    Notes: Summary This investigation has dealt with a light and electron microscopic autoradiographic analysis of the median eminence of the rat following intraventricular infusion of 3H-dopamine. This study has demonstrated that 3H-dopamine, once infused into the mammalian cerebral ventricular system, is rapidly and selectively absorbed by tanycytes and transported to the contact zone within 5 minutes after ventricular infusion. Certain axon terminals in the ependymal, hypendymal, and palisade-contact zones selectively sequester 3H-dopamine as do subpopulations of arcuate neurons. This picture of intense labelling of neurons and axon terminals is discussed with respect to monoaminergic re-uptake mechanisms. Uptake and transport of 3H-DA by tanycytes is discussed.

    Notes: Summary (3H)-Thymidine autoradiography was used to study the DNA synthesis in the stroma and epithelium in the uterus proper and the uterine cervix of neonatal mice treated with oestradiol. It was found that in the epithelium of the uterus proper the DNA synthesis is stimulated between 6 and 12 h after injection of oestradiol and decreases again at 18 h. In the stroma of the uterus proper the DNA synthesis is increased 18 h after oestradiol injection. In the epithelium and stroma of the uterine cervix the DNA synthesis decreases from 5 h and is strongly depressed 18 h after oestradiol treatment.

    Notes: Summary Ornithine decarboxylase, a key enzyme in polyamine biosynthesis and cell growth, has been localized in mouse kidney by autoradiography after administration of radiolabeled α-difluoromethylornithine. This drug is an enzyme-activated irreversible inhibitor of ornithine decarboxylase and forms a covalent bond with the enzyme. It was found that ornithine decarboxylase is present in all cell types studied but that the highest content occurs in the proximal convoluted tubules followed by the distal convoluted tubules and the collecting tubules. The majority of the enzyme is located in the cytoplasm but about 10–15% is present in the nuclei (often associated with nucleolus-like components) of the cells of the proximal and distal convoluted tubules. The labeled ornithine decarboxylase was lost rapidly from both nucleus and cytoplasm of all the cell types examined, and labeling by radioactive α-difluoromethylornithine was greatly reduced if the mice were pretreated for 5 h with cycloheximide to block protein synthesis. These results indicate that ornithine decarboxylase turns over rapidly in all of the cells.

    Notes: Summary The distribution of estradiol-concentrating cells in the pituitary of the platyfish, Xiphophorus maculatus, is studied after the injection of 3H estradiol-17β by thaw-mount autoradiography. Autoradiograms prepared 2–8 h after the injection show nuclear concentration of radioactivity in certain cells of the proximal pars distalis, while no nuclear labeling is found in cells of the rostral pars distalis, pars intermedia and pars nervosa. Radioactively labeled cells are identified as gonadotropes by a combined technique of thaw-mount autoradiography and immunoperoxidase staining with antiserum to ovine LH. Approximately 80% of the immunoreactive LH cells show a concentration of radioactivity in their nuclei. These observations indicate that in teleosts, as in mammals, estradiol has a direct effect on pituitary gonadotropes.

    Notes: Summary Male Wistar rats were injected intravenously with 5-(3H)uridine-labeled lymphocytes isolated from lymph nodes of syngeneic donors and enriched in T cells. After short periods of time (3 to 120 min after injection), labeled lymphocytes were localized in spleen compartments using autoradiography to identify routes of lymphocyte movement from blood into splenic parenchyma and to follow migration pathways of recirculating lymphocytes within the periarterial lymphoid sheath (PALS). Topographical analysis of labeled lymphocytes was performed in specific planes of PALS characterized by the diameter of the arterial vessel and termed PALS large, PALS medium, and PALS small (PALS L, PALS M, PALS S, respectively). Attention was also paid to accumulations of labeled lymphocytes close to the arterial vessel wall. Initially, labeled lymphocytes were localized in PALS S and PALS M near the terminal branching of arterial vessels and in the marginal zone (MZ). We conclude that lymphocytes emigrate from blood into splenic parenchyma within two white pulp compartments: in MZ, and directly within PALS through the wall of capillary vessels. The sequential accumulation of labeled cells near arterial vessels of increasing diameter suggests that the recirculating pool of lymphocytes migrates into the central part of PALS L by two routes: from MZ, and along arterial vessels from PALS S and PALS M.

    Notes: Summary Affinity of lymphoid cells for the microenvironment of germinal centers (GC), as detectable in transfer experiments by rapid homing in spleen GC from the blood, is a capacity expressed by only a subset of lymphoid cells, in particular by those constituting a GC. However, when introduced into the blood stream, these cells do not home into GC of lymph nodes and gut-associated lymphoid tissues. To investigate further this homing inability for high endothelial venule (HEV)-containing lymphoid tissues, GC cells isolated from donor rabbit appendix were labeled in vitro with 3H-leucine and injected into an afferent lymph vessel of recipient popliteal lymph nodes. Draining lymph nodes were removed 15 min to 24 h after cell administration and prepared for radioautography. For reference, the migration of cells isolated from Peyer's patches and thoracic duct lymph was also studied. By use of appendix GC cells, large numbers of labeled cells were found to migrate into GCs of the outer cortex centripetally, i.e., from the subcapsular sinus through the lymphocyte corona into the GC proper. The same was observed for cells from Peyer's patches, although in smaller numbers. Thoracic duct lymphocytes were only localized in the lymphocyte corona and the deep cortex. Thus, appendix GC cells and a subpopulation of cells from Peyer's patches can reach lymph node GC, but only when administered intralymphatically. We conclude that cells expressing affinity for the GC microenvironment do so for both spleen and lymph node GC, but do not have the capacity to interact with the wall of HEV; its implication for the understanding of the dynamics of a GC reaction is discussed.

    Notes: Summary Tritium-labeled 1,25 (OH2) vitamin D3, when injected into vitamin D-deficient adult and pregnant rats is concentrated and retained strongest in nuclei of cells in the outer root sheath of the hair, followed by the stratum granulosum, spinosum, and basale of the epidermis. In the hair follicle, in addition to the most heavily labeled outer root sheath, nuclear labeling exists also in cells of the hair bulb and of the inner root sheath, as well as in basal cells of the sebaceous gland. In contrast, cells of the dermal papilla and the connective tissue of the dermis are generally unlabeled, except for labeled cells in the outer connective tissue sheath at the infundibulum of vibrissae of 20-day fetal rats and a few scattered labeled cells in the dermis, probably macrophages. In the developing hair, in 18- and 20-day fetal rats, a distinct topographic pattern of labeled cells can be seen, which is characteristic of the different stages of hair follicle development. In the hair germ, heavily labeled cells appear first in the stratum spinosum. In the hair peg, they remain in this position in its juxtaepidermal portion; however, when a dermal papilla develops, heavily labeled cells assume a marginal position. This suggests a sequential epidermal-epidermal and mesenchymal-epidermal receptor induction. Injection of tritium labeled 25 (OH) vitamin D3 did not show nuclear concentration in these tissues and excess unlabeled 25 (OH) vitamin D3 — unlike excess 1,25 (OH2) vitamin D3 — did not prevent nuclear uptake of tritium labeled 1,25 (OH2) vitamin D3. The results indicate differential effects of 1,25 (OH2) vitamin D3 on different structures in the epidermis and dermis.

A concomitant increase in nuclear volume and in the incorporation of 35S-cysteine indicates increased synthesis of neurosecretory material.     Notes: Summary Sympathectomy was carried out in rats by injections of guanethidine-sulfate from birth to 14 days of age. At 45 days of age, the activity of osteoblastic cells was monitored by 3H-proline autoradiography. Effectiveness of sympathectomy was verified by light-microscopic examination of superior cervical and celiac ganglia. Grain counts over periosteal osteoblasts of the femoral diaphysis and osteoblasts mesial to the first molar in the mandible demonstrated a significantly reduced uptake of 3H-proline in the sympathectomized rats. The data provide direct evidence of sympathetic influence on osteoblastic activity and suggest that sympathectomy may result in the loss of a trophic influence which is important in the regulation of osteogenesis.

    Notes: Summary The hypothalamus of male and female rats, given 0.3 μg/100 g body weight of 6.7-3H-oestradiol-17β and killed 1 hour after the injection, was examined by autoradiography in order to 1) localize the areas and the cells involved in the uptake of the hormone, and 2) study the intracellular localization of the labelled material. Only nerve cells contained radioactive material while glial and ependymal cells were not significantly labelled. In the anterior hypothalamus, labelled nerve cells were concentrated in areas corresponding to nucleus preopticus medialis and nucleus preopticus, pars suprachiasmatica. The nucleus supraopticus was unlabelled. In the medial basal hypothalamus, neurons corresponding to the nucleus arcuatus and the lateral part of the nucleus ventromedialis showed marked labelling. No significant labelling was observed in the nucleus paraventricularis, pars magnocellularis. Although the individual nerve cells varied in their extent of labelling, the major proportion of the silver grains were consistently concentrated over the nuclei. Castration was not found to influence the results. The findings were essentially the same in male and female rats and appear to suggest that oestradiol exerts a direct effect on nerve cells in certain hypothalamic areas.

