    Notes: In order to investigate whether genetic factors are involved in the response of schizophrenic patients to treatment with γ-type endorphins, we typed 32 Dutch schizophrenic patients for the HLA-A, -B, -C and -DR antigens. The total patient group showed an increase of HLA-Bw4 and HLA-Cw1. A subgroup of 20 paranoid patients showed an increase of HLA-Cw1 and a significant heterogeneity for the HLA-C locus. In 16 patients who responded moderately or markedly to treatment with γ-type endorphins, an increase of HLA-B15/Cw3 and a decrease of HLA-B17 were found as compared to 16 patients with no or a slight response. Moreover, HLA-B15 was particularly increased in those patients who responded markedly and remained free of psychotic symptoms for a period of at least 6 months after treatment with γ-type endorphins (RR = 24.6, Puncorr.= 0.0015).Our results suggest that genetic factors coded for within the HLA region are associated with paranoid schizophrenia, and that HLA-B15/Cw3 is associated with a marked and prolonged response to treatment with γ-type endorphins.

    Notes: We have recently described a murine lymphocyte alloantigen, Ly-21.2, and showed that the gene which controls Ly-21.2 expression is linked to albinism on the 7th chromosome. In order to map the gene more precisely, we have studied the linkage of Ly-21.2 with two other genetic markers on the 7th chromosome, the electrophoretic variant of the β chain of haemoglobin and glucose phosphate isomerase, in (A/J × C57BL/10)F2 mice. The results of these 3-point cross studies show that the gene encoding Ly-21.2 maps 27 units distal to albinism.

    Notes: The identity, or close linkage, or a series of murine alloantigens encoded by the Ly-6 genetic region has been well documented. We have investigated the relationship between two component antigens, Ly-6.2 and H9/25, by generating and characterizing a number of variant cell lines which have altered antigen expression, isolated by chemical mutagenesis and immunoselection of BW5147 lymphoid cells. Cloned cell lines, selected by either antiserum, show loss of both antigens and also fail to express Thy-1, a further genetically linked, but unrelated determinant. The relevance of these findings to furthering understanding of the structure of the Ly-6 region and regulation of antigen expression is discussed.

    Notes: Digestion of bovine erythrocytes (BE) with trypsin (Tr) or chymotrypsin (CTr) revealed Tr and CTr specific' receptor respectively. These receptor reacted with all the isologous, including the autologous, sera. The titres of agglutinability varied for the Tr-digested BE from 1:2 to 1:64 and from zero to 1:128 for the CTr-digested BE. However, the variation between the members of nomozygous twin pairs (MZ) in no case exceeded ±2 score units, the error limits. Agglutination of the CTr-digested BE gave with the PHA lection very similar, (r= 0.964) results to that which was ontained with the isologous sera.The titre of agglutinins showed concordance in all the sera of MZ pairs when tested against the neuraminidas-digested human erythrocytes and the Tr or CTr-digested BE. Their variance due to differences varied between MZ pairs from 60.4 to 74.3%.

    Notes: Murine alloantisera to lymphocyte (Ly) alloantigens also contain antibodies to murine leukaemia viruses (MuLV) in high titre. A number of such sera were extensively absorbed with ecotropic AKR murine leukaemia virus to remove contaqiminating anti-MuLV antibodies. The virus-absorbed and unabsorbed antisera wre then examined by the radioimmune precipitation (RIP) assay for anti-MuLV antibodies, and for anti-Ly antibodies by cytotoxicity assays against specific target cells. The results presented here demonstrate that whilst virus absorption markdely reduced the anti-viraql titres of the alloantisera they ahd no effect on the anti-Ly titres of any of them. This indicated that none of the Ly antigens which was examined cross reacts with the antigens of ecotropic AKR-MuLV and further suggest that Ly antigens are not encoded by the genes of endogenous ecotropic MuLVs. It was also demonstrated that the majority of the constaninating anti-MuLV antibodies present in anti-Ly-6.2 proteins of the virus.

    Notes: The MHC restriction criteria for T cells activating macrophages in vivo and mediating antimicrobiol resistance to Listeria monocytogenes were determined. Antimicribiol resistance could be transferred by T cell in order of decreasing efficiency from syngeneic. RT1A. compatible, RTI.B compatible and RT1 incompatible donors. Alloreactive T cells responding to either A locus or B locus encoded antigens in a graft-versus-host reaction were also able to activate macrophages. Approximately five times as many MLC-reactive precursors responded to B locus alloantigens as to A locus alloantigens, but A-resticted Listeria-specific T cells wre considerably more numerous (or more efficient) in Listeriainfected hosts than were B-restricted, Listeria-specific T cells. This was unexpected, since A-restricted, Listeria-specific T cells failed to transfer delayed hypersensitivity (DTH) to soluble bacterial antigens.

    Notes: This study nvestigates the dicordance between the restriction criteria required for the transfer of cellular resistance to Listeria monocytogenes (LM) and those for the transfer of delayed type hypersensitivity to Listeria antigens. Infective bacteria elicity both RTl.A-restricted T cells and RT1.B.-restricted T cells. Both populations of t cells mediate lymphoblast localization and macrophage accumulation, which are reactions characteristic of delayed type hypersensitivity (DTH), and cause macrophage activation with rapid and efficient bacterial elimination, which is an expression of cellular resistance. If alcohol-killed Listeria organisms (pLMA) are injected, only the RTl.B-restricted T cell subset is triggered. Direct comparison of lymphoblast localization in LM infection sites and the expression of resistance of resistance revealed that efficient resistance may be mediated by small numbers of lymphoblasts and that below a certain threshold there is no correlation between lymphoblast localization and the level of resistance.

    Notes: Four sequential polypeptides containing equimolar amounts of tyrosine, glutamic acid, alanine and glycine were shown to be T lymphocyte-dependent immunogens in inbred guinea-pigs. Poly (Glu-Ala-Tyr-Gly) was immunogenic only in strain 2 guinea-pigs; poly (Glu-Tyr-Ala-Gly) and poly (Ala-Tyr-Glu-Gly) were immunogenic only in strain 13 guinea-pigs; and poly (Ala-Glu-Tyr-Gly) was immunogenic in both inbred strains. The specificity of immune recognition was probed at the T lymphocyte and humoral levels with the heterologous polypepties. Only a few cases of heterologous cross-stimulation or cross-reaction were observed, indcating the great selectivity of immune recognition. The results showed considerable variability in immune recognition from animal to animal. Nevertheless, at the T-cell level, cross-stimulation appears to necessitate that the antigen is itself immunogenic in that strain, whereas at the antibody level, cross-reaction is not similarly restricted. The structural basis for mutual recognition of immunogen and antigen at these levels is discussed.

