    Beschreibung: In this paper, the advantages achievable from the use of two prototype systems that are being developed to increase safety and security in ports are shown. Both systems start by monitoring environmental parameters in harbors, and then process data acquired. The first system has been conceived to be helpful to port communities (port authorities, pilots) to optimize harbor waterside management (ship’s navigation and cargo, dock performances, boat moorings, refloating of stranded ships, water quality control). By monitoring and processing sea level and atmospheric pressure in port areas, it can help port communities, e.g., to choose the best time when a ship with a certain draft can enter or leave a harbor, or to plan the best route inside the basin for that vessel (port safety). The second system, instead, has been designed for port protection purposes: by monitoring and processing the Earth’s magnetic field below the sea surface in harbors (where the natural field is disturbed by a high artificial component), it is able to detect the possible presence of intruders (e.g., divers) swimming underwater in prohibited areas (port security). Here, the results of monitoring and processing activities of the two systems performed in Livorno and La Spezia harbors are shown (Italy). The processing procedures and the graphical interfaces of the systems are based on applications under development by the research team the author belongs to, by using C# and C++ languages; Matlab environment has been employed for simulations.

    Beschreibung: Krill and salps are important for carbon flux in the Southern Ocean, but the extent of their contribution and the consequences of shifts in dominance from krill to salps remain unclear. We present a direct comparison of the contribution of krill and salp faecal pellets (FP) to vertical carbon flux at the Antarctic Peninsula using a combination of sediment traps, FP production, carbon content, microbial degradation, and krill and salp abundances. Salps produce 4-fold more FP carbon than krill, but the FP from both species contribute equally to the carbon flux at 300 m, accounting for 75% of total carbon. Krill FP are exported to 72% to 300 m, while 80% of salp FP are retained in the mixed layer due to fragmentation. Thus, declining krill abundances could lead to decreased carbon flux, indicating that the Antarctic Peninsula could become a less efficient carbon sink for anthropogenic CO2 in future.

    Beschreibung: Coastal water quality in urban cities is increasingly impacted by human activities such as agricultural runoff, sewage discharges, and poor sanitation. However, environmental factors controlling bacteria abundance remain poorly understood. The study employed multiple indicators to assess ten beach water qualities in Ghana during minor wet seasons. Environmental parameters (e.g. temperature, electrical conductivity, total dissolved solids) were measured in situ using the Horiba multiple parameter probe. Surface water samples were collected to measure total suspended solids, nutrients, and chlorophyll-a via standard methods and bacteria determination through membrane filtration. Environmental parameters measured showed no significant variation for the sample period. However, bacteria loads differ significantly (p = 0.024) among the beaches and influenced significantly by nitrate (55.3%, p = 0.02) and total dissolved solids (17.1%, p = 0.017). The baseline study detected an increased amount of total coliforms and faecal indicator bacteria (Escherichia coli and Enterococcus spp.) in beach waters along the coast of Ghana, suggesting faecal contamination, which can pose health risks. The mean ± standard deviations of bacteria loads in beach water are total coliforms (4.06 × 103 ± 4.16 × 103 CFU/100 mL), E. coli (7.06 × 102 ± 1.72 × 103 CFU/100 mL), and Enterococcus spp. (6.15 × 102 ± 1.75 × 103 CFU/100 mL). Evidence of pollution calls for public awareness to prevent ecological and health-related risks and policy reforms to control coastal water pollution. Future research should focus on identifying the sources of contamination in the tropical Atlantic region.

    Beschreibung: Background: Extreme terrestrial, analogue environments are widely used models to study the limits of life and to infer habitability of extraterrestrial settings. In contrast to Earth’s ecosystems, potential extraterrestrial biotopes are usually characterized by a lack of oxygen. Methods: In the MASE project (Mars Analogues for Space Exploration), we selected representative anoxic analogue environments (permafrost, salt-mine, acidic lake and river, sulfur springs) for the comprehensive analysis of their microbial communities. We assessed the microbiome profile of intact cells by propidium monoazide-based amplicon and shotgun metagenome sequencing, supplemented with an extensive cultivation effort. Results: The information retrieved from microbiome analyses on the intact microbial community thriving in the MASE sites, together with the isolation of 31 model microorganisms and successful binning of 15 high-quality genomes allowed us to observe principle pathways, which pinpoint specific microbial functions in the MASE sites compared to moderate environments. The microorganisms were characterized by an impressive machinery to withstand physical and chemical pressures. All levels of our analyses revealed the strong and omnipresent dependency of the microbial communities on complex organic matter. Moreover, we identified an extremotolerant cosmopolitan group of 34 poly-extremophiles thriving in all sites. Conclusions: Our results reveal the presence of a core microbiome and microbial taxonomic similarities between saline and acidic anoxic environments. Our work further emphasizes the importance of the environmental, terrestrial parameters for the functionality of a microbial community, but also reveals a high proportion of living microorganisms in extreme environments with a high adaptation potential within habitability borders.

    Beschreibung: In contrast to the well-recognized permafrost carbon (C) feedback to climate change, the fate of permafrost nitrogen (N) after thaw is poorly understood. According to mounting evidence, part of the N liberated from permafrost may be released to the atmosphere as the strong greenhouse gas (GHG) nitrous oxide (N2O). Here, we report post-thaw N2O release from late Pleistocene permafrost deposits called Yedoma, which store a substantial part of permafrost C and N and are highly vulnerable to thaw. While freshly thawed, unvegetated Yedoma in disturbed areas emit little N2O, emissions increase within few years after stabilization, drying and revegetation with grasses to high rates (548 (133–6286) μg N m−2 day−1; median with (range)), exceeding by 1–2 orders of magnitude the typical rates from permafrost-affected soils. Using targeted metagenomics of key N cycling genes, we link the increase in in situ N2O emissions with structural changes of the microbial community responsible for N cycling. Our results highlight the importance of extra N availability from thawing Yedoma permafrost, causing a positive climate feedback from the Arctic in the form of N2O emissions.

    Beschreibung: The right timing of animal physiology and behaviour ensures the stability of populations and ecosystems. To predict anthropogenic impacts on these timings, more insight is needed into the interplay between environment and molecular timing mechanisms. This is particularly true in marine environments. Using high-resolution, long-term daylight measurements from a habitat of the marine annelid Platynereis dumerilii, we found that temporal changes in ultraviolet A (UVA)/deep violet intensities, more than longer wavelengths, can provide annual time information, which differs from annual changes in the photoperiod. We developed experimental set-ups that resemble natural daylight illumination conditions, and automated, quantifiable behavioural tracking. Experimental reduction of UVA/deep violet light (approximately 370–430 nm) under a long photoperiod (16 h light and 8 h dark) significantly decreased locomotor activities, comparable to the decrease caused by a short photoperiod (8 h light and 16 h dark). In contrast, altering UVA/deep violet light intensities did not cause differences in locomotor levels under a short photoperiod. This modulation of locomotion by UVA/deep violet light under a long photoperiod requires c-opsin1, a UVA/deep violet sensor employing Gi signalling. C-opsin1 also regulates the levels of rate-limiting enzymes for monogenic amine synthesis and of several neurohormones, including pigment-dispersing factor, vasotocin (vasopressin/oxytocin) and neuropeptide Y. Our analyses indicate a complex inteplay between UVA/deep violet light intensities and photoperiod as indicators of annual time.

    Beschreibung: The Antarctic Circumpolar Current (ACC) plays a crucial role in global ocean circulation by fostering deep-water upwelling and formation of new water masses. On geological timescales, ACC variations are poorly constrained beyond the last glacial. Here, we reconstruct changes in ACC strength in the central Drake Passage in vicinity of the modern Polar Front over a complete glacial-interglacial cycle (i.e., the past 140,000 years), based on sediment grain-size and geochemical characteristics. We found significant glacial-interglacial changes of ACC flow speed, with weakened current strength during glacials and a stronger circulation in interglacials. Superimposed on these orbital-scale changes are high-amplitude millennialscale fluctuations, with ACC strength maxima correlating with diatom-based Antarctic winter sea-ice minima, particularly during full glacial conditions. We infer that the ACC is closely linked to Southern Hemisphere millennial-scale climate oscillations, amplified through Antarctic sea ice extent changes. These strong ACC variations modulated Pacific-Atlantic water exchange via the “cold water route” and potentially affected the Atlantic Meridional Overturning Circulation and marine carbon storage.

    Beschreibung: The aim of this paper is to investigate, through fnancial-statement analyzes, the economic fnancial performance of Italian hotels, after the international economic crisis, also con sidering the possible macro-regional diferences. The study focuses the fnancial state ments of 5473 hotels from 2009 to 2018. National data are also disaggregated in the three macro-areas that characterize Italy for diferent social and economic aspects. Anova test and Tukey–Kramer test are used. Results show that the crisis afected proftability. Ital ian hotels have a low capitalization, unable to cope with the large structural investments that require signifcant debts. The proftability indicators record similar trends in the three macro-areas, while the fnancial independence index and the coverage index show signif cant diferent values in the three observed areas. Therefore, in the digital era, Italian hotel industry has all the potential to restructure itself. Here fve ratios are considered to observe medium sized hotels. Future research with other variables will be useful, even on smaller hotels, and the analysis of their trends by cohorts of companies is necessary, as well as the integration of quantitative data with qualitative evidence. This paper encourages the cul ture of temporal sector comparison, re-evaluating the potential of accounting information systems, in order to promote data-based growth and development strategies. Furthermore, it contains indications for government ofcials, as well as for countries in the process of developing the hospitality sector following the example of the Italian experience.

    Beschreibung: During the last ice age, the Northern Hemisphere experienced a series of abrupt millennial-scale climatic changes linked to variations in the strength of the Atlantic Meridional Overturning Circulation and sea-ice extent. However, our understanding of their impacts on decadal-scale climate variability in central Europe has been limited by the lack of high-resolution continental archives. Here, we present a near annual-resolution climate proxy record of central European temperature reconstructed from the Eifel maar lakes of Holzmaar and Auel in Germany, spanning the past 60,000 years. The lake sediments reveal a series of previously undocumented multidecadal climate cycles of around 20 to 150 years that persisted through the last glacial cycle. The periodicity of these cycles suggests that they are related to the Atlantic multidecadal climate oscillations found in the instrumental record and in other climate archives during the Holocene. Our record shows that multidecadal variability in central Europe was strong during all warm interstadials, but was substantially muted during all cold stadial periods. We suggest that this decrease in multidecadal variability was the result of the atmospheric circulation changes associated with the weakening of the Atlantic Meridional Overturning Circulation and the expansion of North Atlantic sea-ice cover during the coldest parts of the last ice age.

    Beschreibung: Synthetic polypeptides prepared from N-carboxyanhydrides (NCAs) of α-amino acids are useful for elucidating the relationship between the primary structure of natural peptides and their immunogenicity. In this study, complex copolypeptide sequences were prepared using a recently developed technique; specifically, the random copolymerization of l-alanine NCA with NCAs of l-glutamic acid 5-benzylester (Bn-Glu NCA), S-benzyl-cysteine (Bn-Cys NCA), O-benzyl-l-serine (Bn-Ser NCA), and l-phenylalanine (Phe NCA) was performed using N-heterocyclic carbene (NHC) catalysts. The NHC-initiated Ala NCA/Bn-Glu NCA and Ala NCA/Bn-Cys NCA copolymerization reactions achieved 90% conversion within 30 min. The reactivity ratio values estimated using the Kelen and Tüdos method show that poly(Bn-Glu-co-Ala) and poly(Bn-Cys-co-Ala) have random repeating units with rich alternating sequences, whereas poly(Bn-Ser-co-Ala) and poly(Phe-co-Ala) contain a larger proportion of Ala-repeating units than Bn-Ser and Phe in random placement.

    Beschreibung: The process of spermatogenesis is complex and systemic, requiring the cooperation of many regulators. However, little is known about how micro RNAs (miRNAs) regulate spermatogenesis in poultry. In this study, we investigated key miRNAs and their target genes that are involved in spermatogenesis in chickens. Next-generation sequencing was conducted to determine miRNA expression profiles in five cell types: primordial germ cells (PGCs), spermatogonial stem cells (SSCs), spermatogonia (Spa), and chicken sperm. Next, we analyzed and identified several key miRNAs that regulate spermatogenesis in the four germline cell miRNA profiles. Among the enriched miRNAs, miRNA-301a-5p was the key miRNA in PGCs, SSCs, and Spa. Through reverse transcription quantitative PCR (RT-qPCR), dual-luciferase, and miRNA salience, we confirmed that miR-301a-5p binds to transforming growth factor-beta 2 (TGFβ2) and is involved in the transforming growth factor-beta (TGF-β) signaling pathway and germ cell development. To the best of our knowledge, this is the first demonstration of miR-301a-5p involvement in spermatogenesis by direct binding to TGFβ2, a key gene in the TGF-β signaling pathway. This finding contributes to the insights into the molecular mechanism through which miRNAs regulate germline cell differentiation and spermatogenesis in chickens.

    Beschreibung: Ursolic acid (UA), a pentacyclic triterpenoid extracted from various plants, inhibits cell growth, metastasis, and tumorigenesis in various cancers. Chemotherapy resistance and the side effects of paclitaxel (PTX), a traditional chemotherapy reagent, have limited the curative effect of PTX in esophageal cancer. In this study, we investigate whether UA promotes the anti-tumor effect of PTX and explore the underlying mechanism of their combined effect in esophageal squamous cell carcinoma (ESCC). Combination treatment with UA and PTX inhibited cell proliferation and cell growth more effectively than either treatment alone by inducing more significant apoptosis, as indicated by increased sub-G1 phase distribution and protein levels of cleaved-PARP and cleaved caspase-9. Similar to the cell growth suppressive effect, the combination of UA and PTX significantly inhibited cell migration by targeting uPA, MMP-9, and E-cadherin in ESCC cells. In addition, combination treatment with UA and PTX significantly activated p-GSK-3β and suppressed the activation of Akt and FOXM1 in ESCC cells. Those effects were enhanced by the Akt inhibitor LY2940002 and inverted by the Akt agonist SC79. In an in vivo evaluation of a murine xenograft model of esophageal cancer, combination treatment with UA and PTX suppressed tumor growth significantly better than UA or PTX treatment alone. Thus, UA effectively potentiates the anti-tumor efficacy of PTX by targeting the Akt/FOXM1 cascade since combination treatment shows significantly more anti-tumor potential than PTX alone both in vitro and in vivo. Combination treatment with UA and PTX could be a new strategy for curing esophageal cancer patients.

