    Notes: We review the origin, evolution, and physical nature of hot gas in elliptical galaxies and associated galaxy groups. Unanticipated recent X-ray observations with Chandra and XMM indicate much less cooling than previously expected. Consequently, many long-held assumptions must be reexamined or discarded and new approaches must be explored. Chief among these are the role of heating by active galactic nuclei, the influence of radio lobes on the hot gas, details of the cooling process, possible relation between the hot and colder gas in elliptical galaxies, and the complexities of stellar enrichment of the hot gas.

    Notes: This review surveys the observed properties of interstellar dust grains: the wavelength-dependent extinction of starlight, including absorption features, from UV to infrared; optical luminescence; infrared emission; microwave emission; optical, UV, and X-ray scattering by dust; and polarization of starlight and of infrared emission. The relationship between presolar grains in meteorites and the interstellar grain population is discussed. Candidate grain materials and abundance constraints are considered. A dust model consisting of amorphous silicate grains, graphite grains, and polycyclic aromatic hydrocarbons is compared with observed emission and scattering. Some issues concerning evolution of interstellar dust are discussed.

    Notes: The status of our current understanding of angular momentum transport in accretion disks is reviewed. The last decade has seen a dramatic increase both in the recognition of key physical processes and in our ability to carry through direct numerical simulations of turbulent flow. Magnetic fields have at once powerful and subtle influences on the behavior of (sufficiently) ionized gas, rendering them directly unstable to free energy gradients. Outwardly descreasing angular velocity profiles are unstable. The breakdown of Keplerian rotation into MHD turbulence may be studied in some numerical detail, and key transport coefficients may be evaluated. Chandra observations of the Galactic Center support the existence of low luminosity accretion, which may ultimately prove amenable to global three-dimensional numerical simulation.

    Notes: Helioseismology has transformed our knowledge of the Sun's rotation. Earlier studies revealed the Sun's surface rotation, but now a detailed observational picture has been built up of the internal rotation of our nearest star. Unlike the predictions of stellar-evolution models, the radiative interior is found to rotate roughly uniformly. The rotation within the convection zone is also very different from prior expectations, which had been that the rotation rate would depend primarily on the distance from the rotation axis. Layers of rotational shear have been discovered at the base of the convection zone and in the subphotospheric layers. Studies of the time variation of rotation have uncovered zonal-flow bands, extending through a substantial fraction of the convection zone, which migrate over the course of the solar cycle, and there are hints of other temporal variations and of a jet-like structure. At the same time, building on earlier work with mean-field models, researchers have made great progress in supercomputer simulations of the intricate interplay between turbulent convection and rotation in the Sun's interior. Such studies are beginning to transform our understanding of how rotation organizes itself in a stellar interior.

    Notes: The launches of the Chandra X-Ray Observatory in June 1999 and the XMM-Newton Observatory in December 1999 opened a new era in X-ray astronomy. Both of these missions incorporate novel diffraction grating spectrometers that are providing the first high-resolution X-ray spectra of most classes of astrophysical sources. The spectra obtained to date exhibit a wealth of discrete detail, yielding sensitive constraints on physical conditions in the emitting plasmas. We review the essential characteristics of these instruments, the basics of X-ray spectral formation in cosmic sources, and the exciting new results that have emerged from Chandra and XMM-Newton grating observations of a wide variety of astrophysical systems.

    Notes: Weak gravitational lensing provides a unique method to map directly the distribution of dark matter in the universe and to measure cosmological parameters. This cosmic-shear technique is based on the measurement of the weak distortions that lensing induces in the shape of background galaxies as photons travel through large-scale structures. This technique is now widely used to measure the mass distribution of galaxy clusters and has recently been detected in random regions of the sky. In this review, we present the theory and observational status of cosmic shear. We describe the principles of weak lensing and the predictions for the shear statistics in favored cosmological models. Next, we review the current measurements of cosmic shear and show how they constrain cosmological parameters. We then describe the prospects offered by upcoming and future cosmic-shear surveys as well as the technical challenges that have to be met for the promises of cosmic shear to be fully realized.

    Notes: Abstract The more violent impacts of water waves on walls create velocities and pressures having magnitudes much larger than those associated with the propagation of ordinary waves under gravity. Insight into these effects has been gained by irrotational-flow computations and by investigating the role of entrained and trapped air in wave impacts. This review focuses on the results of theoretical work, making particular note of the value of considering pressure impulse, and highlights the aspects that are poorly understood.

    Notes: Abstract This paper reviews recent advances in our understanding of the origin and hierarchy of organized flow structures in fluidized beds, distinction between bubbling and nonbubbling systems, and stages of bubble evolution. Experimental data and theory suggest that, at high particle concentrations, the particle-phase pressure arising from flow-induced velocity fluctuations decreases with increasing concentration of particles. This, in turn, implies that nonhydrodynamic stresses must be present to impart stability to a uniformly fluidized bed at very high particle concentrations. There is ample evidence to support an argument that, in commonly encountered gas-fluidized beds, yield stresses associated with enduring particle networks are present in the window of stable bed expansion, where the particles are essentially immobile until bubbling commences. However, some recent data on gas-fluidized beds of agglomerates of cohesive particles suggest that there exists a window of bed expansion where the bed does manifest a smooth appearance to the naked eye and the particles are mobile; at higher gas velocities the bed bubbles visibly. The mechanics of such beds remain to be fully explained.

    Notes: Abstract A review is given of the stability of complex fluids subject to homogeneous states of shearing, a research field that is scarcely two decades old. For the benefit of fluid mechanicians, a brief, somewhat historical overview is presented of material instability in elastoplastic solids, where one finds a considerable body of experiment and a rich source of theoretical concepts including Hadamard instability, strain localization, and nonlocal constitutive models. A survey is then given of recent theoretical and experimental studies of instability with shear banding in various complex fluids, including micellar solutions, particulate suspensions, and rapidly sheared granular media. Various stability analyses are encapsulated in a mathematical dynamical-systems model for constitutive equations of the rate-type, and a general linear-stability theory is given for viscoelastic fluids in unbounded homogeneous shear flows. A general form of (Kelvin) wave-vector stretching is shown to play a key role in the growth of Fourier modes, as illustrated by recent computations for granular shear flow. The Fourier description also provides an explicit representation of higher-gradient (nonlocal) effects as higher-order powers of wave number.

    Notes: Abstract Turbulence is ubiquitous in atmospheric clouds, which have enormous turbulence Reynolds numbers owing to the large range of spatial scales present. Indeed, the ratio of energy-containing and dissipative length scales is on the order of 105 for a typical convective cloud, with a corresponding large-eddy Reynolds number on the order of 106 to 107. A characteristic trait of high-Reynolds-number turbulence is strong intermittency in energy dissipation, Lagrangian acceleration, and scalar gradients at small scales. Microscale properties of clouds are determined to a great extent by thermodynamic and fluid-mechanical interactions between droplets and the surrounding air, all of which take place at small spatial scales. Furthermore, these microscale properties of clouds affect the efficiency with which clouds produce rain as well as the nature of their interaction with atmospheric radiation and chemical species. It is expected, therefore, that fine-scale turbulence is of direct importance to the evolution of, for example, the droplet size distribution in a cloud. In general, there are two levels of interaction that are considered in this review: (a) the growth of cloud droplets by condensation and (b) the growth of large drops through the collision and coalescence of cloud droplets. Recent research suggests that the influence of fine-scale turbulence on the condensation process may be limited, although several possible mechanisms have not been studied in detail in the laboratory or the field. There is a growing consensus, however, that the collision rate and collision efficiency of cloud droplets can be increased by turbulence-particle interactions. Adding strength to this notion is the growing experimental evidence for droplet clustering at centimeter scales and below, most likely due to strong fluid accelerations in turbulent clouds. Both types of interaction, condensation and collision-coalescence, remain open areas of research with many possible implications for the physics of atmospheric clouds.