    Notes: Summary The pituitaries of male and female rats given 0.3 μg of 6.7-3H-oestradiol-17β per 100 g body weight were examined by autoradiography in order to 1) identify the cells responsible for the uptake of the hormone, 2) determine the intracellular distribution of the hormone and quantify the proportions localized within the cytoplasm and nucleus by silver grain counting, and 3) see if sex differences existed in the cellular and intracellular distribution of the hormone. The animals were killed at intervals varying from 1 minute to 8 hours following intravenous or intramuscular injection. A large proportion of pituitary cells having the morphologic characteristics of acidophils, basophils and chromophobes contained radioactive material. Castration cells and acidophils of gonadectomized and lactating rats showed marked labelling. In male and female rats killed 10 minutes after intravenous injection, 84.4 and 83.6 per cent of the cells were labelled. One hour after intramuscular injection, 86.6 and 76.1 per cent of the cells were labelled in males and females, respectively. Thus, a small proportion of the cells remained unlabelled. Labelled cells showed silver grains both in the cytoplasm and over the cell nuclei, but the major proportion of the radioactive material was invariably associated with the cell nuclei in all cell types and at all time intervals. About 65 per cent of the radioactive material was associated with the cell nuclei in animals killed five minutes or one hour after intravenous or intramuscular injection of the hormone. The silver grains appeared to be randomly distributed in both the cytoplasm and over the cell nuclei. In the intermediate lobe and the neurohypophysis, only sparse labelling with random distribution was observed. At the border between the intermediate lobe and the neurohypophysis, labelling of single cells or clusters of cells similar to those in the adenohypophysis was found. The results, which were essentially the same in male and female rats, appear to indicate a direct effect of oestradiol at the pituitary level.

    Notes: Summary In order to localize pineal indoleamine synthesis at the ultrastructural level, an electron microscopic analysis was carried out on rabbit pineal tissue, cultured for several days in a medium containing the inhibitor p-chlorophenylalanine. Furthermore, electron microscopic autoradiography was applied to rabbit pineal tissue cultured in a medium containing the tritium labeled precursor 5-hydroxytryptophan. p-Chlorophenylalanine altered the ultrastructure of the mitochondria in the light pinealocytes only. Incubation with 3H-5-hydroxytryptophan caused a moderate and uniform labeling of the pinealocytes. These data suggest that the synthesis and storage of indoleamines in the rabbit pineal gland do not take place within the conventional membrane-limited cell organelles but more diffusely in the cytosol of the light pinealocytes. The spherical enlargement of the mitochondria in the light pinealocytes after culturing with p-chlorophenylalanine supports the biochemical findings of Hori et al. (1976) that the mitochondria are the carriers of the enzyme tryptophan 5-hydroxylase. The subsequent decarboxylation of 5-hydroxytryptophan to serotonin probably occurs free in the cytosol. The strong labeling of the noradrenergic nerve endings (in contrast to the cholinergic ones) after incubation with 3H-5-hydroxytryptophan demonstrates that the noradrenergic nerve endings in the rabbit pineal gland can, at least in vitro, selectively take up 5-hydroxytryptophan.

    Notes: Summary The localization of gamma-aminobutyric acid (GABA) high-affinity binding sites was investigated in the exocrine and endocrine pancreas of neonatal rats by means of 3H-GABA autoradiography. GABA-binding was identified on Schwann cells and on the cells of the intralobular excretory ducts. In the endocrine part of the pancreas, no labelling was observed except in peripheral islet cells which, on the basis of their scarcity and distribution, could be somatostatin cells. Furthermore, peri-insular innervation showed considerable labelling.

    Notes: Summary Autoradiography of sections from neurohypophyses treated with tritiated clonidine has shown specific binding to α2-adrenoceptor sites in the neurohypophysis but not in the intermediate lobe. Other studies have shown that α2 agonists such as clonidine can cause a fall in circulating antidiuretic hormone; it is therefore possible to speculate that this action could be a direct one on the neurohypophysis since the appropriate binding sites have been shown to exist.

Les résultats sont comparés avec ceux obtenus chez des mâles normaux ayant ou non développé un rein muqueux. Des cellules marquées sont présentes à tous les niveaux des tubules proximaux, mais leur nombre varie considérablement d'un segment à l'autre. Elles sont les plus nombreuses dans la seconde partie du tubule proximal, particulièrement dans sa région distale. Le taux de marquage est modifié dans toutes les régions du néphron, mais les variations sont les plus intenses dans la région distale de la seconde partie du tubule proximal, où l'on observe parfois la formation de nouveaux tubules par bourgeonnement. La comparaison des résultats obtenus dans les deux sexes fait apparaître une différence de réactivité au niveau de la zone proximale de la seconde partie du tubule proximal, où la méthyltestostérone agit fortement chez les femelles, alors qu'elle n'exerce pas d'effet notable chez les mâles. L'augmentation de volume du rein chez le mâle lors de la période de reproduction ne résulte donc pas uniquement d'un gonflement des cellules de la seconde partie du tubule proximal, comme on le pensait généralement. Des phénomènes mitotiques interviennent également dans ce processus, sous l'influence des hormones sexuelles mâles, qui conduisent à un allongement, voire à une multiplication des tubules proximaux.
    Notes: Summary The rate of 3H-thymidine incorporation into different parts of the renal proximal tubule of female sticklebacks treated with methyltestosterone was investigated using high-speed scintillation autoradiography. The results are compared with those from normal males before or after mucous transformation of the kidney. Labelled cells are observed in all parts of the proximal tubule, with marked variations from one segment to another. They are numerous in part 2 of the proximal tubule, particularly in the distal region. Male sex hormones affect the labelling rate in all parts of the nephron, especially in the distal region of part 2 of the proximal tubule. In that particular area, new tubule formation by budding is observed in some individuals, but this process does not appear to be a general one. Correlation between the frequency of these figures and the time of treatment could not be established. Comparing the action of sex hormones in females with that in males reveals a difference in reactivity in the proximal zone of part 2 of the proximal tubule, where methyltestosterone has a strong action in females; in contrast, in “mature” and “immature” males, only a few labelled cells are present in this region. It is concluded that kidney enlargement during the breeding season does not result only from a swelling of cells belonging to part 2 of the proximal tubule, as was generally believed, but also from a lengthening or even a proliferation of the proximal tubules, induced by an increase in mitotic activity controlled by male sex hormones.

    Notes: Summary In order to compare the functional state of the anterior pituitary of spontaneously hypertensive rats (SHR) with that of normotensive Wistar Kyoto rats (WKR), the anterior pituitary was examined by morphometry and autoradiography at the level of electron microscopy. The relative number and the relative volume of thyrotrophs in the anterior pituitary were significantly greater in SHR compared with age-matched WKR at 0, 7, 30–33 days, and 10 months of age, while the relative number of somatotrophs in SHR was significantly smaller at 1 and 10 months of age. Electron microscope autoradiographic analysis of uptake of 3H-lysine by thyrotrophs of both strains at the age of approximately one month showed that 3H-lysine was incorporated into protein and transported finally to secretory granules which migrated to near the cell membrane to be discharged. Silver grains were significantly more numerous over the thyrotrophs of SHR than over those of WKR at 30 min, 1 h, and 4h after the injection of 3H-lysine. The present study has ascertained morphologically that a congenital hypersynthesis of TSH by the anterior pituitary occurs in SHR.

In Xiphophorus helleri, NLB consist of fibrillar and granular material that gradually becomes more granular during meiotic prophase I, and is associated with mitochondrial arrangements during diplotene and dictyate of meiosis. Autoradiographic studies of uridine incorporation into the nucleolus and subsequently into NLB suggest that the latter represent a reserve of ribonucleoproteins that is later used in ribosomal maturation during vitellogenesis.

Approximately 31% of the satellite cells in uninjured muscles appeared labelled after three injections of tritiated thymidine; none of the myonuclei were labelled in the same muscles. After 2 hours, in the duodenum, silver grains are found in many undifferentiated cells, in a few young goblet cells, in some crystal-containing cells, and in some lymphocytes. In the colon after 2 hours silver grains are seen in some undifferentiated cells, and in many young goblet cells. Undifferentiated cells are characterized by a few short microvilli, poorly developed rough-surfaced endoplasmic reticulum, abundant free ribosomes, and a few apical moderately dense granules. In normal animals, absorptive cells seem to arise from undifferentiated cells, and goblet cells — from younger goblet cells. Undifferentiated cells could also become young goblet cells. Crystal-containing cells, which may not be of epithelial origin, proliferate in the epithelium in the adult animal.