    Notes: The topographical relationship of Thy-1, Ly-1, Ly-2, and T200 were examined on the murine thymocyte. The inhibition of the binding of radiolabelled monoclonal antibody after incubation with unlabelled heterologous antibody was used as a measure of the proximity of the target antigens on the cell surface. On unfixed cells, prior incubation with anti-Thy-1.2 impeded the attachment of labelled anti-Ly-1, anti-Ly-2, and anti-T200. Anti-T200, but not anti-Ly-1, or anti-Ly-2, was capable of impeding the attachment of labelled anti-Thy-1.2 on unfixed cells. In general, gentle fixation with paraformaldehyde did not alter these inter-relationships. The possible functional contributions of such supramolecular relationships are discussed.

    Notes: Two examples of mouse monoclonal anti-N are described. The antibodies were derived from mice immunized with sialoglycoprotein extracts of group O MM ss erythrocyte membranes and the probable stimulus for immunization was glycophorin B associated N-antigen. Both antibodies reacted as direct agglutinins but appeared to recognize different epitopes with one having a greater dependence on sialic acid. The antibodies could prove to be valuable alternatives to those reagents used currently for N-blood grouping.

    Notes: Heterodontus francisci (horned shark) and Pseudopleuronectes americanus (winter flounder) were immunized with furyl-oxazolone (furyl-Ox) and phenyl-oxazolone (phenyl-Ox) coupled either to bacteria or protein carriers. The antibodies produced were measured by inactivation of furyl- or phenyl-Ox conjugated bacteriophage, and their affinity and fine specificity were estimated by inhibition of phage inactivation with a series of structurally related hapten analogues. In both species, post-immunization peak titres were 100 to 2000 times higher than preimmunization titres.A number of unique features distinguished Heterodontus antibodies from Pseudopleuronectes or mammalian antibodies. Heterondontus antibodies exhibited a lower affinity for the immunizing hapten (furyl-Ox or phenyl-Ox) and a reduced ability to distinguish the homologous immunogenic hapten from its structural analogues. In addition, Heterodontus antibodies exhibited a lower level of inter-individual variation in affinity and fine specificity than did Pseudopleuronectes or mammalian IgM antibodies; this was especially prominent in anti-furyl-Ox responses. Typically the affinity and fine specificity of Heterodontus antibodies did not change over the 146-day period of immunization and were not influenced by the nature of the carrier. The implications of these findings in terms of the phylogenetic origins of antibody diversity are discussed.

    Notes: The precise physicochemical conditions under which two examples of mouse monoclonal anti-N can be used as blood grouping reagents have been defined. The antibodies were shown to belong to the IgG1 and IgG2b subclasses respectively and both reacted within discrete ranges of temperature and pH with optimal reactions occurring at 20d̀C or less and pH 8.5. Under these conditions and at concentrations of 2-3 μg/ml, the antibodies were used, in parallel with conventional polyclonal antisera, to type a series of random blood donors. The results obtained with one of the monoclonal antibodies and the polyclonal reagents were in perfect agreement and the monoclonal antibody was judged to have considerable potential as an N-grouping reagent.

    Notes: We typed coded sera from 135 healthy controls, seventy-six patients with autoimmne goitrous and seventy-three with atrophic thyroiditis for IgG heavy chain markers (Gm). All subjects were Caucasian from Newfoundland. An increase in the Gm phenotype ag was found in the 149 patients with thyroiditis compared to controls (X12= 5.82, P 〈0.01); significance was, however, not maintained after correction for the number of variables tested. The difference in ag phenotype was more pronounced among the seventy-three patients with atrophic thyroiditis (X12= 8.80 corrected P 〈 0.05). Because the haplotype ag was not significantly increased in this group, we conclude that homozygotes for Gm ag are at an increased risk of developing atrophic thyroiditis.

    Notes: A fourth human blood group chimaera studied in Birmingham is an example of haemopoietic (twin) chimaerism in which the subject was unaware of being a twin. Chimarerism was discovered during routine antenatal serological investigation in which it was shown that the proposita has two red cell populations, one of the rhesus genotype rr, and the other R1r. Further studies showed that she has two populations of lymphocytes, one with the female karyotye, 46XX, and the other with the male karyotype, 46XY. Skin fibroblasts were all 46XX.

    Notes: By crossing and back-crossing three strains of rats, Brown-Norway, Fischer, and PVG, none of which is definitely susceptible to experimental autoimmune encephalomyelitis (EAE), it is possible to obtain fully susceptible animals. Genetic analysis indicates the presence of Ir-genes in at least two of the strains (Fisher and PVG) related to the AgB complex, and genes of ‘resistance’ located on another chromosome. The latter appear to be different in the three strains; also the BN strain, which perhaps does not carry an Ir-EAE gene, apparently carries effective genes of resistance.

    Notes: A new, non-MHC linked alloantigenic membrane antigen on the equine lymphocytes is described. This antigen was characterized with alloantisera in the two-stage microcytotoxicity test and designated as ELy-1 antigen.The frequency of ELy-1 antigen positive animals in various populations is close to 50%. ELy-1 shows an autosomal, dominant inheritance. Since an allelic antigen (s) could not be demonstrated in family studies, it is assumed that only two alleles ELy-I+ and ELy-I- exist. The ELy-1 antigen in positive animals is expressed on both T and B lymphocytes but it is not present on erythrocytes and thrombocytes.The incidence of ELy-1 antigen positive animals is significantly higher in horse groups suffering from chronic bronchitis or from laminitis, compared to clinically healthy controls.