    Notes: Abstract This review describes some of the important developments in the numerical investigation of transition to turbulence in wall-bounded and free shear flows during the past decade. The evolution of numerical techniques and models as well as the advances in our theoretical understanding of the physics of laminar-turbulent transition that were achieved using these tools are described. For wall-bounded flows, particular emphasis is placed on investigations studying various scenarios of "bypass transition" in flows that are asymptotically stable. A brief review of investigations into receptivity and control of transitional flows is included.

    Notes: Abstract We provide an overview of level set methods, introduced by Osher and Sethian, for computing the solution to fluid-interface problems. These are computational techniques that rely on an implicit formulation of the interface, represented through a time-dependent initial-value partial-differential equation. We discuss the essential ideas behind the techniques, the coupling of these techniques to finite-difference methods for incompressible and compressible flow, and a collection of applications including two-phase flow, ship hydrodynamics, and ink-jet-printhead design.

    Notes: Abstract We review the most important theoretical and numerical results obtained in the realm of shell models for the energy-turbulent cascade. We mainly focus here on those results that had or will have some impact on the fluid-dynamics community. In particular, we address the problem of small-scale intermittency by discussing energy-helicity interactions, energy-dissipation multifractality, and universality of intermittency, i.e., independence of anomalous scaling exponents from large-scale forcing and boundary conditions. A multifractal-based description of multiscale and multitime correlation functions in turbulence is also presented. Finally, we also briefly review the analytical difficulties, and hopes, of calculating anomalous exponents.

    Notes: Abstract Vibratory signals of plant-dwelling insects, such as land bugs of the families Cydnidae and Pentatomidae, are produced mainly by stridulation and/or vibration of some body part. Signals emitted by the vibratory mechanisms have low-frequency characteristics with a relatively narrow frequency peak dominant around 100 Hz and differently expressed frequency modulation and higher harmonics. Such spectral characteristics are well tuned to the transmission properties of plants, and the low attenuation enables long-range communication on the same plant under standing wave conditions. Frequencies of stridulatory signals extend up to 10 kHz. In some groups, vibratory and stridulatory mechanisms may be used simultaneously to produce broadband signals. The subgenual organ, joint chordotonal organs, campaniform sensilla and mechanoreceptors, such as the Johnston's organ in antennae, are used to detect these vibratory signals. Species-specific songs facilitate mate location and recognition, and less species-specific signals provide information about enemies or rival mates.

    Notes: Abstract Insect-plant interactions involving the cultivated tomato and its relatives in the genus Lycopersicon have been intensively studied for several decades, resulting in one of the best documented and in-depth examples of the mechanistic complexities of insect-plant interactions, which encompass both herbivores and their natural enemies. Trichome-mediated defenses are particularly significant in L. hirsutum f. glabratum and have been extensively implicated in negative tritrophic effects mediated by direct contact of parasitoids and predators with trichomes, as well as indirect effects mediated through their hosts or prey. Both constitutive and inducible defense traits of L. esculentum exert effects on selected parasitoids and predators. The effects of any particular plant defense trait on parasitoids and predators depend on the specific attributes of the plant trait and the details of the physical, biochemical, and behavioral interaction between the natural enemy, its host (prey), and the plant.

    Notes: Abstract The century-old discovery of the role of Anopheles in human malaria transmission precipitated intense study of this genus at the alpha taxonomy level, but until recently little attention was focused on the systematics of this group. The application of molecular approaches to systematic problems ranging from subgeneric relationships to relationships at and below the species level is helping to address questions such as anopheline phylogenetics and biogeography, the nature of species boundaries, and the forces that have structured genetic variation within species. Current knowledge in these areas is reviewed, with an emphasis on the Anopheles gambiae model. The recent publication of the genome of this anopheline mosquito will have a profound impact on inquiries at all taxonomic levels, supplying better tools for estimating phylogeny and population structure in the short term, and ultimately allowing the identification of genes and/or regulatory networks underlying ecological differentiation, speciation, and vectorial capacity.

    Notes: Abstract Secretions of male accessory glands contain a variety of bioactive molecules. When transferred during mating, these molecules exert wide-ranging effects on female reproductive activity and they improve the male's chances of siring a significant proportion of the female's offspring. The accessory gland secretions may affect virtually all aspects of the female's reproductive activity. The secretions may render her unwilling or unable to remate for some time, facilitating sperm storage and ensuring that any eggs laid will be fertilized by that male's sperm. They may stimulate an increase in the number and rate of development of eggs and modulate ovulation and/or oviposition. Antimicrobial agents in the secretions ensure that the female reproductive tract is a hospitable environment during sperm transfer. In a few species the secretions include noxious chemicals. These are sequestered by developing eggs that are thereby protected from predators and pathogens when laid.

    Notes: Abstract Comparative analysis of the complete genome sequences of 13 baculoviruses revealed a core set of 30 genes, 20 of which have known functions. Phylogenetic analyses of these 30 genes yielded a tree with 4 major groups: the genus Granulovirus (GVs), the group I and II lepidopteran nucleopolyhedroviruses (NPVs), and the dipteran NPV, CuniNPV. These major divisions within the family Baculoviridae were also supported by phylogenies based on gene content and gene order. Gene content mapping has revealed the patterns of gene acquisitions and losses that have taken place during baculovirus evolution, and it has highlighted the fluid nature of baculovirus genomes. The identification of shared protein phylogenetic profiles provided evidence for two putative DNA repair systems and for viral proteins specific for infection of lymantrid hosts. Examination of gene order conservation revealed a core gene cluster of four genes, helicase, lef-5, ac96, and 38K(ac98), whose relative positions are conserved in all baculovirus genomes.

    Notes: Abstract Ideally, integrated pest management should rely on an array of tactics. In reality, the main technologies in use are synthetic pesticides. Because of well-documented problems with reliance on synthetic pesticides, viable alternatives are sorely needed. Physical controls can be classified as passive (e.g., trenches, fences, organic mulch, particle films, inert dusts, and oils), active (e.g., mechanical, polishing, pneumatic, impact, and thermal), and miscellaneous (e.g., cold storage, heated air, flaming, hot-water immersion). Some physical methods such as oils have been used successfully for preharvest treatments for decades. Another recently developed method for preharvest situations is particle films. As we move from production to the consumer, legal constraints restrict the number of options available. Consequently, several physical control methods are used in postharvest situations. Two noteworthy examples are the entoleter, an impacting machine used to crush all insect stages in flour, and hot-water immersion of mangoes, used to kill tephritid fruit fly immatures in fruit. The future of physical control methods will be influenced by sociolegal issues and by new developments in basic and applied research.

