    Notes: A low-energy (5-450 eV) collision-activated dissociation (CAD) study of a series of aliphatic amines revealed that at collision energies above 200 eV, charge-site-initiated fragmentation occurs. The resulting fragment ions can be utilized in the characterization of alkyl substituents of di- and trisubstituted aliphatic amines. In the presence of Cu2+ and a suitable nucleophile, such as n-dodecylamine, N-alkyl protoporphyrins dealkylate to afford copper protoporphyrin and an alkyl-dodecylamine adduct. A CAD study of a number of alkyl-dodecylamine adducts derived from the copper-induced dealkylation of synthetic N-alkyl protoporphyrins, using charge-site-initiated fragment ions, showed that the alkyl group was trapped by the nucleophilic amine present. Subsequently this method was used to identify the alkyl group of a biologically derived N-alkyl protoporphyrin.

    Notes: A CONCEPT 1 SQ hybrid mass spectrometer has been used to show the potential of high resolution mass spectrometry and hybrid tandem mass spectrometry (MS/MS) for dioxin analysis. The usual method chosen for gas chromatography/mass spectrometry (GC/MS) determination of polychlorinated dibenzodioxins (PCDDs) and polychlorinated dibenzofurans (PCDFs) is selected-ion monitoring (SIM) at high resolution, the high resolution imparting a degree of specificity to the analysis. An alternative technique is to use low resolution (1000) gas chromatography/tandem mass spectrometry on a hybrid mass spectrometer, where the specificity of MS/MS is utilized. Data are presented illustrating the potential of both techniques from a single instrument, the CONCEPT 1 SQ.

    Notes: The transmission oscillation properties of parent ions through an RF-only quadrupole are investigated as a function of collision energy, radicfrequency of the quadrupole, and type and pressure of target gas. It is shown that small variations of collision energy can result in substantial differences in daughter-ion spectra obtained from the same parent ion. Since absolute values of collision energy are difficult to determine, reproducible daughter-ion spectra are problematical to acquire.In this work we show that reproduction of daughter-ion spectra, using the transmission properties of the collision cell, is possible. The characteristics of an RF-only quadrupole (Q*) are such that the parent ion intensity at the second detector varies as a function of collision energy and displays sinusoidal properties. By using the nodes and anti-nodes of the parent-ion transmission it is possible to achieve reproducible daughter-ion spectra of a wide range of biopolymers. The tuning of Q* is mass-dependent and not compound-dependent. Hence it is possible to optimize and tune Q* using an isobaric (or almost) standard such as one of the ions of perfluorotri-n-butylamine (electron ionization) or a glycerol cluster ion (fast-atom bombardment mass spectrometry) before subjecting valuable sample to tandem mass spectrometry.

    Notes: The oximes of monosaccharides have not been well characterized by mass spectrometry. Conventional electron ionization (EI) experiments have supported the structures of these compounds but the data produced are usually equivocal (Finch and Merchant, JCS Perkin 1, 1682 (1975)). The application of fast-atom bombardment mass spectrometry to monosaccharide oximes gives molecular weight information. However, little fragmentation is observed and it is not possible to distinguish between stereoisomers. Using fast-atom bombardment tandem mass spectrometry experiments, we have produced daughter-ion spectra which are sufficiently different to enable us to distinguish between the stereoisomers of the oximes of monosaccharides.

    Notes: The introduction of more rigorous regulations aimed at controlling the use of sulphonated dyestuffs and dyestuff intermediates has pointed to the need for screening methods for the presence of these involatile compounds. Although much work has been caried out recently on the mass spectrometric behaviour of these compounds, it has been concerned principally with the production of molecular weight information using novel ionization methods rather than with any attempt to obtain structurally diagnostic fragment ions. In this mass spectrometric investigation of a range of naphthalene sulphonic acid salts, positive-ion and negative-ion fast-atom bombardment (FAB) mass spectra have been obtained and FAB-tandem mass spectrometry (MS/MS) has been used to acquire spectra (daugher-ion, parent-ion and neutral-loss) in an attempt to develop a useful screening method. On known compounds, best results were found when using a negative-ion neutral-loss scan to detect the loss of 64 mass units (corresponding to loss of SO2). This was confirmed by successful use of this scan to screen a sample of sulphonate residues.

