son alimentation interrompue et en restant dans une galerie superficielle, la larve inhibée évite les affrontements et peut se développer lorsque la larve dominante meurt ou se nymphose, puisque la galerie de la larve inhibée n'avait pas été sectionnée. La stratégie de ponte réduit la fréquence de la compétition en uniformisant la distribution des oeufs; elle augmente les chances de survie des larves vaincues avec l'addition des oeufs sur les graines les plus grosses quand les disponibilités sont limitées. L'étude de l'évolution de ces adaptations a été possible puisque, comme on le sait, C. maculatus est génétiquement polymorphe pour les caractères déterminant le comportement de ponte des femelles et la compétition larvaire.
    Notizen: Abstract Larvae of the bean weevil C. maculatus feeding in a single bean apparently respond to vibrations from each other's chewing: one larva feeds normally, the other is inhibited. If the burrows of the larvae intersect, the inhibited larva dies. If the dominant larva pupates or dies without the burrows intersecting, the inhibited larvae then feeds and matures if enough food remains. Since females add second eggs to the largest beans after most beans carry a single egg, competition is most common in the largest available beans, precisely where inhibited larvae can benefit from avoiding a contest.

Les mâles eux aussi ont révélé une activité sexuelle réduite aux photophases courtes, c'est-à-dire qu'ils sont entrés comme les femelles en diapause reproductive. Les photophases critiques pour l'induction de la diapause des mâles et des femelles n'ont pas présenté de différences. Les diapause des mâles et des femelles s'achèvent même sous courtes photophases, mais la diapause mâle était quelque peu plus faible que la diapause femelle. La photophase critique et le taux de diapause ont varié en fonction de l'origine géographique dans l'espèce actuelle. Lors de croisements entre lignées diapausantes et non-diapausantes, la photophase critique et la durée de la diapause ont été héritées quantitativement. A partir de ces expériences et d'expériences précédentes de croisements (Kimura, 1983), quelques modèles de méchnisme d'induction de la diapause de cette espèce sont proposés.
    Notizen: Abstract In Drosophila triauraria Bock & Wheeler (Diptera: Drosophilidae) of which females were known to enter reproductive diapause at short daylengths, males also showed reduced mating activity at short daylengths, i.e., males as well as females entered reproductive diapause. The critical daylength for diapause induction did not differ between females and males. Both male and female diapause ended even under short daylengths, but the male diapause was somewhat weaker than the female diapause. The critical daylength and the diapause rate varied geographically in this species. In the cross between diapausing and non-diapausing strains, the critical daylength and the diapause duration inherited in a quantitative manner. On the basis of the present and previous crossing experiments, some models are proposed on the mechanism of diapause induction of this species.

    Beschreibung / Inhaltsverzeichnis: Summary Bruchidius atrolineatus (Pic) is a widely distributed bruchid in the Sahelian zone which shows a reproductive diapause during the dry season and part of the rainy season. Diapausing females do not produce vitellogenin and their ovaries are reduced to the germarium. Spermatogenesis is very much reduced and male accessory glands are inactive. When these insects were placed in the presence of inflorescences of Vigna unguiculata which were renewed daily, the reproductive diapause of both males and females was interrupted after 15–20 days. Vitellogenesis occurred in the females and spermatogenesis increased in the males whilst their accessory glands became functional. When diapausing bruchids, found in stores of on V. unguiculata seeds during the dry season, were placed near the host plant's inflorescences, diapause was also terminated. In all cases, diapause was not interrupted when the insects were offered dry pods of V. unguiculata in a water-saturated atmosphere. The pollen, which is hardly eaten by this bruchid, did not seem to stimulate termination of diapause. Sensory stimulations derived from the flowers or/and nutritional factors may be the cause of the development of the reproductive organs. After termination of the diapause the males showed normal sexual activity whereas female fecundity was rather low, at least in our experimental conditions. This type of reproductive regulation allows this sahelian bruchid to resume its sexual activity when the host plant's flowers appear in the field at the end of the rainy season. Then the beetles lay their eggs on the pods as soon as the pods are developed.

Il n'y a par contre aucune levée de la diapause chez des bruches placées en présence de gousses sèches de V. unguiculata dans une atmosphère saturée. Le pollen fort peu consommé n'a par contre aucun rôle. Cette régulation du cycle reproducteur de B. atrolineatus par les inflorescences de V. unguiculata permet l'émission des oeufs dès que les gousses commencent à se former à la fin de la saison des pluies.

    Beschreibung / Inhaltsverzeichnis: Résumé Un olfactomètre à tube en Y, dont la description est donnée, a été utilisé pour examiner les réactions d'I. typographus L. par la marche à la présence de phéromones de congénères creusant des galeries dans des bûches d'Epicéa (Picea abies L.). Les insectes dans les bûches ont été répartis en 7 classes séquentielles, depuis l'élaboration de la chambre nuptiale par le mâle (phases 1–3), l'accouplement (4), et la ponte jusqu'à ce que les galeries de ponte aient 5 cm (5–7). Les mâles dans les phases précoces (1–2) étaient attractifs pour les 2 sexes dès 0,5 à 2,5 h après leur introduction, et ils le restaient pendant plus de 2 semaines. Les femelles qui se déplacent par la marche ont préféré nettement les bûches avec des mâles vierges à des bûches non habitées. De même elles ont préféré des bûches avec des mâles unis aux femelles à des bûches sans Ips. Ayant à choisir entre des bûches infestées par des mâles vierges ou expérimentés, les femelles ont préféré nettement les mâles vierges. Les mâle ont présenté une plus faible attraction aussi bien pour les bûches avec mâles vierges qu'avec mâles expérimentés, ils n'ont pas manifesté de préférence entre les 2. Les résultats sont interprétés comme des réponses adaptatives des 2 sexes aux variations dans la libération de phéromones par les insectes pendant les phases successives d'attaque et de colonisation de l'hôte.
    Notizen: Abstract A Y-tube olfactometer is described which was used to assay the response of walking Ips typographus (L.) towards the pheromone from beetles excavating galleries in logs of Norway spruce [Picea abies (L.) Karst]. Beetles in the logs were categorized into seven sequential attack phases. Males in the early attack phases were attractive as early as 0.5 to 2.5 h after their introduction. These logs remained attractive for more than 2 weeks. Walking females strongly preferred logs with unmated males over uninfested logs and over logs containing males and females. They also preferred logs with males and females over uninfested logs. Responding males showed a weaker attraction than females to logs with unmated males. The results are interpreted as adaptive responses of the sexes to variation of pheromone release from beetles in the host during subsequent phases of attack and colonization.

