    Beschreibung: Iron limits phytoplankton growth and hence the biological carbon pump in the Southern Ocean1. Models assessing the impacts of iron on the global carbon cycle generally rely on dust input and sediment resuspension as the predominant sources2, 3. Although it was previously thought that most iron from deep-ocean hydrothermal activity was inaccessible to phytoplankton because of the formation of particulates4, it has been suggested that iron from hydrothermal activity5, 6, 7 may be an important source of oceanic dissolved iron8, 9, 10, 11, 12, 13. Here we use a global ocean model to assess the impacts of an annual dissolved iron flux of approximately 9×108 mol, as estimated from regional observations of hydrothermal activity11, 12, on the dissolved iron inventory of the world’s oceans. We find the response to the input of hydrothermal dissolved iron is greatest in the Southern Hemisphere oceans. In particular, observations of the distribution of dissolved iron in the Southern Ocean3 (Chever et al., manuscript in preparation; Bowie et al., manuscript in preparation) can be replicated in our simulations only when our estimated iron flux from hydrothermal sources is included. As the hydrothermal flux of iron is relatively constant over millennial timescales14, we propose that hydrothermal activity can buffer the oceanic dissolved iron inventory against shorter-term fluctuations in dust deposition.

    Beschreibung: The elemental stoichiometry of sea water and particulate organic matter is remarkably similar. This observation led Redfield to hypothesize that the oceanic ratio of nitrate to phosphate is controlled by the remineralization of phytoplankton biomass1. The Redfield ratio is used universally to quantitatively link the marine nitrogen and phosphorus cycles in numerous biogeochemical applications2,3,4. Yet, empirical and theoretical studies show that the ratio of nitrogen to phosphorus in phytoplankton varies greatly with taxa5,6 and growth conditions7,8,9. Here we present a dynamic five-box ecosystem model showing that non-Redfield utilization of dissolved nitrogen and phosphorus by non-nitrogen-fixing phytoplankton controls the magnitude and distribution of nitrogen fixation. In our simulations, systems dominated by rapidly growing phytoplankton with low nitrogen to phosphorus uptake ratios reduce the phosphorus available for nitrogen fixation. In contrast, in systems dominated by slow-growing phytoplankton with high nitrogen to phosphorus uptake ratios nitrogen deficits are enhanced, and nitrogen fixation is promoted. We show that estimates of nitrogen fixation are up to fourfold too high when non-Redfield uptake stoichiometries are ignored. We suggest that the relative abundance of fast- and slow-growing phytoplankton controls the amount of new nitrogen added to the ocean.

    Beschreibung: Methane hydrate, a potential future energy resource, is known to occur naturally in vast quantities beneath the ocean floor and in permafrost regions. It is important to evaluate how much methane is recoverable from these hydrate reserves. This article introduces the theoretical background of HydrateResSim, the National Energy Technology Laboratory (NETL) methane production simulator for hydrate-containing reservoirs, originally developed for NETL by Lawrence Berkeley National Laboratory (LBNL). It describes the mathematical model that governs the dissociation of methane hydrate by depressurization or thermal stimulation of the system, including the transport of multiple temperature-dependent components in multiple phases through a porous medium. The model equations are obtained by incorporating the multiphase Darcy’s law for gas and liquid into both the mass component balances and the energy conservation equations. Two submodels in HydrateResSim for hydrate dissociation are also considered: a kinetic model and a pure thermodynamic model. Contrary to more traditional reservoir simulations, the set of model unknowns or primary variables in HydrateResSim changes throughout the simulation as a result of the formation or dissociation of ice and hydrate phases during the simulation. The primary variable switch method (PVSM) is used to effectively track these phase changes. The equations are solved by utilizing the implicit time finite-difference method on the grid system, which can properly describe phase appearance or disappearance as well as the boundary conditions. The Newton-Raphson method is used to solve the linear equations after discretization and setup of the Jacobian matrix. We report here the application of HydrateResSim to a three-component, four-phase flow system in order to predict the methane produced from a laboratory-scale reservoir. The first results of HydrateResSim code in a peer-reviewed publication are presented in this article. The numerical solution was verified against the state-of-the art simulator TOUGH+Hydrate. The model was then used to compare twodissociation theories: kinetic and pure equilibrium. Generally, the kinetic model revealed a lower dissociation rate than the equilibrium model. The hydrate dissociation patterns differed significantly when the thermal boundary condition was shifted from adiabatic to constant-temperature. The surface area factor was found to have an important effect on the rate of hydrate dissociation for the kinetic model. The deviation between the kinetic and equilibrium models was found to increase with decreasing surface area factor.