    Notes: Abstract Until the 1990s, Amblyomma americanum was regarded primarily as a nuisance species, but a tick of minor importance as a vector of zoonotic pathogens affecting humans. With the recent discoveries of Ehrlichia chaffeensis, Ehrlichia ewingii, and "Borrelia lonestari," the public health relevance of lone star ticks is no longer in question. During the next 25 years, the number of cases of human disease caused by A. americanum-associated pathogens will probably increase. Based on current trajectories and historic precedents, the increase will be primarily driven by biological and environmental factors that alter the geographic distribution and intensity of transmission of zoonotic pathogens. Sociologic and demographic changes that influence the likelihood of highly susceptible humans coming into contact with infected lone star ticks, in addition to advances in diagnostic capabilities and national surveillance efforts, will also contribute to the anticipated increase in the number of recognized cases of disease.

    Notes: Abstract Controversy exists over ecological risks in classical biological control. We reviewed 10 projects with quantitative data on nontarget effects. Ten patterns emerged: (a) Relatives of the pest are most likely to be attacked; (b) host-specificity testing defines physiological host range, but not ecological range; (c) prediction of ecological consequences requires population data; (d) level of impact varied, often in relation to environmental conditions; (e) information on magnitude of nontarget impact is sparse; (f) attack on rare native species can accelerate their decline; (g) nontarget effects can be indirect; (h) agents disperse from agroecosystems; (i) whole assemblages of species can be perturbed; and (j) no evidence on adaptation is available in these cases. The review leads to six recommendations: Avoid using generalists or adventive species; expand host-specificity testing; incorporate more ecological information; consider ecological risk in target selection; prioritize agents; and pursue genetic data on adaptation. We conclude that retrospective analyses suggest clear ways to further increase future safety of biocontrol.

    Notes: Abstract Recent application of biochemical and molecular techniques to study the genesis of scolytid aggregation pheromones has revealed that bark beetles are primarily responsible for the endogenous synthesis of widely occurring pheromone components such as ipsenol, ipsdienol, and frontalin. Because many of the chemical signals are isoprenoids, the roles of the mevalonate biosynthetic pathway and the enzyme HMG-CoA reductase (HMG-R) have been investigated. This has led to the identification of endothelial cells in the anterior midgut as the site of synthesis and to the concept that de novo pheromone biosynthesis is regulated in part by the positive effect of juvenile hormone III (JHIII) on gene expression for HMG-R. Both the pronounced regulation by JHIII and the expression pattern of eukaryotic HMG-R argue against synthesis of these pheromones by prokaryotes. As the mevalonate pathway and its regulation have been studied in few other insects, broader issues addressed through the study of scolytid pheromone biosynthesis include major step versus coordinate regulation of the pathway and a genomics approach to elucidating the entire pathway and the mode of action of JHIII.

    Notes: Abstract FMRFamide-related peptides (FaRPs) contain a C-terminal RFamide but unique N-terminal extensions. They are expressed throughout the animal kingdom and affect numerous biological activities. Like other animal species, Drosophila melanogaster contains multiple genes that encode different FaRPs. The ease of genetic manipulations, the availability of genomic sequence data, the existence of established bioassays, and its short lifespan make D. melanogaster a versatile experimental organism in which to investigate peptide processing, functions, and signal transduction pathways. Here, the structures, precursor organizations, distributions, and activities of FaRPs encoded by D. melanogaster FMRFamide (dFMRFamide), myosuppressin (Dms), and sulfakinin (Dsk) genes are reviewed, and predictions are made on their signaling pathways and biological functions.

    Notes: Abstract Investigations of plant-herbivore interactions continue to be popular; however, a bias neglecting root feeders may limit our ability to understand how herbivores shape plant life histories. Root feeders can cause dramatic plant population declines, often associated with secondary stress factors such as drought or grazing. These severe impacts resulted in substantial interest in root feeders as agricultural pests and increasingly as biological weed control agents, particularly in North America. Despite logistical difficulties, establishment rates in biocontrol programs are equal or exceed those of aboveground herbivores (67.2% for aboveground herbivores, 77.5% for belowground herbivores) and root feeders are more likely to contribute to control (53.7% versus 33.6%). Models predicting root feeders would be negatively affected by competitively superior aboveground herbivores may be limited to early successional habitats or generalist root feeders attacking annual plants. In later successional habitats, root feeders become more abundant and appear to be the more potent force in driving plant performance and plant community composition. Aboveground herbivores, even at high population levels, were unable to prevent buildup of root herbivore populations and the resulting population collapse of their host plants. Significant information gaps exist about the impact of root feeders on plant physiology and secondary chemistry and their importance in natural areas, particularly in the tropics.

    Notes: Abstract The interactions of two economically important gall midge species, the rice gall midge and the Hessian fly, with their host plants, rice and wheat, respectively, are characterized by plant defense via R genes and insect adaptation via avr genes. The interaction of a third gall midge species, the orange wheat blossom midge, with wheat defense R genes has not yet exhibited insect adaptation. Because of the simple genetics underlying important aspects of these gall midge-grass interactions, a unique opportunity exists for integrating plant and insect molecular genetics with coevolutionary ecology. We present an overview of some genetic, physiological, behavioral, and ecological studies that will contribute to this integration and point to areas in need of study.

    Notes: Abstract Nervous system function is entirely dependent on the intricate and precise pattern of connections made by individual neurons. Much of the insightful research into mechanisms underlying the development of this pattern of connections has been done in insect nervous systems. Studies of developmental mechanisms have revealed critical interactions between neurons and glia, the non-neuronal cells of the nervous system. Glial cells provide trophic support for neurons, act as struts for migrating neurons and growing axons, form boundaries that restrict neuritic growth, and have reciprocal interactions with neurons that govern specification of cell fate and axonal pathfinding. The molecular mechanisms underlying these interactions are beginning to be understood. Because many of the cellular and molecular mechanisms underlying neural development appear to be common across disparate insect species, and even between insects and vertebrates, studies in developing insect nervous systems are elucidating mechanisms likely to be of broad significance.

    Notes: Abstract Many of the most harmful parasitic diseases are transmitted by blood-feeding insect vectors. During this stage of their life cycles, selection pressures favor parasites that can manipulate their vectors to enhance transmission. Strategies may include increasing the amount of contact between vector and host, reducing vector reproductive output and consequently altering vector resource management to increase available nutrient reserves, and increasing vector longevity. Manipulation of these life-history traits may be more beneficial at some phase of the parasite's developmental process than at others. This review examines empirical, experimental, and field-based evidence to evaluate examples of changes in vector behavior and physiology that might be construed to be manipulative. Examples are mainly drawn from malaria-infected mosquitoes, Leishmania-infected sandflies, and Trypanosoma-infected tsetse flies.

    Notes: Abstract Birds host many lineages of symbiotic mites, but the greatest diversity is shown by the three superfamilies of astigmatan feather mites: Analgoidea, Pterolichoidea, and Freyanoidea. Members of this diphyletic grouping have colonized all parts of the avian integument from their ancestral nidicolous habitat. Whereas some clearly feed on feather pith or skin, acting as parasites, other feather mites are paraphages and consume feather oils without causing structural damage. Sexual dimorphism in feather mites is often extreme, and little is known of the function of many elaborate male structures. Abundance and location of vane-dwelling mites is affected by season, temperature, light, humidity, and host body condition. Because transmission between hosts usually depends on host body contact, it is unsurprising that feather mite phylogeny often parallels host phylogeny; however, recent cladistic analyses have also found evidence of host-jumping and "missing the boat" in several mite lineages.