    Notizen: Abstract The insect growth regulator Dimilin®, common name: diflubenzuron, applied at concentrations effective against the cabbage maggot Delia radicum L., did not affect the hatching of eggs of the staphylinid predator and parasite, Aleochara bilineata Gyllenhal. Applied externally in distilled water, Dimilin was non-toxic to first instar larvae, to larval instars developing within host puparia, and to adults. When Dimilin was dissolved in dimethylsulfoxide, then applied to the exoskeleton of host puparia, emergence of A. bilineata adults was suppressed. The conventional insecticide Furadan, at a concentration (0.001% in distilled water) equal to the lowest concentration of Dimilin used, was toxic to both eggs and larvae of A. bilineata.

Des niveaux intermédiaires d'activité ont été obtenus chez les insectes élevés sur maïs jaune, et les niveaux les plus faibles chez ceux élevés sur blé. Bien que les extraits préparés à partir d'orge présentent une activité inhibitrice de deux isoamylases purifiées de S. oryzae, les niveaux des inhibiteurs naturels α-amylase de ces deux enzymes sont environ respectivement 2,2 et 6,1 fois plus concentrés dans le blé. L'ingestion de ces inhibiteurs d'amylase et la formation d'un complexe enzyme inactive/inhibiteur avec l'amylase secrétée antérieurement, peut rendre compte de la plus faible activité de l'amylase chez les charançons consommant du blé. Le niveau d'amylase de S. oryzae est 2 fois plus élevé que celui de S. zeamais pour toutes les souches élevées sur un régime donné. Des niveaux d'activité significativement différents ont été trouvés suivant les souches pour chacune des deux espèces. Puisque l'amylase est la principale hydrolase digestive de ces espèces, l'intensité de la modification des teneurs en amylase par la consommation de céréales peut indiquer leur adéquation comme hôtes potentiels.
    Notizen: Abstract Rank transformation of specific activity values of α-amylase across four strains of Sitophilus oryzae (L.) and four strains of S. zeamais Motschulsky indicates that levels of these predominant enzymes are highest in adults feeding on hulled barley or long-grain brown rice. Intermediate activity levels are found in weevils feeding on yellow corn (maize) and lowest levels are found in wheat-fed weevils. Although extracts prepared from barley contain inhibitory activity against two purified isoamylases from S. oryzae, levels of the naturally-occurring α-amylase inhibitors against these two enzymes are about 2.2-fold and 6.1-fold, respectively, more concentrated in wheat. Ingestion of these amylase inhibitors and formation of an inactive enzyme:inhibitor complex with previously secreted amylase may account for the lower activity of amylase in weevils of both species feeding on wheat. Amylase levels across all strains feeding on a given diet are about 2-fold higher in S. oryzae than in S. zeamais. Significant differences in activity levels were also found between strains in both species. Since α-amylase is a predominant digestive hydrolase in these species, the degree to which cereal diets affect amylase levels may indicate their suitability as potential hosts.

    Notizen: Abstract The attraction of Tomicus piniperda (L.) (Coleoptera: Scolytidae) to Scots pine trees of low and higher vigor with and without previous attacks was monitored with sticky traps. Somewhat higher numbers of beetles were caught on low vigor trees than on trees of higher vigor, indicating differences in olfactory stimuly between the two classes of trees. Many more beetles were caught on trees attacked by T. piniperda and on trees with simulated bark beetle galleries (manually drilled holes) than on control trees with no or only a few attacks. The strong attraction of beetles to attacked trees is attributed to the beetles responding to host volatiles released from the galleries.

    Notizen: Abstract A bioassay was developed to quantify the feeding of adult sweet potato weevils, Cylas formicarius elegantulus (Summers) (Coleoptera; Curculionidae) on the foliage of four cultivars (Centennial, Jewel, Resisto and Regal) of sweet potato (Ipomoea batatas (L.) Lam.) (Convolvulaceae). Weevils fed along the leaf veins, preferring the lower to the upper leaf surface. Males and females had similar levels of feeding. Different levels of feeding by female weevils were observed between cultivars in dual-choice bioassays with Centennial, a susceptible cultivar in field-plot experiments, being most preferred and Resisto least preferred. However, these feeding differences were not observed in no-choice bioassays. Little difference was observed in the leaf surface chemistry of the four cultivars.

    Notizen: Summary Because natural populations ofDrosophila melanogaster are polymorphic for different allozymes of alcohol dehydrogenase (ADH) and becauseD. melanogaster is more tolerant to the toxic effects of ethanol than its sibling speciesD. simulans, information regarding the sensitivities of the different forms of ADH to the products of ethanol degradation are of ecological importance. ADH-F, ADH-S, ADH-71k ofD. melanogaster and the ADH ofD. simulans were inhibited by NADH, but the inhibition was relieved by NAD+. The order of sensitivity of NADH was ADH-F〈ADH-71k, ADH-S〈ADH-simulans with ADH-F being about four times less sensitive than theD. melanogaster enzymes and 12 times less sensitive than theD. simulans enzyme. Acetaldehyde inhibited the ethanolto-acetaldehyde activity of the ADHs, but at low acetaldehyde concentrations ethanol and NAD+ reduced the inhibition. ADH-71k and ADH-F were more subject to the inhibitory action of acetaldehyde than ADH-S and ADH-simulans, with ADH-71k being seven times more sensitive than ADH-S. The pattern of product inhibition of ADH-71k suggests a rapid equilibrium random mechanism for ethanol oxidation. Thus, although the ADH variants only differ by a few amino acids, these differences exert a far larger impact on their intrinsic properties than previously thought. How differences in product inhibition may be of significance in the evolution of the ADHs is discussed.