    Notes: Summary Lipid metabolism in the cells of the renal proximal convoluted tubules (PCT) was investigated in healthy fowls and in fowls with the Fatty Liver and Kidney Syndrome (FLKS). The tissue was fixed at 10–25 min intervals after intravenous injection of 3H-oleic acid. The distribution of autoradiographic grains was analysed by the “circle method”. In normal cells most of the silver grains were associated with the cytoplasmic organelles. Lipid droplets and Golgi elements had the highest specific activity relative to the nuclear activity, which was little above background level. Lysosome-like bodies and mitochondria had lower values. In the cells of the FLKS-affected birds a large proportion of the grains was located over the lipid droplets, which are abundant in this condition. The specific activity of the cytoplasmic organelles was barely 2-fold higher than the nuclear activity. The results suggest that there is a diminished incorporation of esterified fatty acids by the organelles of these cells and that the excess is transferred to the lipid droplets. The identity of low electron density particles observed in the PCT cells of severely affected birds is discussed.

    Notes: Summary The pathway of lymphocyte migration through the white pulp of rat spleen and the relationship of migrating cells to the accessory cells (marginal zone macrophages and interdigitating cells, IDCs) of the white pulp compartments were analysed. Donor lymphocytes were obtained from lymph nodes, enriched for T lymphocytes and labeled in vitro with 5-(3H)uridine. They were injected intravenously into syngeneic recipients from which samples of spleen were taken at short intervals from 3 to 300 min after injection. Autoradiographs of semithin and ultrathin sections showed that, in the internal layer of the marginal zone (MZ), lymphocytes tended to accumulate within some regions in close proximity to marginal-zone macrophages before migrating into the periarteriolar lymphoid sheath (PALS). The lymphocytes enter PALS between protrusions of the accessory cells located in the peripheral area of the sheath. During migration towards the central area of PALS, a close contact between both cell types was noted. In the central area of PALS, preferential accumulation of lymphocytes around IDCs was observed. Labeled lymphocyte distribution within PALS and the rate of cell migration through the white pulp seem to depend on lymphocyte-IDC contact. A common feature of accessory cells which may affect the migration of lymphocytes in both MZ and PALS is the presence of Birbeck granules.

    Notes: Summary The sequence of the synthesis and transport of the organic matrix of spicules has been elucidated in the gorgonian Leptogorgia virgulata by use of 3H-aspartic acid as the tracer in electron-microscopic autoradiography. The entire process of matrix synthesis and transport takes approximately 2 h. It seems that the protein moiety of the organic matrix is synthesized in the RER prior to 5 min following the initial 10 min incubation in the tracer. At the 5 min chase the label is moving from the RER to the Golgi complexes where the carbohydrate moiety of the matrix is presumed to be synthesized. At the 5 to 15 min chases the label is transported out of the Golgi complexes via Golgi vesicles. This phase continues for 30 min. From 60 to 120 min the 3H-aspartic acid moves to the spicules. After 120 min the majority of the label has moved into the spicules. Silver grain counts over both multivesicular and electron-dense bodies remain at relatively low and constant levels over 4 h indicating that neither organelle is involved in the synthesis and transport of the organic matrix.

    Notes: Summary Pineal glands of rat, rabbit and hamster were incubated during day or night in Merlis' fluid containing [3H]-5-hydroxytryptamine (= [3H]-HT) by the use of a 20-min pulse with or without postincubation in “cold” medium for 15, 30, 45 or 60 min. (1) Selective autoradiographic labeling was observed in sympathetic nerve terminals; this reaction was missing after bilateral surgical removal of the superior cervical ganglia. In contrast, a scarce and diffuse labeling was found in pinealocytes (Pi) and interstitial cells (IC) of both untreated and ganglionectomized animals. (2) With the use of thin-layer chromatography, it could be shown in the rat that the well-known indoles of the pineal gland are formed from [3H]-HT. (3) During preparation for electron microscopy (EM), the total loss of indoles from pineal glands was studied by means of liquid-scintillation counting; approximately 57% of the radioactivity of the pineal glands was released into EM-processing solutions, mainly into the glutaraldehyde fixative. In summary, our results show that in this type of experiment with pineal glands of mammals, the routinely used EM-procedures are inadequate to visualize the uptake and metabolism of exogenous indoles in Pi and IC. Furthermore, the data differ considerably from those obtained with the pineal organs of several reptilian and avian species when a similar cytological procedure is used. It appears that protein(s) located in the densely packed vesicles of the pineal cells of sauropsids, homologous to mammalian pinealocytes, may play a crucial role in indole binding (specific indole-binding proteins); this may help to interpret the diverging results obtained in different amniotes.

    Notes: Summary Light-microscopic autoradiographs of the adrenal medulla at various intervals after the intravenous injection of [3H] 5-HTP, [3H] 5-HT, [3H] noradrenaline and [3H] adrenaline have been studied. The distribution of silver grains following [3H] 5-HTP uptake was found to be uniform over each of the two main cell populations, adrenaline-storing (A) cells and noradrenaline-storing (NA) cells in the adrenal medulla, but A cells were twice as active as NA cells in incorporating the isotope, a situation very similar to that found after [3H] dopa uptake. 5-HT administration resulted in a pattern resembling the distribution of [3H] noradrenaline uptake, with A cells being 4 or 5 times more active than NA cells and a gradient of activity from the periphery of the medulla inwards. However, the time-course for the loss of radioactivity was not the same for both amines: levels of 5-HT activity were not significantly reduced after one week whereas the degree of [3H] noradrenaline labelling after one week was less than 10% of that at one hour. Thus 5-HT may be bound to sites in the adrenal medulla normally occupied by noradrenaline but it would appear that the release mechanism is different. There was no evidence of 5-HT uptake by adrenal nerve endings.

    Notes: Summary The process of ribosome assembly in eukaryotes was studied by injecting tritium-labeled ribosomal proteins S6 and eL12 into oocytes of Xenopus laevis. The subcellular distribution of the two proteins was visualized by means of autoradiography in sections of oocytes. Protein S6 but not eL12 was found in the nucleus where it accumulated at the nucleoli. In the presence of actinomycin D the accumulation of S6 at the nucleoli was reduced. In-situ immunofluorescence studies indicated that S6 is located at the nucleoli and eL12 exclusively in the cytoplasm. It appears that S6 is involved in the early ribosomal assembly process at the nucleoli, whereas eL12 is restricted to the cytoplasm where it is incorporated into 60S ribosomal subunits in a late assembly step.

    Notes: Summary The proliferative activity of newborn and adult mouse adrenal medullary cells was determined with light and electron microscopic autoradiography. The H3 thymidine labelling index of 2 weeks old mice adrenal medullary cells was about 9.4 % and declined to less than 1 % in adult mice. In electron microscopic autoradiography labelled norepinephrine as well as epinephrine cells could be seen. Only in 1 and 2 weeks old mice some morphologically undifferentiated cells were visible. In formaldehyde induced fluorescence combined with light microscopic autoradiography the fluorescence intensities of labelled and unlabelled medullary cells were measured. On average the fluorescence intensity of labelled cells was lower than that of unlabelled cells. The differences could be explained by a higher number of autoradiographic silver grains laying on the cytoplasm of labelled cells. These results give evidence that fully differentiated adrenal medullary cells are capable of division.

    Description / Table of Contents: Résumé L'autoradiographie révèle, au niveau du pied, une incorporation massive et sélective de la 3H-Proline dans la ≪glande blanche≫ de Mytilus edulis. Cette étude a permis de suivre le processus qui mène de la synthèse de la sécrétion dans la partie basale des cellules jusqu'a son émission dans le sillon pédieux où elle participe à la formation du filament. La collagénase détruit la presque totalité du marquage, attestant ainsi la nature collagénique du produit sécrété. Les autres glandes pédieuses ainsi que la glande du byssus proprement dite, située à la base du pied, montrent une incorporation très faible, sans commune mesure avec celle de la ≪glande blanche≫. Ceci démontre de façon définitive que le collagène présent dans le filament prend naissance dans cette glande et justifie la dénomination de ≪glande du collagène≫. Des contrôles réalisés dans différentes régions (bords du manteau, manteau, branchies) montrent que l'injection du précurseur dans le bord palléal constitue une méthode satisfaisante pour marquer de façon relativement rapide et différentielle le collagène de la glande.
    Notes: Summary Autoradiographic studies reveal a strong specific incorporation of 3H-Proline in the “white gland” in the foot of Mytilus edulis. The author could trace the radioactive secretory product from its synthesis in the basal part of the cells down to its outflow into the pedial groove where it takes part in the formation of the filament. Purified collagenase takes out radioactivity from the sections. This observation confirms the collagenous nature of the secretion. The other foot-glands as well as the main “byssus gland” located at the base of the foot show but a very weak labelling not comparable with that of the “white gland”. This clearly evidences that the collagen occuring in the filament originates from the latter. The “white gland” may be properly called: “collagen gland”. Control sections through different parts of the body (mantle-edge, mantle, gills) confirm that our injection technique of the precursor into the palleal margin is a suitable method for a rather quick and specific labelling of the glandular collagen.

