    Beschreibung: Abstract: Hydrological conditions at the Kleiner Landgraben Valley (Neubrandenburg, NE Germany) were planned to be modified as a part of the Mecklenburg-Vorpommern peatland conservation program. Before restoring hydrological conditions, extensive top-soil removal from drained fens was also scheduled. By doing so, phosphorus, ammonia and methane release from rewetted fens is pretended to be strongly reduced. Moreover, a marked nitrate export reduction is expected, what will decrease N fluxes into the Baltic Sea. Several field and laboratory experiments were performed in the period 2009-2012 in order to evaluate: a) the influence of agricultural activities in the catchment on nutrient load over the lowland fens and nutrient export from drained fens site; b) the groundwater fluctuation at drained fen, c) the phosphorus release potential from the upper peat layer, and d) the impact of top-soil removal on the level of phosphorus concentrations in the surface water of inundated peatlands. Such previous research served as a basis of a laboratory experiment aiming to investigate the influence of the degree of peat decomposition, temperature and N load over nitrate removal and phosphorus turnover. According to our results, nitrate is efficiently removed from highly degraded peat (up to 90 %), whereas phosphorus release decreased at increasing nitrate load and is negligible in less decomposed peat. Moreover, moderately decomposed peat was also observed to have a high nitrate removal potential. Our results provide useful insights for the management of rewetted fens, and can be applied in another agricultural watershed within the NE Germany glacial landscape.
    Beschreibung: Zusammenfassung: Im Rahmen des Moorschutzprogramms von Mecklenburg-Vorpommern war eine hydrologische Umgestaltung des Kleinen Landgrabentals bei Neubrandenburg geplant. Dazu waren auch großflächige Flachabtorfungen im entwässerten Moor vorgesehen. Diese Maßnahme sollte dazu beitragen, eine hohe Phosphor-, Ammonium- und Methan-Freisetzung nach einer Moorvernässung zu unterbinden. Gleichzeitig sollte eine deutliche Reduzierung der Nitratausträge aus dem Einzugsgebiet des Kleinen Landgrabens erfolgen, um letztlich die N-Belastung der Ostsee zu senken. Im Zeitraum 2009 bis 2012 wurden daher umfangreiche Freiland- und Laboruntersuchungen zu folgenden Themen durchgeführt: (a) Höhe der Nährstoffbelastung des Moores durch das landwirtschaftlich genutzte Einzugsgebiet und des Nährstoffaustrages aus dem entwässerten Moor, (b) Schwankungen der Grundwasserstände im Moor unter entwässerten Bedingungen, (c) Höhe des P-Freisetzungspotenzials im Oberboden und daraus resultierendes P-Austragsrisiko und (d) Einfluss einer Flachabtorfung auf die Höhe der P-Konzentrationen im Freiwasser überstauter Moore. Diese Ergebnisse waren Grundlage für Laborversuche zur Bestimmung der Nitratreduktion sowie der P-Dynamik in Abhängigkeit von Torfqualität, Temperatur und äußerer Nitratbelastung. Es konnte gezeigt werden, dass der Nitratrückhalt besonders hoch in den vererdeten Torfen ausfällt (bis zu 90 %), aber auch mäßig zersetzte Torfe wiesen noch ein großes Nitratrückhaltepotenzial auf. Gleichzeitig wurde in den vererdeten Torfen bei einer hohen Nitratbelastung die Phosphorfreisetzung stark herabgesetzt wobei die Phosphorfreisetzung in mittel zersetzten Torfen generell sehr gering ausfiel. Auf der Basis dieser Ergebnisse wurden konkrete Handlungsempfehlungen für die Wiedervernässungspraxis abgeleitet, die auf andere Regionen des pleistozän geprägten nordostdeutschen Tieflandes übertragbar sind.

    Beschreibung: Temporal lobe epilepsy (TLE) is characterized by debilitating, recurring seizures and an increased risk for cognitive deficits. Mossy cells (MCs) are key neurons in the hippocampal excitatory circuit, and the partial loss of MCs is a major hallmark of TLE. We investigated how MCs contribute to spontaneous ictal activity and to spatial contextual memory in a mouse model of TLE with hippocampal sclerosis, using a combination of optogenetic, electrophysiological, and behavioral approaches. In chronically epileptic mice, real-time optogenetic modulation of MCs during spontaneous hippocampal seizures controlled the progression of activity from an electrographic to convulsive seizure. Decreased MC activity is sufficient to impede encoding of spatial context, recapitulating observed cognitive deficits in chronically epileptic mice.

    Beschreibung: Glia constitute roughly half of the cells of the central nervous system (CNS) but were long-considered to be static bystanders to its formation and function. Here we provide an overview of how the diverse and dynamic functions of glial cells orchestrate essentially all aspects of nervous system formation and function. Radial glia, astrocytes, oligodendrocyte progenitor cells, oligodendrocytes, and microglia each influence nervous system development, from neuronal birth, migration, axon specification, and growth through circuit assembly and synaptogenesis. As neural circuits mature, distinct glia fulfill key roles in synaptic communication, plasticity, homeostasis, and network-level activity through dynamic monitoring and alteration of CNS structure and function. Continued elucidation of glial cell biology, and the dynamic interactions of neurons and glia, will enrich our understanding of nervous system formation, health, and function.

    Beschreibung: To understand how neurons assemble to form functional circuits, it is necessary to obtain a detailed knowledge of their diversity and to define the developmental specification programs that give rise to this diversity. Invertebrates and vertebrates appear to share common developmental principles of neuronal specification in which cascades of transcription factors temporally pattern progenitors, while spatial cues modify the outcomes of this temporal patterning. Here, we highlight these conserved mechanisms and describe how they are used in distinct neural structures. We present the questions that remain for a better understanding of neuronal specification. Single-cell RNA profiling approaches will potentially shed light on these questions, allowing not only the characterization of neuronal diversity in adult brains, but also the investigation of the developmental trajectories leading to the generation and maintenance of this diversity.