The members of this dispersed repetitive family have terminal inverted repeats that sometimes flank a central region. The inverted repeats of all the family members are homologous. The study of the distribution and conservation of the foldback elements in differentDrosophila species shows that this distribution is different from that of the hybrid dysgenesis systems (PM and IR). Sequences homologous to foldback elements were observed by Southern blots and in situ hybridization in all species of themelanogaster subgroup and in some species of themontium andtakahashii subgroups.     Notizen: Summary The sequence of the 16,019 nucleotide-pair mitochondrial DNA (mtDNA) molecule ofDrosophila yakuba is presented. This molecule contains the genes for two rRNAs, 22 tRNAs, six identified proteins [cytochrome b, cytochrome c oxidase subunits I, II, and III (COI-III), and ATPase subunits 6 and 8] and seven presumptive proteins (URF1-6 and URF4L). Replication originates within a region of 1077 nucleotides that is 92.8% A+T and lacks any open reading frame larger than 123 nucleotides. An equivalent to the sequence found in all mammalian mtDNAs that is associated with initiation of second-strand DNA synthesis is not present inD. yakuba mtDNA. Introns are absent fromD. yakuba mitochondrial genes and there are few (0–31) intergenic nucleotides. The genes found inD. yakuba and mammalian mtDNAs are the same, but there are differences in their arrangement and in the relative proportions of the complementary strands of the molecule that serve as templates for transcription. Although theD. yakuba small and large mitochondrial rRNA genes are exceptionally low in G and C and are shorter than any other metazoan rRNA genes reported, they can be folded into secondary structures remarkably similar to the secondary structures proposed for mammalian mitochondrial rRNAs.D. yakuba mitochondrial tRNA genes, like their mammalian counterparts, are more variable in sequence than nonorganelle tRNAs. In mitochrondrial protein genes ATG, ATT, ATA, and in one case (COI) ATAA appear to be used as translation initiation codons. The only termination codon found in these genes is TAA. In theD. yakuba mitochondrial genetic code, AGA, ATA, and TGA specify serine, isoleucine, and tryptophan, respectively. Fifty-nine types of sense codon are used in theD. yakuba mitochondrial protein genes, but 93.8% of all codons end in A or T. Codon-anticodon interactions may include both G-A and C-A pairing in the wobble position. Evidence is summarized that supports the hypothesis that A and T nucleotides are favored at all locations in theD. yakuba mtDNA molecule where these nucleotides are compatible with function.

The development of an indirect flight muscle, the dorsal longitudinal muscle (DLM), has been thought to be responsible for the formation of the distinct thoracic curvature.     Notizen: Summary Embryos of Drosophila melanogaster were irradiated in the presumptive head region with a UV-laser microbeam of 20 μm diameter at two developmental stages, the cellular blastoderm and the extended germ band. The ensuing defects were scored in the cuticle pattern of the head of the first-instar larva, which is described in detail in this paper. The defects caused by irradiating germ band embryos when morphologically recognisable lobes appear in the head region were used to establish the segmental origin of various head structures. This information enabled us to translate the spatial distribution of blastoderm defects into a fate map of segment anlagen. The gnathal segments derive from a region of the blastoderm between 60% and 70% egg length (EL) dorsally and 60% and 80% ventrally. The area anterior to the mandibular anlage and posterior to the stomodaeum is occupied by the small anlagen of the intercalary and antennal segments ventrally and dorsally, respectively. The labrum, which originates from a paired anlage dorsally at 90% EL, is separated from the remaining head segments by an area for which we did not observe cuticle defects following blastoderm irradiation, presumably because those cells give rise to the brain. The dorsal and lateral parts of the cephalo-pharyngeal skeleton appear to be the only cuticle derivatives of the non-segmental acron. These structures derive from a dorso-lateral area just behind the putative brain anlage and may overlap the latter. In addition to the segment anlagen, the regions of the presumptive dorsal pouch, anterior lobe and post-oral epithelium, whose morphogenetic movements during head involution result in the characteristic acephalic appearance of the larva, have been projected onto the blastoderm fate map. The results suggest that initially the head of the Drosophila embryo does not differ substantially from the generalised insect head as judged by comparison of fate map and segmental organisation.

By comparing the morphogenetic map derived from gynandromorphs and the fate map derived from data on the transplantation of fragments of the mature wing imaginal disc, it was observed that no major morphogenetic movements occur in the notum anlage between the stages of the allocation of the disc and the mature disc. The results are consistent with the observations of other authors concerning the larval development of eye-antenna, wing and leg discs.

Both antigens are present in control and 20-hydroxyecdysone treated imaginal discs, although apparent increases in antigen content are associated with hormone treatment. Immunofluorescent staining of whole discs with anti-P116 and anti-P93 reveals increased amounts of both antigens at the surface of hormone-treated discs compared to controls. Both antibodies were used to characterize the expression of their respective antigens during embryonic development, and both antibodies were found to recognize in embryos a third developmentally-regulated antigen with a relative mobility of approximately 220000. Our results indicate, at least in the case of P116 and P93, that 20-hydroxyecdysone-dependent cell surface antigens in imaginal discs may be regulated both by increasing the amounts of constitutively present proteins, and possibly through biochemical modifications, altering the localization of these proteins from a cytoplasmic to a cell surface domain.

    Notizen: Summary In this paper experiments concerning some aspects of the development of pole cells and midgut progenitors in Drosophila are reported. Cells were labelled by injecting horseradish-peroxidase (HRP) in embryos before pole bud formation and transplanted at different stages into unlabelled embryos, where the transplanted cells developed together with the unlabelled cells of the host. The hosts were then fixed and stained at different ages in order to demonstrate the presence of HRP in the progenies of transplanted cells. The main conlusions of the study are as follows. The gonads are the only organ to the formation of which pole cells normally contribute; those pole cells which do not participate in the formation of the gonads are finally eliminated or degenerate. Since the number of primordial germ cells in the gonads is the same irrespective of the number of pole cells present in the embryo, an (unknown) mechanism must exist regulating the final number of pole cells in each of the gonads. After their formation and before reaching the gonads, pole cells have been found to divide only up to two times. With respect to the midgut progenitors, the cells of both anlagen have been found to be committed to develop into midgut, although they behave as equivalent in that they do not apparently distinguish between the anterior and posterior anlage. Midgut progenitors have been found to divide a maximum of three times and to produce two different types of cells, epithelial cells of the midgut wall and spindle-like cells located internally in the gut.

Distinct classes of neurons are distinguishable according to their location, the targets they innervate, the particular shape of their dendrites and their cell size. Those neurons innervating external sensory structures (es) and chordotonal organs (ch) have single dendrites and have been previously described (Ghysen et al. 1986; Dambly-Chaudiere and Ghysen 1986; Campos-Ortega and Hartenstein 1985). We describe here the identity and morphological features of three other classes of neurons in the body segments which have multiple dendrites (md neurons): 1) neurons that give rise to elaborate dendritic arborisations (da neurons); 2) neurons that have bipolar dendrites (bd neurons); 3) neurons that arborize around particular tracheal branches (td neurons). The thoracic hemisegment (T2 and T3) contains 13 da, one bd, one td, 21 es and four ch neurons; the abdominal hemisegment (A1 to A7) contains 14 da, three bd, three td, 15 es and eight ch neurons. The arrangement of the segmented peripheral neurons is highly invariant and provides a favorable assay system for the genetic analysis of neurodevelopment.