    Notes: Summary Hypothalamic tissue from 16 to 18-day fetal rats was transplanted onto the choroidal pia overlying the superior colliculus in adult female rats. After survival periods of 2 weeks to 19 months, brains containing transplants were processed for monoamine fluorescence histochemistry, immunohistochemistry for three neuropeptides (LHRH, somatostatin, neurophysin), or for autoradiography in ovariectomized hosts that received [3H] estradiol. Most of the transplants survived and retained or increased in size; 14 of 25 transplants examined by fluorescence histochemistry were found to contain median eminence-like structures. In almost all of the transplants that were stained for neuropeptides, beaded processes and occasional cell bodies were observed. Although immunoreactive fibers were found near blood vessels, no palisade arrangement typical of the normal median eminence was evident. Each of the hypothalamic transplants on which steroid autoradiography was performed contained clusters of estrophilic neurons, the intensity of labeling of which was comparable to that seen in the host hypothalamus. These results indicate that many characteristic morphological and chemical features of the hypothalamus, which are not evident in the 16 to 18-day fetus, are elaborated in transplants during the survival period in the host. Transplantation of fetal hypothalamus to adult choroidal pia thus appears to be a valuable approach for studying the factors, humoral or neural, that regulate the differentiation of this brain region.

    Notes: Summary Castrated adult male hamsters and castrated adult female rats were injected with either 0.2 μg (hamsters) or 0.5 μg (rats) of 3H-dihydrotestosterone (107 Ci/mmole)/100 grams body weight and killed 11/2 h later. The pituitary glands were removed and processed for both autoradiography and immunocytochemistry (hamster) or only autoradiography (rats). Localization of the androgen was found in 10–15% of the cells of the pars distalis in both species. Only cells that stained for luteinizing hormone (LH) in the hamster's pars distalis concentrated the androgen. Also cells in both the pars intermedia and pars nervosa (1–5%) concentrated the androgen in both species. Although the number of cells that concentrated the androgen in the pars intermedia and pars nervosa was small, this finding may be related to recent physiologic data that suggest that the gonadal steroids may play a role in regulating water retention and natriuresis.

    Notes: Summary The growth of tertiary follicles, i.e., the proliferation of cells in the stratum granulosum and in the capillary network of the theca interna, after injection of ovulation-inducing human chorionic gonadotropin (HCG), was investigated in the rabbit by means of autoradiographic and morphometric methods. Based on the frequency distribution of follicles with different sizes and on the labeling index (LI) of granulosa cells as a function of follicle size and of time prior to and after HCG stimulation, two groups of tertiary tollicles can be distinguished: growing (250–900 μm in diameter) and mature (〉900 μm in diameter) elements. The growth of both groups is influenced by the release of gonadotropins. After HCG stimulation, follicles belonging to the first group grow rapidly. During, and a short time after ovulation, almost all non-ruptured follicles larger than 600 μm in diameter become atretic. Within 35–50 h the ruptured and atretic mature follicles (〉900 μm in diameter) are replaced by follicles out of the group of growing follicles. From these results the following concept for regulation of follicle growth is derived: In principle, all growing follicles possess the potential to develop into mature follicles. When a sufficient number of mature follicles is generated, these mature follicles determine the number of succeeding growing follicles. Follicles that are not required for providing mature follicles become atretic as soon as they reach a diameter of 700 μm. When the majority of mature follicles is lost during ovulation (by rupture or atresia), this inhibition regulated by mature follicles is abolished, and all of the growing follicles again are capable to develop into mature follicles. The relative amount of capillaries in the theca interna of growing and mature follicles remains constant with increasing follicle size. This means that the capillary network grows parallel to the increasing size of follicles. No differences are found between intact and atretic follicles; advanced atretic follicles were excluded from this study. The labeling index (LI) of granulosa cells in the stratum granulosum and of endothelial cells in the theca interna, as a function of follicle size and of time after HCG stimulation, are closely correlated. A change in the LI of granulosa cells is usually followed with a certain delay by a similar alteration of the LI of endothelial cells in the theca interna. This suggests that granulosa cells have a certain regulatory function on capillary growth.

    Notes: Summary As a companion to an earlier study, the morphometric attributes of stimulated (blast-derived) lymphocytes in mouse axillary lymph nodes have been established using stereological and autoradiographic methods. Blast transformation was induced in vivo with dinitrochlorobenzene (DNCB) and stimulated cells were labelled with tritiated thymidine. Random samples of cells were taken for light and electron microscopic morphometry. In comparison to the unstimulated lymphocyte, the stimulated cell increased in size and possessed a greater plasma membrane surface area. Increase in cell size was the result of increases in the volumes of all measured subcellular compartments both in the nucleus and the cytoplasm. Heterogeneity of the stimulated cell population precludes firm conclusions regarding the significance of all these ultrastructural changes, though alterations in cell surface are discussed in the context of known biochemical differences accompanying blastogenesis.

    Notes: Summary The uptake of Ca45 into cells of the dinoflagellateGlenodinium foliaceum was investigated using insoluble compound light microscope autoradiography. The distribution of silver grains showed marked localisation to the dinocaryotic nucleus, with a random scatter of grains over the surrounding protoplasm (cytoplasm and supernumerary nucleus). Correction of grain counts for lateral sensitisation from the dinocaryotic nucleus indicated an isotope concentration 16–32 times greater in this organelle compared to the rest of the cell. Cells labelled for varying periods of time showed differences in the pattern of Ca45 uptake throughout the sample populations, but no increase in the mean level of uptake per cell. This would suggest a rapid incorporation of isotope within 1–2 hours, with little subsequent uptake. The presence of high levels of label after processing with both additive (glutaraldehyde, paraformaldehyde) and coagulative (acetic alcohol) fixatives indicated that the retention of Ca45 in these preparations was not simply a fixation artefact. Although the isotope did not appear to be suitable for (high resolution) electron microscope autoradiography, the intranuclear site of incorporation was demonstrated indirectly using a buffer extraction technique. Prolonged treatment with phosphate buffer resulted in a large scale loss of label from both cytoplasm and dinocaryotic nucleus. The latter appeared to show specific correlation with the loss of (protein) matrix from the chromosomes — as observed under both light and electron microscopy, with no apparent change in either nucleolus or nucleoplasm. This would suggest that incorporated Ca45 in the nucleus was largely confined to the condensed chromatin, where it was combined with the acidic proteins which make up the bulk of the chromatin matrix. The results obtained in this investigation are related to previous studies involving X-ray microanalysis and uptake of Ni63.

    Notes: Summary The incorporation of the chitin precursor N-acetyl-D-(1-3H) glucosamine byH. chlorinus has been studied by light and electron microscopic autoradiography. Light microscopic autoradiography showed that the incorporation occured preferentially at the hyphal apex. Autoradiograms from electron microscopy were quantitatively evaluated to determine the relative radioactivity incorporation between the cell wall and cytoplasm: this showed that (3H) incorporation took place mainly in the plasmalemma-wall complex. However a small amount of N-acetyl glucosamine can enter into the cytoplasmic space and is then transported by endomembranes (Golgi apparatus-vesicles) to the plasmalemma-cell wall interface before polymerization.

Incorporation of label during pollen maturation was periodic with peak RNA synthesis occurring in the uninucleate, nonvacuolate pollen grains and in the vegetative cell of the bicellular pollen grains. During the early stages of germination, isotope incorporation occurred predominantly in the nucleus of the vegetative cell with little or no incorporation in the generative cell. With the appearance of the pollen tube, incorporation of3H-uridine in the vegetative cell nucleus decreased and completely disappeared at later stages of germination. No incorporation of isotope was observed in the sperms formed in the pollen tube by the division of the generative cell. From a comparison of the results of this study with those of previous works on RNA synthesis during pollen embryogenesis in cultured anthers ofH. niger, it is concluded that in contrast to embryogenic development, there is no requirement for sustained RNA synthesis by the generative cell nucleus for normal gametophytic development.

    Notes: Summary Haustorial complexes were isolated from leaves ofPisum sativum infected withErysiphe pisi and exposed to14CO2 for 2 hours. The constituents of the isolated fraction were quantified and ultrastructurally described and the distribution of14C studied by electron microscope autoradiography and statistical treatment. Most (86%) of the isotope in the fraction was associated with haustorial complexes. Three classes of haustorial complexes were distinguished by degree of labelling and ultrastructure. Most of the haustorial complexes were termed healthy (i.e., they showed a normal ultrastructure) and were heavily labelled; necrotic complexes were unlabelled; and a class with intermediate labelling, modified extrahaustorial membrane and usually a normal haustorial cytoplasm was termed pre-necrotic. In healthy haustorial complexes the haustorial lobes and extrahaustorial membrane showed the highest grain densities and the body and extrahaustorial matrix were also significantly labelled. Comparison of the results suggest that ultrastructural modifications leading to necrosis were caused by dehydration which in turn determined reduction in photosynthate transfer. Other factors influencing transport into haustoria and the status of the extrahaustorial matrix are also discussed. U.V. fluorescence microscopy with acetamido-isothiocyanatostilbene salt was used to distinguish healthy and pre-necrotic haustorial complexes. It is recommended as a simple technique to monitor the functional quality of isolated haustorial complexes.