    Notes: Electron-ionization-induced mass spectra of N-monosubstituted 2-phenylacetamides were recorded and their fragmentation patterns were studied by metastable-ion analyses. Representative deuterated analogues of these compounds have also been synthesized and their mass spectra compared with those of the unlabelled parent compounds. The most typical fragmentation for N-alkyl-, N-isoalkyl- and N-cycloalkyl-2-phenylacetamides is cleavage of the bond β to the carbonyl function, resulting in an ion fragment of m/z 92, following the transfer of hydrogen and elimination of a corresponding ketene. The primary fragmentation process for N-aryl substituted 2-phenylacetamides is the loss of an aromatic hydrogen atom from the molecular ion. The other principal fragmentation processes observed with these compounds are discussed.

    Notes: Polycyclic biomarkers such as steranes, diasteranes and tri-, tetra- and pentacyclic terpanes in the hydrocarbon fraction of oils were studied by gas chromatographic/mass spectrometric (GC/MS) analysis in order to correlate six crude oils. Distribution and abundance of these biomarkers were determined for the samples from the Kikinda, Mokrin (Vojvodina) and Požarevac oil fields (two samples for each oil field). The SIM (single-ion monitoring) method was used for the analysis of steranes and terpanes. Individual steranes, terpanes and their stereoisomers were identified by the comparison of obtained retention times with those quoted in the literature. On the basis of the results obtained, it can be concluded that the oils from the Kikinda and Mokrin (Vojvodina) fields belong to the same genetic type, while the oil from the Požarevac field belongs to another type. It was also found that the oils from the Kikinda and Mokrin fields were of a higher degree of maturity than the oils from the Požarevac field.

    Notes: The molecular weights of four linear synthetic peptides, fragments of a snake α-neurotoxin, were measured by 252Cf-plasma desorption mass spectrometry. The fragmentation phenomenon observed at the level of serine and/or threonine residue with a concomitant ion/fragment association is reported for a group of two peptides (B and D) in contrast with the group (A and C) in spite of the high ratio of serine and threonine, namely peptide A. The propensity for specific fragmentation of peptide D seems to be correlated to the repetitive sequence, (Gly-Ser)2. Finally, based on the m/z of the daughter-ions measured, we propose an overall mechanism as an N→O acyl shift analogous to that observed for serine- and threonine- containing peptides in solution chemistry.

    Notes: Double-electron-capture reactions in which singly charged projectile ions A+ convert to negative ions A- in collisions with molecules M are endoergic. Non-zero cross-sections for the reactions occur when the endoergicities fall within a ‘reaction window’. The existence of a window for reactions with CH3Br molecules is demonstrated in this investigation by altering the population of ground and excited states of the resulting CH3Br2+ ions using three projectile ions, S+·, OH+ and F+ which markedly alters the endoergicities of the double-electron-capture reactions. Analysis of the energy-loss spectra indicates that the reaction window for CH3Br is between endoergicities of about 8 and 22 eV. Using F+ as the projectile ion, it is shown that the population of the more highly excited states of CH3Br2+ is enhanced.

    Notes: Ready identification of hydroxy metabolites of cannabichromene (CBC) by mass spectrometry of their trimethylsilyl derivatives is prevented by the dominant fragmentation to give a substituted chromenyl ion; this suppresses ions diagnostic of the position of metabolic hydroxylation. To overcome this difficulty, metabolites were hydrogenated over a rhodium/alumina catalyst to reduce the double bond responsible for chromenyl ion formation and to redirect the fragmentation to the site of metabolic attack. This resulted in the production of abundant diagnostic fragment ions enabling all monohydroxy-CBCs to be readily identified.