    Beschreibung: White adipocytes contribute to energy storage, accumulating lipid droplets, whereas brown and beige adipocytes mainly function in dissipating energy as heat primarily via the action of uncoupling protein 1 (UCP1). Bone morphogenic protein 7 (BMP7) was shown to drive brown adipocyte differentiation in murine interscapular adipose tissue. Here, we performed global RNA-sequencing and functional assays on adipocytes obtained from subcutaneous (SC) and deep-neck (DN) depots of human neck and differentiated with or without BMP7. We found that BMP7 did not influence differentiation but upregulated browning markers, including UCP1 mRNA and protein in SC and DN derived adipocytes. BMP7 also enhanced mitochondrial DNA content, levels of oxidative phosphorylation complex subunits, along with PGC1α and p-CREB upregulation, and fragmentation of mitochondria. Furthermore, both UCP1-dependent proton leak and UCP1-independent, creatine-driven substrate cycle coupled thermogenesis were augmented upon BMP7 addition. The gene expression analysis also shed light on the possible role of genes unrelated to thermogenesis thus far, including ACAN, CRYAB, and ID1, which were among the highest upregulated ones by BMP7 treatment in both types of adipocytes. Together, our study shows that BMP7 strongly upregulates thermogenesis in human neck area derived adipocytes, along with genes, which might have a supporting role in energy expenditure.

    Beschreibung: In this paper, 4,4′-diaminodiphenyl ether and 2,2′-bis (trifluoromethyl)-4,4′-diaminophenyl ether are selected for molecular structure design, and PAI materials are synthesized by acyl chloride method. 2,2′-bis (trifluoromethyl)-4,4′-diaminophenyl ether has the same main chain structure as 4,4′-diaminodiphenyl ether, but the side chain contains two trifluoromethyl groups, which has high fluorine content. PAI terpolymers were prepared by compounding two diamine monomers, and the effects of trifluoromethyl on heat resistance, friction and wear properties, hydrophobic properties and mechanical properties of PAI materials were studied. The results showed that with the increase of trifluoromethyl content, the Tg of PAI material first increased and then changed little, and the Td5% would decrease and the tensile properties would also decrease. The wear mechanism of PAI varied with the content of trifluoromethyl. With the increase of the amount of fluorinated diamine monomer, the adhesive wear degree of PAI materials gradually increased, and reached the maximum when the molar ratio of the two monomers was 5:5, and then decreased gradually. Different trifluoromethyl content had little effect on friction coefficient, and the friction coefficient increased slightly when the molar ratio of 4,4′-diaminodiphenyl ether to 2,2′-bis (trifluoromethyl)-4,4′-diaminophenyl ether is 1:9. With the increase of trifluoromethyl content, the wear of PAI material would increase. With the increase of the amount of trifluoromethyl, the water absorption of PAI material decreased and the water contact angle increased, which indicated that the hydrophobic property of PAI material was improved. To sum up, the results of this study showed that the introduction of trifluoromethyl into the side chain provided an effective way to prepare PAI materials with low water absorption. Considering the comprehensive properties such as heat resistance, friction and wear, tensile properties, etc., the appropriate addition amount is 10–30%.

    Beschreibung: Railway vehicle makers manufacture the bogie frame by welding medium-strength carbon steel sheets. It has been a long-standing practice to perform post-weld heat treatment (PWHT) to remove welding-residual stress, but rail car manufacturers are moving toward producing bogie frames without PWHT. Since securing the fatigue strength of the bogie frame is essential for vehicle operation safety, it is necessary to systematically evaluate the effects of PWHT on hardness, microstructure, mechanical properties, corrosion, fatigue strength, etc. In this study, small-scale welding specimens and full-size components were produced using S355JR used in general structures, automobiles, shipbuilding, railroad vehicles, etc. The effect of PWHT on material properties-the hardness of the base material, heat-affected zone and weld metal, microstructure, shock absorption energy, yield strength, tensile strength, and fatigue were investigated. When the weld specimen was annealed at 590 °C and 800 °C for 1 h, the yield strength and tensile strength of the specimen decreased, but the elongation increased. For specimens not heat-treated, the parent material’s yield strength, the yield strength in HAZ, and the yield strength of the weld metal were 350 MPa, 345 MPa, and 340 MPa. For specimens heat-treated at 590 °C, they were 350 MPa, 345 MPa, and 340 MPa. For specimens heat-treated at 800 °C, they were 350 MPa, 345 MPa, and 340 MPa. Annealing heat treatment of the specimen at 800 °C homogenized the structure of the weldments similar to that of the base material and slightly improved the shock absorption energy. For specimens not heat-treated, the Charpy impact absorption energies at 20 °C of the parent material and weld metal were 291.5 J and 187 J. For specimens heat-treated at 590 °C, they were 276 J and 166 J. For specimens heat-treated at 800 °C, the Charpy impact absorption energy at 20 °C of the parent material was 299 J. PWHT at 590 °C had the effect of slightly improving the fatigue limit of the specimen but lowered the fatigue limit by 10.8% for the component specimen.

    Beschreibung: Dramatic urban land expansion and its internal sub-fraction change during 2000–2020 have taken place in Africa; however, the investigation of their spatial heterogeneity and dynamic change monitoring at the continental scale are rarely reported. Taking the whole of Africa as a study area, the synergic approach of normalized settlement density index and random forest was applied to assess urban land and its sub-land fractions (i.e., impervious surface area and vegetation space) in Africa, through time series of remotely sensed images on a cloud computing platform. The generated 30-m resolution urban land/sub-land products displayed good accuracy, with comprehensive accuracy of over 90%. During 2000–2020, the evaluated urban land throughout Africa increased from 1.93 × 104 km2 to 4.18 × 104 km2, with a total expansion rate of 116.49%, and the expanded urban area of the top six countries accounted for more than half of the total increments, meaning that the urban expansion was concentrated in several major countries. A turning green Africa was observed, with a continuously increasing ratio of vegetation space to built-up area and a faster increment of vegetation space than impervious surface area (i.e., 134.43% vs., 108.88%) within urban regions. A better living environment was also found in different urbanized regions, as the newly expanded urban area was characterized by lower impervious surface area fraction and higher vegetation fraction compared with the original urban area. Similarly, the humid/semi-humid regions also displayed a better living environment than arid/semi-arid regions. The relationship between socioeconomic development factors (i.e., gross domestic product and urban population) and impervious surface area was investigated and both passed the significance test (p 〈 0.05), with a higher fit value in the former than the latter. Overall, urban land and its fractional land cover change in Africa during 2000–2020 promoted the well-being of human settlements, indicating the positive effect on environments.

    Beschreibung: This paper studies the power of online search intensity metrics, measured by Google, for examining and forecasting exchange rates. We use panel data consisting of quarterly time series from 2004 to 2018 and ten international countries with the highest currency trading volume. Newly, we include various Google search intensity metrics to our panel data. We find that online search improves the overall econometric models and fits. First, four out of ten search variables are robustly significant at one percent and enhance the macroeconomic exchange rate models. Second, country regressions corroborate the panel results, yet the predictive power of search intensity with regard to exchange rates vary by country. Third, we find higher prediction performance for our exchange rate models with search intensity, particularly in regard to the direction of the exchange rate. Overall, our approach reveals a value-added of search intensity in exchange rate models.

    Beschreibung: Daily use of wholegrain foods is generally recommended due to strong epidemiological evidence of reduced risk of chronic diseases. Cereal grains, especially the bran part, have a high content of dietary fiber (DF). Cereal DF is an umbrella concept of heterogeneous polysaccharides of variable chemical composition and molecular weight, which are combined in a complex network in cereal cell walls. Cereal DF and its distinct components influence food digestion throughout the gastrointestinal tract and influence nutrient absorption and other physiological reactions. After repeated consumption of especially whole grain cereal foods, these effects manifest in well-demonstrated health benefits. As cereal DF is always consumed in the form of processed cereal food, it is important to know the effects of processing on DF to understand, safeguard and maximize these health effects. Endogenous and microbial enzymes, heat and mechanical energy during germination, fermentation, baking and extrusion destructurize the food and DF matrix and affect the quantity and properties of grain DF components: arabinoxylans (AX), beta-glucans, fructans and resistant starch (RS). Depolymerization is the most common change, leading to solubilization and loss of viscosity of DF polymers, which influences postprandial responses to food. Extensive hydrolysis may also remove oligosaccharides and change the colonic fermentability of DF. On the other hand, aggregation may also occur, leading to an increased amount of insoluble DF and the formation of RS. To understand the structure–function relationship of DF and to develop foods with targeted physiological benefits, it is important to invest in thorough characterization of DF present in processed cereal foods. Such understanding also demands collaborative work between food and nutritional sciences.

    Beschreibung: The COVID-19 pandemic affected consumers’ behavior worldwide. This paper aims to analyze consumers’ sustainable food behavior during the COVID-19 pandemic. The research was based on an online survey during May–October 2020 among 859 Romanian consumers. Principal component analysis and cluster analysis were employed to group the consumers based on their behavior. Furthermore, the binary-logistic regression was used to identify the socio-demographic profile of the identified groups. Based on the cluster analysis, the consumers were grouped into three main groups: indifferent, pro-environment protection, and health concerned. The results indicate a positive attitude towards sustainable food behavior. Consumers’ attitude towards sustainable food choice is mainly influenced by age and education level. The study offers valuable information for future public policy and marketing campaign regarding sustainable food behavior.

    Beschreibung: The increasing use of magnetic nanoparticles as heating agents in biomedicine is driven by their proven utility in hyperthermia therapeutic treatments and heat-triggered drug delivery methods. The growing demand of efficient and versatile nanoheaters has prompted the creation of novel types of magnetic nanoparticle systems exploiting the magnetic interaction (exchange or dipolar in nature) between two or more constituent magnetic elements (magnetic phases, primary nanoparticles) to enhance and tune the heating power. This process occurred in parallel with the progress in the methods for the chemical synthesis of nanostructures and in the comprehension of magnetic phenomena at the nanoscale. Therefore, complex magnetic architectures have been realized that we classify as: (a) core/shell nanoparticles; (b) multicore nanoparticles; (c) linear aggregates; (d) hybrid systems; (e) mixed nanoparticle systems. After a general introduction to the magnetic heating phenomenology, we illustrate the different classes of nanoparticle systems and the strategic novelty they represent. We review some of the research works that have significantly contributed to clarify the relationship between the compositional and structural properties, as determined by the synthetic process, the magnetic properties and the heating mechanism.

    Beschreibung: The significant growth in the use of the Internet and the rapid development of network technologies are associated with an increased risk of network attacks. Network attacks refer to all types of unauthorized access to a network including any attempts to damage and disrupt the network, often leading to serious consequences. Network attack detection is an active area of research in the community of cybersecurity. In the literature, there are various descriptions of network attack detection systems involving various intelligent-based techniques including machine learning (ML) and deep learning (DL) models. However, although such techniques have proved useful within specific domains, no technique has proved useful in mitigating all kinds of network attacks. This is because some intelligent-based approaches lack essential capabilities that render them reliable systems that are able to confront different types of network attacks. This was the main motivation behind this research, which evaluates contemporary intelligent-based research directions to address the gap that still exists in the field. The main components of any intelligent-based system are the training datasets, the algorithms, and the evaluation metrics; these were the main benchmark criteria used to assess the intelligent-based systems included in this research article. This research provides a rich source of references for scholars seeking to determine their scope of research in this field. Furthermore, although the paper does present a set of suggestions about future inductive directions, it leaves the reader free to derive additional insights about how to develop intelligent-based systems to counter current and future network attacks.

    Beschreibung: This paper presents the results of an experimental program aimed at the assessment of the freeze–thaw (F–T) resistance of concrete based on the evaluation of fracture tests accompanied by acoustic emission measurements. Two concretes of similar mechanical characteristics were manufactured for the experiment. The main difference between the C1 and C2 concrete was in the total number of air voids and in the A300 parameter, where both parameters were higher for C1 by about 35% and 52%, respectively. The evaluation of the fracture characteristics was performed on the basis of experimentally recorded load–deflection and load–crack mouth opening displacement diagrams using two different approaches: linear fracture mechanics completed with the effective crack model and the double-K model. The results show that both approaches gave similar results, especially if the nonlinear behavior before the peak load was considered. According to the results, it can be stated that continuous AE measurement is beneficial for the assessment of the extent of concrete deterioration, and it suitably supplements the fracture test evaluation. A comparison of the results of fracture tests with the resonance method and splitting tensile strength test shows that all testing methods led to the same conclusion, i.e., the C1 concrete was more F–T-resistant than C2. However, the fracture test evaluation provided more detailed information about the internal structure deterioration due to the F–T exposure.