    Notes: Summary The uptake of63Ni into cells of the binucleate dinoflagellateGlenodinium foliaceum was investigated using insoluble compound light and electron microscope autoradiography. Cells labelled over a period of 2 hours showed active uptake throughout the whole population, with an increase in mean cell grain count when the labelling period was extended to 4 hours and 24 hours. The mean grain count did not vary with type of fixation (glutaraldehyde, paraformaldehyde or acetic alcohol) suggesting that retention of63 Ni is not a specific fixation-binding artefact. At light microscope level, silver grains were not localised to any major cell component, but with the greater resolution of electron microscope autoradiography, a high degree of localisation was demonstrated in the typical dinoflagellate (dinocaryotic) nucleus-which contained about 83% of the cell label (cytoplasm 16%, supernumerary nucleus 1%). Silver grain distribution within the dinocaryotic nucleus was consistent with some degree of localisation to the condensed chromatin. The autoradiographic results corroborate previous X-ray microanalytical data which demonstrated high levels of transition metals in dinoflagellate nuclei. The distinction between the two types of nucleus inGlenodinium is further emphasised, giving additional support to the concept of a separate phyllogenetic origin of the supernumerary nucleus.

Subsequent to radioactive labelling (32P,33P,35S) of the living part of the root system, the rooted soil is deep-frozen with liquid nitrogen and cut into thin slices. The location of the cut living roots in the soil cross sections can thereupon be determined by means of autoradiography. The method is suitable for various types of soils and can be employed in investigations with pot cultures and in the field. The technique has proved to be particularly effective for the determination of the average distances between roots in the context of investigating root competition for nutrients.

    Notes: Summary Seedlings of millet (Pennisetum americanum (L) K. Schum var. Tift 23 DA × Tift 18 DB) and guineacorn (Sorghum bicolor (L) Moench var. RS' 702) were employed in growth chamber studies to evaluate the effects of various phosphorus (P) levels on their growth and nutrient composition. The growth medium was Hoagland solution containing 0.0, 0.1, 1.0 and 2.5 mM P and 2.2 mM ferrous sulphate-tartate solution. At the end of twenty-four days, two plants from each treatment were supplied with 0.31 μci/ml of radioactive iron (55Fe) (with the same P levels used to grow them) and then allowed to absorb the55Fe for 3 hours after which they were harvested, separated into roots and shoots. The two components were wet-ashed separately, and assayed for55Fe. The rest of the plants in Hoagland solution were also harvested, separately into roots and shoots. The components parts were also wet-ashed separately and assayed for iron (Fe), manganese (Mn), zinc (Zn), copper (Cu) and phosphorous (P). High P concentration in the growth medium inhibited55Fe absorption and translocation in both millet and sorghum but millet was more tolerant of P-induced Fe deficiency than sorghum. Millet and sorghum grew best at 1.0 and 0.1 mM P, respectively. Fe concentrations in plant declined as P levels increased; while Mn increased in both millet and sorghum with increasing P levels. Millet, however, maintained higher concentrations of Fe and Mn. The distribution patter of55Fe, regardless of P levels could be summarized as follows: Roots〉Vascular tissue〉leaf-sheaths〉leaf blade. Less55Fe was absorbed and translocated to shoots at 2.5 mM P, in millet and sorghum.

    Notes: Abstract Histochemical studies on the rostellum ofHymenolepis diminuta revealed diastase-stable, protein/neutral carbohydrate-rich material localized in the rostellum tegument. The remainder of the rostellum, primarily composed of the glycogen-rich myocytons of the rostellum musculature, is protein-poor, but rich in diastase-labile, neutral and acidic carbohydrates. Ultrastructural cytochemical studies, using the periodic acid-thiocarbohydrazideosmium (PATCO) technique, indicated that the granules of the rostellar tegumental cytons and distal cytoplasm are carbohydrate-rich. Lipids are present in the rostellar myocytons but not in the tegumental cytons. Autoradiography using a pulse-labeling with [3H]leucine revealed an apical translocation of tegumental granules, but at a slower rate than had been reported for the strobilar tegument of the same organism. Neither [3H]galactose nor [3H]glucose were incorporated into the rostellar tegumental granules. The function of the secretory glycoprotein(s) produced in the rostellar tegument and its (their) possible role in the regulation of maturation and/or strobilization remain enigmatic.

Experiments were carried out on plexuses that had been freshly isolated from the gut wall or on isolated plexuses that had been maintained as expiant cultures for 7 to 14 days.     Notes: Summary The uptake and incorporation of L-3H-leucine and L-3H-serine in the oesophageal, gastric and pancreatic exocrine glands of Rana pipiens was examined using light microscopic autoradiographic techniques. At 1/2 h, 1 1/2 h and 3 h following injections of amino acids, silver grains are observed over the basal region of the cells comprising the glands. At 6 h after injection, silver grains occur over the apical region of the cells and by 24 h the grains can be observed over the cytoplasm of the cells adjacent to the lumen of the glands, collecting ducts, oesophagus and stomach. These observations indicate that the amino acids are being incorporated into proteins that are then discharged into the lumen of the pancreatic ducts, oesophagus and stomach. The secretory picture observed in the oesophageal glands closely resembles that of the gastric glands. This autoradiographic study supports the suggestion that the two glands are homologous and that they both secrete pepsinogen granules.

    Notes: Summary Previous work in the rabbit has shown that there is a significant flux of plasma hyaluronic acid (HA)1 which is taken up and degraded mainly in the liver but also concentrated in the spleen. Purified 14C-labelled HA of high average molecular wt prepared by biosynthesis from D-[U-14C] glucose was injected i.v. in mice and its tissue distribution was determined by whole-body autoradiography during the next 24 h. As blood levels declined, radioactivity was concentrated in the liver and spleen as found in the rabbit, and also in bone marrow and lymph nodes. Distribution was uniform in liver tissue, concentrated and relatively persistent in the periphery of lymph nodes, and distinctly nodular within the spleen. Analysis of an aqueous liver extract taken 4 h after injection identified 14C in HA, in a macromolecular fraction resistant to fungal hyaluronidase, and in metabolites of low molecular wt. These findings confirm and extend observations based on tissue extraction in rabbits. The pattern of distribution through the body and the restricted localization within spleen and lymph nodes further suggest that HA is absorbed from plasma and tissue fluids by elements of the reticuloendothelial system.

In addition to other auditory nuclei, which showed more or less uniform labeling with the present technique, the n. mesencephalicus lateralis dorsalis (MLD) of the midbrain, as well as field L and parts of the hyperstriatum ventrale in the telencephalon, showed a stripe-pattern of labeling after stimulation with a pure tone. The position and orientation of the tone-activated striped areas in field L, observed after stimulation with different tones, correspond to isofrequency contours obtained with microelectrode recordings. The labeling of the three congruent tonotopically organized layers of field L (L1, L2, and L3) was not uniform along the anterior-posterior axis of the field. Harmonic tones produced multiple reactive stripes each of which corresponded to the stripe characteristic of a particular harmonic presented as a pure tone. The species-specific Iambus-call labeled the tonotopic area of field L that corresponds to the frequency band with the highest energy of the call. The hyperstriatum ventrale generally showed a weaker pattern of labeling that, however, resembled the labeling in field L.

In male and female rats that had been castrated and adrenalectomized for one day all five cell types showed nuclear concentration of label one hour after injection of 3H-estradiol.     Notes: Summary The migration of radiolabeled intravenously injected B lymphocytes through thymus-dependent areas was studied in lymphoid organs of mice with experimentally defined T cell domains (B cell-deprived mice or “T” mice). In the spleen, B cells were found to enter the peri-arteriolar lymphoid sheath (PALS) by two routes: (i) via the marginal zone, and (ii) via reticulin sheaths surrounding terminal arterioles. B cells migrated through the peripheral and central PALS and initiated the formation of primary follicles in the peripheral PALS 6 h after injection. Distinct primary follicles were noted at 18 h after injection of the labeled B cells. After 24 h small numbers of labeled cells were also noted in the efferent lymphatic vessels of the spleen. The reconstitution of B cell compartments in the mesenteric lymph node was delayed compared to the spleen. B cells entered the nodal stroma across the wall of high endothelial venules in the paracortex and by 6 h were found scattered throughout the paracortex. Isolated clusters of a few labeled cells were noted in the outer cortex at 18 h after cell transfer. Defined primary nodules were seen only 24 h after reconstitution. A minority of labeled cells was found at 24 h in the cortico-medullary junctions and in medullary cords. The present study shows that B lymphocytes traverse T cell domains on their way to their own specific B cell compartments. The immunological significance of this particular migration route is discussed in view of data on the cellular cooperation of B cells, T cells and macrophages during the humoral immune response.

    Notes: Summary The localization and retention of radioactivity in the uteri of fetal and newborn guinea pigs was studied after subcutaneous injection of 3H-progesterone into intact and estradiol-primed animals. In the fetal uterus of the animals treated with estradiol, a significant increase in the accumulation of silver grains was observed, mainly localized in the stroma and myometrium, but very little is detected in the uterine gland. On the other hand, in the uteri of newborns, the effect of estradiol on radioactivity retention was significantly less intense and the radioactivity is mainly localized in the epithelium and the uterine gland. These data correlate well with the quantitative evaluation of the concentration of progesterone-specific-binding sites in the non-treated and in the estradiol-primed fetal and newborn guinea pigs.