    Beschreibung: Clinical observations indicate that the paramedian region of the thalamus is a critical node for controlling wakefulness. However, the specific nucleus and neural circuitry for this function remain unknown. Using in vivo fiber photometry or multichannel electrophysiological recordings in mice, we found that glutamatergic neurons of the paraventricular thalamus (PVT) exhibited high activities during wakefulness. Suppression of PVT neuronal activity caused a reduction in wakefulness, whereas activation of PVT neurons induced a transition from sleep to wakefulness and an acceleration of emergence from general anesthesia. Moreover, our findings indicate that the PVT–nucleus accumbens projections and hypocretin neurons in the lateral hypothalamus to PVT glutamatergic neurons’ projections are the effector pathways for wakefulness control. These results demonstrate that the PVT is a key wakefulness-controlling nucleus in the thalamus.

    Beschreibung: The salience of behaviorally relevant stimuli is dynamic and influenced by internal state and external environment. Monitoring such changes is critical for effective learning and flexible behavior, but the neuronal substrate for tracking the dynamics of stimulus salience is obscure. We found that neurons in the paraventricular thalamus (PVT) are robustly activated by a variety of behaviorally relevant events, including novel ("unfamiliar") stimuli, reinforcing stimuli and their predicting cues, as well as omission of the expected reward. PVT responses are scaled with stimulus intensity and modulated by changes in homeostatic state or behavioral context. Inhibition of the PVT responses suppresses appetitive or aversive associative learning and reward extinction. Our findings demonstrate that the PVT gates associative learning by providing a dynamic representation of stimulus salience.

    Beschreibung: Animals constantly update their body posture to meet behavioral demands, but little is known about the neural signals on which this depends. We therefore tracked freely foraging rats in three dimensions while recording from the posterior parietal cortex (PPC) and the frontal motor cortex (M2), areas critical for movement planning and navigation. Both regions showed strong tuning to posture of the head, neck, and back, but signals for movement were much less dominant. Head and back representations were organized topographically across the PPC and M2, and more neurons represented postures that occurred less often. Simultaneous recordings across areas were sufficiently robust to decode ongoing behavior and showed that spiking in the PPC tended to precede that in M2. Both the PPC and M2 strongly represent posture by using a spatially organized, energetically efficient population code.

    Beschreibung: Memory resides in engram cells distributed across the brain. However, the site-specific substrate within these engram cells remains theoretical, even though it is generally accepted that synaptic plasticity encodes memories. We developed the dual-eGRASP (green fluorescent protein reconstitution across synaptic partners) technique to examine synapses between engram cells to identify the specific neuronal site for memory storage. We found an increased number and size of spines on CA1 engram cells receiving input from CA3 engram cells. In contextual fear conditioning, this enhanced connectivity between engram cells encoded memory strength. CA3 engram to CA1 engram projections strongly occluded long-term potentiation. These results indicate that enhanced structural and functional connectivity between engram cells across two directly connected brain regions forms the synaptic correlate for memory formation.

    Beschreibung: Consolidation of spatial and episodic memories is thought to rely on replay of neuronal activity sequences during sleep. However, the network dynamics underlying the initial storage of memories during wakefulness have never been tested. Although slow, behavioral time scale sequences have been claimed to sustain sequential memory formation, fast ("theta") time scale sequences, nested within slow sequences, could be instrumental. We found that in rats traveling passively on a model train, place cells formed behavioral time scale sequences but theta sequences were degraded, resulting in impaired subsequent sleep replay. In contrast, when the rats actively ran on a treadmill while being transported on the train, place cells generated clear theta sequences and accurate trajectory replay during sleep. Our results support the view that nested sequences underlie the initial formation of memory traces subsequently consolidated during sleep.

In particular, which of the two scenarios underlies the attenuation of remote (month-old) fear memories is completely unknown, despite the impetus to better understand this process against the backdrop of enduring traumata. We found—chemogenetically and in an engram-specific manner—that effective remote fear attenuation is accompanied by the reactivation of memory recall–induced neurons in the dentate gyrus and that the continued activity of these neurons is critical for fear reduction. This suggests that the original memory trace of fear actively contributes to remote fear attenuation.

    Beschreibung: Why are some visual stimuli consciously detected, whereas others remain subliminal? We investigated the fate of weak visual stimuli in the visual and frontal cortex of awake monkeys trained to report stimulus presence. Reported stimuli were associated with strong sustained activity in the frontal cortex, and frontal activity was weaker and quickly decayed for unreported stimuli. Information about weak stimuli could be lost at successive stages en route from the visual to the frontal cortex, and these propagation failures were confirmed through microstimulation of area V1. Fluctuations in response bias and sensitivity during perception of identical stimuli were traced back to prestimulus brain-state markers. A model in which stimuli become consciously reportable when they elicit a nonlinear ignition process in higher cortical areas explained our results.

    Beschreibung: The specific effects of sleep on synaptic plasticity remain unclear. We report that mouse hippocampal sharp-wave ripple oscillations serve as intrinsic events that trigger long-lasting synaptic depression. Silencing of sharp-wave ripples during slow-wave states prevented the spontaneous down-regulation of net synaptic weights and impaired the learning of new memories. The synaptic down-regulation was dependent on the N -methyl- d -aspartate receptor and selective for a specific input pathway. Thus, our findings are consistent with the role of slow-wave states in refining memory engrams by reducing recent memory-irrelevant neuronal activity and suggest a previously unrecognized function for sharp-wave ripples.