    Beschreibung: Through proper arranging of a hybrid combination of longitudinal fiber reinforced polymer (FRP) bars and steel bars in the tensile region of the beam, the advantages of both FRP and steel materials can be sufficiently exploited to enhance the flexural capacity and ductility of a concrete beam. In this paper, a methodology for the flexural strength design of hybrid FRP-steel reinforced concrete (RC) beams is proposed. Firstly, based on the mechanical features of reinforcement and concrete and according to the latest codified provisions of longitudinal reinforcement conditions to ensure ductility level, the design-oriented allowable ranges of reinforcement ratio corresponding to three common flexural failure modes are specified. Subsequently, the calculation approach of nominal flexural strength of hybrid FRP-steel RC beams is established following the fundamental principles of equilibrium and compatibility. In addition to the common moderately-reinforced beams, the proposed general calculation approach is also applicable to lightly-reinforced beams and heavily-reinforced beams, which are widely used but rarely studied. Furthermore, the calculation process is properly simplified and the calculation accuracy is validated by the experimental results of hybrid FRP-steel RC beams in the literature. Finally, with the ductility analysis, a novel strength reduction factor represented by net tensile steel strain and reinforcement ratio is proposed for hybrid FRP-steel RC beams.

    Beschreibung: There have been numerous recent studies on improving the mechanical properties and durability of cement composites by mixing them with functional polymers. However, research into applying modified biopolymer such as catechol-functionalized chitosan to cement mortar or concrete is rare to the best of our knowledge. In this study, catechol-functionalized chitosan (Cat-Chit), a well-known bioinspired polymer that imitates the basic structures and functions of living organisms and biological materials in nature, was synthesized and combined with cement mortar in various proportions. The compressive strength, tensile strength, drying shrinkage, accelerated carbonation depth, and chloride-ion penetrability of these mixes were then evaluated. In the ultraviolet–visible spectra, a maximum absorption peak appeared at 280 nm, corresponding to catechol conjugation. The sample containing 7.5% Cat-Chit polymer in water (CPW) exhibited the highest compressive strength, and its 28-day compressive strength was ~20.2% higher than that of a control sample with no added polymer. The tensile strength of the samples containing 5% or more CPW was ~2.3–11.5% higher than that of the control sample. Additionally, all the Cat-Chit polymer mixtures exhibited lower carbonation depths than compared to the control sample. The total charge passing through the samples decreased as the amount of CPW increased. Thus, incorporating this polymer effectively improved the mechanical properties, carbonation resistance, and chloride-ion penetration resistance of cement mortar.

    Beschreibung: Charcot-Marie-Tooth disease type 2A (CMT2A) is the most common hereditary axonal neuropathy caused by mutations in MFN2 encoding Mitofusin-2, a multifunctional protein located in the outer mitochondrial membrane. In order to study the effects of a novel MFN2K357T mutation associated with early onset, autosomal dominant severe CMT2A, we generated a knock-in mouse model. While Mfn2K357T/K357T mouse pups were postnatally lethal, Mfn2+/K357T heterozygous mice were asymptomatic and had no histopathological changes in their sciatic nerves up to 10 months of age. However, immunofluorescence analysis of Mfn2+/K357T mice revealed aberrant mitochondrial clustering in the sciatic nerves from 6 months of age, in optic nerves from 8 months, and in lumbar spinal cord white matter at 10 months, along with microglia activation. Ultrastructural analyses confirmed dysmorphic mitochondrial aggregates in sciatic and optic nerves. After exposure of 6-month-old mice to lipopolysaccharide, Mfn2+/K357T mice displayed a higher immune response, a more severe motor impairment, and increased CNS inflammation, microglia activation, and macrophage infiltrates. Overall, ubiquitous Mfn2K357T expression renders the CNS and peripheral nerves of Mfn2+/K357T mice more susceptible to mitochondrial clustering, and augments their response to inflammation, modeling some cellular mechanisms that may be relevant for the development of neuropathy in patients with CMT2A.

    Beschreibung: The North Railroad Avenue Plume, discovered in 1989, contained chlorinated solvent groundwater plumes extending over 23.5 hectares (58 acres) and three hydrostratigraphic units. The source contaminant, tetrachloroethene, stemmed from release at a dry cleaner/laundromat business. The anaerobic biodegradation byproducts trichloroethene, isomers of dichloroethene (DCE), and vinyl chloride were detected in groundwater samples collected prior to remedial action. The impacted aquifers are the sole source drinking water aquifers for the communities near the site. Following the remedial investigation and feasibility study, the selected alternative for full-scale remedial action at the site was enhanced reductive dichlorination (ERD) focused on four treatment areas: the shallow source zone, the shallow hotspot area, the shallow downgradient area, and the deep zone. Pilot testing, which was conducted in the source zone and hotspot areas, is the subject of this paper. The primary objectives of the pilot test were to obtain the necessary information to select an ERD treatment formulation, dose, and frequency of dosing for use during full-scale remedial action, as well as to refine the site’s hydrogeologic conceptual site model and design parameters. Four (4) test cells, each of which contained well pairs of injection and downgradient extraction wells, were used to test ERD bio-amendment formulations: ethyl lactate, dairy whey, emulsified vegetable oil (EVO), and a combination of EVO and a hydrogen gas infusion. A conservative tracer, bromide, was added to the recirculation flow to record tracer breakthrough, peak, and dissipation at extraction wells. The results of these dipole tracer tests were used to reassess the hydraulic conductivity and hydrodynamic dispersity used in the remedial design. In addition to water quality analyses of contaminants and substrates, groundwater samples were also analyzed for biological analyses before, during, and after the addition of bioamendment. Analyses of phospholipid fatty acids and deoxyribonucleic acid (DNA) extracts from fresh groundwater samples informed decisions on the capacity for complete ERD without DCE stalling and tracked the shifts in the bacterial and archaeal taxonomy and phylogeny stemming from the addition of bioamendments. The pilot test concluded that EVO was the most suitable, considering (1) support of the native microbial consortia for ERD, (2) mechanics and hydraulics of the remediation system, and (3) sustainability/retention of the substrate in the subsurface. Along with EVO, the addition of a nutrient broth derived from brewery waste accelerated and sustained the desired conditions and microbial diversity and population levels. The pilot test results were also used to assess the utilization kinetics of the injected substrates based on total organic carbon (TOC) concentrations measured in the groundwater. After determining that substrate utilization followed Monod kinetics, a TOC threshold at 300 milligrams per liter, equivalent to approximately twice its half-saturation constant was established. Full scale treatment dosing and dose frequency were designed around this threshold, assuming the maximum substrate utilization would yield optimum ERD.

    Beschreibung: Strong evidence from studies on primates and rodents shows that changes in pupil diameter may reflect neural activity in the locus coeruleus (LC). Pupillometry is the only available non-invasive technique that could be used as a reliable and easily accessible real-time biomarker of changes in the in vivo activity of the LC. However, the application of pupillometry to preclinical research in rodents is not yet fully standardized. A lack of consensus on the technical specifications of some of the components used for image recording or positioning of the animal and cameras have been recorded in recent scientific literature. In this study, a novel pupillometry system to indirectly assess, in real-time, the function of the LC in anesthetized rodents is presented. The system comprises a deep learning SOLOv2 instance-based fast segmentation framework and a platform designed to place the experimental subject, the video cameras for data acquisition, and the light source. The performance of the proposed setup was assessed and compared to other baseline methods using a validation and an external test set. In the latter, the calculated intersection over the union was 0.93 and the mean absolute percentage error was 1.89% for the selected method. The Bland–Altman analysis depicted an excellent agreement. The results confirmed a high accuracy that makes the system suitable for real-time pupil size tracking, regardless of the pupil’s size, light intensity, or any features typical of the recording process in sedated mice. The framework could be used in any neurophysiological study with sedated or fixed-head animals.

    Beschreibung: Along with the cells that are exposed to radiation, non-irradiated cells can unveil radiation effects as a result of intercellular communication, which are collectively defined as radiation induced bystander effects (RIBE). Exosome-mediated signalling is one of the core mechanisms responsible for multidirectional communication of tumor cells and their associated microenvironment, which may result in enhancement of malignant tumor phenotypes. Recent studies show that exosomes and exosome-mediated signalling also play a dynamic role in RIBE in cancer cell lines, many of which focused on altered exosome cargo or their effects on DNA damage. However, there is a lack of knowledge regarding how these changes in exosome cargo are reflected in other functional characteristics of cancer cells from the aspects of invasiveness and metastasis. Therefore, in the current study, we aimed to investigate exosome-mediated bystander effects of 2 Gy X-ray therapeutic dose of ionizing radiation on the invasive potential of MCF-7 breast cancer cells in vitro via assessing Matrigel invasion potential, epithelial mesenchymal transition (EMT) characteristics and the extent of glycosylation, as well as underlying plausible molecular mechanisms. The findings show that exosomes derived from irradiated MCF-7 cells enhance invasiveness of bystander MCF-7 cells, possibly through altered miRNA and protein content carried in exosomes.

    Beschreibung: Chronic inflammation promotes endothelial plasticity, leading to the development of several diseases, including fibrosis and cancer in numerous organs. The basis of those processes is a phenomenon called the endothelial–mesenchymal transition (EndMT), which results in the delamination of tightly connected endothelial cells that acquire a mesenchymal phenotype. EndMT-derived cells, known as the myofibroblasts or cancer-associated fibroblasts (CAFs), are characterized by the loss of cell–cell junctions, loss of endothelial markers, and gain in mesenchymal ones. As a result, the endothelium ceases its primary ability to maintain patent and functional capillaries and induce new blood vessels. At the same time, it acquires the migration and invasion potential typical of mesenchymal cells. The observed modulation of cell shape, increasedcell movement, and invasion abilities are connected with cytoskeleton reorganization. This paper focuses on the review of current knowledge about the molecular pathways involved in the modulation of each cytoskeleton element (microfilaments, microtubule, and intermediate filaments) during EndMT and their role as the potential targets for cancer and fibrosis treatment.

    Beschreibung: Calmodulin (CaM), as an important factor in the calcium signaling pathway, is widely involved in plant growth and development regulation and responses to external stimuli. In this study, the full-length sequence of the ScCaM gene (GenBank: GQ246454) was isolated from the leaves of a Saccharum spp. hybrid. Prokaryotic expression showed that ScCaM could be solubly expressed and purified in Escherichia coli BL21. Subcellular localization confirmed that ScCaM was localized in the plasma membrane and nucleus of cells. A quantitative reverse transcription polymerase chain reaction (qRT-PCR) analysis revealed that ScCaM can be induced by various stresses, including sodium chloride (NaCl), chromium trichloride (CrCl3), salicylic acid (SA), and methyl jasmonate (MeJA). Ectopic expression in Arabidopsis thaliana demonstrated that ScCaM can affect the growth and development of transgenic plants. Moreover, the qRT-PCR analysis indicated that the overexpression of the allogenic ScCaM gene inhibits the expression of AtSTM, leading to the phenomenon of multiple-tillering in transgenic A. thaliana. The present study provided valuable information and facilitates further investigation into the function of ScCaM in the future.

    Beschreibung: Essential Oils (EOs) are expensive hydrocarbons produced exclusively by specific species in the plant kingdom. Their applications have deep roots in traditional herbal medicine, which lacks scientific evidence. Nowadays, more than ever, there is a growing global interest in research-based discoveries that maintain and promote health conditions. Consequently, EOs became a much attractive topic for both research and industry, with revenues reaching billions of dollars annually. In this work, we provide key guidance to all essential oil-bearing plants growing in the United Arab Emirates (UAE). The comprehensive data were collected following an extensive, up-to-date literature review. The results identified 137 plant species, including indigenous and naturalized ones, in the UAE, citing over 180 published research articles. The general overview included plant botanical names, synonyms, common names (Arabic and English), families and taxonomic authority. The study acts as a baseline and accelerator for research, industry and discoveries in multiple disciplines relying on essential oil-bearing plants.

    Beschreibung: This paper considers a variant of the Vehicle Routing Problem with Time Windows, with site dependencies, multiple depots and outsourcing costs. This problem is the basis for many technician routing problems. Having both site-dependency and time window constraints lresults in difficulties in finding feasible solutions and induces highly constrained instances. Matheuristics based on Mixed Integer Linear Programming compact formulations are firstly designed. Column Generation matheuristics are then described by using previous matheuristics and machine learning techniques to stabilize and speed up the convergence of the Column Generation algorithm. The computational experiments are analyzed on public instances with graduated difficulties in order to analyze the accuracy of algorithms for ensuring feasibility and the quality of solutions for weakly to highly constrained instances. The results emphasize the interest of the multiple types of hybridization between mathematical programming, machine learning and heuristics inside the Column Generation framework. This work offers perspectives for many extensions of technician routing problems.

    Beschreibung: Methionine restriction (MetR) is an efficient method of amino acid restriction (AR) in cells and organisms that induces low energy metabolism (LEM) similar to caloric restriction (CR). The implementation of MetR as a therapy for cancer or other diseases is not simple since the elimination of a single amino acid in the diet is difficult. However, the in vivo turnover rate of cysteine is usually higher than the rate of intake through food. For this reason, every cell can enzymatically synthesize cysteine from methionine, which enables the use of specific enzymatic inhibitors. In this work, we analysed the potential of cysteine restriction (CysR) in the murine cell line L929. This study determined metabolic fingerprints using mass spectrometry (LC/MS). The profiles were compared with profiles created in an earlier work under MetR. The study was supplemented by proliferation studies using D-amino acid analogues and inhibitors of intracellular cysteine synthesis. CysR showed a proliferation inhibition potential comparable to that of MetR. However, the metabolic footprints differed significantly and showed that CysR does not induce classic LEM at the metabolic level. Nevertheless, CysR offers great potential as an alternative for decisive interventions in general and tumour metabolism at the metabolic level.

    Beschreibung: Hot pepper (Capsicum annum L.) is a major spice crop and is used worldwide for its nutritional value. In the field, its plant is susceptible to various fungal diseases, including fusarium wilt, caused by soil-borne fungus Fusarium oxysporum f. sp. capsici, which can survive in the soil for several years. The infected plant can be recognized by the yellowing of older leaves and downward curling of apical shoots, followed by plant wilting and ultimately the death of the plant. The resistance mechanism in plants is controlled by a single dominant gene, and conventional plant breeding techniques are used to develop a wilt-resistant germplasm. Non-conventional techniques such as gene pyramiding and expression enhancement of antifungal genes could be used to shorten the time to develop resistance against fusarium wilt in hot peppers. In this review, we discuss different aspects of the disease and the molecular basis of resistance in chili/hot pepper plants. Furthermore, this review covers the scope of conventional and non-conventional breeding strategies and different management approaches used to tackle the disease.