    Notes: Analysis of the profiles of isotope clusters can be used to determine the charge state of ions in thermospray mass spectrometry. This observation was verified by comparison of experimental results with theoretical calculations. The applicability of this technique was illustrated by the characterization of peptides whose molecular weights were in the range of 500 to 5000 Da. Using thermospray ionization on a quadrupole mass spectrometer, [M + 2H]2+ and [M + 3H]3+ ions of the peptides were identified. These multiply charged ions enabled us to characterize molecules with molecular weights exceeding the normal mass range of the instrument.

    Notes: To date, methods for calculating the trajectories of ions in radio frequency (RF) multipole fields have involved solving, analytically or numerically, the appropriate equations of motion. Another method is presented here whereby a non-ideal distribution of field may be modelled and the position of an ion can be tracked, as it traverses the multipole. The validity of the calculations is discussed and some of the properties of collision cells are explained. Fragmentation processes within the cell are modelled showing that the focusing properties within the cell have a striking effect on parent- and daughter-ion transmission.

    Notes: The particle beam liquid chromatography/mass spectrometry (LC/MS) interface has been adapted to a double-focusing sector instrument of reversed geometry. Electron ionizaition (EI) mass spectra of single components injected through a loop or of mixtures separated via a reversed-phase LC column have been recorded. These are superimposable with EI mass spectra obtained via a direct-insertion probe. Sensitivity tests show that high picogram and low nanogram amounts of samples can be detected. Complete high-resolution mass spectra on standards (single components and mixtures) were recorded. Good accuracy (better than 5 mmu) was obtained in most cases on 5 μg samples. To illustrate the versatility of the system, mass-analysed ion kinetic energy scans and linked scans at constant B/E were obtained on the molecular ion of cholesterol (2.5 μg and 500 pg). Parameters relating to various parts of a commercial interface are briefly reported.

    Notes: The 70 eV electron ionization mass spectra of six cyclohexane fused 2-N-phenyliminoperhydro-1,3-oxazines and six related thiazines were recorded and their fragmentation patterns were studied by metastable-ion analysis and exact mass measurement. All the compounds examined lost a hydrogen atom from their molecular ion in a process best described as intramolecular cyclization. The most typical fragmentations were the loss of various alkyl radicals from the M+· or [M—H]+ ions with the simultaneous migration of one or more hydrogen atoms from the carbocyclic part to the heterocyclic part of the molecule. N-Substitution clearly affected the type and extent of different reactions. In general, the fragmentation patterns of the thiazine derivatives were more complicated than those of the related oxazines. In all cases studied, the cis- and trans-fused isomers gave rise to so similar spectra that their adequate differentiation was not possible.

    Notes: The matrix-assisted desorption/ionization of large biomolecules with a wavelength of 2.94 μm in the infrared is reported. A mechanically Q-switched Erbium-YAG laser with a pulse width of ca. 200 ns was used for the experiments. A large variety of matrices have been used successfully, among them all matrices found useful for ultraviolet desorption and, in addition, carboxylic acids, glycerol and urea. Desorption of molecules at this wavelength and caffeic acid as a matrix results in ions with up to 13 charges per ion.

    Notes: The electron-ionization induced fragmentation of diphenylacetylene and its d1, d2, d5 and d10 analogues has been studied by mass spectrometry. The main fragment ions found in the mass spectrum of diphenylacetylene are due to the loss of two hydrogen radicals in a stepwise process and expulsion of C2H2 from the molecular ion in parallel fragmentation pathways producing the fragment ions at m/z 176 and m/z 152, respectively, the latter having possibly a biphenylene radical cation structure. Knowing the actual composition of deuterated derivatives, and the fragmentation pattern of diphenylacetylene, the distribution of the label in these ions was calculated under the assumption that this distribution is statistical. The final results were compared with the experimentally observed spectra of deuterated compounds. This comparison favours the assumption of hydrogen randomization prior to m/z 176 and m/z 152 formation. In contrast to the loss of hydrogen radicals and of C2H2, the formation of m/z 139, 126 and 102 does not involve any major hydrogen scrambling between the phenyl rings.