    Beschreibung: Growth, protein amount, and activity levels of metabolic pathways in Trichodesmium are influenced by environmental changes such as elevated pCO2 and temperature. This study examines changes in the expression of essential metabolic genes in Trichodesmium grown under a matrix of pCO2 (400 and 900 µatm) and temperature (25 and 31°C). Using RT-qPCR, we studied 21 genes related to four metabolic functional groups: CO2 concentrating mechanism (bicA1, bicA2, ccmM, ccmK2, ccmK3, ndhF4, ndhD4, ndhL, chpX), energy metabolism (atpB, sod, prx, glcD), nitrogen metabolism (glnA, hetR, nifH), and inorganic carbon fixation and photosynthesis (rbcL, rca, psaB, psaC, psbA). nifH and most photosynthetic genes exhibited relatively high abundance and their expression was influenced by both environmental parameters. A two to three orders of magnitude increase was observed for glnA and hetR only when both pCO2 and temperature were elevated. CO2 concentrating mechanism genes were not affected by pCO2 and temperature and their expression levels were markedly lower than that of the nitrogen metabolism and photosynthetic genes. Many of the CO2 concentrating mechanism genes were co-expressed throughout the day. Our results demonstrate that in Trichodesmium, CO2 concentrating mechanism genes are constitutively expressed. Co-expression of genes from different functional groups were frequently observed during the first half of the photoperiod when oxygenic photosynthesis and N2 fixation take place, pointing at the tight and complex regulation of gene expression in Trichodesmium. Here we provide new data linking environmental changes of pCO2 and temperature to gene expression in Trichodesmium. Although gene expression indicates an active metabolic pathway, there is often an uncoupling between transcription and enzyme activity, such that transcript level cannot usually be directly extrapolated to metabolic activity.

    Beschreibung: The two commonly applied methods to assess dinitrogen (N2) fixation rates are the 15N2-tracer addition and the acetylene reduction assay (ARA). Discrepancies between the two methods as well as inconsistencies between N2 fixation rates and biomass/growth rates in culture experiments have been attributed to variable excretion of recently fixed N2. Here we demonstrate that the 15N2-tracer addition method underestimates N2 fixation rates significantly when the 15N2 tracer is introduced as a gas bubble. The injected 15N2 gas bubble does not attain equilibrium with the surrounding water leading to a 15N2 concentration lower than assumed by the method used to calculate 15N2-fixation rates. The resulting magnitude of underestimation varies with the incubation time, to a lesser extent on the amount of injected gas and is sensitive to the timing of the bubble injection relative to diel N2 fixation patterns. Here, we propose and test a modified 15N2 tracer method based on the addition of 15N2-enriched seawater that provides an instantaneous, constant enrichment and allows more accurate calculation of N2 fixation rates for both field and laboratory studies. We hypothesise that application of N2 fixation measurements using this modified method will significantly reduce the apparent imbalances in the oceanic fixed-nitrogen budget.