    Beschreibung: Background: Several immunotherapy (IT) agents are FDA approved for treatment of melanoma and non-small-cell lung cancer (NSCLC). The addition of stereotactic radiosurgery (SRS) or stereotactic body radiation therapy (SBRT) to immunotherapy looks promising. A systematic review was conducted to evaluate the possible synergistic effects of immune checkpoints inhibitors (ICIs) and stereotactic radiation therapy in melanoma and NSCLC. Materials and methods: Pubmed databases from January 2010 to December 2020 were reviewed to identify English language studies reporting control of local and abscopal effect of the combination of ICI-SBRT/SRS in metastatic NSCLC and melanoma cancer. The inclusion criteria were followed according to PICO criteria. Results: Thirty-nine articles were included of the 2141 initial results. The reported rates for local control were 16.5–100% and 40–94% in brain and extracerebral metastases, respectively. Distant/abscopal response rates were 1–45% in extracerebral metastases. Abscopal effect could not be evaluated in brain metastases because it was not reported in studies. Treatments were well tolerated with few grade 4 toxicities and no grade 5. Conclusions: The combined treatment of ICI-SBRT/SRS achieves high local control and non-negligible abscopal response in patients with extracerebral metastases, with its benefit in cerebral metastases being more controversial. Clinical trials are needed to better characterize the potential synergism.

    Beschreibung: Dittrichia viscosa plants were grown hydroponically with different concentrations of Sb. There was preferential accumulation of Sb in roots. Fe and Cu decreased, while Mn decreased in roots but not in leaves. Chlorophyll content declined, but the carotenoid content increased, and photosynthetic efficiency was unaltered. O2●− generation increased slightly, while lipid peroxidation increased only in roots. H2O2, NO, ONOO−, S-nitrosothiols, and H2S showed significant increases, and the enzymatic antioxidant system was altered. In roots, superoxide dismutase (SOD) and monodehydroascorbate reductase (MDAR) activities declined, dehydroscorbate reductase (DHAR) rose, and ascorbate peroxidase (APX), peroxidase (POX), and glutathione reductase (GR) were unaffected. In leaves, SOD and POX increased, MDAR decreased, and APX was unaltered, while GR increased. S-nitrosoglutathione reductase (GSNOR) and l-cysteine desulfhydrilase (l-DES) increased in activity, while glutathione S-transferase (GST) decreased in leaves but was enhanced in roots. Components of the AsA/GSH cycle decreased. The great capacity of Dittrichia roots to accumulate Sb is the reason for the differing behaviour observed in the enzymatic antioxidant systems of the two organs. Sb appears to act by binding to thiol groups, which can alter free GSH content and SOD and GST activities. The coniferyl alcohol peroxidase activity increased, possibly to lignify the roots’ cell walls. Sb altered the ROS balance, especially with respect to H2O2. This led to an increase in NO and H2S acting on the antioxidant system to limit that Sb-induced redox imbalance. The interaction NO, H2S and H2O2 appears key to the response to stress induced by Sb. The interaction between ROS, NO, and H2S appears to be involved in the response to Sb.

    Beschreibung: Non-coding RNAs (ncRNAs) are emerging therapeutic tools but there are barriers to their translation to clinical practice. Key issues concern the specificity of the targets, the delivery of the molecules, and their stability, while avoiding “on-target” and “off-target” side effects. In this “ncRNA in therapeutics” issue, we collect several studies of the differential expression of ncRNAs in cardiovascular diseases, bone metabolism-related disorders, neurology, and oncology, and their potential to be used as biomarkers or therapeutic targets. Moreover, we review recent advances in the use of antisense ncRNAs in targeted therapies with a particular emphasis on their basic biological mechanisms, their translational potential, and future trends.

    Beschreibung: Coffee is a beverage that is very popular all over the world. Its pro-health effect has been demonstrated in many publications. This drink can counteract the effects of oxidative stress thanks to its antioxidant properties. The aim of this study was to collect data on the content of microelements with antioxidant activity (manganese, zinc, copper, iron) in coffee infusions, taking into account various factors. The study considered publications from the years 2000–2020 found in Google Scholar and PubMed databases. It was noted that coffee can provide up to 13.7% of manganese requirements per serving, up to 4.0% and 3.1% of zinc requirements for women and men, up to 2.7% and 2.1% of copper requirements for women and men, and up to 0.4% and 0.6% of iron requirements for women and men. Coffee infusions can also be a source of fluoride (up to 2.5%), chromium (up to 0.4% of daily intake for women and 0.2% for men), and cobalt (up to 0.1%). There are no data in the literature regarding the content of selenium in coffee infusions. The origin of coffee beans and the type of water used (especially regarding fluoride) may have an impact on the content of minerals in infusions. The brewing method does not seem to play an important role. As it is a very popular beverage, coffee can additionally enrich the diet with such micronutrients as manganese, zinc, and copper. This seems beneficial due to their antioxidant properties, however the bioavailability of these elements of coffee should be taken into account. It seems necessary to carry out more research in this area.

    Beschreibung: We are reporting on the impact of air annealing temperatures on the physicochemical properties of electrochemically synthesized cadmium selenium telluride (CdSe0.6Te0.4) samples for their application in a photoelectrochemical (PEC) solar cell. The CdSe0.6Te0.4 samples were characterized with several sophisticated techniques to understand their characteristic properties. The XRD results presented the pure phase formation of the ternary CdSe0.6Te0.4 nanocompound with a hexagonal crystal structure, indicating that the annealing temperature influences the XRD peak intensity. The XPS study confirmed the existence of Cd, Se, and Te elements, indicating the formation of ternary CdSe0.6Te0.4 compounds. The FE-SEM results showed that the morphological engineering of the CdSe0.6Te0.4 samples can be achieved simply by changing the annealing temperatures from 300 to 400 °C with intervals of 50 °C. The efficiencies (ƞ) of the CdSe0.6Te0.4 photoelectrodes were found to be 2.0% for the non-annealed and 3.1, 3.6, and 2.5% for the annealed at 300, 350, and 400 °C, respectively. Most interestingly, the PEC cell analysis indicated that the annealing temperatures played an important role in boosting the performance of the photoelectrochemical properties of the solar cells.

    Beschreibung: Several investigations on senescence and its causative role in aging have underscored the importance of developing senotherapeutics, a field focused on killing senescent cells and/or preventing their accumulation within tissues. Using polyphenols in counteracting senescence may facilitate the development of senotherapeutics given their presence in the human diet, their confirmed tolerability and absence of severe side effects, and their role in preventing senescence and inducing the death of senescent cells. Against that background, we evaluated the effect of piceatannol, a natural polyphenol, on the senescence of mesenchymal stromal cells (MSCs), which play a key role in the body’s homeostasis. Among our results, piceatannol reduced the number of senescent cells both after genotoxic stress that induced acute senescence and in senescent replicative cultures. Such senotherapeutics activity, moreover, promoted the recovery of cell proliferation and the stemness properties of MSCs. Altogether, our findings demonstrate piceatannol’s effectiveness in counteracting senescence by targeting its associated pathways and detecting and affecting P53-dependent and P53-independent senescence. Our study thus suggests that, given piceatannol’s various mechanisms to accomplish its pleiotropic activities, it may be able to counteract any senescent phenotypes.

    Beschreibung: The nature of genome organization into two basic structural compartments is as yet undiscovered. However, it has been indicated to be a mechanism of gene expression regulation. Using the classification approach, we ranked genomic marks that hint at compartmentalization. We considered a broad range of marks, including GC content, histone modifications, DNA binding proteins, open chromatin, transcription and genome regulatory segmentation in GM12878 cells. Genomic marks were defined over CTCF or RNAPII loops, which are basic elements of genome 3D structure, and over 100 kb genomic windows. Experiments were carried out to empirically assess the whole set of features, as well as the individual features in classification of loops/windows, into compartment A or B. Using Monte Carlo Feature Selection and Analysis of Variance, we constructed a ranking of feature importance for classification. The best simple indicator of compartmentalization is DNase-seq open chromatin measurement for CTCF loops, H3K4me1 for RNAPII loops and H3K79me2 for genomic windows. Among DNA binding proteins, this is RUNX3 transcription factor for loops and RNAPII for genomic windows. Chromatin state prediction methods that indicate active elements like promoters, enhancers or heterochromatin enhance the prediction of loop segregation into compartments. However, H3K9me3, H4K20me1, H3K27me3 histone modifications and GC content poorly indicate compartments.

    Beschreibung: New technologies have allowed traditional map production criteria to be modified or even subverted. Starting from the communication sciences—journalism in particular—and digital humanities via the history of communication, we show how to use interactive digital maps for the production and publication of knowledge through and/or for participation. Firstly, we establish the theoretical-conceptual framework necessary to base the practices, dividing the elements into three areas: interactive maps and knowledge production (decentralization, pluralization, reticularization, and humanization), maps as instruments to promote political and social participation (egalitarianism, horizontality, and criticism), and maps as instruments for the visualization of data that favors the user experience (interactivity, multimediality, reticularity of reading, and participation). Next, we present two cases that we developed to put into practice the theoretical concepts that we established: the Mapa Infoparticipa (Infoparticipa Map), which shows the results of the evaluation of the transparency of public administrations, and the Ciutadania Plural (Plural Citizenship) web platform for the production of social knowledge about the past and the present. This theoretical and practical model shows the possibilities of interactive maps as tools to promote political participation and as instruments for the construction of social knowledge in a collaborative, participatory, networked way.

    Beschreibung: Aquaculture has grown rapidly in the field of food industry in recent years; however, it brought many environmental problems, such as water pollution and reclamations of lakes and coastal wetland areas. Thus, the evaluation and management of aquaculture industry are needed, in which accurate aquaculture mapping is an essential prerequisite. Due to the difference between inland and marine aquaculture areas and the difficulty in processing large amounts of remote sensing images, the accurate mapping of different aquaculture types is still challenging. In this study, a novel approach based on multi-source spectral and texture features was proposed to map simultaneously inland and marine aquaculture areas. Time series optical Sentinel-2 images were first employed to derive spectral indices for obtaining texture features. The backscattering and texture features derived from the synthetic aperture radar (SAR) images of Sentinel-1A were then used to distinguish aquaculture areas from other geographical entities. Finally, a supervised Random Forest classifier was applied for large scale aquaculture area mapping. To address the low efficiency in processing large amounts of remote sensing images, the proposed approach was implemented on the Google Earth Engine (GEE) platform. A case study in the Pearl River Basin (Guangdong Province) of China showed that the proposed approach obtained aquaculture map with an overall accuracy of 89.5%, and the implementation of proposed approach on GEE platform greatly improved the efficiency for large scale aquaculture area mapping. The derived aquaculture map may support decision-making services for the sustainable development of aquaculture areas and ecological protection in the study area, and the proposed approach holds great potential for mapping aquacultures on both national and global scales.

    Beschreibung: In China, peanut sprouts are popular among consumers as functional vegetables. This study reports the change in total phenolic content (TPC), total flavonoid content (TFC), monomeric anthocyanin content (MAC), vitamin C, trans-resveratrol content, antioxidant capacities, and phenolic profile of three different varieties of peanut during 8 days of germination. The TPC, TFC, and antioxidant capacity of peanut samples were reduced and then increased with an increase in germination time. TFC values were highly correlated with 2,2-diphenyl-1-picrylhydrazyl (DPPH) and ferric reducing antioxidant power (FRAP) values. MAC values of peanuts were first increased and then decreased during 8 days of germination. The TFC, DPPH, and FRAP values of germinated peanuts were lower compared to the non-germinated peanut. Germination of peanut samples enhanced the total phenolic acids and trans-resveratrol content, but the vitamin C content of peanut sprouts was lower than ungerminated peanuts.

    Beschreibung: Plicosepalus acacia (Fam. Loranthaceae) has been reported to possess hypoglycemic, antioxidant, antimicrobial, and anti-inflammatory effects. Liquid chromatography combined with tandem mass spectrometry (LC-MS/MS) analysis revealed the presence of a high content of polyphenolic compounds that are attributed to the therapeutic effects of the crude extract. In addition, methyl gallate and quercetin were detected as major phytomedicinal agents at concentrations of 1.7% and 0.062 g%, respectively, using high-performance thin layer chromatography (HPTLC). The present study investigated the effect of the P. acacia extract and its isolated compounds, methyl gallate and quercetin, on hind limb ischemia induced in type 1 diabetic rats. Histopathological examination revealed that treatment with P. acacia extract, methyl gallate, and quercetin decreased degenerative changes and inflammation in the ischemic muscle. Further biochemical assessment of the hind limb tissue showed decreased oxidative stress, increased levels of nitric oxide and endothelial nitric oxide synthase (eNOS), and enhancement of the levels of heme oxygenase-1 (HO-1) and vascular endothelial growth factor (VEGF) in the groups treated with methyl gallate and quercetin. Expression levels of hypoxia inducible factor-1 alpha (HIF-1α), VEGF, fibroblast growth factor-2 (FGF-2), and miR-146a were upregulated in the muscle tissue of methyl gallate- and quercetin-treated groups along with downregulation of nuclear factor kappa B (NF-κB). In conclusion, P. acacia extract and its isolated compounds, methyl gallate and quercetin, mediated therapeutic angiogenesis in diabetic hind limb ischemia.