The results showed a decrease in the radioactivity of the cell coat on the microvilli and an increase in the Golgi apparatus and in the apical vesicles and tubules. This divergence is attributed to a colchicine-induced impairment of the normal transport of cell-coat glycoproteins from the Golgi apparatus, via the apical vesicles and tubules, to the apex of the cell. The radioactivity of the lysosome-like bodies in the absorptive cells cultured with colchicine also increased. This finding supports a crinophagic function of these organelles in the degradation of excess cell-coat material. Male and female rats castrated for 14 days show nuclear concentration of label in all five cell types one h after injection of 3H-estradiol, whether progesterone treated or not. The order of labeling intensity is gonadotropes ≧ lactotropes = somatotropes 〉 thyrotropes = corticotropes. Progesterone treatment induces a dramatic sexual dimorphism in estrogen uptake; it significantly increases 3H-estrogen uptake in all female cell types. In males, progesterone decreases uptake in gonadotropes while not altering uptake in other cell types.

    Notes: Summary Autoradiographic experiments using 3H-thymidine were designed to analyse cell proliferation which occurs in skeletal muscle after denervation and after tenotomy. In mouse tibialis anterior and tongue muscles during the first 24 h after denervation or tenotomy labelling levels were low and did not differ significantly from sham operated control muscles. By 48 h after denervation and tenotomy of tibialis anterior muscles, increased levels of labelling occurred in both muscle and connective tissue nuclei. Daily pulse labelling for 7 days after denervation produced a labelling level which was 8 times that of sham operated controls, 25–30% of the total nuclear population being labelled. Denervated muscles had twice the level of labelling compared to tenotomised muscles. These results provide conclusive evidence that both denervation and tenotomy stimulate cell proliferation in skeletal muscle and it is suggested that the increased numbers of labelled muscle nuclei are likely to be the result of mitotic activity in muscle satellite cells.

This interference involves structural alterations of cellular membrane which do not include a reduced Con A-binding capacity. This conclusion is supported by the observation of deformities in both nuclear and cytoplasmic membranes in Con A-activated lymphocytes subsequently treated with CB. The high incidence of membrane blebs and pseudomyelin bodies in the cytoplasm points to a general effect of CB on the structural organization of membrane which may secondarily interfere with some specific event such as generation or transfer of signals for activation or cytokinesis.

A dorsomedial continuum of arcuate and periventricular neurons selectively sequester 3H L-dopa 20 min following its intraventricular infusion. By 40 and 60 min following the infusion labelling of neurons is minimal and supports the notion of rapid degradation. Other cell compartments such as tanycytes demonstrate uptake of 3H L-dopa. The ultrastructural localization and distribution of radiolabelled L-dopa (or its metabolites) in the rodent hypothalamus is discussed with respect to mechanisms and cell compartments involved in neuroendocrine regulatory processes.

    Notes: Summary In vitro-uptake and localization of 3H-5-hydroxytryptophan and 3H-dihydroxyphenylalanine have been investigated by means of light microscopic autoradiography in the central nervous system of the freshwater mussel, Anodonta cygnea L. Accumulation of the labelled monoamine precursors could be observed both over neuronal perikarya and neuropil in the ganglia. 3H-5-hydroxytryptophan and 3H-dihydroxyphenylalanine were taken up by neurons characterized by a small size (10–20 μm), a polygonal form and an intense staining with toluidine blue. Labelled axon branches situated either in the neuropil or between nerve cells in the cortex exhibited only a moderate activity, frequently of a diffuse character. After pretreatment for 24 h with colchicine, silver grains were also seen over the neuronal perikarya and some axon profiles. This shows that local uptake of the monoamine precursors and, presumably, the synthesis of the monoamines takes place both at perikaryal and axonal/terminal level. The nerve cells accumulating the monoamine precursors contain a nucleus with rich chromatin content and a deeply invaginated nuclear envelope. Further ultrastructural characteristics of these neurons are their well-developed rough endoplasmic reticulum system, a great number of mitochondria and glycogen granules, and the presence of dense-cored vesicles of different types. All these features are responsible for the electron-dense appearance of the cytoplasm.

    Notes: Summary This study provides information on the rates of DNA synthesis, sites of DNA synthesis, and DNA content of the avian salt gland during the osmoticstressing (plasma membrane synthesis) and destressing (plasma membrane turnover) cycle, in an effort to better understand the relationship of cell turnover to the initial events in plasma membrane amplification, differentiation, and turnover. The rate of DNA synthesis increases 12–24 h after the onset of osmotic stress, is maximal at about 24 h of osmotic stress, and decreases thereafter in fully stressed and destressed glands. The maximum DNA and protein content, and the maximum protein/DNA ratio are obtained after about 3 days of stress. Autoradiograms show that at 24 h of stress 70–80% of DNA synthesis occurs in connective tissue cells and 20–30% in parenchymal cells, but by 6 days of stress, synthesis occurs about equally in these cell groups. Because destressing is characterized by a large decrease in plasma membrane and in glandular protein, but by little DNA turnover or loss, the loss of plasma membrane is likely due to some type of cell dedifferentiation rather than cell turnover.

    Notes: Summary Immunocytochemical and autoradiographic localization of thyrotropin-releasing hormone (TRH)-and 3H-TRH-binding sites was studied in the arcuate nucleus-median eminence region of the rat. TRH-like immunoreactivity was found in dense granular vesicles (90–140 nm in diameter) in TRH-like immunoreactive nerve fibers and terminals. In the median eminence, the immunoreactive terminals were observed to be in direct contact with the perivascular basal lamina of the portal vessel and to form synaptoid contacts with tanycytes. In the arcuate nucleus, the immunoreactive terminals were often found to form axosomatic and axo-axonic, and/or axo-dendritic synapses. The uptake of tritiated TRH into the nerve fibers and terminals of the median eminence was also observed by autoradiography and the distribution and localization of silver grains in them were analyzed quantitatively by circle analysis. Thirty minutes after intraventricular infusion of 3H-TRH, radioactive labeling occurred in type-2 and 3-nerve fibers and terminals containing dense granular vesicles in the median eminence. It is therefore suggested that the neurons labeled after 3H-TRH infusion possess certain functions as physiological recognition sites or receptors for TRH.

    Notes: Summary The cellular and subcellular localization of radioactivity in the brain of immature female rats was determined by dry-mount autoradiography 2 h after iv injection of 1.0 μg of (monethyl-3H) diethylstilboestrol per 100 g body weight. A specific topographic pattern of nuclear concentration of the synthetic oestrogen was obtained similar to that for 3H-oestradiol-17β in specific neurons of the basal hypothalamus, preoptic region and amygdala. In competition experiments, the nuclear concentration of radioactivity in all areas studied was inhibited by unlabeled oestradiol, while unlabeled testosterone had no effect. These data suggest that although oestradiol can bind to androgen receptors, the oestrogen receptor itself can account for the localization seen after the injection of 3H-oestradiol.

We performed autoradiographic studies using 3H-estradiol-17β and 3H-dihydrotestosterone on male and female baboons. Discrete sites of localization of exposed photographic emulsion were observed over nuclei of cells of the islets of Langerhans of the pancreases of baboons injected with estrogen but not over those of baboons injected with androgen. These observations that islet cells contain specific receptors for estrogen when combined with the clinical observations, suggest that estrogens have a direct effect on the islet cells that may modulate the release of insulin.

A total of 12 baboons were divided into two groups, each containing three males and three females. Each animal in one group, both males and females, was injected intravenously with 1 μg/kg body weight of 3H-estradiol while those in the second group were each injected with 1 μg/kg body weight of 3H-dihydrotestosterone. As controls, one male and one female from each group also received a dose of 100 μg/kg body weight of the corresponding unlabeled steroid. One and a half hours after the injections, the animals were sacrificed and the spleen, thymus, and inguinal lymph nodes removed and processed for autoradiography. The localization of 3H-estradiol was similar in both males and females. In the thymus fibroblasts and epithelio-reticular cells, but not thymocytes, localized 3H-estradiol. In lymph node and spleen, nonlymphoid tissue concentrated the labeled estrogen. Additionally, in the paracortical region of the lymph node, an unknown cell type was labeled with estrogen. Only one male baboon demonstrated nuclear localization of 3H-dihydrotestosterone. This was observed in the reticular cells in the spleen and lymph nodes. The same cell type in the organs of the remaining animals was unlabeled.