    Beschreibung: Concerns about the slow pace of climate mitigation have led to renewed dialogue about solar-radiation management, which could be achieved by adding reflecting aerosols to the stratosphere1, 2, 3, 4, 5, 6. Modelling studies suggest that solar-radiation management could produce stabilized global temperatures and reduced global precipitation4, 5, 6. Here we present an analysis of regional differences in a climate modified by solar-radiation management, using a large-ensemble modelling experiment that examines the impacts of 54 scenarios for global temperature stabilization. Our results confirm that solar-radiation management would generally lead to less extreme temperature and precipitation anomalies, compared with unmitigated greenhouse gas emissions. However, they also illustrate that it is physically not feasible to stabilize global precipitation and temperature simultaneously as long as atmospheric greenhouse gas concentrations continue to rise. Over time, simulated temperature and precipitation in large regions such as China and India vary significantly with different trajectories for solar-radiation management, and they diverge from historical baselines in different directions. Hence, it may not be possible to stabilize the climate in all regions simultaneously using solar-radiation management. Regional diversity in the response to different levels of solar-radiation management could make consensus about the optimal level of geoengineering difficult, if not impossible, to achieve.

    Beschreibung: The growth of oceanic plates at mid-ocean ridges, crustal accretion, occurs by a combination of magmatic and tectonic processes. Magmatic processes along ridges spreading at fast, intermediate and slowrates, continually add volcanic material to a centrally located spreading axis. This creates a narrowband of young volcanic rocks. However, at ridges spreading at ultraslow rates, diminished volcanism allows entire blocks of mantle to spread on the sea floor by tectonic processes. Remote imaging has advanced our observational understanding of crustal accretion, but temporal constraints are required to quantitatively understand ultraslow-spreading ridge construction. Here, we use U-series eruption ages of volcanic rocks collected from the ultraslow-spreading Southwest Indian Ridge. Unexpectedly, we find young volcanic eruption ages that are broadly dispersed throughout the rift valley, indicating that crustal accretion of young volcanic rocks is not confined to a narrow central spreading axis. As areas of young volcanism are observed close to distinct fault surfaces, we propose that the widely dispersed volcanism may result frommagma rising along faults. Our results indicate that axial-centric spreading models may not accurately describe crustal accretion at ultraslow-spreading ridges, prompting the re-evaluation of these models.

    Beschreibung: Marine-derived fungi have been shown in recent years to produce a plethora of new bioactive secondary metabolites, some of them featuring new carbon frameworks hitherto unprecedented in nature. These compounds are of interest as new lead structures for medicine as well as for plant protection. The aim of this protocol is to give a detailed description of methods useful for the isolation and cultivation of fungi associated with various marine organisms (sponges, algae and mangrove plants) for the extraction, characterization and structure elucidation of biologically active secondary metabolites produced by these marine-derived endophytic fungi, and for the preliminary evaluation of their pharmacological properties based on rapid 'in house' screening systems. Some results exemplifying the positive outcomes of the protocol are given at the end. From sampling in marine environment to completion of the structure elucidation and bioactivity screening, a period of at least 3 months has to be scheduled.

    Beschreibung: Whether the characteristics of tropical cyclones have changed or will change in a warming climate — and if so, how — has been the subject of considerable investigation, often with conflicting results. Large amplitude fluctuations in the frequency and intensity of tropical cyclones greatly complicate both the detection of long-term trends and their attribution to rising levels of atmospheric greenhouse gases. Trend detection is further impeded by substantial limitations in the availability and quality of global historical records of tropical cyclones. Therefore, it remains uncertain whether past changes in tropical cyclone activity have exceeded the variability expected from natural causes. However, future projections based on theory and high-resolution dynamical models consistently indicate that greenhouse warming will cause the globally averaged intensity of tropical cyclones to shift towards stronger storms, with intensity increases of 2–11% by 2100. Existing modelling studies also consistently project decreases in the globally averaged frequency of tropical cyclones, by 6–34%. Balanced against this, higher resolution modelling studies typically project substantial increases in the frequency of the most intense cyclones, and increases of the order of 20% in the precipitation rate within 100 km of the storm centre. For all cyclone parameters, projected changes for individual basins show large variations between different modelling studies.