    Notes: In standard pulsed ion cyclotron resonance mass spectrometry (ICRMS) mass spectra are generated by converting the time domain signal to the frequency or mass domain by fast Fourier transform (FFT). The FFT requires long acquisition times to acquire high resolution mass spectra. In ICRMS, long acquisition times introduce distortions in the peak intensities of the transformed spectrum that lead to errors in ion abundance. We report here the use of linear prediction methods on data sets collected during much shorter acquisition times to determine actual ion abundances more accurately.

    Notes: A new geometric model has been developed and applied to the deconvolution of ion kinetic-energy release signals observed from fragmentation of metastable ions in the second field-free region of a reversed geometry mass spectrometer. The model takes account of: (i), fragmentations occurring throughout the entire length of the field-free region; (ii), the difference in ion beam intensity before and after the intermediate slit; (iii), the slight difference in angular acceptance between backward- and forward-scattered product ions; (iv), fragmentations of singly and multiply charged ions; and (v), ion kinetic-energy releases of any magnitude.     Notes: Studies on combined supercritical-fluid chromatography (SFC) using a packed column, with fast-atom bombardment mass spectrometry (FAB-MS) are described. The interfacing was successfully carried out by simply coupling a frit interface between the liquid chromatograph (LC) and the mass spectrometer. The interface was equipped with a pneumatic splitter and post-column addition device delivering FAB matrix material to a release valve which maintained a constant back-pressure at the column outlet. Using a 1% glycerol solution as a matrix compound in the release valve, the SFC/FAB-MS combination operates as well as a normal LC/MS instrument. The performance was demonstrated with a mixture of α-, β- and γ-tocopherol and Triton X-100 (poly(ethylene glycol) p-isooctylphenyl ether).

    Notes: A method is described whereby, under software control, the scan function employed to collisionally excite ions in an ion-trap mass spectrometer is regulated so as to obtain parent ion (fixed product) tandem mass spectral data. At the same time, constant neutral-loss information is also provided. Key features are the automatic ‘intelligent’ selection of parent ions from observation of the initial mass spectrum and the scanning of the applied ‘tickle’ frequency in order to locate the precise resonance points for excitation of these ions. Results for the model compounds perfluorotributylamine (‘FC43’) and n-butylbenzene are reported and complicating effects such as the unimolecular decay of the precursor ion species and charge exchange between product ions and the original sample material are discussed.

    Notes: The abuse of nortestosterone in sport is an important problem in doping-control analysis. In order to detect the main urinary metabolite of nortestosterone, norandrosterone (NA), sensitive and specific methodology is necessary. In this context the use of a low-cost mass spectrometric detector such as the Finnigan MAT ion-trap detector (ITD) was studied. The electron ionization (EI) and positive-ion chemical ionization (PICI) mass spectra of the methoxime-trimethylsilyi, trimethylsilyl-enol trimethylsilyl ether and pentafluoropropionic ester derivatives of NA are described. The limits of detection of these derivatives are compared with those obtained by the Hewlett-Packard mass selective detector (MSD), another low-cost mass spectrometric detector and operating only in the EI mode. For the derivatives of the reference standard of NA the ITD has in the EI mode the same limit of detection, in the range of 0.5 to 1 ng injected on the column, as the MSD. However, under these conditions the ITD provides more spectrometric information, because it gives full scan data. Moreover, with the same or even improved limits of detection the ITD can operate in the PICI mode. On the other hand, for the analysis of NA isolated from urine samples, the performance of the MSD was better than that of the ITD. The ion trapping technique is probably limited when the chemical background is high.