    Notes: Summary Light microscopic autoradiography performed subsequent to intraocular injection of 3H-leucine revealed silver grains (SG) above axons of the optic tract which could be followed into the ventral and caudal portion of the suprachiasmatic nuclei (SCN) and above the contralateral anterior hypothalamic nucleus (AHN). By high resolution photometric measurement and computer processing the labelled areas were analysed, thus yielding statistical data of the relative grain distribution. The highest SG density was found in the ventrolateral part of both SCN (SCvl), confirming earlier reports concerning retinohypothalamic connections. That area exhibiting a cytoarchitecture different from the remaining nucleus was traversed, however, by numerous labelled axons. In the caudal part of both SCN a specific projection field of retinal fibres could be located. Here, almost no traversing fibres contribute to the rather circumscribed marked area. In the ventral part of the contralateral AHN, diffuse labelling well above background levels could be observed. Distinction between bypassing and terminating fibres within the SCvl could not be made using light microscopy. Analysis of SG distribution of the SCvl with electron microscopic autoradiography revealed a specific localization of SG within presynaptic terminals containing clear vesicles and pale mitochondria.

    Notes: Summary The LH(HCG) receptors in the ovaries of immature rats which were either untreated, or primed with PMSG and HCG, have been studied with a histochemical method which has proved to be as effective as when earlier used in the rat testis. This method, which consists of the topical application of 125I-HCG to picric acid-formaldehyde (PAF) fixed frozen sections followed by autoradiography, is also suitable for quantitative studies on the distribution of receptors. In the ovary of the immature 26 days old rat, the LH(HCG) receptors are localized exclusively in the interstitial and thecal tissues. After PMSG treatment many receptors appear in the granulosa of the large antral follicles. These receptors are most numerous in the outer layers of cells and least numerous in the inner. At the same time there are fewer receptors in the thecal and interstitial cells which have undergone the process of luteinization. After PMSG and HCG treatment the newly formed corpora lutea have few receptors, but these become progressively more numerous on subsequent days. It is suggested that, in the rat, the luteinization of the ovarian LH-target cells is associated with an initial decrease in the number of their LH(HCG) receptors.

    Notes: Summary Light- and electron-microscopic autoradiographic studies were carried out on Owenia to detect selectively catecholaminergic and indolaminergic neurons at two appropriate body levels according to the regenerative properties of this annelid, i.e. in the 1st and the 4th abdominal segments. Autoradiographically, intense 3H-5-hydroxytryptamine accumulation is seen in the 1st abdominal segment, and a less intense autoradiographic reaction in the 4th segment. Roughly, a similar difference in the distribution of the label is seen at both levels following 3H-noradrenaline administration. These observations, confirmed by a quantitative uptake analysis, suggest the presence of catecholaminergic and indolaminergic neurons in Owenia.

    Notes: Summary Neither homogenates nor frozen ovarian sections of rats aged 1, 5 and 7 days demonstrated specific binding of 125I-HCG. However, from day 10 to day 35, the homogenates could bind specifically, with an equilibrium association constant in the range of 0.9–2.7 × 1010 M−1. The binding capacity increased from 5.8 fmol/mg tissue at day 10 to 15.7 fmol/mg tissue at day 35. As revealed by autoradiography, the frozen ovarian sections from 10-day-old rats showed 125I-HCG binding localized over the interstitial and thecal tissues and, in the older age groups, also in the granulosa cells of follicles larger than 500 μm in diameter. These results indicate that LH/HCG receptors appear in the rat ovary at the beginning of the second postnatal week, and that the interstitial cells are the main site of action of LH before puberty.

    Notes: Summary The marrow cavity of the rat tibia was mechanically evacuated and autoimplanted to the subcutaneous tissue. The regenerative process which restored the integrity of marrow stroma and hemopoiesis, was morphometrically evaluated in whole mount of tibia. Following evacuation, the clot filled the cavity. The granulation tissue then appeared and expanded, penetrating and replacing the clot. The fibroblasts of the granulation tissue differentiated into osteoblasts forming osteoid bone. Within its interstices, the primordial marrow consisting of loose connective tissue and vascular sinuses appeared and hemopoiesis resumed. Expansion of hemopoiesis resulted in the resorption of bone and within three weeks the tibial cavity was restored to the pre-evacuation state. Autoradiography indicated that the labeling index was initially high in fibroblasts and osteoblasts but was subsequently reduced while it increased in osteocytes, cells of Haversian canals, stromal and hemopoietic cells of marrow. The finding is in disagreement with the view that the regenerative process originates from the Haversian canal. When the label was introduced on day 4 post-operatively, it subsequently appeared in osteocytes, cells of Haversian canal, stromal elements of the marrow, but not in the hemopoietic cells. This indicates complete dissociation of marrow stroma and hemopoietic stem cell.

    Notes: Summary The uptake and retention of radiolabelled dihydrotestosterone by the pituitary gland was examined in the rhesus monkey. Two animals were given an intravenous injection of 1.0μg/kg 3H-dihydrotestosterone (DHT) alone while one monkey received both the labelled androgen and 100μg/kg of unlabelled steroid. One and a half hours later, they were sacrificed. The pituitary glands were removed and processed for autoradiography and immunocytochemistry. Autoradiographic localization of DHT was discernible in the partes nervosa, intermedia and distalis, albeit the highest concentration of radiolabelled cells was noted in the pars distalis. Immunocytochemical staining with antibodies to rat PRL, human TSHβ and ovine LHβ revealed a population of steroid-concentrating cells that contained TSH and a second group that contained LH. None of the cells that reacted with the anti-PRL serum were radiolabelled.

    Notes: Summary Continuous treatment of developing eggs of the polychete Ophryotrocha labronica with α-methylornithine, which inhibits synthesis of putrescine, led to arrest of development at gastrulation. The present ultrastructural analysis suggests that the arrest of development is due to failure to form nuclei, and thus reveals a possible role for putrescine in nucleolar formation. Further support for this contention was provided by means of electron-microscopical autoradiography. It was found that newly synthesized putrescine, derived from administered 3H-ornithine, labeled the nucleoli intensely at the time of their normal appearance during gastrulation, the time at which the rate of endogenous putrescine synthesis is maximal. These observations have led to the conclusion that putrescine synthesis may be directly involved in formation of nucleoli.

At 15, 30, 60, 90, or 150 days after surgery, the mice were injected with 2μCi of 3H-proline (sp. act. 1.0 Ci/mM) per g of body weight and killed 15, 30, or 60 min later. Autoradiographs were prepared from 5μm decalcified sagittal sections of mandibles and grain counts made over periosteal osteoblasts mesial to the first molar. In denervated mandibles, osteoblasts incorporated less isotope compared to controls with differences being maximal at the early intervals. These differences became attenuated with time, possibly due to an intrinsic compensatory mechanism, secondary to neurotrophic regulation.

    Notes: Summary Proteins filtered in the renal glomeruli are reabsorbed by the proximal tubule and catabolized in the lysosomes. On the basis of studies on isolated flounder tubules it has been suggested that, in addition to this catabolism, a transtubular transport of intact protein (lysozyme) also occurs. The present study demonstrates that significant amounts of lysozyme are reversibly bound to the peritubular side of isolated tubules. Electron microscopic autoradiography demonstrates that the protein is located in the basement membrane and intercellular spaces. After in vivo injection, 125-Ilysozyme is mainly located in endocytic vacuoles of the first proximal segment, but also over the basal part of the cells. Since a significant peritubular binding of lysozyme is demonstrated in vitro, it is suggested that a similar binding of tracer protein originating from the peritubular capillaries might occur in vivo and that subsequent release of this protein in vitro might simulate transtubular transport. It is therefore concluded that release of tracer protein from isolated kidney tubules does not conclusively demonstrate transtubular transport of intact protein in experimental systems in which peritubular binding of protein can be demonstrated.

    Notes: Summary The migration pattern of germinal center cells of the rabbit appendix was studied and compared with that of appendix dome cells, spleen cells, thymus cells and thoracic duct lymphocytes. To discriminate T-and B-cell migration pathways, normal or T-cell-depleted rabbits were used as donors. Cell suspensions were labeled in vitro with 3H-leucine followed by intravenous transfer. The migration of labeled cells in lymphoid organs was studied using autoradiography, particular attention being paid to the spleen of the recipient. B-cells from the appendix dome, spleen and thoracic-duct lymph migrate to primary follicles or the corona of secondary follicles via thymus-dependent areas of peripheral lymphoid organs. In contrast, a B-cell subpopulation from the germinal centers of the appendix migrates to the center of splenic primary follicles and into germinal centers. The migration of germinal center cells to splenic follicle centers is not enhanced by specific antigens. The migration properties of B-cells, possibly changing during differentiation, may be instrumental in the two types of immune reactions, i.e., plasma-cell reaction and germinal-center reaction.