    Notes: Previously, it was reported that the immune response to myoglobin (Mb) was under genetic control, with the response to each site being under separate Ir-gene control. Here we have investigated the effect of antigen dose on the control of the antibody response to the five antigenic sites of sperm-whale Mb to determine whether or not the overcoming of genetic control by antigen dose has a uniform effect on all five antigenic sites.The antibody response to sperm whale myoglobin (Mb) and its five antigenic was measured in the following inbred strains of mice, C57BL/6J, AKR and SWR/J. These strains of mice are low responders to Mb following immunization with 50 μg, responding only to site 4. After immunization with 200 μg Mb:C57BL/6J mice are high responders to Mb and respond to antigenic sites 1, 3,4 and 5; AKR mice are high responders to Mb and respond to antigenic sites 1 and 4; SWR/J mice are high responders to Mb and respond to all five antigenic sites. It was concluded that the genetic control of the immune response to Mb and its synthetic antigenic sites is dependent on antigen dose. Also, these studies have enabled us for the first time to separate the response to site 1 from the response to site 2 and thus have conclusively established that sites 1 and 2 are controlled by separate Ir-genes.

    Notes: Immnoglobulin allotypes (Gm) were analysed in 40 multiple sclerois (MS) patients and the distribution of phenotypes compared to that in 1220 healthy controls. The frequencies of Gm(1) and Gm(1, 2) are significantly increased in the patietns suggesting that the presence of the Gm3;5,13,14 haplotype confers a resistance to the development of MS.

    Notes: Simian B and T lymphoid cell lines were shown to maintain surface markers found on mature lymphocytes in vivo. The T lymphoid cell lines expressed Ia-like antigens on their surfaces, further suggesting that they represent mature, activated T cells. These Ia antigens show a structural similarity to Ia on human cells although some diversity exists. The Ia antigen expressed on T lymphoid cell lines was shown to be very similar to those on B lymphoid cell lines. Owl monkey and marmoset T lymphoid cell lines were also shown to express a VH immunoglobulin-related determinant, a marker which is thought to be associated with T cell antigen receptor. Owl monkey and marmoset T cell lines express a surface antigen which identifies the sheep erythrocyte receptor on human T cells and some of these lines express an antigen found on human helper T cells. It is noteworthy that substantial conservation of surface components has occurred within primate evolution such that monoclonal antibodies to human Ia, OKT-11a and Leu 3a markers can be used to type lymphocytes of lower primates.

    Notes: Immunogenetics of the Basilea allotype were investigated. It was shown that the Basilea gene is controlled at a locus identical with, or closely linked to, the Ab kappa allotypic locus. Basilea-positive molecules have been physically separated by immunoabsorption from molecules carrying the λ chain markers, c7 and c21. Injection of newborn Basilea homozygotes with anti-Basilea immune serum results in suppression of the synthesis of Basilea-positive molecules, but does not affect the synthesis of the λ chain, or of the heavy chain allotypic markers. Allotype-suppressed Basilea homozygotes do not recognize either the Basilea allotype or the kappa isotype as ‘self’ and make anti-Basilea and anti-kappa antibody upon immunization with Basilea antigen. The specificity of this antibody was identical with that of a heterologous anti-kappa immune serum raised in a goat and made specific by absorption with purified lambda material. These results prove that Basilea gene products are of kappa isotype and are controlled at the Ab allotypic locus.

    Notes: The B6-C.H-2bm12 (bm12) strain has previously been described as a spontaneously occurring mutant wherein the parental C57BL/6 Iab specificities (Ia.3, 8, 9, 15 and 20) are absent or reduced in amount. To further characterize the bm12 mutant the antigenic phenotype of LPS-activated bm12 blast cells was determined with monoclonal and conventional anti-Ia antibodies. By direct testing and absorption the public Ia.8, 9 and 15 specificities were readily detected on both parental C57BL/6 and mutant bm12 blast cells, and the amounts of these antigens on activated bm12 cells were similar to or less than those found on C57BL/6 blast cells. The findings demonstrate that the Ia-1 mutation has not resulted in an absolute loss of these specificities, but has altered their expression, or their accessibility to antibody on resting cells. By contrast, the private Ia. 20 specificity and another specificity (designated Iab) could not be detected on bm12 blast cells.

    Notes: Data presented here defines the map of H-2 associated genes which affect the glucocorticoid-induced cleft palate frequency to the regions H-2K to I-B and G to H-2D. This was done by observing in four congenic strains the frequency of cleft palate induced by 160 mg/kg of dexamethasone administered to pregnant females on day 12 of gestation. The strains used were B10.A/SgSn, C57BL/10Sn, B10.A(5R)/SgSn and B10.A(18R)/Sg. Additionally the cleft palate frequency was observed in a large number of litters from saline-treated pregnant females. The cleft palate frequency in this control was very low and the rank of strains was the same as for the glucocorticoid-induced cleft palate frequency. This observation suggests that the induced cleft palate frequency reflects the spontaneous occurrence of isolated cleft palate in these strains.

    Notes: Experiments were conducted to investigate the non-H-2 genetic effects on experimental autoimmune thyroiditis (EAT). Strains having C3H or BALB backgroud in general produced higher autoimmune responses to mouse thyroglobulin (MTg) than the B10 or A strains. Comparisons of C3H and B10 congenic strains carrying similar H-2 haplotypes demonstrated that the C3H congenics had significantly higher MTg antibody titres and more severe thyroid damage, even when the strains carry the low responder H-2 haplotypes. These observations show that non-H-2 gene(s) influences EAT, in addition to genes in the MHC.

    Notes: Age-dependent changes, during adult life, in suppressor capacity and B cell responsiveness have been known for some time. In this paper, an age dependent change in helper function, is being reported. A Th-2 type of helper effect can be observed in tissue culture of spleen cells from animals sensitized with a macromolecule. An indirect plaque-forming response to the hapten occurs when the spleen cells are exposed to the sensitizing macromolecule and to the hapten conjugated to a carrier, structurally unrelated to the sensitizing macromolecule. There is considerable polymorphism in the age at which this effect is first demonstrable and at which it reaches mature levels. Mice of inbred strains, with various defects in the suppressor cell circuit (SJL/J, MRL/MpJ-lpr/lpr, BXSB/MpJ) show an accelerated development of Th-2 helper capacity in the indirect plaque-forming response.