Relative labeling was greater in eleven peptides and lower in three peptides of evaginated discs compared to unevaginated discs. These results are compared to the effects of 20-hydroxyecdysone (20-HOE) on metabolic labeling of membrane proteins fractionated from imaginal discs, and on cell surface labeling of a hormone-responsive Drosophila tissue culture line. A group of 35S-methionine labeled membrane fraction peptides whose metabolic labeling is 20-HOE dependent have isoelectric points and apparent molecular weights very similar to those of a group of proteins only labeled in iodinated evaginated discs, supporting the conclusion that these are hormone-dependent, cell surface proteins (Rickoll and Fristrom 1983). Based upon two-dimensional gel electrophoretic and immunological criteria three of the proteins showing increased labeling in evaginated discs are related to three proteins induced by 20-HOE in tissue culture cells. Two different subsets of radiolabeled peptides were observed in the imaginal discs based upon detergent solubility. Some of the proteins which are soluble in NP-40 plus urea but insoluble in NP-40 alone may be localized in the basal lamina of the imaginal discs, a structure which labels heavily with 125I and is lacking in tissue culture cells. In discs, the majority of hormone-dependent changes in radiolabeled peptides were seen in the fraction solubilized by NP-40 and urea with a sulfhydryl reducing agent, while in tissue culture cells, the majority of differences is seen in the fraction solubilized by NP-40 only. We speculate that these proteins may be involved in similar processes, e.g., cell rearrangement, that occur during both disc morphogenesis and 20-HOE induced aggregation in tissue culture cells.

    Notizen: Summary A comparison of pupation-temperature range was made in the laboratory on a temperature gradient (3–38°C) using 12 species ofDrosophila representing four species groups and four different ecological backgrounds (temperate-montane forest:virilis group; desert;repleta group; cosmopolitan:melanogaster group; tropical forest:willistoni group). Within groups, differences are found which usually reflect species' distributions. Comparisons of species' mating-, oviposition- and pupation-temperature ranges reveal that pupation most-often occurs at temperatures beyond those for mating and oviposition. Each species reflects a different combination of temperature effects. Individual species have different temperature-limits for mating, oviposition and pupation. Temperatures permissive for one response are not predictive of limits on other responses. Among species, temperature can affect a particular response differently. Within groups, species differences can be at high and/or low temperatures for any response, and temperature effects among closely related species can manifest themselves in one, or any combination of responses. One cannot predict which responses will be most and least limited, or at which end of the temperature scale a response will be most limited. Among groups,common, but notabsolute temperature ranges generally correspond to the geographic distributions and ecological backgrounds of the species triads. The evaluation of temperature effects on species, based on a single activity, may not be adequate for predicting adaptive strategies.

We have previously shown that reductions in gap junctions are correlated with the hyperplasia (Ryerse and Nagel 1984a). We report here that reductions in gap junction surface density, number and percent of the lateral plasma membrane area precede the onset of tissue hyperplasia as defined by the gross appearance of tissue overgrowth in the wing pouch and an increase in cell number. Gap junction reductions begin soon after temperature upshift and become significantly different from non-shifted controls by 16 h. Direct cell counts indicate that there is no difference in the total number of cells in experimental vs control discs until after 16 h when the 28° C discs begin to grow rapidly with a cell doubling time of about 6 h as compared with about 13 h for the 20°C controls. The finding that gap junction reductions precede the onset of tissue hyperplasia is consistent with the idea that gap junctions play a regulatory role in growth control and pattern formation and strengthens our hypothesis (Ryerse and Nagel 1984b) that a minimum number and a specific distribution of gap junctions are required for normal development.

    Notizen: Summary Genetically marked maroon-like (mal) clones were induced by mitotic recombination with X-rays at the blastoderm stage in mal/mal + heterozygotes and were analysed in differentiated Malpighian tubules (MT). Marked cells were not confined to single anterior (MA) or posterior (MP) tubules, but were distributed among the four tubules. About 70% of the clones with two or more cells were fragmented, i.e. mal cells were separated by wild-type cells. Since the clones contain, on average, 6 cells and the differentiated MT consist of 484 cells (2 × 136 MA cells, 2 × 106 MP cells), we estimate that there are about 80 cells in the blastoderm anlage which on average pass through two to three mitoses. With increasing radiation doses (254 R, 635 R, 1270 R) a linear increase in clone frequency is observed. The mean sizes and size distributions of clones, however, remain unchanged. Since the increasing radiation dose also results in fewer differentiated Malpighi cells, we assume that regeneration does not occur. Therefore, size distributions of marked clones presumably represent real mitotic patterns in normogenesis. We suggest that essentially three successive mitoses take place, with a decreasing fraction of cells showing mitotic activity. Only a small fraction of cells goes through a fourth or even a fifth mitosis. Marked non-Minute clones induced in Minute heterozygotes are more frequent, but are not larger than non-Minute clones in wild-type background. Therefore, compartment boundaries cannot be recognized by this method. However, frequencies of marked cells found simultaneously in MA and MP pairs or in several single tubules of the same individuals are significantly higher than frequencies of multiple recombination events predicted by the Poisson distribution. From this, we conclude that neither the MA pair nor the MP pair nor single tubules represent compartments of the MT anlage.

Cells from various larval anlagen of donor embryos labelled with a lineage tracer are individually transplanted from defined positions into similar, or different, positions in unlabelled hosts. The clones produced by these cells can be seen in whole mounts or in sections of fixed material, when using a histochemical marker (i.e. HRP), and/or in living embryos, when using fluorescent lineage tracers. The characteristics of the clones disclose lineage parameters, such as division patterns, morphogenetic movements and differentiation. The method is especially useful for testing the respective roles of positional information and cell lineage on the commitment of progenitor cells by transplanting these cells into heterotopic positions or into hosts of different genotypes.

    Notizen: Summary In this paper we describe a new allele of suppressor of forked, su(f) hd37, referred to as hd37, which was isolated in a hybrid dysgenesis mutation screen and is shown to be P induced by its high frequency of reversion in hybrid dysgenic crosses, and by in situ hybridization. hd37 suppresses forked and fails to complement the forked suppression of known su(f) alleles. However, it complements the recessive lethality of alleles in both of the su(f) lethal complementation groups. We also describe a new phenotypic effect of su(f) alleles, the enhancement of Minute(3)i 55. Recessive lethal alleles enhance the lethal effects of this Minute, but hd37 does not. The temperature sensitive period for forked bristle suppression by hd37 was found to be very narrow, consisting of a short interval (12–18 h) immediately before bristle formation. These results suggest that the several genetic functions associated with this locus may be genetically separable.

This growth continued over the first 8 days of adult life and in the young adults sperm were not produced until the testes increased approximately threefold in length to about 28 mm. The length of testes is correlated with genetic factors on the X and Y chromosomes. In males lacking a Y chromosome (X/O) or the short arm (YS) of the Y chromosome (X/YL) the testes were about half the length of testes of control males (X/Y) or double Y males (X/Y/Y). Males with deletions of the distal YL chromosome arm had testicular lengths equivalent to the controls. Males with short testes (X/O and X/YL) showed disruptions to spermatogenesis at meiosis and an absence of normal spermatid elongation. Reduction of active ribosomal RNA genes on the X chromosome in X/O caused an increased expression ofbobbed (bb) and a corresponding reduction in length of testes. Severelybobbed X/O males had very few cysts of spermatogonia and these cysts did not develop into primary spermatocytes.