    Beschreibung: Background: Emerging data suggest that more aggressive beta-lactam PK/PD targets could minimize the occurrence of microbiological failure and/or resistance development. This study aims to assess whether a PK/PD target threshold of continuous infusion (CI) beta-lactams may be useful in preventing microbiological failure and/or resistance development in critically ill patients affected by documented Gram-negative infections. Methods: Patients admitted to intensive care units from December 2020 to July 2021 receiving continuous infusion beta-lactams for documented Gram-negative infections and having at least one therapeutic drug monitoring in the first 72 h of treatment were included. A receiver operating characteristic (ROC) curve analysis was performed using the ratio between steady-state concentration and minimum inhibitory concentration (Css/MIC) ratio as the test variable and occurrence of microbiological failure as the state variable. Area under the curve (AUC) and 95% confidence interval (CI) were calculated. Independent risk factors for the occurrence of microbiological failure were investigated using logistic regression. Results: Overall, 116 patients were included. Microbiological failure occurred in 26 cases (22.4%). A Css/MIC ratio ≤ 5 was identified as PK/PD target cut-off with sensitivity of 80.8% (CI 60.6–93.4%) and specificity of 90.5% (CI 74.2–94.4%), and with an AUC of 0.868 (95%CI 0.793–0.924; p 〈 0.001). At multivariate regression, independent predictors of microbiological failure were Css/MIC ratio ≤ 5 (odds ratio [OR] 34.54; 95%CI 7.45–160.11; p 〈 0.001) and Pseudomonas aeruginosa infection (OR 4.79; 95%CI 1.11–20.79; p = 0.036). Conclusions: Early targeting of CI beta-lactams at Css/MIC ratio 〉 5 during the treatment of documented Gram-negative infections may be helpful in preventing microbiological failure and/or resistance development in critically ill patients.

    Beschreibung: The reniform nematode, Rotylenchulus reniformis (Linford and Oliveira), remains a common, widespread nematode pest of cotton across the southern United States. Trials were conducted during 2017 at three non-irrigated locations: one location in Hamilton, MS, and two locations in Tchula, MS, in field settings with a history of cotton production and documented economically-damaging reniform nematode populations. Trials were designed to evaluate the response of two cotton cultivars to in-furrow nematicides consisting of aldicarb, 1,3-dichloropropene, and a non-treated control applied for nematode suppression. No significant interactions between cotton cultivar and nematicide were observed. However, treatment with 1,3-dichloropropene produced greater plant biomass, and plant height compared to aldicarb-treated cotton and the nontreated. Nematode densities were suppressed with the use of 1,3-dichloropropene compared to aldicarb and the non-treated control. The use of 1,3-dichloropropene resulted in positive early-season plant growth responses; however, these responses did not translate into greater yield.

    Beschreibung: Monitoring gait patterns in daily life will provide a lot of biological information related to human health. At present, common gait pressure analysis systems, such as pressure platforms and in-shoe systems, adopt rigid sensors and are wired and uncomfortable. In this paper, a biomimetic porous graphene–SBR (styrene-butadiene rubber) pressure sensor (PGSPS) with high flexibility, sensitivity (1.05 kPa−1), and a wide measuring range (0–150 kPa) is designed and integrated into an insole system to collect, process, transmit, and display plantar pressure data for gait analysis in real-time via a smartphone. The system consists of 16 PGSPSs that were used to analyze different gait signals, including walking, running, and jumping, to verify its daily application range. After comparing the test results with a high-precision digital multimeter, the system is proven to be more portable and suitable for daily use, and the accuracy of the waveform meets the judgment requirements. The system can play an important role in monitoring the safety of the elderly, which is very helpful in today’s society with an increasingly aging population. Furthermore, an intelligent gait diagnosis algorithm can be added to realize a smart gait monitoring system.

    Beschreibung: Clinical outcomes of COVID-19 patients are worsened by the presence of co-morbidities, especially cancer leading to elevated mortality rates. SARS-CoV-2 infection is known to alter immune system homeostasis. Whether cancer patients developing COVID-19 present alterations of immune functions which might contribute to worse outcomes have so far been poorly investigated. We conducted a multi-omic analysis of immunological parameters in peripheral blood mononuclear cells (PBMCs) of COVID-19 patients with and without cancer. Healthy donors and SARS-CoV-2-negative cancer patients were also included as controls. At the infection peak, cytokine multiplex analysis of blood samples, cytometry by time of flight (CyTOF) cell population analyses, and Nanostring gene expression using Pancancer array on PBMCs were performed. We found that eight pro-inflammatory factors (IL-6, IL-8, IL-13, IL-1ra, MIP-1a, IP-10) out of 27 analyzed serum cytokines were modulated in COVID-19 patients irrespective of cancer status. Diverse subpopulations of T lymphocytes such as CD8+T, CD4+T central memory, Mucosal-associated invariant T (MAIT), natural killer (NK), and γδ T cells were reduced, while B plasmablasts were expanded in COVID-19 cancer patients. Our findings illustrate a repertoire of aberrant alterations of gene expression in circulating immune cells of COVID-19 cancer patients. A 19-gene expression signature of PBMCs is able to discriminate COVID-19 patients with and without solid cancers. Gene set enrichment analysis highlights an increased gene expression linked to Interferon α, γ, α/β response and signaling which paired with aberrant cell cycle regulation in cancer patients. Ten out of the 19 genes, validated in a real-world consecutive cohort, were specific of COVID-19 cancer patients independently from different cancer types and stages of the diseases, and useful to stratify patients in a COVID-19 disease severity-manner. We also unveil a transcriptional network involving gene regulators of both inflammation response and proliferation in PBMCs of COVID-19 cancer patients.

    Beschreibung: The aim of this study was to compare the surface roughness and gloss of polymer-infiltrated ceramics after simulated in vitro toothbrushing in different storage mediums. Four polymer- infiltrated ceramics were evaluated, Lava ultimate (LU), Vita enamic (EN), Shofu (SH), and Crystal ultra (CU). The control group was a feldspathic ceramic, Vita Mark II (VM). One hundred and twenty specimens (12 × 14 × 2.5 mm) were prepared using a precision saw. For each material (n = 24), the specimens were allocated into two groups, polished and stained. The specimens of each group were stored (for 7 days) in either citric acid (0.2N) or distilled water. Data for surface gloss (ΔE*SCE-SCI) and roughness (Ra) were evaluated before (baseline) and after simulated toothbrushing. For toothbrushing simulation, a toothpaste slurry containing a toothpaste of 100 relative dentin abrasion (RDA) and 0.3 ml distilled water was used for 3650 cycles (7300 strokes) for each specimen. Data were analyzed using t-test and ANOVA. A p-value of ≤ to 0.05 was considered significant. The highest mean value of surface gloss was identified in CU (stained—water) (4.3 (0.47)) (ΔE*) and EN (stained—acid) (4.3 (1.00)) (ΔE*) specimens, whereas the lowest mean value was shown by SH (stained—acid) (2.04 (0.42)) (ΔE*) samples. The highest mean value of surface roughness was observed in SH (0.40 (0.99)) Ra (stained—acid) whereas the lowest in VM (0.13 (0.039)) Ra (polished— water). A significant difference (p 〈 0.05) was observed in surface roughness and gloss between the materials with simulated toothbrushing, except in VM and LU, respectively. Therefore, it can be concluded that simulated toothbrushing impacts on surface roughness and gloss, irrespective of the storage medium.

    Beschreibung: In the past few decades, ZrN thin films have been identified as wear resistant coatings for tribological applications. The mechanical and tribological properties of ZrN thin layers depend on internal stress induced by the adopted deposition techniques and deposition parameters such as pressure, temperature, and growth rate. In sputtering deposition processes, the selected target voltage waveform and the plasma characteristics also play a crucial influence on physical properties of produced coatings. In present work, ZrN thin films, obtained setting different values of duty cycle in a reactive bipolar pulsed dual magnetron sputtering plant, were investigated to evaluate their residual stress through the substrate curvature method. A considerable progressive increase of residual stress values was measured at decreasing duty cycle, attesting the significant role of voltage waveform in stress development. An evident correlation was also highlighted between the values of the duty cycle and those of wear factor. The performed analysis attested an advantageous effect of internal stress, having the samples with high compressive stress, higher wear resistance. A downward trend for wear rate with the increase of internal residual stress was observed. The choice of suitable values of duty cycle allowed to produce ceramic coatings with improved tribological performance.

    Beschreibung: Individual identification plays an important part in disease prevention and control, traceability of meat products, and improvement of agricultural false insurance claims. Automatic and accurate detection of cattle face is prior to individual identification and facial expression recognition based on image analysis technology. This paper evaluated the possibility of the cutting-edge object detection algorithm, RetinaNet, performing multi-view cattle face detection in housing farms with fluctuating illumination, overlapping, and occlusion. Seven different pretrained CNN models (ResNet 50, ResNet 101, ResNet 152, VGG 16, VGG 19, Densenet 121 and Densenet 169) were fine-tuned by transfer learning and re-trained on the dataset in the paper. Experimental results showed that RetinaNet incorporating the ResNet 50 was superior in accuracy and speed through performance evaluation, which yielded an average precision score of 99.8% and an average processing time of 0.0438 s per image. Compared with the typical competing algorithms, the proposed method was preferable for cattle face detection, especially in particularly challenging scenarios. This research work demonstrated the potential of artificial intelligence towards the incorporation of computer vision systems for individual identification and other animal welfare improvements.

    Beschreibung: Wine batch distillation in a copper pot still heated directly by combustion is unusual. It is a niche sector. Few studies are available on the subject and even fewer have studied it from a thermal point of view. In a context of ecology awareness and regularly increasing gas taxes, finding less energy consuming and more performing solutions has become necessary. Two methods already exist to recover energy for such pot stills, however improvements could be made. The present study focuses on the heating zone of an experimental pot still. First, a thermal diagnosis based on a nodal model is conducted in transient regime. The major sites of energy expenditure and waste are thus identified and quantified. Results highlight significant losses both in combustion smoke and by conduction to the environment resulting from a limited thermal performance of the system. Proposals are then made to enhance it. For instance, the preheating could be optimized and more thermal waste could be recovered.

    Beschreibung: We explored gender differences in sexual behavior, and their relevance to mental health among high school students in South Korea. This study was based on data from the 14th Korea Youth Risk Behavior Web-Based Survey (2018). The subjects were 2460 high school students who reported sexual experiences. Student′s t-test, ANOVA, and Rao-Scott chi-squared test were performed to identify the significance of the differences. Boys had earlier sexual debuts (Rao-Scott χ2 = 53.55, p 〈 0.001), a lower frequency of using contraceptives (Rao-Scott χ2 = 26.57, p 〈 0.001), and lower rates of sex education in school (Rao-Scott χ2 = 11.20, p = 0.004). With respect to mental health factors, there was a difference in suicidality according to sexual risk behaviors, with a stronger association found in boys. In girls, there was an association between pregnancy experiences and suicidal ideation (Rao-Scott χ2 = 9.90, p = 0.003), plans (Rao-Scott χ2 = 17.25, p 〈 0.001), and attempts (Rao-Scott χ2 = 23.11, p 〈 0.001). Our findings suggest differences by gender and age group in the association between sexual behavior and mental health. It is necessary to devise a sex education strategy for adolescents considering gender and early versus late adolescent development period.

    Beschreibung: Indonesia has 14 million ha of degraded and marginal land, which provides very few benefits for human wellbeing or biodiversity. This degraded land may require restoration. The leguminous tree Pongamia pinnata syn. Milettia pinnata (pongamia) has potential for producing biofuel while simultaneously restoring degraded land. However, there is limited information on this potential for consideration. This paper aims to address the scientific knowledge gap on pongamia by exploring its potential as a biofuel and for restoring degraded land in Indonesia. We applied a literature review to collect relevant information of pongamia, which we analyzed through narrative qualitative and narrative comparative methods with careful compilation and scientific interpretation of retrieved information. The review revealed that pongamia occurs naturally across Indonesia, in Sumatra, Java, Bali, Nusa Tenggara and Maluku. It can grow to a height of 15–20 m and thrive in a range of harsh environmental conditions. Its seeds can generate up to 40% crude pongamia oil by weight. It is a nitrogen-fixing tree that can help restore degraded land and improve soil properties. Pongamia also provides wood, fodder, medicine, fertilizer and biogas. As a multipurpose species, pongamia holds great potential for combating Indonesia’s energy demand and restoring much of the degraded land. However, the potential competition for land and for raw material with other biomass uses must be carefully managed.

    Beschreibung: Peptides are increasingly being developed for use as therapeutics to treat many ailments, including cancer. Therapeutic peptides have the advantages of target specificity and low toxicity. The anticancer effects of a peptide can be the direct result of the peptide binding its intended target, or the peptide may be conjugated to a chemotherapy drug or radionuclide and used to target the agent to cancer cells. Peptides can be targeted to proteins on the cell surface, where the peptide–protein interaction can initiate internalization of the complex, or the peptide can be designed to directly cross the cell membrane. Peptides can induce cell death by numerous mechanisms including membrane disruption and subsequent necrosis, apoptosis, tumor angiogenesis inhibition, immune regulation, disruption of cell signaling pathways, cell cycle regulation, DNA repair pathways, or cell death pathways. Although using peptides as therapeutics has many advantages, peptides have the disadvantage of being easily degraded by proteases once administered and, depending on the mode of administration, often have difficulty being adsorbed into the blood stream. In this review, we discuss strategies recently developed to overcome these obstacles of peptide delivery and bioavailability. In addition, we present many examples of peptides developed to fight cancer.

    Beschreibung: Cholera remains a heavy burden worldwide, especially in Sub-Saharan African countries, which account for the majority of the reported cases on the continent. In this study, a 27-year retrospective analysis of cholera epidemics in Guinea-Bissau was performed in order to highlight major stressors fueling the trigger and dissemination of the disease. Although the role of environmental factors did not always have the same degree of importance for the onset of epidemics, a cholera seasonal pattern was clearly perceived, with most of the reported cases occurring during the wet season. The generated theoretical hypothesis indicated rainfall above climatological average, associated with a lack of WASH (water, sanitation and hygiene) infrastructure, and the occurrence of concomitant epidemics in neighboring countries as the key indicators for optimal conditions for cholera to thrive in Guinea-Bissau. Warmer air temperature, the increase in sea surface temperature, and the decrease in salinity in the coastal areas may also contribute to the emergence and/or aggravation of cholera events. Prediction of the conditions favorable for cholera growth and identification of risk pathways will allow the timely allocation of resources, and support the development of alert tools and mitigation strategies.