    Notes: Summary The uptake and retention of radiolabeled estradiol by the uterus was examined in the armadillo. One pregnant and two non-pregnant armadillos were treated with 1.4 μg/kg body weight of 3H-estradiol (E2) by injection into the left ventricle, and one non-pregnant animal was injected with both the labeled hormone and 140 μg/kg body weight of unlabeled E2. One and a half hour after injection, the animals were sacrificed and the uteri were removed and processed for autoradiography. In the non-pregnant animals, nuclear localization was observed in the interstitial cells and glandular epithelium of the endometrium and the connective tissue cells and smooth muscle of the myometrium. Additionally, there was a gradation of uptake in the epithelial cells of the endometrium in that the glandular cells of the basal region were heavily labeled, while those cells in the sinusoidal, and luminal regions contained successively less label. The luminal cells were poorly labeled. In the pregnant female, the smooth muscle and glandular cells hypertrophied and their nuclei contained less label than was observed in the non-pregnant animals. The arteries of the myometrium were more easily distinguished in the pregnant animals and the nuclei of the endothelial cells and smooth muscle were more consistently labeled than those of the non-pregnant armadillos.

    Notes: Summary The uptake of γ-aminobutyric acid (GABA) in the thyroid gland of the rat was studied autoradiographically following in vitro incubation. High-affinity GABA uptake was localized in follicle cells, whereas C cells (parafollicular cells) in general did not accumulate GABA by high-affinity transport. The follicle cells were also the main sites of low-affinity GABA uptake. Additionally, some nerve fibres were found to accumulate GABA. The predominant localization of GABA uptake in follicle cells is discussed in view of a presumed role of GABA in thyroid function.

    Notes: Summary After injection of radiolabeled 1,25 (OH)2 vitamin D3, nuclear concentration of radioactivity is observed in parenchymal cells of the parathyroid gland in pregnant, adult male, and 10-day male neonatal rats. In competition studies with unlabeled 1,25 (OH)2 vitamin D3, but not with 25 (OH) vitamin D3, nuclear uptake is prevented. Experiments with 3H 25 (OH) vitamin D3, in contrast to 3H 1,25 (OH)2 vitamin D3, do not show nuclear concentration in cells of the parathyroid. The results of the autoradiographic studies suggest the presence of receptors for a direct effect of 1,25 (OH)2 vitamin D3 on the parathyroid gland for modulation of parathyroid hormone secretion.

    Notes: Summary Autoradiographic studies showed that in the rat hippocampus “microglia-like reactive cells” (MRC) and astrocytes are capable of proliferation in response to kainic acid (KA)-induced lesions. A marked increase in the number of labeled MRC was observed during the first four days after the induction of the KA-lesion. A proliferative response of astrocytes occurred at two days after the KA-lesion. After the induction of a KA-lesion brain macrophages and oligodendrocytes were only slightly labeled with 3H-thymidine. It appears likely that MRC is the main cellular element responding to this type of lesion.

    Notes: Summary The movement of labelled glucose along the digestive tract of two ascidian species, Sidnyum argus and Dendrodoa grossularia is compared. In Dendrodoa grossularia, glucose absorption takes place in all parts of the digestive tract except the oesophagus. Labelled glucose passes through the digestive tract in less than 12 h, and it accumulates in the pyloric ampullae in less than 3 days. In this species, the pyloric gland has more capacity to stock glucidic compounds than other tissues. Under normal feeding conditions, after five days, there is no transfer of labelled glucose to other tissues. Whatever the substances given to Sidnyum argus, its pyloric gland never accumulates labelled products.

    Notes: Summary Autoradiography after pulse labelling with [3H] thymidine was applied to investigate the proliferation processes in the granulosa and theca related to follicular atresia of the dog ovary during metestrus. The number of proliferating cells depends on the follicle type and its atretic stage. There is less proliferation in smaller secondary follicles than either in larger ones or tertiary follicles. While in early atresia tertiary follicles show the highest labelling indices, in advanced atresia the larger secondary follicles are those with the highest values. For each follicle type a decline in the labelling indices can be observed from early to terminal atresia. Tertiary follicles show a precipitous decrease in the labelling index between early and advanced atresia. There is a continuous gradient of proliferation from the center of the follicle over the peripheral granulosa to the theca. In tertiary follicles, an inverse correlation between labelling and necrosis of granulosa cells can be observed.

    Notes: Summary Intraperitoneal injection of p-chlorophenylalanine (pCPA) methylester (100 mg/kg body weight) results in an activation of the lysosomal system of the secretory cells in the rat seminal vesicle and an elevation of the activities of lysosomal enzymes within 15 min following the injection. Large autophagic vacuoles are formed, sequestering rough endoplasmic reticulum and part of the Golgi apparatus within 2 h. Shortly after the activation of the lysosomal system an elevation of both DNA and protein synthesis is measured biochemically. 6 h subsequent to the injection a wave of mitoses of the secretory cells begins, reaching a maximum 6 h later and then declining within 3 h. About 12 h following the injection a second rise in lysosomal activity begins, declining within 24 h. The entire sequence of lysosomal and proliferative activities is inhibited in antiandrogen-pretreated rats. Deduced from these findings the following hypothesis of growth regulation of the accessory sex glands is advanced: enhanced loss of intracellular material during autophagocytosis diminishes the intracellular concentration of a substance curtailing cell division below its effective threshold resulting in division of the secretory cells. The prerequisites of this mechanism are (i) a sufficient distributive capacity of the stroma for hormones (androgens) and metabolic precursors, and (ii) sufficient capacity of the basal cells for transporting the precursors to the secretory cells. Sloughing of the secretory cells separates them from these auxiliary structures (stroma and basal cells) and enables the basal cells to divide.

    Description / Table of Contents: Zusammenfassung An zwei Kollektiven von insgesamt 120 weiblichen Ratten wurde der Einfluß des ovariellen Zyklus auf die Proliferationsaktivität der Nebennierenrinde untersucht. Es zeigte sich, daß in der Zona glomerulosa und Zona fasiculata rhythmische Schwankungen der Zahl DNA-synthetisierender Zellen auftreten. Das Maximum 3H-Thymidin-markierter Kerne der Glomerulosa liegt im Proöstrus; die Fasziculata weist ein Minimum im Östrus auf. Dem Maximum markierter Kerne in der Glomerulosa folgt eine Mitosewelle im Östrus. In der Reticularis konnten keine rhythmischen Schwankungen der Zahl markierter Zellkerne nachgewiesen werden. Die möglichen endokrinen Regulationsmechanismen werden diskutiert.
    Notes: Summary In a total of 120 rats the number of 3H-Tdr-labelled nuclei of the adrenocortex was counted during various phases of the estrous cycle. In two separate experiments cyclic variations in the number of DNA synthesizing cells were found. The labelling index of the zona glomerulosa showed a significant maximum in proestrus whereas in zona fasciculata a minimum was revealed in estrus. In the glomerulosa the maximum of labelled cells was followed by a mitotic peak in estrus. Due to very low numbers of labelled cells no rhythm was found in the reticularis. The possible endocrine regulations of this endogenous rhythm of adrenocortical cell replication are discussed.

    Description / Table of Contents: Résumé La localisation au niveau du cerveau de souris de la corticostérone, qui est le glucocorticoide naturel chez la souris, et de la cortexolone, démontrée comme étant un antagoniste des glucocorticoides, est étudiée par autoradiographie 30 min après injection in vivo des composés tritiés. Après injection de 3H-corticosterone (3HB), la radioactivité se concentre préférentiellement dans des noyaux cellulaires de plusieurs structures du système limbique et dans les noyaux de certains neurones du cortex cérébral et du bulbe rachidien. Cette concentration nucléaire est abolie après injection de 3H-corticostérone en présence d'un excès de corticostérone non radioactive. Après injection de 3H-cortexolone (3HS), une distribution diffuse de la radioactivité est observée dans tout le cerveau, cependant, une concentration plus élevée de grains d'argent est présente dans la partie ventrale du nucleus arcuatus et dans l'infundibulum. Après injection de 3H-cortexolone en présence d'un excès de cortexolone non radioactive, cette accumulation préférentielle des grains est abolie. L'accumulation de la 3H-cortexolone dans la région hypothalamique suggère que la cortexolone se concentre préférentiellement dans la région cérébrale qui contient les sites de liaison de la dexaméthasone et de plus, les résultats autoradiographiques montrent que le complexe cortexolone-récepteur ne s'accumule pas dans le noyau cellulaire.
    Notes: Summary The localization in the mouse brain of corticosterone, the natural glucocorticoid in the mouse, and cortexolone, reported to be a glucocorticoid antagonist, was studied by autoradiography 30 min after in vivo administration of the tritiated compounds. After 3H-corticosterone (3HB) injection, radioactivity was preferentially concentrated in cell nuclei of several structures within the limbic system, and in nuclei of certain neurones of the cerebral cortex and medulla oblongata. This nuclear concentration was abolished after injection of 3H-corticosterone with an excess of unlabelled corticosterone. After 3H-cortexolone (3HS) injection, a diffuse radioactivity was observed throughout the brain. However, a higher concentration of grains was present in the ventral nucleus arcuatus and in the infundibulum. When excess unlabelled cortexolone was administered with 3H-cortexolone this preferential accumulation of grains was abolished. The accumulation of 3H-cortexolone in the medial basal hypothalamic region suggests that cortexolone concentrates preferentially in dexamethasone (DM) target regions, and in addition the autoradiographic results show that the cortexolone-receptor complex does not accumulate in the cell nucleus.