    Notizen: Summary Transformations of tumorous-head Drosophila melanogaster were examined in order to investigate whether head structures were replaced by specific abdominal structures. Heads selected for the presence of genital structures were analyzed in detail. Female abnormalities included any combination of vaginal teeth, vulvar papillae, sensilla trichodea, abdominal tergites 6 (T6), 7 (T7), 8 (T8) and anal plate. Anal plate was observed in the prefrons and rostral membrane, while all other genital structures were intimately associated with modified shingle cuticle. Male abnormalities included transformation of antennal structures to penis, clasper teeth, lateral plate, anal plate and eye to T6. The distribution of each type of homeotic structure was confined to general regions of the eye-antenna, with no precise dividing lines between them. However, the spatial sequence of homeotic structures in the eye-antenna was generally the same as the sequence of the same structures in the posterior abdomen.

We show that the complete pattern results from the superimposition of two independent subpatterns, one of which depends on the achaete gene and the other on scute. The scute subpattern comprises several clusters of sensilla, most of which seem to require the presence of control regions located upstream of the transcribed region. This is in contrast with the pattern of scute-dependent bristles, most of which depends on control elements located downstream of the transcribed region.

    Notizen: Summary In Drosophila melanogaster the neuroblasts separate from epidermoblasts to give rise to the neural primordium. This process is under the control of several genes. The group of the so-called neurogenic genes is required for epidermal development; other genes, comprising those of the achaete-scute complex and daughterless, are required for neural development. We have studied the relationships between both groups of genes in two different ways. We have analyzed the phenotype of double-mutant embryos and our results show that the neural hyperplasia caused by neurogenic mutations can be partially prevented if a mutation in one of the other genes is present in the same genome. Only the neural cells that do not require the function of a particular gene of the achaete-scute complex in the wild-type seem to develop to a neural fate in the double mutant embryos. At least some of the genetic interactions affect the transcriptional level, as shown by in situ hybridization, since the territories of transcription of the achaetescute genes are expanded in neurogenic mutants. All cells of the neurogenic region of the double mutants apparently initiate neural development. However, during later development some of these cells switch their fate either to epidermogenesis or to cell death and this leads to the final phenotype of the double mutants. We discuss these results with respect to the events of early neurogenesis.

    Notizen: Summary The temperature-sensitive mutation shibire (shi) in Drosophila melanogaster is thought to disrupt membrane recycling processes, including endocytotic vesicle pinch-off. This mutation can perturb the development of nerves and muscles of the adult escape response. After exposure to a heat pulse (6 h at 30° C) at 20 h of pupal development, adults have abnormal flight muscles. Wing depressor muscles (DLM) are reduced in number from the normal six to one or two fibers, and are composed of enlarged fibers that appear to represent fiber fusion; large spaces devoid of muscle fibers suggested fiber deletion. The normal five motor axons are present in the peripheral nerve PDMN near the ganglion. However, while some motor axons pass dorsally to the extant fibers, other motor axons lacking end targets pass into an abnormal posterior branch and terminate in a neuroma, i.e., a tangle of axons and glia without muscle target tissue. Hemisynapses are common in axons of the proximal PDMN and within the neuroma, but they are rarely seen in control (no heat pulse) shi or wild-type flies. All surviving muscle fibers are innervated; no muscle tissue exists without innervation. Fibrillar fine structure and neuromuscular synapses appear normal. Fused fibers have dual innervation, suggesting correct and specific matching of target tissue and motor axons. Motor axons lacking target fibers do not innervate erroneous targets but instead terminate in the neuroma. These results suggest developmental constraints and rules, which may contribute to the orderly, stereotyped development in the normal flight system. The nature of the anomalies inducible in the flight motor system in shi flies implies that membrane recycling events at about 20 h of pupal development are critical to the formation of the normal adult nerve-muscle pattern for DLM flight muscles.

    Notizen: Summary Cell-cell interactions are involved in mediating developmental fate. An example is the decision of the neuroectodermal cells of Drosophila to develop as neural or epidermal progenitors, where cellular interactions participate in the process of acquisition of either cell fate. The results of heterochronic cell transplantations we describe here suggest that both neuroblasts and epidermoblasts are not irreversibly committed to a particular developmental fate. Rather, they retain the ability to interact with neighbouring cells and, under our experimental conditions, are capable of switching their fate during a relatively long period of time, i.e. until the end of embryonic stage 11.

This transformation suggests a reduced activity of the Sex combs reduced (Scr) gene on the basis of gene dosage studies, as well as enhanced expression of the phenotypes of the weak Scr − alleles in Rg-bx − larvae. Morphological observations of adult flies doubly heterozygous for Rg-bx and Scr mutations also suggest the enhancement of an aspect of Scr adult phenotypes. I conclude that the Rg-bx + gene function is required for the optimal expression of the Scr gene in larval and imaginai cells.

    Notizen: Summary Random insertions of a promotor fused to a reporter gene, such as Lac-Z, reveal regulatory sequences that confer temporal and spatial patterns of gene expression in eukaryotes. These patterns may reflect the activity of a neighbouring gene and thus lead to the isolation of new genes essential for normal development. Here, we demonstrate that this hypothesis is true for an insertion into the well characterized segmentation gene, hairy, in Drosophila. The insertion is homozygous lethal and fails to complement other hairy alleles, giving the phenotype described for hairy mutations. The insertion is located at 66D on the polytene chromosome map, is within 300–600 bp 5′ to the first hairy exon, and is orientated in the same sense (5′-3′) as the hairy transcription unit. Expression of β-galactosidase (β-gal), deriving from the insertion, follows closely the spatio-temporal patterns of expression of hairy gene product during embryogenesis. In addition, other sites of β-galactosidase expression are shown in the third larval instar stage and in the adult ovary. The results show that some insertions, giving restricted patterns of reporter gene expression, will reflect the temporo-spatial activity of a nearby gene.

    Notizen: Summary We have examined the metamorphosis of the wing imaginal disc of Drosophila during culture in vitro in the continuous presence of 20-hydroxy ecdysone (0.1 μg/ ml). We find that the sequence of cellular changes in the wing blade during culture closely match those occurring in situ, involving two periods at which the dorsal and ventral surfaces are joined only by cell processes containing trans-alar microtubule arrays. Good pupal and imaginal cuticle secretion is found in this system.