    Notes: BALB/cAnN and C3H/HeN mice were primed and challenged with DNP-Ficoll, a thymic independent (TI-2) antigen. They developed significant carrier-specific IgG memory responses: DNP-pneumococcal polysaccharide or DNP-hemocyanin challange did not elicit memory responses.     Notes: D-25 is a H-2 alloantiserum produced in (B10.D2 x C3H.NB) (H-2d x H-2p)F1 mice after immunization with B10.RIII(H-2r) cells, and which is known to recognize the H-2.25 public antigen on H-2k haplotypes. The ‘anomalous’ reaction of D-25 with a partially purified deoxycholate-solubilized glycoprotein of B10-1 (H-2b) fibrosarcoma was studied with various biochemical techniques. The 125I-labelled precipitates were analysed both in one- and two-dimensional SDS-PAGE and by a partial proteolysis peptide mapping (Cleveland's mapping). The results indicate first that the D-25-related antigen was a genuine H-2 antigen normally associated with the B2-microglobulin, second that this antigen was borne by the Kb but not Db gene products, and finally that D-25 was able to precipitate the same antigen on normal H-2b spleen cells. We conclude that this serum recognized a normal H-2 specificity shared by H-2b and H-2r haplotypes and tentatively identified as the public antigen H-2.54 which for the first time could be assigned to the K but not to the D region of the H-2b haplotype.

    Notes: The study of the distribution of IgG subclasses in a group of Iraqi Arabs has revealed marked elevation in the levels of IgG3 associated with low levels of IgG1. Furthermore, individuals homozygous for the G3m(b) marker were found to have significantly higher levels of IgG3 than individuals heterozygous for this marker. Results are discussed in relation to other similar reports.

    Notes: Among 290 F2 progeny of an r10 x ACP cross were two recombinants which allowed the loci for glyoxalase-1 and neuraminidase-1 to be mapped relative to the RT1.A and dw-3 loci in the major histocompatibility complex (MHC) of the rat. In 673 progeny of the same cross there was a recombinant between ft and dw-3, and in 403 progeny of the backcross BY1 x (BY1 x BDIX)F1 there was another recombinant between ft and dw-3. These data, combined with those from previous studies, provide the information for constructing a detailed map of the rat major histocompatibility complex: the gene order and size in the rat are very similar to those in the mouse and different from those in man and in the other species that have been studied. Comparison of the structures of the MHC in the various species leads to a hypothesis about the evolution of the MHC which involves sequential duplications of the genes coding for class I and class II loci and an inversion in the prototypic muridae which placed the class II loci between the class I loci.

    Notes: The following attributes of the immune response were studied from nine patients with different numbers of X chromosomes: serum immunoglobulin levels, C3 and C4 concentrations, the presence of autoantibodies, phagocytosis, killing of bacteria, the chemotactic response of neutrophils. the in vitro response of lymphocytes to PHA, ConA, PwM, PPD and oidiomycin; and the proportion of T lymphocytes was determined. The number of X chromosomes varied from one (45,X) to four (48,XXXX). No evidence of severe dysfunction in the immune system was found in any of the patients. The correlation coefficients between the serum concentration of IgM and IgG and the number of X chromosomes present were statistically significant (r = 0.691, P 〈 0.05, and r = 0.714, P 〈 0.05, respectively). The serum IgA concentration showed a tendency towards a negative correlation. The concentrations of neither IgD nor IgE correlated to the number of X chromosomes. It seems obvious that the number of X chromosomes is positively associated with the concentration of serum IgG and IgM. The results support the hypothesis that antibody production is at least partially controlled by genes located in the X chromosome.

    Notes: A group AB mother (Mrs P.D.) gave birth to a group O female baby (C.D.). Extensive study of the blood group genetic markers in both the parents and the child, carried out on several occasions, showed nothing unusual outside the ABO system. Mrs P.D. then, gave birth to a second female baby who was also group O. Mrs P.D. had normal amounts of A, B, H and Lewis antigens in her saliva. The H, A and B agglutinability of her red cells was in the range of normal A2B group. This A2B blood group was characterized by very low A gene-specified glycosyltransferase activity in serum. Moreover this activity was undettable in red blood cell membranes. These results are discussed in the light of various hypotheses in order to explain this unusual transmission of ABO blood group.

    Notes: Because of the inconsistency in published results concerning the serological detection of cell surface antigens coded for by the t-complex, a cell-mediated lymphocytotoxicity (CML) assay, secondary CML, was used in a search for t-antigens. By sensitizing C3H. Ttf (C3H. Brachyury, tufted) with the congenic strain C3H. Ttf/tw18 splenic cells, a response against lipopolysaccharide (LPS) stimulated splenic cells from C3H. Ttf/tw18 mice is obtained. The locus coding for the antigen detected by this reaction lies to the left of tf on the murine seventeenth chromosome. The secondary CML response to this antigen is H-2 restricted and detects an antigen on all t-haplotypes tested: tw18, tw18tf, t12, t6, th2tf, and tw5.

    Notes: Four OLA factors of the ovine histocompatibility complex and the OLX-5 factor were recognized in hamster cell x sheep fibroblast hybrids, by means of absorption of OLA reagents with hybrid cells. Segregation of these factors could be studied in eighteen independent hybrids. In the case described, the four OLA factors were distributed into two haplotypes isolated in some hybrids. The heterozygous OLX-5 factor was linked to only one haplotype; nevertheless a dissociation, occurred in one hybrid (relative to five simultaneous transmissions), is in agreement with a previous genetic study showing a loose linkage between the OLA complex and the OLX locus.

    Notes: Selective breedings of mice were carried out for quantitative antibody responsiveness to flagellar Ag., f (Selection III) or somatic Ag., s (Selection IV) of two non cross-reaction Salmonellae (Salm. tm., Salm. or.) alternated for immunization of consecutive generations. At the selection limit, these selections produced homozygous high (H) and low (L) responder lines for the character investigated: peak agglutinin response to optimal secondary immunization. The responsiveness to both f and s Ags. is submitted to polygenic regulation. The heritability (h2) realized during the selective breeding was 0.37 ± 0.07 for the response to f Ag. and 0.40 ± 0.1 for the response to s Ag. The respective part of genetic and environmental variance in F2 hybrids was 64% and 36% in selection III and 61% and 39% in selection IV. In the two selections, the dominance variance is negligible (〈1%), therefore the genetic variance is essentially additive. The additive variance calculated as the heritable fraction of the F2 hybrid variance is somewhat lower, the reason for this difference is discussed. The quantitative antibody response to f Ag. in selection III is controlled by about seven independent loci. The antibody response to s Ag. in selection IV is controlled by about four independent loci. A possible association of relevant genes with the H-2 locus was investigated. In selection III, no significant participation of H-2 linked genes, in the regulation of responses to f and s Ags. of Salm, tm and Salm. or. could be demonstrated. In selection IV a partial contribution of H-2 linked genes was observed concerning responsiveness to both f and s Ags. of Salm. tm.. but not to Salm. or. Ags. The H-2 effect accounts for 25% of the total interline difference.