    Notes: Abstract Genomics is the study of the structure and function of the genome: the set of genetic information encoded in the DNA of the nucleus and organelles of an organism. It is a dynamic field that combines traditional paths of inquiry with new approaches that would have been impossible without recent technological developments. Much of the recent focus has been on obtaining the sequence of entire genomes, determining the order and organization of the genes, and developing libraries that provide immediate physical access to any desired DNA fragment. This has enabled functional studies on a genome-wide level, including analysis of the genetic basis of complex traits, quantification of global patterns of gene expression, and systematic gene disruption projects. The successful contribution of genomics to problems in applied entomology requires the cooperation of the private and public sectors to build upon the knowledge derived from the Drosophila genome and effectively develop models for other insect Orders.

    Notes: Abstract Lacking a comprehensive fossil record, solitary representatives of the taxa, and/or a definitive phylogeny of closely related insects, comparison of the life history and social biology of basal, living groups is one of the few available options for developing inferences regarding the early eusocial evolution of ants and termites. Comparisons of a select group of basal formicid and isopteran taxa suggest that the reproductive organization of colonies and their patterns of division of labor were particularly influenced, in both groups, by nesting and feeding ecology. Opportunities for serial inheritance of the nest structure and colony population by kin may have been significant in the evolution of multiple reproductive forms and options. Disease has been a significant factor in the evolution of social organization in ants and termites, but the adaptive mechanisms of infection control differ. Evaluations of the convergent and divergent social biology of the two taxa can generate novel domains of research and testable hypotheses.

    Notes: Abstract Neonicotinoids, the most important new class of synthetic insecticides of the past three decades, are used to control sucking insects both on plants and on companion animals. Imidacloprid (the principal example), nitenpyram, acetamiprid, thiacloprid, thiamethoxam, and others act as agonists at the insect nicotinic acetylcholine receptor (nAChR). The botanical insecticide nicotine acts at the same target without the neonicotinoid level of effectiveness or safety. Fundamental differences between the nAChRs of insects and mammals confer remarkable selectivity for the neonicotinoids. Whereas ionized nicotine binds at an anionic subsite in the mammalian nAChR, the negatively tipped ("magic" nitro or cyano) neonicotinoids interact with a proposed unique subsite consisting of cationic amino acid residue(s) in the insect nAChR. Knowledge reviewed here of the functional architecture and molecular aspects of the insect and mammalian nAChRs and their neonicotinoid-binding site lays the foundation for continued development and use of this new class of safe and effective insecticides.

    Notes: Abstract There are three major classes of insect genetic systems: those with diploid males (diplodiploidy), those with effectively haploid males (haplodiploidy), and those without males (thelytoky). Mixed systems, involving cyclic or facultative switching between thelytoky and either of the other systems, also occur. I present a classification of the genetic systems of insects and estimate the number of evolutionary transitions between them that have occurred. Obligate thelytoky has arisen from each of the other systems, and there is evidence that over 900 such origins have occurred. The number of origins of facultative thelytoky and the number of reversions from obligate thelytoky to facultative and cyclic thelytoky are difficult to estimate. The other transitions are few in number: five origins of cyclic thelytoky, eight origins of obligate haplodiploidy (including paternal genome elimination), the strange case of Micromalthus, and the two reversions from haplodiploidy to diplodiploidy in scale insects. Available evidence tends to support W.D. Hamilton's hypothesis that maternally transmitted endosymbionts have been involved in the origins of haplodiploidy. Bizarre systems of extrazygotic inheritance in Sternorrhyncha are not easily accommodated into any existing classification of genetic systems.

    Notes: Abstract Gustatory receptors associated with feeding in phytophagous insects are broadly categorized as phagostimulatory or deterrent. No phytophagous insect is known that tastes all its essential nutrients, and the ability to discriminate between nutrients is limited. The insects acquire a nutritional balance largely "adventitiously" because leaves have an appropriate chemical composition. Sugars are the most important phagostimulants. Plant secondary compounds are most often deterrent but stimulate phagostimulatory cells if they serve as host-indicating sign stimuli, or if they are sequestered for defense or used as pheromone precursors. The stimulating effects of chemicals are greatly affected by other chemicals in mixtures like those to which the sensilla are normally exposed. Host plant selection depends on the balance of phagostimulatory and deterrent inputs with, in some oligophagous and monophagous species, a dominating role of a host-related chemical. Evolution of phytophagy has probably involved a change in emphasis in the gustatory system, not fundamentally new developments. The precise role of the gustatory systems remains unclear. In grasshoppers, it probably governs food selection and the amounts eaten, but in caterpillars there is some evidence that central feedbacks are also involved in regulating the amount eaten.

    Notes: Abstract CD8 T cells respond to viral infections but also participate in defense against bacterial and protozoal infections. In the last few years, as new methods to accurately quantify and characterize pathogen-specific CD8 T cells have become available, our understanding of in vivo T cell responses has increased dramatically. Pathogen-specific T cells, once thought to be quite rare following infection, are now known to be present at very high frequencies, particularly in peripheral, nonlymphoid tissues. With the ability to visualize in vivo CD8 T cell responses has come the recognition that T cell expansion is programmed and, to a great extent, independent of antigen concentrations. Comparison of CD8 T cell responses to different pathogens also highlights the intricate relationship between microbially induced innate inflammatory responses and the kinetics, magnitude, and character of long-term T cell responses. This review describes recent progress in some of the major murine models of CD8 T cell-mediated immunity to viral, bacterial, and protozoal infection.

    Notes: Abstract A functional immune system requires the selection of T lymphocytes expressing receptors that are major histocompatibility complex restricted but tolerant to self-antigens. This selection occurs predominantly in the thymus, where lymphocyte precursors first assemble a surface receptor. In this review we summarize the current state of the field regarding the natural ligands and molecular factors required for positive and negative selection and discuss a model for how these disparate outcomes can be signaled via the same receptor. We also discuss emerging data on the selection of regulatory T cells. Such cells require a high-affinity interaction with self-antigens, yet differentiate into regulatory cells instead of being eliminated.

    Notes: Abstract In the absence of antiretroviral treatment, HIV-1 establishes a chronic, progressive infection of the human immune system that invariably, over the course of years, leads to its destruction and fatal immunodeficiency. Paradoxically, while viral replication is extensive throughout the course of infection, deterioration of conventional measures of immunity is slow, including the characteristic loss of CD4+ T cells that is thought to play a key role in the development of immunodeficiency. This conundrum suggests that CD4+ T cell-directed viral cytopathicity alone cannot explain the course of disease. Indeed, recent advances now indicate that HIV-1 pathogenesis is likely to result from a complex interplay between the virus and the immune system, particularly the mechanisms responsible for T cell homeostasis and regeneration. We review these data and present a model of HIV-1 pathogenesis in which the protracted loss of CD4+ T cells results from early viral destruction of selected memory T cell populations, followed by a combination of profound increases in overall memory T cell turnover, damage to the thymus and other lymphoid tissues, and physiological limitations in peripheral CD4+ T cell renewal.