    Beschreibung: The hydrographic properties of the North Atlantic Ocean changed significantly from 1950 to 2000: the subtropics warmed and became more saline, whereas the subpolar ocean cooled and freshened. These changes directly affect the storage of heat and fresh water in the ocean, but their consequences for ocean dynamics are determined by the resultant changes in seawater density. Here we use historical hydrographic data to show that the overall seawater density in the North Atlantic basin decreased during this 50-year period. As a result of these density changes, sea-surface heights changed in a spatially varying pattern with typical rates of 2 mm yr−1, in broad agreement with tide-gauge measurements. Melding the observed density fields within a numerical model we find a slight weakening in the overturning of the subtropical gyre by −1.5±1 Sv and a slight strengthening in the overturning of the subpolar gyre by +0.8±0.5 Sv. These gyre-specific changes run counter to the canonical notion of a single, basin-scale overturning cell and probably reflect interannual and decadal trends rather than any long-term climate trend. We conclude that gyre dynamics strongly affect temperature and salinity changes that translate into changes in the meridional overturning circulation.

    Beschreibung: Background Surface waters of aquatic environments have been shown to both evolve and consume hydrogen and the ocean is estimated to be the principal natural source. In some marine habitats, H2 evolution and uptake are clearly due to biological activity, while contributions of abiotic sources must be considered in others. Until now the only known biological process involved in H2 metabolism in marine environments is nitrogen fixation. Principal Findings We analyzed marine and freshwater environments for the presence and distribution of genes of all known hydrogenases, the enzymes involved in biological hydrogen turnover. The total genomes and the available marine metagenome datasets were searched for hydrogenase sequences. Furthermore, we isolated DNA from samples from the North Atlantic, Mediterranean Sea, North Sea, Baltic Sea, and two fresh water lakes and amplified and sequenced part of the gene encoding the bidirectional NAD(P)-linked hydrogenase. In 21% of all marine heterotrophic bacterial genomes from surface waters, one or several hydrogenase genes were found, with the membrane-bound H2 uptake hydrogenase being the most widespread. A clear bias of hydrogenases to environments with terrestrial influence was found. This is exemplified by the cyanobacterial bidirectional NAD(P)-linked hydrogenase that was found in freshwater and coastal areas but not in the open ocean. Significance This study shows that hydrogenases are surprisingly abundant in marine environments. Due to its ecological distribution the primary function of the bidirectional NAD(P)-linked hydrogenase seems to be fermentative hydrogen evolution. Moreover, our data suggests that marine surface waters could be an interesting source of oxygen-resistant uptake hydrogenases. The respective genes occur in coastal as well as open ocean habitats and we presume that they are used as additional energy scavenging devices in otherwise nutrient limited environments. The membrane-bound H2-evolving hydrogenases might be useful as marker for bacteria living inside of marine snow particles.

    Beschreibung: Background: Regime shifts are abrupt changes encompassing a multitude of physical properties and ecosystem variables, which lead to new regime conditions. Recent investigations focus on the changes in ecosystem diversity and functioning associated to such shifts. Of particular interest, because of the implication on climate drivers, are shifts that occur synchronously in separated basins. Principal Findings: In this work we analyze and review long-term records of Mediterranean ecological and hydro-climate variables and find that all point to a synchronous change in the late 1980s. A quantitative synthesis of the literature (including observed oceanic data, models and satellite analyses) shows that these years mark a major change in Mediterranean hydrographic properties, surface circulation, and deep water convection (the Eastern Mediterranean Transient). We provide novel analyses that link local, regional and basin scale hydrological properties with two major indicators of large scale climate, the North Atlantic Oscillation index and the Northern Hemisphere Temperature index, suggesting that the Mediterranean shift is part of a large scale change in the Northern Hemisphere. We provide a simplified scheme of the different effects of climate vs. temperature on pelagic ecosystems. Conclusions: Our results show that the Mediterranean Sea underwent a major change at the end of the 1980s that encompassed atmospheric, hydrological, and ecological systems, for which it can be considered a regime shift. We further provide evidence that the local hydrography is linked to the larger scale, northern hemisphere climate. These results suggest that the shifts that affected the North, Baltic, Black and Mediterranean (this work) Seas at the end of the 1980s, that have been so far only partly associated, are likely linked as part a northern hemisphere change. These findings bear wide implications for the development of climate change scenarios, as synchronous shifts may provide the key for distinguishing local (i.e., basin) anthropogenic drivers, such as eutrophication or fishing, from larger scale (hemispheric) climate drivers.