    Notes: A combined liquid chromatography/mass spectrometry (LC/MS) method using ion-spray ionization is described for the sensitive determination of okadaic acid and dinophysistoxin-1, the principal toxins implicated in cases of diarrhetic shellfish poisoning. The method is used to confirm the presence of okadaic acid in a culture of the dinoflagellate species Prorocentrum concavum and in natural populations of dinoflagellates from eastern Canadian waters. Several analogues of the toxin have also been detected. Quantitative results are comparable to those obtained by high performance liquid chromatography with fluorescence detection. With slight modifications, the LC/MS method is also used to analyse the fluorescent anthrylmethyl ester of okadaic acid. Preliminary tandem mass spectrometric results, obtained using fast-atom bombardment ionization, are presented for okadaic acid.

    Notes: Negative-ion fast-atom bombardment mass spectrometry was evaluated as a means for quantitative analysis of individual molecular species of glycerophosphatidylserine (GPS). With 200 ng of 1,2-dipalmitoyl-sn-glycero-3-phosphoserine (GPS di 16:0) as an internal standard, the calibration curve for 1-stearoyl-2-oleoyl-sn-glycero-3-phosphoserine (GPS 18:0-18:1) was linear over a range of 50-1000 ng (signal-to-noise ratio 〉4), with a correlation coefficient of 0.998. When the curve was derived from a mixture of GPS molecular species, only a slight variation in the ion intensity ratio of GPS 18:0-18:1 to GPS di 16:0 occurred. The method provides the possibility of quantifying GPS 18:0-18:1 among GPS species in the low nanogram range.

    Notes: Collision-induced dissociation of nitromethane molecular ions to NO+ and NO2+ has been studied at 19 eV center-of-mass collision energy using a crossed-beam tandem mass spectrometer. Center-of-mass velocity contour maps for the two dissociation processes are distinctly different; NO2+ has its intensity maximum at 7 degrees with energy transfer endothermicities of 1.8 eV while NO+ has intensity maxima at zero and 8 degrees with energy transfer endothermicities of 0.3 and 5.5 eV, respectively. The observation of two distinct peaks in the NO+ contour map clearly demonstrates that nitromethane ions excited to 5.5 eV internal energy do not dissociate from the ground state potential surface. Dissociation is faster than intramolecular relaxation, in contradiction to the fundamental postulate of statistical theories (RRKM, QET) for unimolecular dissociation.

    Notes: Evaporation of crystalline LiBF4 was studied by Knudsen-cell mass spectrometry in the temperature range 70-140°C. Only BF3 molecules were identified in the saturated vapour. The temperature dependence of BF3 pressure was measured and ΔrH°374 = 105.4 ± 9.6 kJ mol-1 for the reaction LiBF4(c) = LiF(c) + BF3(g) was obtained from the second-law analysis. The corresponding heat of formation ΔfH° (LiBF4, c, 374) = -1856.4 ± 10.5 kJ mol-1 was evaluated and compared with Literature data.

    Notes: The factors affecting the yield of high-mass molecules by matrix-assisted ultraviolet laser volatilization are examined in a simple model. The key material factors appear to be a low heat of sublimation, subcritical concentration of the guest molecules and a high irradiance input in a short time compared to the sublimation induction period. The model is homogeneous in that the energy density is taken to be uniform within the ‘hot region’ of the matrix. The two competing effects are the rates of energy transfer from the matrix to the guest molecules and the desorption by sublimation. It is the bottleneck for energy transfer to the embedded guest molecules that makes their energy content lag behind that of the matrix. This is particularly the case for an initially cold sample. When a sufficiently high rate of sublimation can be achieved (e.g., using a high-power laser), the guest molecules (or adduct ions) will desorb internally cold and will thus not fragment. Numerical simulations of the sublimation kinetics using realistic laser and material parameters support the conclusions and delineate the ranges of the critical factors.