    Notes: Abstract IL-13 was first recognized for its effects on B cells and monocytes, where it upregulated class II expression, promoted IgE class switching and inhibited inflammatory cytokine production. It was also thought to be functionally redundant with IL-4. However, studies conducted with knockout mice, neutralizing antibodies, and novel antagonists demonstrate that IL-13 possesses several unique effector functions that distinguish it from IL-4. Resistance to most gastrointestinal nematodes is mediated by type-2 cytokine responses, in which IL-13 plays a dominant role. By regulating cell-mediated immunity, IL-13 modulates resistance to intracellular organisms including Leishmania major, Leishmania mexicana, and Listeria monocytogenes. In the lung, IL-13 is the central mediator of allergic asthma, where it regulates eosinophilic inflammation, mucus secretion, and airway hyperresponsiveness. Manipulation of IL-13 effector function may also prove useful in the treatment of some cancers like B-cell chronic lymphocytic leukemia and Hodgkin's disease, where IL-13 modulates apoptosis or tumor cell growth. IL-13 can also inhibit tumor immunosurveillance. As such, inhibitors of IL-13 might be effective as cancer immunotherapeutics by boosting type-1-associated anti-tumor defenses. Finally, IL-13 was revealed as a potent mediator of tissue fibrosis in both schistosomiasis and asthma, which indicates that it is a key regulator of the extracellular matrix. The mechanisms that regulate IL-13 production and/or function have also been investigated, and IL-4, IL-12, IL-18, IFN-gamma, IL-10, TGF-beta, TNF-alpha, and the IL-4/IL-13 receptor complex play important roles. This review highlights the effector functions of IL-13 and describes multiple pathways for modulating its activity in vivo.

    Notes: Abstract Collectins and ficolins, present in plasma and on mucosal surfaces, are humoral molecules of the innate immune systems, which recognize pathogen-associated molecular patterns. The human collectins, mannan-binding lectin (MBL) and surfactant protein A and D (SP-A and SP-D), are oligomeric proteins composed of carbohydrate-recognition domains (CRDs) attached to collagenous regions and are thus structurally similar to the ficolins, L-ficolin, M-ficolin, and H-ficolin. However, they make use of different CRD structures: C-type lectin domains for the collectins and fibrinogen-like domains for the ficolins. Upon recognition of the infectious agent, MBL and the ficolins initiate the lectin pathway of complement activation through attached serine proteases (MASPs), whereas SP-A and SP-D rely on other effector mechanisms: direct opsonization, neutralization, and agglutination. This limits the infection and concurrently orchestrates the subsequent adaptive immune response. Deficiencies of the proteins may predispose to infections or other complications, e.g., reperfusion injuries or autoimmune diseases. Structure, function, clinical implications, and phylogeny are reviewed.

    Notes: Abstract The T helper lymphocyte is responsible for orchestrating the appropriate immune response to a wide variety of pathogens. The recognition of the polarized T helper cell subsets Th1 and Th2 has led to an understanding of the role of these cells in coordinating a variety of immune responses, both in responses to pathogens and in autoimmune and allergic disease. Here, we discuss the mechanisms that control lineage commitment to the Th1 phenotype. What has recently emerged is a rich understanding of the cytokines, receptors, signal transduction pathways, and transcription factors involved in Th1 differentiation. Although the picture is still incomplete, the basic pathways leading to Th1 differentiation can now be understood in in vitro and a number of infection and disease models.

    Notes: Abstract Dendritic cells (DCs) have several functions in innate and adaptive immunity. In addition, there is increasing evidence that DCs in situ induce antigen-specific unresponsiveness or tolerance in central lymphoid organs and in the periphery. In the thymus DCs generate tolerance by deleting self-reactive T cells. In peripheral lymphoid organs DCs also induce tolerance to antigens captured by receptors that mediate efficient uptake of proteins and dying cells. Uptake by these receptors leads to the constitutive presentation of antigens on major histocompatibility complex (MHC) class I and II products. In the steady state the targeting of DC antigen capture receptors with low doses of antigens leads to deletion of the corresponding T cells and unresponsiveness to antigenic rechallenge with strong adjuvants. In contrast, if a stimulus for DC maturation is coadministered with the antigen, the mice develop immunity, including interferon-gamma-secreting effector T cells and memory T cells. There is also new evidence that DCs can contribute to the expansion and differentiation of T cells that regulate or suppress other immune T cells. One possibility is that distinct developmental stages and subsets of DCs and T cells can account for the different pathways to peripheral tolerance, such as deletion or suppression. We suggest that several clinical situations, including autoimmunity and certain infectious diseases, can be influenced by the antigen-specific tolerogenic role of DCs.

    Notes: Abstract B cell chronic lymphocytic leukemia (B-CLL) is an accumulative disease of slowly proliferating CD5+ B lymphocytes that develops in the aging population. Whereas some patients with B-CLL have an indolent course and die after many years from unrelated causes, others progress very rapidly and succumb within a few years from this currently incurable leukemia. Over the past decade studies of the structure and function of the B cell antigen receptor (BCR) used by these leukemic cells have helped redefine the nature of this disease. In this review we summarize and reinterpret several aspects of these BCR-related studies and how they might relate to the disease. In particular, we address the ability of antigens to select out and drive B cell clones from the normal state to overt leukemic cells by binding to BCRs that are relatively unique and characteristic of B-CLL cells. The differential capacity of some B-CLL cases to continue to transduce signals through the BCR during the leukemic phase and the consequences for the in vivo biology of the leukemic clone is also considered. Finally, we discuss current and emerging views of the cellular origin of B-CLL cells and the differentiation pathways down which we believe these cells progress.

    Notes: Abstract The ability of the myocardium to successfully compensate for and adapt to environmental stress ultimately determines whether the heart will decompensate and fail or maintain preserved function. Despite the importance of the myocardial response to environmental stress, very little is known with respect to the biochemical mechanisms that are responsible for mediating and integrating the stress response in the heart. In the present review we summarize recent experimental material suggesting that the cytokines expressed within the myocardium in response to environmental injury, namely tumor necrosis factor (TNF), interleukin-1 (IL-1), and the interleukin-6 (IL-6) family, play an important role in initiating and integrating homeostatic responses. However, these stress-activated cytokines all have the potential to produce cardiac decompensation when expressed at sufficiently high concentrations. Accordingly, the theme to emerge from this review is that the short-term expression of stress-activated cytokines within the heart may be an adaptive response to stress, whereas long-term expression of these molecules may be frankly maladaptive by producing cardiac decompensation.

    Notes: Abstract Conventional kinesin is the prototypic member of a family of diverse proteins that use the chemical energy of ATP hydrolysis to generate force and move along microtubules. These proteins, which are involved in a wide range of cellular functions, have been identified in protozoa, fungi, plants, and animals and possess a high degree of sequence conservation among species in their motor domains. The biochemical properties of kinesin and its homologues, in conjunction with the recently solved three-dimensional structures of several kinesin motors, have contributed to our understanding of the mechanism of kinesin movement along microtubules. We discuss several models for movement, including the hand-over-hand, inchworm, and biased diffusion models of processive movement, as well as models of nonprocessive movement.