    Beschreibung: Memories are integrated into interconnected networks; nevertheless, each memory has its own identity. How the brain defines specific memory identity out of intermingled memories stored in a shared cell ensemble has remained elusive. We found that after complete retrograde amnesia of auditory fear conditioning in mice, optogenetic stimulation of the auditory inputs to the lateral amygdala failed to induce memory recall, implying that the memory engram no longer existed in that circuit. Complete amnesia of a given fear memory did not affect another linked fear memory encoded in the shared ensemble. Optogenetic potentiation or depotentiation of the plasticity at synapses specific to one memory affected the recall of only that memory. Thus, the sharing of engram cells underlies the linkage between memories, whereas synapse-specific plasticity guarantees the identity and storage of individual memories.

    Beschreibung: The tuberal nucleus (TN) is a surprisingly understudied brain region. We found that somatostatin (SST) neurons in the TN, which is known to exhibit pathological or cytological changes in human neurodegenerative diseases, play a crucial role in regulating feeding in mice. GABAergic tuberal SST ( TN SST) neurons were activated by hunger and by the hunger hormone, ghrelin. Activation of TN SST neurons promoted feeding, whereas inhibition reduced it via projections to the paraventricular nucleus and bed nucleus of the stria terminalis. Ablation of TN SST neurons reduced body weight gain and food intake. These findings reveal a previously unknown mechanism of feeding regulation that operates through orexigenic TN SST neurons, providing a new perspective for understanding appetite changes.

    Beschreibung: Episodic memories are encoded by a sparse population of hippocampal neurons. In mice, optogenetic manipulation of this memory engram established that these neurons are indispensable and inducing for memory recall. However, little is known about their in vivo activity or precise role in memory. We found that during memory encoding, only a fraction of CA1 place cells function as engram neurons, distinguished by firing repetitive bursts paced at the theta frequency. During memory recall, these neurons remained highly context specific, yet demonstrated preferential remapping of their place fields. These data demonstrate a dissociation of precise spatial coding and contextual indexing by distinct hippocampal ensembles and suggest that the hippocampal engram serves as an index of memory content.

    Beschreibung: Brain size variation over primate evolution and human development is associated with shifts in the proportions of different brain regions. Individual brain size can vary almost twofold among typically developing humans, but the consequences of this for brain organization remain poorly understood. Using in vivo neuroimaging data from more than 3000 individuals, we find that larger human brains show greater areal expansion in distributed frontoparietal cortical networks and related subcortical regions than in limbic, sensory, and motor systems. This areal redistribution recapitulates cortical remodeling across evolution, manifests by early childhood in humans, and is linked to multiple markers of heightened metabolic cost and neuronal connectivity. Thus, human brain shape is systematically coupled to naturally occurring variations in brain size through a scaling map that integrates spatiotemporally diverse aspects of neurobiology.

    Beschreibung: Plasticity of cortical responses in vivo involves activity-dependent changes at synapses, but the manner in which different forms of synaptic plasticity act together to create functional changes in neurons remains unknown. We found that spike timing–induced receptive field plasticity of visual cortex neurons in mice is anchored by increases in the synaptic strength of identified spines. This is accompanied by a decrease in the strength of adjacent spines on a slower time scale. The locally coordinated potentiation and depression of spines involves prominent AMPA receptor redistribution via targeted expression of the immediate early gene product Arc. Hebbian strengthening of activated synapses and heterosynaptic weakening of adjacent synapses thus cooperatively orchestrate cell-wide plasticity of functional neuronal responses.

    Beschreibung: Neural stem and progenitor cells (NSPCs) generate neurons throughout life in the mammalian hippocampus. We used chronic in vivo imaging and followed genetically labeled individual NSPCs and their progeny in the mouse hippocampus for up to 2 months. We show that NSPCs targeted by the endogenous Achaete-scute homolog 1 (Ascl1) promoter undergo limited rounds of symmetric and asymmetric divisions, eliciting a burst of neurogenic activity, after which they are lost. Further, our data reveal unexpected asymmetric divisions of nonradial glia-like NSPCs. Cell fates of Ascl1-labeled lineages suggest a developmental-like program involving a sequential transition from a proliferative to a neurogenic phase. By providing a comprehensive description of lineage relationships, from dividing NSPCs to newborn neurons integrating into the hippocampal circuitry, our data offer insight into how NSPCs support life-long hippocampal neurogenesis.

    Beschreibung: An animal’s awareness of its location in space depends on the activity of place cells in the hippocampus. How the brain encodes the spatial position of others has not yet been identified. We investigated neuronal representations of other animals’ locations in the dorsal CA1 region of the hippocampus with an observational T-maze task in which one rat was required to observe another rat’s trajectory to successfully retrieve a reward. Information reflecting the spatial location of both the self and the other was jointly and discretely encoded by CA1 pyramidal cells in the observer rat. A subset of CA1 pyramidal cells exhibited spatial receptive fields that were identical for the self and the other. These findings demonstrate that hippocampal spatial representations include dimensions for both self and nonself.

    Beschreibung: Prior studies have demonstrated that correlated variability changes with cognitive processes that improve perceptual performance. We tested whether correlated variability covaries with subjects’ performance—whether performance improves quickly with attention or slowly with perceptual learning. We found a single, consistent relationship between correlated variability and behavioral performance, regardless of the time frame of correlated variability change. This correlated variability was oriented along the dimensions in population space used by the animal on a trial-by-trial basis to make decisions. That subjects’ choices were predicted by specific dimensions that were aligned with the correlated variability axis clarifies long-standing paradoxes about the relationship between shared variability and behavior.