    Notizen: Summary We describe the results of cell transplantation experiments performed to investigate mesodermal lineages in Drosophila melanogaster, particularly the lineages of the somatic muscles, the visceral muscles and the fat body. Cells to be transplanted were labelled by injecting a mixture of horseradish peroxidase (HRP) and fluorescein-dextran (FITC) in wild-type embryos at the syncytial blastoderm stage. For transplantation cells were removed from the ventral furrow, 8–12 min after the start of gastrulation, and individually transplanted into homotopic or heterotopic locations of unlabelled wild-type hosts of the same age. HRP labelling in the resulting cell clones was demonstrated histochemically in the fully developed embryo; histotypes could be distinguished without ambiguity. Mesodermal cells were already found to be committed to mesodermal fates at the time of transplantation. They developed only into mesodermal derivatives and did not integrate in non-mesodermal organs upon heterotopical transplantation. No evidence was found for commitment to any particular mesodermal organ at the time of transplantation. The majority of somatic muscle clones contributed cells to only one segment. However, clones were not infrequently distributed through two or even three segments. Clones of fat body cells were generally restricted to a small region. However, cells of clones of visceral musculature were widely distributed. With respect to the proliferative abilities of transplanted cells the clones were difficult to interpret due to the syncytial character of the somatic musculature and the fact that the organization of the other organs is poorly understood. Evidence from histological observations of developing normal embryos indicates only three mitoses for mesodermal cells. Clones larger than seven cells were not found when embryos were fixed previous to germ-band shortening; larger clones were found in the fat body and visceral musculature after fixing the embryos at the end of organogenesis. Quantitative considerations suggest that a few mesodermal cells might perform more than three mitoses.

    Notizen: Summary Drosophila embryos, exposed to ether between 1 and 4 h after oviposition, develop defects ranging from the complete lack of segmentation to isolated gaps in single segments. Between these extremes are varying extents of incomplete and abnormal segmentation. On the basis of both their temporal and spatial characteristics, five major phenotype classes may be distinguished: headless — unsegmented or incompletely segmented anteriorly; gap — interruptions of segmentation not obviously periodic; alternating segment gaps — interruptions with double segment periodicities; fused segments; and short segments — truncations with single segment periodicities. Many defects resemble known mutant phenotypes. The disturbances in segmentation are predominantly global and frequently accompanied by alterations in segment specification, such that the segments obtained show no resemblance to the normal homologues. These features, together with the distinctive spatiotemporal characteristics of the defects, all point to segmentation as a dynamic process. The regular spacing of the segments and the fact that the entire range of defects is inducible by ether are further consistent with the hypothesis that at least part of the segmentation process may consist of physicochemical reactions coordinated over the whole body. The relationship between our data and data from genetic and other analyses are briefly discussed.

Anti-HRP antibody staining has shown that larval epidermal sensilla of homozygous mutant embryos occur in increased numbers, which depend on the allele considered. This increase is apparently due to the development into sensory organs of cells which in the wild-type would have developed as non-sensory epidermis. Thus, neurogenic genes act whenever developing cells have to decide between neurogenic and epidermogenic fates, both in central and peripheral nervous systems. Different regions of the ectodermal germ layer are distinguished with respect to their neurogenic abilities.

    Notizen: Summary Mutations in seven different maternal-effect loci on the second chromosome of Drosophila melanogaster all cause alterations in the anterior-posterior pattern of the embryo. Mutations in torso (tor) and trunk (trk) delete the anterior- and posterior-most structures of the embryo. At the same time they shift cellular fates which are normally found in the subterminal regions of the embryo towards the poles. Mutations in vasa (vas), valois (vls), staufen (stau) and tudor (tud) cause two embryonic defects. For one they result in absence of polar plasm, polar granules and pole cells in all eggs produced by mutant females. Secondly, embryos developing inside such eggs show deletions of abdominal segments. In addition, embryos derived from staufen mothers lack anterior head structures, embryos derived from valois mothers frequently fail to cellularize properly. Mutations in exuperantia (exu) cause deletions of anterior head structures, similar to torso, trunk and staufen. However in exu, these head structures are replaced by an inverted posterior end which comprises posterior midgut, proctodeal region, and often malpighian tubules. The effects of all mutations can be traced back to the beginning stages of gastrulation, indicating that the alterations in cellular fates have probably taken place by that time. Analysis of embryos derived from double mutant mothers suggests that these three phenotypic groups of mutants interfere with three different, independent pathways. All three pathways seem to act additively on the system which specifies anterior-posterior cellular fates within the egg.

    Notizen: Summary The fine structure of the indirect flight muscles was studied by electron microscopy in the following Notch locus mutants of Drosophila melanogaster reared at 18° C or 29° C for 6 days after eclosion: Ax 16172/Ax16172, Ax28/ Ax28, l(1)Nts1/l(1)Nts1,l(1)Nts1/Y and in wild-type controls. The flies were raised up to eclosion at 25° C or 18° C. It was observed that the l(1)Nts1 flies gradually became flightless within a few days if reared at 29° C as adults, and gross changes in the fine structure of the flight muscles were also observed in flies of this genotype. Peripheral myofilaments of myofibrils were disarranged and the mitochondria diminutive. At 18° C the flight muscles remained normal. In all of the Abruptex (Ax) combinations the flight muscles remained similar to the wild-type controls at both 18° C and 29° C, i.e. they were normal. The results suggest that the Notch gene is active in adult flies in addition to its activity during embryonic, larval and pupal stages, and is directly or indirectly involved in the adult development of the muscle tissue.

    Notizen: Summary The Y chromosome of Drosophila hydei carries information that is necessary for the development of the spermatozoa. In primary spermatocytes Y chromosomal genes become active: five of the male fertility factors form giant lampbrush loops. Our prior work indicated interactions between the Y chromosomal genes and autosomal loci. It is of interest to identify loci regulating the activity of the Y chromosomal genes. We, therefore, screened a total of about 14,000 chromosomes (X, 2, 3 and 4) for mutations that interfere with the expression of the lampbrush loops. Two mutations with substantial effects on the loop morphology were recovered. One of them, a recessive male sterile mutation (ms (3) 5) on chromosome 3, is described in this paper. Its homozygous state results in a complete absence of all Y chromosomal lampbrush loops at 26° C; at 18° C the loops are formed. Temperature shifts with homozygous males indicate that the function early during the spermatogonial stage is crucial for the development of lampbrush loops in the primary spermatocyte. Meiosis is entirely absent in the male, but normal in females. Females homozygous for ms (3) 5 display a maternal effect, which reduces the viability and fertility of homozygous daughters and produces sons with signs of intersexuality. Linkage studies indicated that the effect on the male germ line and the maternal effects cannot be separated and may hence be induced by a single gene.