    Notes: A monoclonal antibody was produced which binds specifically to Type 2 H antigen of the ABO blood group system. The antibody, H11, is an IgM molecule which reacts by direct agglutination of red cells with the same pattern as other anti-H reagents such as Ulex europaeus lectin. The specificity was determined by inhibition and adsorption with chemically defined oligosaccharides. H11 also reacted with poly(glycosyl)ceramide purified from group O red cells and glycoprotein H purified from human stomach mucosa and meconium. H11 differs from Ulex lectin, however, in that it was not inhibited by saliva from ABH secretor individuals or by glycoprotein H purified from human ovarian cysts or submaxillary glands.

    Notes: The number of cells in preimplantation mouse embryos of different H-2 haplotypes was analysed. It was found that embryos of the H-2k haplotype have fewer cells per embryo than those of the H-2b haplotype. By analysing reciprocal congenic pairs of mice it was demonstrated unequivocally that slow development is linked to the H-2k haplotype and fast development to the H-2b haplotype. The gene(s) in the H-2 complex which influence early mouse embryo development have been named Ped: preimplantation embryo development. Analysis of F1 hybrid embryos showed that fast development is dominant. Reciprocal F1 crosses yielded identical results, which indicated there was no apparent effect of the maternal egg cytoplasm on Ped gene expression. Analysis of F2 and back-cross embryos was consistent with the interpretation that there is a major gene located in the H-2 complex (Ped), which is modified by environment and genetic background, that influences early mouse embryo development.

    Notes: Allophenic mice were produced by aggregating 8-cell embryos from a low responder (CBA) and a high responder (DBA/1) strain to the synthetic amino acid polymer GLø. In some strains of mice, the immune response to this polymer is controlled by two genes, α and β, which map to the IE/C and IA subregions of the H-2 complex, respectively. The CBA strain is α+β - whereas the DBA/1 strain can be considered to be α+β+. These two strains also differ in immunoglobulin allotype: CBA is allotype a and DBA/1 is allotype c. Eight CBA DBA/1 allophenic mice were immunized with GLø. Four of the mice gave a high secondary response to the polymer. The alloptype of the angibody from these mice was determined by affinity chromatography, and it was found that only allotype c was present. This means that all of the antibody was produced by high responder DBA/1 cells. Thus, there is no evidence for the cooperation of histoincompatible cells in the mounting of an immune response by allophenic mice.

    Notes: Experiments were conducted to determine if non-H-2 gene effects could be demonstrated in mice which had been primed to either the α-subunit or β-subunit of human haemoglobin. It was found that C3H.SW (H-2b) and Balb/c (H-2d) mice are low responder mice to α-chain of a haemoglobin when compared to H-2 identical B10 (H-2b) and B10.D2(H-2d) mice. B120.S and A.SW (both H-2s) are responsive to β-chain challenge while Balb/c mice are low responders in contrast to high responder B10.D2 mice. Ly-1+ cells were demonstrated to be required (by cell depletion experiments) for an in vitro T-cell proliferative response to either subunit.     Notes: Rabbits homozygous at the Ab kappa chain allotypic locus (Ab4/Ab4 and Ab9/Ab9) were immunized with negatively and positively charged antigens. The negatively charged antigens were bovine serum albumin (BSA; pI = 4.9), ovalbumin (OV; pI = 4.9) and two synthetic polypeptides: copolymer L-glutamic acid L-tyrosine (abbreviated TG; pI = 4.8) and multichain poly (L-tyrosine: L-glutamic acid) poly D-alanine: poly L-lysine (poly L-lysine backbone) (abbreviated (TG) - AL; pI = 4.8). The positively charged antigen was hen egg lysozyme (LYS; pI = 10.2).Homozygous Ab9 rabbits responding to negatively charged antigens made less antibody than did Ab4 homozygotes. In contrast, both groups of animals responded equally well to positively charged lysozyme. The relative electric charges of the antibodies were assessed by Sephadex A-50 chromatography. The charge distribution was found to depend on the charge of the eliciting antigen. The positively charged antibodies of Ab9/Ab9 rabbits were not nearly as positively charged as those raised in Ab4/Ab4 rabbits.The difference in average electric charges and in the range of these charges between Ig Ab4 and Ig Ab9 explain quantitative differences in antibody formation in response to negatively charged antigens and may be a contributing factor to the ‘pecking order’, i.e. unequal phenotypic expression of ligh chain genes in Ab4/Ab9 heterozygotes.

    Notes: Virus infected cells can carry HLA antigens not demonstrated in uninfected cells. In a wider context, it is known that tumour cells in the mouse can exhibit H2-like antigens of foreign haplotypes which they resemble. Our aim was to investigate, using an absorption method, the HLA antigens of ten leukaemic cells and to compare these results with those of normal lymphocytes. The following sera was used for absorption: anti-B5 serum, anti-B7 serum, anti-B12 serum. These sera were absorbed on the ten leukaemic cells and fifteen healthy lymphocytes. An anti-A1 + B8 serum and an anti-A2 serum were absorbed on the ten leukaemic cells, but there was not enough serum to carry out the similar absorption with the healthy lymphocytes. Two points emerged from these results. (1) Leukaemia cells absorbed anti-HLA antibodies as effectively as healthy lymphocytes when the cells carried the antigen corresponding to the serum specificity. The curves obtained with leukaemia cells were comparable to those of healthy cells. (2) When the cells were not carriers of the antigens corresponding to the antibody's specificity, leukaemia cells were also capable of absorbing antibodies, unlike healthy lymphocytes which had no or poor absorption.

    Notes: Expression of H-2Kk and H-2Dk molecules was studied by indirect immunofluorescence on vaccinia virus-infected Ls cells (derived from the L929 cell line), using D-23b and ASA-21 monospecific alloantisera, respectively directed against the H-2.23 and H-2.32 private specificities of the H-2k haplotype. Our study demonstrates that the increase in vaccinia virus-induced antigens on the membrane of infected cells (as a function of the multiplicity of infection) is associated with a concomitant decrease in the expression of both H-2Kk and H-2Dk serologically defined private specificities. Absorption experiments of D-23b and ASA-21 sera, using infected or uninfected Ls cells, also indicate that after virus infection, H-2Kk and H-2Dk private specificities are equally altered in their serological expression. We finally show that this alteration consists in a mere quantitative decrease of H-2 molecules, since the cytolytic capacity of a rabbit anti-H-2 serum, specifically reacting against the constant part of the heavy chains of H-2 molecules, was significantly more absorbed by uninfected cells than by infected cells. We conclude that no preferential decrease in the expression of H-2 Kk and H-2 Dk molecules is induced during cell infection by vaccinia virus.