In heterodyne mode FT-ICR mass spectrometry, using quadrature reference signals, a complex ICR signal consisting of two quadrature reference signals, a complex ICR signal consisting of two quadrature signal components can be acquired easily. By Fourier transform of the signal, the reflection of peaks about the Nyquist frequency in the frequency-domain spectrum can be eliminated. As a result, the available bandwidth of the spectrum is doubled. Validity of this quadrature heterodyne method is confirmed by the experimentally obtained spectrum of Xe+ ions.

    Notes: Thermospray ionization with repeller-induced collisionally activated dissociation (repeller-CAD) has been used to acquire mass spectra of a series of dyes. These were obtained by applying a voltage of 400 V on the wire repeller. The mass spectra contained a large number of fragment ions which were useful for structural elucidation. Some of these fragment ions were found to be similar to those obtained by thermospray tandem mass spectrometry with CAD and some of them were similar to those obtained by electron ionization using a particle beam liquid chromatograph/mass spectrometry interface. The results indicate that more than one mechanism is involved in the fragmentation process.

    Notes: Mechanisms related to high-mass ion detection were studied using matrix-assisted UV laser desorption for ion generation and a tandem (MS/MS) time-of-flight mass spectrometer for the analysis of secondary species formed at the conversion dynodes. Secondary-electron emission was found to be less pronounced with increasing molecular weight. Instead small secondary ions generated by collision of primary ions with the conversion dynodes appeared to be the dominant process for the detection of ions of masses up to several hundred thousand Da. The use of microchannel plate detectors has been shown to become less effective for high molecular weight applications. Mass spectra of secondary ions emitted from a conversion dynode bombarded with keV primary ions have been recorded. Regardless of the composition of the primary molecules, the secondary mass spectra were similar. Sputtering of surface adsorbates is assumed to be the major mechanism of secondary ion formation, rather than surface induced dissociation or fragmentation of the primary ions.

    Notes: Variously substituted aryl ketones served as models to gain insight into the collisional-activation process in an ion-trap mass spectrometer. The effects of different targets (helium, argon) at different pressures were examined. The investigation also involved the comparison of the results of tandem mass spectrometry experiments in the ion trap with those obtained in high-energy collision and mass-analysed ion kinetic energy experiments in a sector instrument. Use of argon and low helium pressures in the ion trap results in both an increase of the average internal energy of the activated ion and, presumably, a less broad energy deposition profile. The results indicate that the maximum value of internal energy achieved by an ion through multiple collisions in the ion-trap mass spectrometer is limited by the presence of low-energy reaction channels.

    Notes: The mass spectral fragmentation of 2-adamantanone has been investigated using its specifically labelled monodeuterated derivatives (in positions 1-, 4-exo-, 4-endo-, 5- and 6-) and dideuterated derivatives (in positions 4,4- and 6,6-). The mass spectrum of 2-adamantanone is generally poor in fragments showing the molecular ion as the base peak, the characteristic m/z 117, 104, 93, 91, 80, 79, 72, 67 and 53 fragments, as well as an unusual loss of water. The mass spectra of deuterated compounds favours the assumption of hydrogen randomization prior to the fragmentation of the molecular ion. In contrast, the formation of [C4H8O]+·, m/z 72 does not involve any major hydrogen scrambling.

Extracts obtained from Sedum acre L. were investigated for the presence of alkaloids by gas chromatography/mass spectrometry. Identification has been carried out on the basis of retention time, molecular ion mass and characteristic fragmentation. Investigated extracts contained unknown alkaloids which belong to the piperidine group of sedum alkaloids. Mass spectral fragmentation patterns and a proposed structure for the unknown alkaloids are discussed.

    Notes: Experimental variables affecting the daughter-ion spectra of a series of protonated peptides [MH]+, produced by fast-atom bombardment ionization, using a low energy (0-450 eV) quadruple collision cell are investigated. The parameters studied include target gas pressure, collision energy, cross-sectional area and acidity of the target gas. The results show that low-mass immonium ions are preferentially formed both at high collision energies (〉 200 eV) and at target gas pressures 〉 10-6 mBar (where multiple collisions occur in the gas cell). Positive fragment ion abundance is maximized when acidic gases are used as the target gases, and this is rationalized on the basis of a proton-transfer reaction from the target gas to the amide nitrogen of the peptide bond promoting fragmentation.