The transcripts of this gene, thought to encode a basement membrane type IV collagen, are mainly accumulated during ecdysis in wandering haemocytes.     Notizen: Summary A gene Brista has been identified in chromosome 2R, in the region 60D11-E4, in which mutations cause homoeotic transformation of distal antennal structures to distal leg derivatives, and in which certain alleles also lead to upsets in the formation of distal elements of the legs. This gene is haploinsufficient for the homoeotic phenotype. Several putative null and two hypomorphic alleles have been recovered. The effects of exposure to the non-permissive temperature of a temperature-sensitive allele are cummulative and depend upon the length of the exposure during the period of antennal cell proliferation. It is suggested that this gene contributes to the stability of the state of determination in distal domain of the antennal and leg discs, and its relationship to other genes with similar mutant phenotype is discussed.

    Notizen: Summary The segmental organisation of the tail region in the embryo of Drosophila melanogaster, which is defined here as the epidermal region posterior to the boundary between abdominal segments A7 and A8, has been investigated by means of ultraviolet (UV) laser fate-mapping and phenotypic analysis of embryonic mutants that alter the segmental pattern of the larval cuticle. Wild-type embryos were irradiated in the presumptive tail region with a UV- laser microbeam of 20 μm diameter at the blastoderm stage. The ensuing defects were scored in the cuticle pattern of the tail region of the first-instar larva, which is described in detail in this paper. The spatial distribution of defect frequencies was used to construct a blastoderm fate-map of the cuticle structures of the larval tail region. The segmental origin of the larval tail structures was inferred from the phenotypic analysis of segmentation and homoeotic mutants, which revealed pattern repetition throughout the embryonic tail region corresponding to four segment anlagen, A8 to A11, and a non-segmental telson. These data enabled the transformation of the blastoderm fate-map of cuticle structures into a map of tail segment anlagen. The tail anlage occupies about 10% of the egg length (EL), bounded by segment A7 anteriorly at 20% EL and by the proctodaeum posteriorly at 10% EL, as measured from the posterior pole. The anlagen of segments A8 and A9 appear to be narrow dorso-ventral strips of blastoderm cells similar to the anlagen of the trunk segments, whereas the anlagen of A10 and A11 are smaller and produce fewer pattern elements. The telson is represented in the cuticle by the tuft which derives from a very dorsal posterior position. The antero-posterior axis of the entire tail anlage appears curved upward posteriorly. Differences in the mode of development between tail and trunk segments are discussed, as are similarities of larval and imaginal tail development in Drosophila. Comparison with tail development in other insects suggests that, during evolution, the transition from semi-long-germ to long-germ development modified the organisation of the tail region without affecting its primary subdivision into metameric units.

    Notizen: Summary The peripheral nervous system of embryos homozygous for prd, ftz, en and bxd was examined for defects and transformations in the segment-specific pattern of sensilla and peripheral nerves. This analysis permitted me to assign a distinct subset of sensilla to any of the three genetically and morphologically defined compartments s, a and p of each segment. In the wild-type embryonic segments, sensory axons deriving from sensilla of different compartments form a part of the common peripheral nerves. In the composite segments of prd and ftz mutant embryos, subsets of sensilla of two neighbouring segments are combined. Nevertheless, the axons of sensilla of different segmental identity are able to fasciculate and to form afferent nerves, which connect in an apparently normal fashion to the central nervous system. It is concluded that in the Drosophila embryo compartmental and segmental identity of sensory organs has no influence on the trajectories of sensory axons.

    Notizen: Summary Drosophila melanogaster embryos were injected before the blastoderm stage with conditioned media from several male Burkitt's lymphoma human cell lines and the Daudi cell line. Such injections do not have any effect on the male genital apparatus or on the female tract. The Daudi conditioned medium modifies the ovarian morphogenesis of the flies and the rudimentary ovaries obtained look like nymphal gonads. Moreover, they have a drastically reduced number of germ cells. The ovaries that looked functional contain numerous necrotic germ cells and the mean number of ovarioles per fly is significantly smaller than that of the controls. The abnormalities observed resemble the results of experimental and genetic lack of germ cells. They disappear at very high dilution (1×10−6).

    Notizen: Summary The legs of flies from 16 different mutant strains ofDrosophila melanogaster were examined for abnormal cuticular polarities and extra joints. The strains were chosen for study because they manifest abnormal cuticular polarities in some parts of the body (10 strains) or because they have missing or defective tarsal joints (6 strains). All but three of the stocks were found to exhibit misorientations of either the bristles, hairs, or “bract-socket vectors” on the legs. The latter term denotes an imaginary vector pointing from a hairlike structure called a “bract” to the bristle socket with which it is associated. On the legs of wild-type flies nearly all such vectors point distally, as do the bristles and hairs. In the mutant flies, the most common vector misorientation is a 180° reversal. When the bract-socket vectors of adjacent bristle sites in the same bristle row point toward one another, the distance between the sites is frequently abnormally large, whereas when the vectors point in opposite directions, the interval is frequently abnormally small. This correlation is interpreted to mean that bristle cells actively repel one another via cytoplasmic extensions that are longer in the direction of the bract-socket vector than in the opposite direction. Repulsive forces of this kind may be responsible for “fine-tuning” the regularity of bristle spacing in wild-type flies. Extra tarsal joints were found in eight of the 16 strains. A ninth strain completely lacking tarsal joints appears in some cases to have an extra tibia-basitarsus joint in its tibia. Whereas the tarsi of wild-type flies contain four joints, the tarsi ofspiny legs mutant flies contain as many as eight joints. In this extreme extra-joint phenotype, four of the joints correspond to the normal wild-type joints, and there is an extra joint in every tarsal segment except the distal-most (fifth) segment. Nearly all such ectopic extra joints have inverted polarity. In other strains the extra tarsal joints are located mainly at the wild-type joint sites, and joints of this sort have wild-type polarity. The alternation of normal and inverted (extra) joints inspiny legs resembles the alternation of normal and inverted (extra) body segment boundaries in the embryonic-lethal mutantpatch, suggesting that tarsal and body segmentation may share a common patterning mechanism.

Wild-type cytoplasm retains the rescuing activity longer than the poly(A)+ RNA fraction does, and cytoplasm is always more effective in provoking the rescue than poly(A)+ RNA. Mosaic females containing spz germ cells surrounded by spz + tissues were generated by pole cell transplantations; a mutant genotype in the germ cells is sufficient to produce all aspects of the spz mutant phenotype, suggesting that the maternal source of spz gene product is the germ line.