    Beschreibung: It has long been hypothesized that aging and neurodegeneration are associated with somatic mutation in neurons; however, methodological hurdles have prevented testing this hypothesis directly. We used single-cell whole-genome sequencing to perform genome-wide somatic single-nucleotide variant (sSNV) identification on DNA from 161 single neurons from the prefrontal cortex and hippocampus of 15 normal individuals (aged 4 months to 82 years), as well as 9 individuals affected by early-onset neurodegeneration due to genetic disorders of DNA repair (Cockayne syndrome and xeroderma pigmentosum). sSNVs increased approximately linearly with age in both areas (with a higher rate in hippocampus) and were more abundant in neurodegenerative disease. The accumulation of somatic mutations with age—which we term genosenium—shows age-related, region-related, and disease-related molecular signatures and may be important in other human age-associated conditions.

    Beschreibung: Cross-talk between the nervous and immune systems has been well described in the context of adult physiology and disease. Recent advances in our understanding of immune cell ontogeny have revealed a notable interplay between neurons and microglia during the prenatal and postnatal emergence of functional circuits. This Review focuses on the brain, where the early symbiotic relationship between microglia and neuronal cells critically regulates wiring, contributes to sex-specific differences in neural circuits, and relays crucial information from the periphery, including signals derived from the microbiota. These observations underscore the importance of studying neurodevelopment as part of a broader framework that considers nervous system interactions with microglia in a whole-body context.

    Beschreibung: Grid cells are neurons active in multiple fields arranged in a hexagonal lattice and are thought to represent the "universal metric for space." However, they become nonhomogeneously distorted in polarized enclosures, which challenges this view. We found that local changes to the configuration of the enclosure induce individual grid fields to shift in a manner inversely related to their distance from the reconfigured boundary. The grid remained primarily anchored to the unchanged stable walls and showed a nonuniform rescaling. Shifts in simultaneously recorded colocalized grid fields were strongly correlated, which suggests that the readout of the animal’s position might still be intact. Similar field shifts were also observed in place and boundary cells—albeit of greater magnitude and more pronounced closer to the reconfigured boundary—which suggests that there is no simple one-to-one relationship between these three different cell types.

    Beschreibung: Neuronal synapse formation and remodeling are essential to central nervous system (CNS) development and are dysfunctional in neurodevelopmental diseases. Innate immune signals regulate tissue remodeling in the periphery, but how this affects CNS synapses is largely unknown. Here, we show that the interleukin-1 family cytokine interleukin-33 (IL-33) is produced by developing astrocytes and is developmentally required for normal synapse numbers and neural circuit function in the spinal cord and thalamus. We find that IL-33 signals primarily to microglia under physiologic conditions, that it promotes microglial synapse engulfment, and that it can drive microglial-dependent synapse depletion in vivo. These data reveal a cytokine-mediated mechanism required to maintain synapse homeostasis during CNS development.

    Beschreibung: Thorndike’s law of effect states that actions that lead to reinforcements tend to be repeated more often. Accordingly, neural activity patterns leading to reinforcement are also reentered more frequently. Reinforcement relies on dopaminergic activity in the ventral tegmental area (VTA), and animals shape their behavior to receive dopaminergic stimulation. Seeking evidence for a neural law of effect, we found that mice learn to reenter more frequently motor cortical activity patterns that trigger optogenetic VTA self-stimulation. Learning was accompanied by gradual shaping of these patterns, with participating neurons progressively increasing and aligning their covariance to that of the target pattern. Motor cortex patterns that lead to phasic dopaminergic VTA activity are progressively reinforced and shaped, suggesting a mechanism by which animals select and shape actions to reliably achieve reinforcement.

    Notizen: Abstract In West Africa, Uscana lariophaga (Hymenoptera: Trichogrammatidae) parasitizes the eggs of Callosobruchus maculatus (Coleoptera: Bruchidae), an important pest of stored cowpea. The impact of the egg parasitoid on pest populations was evaluated in clay pots used in traditional storage in Niger. At the beginning of the storage period cowpeas were infested with different densities of larval instars and adults of C. maculatus and inoculated with one density of U. lariophaga. The higher the initial densities of C. maculatus, the better the egg parasitoid was able to establish itself and to control the population of C. maculatus, limiting the damage to cowpea. After three months of storage, the egg parasitoid significantly reduced the number of C. maculatus adults by 68 at the lowest and 86 percent at the highest initial density of the beetle; the percentage of damaged beans was reduced by 13 and 19% respectively.

    Notizen: Abstract Entomopathogenic nematodes (Rhabdita: Heterorhabditidae and Steinernematidae) have been effective as inundative biological control agents of scarab larvae (Coleoptera: Scarabaeidae) in turfgrass. Entomopathogenic nematodes also occur naturally in turfgrass and endemic or inoculated populations may be able to provide effective long-term control. Variation in Heterorhabditis bacteriophora and Steinernema carpocapsae spatial and temporal distribution along transects placed at different turfgrass sites in central New Jersey, USA, was investigated. H. bacteriophora tended to be recovered from fewer sections in a transect than S. carpocapsae, but the two species, overall, did not differ in patchiness of distribution. In one transect with a H. bacteriophora population S. feltiae was also recovered, but the two populations seldom overlapped spatially. In transects with adequate scarab larvae density for analysis, H. bacteriophora density and Popillia japonica larvae density were inversely correlated. This suggests that endemic H. bacteriophora populations may suppress P. japonica populations. In one transect, an epizootic of H. bacteriophora in an undetermined host may have occurred. Edaphic factors were relatively uniform along transects and were, at most, weakly correlated with nematode recovery. Uniform inoculative releases of H. bacteriophora tended to return to patterns of distribution typical of endemic populations.