    Notes: We investigated the capacity of anti-I-Ak and anti-I-E/Ck monoclonal antibodies (m.Ab.) to inhibit T lymphocyte proliferative responses to soluble antigen (Keyhole limpet hemocyanin), alloantigens (H-2 or non-H-2 related) and a mitogen (Concanavalin A). Surprisingly, specific inhibition was observed in all circumstances, and with both anti-I-Ak and anti-I-E/Ck m.Ab., whether the responses tested were I restricted in cell mixing experiments or not.The significance of the inhibition by anti-Ia m.Ab. of non-Ia-restricted responses is still not completely understood. These results, however, strongly suggest that in vitro inhibition by anti-Ia antibodies of T cell proliferative responses does not necessarily indicate I restriction of the presentation to T lymphocytes of the corresponding antigen.

    Notes: A heterophile rheumatoid factor (RF) response to LPS was measured in mice by means of a plaque assay using target cells coated with bovine IgG. CBA mice were high responders and A, and DBA/2 were low responders, while strains C57B1, and LP/J showed low to intermediate responsiveness. Use of appropriate congenic strains confirmed that responsiveness is not closely associated with either H-2 or Igh. Failure to respond is not likely to be due to a V-region deficiency, nor can it be readily circumvented, by heterosis (complementation) or the use of high doses of LPS or an extra virulent form of LPS. Breeding experiments show a quantitative trait with maternal responder status having a measurably positive effect.

    Notes: The T cell alloantigens A.R.T.-1, A.R.T.-2, Pta, Ag-F and RT-Ly-2 were examined for linkage to albinism and the haemoglobin variant Hbb, and for cell expression similarities in the rat. The A.R.T.-1 alloantigen was not linked to either A.R.T.-2, albinism or Hbb. The A.R.T.-2 alloantigen was demonstrated to be closely associated to albinism and Hbb with a recombination frequency of approximately 3% with albinism and 1% with Hbb. The four recombinants (out of 104 animals examined) were subsequently analysed with alloantisera to Ag-F, Pta and RT-Ly-2. These alloantisera detected an antigenic system(s) which appeared to co-segregate with A.R.T.-2 in the recombinants studied Characterization of the specific T cell populations expressing each alloantigen demonstrated that removal of the cells bearing any one of the alloantigens removed all the cells expressing any of the other alloantigens. These results demonstrate that the four alloantigenic systems A.R.T.-2, Pta, Ag-F and RT-Ly-2 are located in the same genetic region of linkage group I, and appear to be expressed on the same peripheral T cell subpopulation(s) in the rat.

    Notes: Abstract. In the intestinal epithelium of 3–5-summer old tench, Tinca tinca L., an unknown Eimeria sp. developing white, pinhead-sized nodules was found. Oocysts were shed in a non-sporulated state, but at 20°C in tap water they completed sporulation and resembled Eimeria subepithelialis, a parasite of common carp. During the period of examination, in April and May, only young oocysts and gamogonic stages developing in the epithelium of the mucosa were detected. Despite the morphological similarities and nodular occurrence there are significant differences between the nodular coccidiosis of the tench and common carp.

    Notes: Abstract. Post-spawning mortalities of 2- to 3-year old female rainbow trout, Salmo gairdneri Richardson, at a Newfoundland hatchery were studied. The fish were found to have a mixed bacterial infection involving primarily a Lactobacillus sp. (a Gram-positive, chain-forming coccobacillus) and to a lesser degree Enterobacteriaceae, Aeromonas hydrophila and Pseudomonas fluorescens. The bacteria were isolated from the visceral organs, kidney, and ascitic fluid present in the coelomic cavity. The fish had extensive degeneration and necrosis of the liver, spleen and kidney, and sloughing of intestinal epithelium, but whether these were caused directly by the bacterial infection was not established because the fish appeared to have received substantial stress from the mechanical stripping and from the retention of dead, unshed eggs in the coelomic cavity. Tissue sections suggested that substantial growth of the Lactobacillus sp. was occurring within these unshed eggs. The present study describes this unusual observation and compares the Lactobacillus sp. to two other Lactobacillus isolates from cultured trout.

An ultrastructural study of Myxobolus exiguus Thelohan, 1895 using interlamellar and subdermal plasmodia from thick-lipped grey mullet, Crenimugil labrosus, revealed differences in the structure of the plasmodia and host tissue reaction. Both plasmodia showed structural evidence of pinocytotic activity, but the surface area of subdermal plasmodia was considerably increased by digitiform extensions of the ectoplasm. A filamentous glycocalyx was a feature of the surface membrane of plasmodia, being more distinct on the interlamellar form. Sporo-genesis involved two morphologically distinct types of generative cells, producing a disporous pansporoblast. Mature spores were oval, measuring 9-0 μm×7.5 μm. Sporogenesis and capsulogenesis were essentially similar to other myxosporea. In the juvenile fish examined the host tissue reaction was slight, involving hyperplasia of epithelial cells associated with interlamellar plasmodia and some collagen deposition and localized melanization associated with subdermal plasmodia.

    Notes: Abstract. An outbreak of disease in a British trout farm was identified as fish botulism. The diagnosis was established by demonstration of toxin in the serum and intestinal contents of fish, and was confirmed by the production of experimental botulism in trout inoculated with toxin from a strain of Clostridium botulinum type E isolated from intestinal contents of a sick fish. The outbreak was controlled and eliminated within 1 month by conservative measures which comprised the emptying, cleaning and heavy liming of affected ponds; intensive examination of all ponds for immediate removal of dead and sick fish and, on public health grounds, the banning of movement of fish outwith the farm. This episode, although of considerable financial consequence to the farmer, was shown to have no public health significance.