    Notes: Abstract DNA microarray technology is revolutionizing many aspects of biological research, allowing the expression of many thousands of gene transcripts to be monitored simultaneously. This provides powerful tools for the genome-wide correlation of gene transcript levels with physiological responses and alterations in physiological states. To date, microarray analyses have been applied almost exclusively to a few model species for which the abundant gene sequence data permit the fabrication of whole-genome microarrays. However, many interesting physiological traits and responses are poorly expressed or absent in model species and may be better illustrated in nonmodel organisms. Comparative approaches to understanding function traditionally focus on species that by virtue of their unusual adaptations, lifestyles, and phylogeny are particularly suited to address a specific biological process or problem. In this review, we show that microarray technology can be successfully applied to these nonmodel species and used to generate new insights of comparative and evolutionary significance into animal function.

    Notes: Abstract Here we explore the physiologic role of leptin as a liporegulatory hormone responsible for maintaining intracellular homeostasis in the face of wide variations in caloric intake. Normally, rats can tolerate a 60% fat diet because 96% of the surplus fat is deposited in adipocytes. In contrast, when leptin is congenitally absent or inactive, even on a normal diet, unutilized dietary fat is deposited in nonadipose tissues, causing dysfunction (lipotoxicity) and possible cell death (lipoapoptosis). We theorize that in diet-induced obesity, acquired leptin resistance may also develop as the result of increase in certain leptin resistance factors. Acquired leptin resistance occurs in aging, obesity, Cushing's syndrome, and acquired lipodystrophy, and preliminary evidence suggests that ectopic lipid deposition is increased. We speculate that the metabolic syndrome may be the human equivalent of the lipotoxic syndrome of rodents.

    Notes: Abstract The regulation of acid secretion in the stomach involves a complex network of factors that stimulate secretion in response to the ingestion of a meal and maintain homeostasis of gastric pH. Genetically engineered mouse models have provided a new opportunity to investigate the importance and function of specific molecules and pathways involved in the regulation of acid secretion. Mouse mutants with disruptions in the three major stimulatory pathways for acid secretion in parietal cells, gastrin, histamine, and acetylcholine, have been generated. Disruption of the gastrin pathway results in a major impairment in both basal and induced acid secretion. Histamine and acetylcholine pathway mutants also have significant alterations in acid secretion, although the impairment does not appear to be as severe as in gastrin pathway mutants, perhaps due in part to the hypergastrinemia that occurs. Mice with a disruption in the somatostatin pathway have increased gastric acid secretion, which confirms an important negative regulatory role for this factor. This review discusses these genetically engineered mouse models, as well as others, that provide insight into the complex regulation of in vivo gastric acid secretion. The regulation of growth and cellular morphology of the stomach in these mouse models is also presented. In addition, transgene promoters that are expressed in the gastric epithelium are discussed because these promoters will be important tools to alter cellular physiology in new mouse models in the future.

    Notes: Abstract Mechanosensation has been studied for decades, but understanding of its molecular mechanism is only now emerging from studies in Caenorhabditis elegans and Drosophila melanogaster. In both cases, the entry point proved to be genetic screens that allowed molecules needed for mechanosensation to be identified without any prior understanding of the likely components. In C. elegans, genetic screens revealed molecules needed for touch sensation along the body wall and other regions of force sensitivity. Members of two extensive membrane protein families have emerged as candidate sensory mechanotransduction channels: mec-4 and mec-10, which encode amiloride-sensitive channels (ASCs or DEG/ENaCs), and osm-9, which encodes a TRP ion channel. There are roughly 50 other members of these families whose functions in C. elegans are unknown. This article classifies these channels in C. elegans, with an emphasis on insights into their function derived from mutation. We also review the neuronal cell types in which these channels might be expressed and mediate mechanotransduction.

    Notes: Abstract To most people, concerns over the link between lipids and cardiovascular health most likely end with monitoring their daily consumption of dietary fats. However, it has become increasingly clear that, in addition to effects on adult cardiovascular physiology, lipids also play key roles in the formation of a functioning cardiovascular system. The lysophospholipids, lysophosphatidic acid (LPA) and sphingosine 1-phosphate (S1P), have come to the forefront as developmental and physiological regulators of the cardiovascular system. In this review, we discuss the function of the G protein-coupled receptors responsible for transducing LPA and S1P signals during development of the vertebrate cardiovascular system, focusing first on their role in angiogenesis and then on their function during embryonic development.

    Notes: Abstract Recent advances-both experimental and theoretical-provide a tentative image of the structures in Ca channels that make them exceptionally selective. The image is very different from K channels, which obtain high selectivity with a rigid pore that tightly fits K+ ions and is lined by carbonyl oxygens of the polypeptide backbone. Ca channels rely on four glutamate residues (the EEEE locus), whose carboxyl side chains likely reach into the pore lumen to interact with passing Ca2+ ions. The structure is thought to be flexible, tightly binding a single Ca2+ ion in order to block Na+ flux but rearranging to interact with multiple Ca2+ ions to allow Ca2+ flux. The four glutamates are not equivalent, a fact that seems important for Ca2+ permeation. This review describes the experimental evidence that leads to these conclusions and the attempts by theorists to explain the combination of high selectivity and high flux that characterizes Ca channels.

    Notes: Abstract This review addresses the mechanisms by which mitochondrial structure and function are regulated, with a focus on vertebrate muscle. We consider the adaptive remodeling that arises during physiological transitions such as differentiation, development, and contractile activity. Parallels are drawn between such phenotypic changes and the pattern of change arising over evolutionary time, as suggested by interspecies comparisons. We address the physiological and evolutionary relationships between ATP production, thermogenesis, and superoxide generation in the context of mitochondrial function. Our discussion of mitochondrial structure focuses on the regulation of membrane composition and maintenance of the three-dimensional reticulum. Current studies of mitochondrial biogenesis strive to integrate muscle functional parameters with signal transduction and molecular genetics, providing insight into the origins of variation arising between physiological states, fiber types, and species.

    Notes: Abstract To examine the role of the insulin receptor in fuel homeostasis, we and others have carried out genetic ablation studies in mice. Mice lacking insulin receptors are born with normal features, but develop early postnatal diabetes and die of ketoacidosis. In contrast, mice lacking insulin receptors in specific cell types as a result of conditional mutagenesis develop mild metabolic and reproductive abnormalities. These experiments have uncovered novel functions of insulin receptors in tissues such as brain and pancreatic beta-cells. Combined knockout studies of insulin and Igf1 receptors indicate that the insulin receptor also promotes embryonic growth. Experimental crosses of mice with insulin receptor haploinsufficiency have been instrumental to the genetic analysis of insulin action by enabling us to assign specific roles to different insulin receptor substrates and identify novel elements in insulin signaling.

    Notes: This review examines the convergence of recent anthropological interests in gender, labor, and globalization. Attention to gender and gender inequality offers a productive strategy for the analysis of globalizing processes and their local variations and contestations. Contemporary ethnographic research explores multiple dimensions of labor and gender inequalities in the global economy: gendered patterns of labor recruitment and discipline, the transnational mobility and commodification of reproductive labor, and the gendered effects of international structural adjustment programs, among others. New and continuing research explores the diverse meanings and practices that produce a gendered global labor force, incorporating the perspectives of men and women, masculinities and femininities, and examines how these processes of gender and labor inequality articulate with other structures of subordination (such as ethnicity and nationality) to shape lived experiences of work and livelihood, exploitation and struggle, around the world.