    Notizen: Abstract Adult females of the predatory mite Phytoseiulus persimilis Athias-Henriot are strongly attracted to infochemicals released by plants infested with their prey, the two-spotted spider mite (Tetranychus urticae Koch), thereby effectively locating their prey. However, we found a consistently lower degree of attraction to these infochemicals for a population of P. persimilis, which is called non-responding population. Here we demonstrate that this low degree of attraction is a contagious phenomenon and that it cannot be explained by differences in abiotic conditions, physiological state and experience of predators or by genetic differences between predator populations. Female predators exposed to dead conspecifics of the non-responding population and their products showed a lower degree of attraction to plant odours and a higher mortality than predators exposed to products of a living conspecific of the non-responding population. This was true 6–7 days after contact with dead conspecifics and their products whereas 2 days after contact no effects were detected. The present results are discussed in view of our hypothesis that the change in foraging behaviour as well as the high mortality rate are symptoms of a contagious disease affecting the non-responding population.

    Notizen: Abstract In a laboratory study two coccinellid species, Coleomegilla maculata (DeGeer) and Harmonia axyridis Pallas, completed preimaginal development on lacewing eggs, Chrysoperla carnea Stephens or pea aphids, Acyrthosiphon pisum (Harris) in similar times. Preimaginal survival on C. carnea eggs was similar to survival on A. pisum for all stages of C. maculata and H. axyridis. Coccinellid adults that developed on C. carnea eggs were smaller than adults reared on A. pisum. Coccinella septempunctata L. did not complete preimaginal development on C. carnea eggs. Chrysoperla carnea preimaginal developmental time was approximately 20 days when fed either C. maculata eggs or A. pisum. Chrysoperla carnea fed C. maculata eggs developed into smaller adults, compared to adults reared as larvae on A. pisum, Ostrinia nubilalis (Hübner) eggs, or A. pisum alternated daily with O. nubilalis eggs. C. carnea did not complete preimaginal development on H. axyridis eggs. Cannibalism occurred more frequently between C. carnea third instars than between C. maculata fourth instars. When a C. carnea third instar was paired with a C. maculata fourth instar, more C. maculata were preyed upon by C. carnea, regardless of the herbivorous prey density. In the field these two predator species may negatively affect each other and reduce their suppression of pest densities.

    Notizen: Abstract The supercooling point (SCP) of an insect model, the lady beetle Hippodamia convergens Guérin-Menéville (Coleoptera, Coccinellidae) was markedly elevated by treatment with aqueous suspensions of the filamentous, ice nucleation active (INA) fungi Fusarium avenaceum and slightly elevated by Fusarium acuminatum. Addition of the surfactant Tween 80 to the fungal suspensions further reduced the supercooling capacity of adult beetles. When used alone the surfactant Triton X-100 produced a greater SCP elevation than Tween 20 or Tween 80. The emulsifier gum arabic was ineffective in elevating beetle SCPs when applied alone and when added to INA fungal preparations it decreased their efficacy. Aqueous suspensions of both viable sporulating and viable pleomorphic (a permanent, degenerative, nonsporulating cultural state) forms of both fungal species were more effective in elevating the SCP than killed preparations except for the pleomorphic F. acuminatum suspension in which the killed form was slightly more active. Application of INA fungi applied in combination with surfactants may be useful in the development of methods for the biological control of overwintering freeze-susceptible insect pests by decreasing their capacity to avoid lethal freezing by supercooling.

    Notizen: Abstract Previous field experiments indicated that the presence of the bird cherry-oat aphid, Rhopalosiphum padi (L.), on perennial grasses can decrease the effectiveness of predatory lacewings, Chrysoperla plorabunda (Fitch), in reducing populations of the Russian wheat aphid, Diuraphis noxia (Mordvilko). We tested the hypothesis that R. padi deflects predation away from D. noxia because it feeds in sites that are more accessible to predators. We quantified the behavior of lacewing larvae on crested wheatgrass plants bearing either D. noxia alone or an equal mixture of D. noxia and R. padi. On non-flowering plants, R. padi typically occurred on leaf sheaths or open blades, and was encountered and captured more often than D. noxia, which usually fed within immature, rolled leaves. Overall time-budgets of lacewings did not differ between the pure-D. noxia and mixed-species treatments, but 〉75% of the time spent consuming aphids in the mixed-species treatment was devoted to R. padi. On flowering plants, D. noxia usually aggregated on the flag leaf below the inflorescence, whereas R. padi occurred mostly on leaf sheaths. Predators again captured R. padi more often than D. noxia, and spent more time consuming aphids in the mixed-species treatment than in the pure-D. noxia treatment. These behavioral observations support the hypothesis that non-target prey can hamper the short-term effectiveness of biological-control agents against D. noxia.

    Notizen: Abstract To examine the influence of phosphorus on the commercial V2O5(WO3)/TiO2 SCR catalyst, measurements were carried out by means of infrared and Raman spectroscopy, XPS, and NO reduction measurement as a function of phosphorus loading. Phosphorus added to the catalyst was found to disperse well over the catalyst without a significant agglomeration up to 5 wt% P2O5 addition. The number of the hydroxyl groups bonded to the vanadium and titanium species decreased readily with increasing amount of phosphorus. Correspondingly, the hydroxyl groups bonded to the phosphorus species were formed. NH3 adsorbed on both hydroxyl groups bonded to vanadium and phosphorus as ammonium ions, implying that the P–OH groups formed are also responsible for the Brønsted acidity. The NO reduction activity was found to be decreased with increasing amount of phosphorus; however, the influence of phosphorus was relatively small irrespective of the large amount of phosphorus addition. The deactivation might be caused by the change in the nature of the surface hydroxyl groups as Brønsted acid sites. Phosphorus species might partially wrap the surface V=O and W=O groups, which might also contribute to the deactivation.