    Notes: Endothermic reactions between leucine enkephalin and ammonia have been carried out at very low beam energies, and compared with activation obtained by conventional collision-induced dissociation at both low and high energies. The similarity between the high-energy and the reactive-collisions spectra suggest that comparable energies are imparted by these two methods. The upper limit of the collision energy deposited by reactive collisions is only 0.30 eV. In addition, a substantial increase in the collection efficiency is observed with endothermic reactions, with the sum of the fragment ions exceeding the intensity of the surviving parent ions.

    Notes: Data are presented showing the use of thermospray ionization to generate multiply charged ions for a number of proteins over the molecular weight range 5000-30000. These measurements were carried out on an unmodified, commercially available thermospray ion source. Between +3 and +19 charges were observed for a series of model proteins. Data can be obtained on as little as 100 pmol of sample. The accuracy and precision of the molecular weight determinations for these proteins is similar to those previously reported for these samples using electrospray or ionspray ionization. The formation of multiply charged ions from proteins via thermospray ionization occurs over a wide range of aqueous/organic solvent compositions, facilitating its interfacing with high-performance liquid chromatography using conventional diameter columns.

    Notes: The possibility of performing matrix-assisted laser desorption experiments in transmission geometry is demonstrated for two neuropeptides (substance P and bombesin), for six analogs of the melanocyte-stimulating hormone (MSH) core and for coliagenase enzyme substrates. Positive- and negative-ion spectra of several peptides are produced without the presence of a metallic substrate. Cationized quasi-molecular ions are abundant in the positive spectra. Peak broadening in the high-mass range can be the consequence of overlapping molecular and adduct ions. The presence of synthesis by-products can be identified readily from the spectra. Ultimately, picogram detection limits are possible for important bioactive peptides and other large molecules. Because of the clearly demonstrated matrix-assisted laser ionization in a homogeneous environment, metal substrate participation in the volatilization mechanism seems less likely.

    Notes: This paper reports the application of matrix-assisted UV-laser induced desorption (LID) mass spectrometry for the analysis of some extracellular multi-domain cellulolytic enzymes, in the molecular weight range 5000-90 000 u, produced by the fungi Phanerochaete chrysosporium and Trichoderma reesei. A high sensitivity (1-10pmol of protein), has been attained in obtaining the mass spectra of these highly glycosylated enzymes and some of their peptides, with carbohydrate content in the 10-40% range. The desorption of samples containing the enzymes mixed with a protein having an accurately known molecular weight - an internal calibrant - is also demonstrated. Spectra from complete enzymes and their functional domains are presented to explore the possibilities of employing LID for obtaining structural information.

    Notes: Three hydroxyindole isomers were investigated by conventional electron ionization (EI) mass spectrometry and by collision-activation (CA) experiments. Although the EI mass spectra of the three isomers are virtually superimposable, 3-hydroxyindole can easily be differentiated from the others by single-energy collision spectroscopy. Only by energy-resolved mass spectrometry (ERMS) could the characterization of the 4- and 5- hydroxyindole isomers be achieved. ERMS, which in the ion-trap mass spectrometer can be performed by different methods, was achieved by changing both the AC supplementary voltage and the βz value at which the collision experiments were performed. The first method proved to be the most effective in the present case. Further collision experiments, carried out at a range of AC voltage durations did not yield energy-resolved data.