    Beschreibung: Dense water formed over the Antarctic continental shelf rapidly descends into the deep ocean where it spreads throughout the global ocean as Antarctic Bottom Water1, 2. The coldest and most voluminous component of this water mass is Weddell Sea bottom water1, 3, 4, 5, 6, 7. Here we present observations over eight years of the temperature and salinity stratification in the lowermost ocean southeast of the South Orkney Islands, marking the export of Weddell Sea bottom water. We observe a pronounced seasonal cycle in bottom temperatures, with a cold pulse in May/June and a warm one in October/November, but the timing of these phases varies each year. We detect the coldest bottom water in 1999 and 2002, whereas there was no cold phase in 2000. On the basis of current velocities and water mass characteristics, we infer that the pulses originate from the southwest Weddell Sea. We propose that the seasonal fluctuations of Weddell Sea bottom-water properties are governed by the seasonal cycle of the winds over the western margin of the Weddell Sea. Interannual fluctuations are linked to the variability of the wind-driven Weddell Sea gyre and hence to large-scale climate phenomena such as the Southern Annular Mode and El Niño/Southern Oscillation.

    Beschreibung: Large amounts (estimates range from 70 Tg per year to 300 Tg per year) of the potent greenhouse gas methane are oxidized to carbon dioxide in marine sediments by communities of methanotrophic archaea and sulphate-reducing bacteria1, 2, 3, and thus are prevented from escaping into the atmosphere. Indirect evidence indicates that the anaerobic oxidation of methane might proceed as the reverse of archaeal methanogenesis from carbon dioxide with the nickel-containing methyl-coenzyme M reductase (MCR) as the methane-activating enzyme4, 5. However, experiments showing that MCR can catalyse the endergonic back reaction have been lacking. Here we report that purified MCR from Methanothermobacter marburgensis converts methane into methyl-coenzyme M under equilibrium conditions with apparent Vmax (maximum rate) and Km (Michaelis constant) values consistent with the observed in vivo kinetics of the anaerobic oxidation of methane with sulphate6, 7, 8. This result supports the hypothesis of ‘reverse methanogenesis’4, 9 and is paramount to understanding the still-unknown mechanism of the last step of methanogenesis. The ability of MCR to cleave the particularly strong C–H bond of methane without the involvement of highly reactive oxygen-derived intermediates is directly relevant to catalytic C–H activation, currently an area of great interest in chemistry10, 11, 12, 13.

    Beschreibung: Instrumental records, proxy data and climate modelling show that multidecadal variability is a dominant feature of North Atlantic sea-surface temperature variations1–4, with potential impacts on regional climates. To understand the observed variability and to gauge any potential for climate predictions it is essential to identify the physical mechanisms that lead to this variability, and to explore the spatial and temporal characteristics of multidecadal variability modes. Here we use a coupled ocean–atmosphere general circulation model to show that the phasing of the multidecadal fluctuations in the North Atlantic during the past 600 years is, to a large degree, governed by changes in the external solar and volcanic forcings. We find that volcanoes play a particularly important part in the phasing of the multidecadal variability through their direct influence on tropical sea-surface temperatures, on the leading mode of northern-hemisphere atmosphere circulation and on the Atlantic thermohaline circulation. We suggest that the implications of our findings for decadal climate prediction are twofold: because volcanic eruptions cannot be predicted a decade in advance, longer-term climate predictability may prove challenging, whereas the systematic post-eruption changes in ocean and atmosphere may hold promise for shorter term climate prediction.