    Notizen: Abstract Diatom analyses were undertaken of sediment cores covering a range of water depths in a small eutrophic lake (Lough Augher, Co. Tyrone, N. Ireland). The significance of between-core variability in diatom relative frequency stratigraphy was assessed by Canonical Correspondence Analysis (CCA) where the ordination axes were constrained to external environmental variables (sediment depth, core location coordinates, water depth, effective fetch, distance-from-shore and distance-from-inflow). After the removal of the effect of sediment age by partialling it out, the resultant first two axes from the partial-CCA were significantly correlated with water depth and distance-from-shore, indicating non-uniform diatom stratigraphies across the lake. Despite this variability, all cores show the same succession of species and, therefore, record the eutrophication of the lake. Diatom-inferred total phosphorus (DI-TP) was inferred for six cores using weighted averaging regression and calibration. Apart from considerable differences of DI-TP in surficial sediment samples, there was good between-core repeatability of DI-TP profiles. These data support the use of DI-TP for establishing background nutrient concentrations for lakes, and associated implications for lake restoration schemes using single cores. Comparisons of DI-TP profiles and total diatom accumulation rate data for the individual cores indicate that diatom production peaked prior to the maximum TP concentrations in the lake.

    Notizen: Abstract Phosphorus accumulation rates in depositional zone sediments of Lake Okeechobee were determined in 11 mud-zone cores and two peat-zone cores dated by 210Pb. Although difficulties were encountered in interpreting 210Pb data from some sites, reliable dating of sediments from the mud zone of this shallow lake is possible. Sediment accumulation rates in this zone have increased during the present century by an average of about twofold, and accumulation of organic sediments in the lake during pre-settlement times apparently was much slower than during the past century. Concentrations of all forms of sedimentary P but especially nonapatite inorganic-P and organic-P also have increased since pre-settlement times and especially since about 1940. Annual P accumulation rates in the lake's sediments have increased about fourfold during the 1900s, with most of the increase occurring in the past 40–50 years. The recent accumulation rate of sedimentary P (past ~ 10 years) agrees within a factor of 1.5 with the net retention of P in the lake calculated from published input-output mass balances.

It has been the target of successful classical biocontrol programmes utilizing an insect natural enemy collected in Brazil and introduced into Australia, Papua New Guinea and South Africa. Little attention was given to the fungal pathogens associated with this weed as potential biocontrol agents. An 11-month field survey in the state of Rio de Janeiro involving repeated visits to 12 sites, and a 2-month survey in the Amazonas Basin (Amazonas state) in Brazil, yielded only one pathogenic fungus on P. stratioites. This was identified as Cercospora pistiae. No previous record of this species has been made in Brazil nor in the New World. The fungus is described and its taxonomy and biocontrol potential discussed herein. C. pistiae is a damaging pathogen of water lettuce. Its distribution was restricted to two interconnected sites in Rio de Janeiro. A literature and herbaria survey yielded few fungi associated to water lettuce (8 species), probably reflecting a lack of adequate collection in the centre of origin of the plant.

    Notizen: Abstract A suggested increase in the growth of macrophytic algae within the Ythan estuary (N.E. Scotland) over recent years has been linked to the increased amounts of nitrogen in the form of NO3–N entering the estuary from the river. The increased NO3 concentration in the river has been associated with recent changes in farming practices in this predominantly agricultural catchment. Terrestrially derived phosphorus is also considered to contribute increasingly to eutrophication of fresh waters. Historical agricultural census data together with appropriate surveys of fertilizer practice were used to calculate the total quantities of fertilizer and manure derived N and P applied annually over the wholeYthan catchment during the period 1960 – 1990. While the total agricultural land area has remained similar, significant changes in cropping practice have occurred. In particular, a greater proportion of land is given to autumn sown crops while the area of grassland has declined. These changes in farming practice are associated with differences in both the total amounts and timing of fertilizer applied. The use of inorganic N in the catchment has trebled since 1960 and is currently approximately 6400 tonnes (104 kg N/ha). The use of P has decreased by more than a quarter to 1274 tonnes (21 kg P/ha) over a similar time period. There has been no obvious change in total quantity of N and P derived from animal manures, estimated to be 44 and 11 kg per ha, respectively, when averaged over the area of agricultural land. Cattle and sheep numbers have remained relatively constant and together account for approximately 80% of the manure N and 70% of the manure P produced annually. However, poultry have declined by 70% since 1960 while pig numbers have increased six-fold. The average annual application rate of manure derived N over the whole catchment (44 kg/ha) is considerably below that proposed at the farm scale in the EC Nitrate Directive (210–170 kg/ha). However, on a local scale difficulties may arise for large manure producing concerns such as dairy or pig units.

    Notizen: Abstract A severe root rot disease of cotton caused by Pythium carolinianum was diagnosed in a cotton field in Beni-Musa village, 20 km southwest of El-Minia city, Egypt, during the summer of 1996. This was the first reported isolation of this fungus in Egypt. In the light of the importance of the cotton industry in Egypt, research was initiated to develop a biocontrol agent against Pythium carolinianum. In vitro agar plate technique identified a Pseudomonas fluorescens strain that was highly antagonist to Pythium carolinianum. Subsequent plant growth experiments establish that substantial disease control could be obtained by applying Pseudomonas fluorescens to the soil. Optimal control was obtained by mixing the bacteria with the soil rather than by dipping the cotton roots in the bacterial suspension immediately before planting. Disease was more severe in autoclaved soil than in nonsterile soil.