    Notizen: Summary Horseradish peroxidase (HRP) was used as an intracellular marker to follow the fate ofDrosophila embryonic cells. Injections were performed into 6- to 8-min-old gastrulae, when the cells have lost their connections with the yolk sack. Staining for HRP was made at the stage of germ band shortening, when most of the larval structures can be identified. By injecting at approximately 85 different sites we were able to locate the anlagen of the following structures: (a) epidermis: abdominal, thoracic and gnathal segments, dorsal ridge, procephalic lobe, clypeolabrum; (b) gut: pharynx, oesophagus, anterior and posterior midgut, proctodeum, Malpighian tubules and salivary glands; (c) central nervous system: abdominal, thoracic, sub-oesophageal and supra-oesophageal ganglia; (d) amnioserosa. The method proved to be useful in order to analyse morphogenetic movements. The anterior portion of the anterior midgut is shown to be formed from cells which invaginate with the stomodeum; the procephalic field is shown to rotate.

We study this phenomenon as a way to understand the normal mechanisms involved in the spatial regulation of homeotic gene expression. To this end we have investigated the distribution of Ultrabithorax (Ubx) proteins in imaginai discs mutant for hypomorphic and neomorphic alleles of Ubx and alleles of trans-acting genes. We find that the morphological discontinuities observed in the adult transformations are caused by corresponding new patterns of the Ubx proteins in the imaginai anlagen. These novel patterns of Ubx proteins are understood as a consequence of a process of reinforcement-extinction of Ubx expression. The evidence suggesting that this process results from a positive feed-back loop and cell-cell interactions is discussed.

    Notizen: Summary Fused is a segmentation gene belonging to the segment-polarity class. Mutations at thefused locus are known to display pleiotropic effects, causing zygotically determined anomalies of ovaries and of some adult cuticular structures, and maternally determined embryonic segmentation defects. In order to determine the amorphic phenotype offused and to study the genetical basis of its pleiotropy, newfused alleles (18 viable and 11 lethal) were isolated. The phenotype of these mutants and of others already known are described, taking into account zygotic and maternal effects. The main results provided by this analysis are as follows. Firstly, allfused alleles show the whole complex fused phenotype, and a good correlation is observed between the strength of the wing and segmentation defects, suggesting that a single function is involved in both processes. Secondly, all embryonic and larval lethals carry deficiencies which allow us to localizefused between the 17C4 and 17D2 bands of the X-chromosome. Thirdly, the 24 viable and 2 pupal lethals examined behave as point mutants, as shown cytologically or by Southern blot analysis. However, only one of them, the pupal lethalfu mH63 was proven to carry a nullfused allele, since it displays in germ-line clones a strong maternal phenotype and a very low zygotic rescue, similar to those of the small deficiencyDf(1)fu z4. The phenotype of the amorphic mutant indicates that zygotic ezpression offused is required for normal metamorphosis, while maternal expression is necessary for a normal segmentation pattern, since a complete loss offused expression during oogenesis cannot be compensated zygotically.

    Notizen: Summary Some aspects of neural and epidermal cell lineages during embryogenesis of Drosophila melanogaster were studied by transplanting horseradish-peroxidase-(HRP-) labelled ectodermal cells from young gastrula donors into host embryos of similar ages. Heterotopic transplantations permitted us to assess the degree of commitment already attained by the transplanted cells. The resulting cell clones showed normal characteristics of cytodifferentiation and cell number. The results indicate that epidermal progenitors perform a maximum of three mitoses during embryonic development, whereas neuroblasts may perform more than ten mitoses. Clone size distribution is in both cases scattered, suggesting either a rather irregular mitotic pattern or cell death. As indicated by heterotopic transplantations, the neurogenic ectoderm for the ventral nervous system exhibits different neurogenic abilities in its different regions, decreasing from medial to lateral; we discuss the hypothesis that some medially located cells of the young gastrulating embryo could be committed towards the neural fate before segregating from the ectoderm. On the other hand, the cells of the dorsal ectodermal regions at the same stage seem to be indifferent with respect to commitment, for they are able to give rise to central neural lineages following their transplantation in the neurogenic region.

    Notizen: Summary The developmental potential of the cells of the somatic follicular epithelium (follicle cells) was studied in mutants in which the differentiation of the germ-line cells is blocked at different stages of oogenesis. In two mutants, sn 36a and kelch, nurse cell regression does not occur, yet the follicle cells around the small oocyte continue their normal developmental program and produce an egg shell with micropylar cone and often deformed operculum and respiratory appendages. Neither the influx of nurse cell cytoplasm into the oocyte nor the few follicle cells covering the nurse cells are apparently required for the formation of the egg shell. In the tumor mutant benign gonial cell neoplasm (bgcn) the follicle cells can also differentiate to some extent although the germ-line cells remain morphologically undifferentiated. Vitelline membrane material was synthesized by the follicle cells in some bgcn chambers and in rare cases a columnar epithelium, which resembled morphologically that of wild-type stage-9 follicles, formed around the follicle's posterior end. The normal polarity of the follicular epithelium that is characteristic for mid-vitellogenic stages may, therefore, be established in the absence of morphologically differentiating germ-line cells. However, the tumorous germ-line cells do not constitute a homogeneous cell population since in about 30% of the analyzed follicles a cell cluster at or near the posterior pole can be identified by virtue of its high number of concanavalin A binding sites. This molecular marker reveals an anteroposterior polarity of the tumorous chambers. In follicles mutant for both bgcn and the polarity gene dicephalic the cluster of concanavalin A-stained germ-line cells shifts to more anterior positions in the follicle.

    Notizen: Summary The gastrulation defective (gd) locus is a maternally expressed gene in Drosophila required for normal differentiation of structures along the embryonic dorso-ventral axis. Cuticular defects of the offspring from females with different combinations of gd alleles comprised a phenotypic continuum. Complementation among several alleles produced normal offspring while progressively more severe mutations produced a graded loss of structures from ventral, and then lateral, blastoderm cells. The most severely affected embryos consisted entirely of structures derived from dorsal blastoderm cells. Histological examination of staged siblings from selected allelic combinations showed that internal tissues were similarly affected. The tissues observed in amorphic embryos support new, more dorsal, assignments of fate map positions for blastoderm precursors of the cephalopharyngeal apparatus, hindgut and ventral nerve cord. The loss of ventral and lateral structures did not occur through cell death and appeared to involve a change in blastoderm cell fate. A direct effect of the mutations on blastoderm cell determination, however, was insufficient to explain the development of the dorsalized embryos. Intermediate phenotypes suggested that cell interactions or movements associated with morphogenesis are required for the determination of some cell fates in the dorsoventral axis. Thus, the developmental fate of all blastoderm cells may not be fixed at the time of blastoderm formation.