Organ samples were collected from plaice, Pleuronectes platessa L., captured in the highly oiled Aber Wrac'h and Aber Benoit at five intervals during the period 1978-1980, following the Amoco Cadiz crude oil spill. Reference plaice were obtained along the western and southern coasts of Brittany. Tissue samples were fixed, processed and stained by routine histologic procedures for qualitative and quantitative histopathological examination. The predominant lesions and conditions which characterized fish from the oiled estuaries were: fin and tail necrosis, hyperplasia and hypertrophy of gill lamellar mucous cells, gastric gland degeneration, decreased hepatocellular vacuolation (lipid), increased concentration of hepatic macrophage centres and lateral trunk muscle fibre degeneration. Dilation of Bowman's space, glomerular hypertrophy and abdominal muscle fibre degeneration were pronounced at the last sampling interval. Other types of lesions were seen at a lower frequency: hyperplasia and fusion of gill lamellar epithelium, telangiectasis of gill capillaries, hepatopancreatic degeneration and hepatic necrosis. Fish muscle and liver collected for biochemical analysis to detect the presence or absence of petroleum hydrocarbons were negative for Amoco Cadiz oil fractions. At the same time oysters, Crassostrea gigas Thunberg, collected from the oiled estuaries throughout the study period, showed high levels of Amoco Cadiz oil fractions. These data, along with the type and frequency of lesions observed, suggested that the plaice had been chronically-exposed to petroleum hydrocarbons of Amoco Cadiz origin. Some relief from chronic pollutant exposure was indicated by a decrease in gill mucous cell concentration at the last sampling interval. An increase in abdominal muscle fibre degeneration and an increase in the frequency of dilation of Bowman's space with glomerular hypertrophy at the last sampling interval suggested a latent response to the polluting agent.

    Notes: Abstract. The clinical signs and histopathology of Saprolegnia sp. [related to S. diclina Humphrey (syn. S. parasitica Coker)] are described in cultured Anguilla anguilla L. elvers under an intensive production system. The main lesions observed were loss of epithelium leading to ulceration, oedema and myofibrillar degenerative changes of the muscle mass. The oedema resulted in swelling of the inter-myotomal connective tissue, loss of nuclei and minor host reaction. Traumatic lesions with subsequent bacterial infections had a mononuclear inflammatory response. It is suggested that the rapid extension of the lesion is due to the loss of integrity of the integument and the widespread oedema, which altered the viability of tissues and assisted the Saprolegnia infection. The condition was fatal for infected elvers.

    Notes: Abstract. The pathogenesis of IPNV infection was examined in young brook trout following intraperitoneal inoculation. Fish were sampled sequentially and the distribution of virus determined by virus isolation, histopathology and immunofluorescence. Following intraperitoneal inoculation, virus entered the peritoneal cavity and by 2 days post-inoculation virus had interacted with the pancreatic exocrine cells. Replication in these cells resulted in the production of IPNV-specific antigen, necrosis of infected cells, and spread of infectious virus to adjacent tissue. Areas of viral replication were at first multifocal but tended to merge as they grew. Eventually, most of the acinar tissue became involved and only small pockets of normal acinar cells remained. Twelve-week old brook trout survived with only a small amount of functional exocrine pancreas and limited virus replication continued in this tissue for weeks. Extensive virus replication appeared to take place only in the pancreas although viral antigen and slight pathological changes were found in the renal interstitium and livers of some fish.

Pale and swollen gills were observed in 23 of 240 Laval's eelpouts, Lycodes lavalaei Vladykov and Tremblay (Perciformes), taken from several locations off the coasts of Newfoundland and southern Labrador. Light microscopic study of histological sections revealed clusters of unidentified cells (‘X-cells’) in the basal region of secondary gill lamellae and extending along the latter, between the epithelium and lamellar capillaries. In the electron microscope two types of X-cells were enclosed in a network of cytoplasmic processes from dense ‘envelope’ cells. Both types of X-cells were bound by a densely-coated limiting membrane and had nuclei with large, dense nucleoli, numerous mitochondria with short vesicular cristae, lipid inclusions, abundant smooth endoplasmic reticulum and small dense inclusions. The Type II X-cells were larger, contained a much greater concentration of lipid and small dense inclusions, and an additional inclusion with a dense core surrounded by a light halo. The X-cells are compared with strikingly similar cells described from epidermal and pseudobranch tumours of marine fish. Although transformation of a host cell by virus or possibly pollution has been suggested, neither the nature nor the origin of the X-cells can be determined.

The inactivation rates in the aquatic environment of two fish pathogens, infectious pancieatic necrosis virus (IPNV) and infectious haematopoietic necrosis virus (IHNV), were compared with that of poliovirus type 1 as a representative of the human enterovirus group. The survival studies were performed using untreated fresh, estuarine and sea water samples held at 15 and 20°C. The results indicated that the salmonid viruses survived longer than poliovirus in the saline waters, whereas in fresh water poliovirus was the most stable of the three viruses. IPN virus proved to be most stable in estuarine water at 15°C, whereas the survival of IHN virus was favoured in fresh water. We also observed that at 20°C the inactivation rate for each virus was independent of salt concentration in estuarine and sea water. Although temperature exhibited a marked effect on virus stability in fresh water, the salmonid viruses presented similar survival patterns at both temperatures in sea water. In general the period of greatest viral inactivation correlated with higher bacterial numbers in the waters.

    Notes: Abstract. The sensitivity of Lagenidium spp. isolated from Penaeus monodon and Scylla serrata to 34 antimycotic compounds was determined. Mycostatic effects were evaluated from observations of chemicals on the development of vesicles, zoospores and mycelial growth and on these bases three classes of mycostatic effect were recognized. Zoospores were inhibited by 〈 1 mg/1 of clotrimazole, crystal violet, econazole nitrate, malachite green, treflan and trifluralin; 1-5 mg/1 of benzalkonium chloride, daconil, domestic detergent, basic fuchsin and furanace; and 10-50 mg/1 benlate, formalin, griseofulvin, phenol and potassium permanganate. Variable mycostatic levels, however, were exhibited with copper sulphate, 2,4-D, hydrogen peroxide, methylene blue, pimaricin, resiguard and tolnaftate while amphotericin, iodine, nystatin, triacetin, boric acid, fungitox and PVP-iodine proved to be poor antifungal agents.

Isolated areas of the skin and gills of rainbow and brown trout were subjected to hyperosmotic infiltration with E. coli both in vitro and in vivo and examined for the passage of bacteria. Bacteria were recovered from the isolated gill arches only after treatment with balanced salt solution at x 10 concentration, followed by the bacterial suspension. All the skin flap experiments were negative. Bacteria were recovered from the blood offish when that part of the animal anterior to the pectoral fins was exposed to the infiltrating bacteria. They were only recovered when the animal was exposed to the full hyperosmotic infiltration schedule. The evidence strongly supports the view that the bacteria enter the animal through the gills.