    Beschreibung: In the last decades, silk fibroin and wool keratin have been considered functional materials for biomedical applications. In this study, fabrics containing silk fibers from Bombyx mori and Tussah silk fibers from Antheraea pernyi, as well as wool keratin fabrics, were grafted with phosmer CL and phosmer M (commercial names, i.e., methacrylate monomers containing phosphate groups in the molecular side chain) with different weight gains. Both phosmers were recently proposed as flame retarding agents, and their chemical composition suggested a possible application in bone tissue engineering. IR and Raman spectroscopy were used to disclose the possible structural changes induced by grafting and identify the most reactive amino acids towards the phosmers. The same techniques were used to investigate the nucleation of a calcium phosphate phase on the surface of the samples (i.e., bioactivity) after ageing in simulated body fluid (SBF). The phosmers were found to polymerize onto the biopolymers efficiently, and tyrosine and serine underwent phosphorylation (monitored through the strengthening of the Raman band at 1600 cm−1 and the weakening of the Raman band at 1400 cm−1, respectively). In grafted wool keratin, cysteic acid and other oxidation products of disulphide bridges were detected together with sulphated residues. Only slight conformational changes were observed upon grafting, generally towards an enrichment in ordered domains, suggesting that the amorphous regions were more prone to react (and, sometimes, degrade). All samples were shown to be bioactive, with a weight gain of up to 8%. The most bioactive samples contained the highest phosmers amounts, i.e., the highest amounts of phosphate nucleating sites. The sulphate/sulphonate groups present in grafted wool samples appeared to increase bioactivity, as shown by the five-fold increase of the IR phosphate band at 1040 cm−1.

    Beschreibung: Difenoconazole is one of the most commonly used fungicides to prevent and treat plant diseases caused by certain fungi. Due to increasing usage, more difenoconazole has been released into the environment and caused environment pollution. However, the potential impact of difenoconazole on plant growth and development and its involved mechanism are unclear. In this study, we discovered that difenoconazole exposure significantly inhibited plant growth, evidenced by the decrease in root dry weight, total root length, and surface area by 20–70%, 43–73%, and 26–66%, respectively, under different regimes of treatment concentrations and periods. Difenoconazole exposure also significantly inhibited shoot growth and development by decreasing 33–61% of the shoot dry weight and 50–65% of the leaf area. Difenoconazole exposure induced plant leaf cells to generate more ROS (O2•− and H2O2) and MDA, which resulted in a decreased chlorophyll content and then inhibited leaf photosynthesis. Difenoconazole exposure also induced the activities of superoxide dismutase (SOD), catalase (CAT), guaiacol peroxidase (G-POD), and ascorbate peroxidase (APX) in the roots and leaves of the wheat seedlings. SOD and APX activities were higher and more stable in the roots than those in the leaves. Based on our study, plant roots exhibited a more pronounced superoxide radical scavenging ability than plant leaves. In summary, difenoconazole exposure caused oxidative stress, reduced chlorophyll biosynthesis and functions, and then inhibited wheat plant growth and development.

    Beschreibung: If the brain structure is assessed at neonatal intensive care units, covert clinical events related with subtle brain injury might be identified. The reduced scattering coefficient of near-infrared light (μS’) obtained using time-resolved near-infrared spectroscopy from the forehead of infants is associated with gestational age, body weight and Apgar scores, presumably reflecting subtle changes of the brain related to foetal growth and birth transition. One hundred twenty-eight preterm and term infants were studied to test whether μS’ obtained from the head at term-equivalent age is associated with foetal growth, birth transition and nutritional status after birth, which are key independent variables of developmental outcomes.     Beschreibung: Plastic waste management has become a global issue. Polyethylene (PE) is the most abundant synthetic plastic worldwide, and one of the most resistant to biodegradation. Indeed, few bacteria can degrade polyethylene. In this paper, the transcriptomic analysis unveiled for the first time Rhodococcus opacus R7 complex genetic system based on diverse oxidoreductases for polyethylene biodegradation. The RNA-seq allowed uncovering genes putatively involved in the first step of oxidation. In-depth investigations through preliminary bioinformatic analyses and enzymatic assays on the supernatant of R7 grown in the presence of PE confirmed the activation of genes encoding laccase-like enzymes. Moreover, the transcriptomic data allowed identifying candidate genes for the further steps of short aliphatic chain oxidation including alkB gene encoding an alkane monooxygenase, cyp450 gene encoding cytochrome P450 hydroxylase, and genes encoding membrane transporters. The PE biodegradative system was also validated by FTIR analysis on R7 cells grown on polyethylene.

    Beschreibung: C-phycocyanin (CPC), a blue pigment protein, is an indispensable component of giant phycobilisomes, which are light-harvesting antenna complexes in cyanobacteria that transfer energy efficiently to photosystems I and II. X-ray crystallographic and electron microscopy (EM) analyses have revealed the structure of CPC to be a closed toroidal hexamer by assembling two trimers. In this study, the structural characterization of non-conventional octameric CPC is reported for the first time. Analyses of the crystal and cryogenic EM structures of the native CPC from filamentous thermophilic cyanobacterium Thermoleptolyngbya sp. O–77 unexpectedly illustrated the coexistence of conventional hexamer and novel octamer. In addition, an unusual dimeric state, observed via analytical ultracentrifugation, was postulated to be a key intermediate structure in the assemble of the previously unobserved octamer. These observations provide new insights into the assembly processes of CPCs and the mechanism of energy transfer in the light-harvesting complexes.

    Beschreibung: One of the main limitations in state-of-the art solid-state quantum processors is qubit decoherence and relaxation due to noise from adsorbates on surfaces, impurities at interfaces, and material defects. For the field to advance towards full fault-tolerant quantum computing, a better understanding of these microscopic noise sources is therefore needed. Here, we use an ultra-high vacuum package to study the impact of vacuum loading, UV-light exposure, and ion irradiation treatments on relaxation and coherence times, as well as slow parameter fluctuations of flux tunable superconducting transmon qubits. The treatments studied do not significantly impact the relaxation rate Γ1 and the echo decay rate $${{{Gamma }}}_{2,{{{ m{SS}}}}}^{{{{ m{e}}}}}$$ Γ 2 , SS e at the sweet spot, except for Ne ion bombardment which reduces Γ1. In contrast, flux noise parameters are improved by removing magnetic adsorbates from the chip surfaces with UV-light and NH3 treatments. Additionally, we demonstrate that SF6 ion bombardment can be used to adjust qubit frequencies in situ and post-fabrication without affecting qubit relaxation and coherence times at the sweet spot.

    Beschreibung: α-Synuclein (α-Syn) can form different fibril strains with distinct polymorphs and neuropathologies, which is associated with the clinicopathological variability in synucleinopathies. How different α-syn fibril strains are produced and selected under disease conditions remains poorly understood. In this study, we show that the hereditary mutation G51D induces α-syn to form a distinct fibril strain in vitro. The cryogenic electron microscopy (cryo-EM) structure of the G51D fibril strain was determined at 2.96 Å resolution. The G51D fibril displays a relatively small and extended serpentine fold distinct from other α-syn fibril structures. Moreover, we show by cryo-EM that wild-type (WT) α-syn can assembly into the G51D fibril strain via cross-seeding with G51D fibrils. Our study reveals a distinct structure of G51D fibril strain triggered by G51D mutation but feasibly adopted by both WT and G51D α-syn, which suggests the cross-seeding and strain selection of WT and mutant α-syn in familial Parkinson’s disease (fPD).

    Beschreibung: A batch of pine timber sawn from butt, middle and top logs was strength graded with the visual method (classification into grading classes KW—best quality, KS—medium quality, KG—inferior quality and Reject) and with the machine strength grading method—performed with the use of a mobile timber grader (classification into C strength classes). We compared the efficiency of grading classes and strength classes, depending on the type of log from which the material was obtained (butt, middle, top). Next, a strength grading machine was used to measure the modulus of elasticity in bending (MOE) and static bending strength (MOR). The ANOVA confirmed the influence of both the log type (butt, middle, top), the C strength class, and the visual strength grading class on the values of density (DEN) and MOR. Timber density and MOR decreased from the butt log section to the top log section. The ANOVA confirmed the influence of log type on MOE values, but only to a limited extent.

    Beschreibung: The manipulation of light intensity by shade nets can lead to exchanges in the phytochemical quality and antioxidants ofsome herbs. This study aimed to determine whether shading by pearl nets (50% shade index) could improve the bioactive compounds in several medicinal herbs such as thyme (Thymus vulgaris L.), marjoram (Origanum majorana L.), oregano (Origanum vulgare L.), lemon balm (Melissa officinalis L.), and peppermint (Mentha piperita L.), and their correspondingherbal extracts during a four-week maceration process in ethanol. Oregano and thyme provided the highest yield of total extractive substances (TES) from both shaded and non-shaded plants. Among all studied herbs, the highest level of antioxidants, expressed as total phenolic content (TPC) was found in extracts from shaded plants of lemon balm. Herbal extracts produced from non-shaded thyme and marjoram had higher flavonoid contents compared to herbal extracts from shaded plants. Accumulation of the investigated secondary metabolites depends more on the specificity of a plant species rather than light intensity. This study was an initial step in the production of medicinal plants with an increased quantity of antioxidant and other bioactive compounds during the maceration process in extracts, aimed to be used as natural alcoholic productwith added value.

    Beschreibung: A novel, environmentally friendly, fast, and flexible polishing process for Nitinol parts is presented in this study. Nitinol samples with both superelastic and shape memory properties at room temperature were investigated. The chemical contamination and surface roughness of superelastic Nitinol plates were examined before and after plasma electrolytic polishing. The shift in phase transformation temperature and tensile strength before and after the polishing process were analysed using Nitinol wire with shape memory properties. The obtained experimental results were compared to the data obtained on reference samples examined in the as-received condition. It was found that plasma electrolytic polishing, when the right process parameters are applied, is capable of delivering Nitinol parts with extremely high surface quality. Moreover, it was experimentally proven that plasma electrolytic polishing does not have a negative impact on functionality or mechanical properties of polished parts.

    Beschreibung: Background: Patients with primary antibody deficiencies are at risk in the current COVID-19 pandemic due to their impaired response to infection and vaccination. Specifically, patients with common variable immunodeficiency (CVID) generated poor spike-specific antibody and T cell responses after immunization. Methods: Thirty-four CVID convalescent patients after SARS-CoV-2 infection, 38 CVID patients immunized with two doses of the BNT162b2 vaccine, and 20 SARS-CoV-2 CVID convalescents later and immunized with BNT162b2 were analyzed for the anti-spike IgG production and the generation of spike-specific memory B cells and T cells. Results: Spike-specific IgG was induced more frequently after infection than after vaccination (82% vs. 34%). The antibody response was boosted in convalescents by vaccination. Although immunized patients generated atypical memory B cells possibly by extra-follicular or incomplete germinal center reactions, convalescents responded to infection by generating spike-specific memory B cells that were improved by the subsequent immunization. Poor spike-specific T cell responses were measured independently from the immunological challenge. Conclusions: SARS-CoV-2 infection primed a more efficient classical memory B cell response, whereas the BNT162b2 vaccine induced non-canonical B cell responses in CVID. Natural infection responses were boosted by subsequent immunization, suggesting the possibility to further stimulate the immune response by additional vaccine doses in CVID.

    Beschreibung: Deviations from Brownian motion leading to anomalous diffusion are found in transport dynamics from quantum physics to life sciences. The characterization of anomalous diffusion from the measurement of an individual trajectory is a challenging task, which traditionally relies on calculating the trajectory mean squared displacement. However, this approach breaks down for cases of practical interest, e.g., short or noisy trajectories, heterogeneous behaviour, or non-ergodic processes. Recently, several new approaches have been proposed, mostly building on the ongoing machine-learning revolution. To perform an objective comparison of methods, we gathered the community and organized an open competition, the Anomalous Diffusion challenge (AnDi). Participating teams applied their algorithms to a commonly-defined dataset including diverse conditions. Although no single method performed best across all scenarios, machine-learning-based approaches achieved superior performance for all tasks. The discussion of the challenge results provides practical advice for users and a benchmark for developers.

    Beschreibung: RUNX2 encodes the master bone transcription factor driving skeletal development in vertebrates, and playing a specific role in craniofacial and skull morphogenesis. The anatomically modern human (AMH) features sequence changes in the RUNX2 locus compared with archaic hominins’ species. We aimed to understand how these changes may have contributed to human skull globularization occurred in recent evolution. We compared in silico AMH and archaic hominins’ genomes, and used mesenchymal stromal cells isolated from skull sutures of craniosynostosis patients for in vitro functional assays. We detected 459 and 470 nucleotide changes in noncoding regions of the AMH RUNX2 locus, compared with the Neandertal and Denisovan genomes, respectively. Three nucleotide changes in the proximal promoter were predicted to alter the binding of the zinc finger protein Znf263 and long-distance interactions with other cis-regulatory regions. By surface plasmon resonance, we selected nucleotide substitutions in the 3’UTRs able to affect miRNA binding affinity. Specifically, miR-3150a-3p and miR-6785-5p expression inversely correlated with RUNX2 expression during in vitro osteogenic differentiation. The expression of two long non-coding RNAs, AL096865.1 and RUNX2-AS1, within the same locus, was modulated during in vitro osteogenic differentiation and correlated with the expression of specific RUNX2 isoforms. Our data suggest that RUNX2 may have undergone adaptive phenotypic evolution caused by epigenetic and post-transcriptional regulatory mechanisms, which may explain the delayed suture fusion leading to the present-day globular skull shape.