    Notes: Positive-ion time-of-flight mass spectra of sodium salts of oligodeoxyribonucleic acids (DNA), and a protein, lysozyme, have been obtained using pulsed-laser ablation and ionization of frozen aqueous solutions from a cooled copper substrate. The efficiency of positive-ion formation was found to increase when the ablating laser was tuned to a frequency corresponding to resonant electronic transitions in sodium or copper atoms. Molecular ions of double-stranded oligomeric nucleic acids, with little fragmentation, were observed at masses up to ∼18 500 Da. Single-stranded nucleic acids could be fragmented by lowering the laser power-density to reduce the the substrate heating rate. A dimer ion of lysozyme, at mass ∼29 000 Da, was detected at an impact energy on the electron multiplier of only 3.5 keV.

    Notes: Field-desorption mass spectrometry has been used to characterize polystyrene oligomers of molecular weight up to 15 000 Da. The technique shows good agreement with molecular weight data calculated using vapour-phase osmometry and gel-permeation chromatography but in addition provides much more detailed information in terms of oligomer distribution and the nature of end groups present in the oligomers.

    Notes: Radical adducts of 5,5-dimethyl-1-pyrroline N-oxide (DMPO) with hydroxyl, methanol-derived, and ethanol-derived radicals were detected by a combination of liquid chromatography with either electron paramagnetic resonance or thermospray mass spectrometry (LC/EPR or LC/TSP-MS) in the Fenton system (with methanol or ethanol). One radical adduct was observed in the reaction of DMPO with the hydroxyl radical or the methanol-derived radical, while two adducts were detected in the reaction of DMPO with ethanol-derived radicals. The LC/TSP-MS spectra showed quasi-molecular ions [M+H]+ at m/z 146 and m/z 160 for the methanol-derived and ethanol-derived radical adducts, respectively, and an apparent molecular ion M+ at m/z 130 for the hydroxyl radical adduct. Use of methyl-D3 alcohol (CD3OH) and ethyl-D5 alcohol (CD3CD2OH) indicated that carbon-centered radicals are formed. Experiments with partially deuterated ethanol (CD3CH2OH and CH3CD2OH) indicated that the two adducts observed in the reaction of DMPO with ethanol-derived radicals correspond to the two diastereomeric adducts of DMPO with the α-hydroxyethyl free radical.

    Notes: The major objective of this investigation was to find a thermospray buffer that would decrease ion/molecule reactions in the gas phase. This would cause solution chemistry and ion evaporation to be major parameters controlling ion production. Using triethylammonium formate, protonation by ion/molecule reaction in the gas phase was decreased as indicated by the effect of solution pH on ion production. The detection limits in the full-scan mode for some amphoteric compounds and acids were one magnitude lower than those obtained with ammonium acetate or ammonium formate buffers. A few examples of sensitive full-scan detection demonstrate the usefulness of this novel buffer for negative-ion thermospray liquid chromatography/mass spectrometry.

    Notes: Unimolecular dissociations of metastable methylsilyl cations CH3SiH3+ are investigated by mass-analysed ion kinetic energy (MIKE) spectrometry. The predominant fragmentation is H2 loss which proceeds via at least two distinct mechanisms with substantially different kinetic energy releases. Their reaction mechanisms are inferred from the results of experiments and MNDO calculations. The MIKE spectrum of the deuterium-labelled ion, CH3SiD2+ indicates that the H and D atoms are randomized before dissociation.

    Notes: A tandem mass spectrometer equipped with the pneumatically-assisted electrospray (ion spray) liquid chromatography/mass spectrometry interface and an atmospheric-pressure ionization source was used for characterization of underivatized cyclic oligosaccharides. Protonated singly charged molecular ions were generated via ion-evaporation mechanism(s). The formation of doubly charged molecular ions from the model compounds appears to be related to the channel diameter of these cyclic oligosaccharides. Tandem mass spectrometry (MS/MS) was used to study collision-induced dissociation of the protonated molecule and showed exclusive cleavage of the glycosidic bond. The molecular weights of these oligosaccharides as well as the number of sugar units were confirmed by MS/MS. The potential application of MS/MS for target compound analysis was demonstrated by the parent-ion scan method with a synthetic mixture.