The present report deals with experiments concerning the theoretical basis for development of a live, attenuated vaccine against viral haemorrhagic septicaemia (VHS) of rainbow trout, Salmo gairdneri Richardson, on the basis of a virus strain derived from the Egtved virus reference strain F, the pathogenicity of which has been strongly reduced by in vitro passage. It is shown that the low pathogenicity of the virus (designated the Reva strain) is a genetically stable feature and that the protection against VHS induced by infection with the virus is due to an immune response. Following immunological priming at 10°C partial protection developed at 5° 10° and 15° but not at 20°C, whereas neutralizing antibody was produced at all four temperatures by some of the fish. The duration of virus persistence in trout was found to be inversely proportional to the water temperature.

Infection trials using two serotypes of VHS viruses (type 1 and 23/75) demonstrated that Atlantic salmon fry were susceptible to the disease when injected intraperitoneally (i.p.) with 103 pfu of virus/fish but resistant to infection by a bath method when exposed for 3 h in water containing 5 × 104 pfu of virus/ml. In the i.p.-infected fish, mortality reached 78 and 67% within 13 days with VHSV1 nad 23/75 serotypes, respectively. High virus yields were recovered from infected fish and virus shedding was demonstrated by the onset of VHS in rainbow trout kept in the outflow water from the aquaria containing infected salmon. Neither mortality nor virus shedding occurred in salmon infected by the water route but virus multiplication was demonstrated in 2 of 60 fish with VHSV1 and 3 of 60 fish with virus 23/75. On day 79 post-infection the sera from surviving salmon of both i.p. and bath infection trials exhibited good neutralizing titres (around 1000) against the homologous viruses.

Exotoxins prepared from streptococcal bacteria of Seriola spp. origin were grown in fluid culture and fractionates of the supernatant used in toxicity studies in Seriola by oral and percutaneous exposure. A recurring disease known as ‘black patch necrosis’ (BPN) has been the cause of severe mortalities in 0-group and 1-group Dover sole during the period 1974-1978 at the Sea Fish Industry Authority's Marine Cultivation Unit at Hunter-ston, Ayrshire, Scotland. Samples of diseased and healthy Dover sole tissue were examined microbiologically and a long thin filamentous Gram-negative organism, strongly resembling Flexibacter columnar's, was repeatedly isolated from diseased tissue but was not found in healthy tissue. This organism was pathogenic to Dover sole, producing 100% mortality in 96 h at 17.5°C (±2°C) and the organism was reisolated from all the infected fish. The pathogenic isolate and reisolate were examined and compared with reference strain NCMB 1038 of Flexibacter columnarts, formerly known as Chondrococcus columnaris or Cytophaga columnaris. The isolate, reisolate and type culture were identical with respect to cell morphology, oxidase test, catalase production, production of H2S, casein hydrolysis, gelatin liquefaction, hydrolysis of Tween 20, degradation of tyrosine and starch hydrolysis. All bacterial types showed similar patterns of sensitivity to a range of antibiotics. It is proposed that a Flexibacter columnari's-like organism is associated with the condition in farmed Dover sole known as ‘black patch necrosis’.

    Notes: Abstract. The exact cellular site of replication of infectious pancreatic necrosis virus (IPNV) in carrier fish is unknown. In order to determine if IPNV replicates in trout leucocytes, we purified leucocytes from normal (non-carrier) trout and separated the cells into an adherent and a non-adherent population. IPNV replicated in less than 0-01 % of the adherent leucocytes with a yield of about 400 p.f.u./cell. IPNV also became associated with less than 0.07% of the non-adherent leucocytes; either IPNV did not replicate in these cells or the yield was, at best, only a few p.f.u./cell. Trout persistently infected with IPNV (carrier fish) were tested for the presence of IPNV in leucocytes by co-cultivating with a sensitive fish cell line; this same population of trout was also tested for IPNV by organ sampling using standard methods. Ninety-eight per cent of the trout were positive for IPNV by organ sampling, but only 75 % yielded IPNV from leucocytes. Thus a blood sample from a living fish can be used to detect the presence of IPNV.

    Notes: Abstract. An epizootic of lymphoma in northern pike, Esox lucius L., occurring in the Åland Islands in Finnish waters of the Baltic Sea is described. Macroscopic inspection of 19 tumorous pike, 17 males and two females, as well as tissue observations by light and electron microscopy showed that this lymphoma is similar to those previously reported in esocid fishes from North America, Ireland and Sweden. Haematoxylin and eosin-stained light microscopic sections of tumour tissue suggested its classification as a lymphoblastic or stem cell lymphoma. Tumour cells cultured during several months at 4°C showed morphological features of differentiation and resembled plasma cells. No conclusive evidence of the presence of viruses in the lesion was obtained.

Continuous visual observations of the signs of development of UDN in fish kept in aquaria were correlated with light microscopical, transmission and scanning electron microscopical results. The initial signs of the disease are circles of pathologically-changed epidermis. The tight junctions of the squamous cells within these mucus-free areas disintegrate resulting in the loss of the protective function of the zonula occludens. Subsequently, the intercellular spaces dilate and communicate with the exterior. Necrosis of the epidermal cells due to hydromineral disturbances occur simultaneously with fungal infections and marked responses of the melanophores. It is suggested that the fungal infections are triggered by metabolites of the necrotic epidermal cells. The epidermal cells are shed and the fungus determines the further course of the disease which terminates in large ulcers covered with fungal hyphae. No substantial evidence for the presence of a virus could be recorded at any stage of the disease. It is concluded that the disease is a squames and the fungus appears as an opportunist which causes the eventual death of the fish.

Incubation temperatures of 11°, 18° and 28° did not substantially affect biochemical reactions of either virulent or avirulent forms of Aeromonas salmonicida subspecies salmonicida. The only change observed, amygdalin fermentation, was positive at 11° and 18° but negative at 28°C. Several isolates utilized sucrose, a characteristic not normally recognized for A. salmonicida subspecies salmonicida. Antimicrobial susceptibility screening indicated resistance to novobiocin increased at the higher incubation temperatures. Standardized drug sensitivity testing procedures and precise zone diameter interpretive standards for bacterial fish pathogens are needed.