    Beschreibung: Biological degradation of Polyethylene terephthalate (PET) plastic and assimilation of the corresponding monomers ethylene glycol and terephthalate (TPA) into central metabolism offers an attractive route for bio-based molecular recycling and bioremediation applications. A key step is the cellular uptake of the non-permeable TPA into bacterial cells which has been shown to be dependent upon the presence of the key tphC gene. However, little is known from a biochemical and structural perspective about the encoded solute binding protein, TphC. Here, we report the biochemical and structural characterisation of TphC in both open and TPA-bound closed conformations. This analysis demonstrates the narrow ligand specificity of TphC towards aromatic para-substituted dicarboxylates, such as TPA and closely related analogues. Further phylogenetic and genomic context analysis of the tph genes reveals homologous operons as a genetic resource for future biotechnological and metabolic engineering efforts towards circular plastic bio-economy solutions.

    Beschreibung: Understanding the trajectory, duration, and determinants of antibody responses after SARS-CoV-2 infection can inform subsequent protection and risk of reinfection, however large-scale representative studies are limited. Here we estimated antibody response after SARS-CoV-2 infection in the general population using representative data from 7,256 United Kingdom COVID-19 infection survey participants who had positive swab SARS-CoV-2 PCR tests from 26-April-2020 to 14-June-2021. A latent class model classified 24% of participants as ‘non-responders’ not developing anti-spike antibodies, who were older, had higher SARS-CoV-2 cycle threshold values during infection (i.e. lower viral burden), and less frequently reported any symptoms. Among those who seroconverted, using Bayesian linear mixed models, the estimated anti-spike IgG peak level was 7.3-fold higher than the level previously associated with 50% protection against reinfection, with higher peak levels in older participants and those of non-white ethnicity. The estimated anti-spike IgG half-life was 184 days, being longer in females and those of white ethnicity. We estimated antibody levels associated with protection against reinfection likely last 1.5-2 years on average, with levels associated with protection from severe infection present for several years. These estimates could inform planning for vaccination booster strategies.

    Beschreibung: One of the challenges in the future of food production, amidst increasing population and decreasing resources, is developing a sustainable food production system. It is anticipated that robotics will play a significant role in maintaining the food production system, specifically in labor-intensive operations. Therefore, the main goal of this project is to develop a robotic fruit harvesting system, initially focused on the harvesting of apples. The robotic harvesting system is composed of a six-degrees-of-freedom (DOF) robotic manipulator, a two-fingered gripper, a color camera, a depth sensor, and a personal computer. This paper details the development and performance of a visual servo system that can be used for fruit harvesting. Initial test evaluations were conducted in an indoor laboratory using plastic fruit and artificial trees. Subsequently, the system was tested outdoors in a commercial fruit orchard. Evaluation parameters included fruit detection performance, response time of the visual servo, and physical time to harvest a fruit. Results of the evaluation showed that the developed visual servo system has the potential to guide the robot for fruit harvesting.

    Beschreibung: In this study, we develop physics-informed neural networks (PINNs) to solve an isothermal fixed-bed (IFB) model for catalytic CO2 methanation. The PINN includes a feed-forward artificial neural network (FF-ANN) and physics-informed constraints, such as governing equations, boundary conditions, and reaction kinetics. The most effective PINN structure consists of 5–7 hidden layers, 256 neurons per layer, and a hyperbolic tangent (tanh) activation function. The forward PINN model solves the plug-flow reactor model of the IFB, whereas the inverse PINN model reveals an unknown effectiveness factor involved in the reaction kinetics. The forward PINN shows excellent extrapolation performance with an accuracy of 88.1% when concentrations outside the training domain are predicted using only one-sixth of the entire domain. The inverse PINN model identifies an unknown effectiveness factor with an error of 0.3%, even for a small number of observation datasets (e.g., 20 sets). These results suggest that forward and inverse PINNs can be used in the solution and system identification of fixed-bed models with chemical reaction kinetics.

    Beschreibung: Klebsiella pneumoniae is an increasing threat to public health and represents one of the most concerning pathogens involved in life-threatening infections. The resistant and virulence determinants are coded by mobile genetic elements which can easily spread between bacteria populations and co-evolve with its genomic host. In this study, we present the full genomic sequences, insertion sites and phylogenetic analysis of 150 prophages found in 40 K. pneumoniae clinical isolates obtained from an outbreak in a Portuguese hospital. All strains harbored at least one prophage and we identified 104 intact prophages (69.3%). The prophage size ranges from 29.7 to 50.6 kbp, coding between 32 and 78 putative genes. The prophage GC content is 51.2%, lower than the average GC content of 57.1% in K. pneumoniae. Complete prophages were classified into three families in the order Caudolovirales: Myoviridae (59.6%), Siphoviridae (38.5%) and Podoviridae (1.9%). In addition, an alignment and phylogenetic analysis revealed nine distinct clusters. Evidence of recombination was detected within the genome of some prophages but, in most cases, proteins involved in viral structure, transcription, replication and regulation (lysogenic/lysis) were maintained. These results support the knowledge that prophages are diverse and widely disseminated in K. pneumoniae genomes, contributing to the evolution of this species and conferring additional phenotypes. Moreover, we identified K. pneumoniae prophages in a set of endolysin genes, which were found to code for proteins with lysozyme activity, cleaving the β-1,4 linkages between N-acetylmuramic acid and N-acetyl-D-glucosamine residues in the peptidoglycan network and thus representing genes with the potential for lysin phage therapy.

To date, in Slovenia, there were only a few outdated studies of dietary fibre intake; therefore, we explored the dietary fibre intake using food consumption data collected in the SI.Menu project. Following the EU Menu methodology, data were collected on representative samples of adolescents, adults, and elderlies using a general questionnaire, a food propensity questionnaire, and two 24 h recalls.     Beschreibung: Education for Sustainable Development (ESD) and Entrepreneurial Education (EE) are quite abstract and demanding concepts for teacher students. Yet, Key Sustainability Competences and Entrepreneurial Competences entail important qualities of future citizens and workers, and teacher students should become prepared to accommodate education for these competencies in their teaching practice. This paper explores teacher students’ process of sense-making of sustainable development and how becoming a teacher who practices ESD connects with entrepreneurship. EE serves as a good mirroring surface to ESD as they both have their roots in Transformative Learning (TL) but pursue transformation towards different goals. The case study follows the vocational teacher education (VTE) students’ sensemaking of Sustainable Development as a part of teacher’s work during one semester which included integrated Thematic Studies of Sustainable Development. The qualitative content analysis of students’ texts focused on signs of transformative learning and was guided by the dimensions of sustainable development and learning goals set for teacher’s sustainability competences in the VTE curriculum. The results indicate that transformative learning is possible. Furthermore, they address the importance of certain entrepreneurial capabilities in the actualization of change agency.

    Beschreibung: Floods are a known natural hazard in Germany, but the amount of precipitation and ensuing high death toll and damages after the events especially from 14 to 15 July 2021 came as a surprise. Almost immediately questions about failure in the early warning chains and the effectiveness of the German response emerged, also internationally. This article presents lessons to learn and argues against a blame culture. The findings are based on comparisons with findings from previous research projects carried out in the Rhein-Erft Kreis and the city of Cologne, as well as on discussions with operational relief forces after the 2021 events. The main disaster aspects of the 2021 flood are related to issuing and understanding warnings, a lack of information and data exchange, unfolding upon a situation of an ongoing pandemic and aggravated further by critical infrastructure failure. Increasing frequencies of flash floods and other extremes due to climate change are just one side of the transformation and challenge, Germany and neighbouring countries are facing. The vulnerability paradox also heavily contributes to it; German society became increasingly vulnerable to failure due to an increased dependency on its infrastructure and emergency system, and the ensuing expectations of the public for a perfect system.

    Beschreibung: This article aims to assess how fiscal and financial incentives and government support conditioned the profitability of Portuguese SMEs between 2010 and 2019. The high tax and financial burdens on SMEs have consequences for sustainability and business development. Thus, the study analyzes different incentives provided by the Portuguese government to ease this burden and improve business profitability. The study uses panel data with fixed effects using five different sources of information from five internal tax grant types, three different European Union program financial subventions, and three national budget-specific expenses. The results obtained suggest that tax incentives influence the profitability of SMEs; however, government incentives do not have any impact. The QREN (financial) incentives positively decide the ROA and negatively impact the ROE, contributing to sustainable performance. Portugal 2020 incentives have a weak effect on the first years, improving in the following years. However, the incentive related to R&D is not relevant. This work aims to contribute to decision making for managers, shareholders, and government entities, allowing them to choose those measures that could increase the company’s added value, and for governments, as a tool to select incentives that will most benefit SMEs’’ profitability. This work identifies the key incentives that impact companies’ profitability.

    Beschreibung: Amniotic membrane is an effective corneal reconstruction material in veterinary surgery. Cryopreserved amniotic membrane is widely used in practice. Properties of cryopreserved canine amniotic membranes are currently not well studied. This study aimed to compare three properties between canine amniotic membranes cryopreserved for 7 days and 30 days, including tensile strength, transparency, and cell viability. After their respective cryopreservation time, stress–strain curves of the cryopreserved membranes’ tensile strength were assessed using a universal testing machine. Both groups produced J-shaped stress–strain curves with statistically comparable parameters, including maximum stress, strain, and Young’s modulus. The percentage of cell viability was observed by trypan blue staining under a light microscope. Membrane transparency was tested with a spectrophotometer. Transparency tests showed high levels of light transmission and low haze, with no statistical difference between groups. Cell viability was statistically lower in the 30-day cryopreserved group. Tensile strength and transparency of cryopreserved CAM were not significantly impeded for up to 30 days. For CAM to be used as an alternative corneal transplant material in veterinary and regenerative medicine, further research on cell biology, biomechanical properties of the membrane, and cell viability should be conducted.

    Beschreibung: Over the last few decades, advanced manufacturing and additive printing technologies have made incredible inroads into the fields of engineering, transportation, and healthcare. Among additive manufacturing technologies, 3D printing is gradually emerging as a powerful technique owing to a combination of attractive features, such as fast prototyping, fabrication of complex designs/structures, minimization of waste generation, and easy mass customization. Of late, 4D printing has also been initiated, which is the sophisticated version of the 3D printing. It has an extra advantageous feature: retaining shape memory and being able to provide instructions to the printed parts on how to move or adapt under some environmental conditions, such as, water, wind, light, temperature, or other environmental stimuli. This advanced printing utilizes the response of smart manufactured materials, which offer the capability of changing shapes postproduction over application of any forms of energy. The potential application of 4D printing in the biomedical field is huge. Here, the technology could be applied to tissue engineering, medicine, and configuration of smart biomedical devices. Various characteristics of next generation additive printings, namely 3D and 4D printings, and their use in enhancing the manufacturing domain, their development, and some of the applications have been discussed. Special materials with piezoelectric properties and shape-changing characteristics have also been discussed in comparison with conventional material options for additive printing.

    Beschreibung: The aim of the study was to determine the effectiveness of selected seven commercial essential oils (EsO) (grapefruit, lemongrass, tea tree (TTO), thyme, verbena, cajeput, and Litsea cubeba) on isolates of common Central European parasitic fungal species of Fusarium obtained from infected wheat kernels, and to evaluate the oils as potential natural fungicides. The study was conducted in 2 stages. At each stage, the fungicidal activity of EsO (with concentrations of 0.025; 0.05; 0.125; 0.25; 0.50; 1.0, and 2.0%) against Fusarium spp. was evaluated using the disc plate method and zones of growth inhibition were measured. At the first stage, the fungistatic activity of EsO was evaluated against four species of Fusarium from the Polish population (F. avenaceum FAPL, F. culmorum FCPL, F. graminearum FGPL and F. oxysporum FOPL). The correlation coefficient between the mycelial growth rate index (T) and the fungistatic activity (FA) was calculated. At the second stage, on the basis of the mycelium growth rate index, the effectiveness of the EsO in limiting the development of Fusarium isolates from the German population (F. culmorum FC1D, F. culmorum FC2D, F. graminearum FG1D, F. graminearum FG2D and F. poae FP0D) was assessed. The first and second stage results presented as a growth rate index were then used to indicate essential oils (as potential natural fungicides) effectively limiting the development of various common Central European parasitic species Fusarium spp. Finally, the sensitivity of four Fusarium isolates from the Polish population and five Fusarium isolates from the German population was compared. The data were compiled in STATISTICA 13.0 (StatSoft, Inc, CA, USA) at the significance level of 0.05. Fusarium isolates from the German population were generally more sensitive than those from the Polish population. The sensitivity of individual Fusarium species varied. Their vulnerability, regardless of the isolate origin, in order from the most to the least sensitive, is as follows: F. culmorum, F. graminearum, F. poae, F. avenaceum and F. oxysporum. The strongest fungicidal activity, similar to Funaben T, showed thyme oil (regardless of the concentration). Performance of citral oils (lemongrass and Litsea cubeba) was similar but at a concentration above 0.025%.

    Beschreibung: Metabolic and hormonal outcomes of polycystic ovary syndrome (PCOS) have implications on telomere biology and physical activity may prevent telomere erosion. We sought to observe the effects of continuous (CAT) and intermittent (IAT) aerobic training on telomere length, inflammatory biomarkers, and its correlation with metabolic, hormonal, and anthropometric parameters of PCOS. This randomized controlled clinical trial study included 87 PCOS randomly stratified according to body mass index (BMI) in CAT (n = 28), IAT (n = 29) and non-training control group (CG, n = 30). The exercises were carried out on a treadmill, three times per week for 16 weeks. The participants’ anthropometric characteristics and biochemical and hormonal concentrations were measured before and after aerobic training or observation period, as the telomere length that was evaluated using quantitative real-time PCR. Four months of aerobic exercises (CAT or IAT) did not alter telomere length and inflammatory biomarkers in PCOS women. Obesity index as BMI and waist circumference (WC), and inflammatory biomarkers negatively affect telomeres. The hyper-andro-genism measured by testosterone levels was reduced after both exercises (CAT, p ≤ 0.001; IAT, p = 0.019). In particular, the CAT reduced WC (p = 0.045), hip circumference (p = 0.032), serum cholesterol (p ≤ 0.001), and low-density lipoprotein (p = 0.030). Whereas, the IAT decreased WC (p = 0.014), waist-to-hip ratio (p = 0.012), free androgen index (FAI) (p = 0.037). WC (p = 0.049) and body fat (p = 0.015) increased in the non-training group while total cholesterol was reduced (p = 0.010). Booth exercises reduced obesity indices and hyperandrogenism on PCOS women without changes in telomere length or inflammatory biomarkers.