Thirty-five references in the scientific literature were found principally dealing with acarine predators. The first paper to point out the specific value of Cheyletus eruditus as a predator of acaroid mites appeared in 1912. In 1965 biological control utilizing C. eruditus was put into practice for the first time and in 1986 a method of mass rearing the predator was developed.

    Notizen: Abstract The biological control of pest tephritid flies using parasitoids has been successful in relatively few subtropical and tropical regions. The best documented successes were in Hawaii and Florida, USA, Fiji and southern Europe. There were relatively limited successes in Australia, Costa Rica and Mexico. With the accidental establishment of new pest tephritids, such as Bactrocera latifrons (Hendel) in Hawaii or Bactrocera papayae Drew and Hancock in Australia, foreign exploration for new parasitoids is essential. A renewal of interest in classical biological control has recently occurred, although not at the same level as in the 1940s and 1950s. New parasitoid species are currently being obtained for several tephritids, such as Ceratitis capitata (Weidemann), B. latifrons and Anastrepha suspensa (Loew). The advances in mass rearing and quality control technology for parasitoids has enabled researchers to perform large-scale field testing of these parasitoids to determine the potential of augmentative releases. Numerous studies on the augmentative release of parasitoids have been done. Historically, the larval—pupal parasitoid Diachasmimorpha longicaudata (Ashmead) has been the most frequently studied parasitoid, due to the ease of rearing this species. However, recent successes in rearing other species with different biologies, e.g. the egg—pupal parasitoid Biosteres arisanus (Sonan), the gregarious eulophid, Tetrastichus giffardianus (Silvestri) and the pupal parasitoid, Coptera sp., will enable researchers to broaden the repertoire of parasitoid species for the future. Both successes and failures in augmentative releases have occurred. Without knowledge of the behavioural ecology of parasitoids, the reasons for success or failure can only be surmised. Recent research on the dispersal and host habitat finding of tephritid parasitoids will provide insights into improving augmentation and conservation strategies. The integrated pest management (IPM) of tephritid flies in the tropics has been less well-developed than research and programmes for tephritids in temperate zones. Significant emphasis is now being placed on the development of quarantine treatment methods that are environmentally sound; thus IPM will take a larger role. Several existing pest management strategies are reviewed which show the potential for compatibility with the activities of tephritid parasitoids. These include trap cropping, insecticides with selective toxicity to the target pests, mass trapping with parapheromones, the sterile insect technique and field sanitation.

    Notizen: Abstract The distribution and dynamics of phosphorus have been studied in the mangroves of Sepetiba Bay, Brazil. Leaf fall contributes 3.0 kg P ha=1yr=1to the sediment. The total above ground biomass of the R. mangle stand was about 65.3 t ha=1, the P accumulation was 3.9 kg P ha=1where 63% of the total P-biomass was accumulated in the leaves. The biomass of below ground roots was about 8.2 tha=1 and accumulated 16% of total P-biomass. Sediment contained 452 kg P ha=1 where P combined with calcium (P-Ca) was the main fraction (260 kg ha=1). The annual flux of P as litter fall was small (〈 1%) compared to total P in the sediment reservoir. The annual export of P by macrodetritus corresponds to 0.05% of the total sediment reservoir.

    Notizen: Abstract Alfalfa weevil, Hypera postica (Gyllenhall) (Coleoptera: Curculionidae), is of Eurasian origin. This destructive pest of alfalfa (lucerne) was accidentally established in North America on three separate occasions. These introductions are commonly identified in the literature as biological strains: western alfalfa weevil, Egyptian alfalfa weevil (=Hypera brunneipennis Boheman), and eastern alfalfa weevil. Alfalfa weevil has been the target of classical biological control almost since its discovery in North America more than 90 years ago. These efforts have resulted in establishment of at least nine exotic parasitoids and egg predators: Bathyplectes curculionis (Thomson), B. anurus (Thomson) and B. stenostigma (Thomson) (Hymenoptera: Ichneumonidae); Microctonus aethiopoides Loan and M. colesi Drea (Hymenoptera: Braconidae), the latter of undetermined origin; Oomyzus incertus (Ratzenberg) (Hymenoptera: Eulophidae); Dibrachoides dynastes (Forester) and Peridesmia discus (Walker) (Hymenoptera: Pteromalidae); and Anaphes luna (Girault) (Hymenoptera: Mymaridae). A fungal pathogen, Zoophthora phytonomi Arthur (Phycomycetes: Entomophthoraceae), of undetermined origin, is becoming an increasingly important alfalfa weevil mortality factor. Most major USA alfalfa production areas now benefit from a complex of alfalfa weevil biological control agents. Collectively, these agents have effected substantial reduction in the economic importance of alfalfa weevil across the northern USA However, biological agents provide only partial control of alfalfa weevil, and importance of their contribution differs considerably with production area. Still, the benefits achieved, especially from reduced need for insecticides in alfalfa production, mark this as one of the great success stories of classical biological control in North America.