    Notes: Our understanding of developmental biology burgeoned during the last decade. This review summarizes recent advances, provides definitions and explanations of some basic principles, and does so in a way that will aid anthropologists in understanding their profound implications. Crucial concepts, such as developmental fields, selector and realizator genes, cell signaling mechanisms, and gene regulatory elements are briefly described and then integrated with the emergence of skeletal morphology. For the postcranium, a summary of events from limb bud formation, the appearance of anlagen, the expression of Hox genes, and the fundamentals of growth plate dynamics are briefly summarized. Of particular importance are revelations that bony morphology is largely determined by pattern formation, that growth foci such as physes and synovial joints appear to be regulated principally by positional information, and that variation in these fields is most likely determined by cis-regulatory elements acting on restricted numbers of anabolic genes downstream of selectors (such as Hox). The implications of these discoveries for the interpretation of both contemporary and ancient human skeletal morphology are profound. One of the most salient is that strain transduction now appears to play a much reduced role in shaping the human skeleton.     Notes: The social brain (or Machiavellian Intelligence) hypothesis was proposed to explain primates' unusually large brains: It argues that the cognitive demands of living in complexly bonded social groups selected for increases in executive brain (principally neocortex). The evidence for this and alternative hypotheses is reviewed. Although there remain difficulties of interpretation, the bulk of the evidence comes down in favor of the social brain hypothesis. The extent to which the cognitive demands of bonding large intensely social groups involve aspects of social cognition, such as theory of mind, is explored. These findings are then related to the evolution of social group size, language, and culture within the hominid lineage.

    Notes: Abstract We present preliminary evidence for a ~10,000-year earthquake record from two major fault systems based on sediment cores collected along the continental margins of western North America. New stratigraphic evidence from Cascadia demonstrates that 13 earthquakes ruptured the entire margin from Vancouver Island to at least the California border since the eruption of the Mazama ash 7700 years ago. The 13 events above this prominent stratigraphic marker have an average repeat time of 600 years, and the youngest event ~300 years ago coincides with the coastal record. We also extend the record of past earthquakes to the base of the Holocene (at least 9800 years ago), during which 18 events correlate along the same region. The sequence of Holocene events in Cascadia appears to contain a repeating pattern of events, a tantalizing first look at what may be the long-term behavior of a major fault system. The northern California margin cores show a cyclic record of turbidite beds that may represent Holocene earthquakes on the northern segment of the San Andreas Fault. Preliminary results are in reasonably good agreement with onshore paleoseismic data that indicate an age for the penultimate event in the mid-1600s at several sites and the most likely age for the third event of ~AD 1300.

    Notes: Abstract Considerable progress has been made over the past decade in understanding the static rheological properties of granitic magmas in the continental crust. Changes in H2O content, CO2 content, and oxidation state of the interstitial melt phase have been identified as important compositional factors governing the rheodynamic behavior of the solid/fluid mixture. Although the strengths of granitic magmas over the crystallization interval are still poorly constrained, theoretical investigations suggest that during magma ascent, yield strengths of the order of 9 kPa are required to completely retard the upward flow in meter-wide conduits. In low Bagnold number magma suspensions with moderate crystal contents (solidosities 0.1 〈=phi〈= 0.3), viscous fluctuations may lead to flow differentiation by shear-enhanced diffusion. AMS and microstructural studies support the idea that granite plutons are intruded as crystal-poor liquids (phi〈= 50%), with fabric and foliation development restricted to the final stages of emplacement. If so, then these fabrics contain no information on the ascent (vertical transport) history of the magma. Deformation of a magmatic mush during pluton emplacement can enhance significantly the pressure gradient in the melt, resulting in a range of local macroscopic flow structures, including layering, crystal alignment, and other mechanical instabilities such as shear zones. As the suspension viscosity varies with stress rate, it is not clear how the timing of proposed rheological transitions formulated from simple equations for static magma suspensions applies to mixtures undergoing shear. New theories of magmas as multiphase flows are required if the full complexity of granitic magma rheology is to be resolved.

    Notes: Abstract Is El Nino one phase of a continual, self-sustaining natural mode of the coupled ocean-atmosphere that has La Nina as the complementary phase? Or is El Nino a temporary departure from "normal" conditions "triggered" by a random disturbance such as a burst of westerly winds? A growing body of evidence-stability analyses, studies of the energetics, simulations that reproduce the statistics of sea surface temperature variations in the eastern equatorial Pacific-indicates that reality corresponds to a compromise between these two possibilities: The observed Southern Oscillation between El Nino and La Nina corresponds to a weakly damped mode that is sustained by random disturbances. This means that the predictability of El Nino is limited by the continual presence of "noise" so that forecasts should be probabilistic. The Southern Oscillation is also subject to decadal modulations. How it will be influenced by global warming is a matter of considerable uncertainty.

    Notes: Abstract Behavioral, anatomical, and physiological approaches can be integrated in the study of sound localization in barn owls. Space representation in owls provides a useful example for discussion of place and ensemble coding. Selectivity for space is broad and ambiguous in low-order neurons. Parallel pathways for binaural cues and for different frequency bands converge on high-order space-specific neurons, which encode space more precisely. An ensemble of broadly tuned place-coding neurons may converge on a single high-order neuron to create an improved labeled line. Thus, the two coding schemes are not alternate methods. Owls can localize sounds by using either the isomorphic map of auditory space in the midbrain or forebrain neural networks in which space is not mapped.

    Notes: Abstract Alzheimer's disease, Parkinson's disease, and motor neuron disease share a propensity to occur with increasing age and as either a sporadic or a familial disorder. A number of behavioral and environmental risk factors have been proposed for each disorder, but most associations lack consistency and specificity. Over the last decade the remarkable frequency of these disorders has become apparent, and the identification of mutations in genes has provided the means to understand their pathogenesis. Better and more accurate means to characterize and diagnose these diseases has greatly facilitated analytic epidemiology. The analysis of behavioral and genetic factors that may lower disease risk has led to clinical trials that are either in progress or being planned with the aim of preventing these disorders.

    Notes: Abstract A significant number of individuals have unexplained difficulties with acquiring normal speech and language, despite adequate intelligence and environmental stimulation. Although developmental disorders of speech and language are heritable, the genetic basis is likely to involve several, possibly many, different risk factors. Investigations of a unique three-generation family showing monogenic inheritance of speech and language deficits led to the isolation of the first such gene on chromosome 7, which encodes a transcription factor known as FOXP2. Disruption of this gene causes a rare severe speech and language disorder but does not appear to be involved in more common forms of language impairment. Recent genome-wide scans have identified at least four chromosomal regions that may harbor genes influencing the latter, on chromosomes 2, 13, 16, and 19. The molecular genetic approach has potential for dissecting neurological pathways underlying speech and language disorders, but such investigations are only just beginning.

    Notes: Abstract Over the past two decades significant progress has been made toward understanding the neural basis of primate decision making, the biological process that combines sensory data with stored information to select and execute behavioral responses. The most striking progress in this area has been made in studies of visual-saccadic decision making, a system that is becoming a model for understanding decision making in general. In this system, theoretical models of efficient decision making developed in the social sciences are beginning to be used to describe the computations the brain must perform when it connects sensation and action. Guided in part by these economic models, neurophysiologists have been able to describe neuronal activity recorded from the brains of awake-behaving primates during actual decision making. These recent studies have examined the neural basis of decisions, ranging from those made in predictable sensorimotor tasks to those unpredictable decisions made when animals are engaged in strategic conflict. All of these experiments seem to describe a surprisingly well-integrated set of physiological mechanisms that can account for a broad range of behavioral phenomena. This review presents many of these recent studies within the emerging neuroeconomic framework for understanding primate decision making.