    Beschreibung: Tea (Camellia sinensis) and herbal tea have been recognized as rich sources of bioactive constituents with the ability to exert antioxidant actions. The aims of this study were to analyze phenolic, carotenoid and saccharide contents in a set of Vietnamese tea and herbal tea and compare the results with those of green and black teas marketed in the U.S. In total, 27 phenolics, six carotenoids and chlorophylls, and three saccharides were quantitatively identified. Catechins, quercetin glycosides and chlorogenic acid were the predominating phenolics in the teas, with the concentrations following the order: jasmine/green teas 〉 oolong tea 〉 black tea. Lutein was the dominant carotenoid in the teas and its concentrations were generally found to be higher in the jasmine and green teas than in the oolong and black teas. The study showed that the green teas originating in Vietnam had much higher levels of phenolics and carotenoids than their counterparts stemming from another country. The application of partial least squares discriminant analysis (PLS-DA) as a chemometric tool was able to differentiate phenolic profiles between methanolic extracts and tea infusions. Through principal component analysis (PCA), the similarities and dissimilarities among the jasmine, green, oolong, black teas and herbal teas were depicted.

    Beschreibung: Gait and physical fitness are related to cognitive function. A decrease in motor function and physical fitness can serve as an indicator of declining global cognitive function in older adults. This study aims to use machine learning (ML) to identify important features of gait and physical fitness to predict a decline in global cognitive function in older adults. A total of three hundred and six participants aged seventy-five years or older were included in the study, and their gait performance at various speeds and physical fitness were evaluated. Eight ML models were applied to data ranked by the p-value (LP) of linear regression and the importance gain (XI) of XGboost. Five optimal features were selected using elastic net on the LP data for men, and twenty optimal features were selected using support vector machine on the XI data for women. Thus, the important features for predicting a potential decline in global cognitive function in older adults were successfully identified herein. The proposed ML approach could inspire future studies on the early detection and prevention of cognitive function decline in older adults.

    Beschreibung: According to the current European scenario, cultural, creative, and community-led policies play an increasingly important role in influencing local resources, systems, and infrastructures management and demand a novel approach in governing, financing, and monitoring urban regeneration processes. Therefore, cities become contexts where cultural and creative practices can be implemented, integrating social cohesion principles based on communities, shared values, and collaborative decision-making approaches, with particular attention to enhancing cultural heritage, mainly unused or underutilised. The purpose of this research is to explore how the Cultural and Creative Cities Monitor (CCCM) methodological framework, developed by the Joint Research Centre of the European Commission, can be integrated at the local scale to assess the impacts of urban regeneration processes in an interactive and dynamic way, through the data emerging from the monitoring of urban regeneration experiences activated with the communities. The paper describes the “Play ReCH (Re-use Cultural Heritage)” approach, that promotes a process of collaboration, gamification, and innovation in cultural heritage reuse, as an opportunity to test how cultural, creative, and community-led urban strategies can support the enhancement of heritage generating enabling environments and culturally vibrant contexts. The Play ReCH approach and the “Hack the City Salerno” mission, activated in the Salerno historic centre (Italy), open the reflection on some relevant issues related to how citizens become makers of cultural and creative cities’ policies, and contribute to evaluating and monitoring their implementation at diverse urban scales. The Play ReCH mission underlines how new evidence suggests declining the CCCM conceptual framework and related urban policies assessment, co-defining suitable community-based indicators.

    Beschreibung: Lignocellulosic biomass is an important source of renewable energy and a potential replacement for fossil fuels. In this work, the X-ray fluorescence (XRF) method was used to analyze the elemental composition of raw and pretreated lignocellulosic biomass of cassava peels, corn cobs, rice husks, sugarcane bagasse, yam peels, and mixtures of cassava peels and yam peels, corn cobs and rice husks and all five biomass samples combined. The influence of particle size on elemental properties was investigated by screening the selected biomass into two size fractions, of an average of 300 and 435 µm, respectively. The total concentration of Mg, Al, Si, P, S, Cl, Ca, Ti, Cr, Mn, Fe, Co, Cu, Zn, Sn, Ni, Br, Mo, Ba, Hg, and Pb were determined for each of the biomass samples before and after the different pretreatments adopted in this study. From the results of the analysis, there was a significant reduction in the concentration of calcium in all the analyzed biomass after the alkaline pretreatment with rice husks biomass having the lowest concentration of 66 ppm after the alkaline pretreatment. The sulfur content of the acid pretreated biomass increased considerably which is likely due to the sulfuric acid used for the acid pretreatment. The fact that a mixture of biomass feedstock affects the properties of the biomass after pretreatment was validated in the mixed biomass of cassava peels and yam peels biomass as an example. The concentration of Mg in the mixed biomass was 1441 ppm but was 200 ppm and 353 ppm in individual cassava peels and yam peels respectively. The results of this study demonstrated that pretreated mixtures of biomass have varied elemental compositions, which could be an important factor affecting downstream processes, especially if a hybrid feedstock is used in a large-scale application.

    Beschreibung: Wildfires considerably affect forest ecosystems. However, there is a lack of data on the post-fire status of insect communities in these ecosystems. This paper presents results of a study conducted in 2019 which considered the post-fire status of the insect fauna in a Protected Area, Mordovia State Nature Reserve (Republic of Mordovia, centre of European Russia), considered as regional hotspot of insect diversity in Mordovia. We sampled insects on intact (unburned, control) and fire-damaged (burnt in 2010) sites and compared the alpha-diversity between sites. In total, we sampled and analysed 16,861 specimens belonging to 11 insect orders, 51 families and 190 species. The largest orders represented in the samples were Coleoptera (95 species), Diptera (54 species), Hymenoptera (21 species), and Neuroptera (11 species). Other insect orders were represented by between one and four species. The largest four orders (Coleoptera, Lepidoptera, Diptera and Hymenoptera) represented 96.7% of all studied specimens. We found that in the ninth year after low intensity surface fire damage, the insect diversity had returned to a similar level to that of the control (unburned) sites. Sites damaged by crown wildfire differed considerably from other sites in terms of a negative impact on both species diversity and the number of specimens. This indicates the serious effect of the crown fires on the biodiversity and consequent long-term recovery of the damaged ecosystem.

    Beschreibung: Allophanic tephra-derived soils can sequester sizable quantities of soil organic matter (SOM). However, no studies have visualized the fine internal porous structure of allophanic soil microaggregates, nor studied the carbon structure preserved in such soils or paleosols. We used synchrotron radiation-based transmission X-ray microscopy (TXM) to perform 3D-tomography of the internal porous structure of dominantly allophanic soil microaggregates, and carbon near-edge X-ray absorption fine-structure (C NEXAFS) spectroscopy to characterize SOM in ≤ 12,000-year-old tephra-derived allophane-rich (with minor ferrihydrite) paleosols. The TXM tomography showed a vast network of internal, tortuous nano-pores within an allophanic microaggregate comprising nanoaggregates. SOM in the allophanic paleosols at four sites was dominated by carboxylic/carbonyl functional groups with subordinate quinonic, aromatic, and aliphatic groups. All samples exhibited similar compositions despite differences between the sites. That the SOM does not comprise specific types of functional groups through time implies that the functional groups are relict. The SOM originated at the land/soil surface: ongoing tephra deposition (intermittently or abruptly) then caused the land-surface to rise so that the once-surface horizons were buried more deeply and hence became increasingly isolated from inputs by the surficial/modern organic cycle. The presence of quinonic carbon, from biological processes but vulnerable to oxygen and light, indicates the exceptional protection of SOM and bio-signals in allophanic paleosols, attributable both to the porous allophane (with ferrihydrite) aggregates that occlude the relict SOM from degradation, and to rapid burial by successive tephra-fallout, as well as strong Al-organic chemical bonding. TXM and C NEXAFS spectroscopy help to unravel the fine structure of soils and SOM and are of great potential for soil science studies.

    Beschreibung: Device-independent quantum key distribution (DIQKD) provides the strongest form of secure key exchange, using only the input–output statistics of the devices to achieve information-theoretic security. Although the basic security principles of DIQKD are now well understood, it remains a technical challenge to derive reliable and robust security bounds for advanced DIQKD protocols that go beyond the previous results based on violations of the CHSH inequality. In this work, we present a framework based on semidefinite programming that gives reliable lower bounds on the asymptotic secret key rate of any QKD protocol using untrusted devices. In particular, our method can in principle be utilized to find achievable secret key rates for any DIQKD protocol, based on the full input–output probability distribution or any choice of Bell inequality. Our method also extends to other DI cryptographic tasks.

    Beschreibung: The applicability of high voltage electrical discharges for the decomposition of CO2 has been extensively demonstrated. In this study, a new AC parallel-plate plasma reactor is presented which was designed for this purpose. Detailed experimental characterization and simulation of this reactor were performed. Gas chromatography of the exhaust gases enabled calculation of the CO2 conversion and energy efficiency. A conversion factor approximating 25% was obtained which is higher in comparison to existing plasma sources. Optical emission spectroscopy enabled the determination of the emission intensities of atoms and molecules inside the plasma and characterization of the discharge. The Stark broadening of the Balmer hydrogen line Hβ was used for the estimation of the electron density. The obtained densities were of the order of 5 × 1014 cm−3 which indicates that the electron kinetic energy dominated the discharge. The rotational, vibrational, and excitation temperatures were determined from the vibro-rotational band of the OH radical. A 2-temperature plasma was found where the estimated electron temperatures (~18,000 K) were higher than the gas temperatures (~2000 K). Finally, a 2-D model using the fluid equations was developed for determining the main processes in the CO2 splitting. The solution to this model, using the finite element method, gave the temporal and spatial behaviors of the formed species densities, the electric potential, and the temperatures of electrons.

    Beschreibung: Enzymatic synthesis of aromatic biobased polyesters is a recent and rapidly expanding research field. However, the direct lipase-catalyzed synthesis of polyesters from ferulic acid has not yet been reported. In this work, various ferulic-based monomers were considered for their capability to undergo CALB-catalyzed polymerization. After conversion into diesters of different lengths, the CALB-catalyzed polymerization of these monomers with 1,4-butanediol resulted in short oligomers with a DPn up to 5. Hydrogenation of the double bond resulted in monomers allowing obtaining polyesters of higher molar masses with DPn up to 58 and Mw up to 33,100 g·mol−1. These polyesters presented good thermal resistance up to 350 °C and Tg up to 7 °C. Reduction of the ferulic-based diesters into diols allowed preserving the double bond and synthesizing polyesters with a DPn up to 19 and Mw up to 15,500 g·mol−1 and higher Tg (up to 21 °C). Thus, this study has shown that the monomer hydrogenation strategy proved to be the most promising route to achieve ferulic-based polyester chains of high DPn. This study also demonstrates for the first time that ferulic-based diols allow the synthesis of high Tg polyesters. Therefore, this is an important first step toward the synthesis of competitive biobased aromatic polyesters by enzymatic catalysis.

    Beschreibung: In the differential diagnosis of nonspecific white matter lesions (NSWMLs) detected on magnetic resonance imaging (MRI), multiple sclerosis (MS) should be taken into consideration. Optical coherence tomography (OCT) is a promising tool applied in the differential diagnostic process of MS. We tested whether OCT may be useful in distinguishing between MS and NSWMLs patients. In patients with MS (n = 41) and NSWMLs (n = 19), the following OCT parameters were measured: thickness of the peripapillary Retinal Nerve Fibre Layer (pRNFL) in superior, inferior, nasal, and temporal segments; thickness of the ganglion cell-inner plexiform layer (GCIPL); thickness of macular RNFL (mRNFL); and macular volume (MV). In MS patients, GCIPL was significantly lower than in NSWMLs patients (p = 0.024). Additionally, in MS patients, mRNFL was significantly lower than in NSWMLs patients (p = 0.030). The average segmental pRNFL and MV did not differ between MS and NSWMLs patients (p 〉 0.05). GCIPL and macular RNFL thinning significantly influenced the risk of MS (18.6% [95% CI 2.7%, 25.3%]; 27.4% [95% CI 4.5%, 62.3%]), and reduced GCIPL thickness appeared to be the best predictor of MS. We conclude that OCT may be helpful in the differential diagnosis of MS and NSWMLs patients in real-world settings.

    Beschreibung: In many embedded systems, we face the problem of correlating signals characterising device operation (e.g., performance parameters, anomalies) with events describing internal device activities. This leads to the investigation of two types of data: time series, representing signal periodic samples in a background of noise, and sporadic event logs. The correlation process must take into account clock inconsistencies between the data acquisition and monitored devices, which provide time series signals and event logs, respectively. The idea of the presented solution is to classify event logs based on the introduced similarity metric and deriving their distribution in time. The identified event log sequences are matched with time intervals corresponding to specified sample patterns (objects) in the registered signal time series. The matching (correlation) process involves iterative time offset adjustment. The paper presents original algorithms to investigate correlation problems using the object-oriented data models corresponding to two monitoring sources. The effectiveness of this approach has been verified in power consumption analysis using real data collected from the developed Holter device. It is quite universal and can be easily adapted to other device optimisation problems.