    Beschreibung: The chemical composition of the Caribbean sponge Pandaros acanthifolium was reinvestigated and led to the isolation of 12 new steroidal glycosides, namely, pandarosides E-J (1-6) and their methyl esters (7-12). Their structures were determined on the basis of extensive spectroscopic analyses, including two-dimensional NMR and HRESIMS data. Like the previously isolated pandarosides A-D (13-16), the new compounds 1-12 share an unusual oxidized D-ring and a cis C/D ring junction. The absolute configurations of the aglycones were assigned by interpretation of CD spectra, whereas the absolute configurations of the monosaccharide units were determined by chiral GC analyses of the acid methanolysates. The majority of the metabolites showed in vitro activity against three or four parasitic protozoa. Particularly active were the compounds 3 (pandaroside G) and its methyl ester (9), which potently inhibited the growth of Trypanosoma brucei rhodesiense (IC(50) values 0.78 and 0.038 mu M, respectively) and Leishmania donovani (IC(50)'s 1.3 and 0.051 mu M, respectively).

    Beschreibung: Polycomb group (PcG) proteins act as evolutionary conserved epigenetic mediators of cell identity because they repress transcriptional programs that are not required at particular developmental stages. Each tissue is likely to have a specific epigenetic profile, which acts as a blueprint for its developmental fate. A hallmark for Polycomb Repressive Complex 2 (PRC2) activity is trimethylated lysine 27 on histone H3 (H3K27me3). In plants, there are distinct PRC2 complexes for vegetative and reproductive development, and it was unknown so far whether these complexes have target gene specificity. The FERTILIZATION INDEPENDENT SEED (FIS) PRC2 complex is specifically expressed in the endosperm and is required for its development; loss of FIS function causes endosperm hyperproliferation and seed abortion. The endosperm nourishes the embryo, similar to the physiological function of the placenta in mammals. We established the endosperm H3K27me3 profile and identified specific target genes of the FIS complex with functional roles in endosperm cellularization and chromatin architecture, implicating that distinct PRC2 complexes have a subset of specific target genes. Importantly, our study revealed that selected transposable elements and protein coding genes are specifically targeted by the FIS PcG complex in the endosperm, whereas these elements and genes are densely marked by DNA methylation in vegetative tissues, suggesting that DNA methylation prevents targeting by PcG proteins in vegetative tissues. In this study, we established the cell type–specific epigenome profile of PcG activity in the endosperm. The endosperm has reduced levels of DNA methylation, and based on our data we propose that PcG proteins are specifically targeted to hypomethylated sequences in the endosperm. Among these endosperm-specific PcG targets are genes with functional roles in endosperm cellularization and chromatin architecture, implicating a fundamental role of PcG proteins in regulating endosperm development. Importantly, we identified transposable elements and genes among the specific PcG targets in the endosperm that are densely marked by DNA methylation in vegetative tissues, suggesting an antagonistic placement of DNA methylation and H3K27me3 at defined sequences.

    Beschreibung: The Macrourid fish roundnose grenadier, Coryphaenoides rupestris, is one of the most common benthopelagic fishes on the northern mid-Atlantic Ridge. The ecology of the species is comparatively well studied in continental slope waters of the North Atlantic, but not on the mid-Atlantic Ridge, which is a central mid-ocean area of its distribution. In total, 166 specimens from the RV G.O. Sars cruise in July 2004 were examined. The diet mainly comprised cephalopods, pelagic shrimps and fish. Pelagic and benthopelagic copepods were the most numerous prey, but did not contribute much on a weight basis. Cephalopods were by far the most important prey of the small grenadiers, while shrimps and fish became increasingly significant with increasing size. Previous studies from other areas have also found pelagic prey to be important, but in contrast to this study, cephalopods were generally of less importance. The study was an element of more wide-ranging food-web studies of the mid-Atlantic Ridge macro- and megafauna communities within the international MAR-ECO project.