    Notizen: Abstract Although the vast majority of orthopterans are not pests, some species have the potential to cause serious damage to human interests. Management of pest populations frequently conflicts with conservation of orthopteran species and processes, particularly when the pest species or its ecological processes are susceptible to extinction or when the pest population is coincident with non-target orthopterans. With respect to chemical control, the greatest hazards are the broad-spectrum, highly lethal properties of most agents, which can be mitigated with formulation and application methods. Biological control risks permanent, large-scale changes to orthopteran species and processes which can be minimized with bioinsecticidal and other short-lived or selective formulations and reliable host-range testing. Cultural control may have large-scale, broad-spectrum impacts to non-target orthopterans, but these hazards can be diminished by appropriate testing and monitoring. Mechanical control methods may be impractically labour intensive, but they are highly target specific and therefore warrant further consideration. Social control measures such as education, insurance and compensation programmes appear to have little direct potential for harm to orthopteran conservation, but the complex socioeconomic and, ultimately, environmental consequences of such programmes have not been assessed. The melding of orthopteran pest management and conservation requires that we perceive these insects and their ecological processes to be vital elements of sustainable agroecosystems. Our management of orthopterans (both non-target and pest populations) must focus on keeping good stewards on the land.

    Notizen: Summary Plants of early potato cultivars were sampled at successive harvests during tuber development and individual tubers were analysed for α-solanine and α-chaconine using high-performance liquid chromatography. Mean tuber total glycoalkaloid concentration (α-solanine plus α-chaconine) per plant decreased with time and statistically significant differences between cultivars were observed in the patterns of decline during tuber growth. The mean tuber ratio of α-chaconine to α-solanine increased during tuber development and was also affected by genotype. Total glycoalkaloid concentration for individual tubers of marketable sizes (up to 50 g fresh weight) exceeded the safety limit of 20 mg per 100 g fresh weight for cultivars Home Guard and British Queen but not for Rocket. These differences were attributable to differences in both rates and patterns of glycoalkaloid accumulation during tuber development, although in all cultivars results were consistent with glycoalkaloid accumulation occurring in the expanding peripheral cell layers of tubers for a considerable period after tuber initiation.

    Notizen: Summary Potato, wheat and sugar beet were grown in a growth chamber in tanks with flowing solutions containing K-concentrations of 1.5, 5, 15, 50 and 200 μM to find reasons for different K contents of solution needed to reach 90% maximum dry matter accumulation (external K requirement). All species showed variation in dry matter accumulation with varying K levels. To reach 90% maximum dry matter accumulation potato needed 40 μM K, whereas wheat and sugar beet needed 6.4 and 4.4 μM K respectively. These differences in external K requirement could not be explained by differences in their internal K requirement since the internal requirement of potato (7.7% K) was about one and a half times of that of sugar beet (5.4%) but its external requirement was nine times of that of sugar beet. At low K supply potato plants showed a lower influx and a reduced root growth as compared with wheat and sugar beet.

    Notizen: Summary In an epiphytotic year of early blight, data were recorded on its incidence in 72 progenies (18 females x 4 males) and their parents planted in a randomised complete block design at the Punjab Agricultural University, Ludhiana, India. Combining ability analysis showed that both additive and non-additive gene action were important, with a preponderance of the latter in the inheritance of resistance to early blight. In general, progenies were as good as or better than mid-parent values. Crosses involving both or at least one parent with good general combining ability resulted in heterotic early blight resistant progenies. Promising parents and crosses were identified.

    Notizen: Summary A test procedure for PSTVd is described based on immobilisation of plant sap on filter paper, by dotting or tissue printing followed by RT-PCR. Tests were carried out using primarily and secondarily infected potato plants, primarily infected in vitro plants, and potato tubers. Print PCR was shown to be suitable for testing large samples of potato plants whereas dot PCR is recommended for in vitro plantlets and tuber tissue. Bulking one infected plant to 4 or 9 healthy plants gave reliable results with secondarily infected potato plants, but sometimes the test failed to detect PSTVd in primarily infected in vitro plants. Dotted and printed paper squares could be stored at 4°C for at least 2 weeks in Triton X-100 solution or under dry conditions. Storing at room temperature can lead to unreliable results.

    Notizen: Summary Using potato parental lines homozygous at a locus or loci controlling resistance to potato leafroll virus (PLRV) can give advantages in the selection of resistant forms. In order to identify homozygous diploid clones their test-cross families were evaluated. All the clones that were test-crossed expressed resistance in primarily- and secondarily-infected plants and etiolated sprouts, and were derived from mating genotypes highly resistant to PLRV. Genotypes from test-cross families varied in resistance to PLRV, and one family was found which had only resistant genotypes, suggesting that the resistant parent of this progeny was homozygous at resistance loci. Evidence was gathered that resistance in some diploid clones may result from resistance to virus multiplication as well as restricted virus transport from leaves to tubers.

    Notizen: Summary Methods for quantifying external damage and bruising can be inaccurate and result in an index that is difficult to interpret. A new index was derived by taking measurements of peeled slices from cv. Record and developing a mathematical model to predict the percentage volume removed per peeler stroke for a range of potato cultivars. It was found that an average of 1.25% of the tuber volume was removed per peeler stroke. By counting the number of peeler strokes to remove damaged tissue and multiplying by 1.25, an estimate of the percentage tuber volume lost due to bruising or external damage can be obtained. The index can accommodate most types of damage and result in an index that is readily understood.

    Notizen: Summary Variation in aggressiveness to tubers among isolates ofPhytophthora infestans sampled from three potato growing regions in the Netherlands was compared. Variation in the ability to infect tubers of cv. Bintje was found between isolates of each of the three regional populations. The most aggressive isolate of the old population matched the average level of the new population in its ability to infect tubers. As a consequence, the commonly used reference isolate VK 6C can no longer be considered to be representative of the present population ofP. infestans. Therefore it is recommended that testing tuber resistance for the official list of potato cultivars with this isolate should be discontinued. Tuber infection and subsequent spread of the fungus in the tuber tissues were not found to be correlated. The components of tuber pathogenicity studied were not correlated to pathogenicity factors in the foliage, as measured under growth chamber conditions.