    Notes: Abstract Transfer cells are plant cells with secondary wall ingrowths. These cells are ubiquitous, occurring in all plant taxonomic groups and in algae and fungi. Transfer cells form from differentiated cells across developmental windows and in response to stress. They are considered to play a central role in nutrient distribution by facilitating high rates of transport at bottlenecks for apo-/symplasmic solute exchange. These properties are conferred by their unique structural features-an invaginated secondary wall ensheathed by an amplified area of plasma membrane enriched in a suite of solute transporters. Recent development of transfer cell experimental systems, combined with technologies to image the three-dimensional structure of wall ingrowths, is allowing identification of inductive and regulatory signals, discovery of sequential processes involved in their differentiation, and a search for transfer cell identity genes. A model of key events in differentiation of a transfer cell is presented to highlight areas for future investigation.

    Notes: Abstract The term apomixis encompasses a suite of processes whereby seeds form asexually in plants. In contrast to sexual reproduction, seedlings arising from apomixis retain the genotype of the maternal parent. The transfer of apomixis and its effective utilization in crop plants (where it is largely absent) has major advantages in agriculture. The hallmark components of apomixis include female gamete formation without meiosis (apomeiosis), fertilization-independent embryo development (parthenogenesis), and developmental adaptations to ensure functional endosperm formation. Understanding the molecular mechanisms underlying apomixis, a developmentally fascinating phenomenon in plants, is critical for the successful induction and utilization of apomixis in crop plants. This review draws together knowledge gained from analyzing ovule, embryo, and endosperm development in sexual and apomictic plants. It consolidates the view that apomixis and sexuality are closely interrelated developmental pathways where apomixis can be viewed as a deregulation of the sexual process in both time and space.

    Notes: Abstract Many disease resistance (R) proteins of plants detect the presence of disease-causing bacteria, viruses, or fungi by recognizing specific pathogen effector molecules that are produced during the infection process. Effectors are often pathogen proteins that probably evolved to subvert various host processes for promotion of the pathogen life cycle. Five classes of effector-specific R proteins are known, and their sequences suggest roles in both effector recognition and signal transduction. Although some R proteins may act as primary receptors of pathogen effector proteins, most appear to play indirect roles in this process. The functions of various R proteins require phosphorylation, protein degradation, or specific localization within the host cell. Some signaling components are shared by many R gene pathways whereas others appear to be pathway specific. New technologies arising from the genomics and proteomics revolution will greatly expand our ability to investigate the role of R proteins in plant disease resistance.

    Notes: Abstract In the present genomics era, powerful reverse-genetic strategies are needed to elucidate gene and protein function in the context of a whole organism. However, most current techniques lack the generality and high-throughput potential of descriptive genomic approaches, such as those that rely on microarray hybridization. For example, in plant research, effective insertional mutagenesis and transgenic methods are limited to relatively few species or are inefficient. Fortunately, single-nucleotide changes can be induced in any plant by using traditional chemical mutagens, and progress has been made in efficiently detecting changes. Because base substitutions in proteins provide allelic series, and not just knockouts, this strategy can yield refined insights into protein function. Here, we review recent progress that has been made in genome-wide screening for point mutations and natural variation in plants. Its general applicability leads to the expectation that traditional mutagenesis followed by high-throughput detection will become increasingly important for plant functional genomics.

    Notes: Abstract The lignin biosynthetic pathway has been studied for more than a century but has undergone major revisions over the past decade. Significant progress has been made in cloning new genes by genetic and combined bioinformatics and biochemistry approaches. In vitro enzymatic assays and detailed analyses of mutants and transgenic plants altered in the expression of lignin biosynthesis genes have provided a solid basis for redrawing the monolignol biosynthetic pathway, and structural analyses have shown that plant cell walls can tolerate large variations in lignin content and structure. In some cases, the potential value for agriculture of transgenic plants with modified lignin structure has been demonstrated. This review presents a current picture of monolignol biosynthesis, polymerization, and lignin structure.

By their nature, these technologies reveal unexpected properties of biological systems. A second and more challenging aspect of omic technologies is the refined analysis of quantitative dynamics in biological systems. For metabolomics, gas and liquid chromatography coupled to mass spectrometry are well suited for coping with high sample numbers in reliable measurement times with respect to both technical accuracy and the identification and quantitation of small-molecular-weight metabolites. This potential is a prerequisite for the analysis of dynamic systems. Thus, metabolomics is a key technology for systems biology.     Notes: Abstract Phosphorylation and dephosphorylation of a protein often serve as an "on-and-off" switch in the regulation of cellular activities. Recent studies demonstrate the involvement of protein phosphorylation in almost all signaling pathways in plants. A significant portion of the sequenced Arabidopsis genome encodes protein kinases and protein phosphatases that catalyze reversible phosphorylation. For optimal regulation, kinases and phosphatases must strike a balance in any given cell. Only a very small fraction of the thousands of protein kinases and phosphatases in plants has been studied experimentally. Nevertheless, the available results have demonstrated critical functions for these enzymes in plant growth and development. While serine/threonine phosphorylation is widely accepted as a predominant modification of plant proteins, the function of tyrosine phosphorylation, despite its overwhelming importance in animal systems, had been largely neglected until recently when tyrosine phosphatases (PTPs) were characterized from plants. This review focuses on the structure, regulation, and function of protein phosphatases in higher plants.

    Notes: Abstract Cellular and whole organism iron homeostasis must be balanced to supply enough iron for metabolism and to avoid excessive, toxic levels. To perform iron uptake from the environment, iron distribution to various organs and tissues, and iron intracellular compartmentalization, various membranes must be crossed by this metal. The uptake and transport of iron under physiological conditions require particular processes such as chelation or reduction because ferric iron has a very low solubility. The molecular actors involved in iron acquisition from the soil have recently been characterized. A few candidates belonging to various gene families are hypothesized to play major roles in iron distribution throughout the plant. All these transport activities are tightly regulated at transcriptional and posttranslational levels, according to the iron status of the plant. These coordinated regulations result from an integration of local and long-distance transduction pathways.

For Lolium temulentum, which flowers in response to a single long day (LD), GAs are a transmitted signal, their content increasing in the leaf early in the LD and then, hours later, at the shoot apex. There is a continuous trail of evidence of hormonal action of these GAs for L. temulentum and support for a similar role in the flowering of other LD-responsive temperate grasses and cereals. A characteristic of the initial flowering responses of grasses and cereals is their limited stem elongation. Interestingly, it is GAs with low effectiveness for stem elongation, GA5 and GA6, that reach the shoot apex and, structurally, are probably not degraded by 2-oxidase enzymes. By contrast, GA1 and GA4 cause stem elongation, may be inactive for floral evocation, and do not reach the vegetative shoot apex apparently because of susceptibility to degradation. However, GA4 can be florally active if protected against 2-oxidases either structurally or by using a 2-oxidase inhibitor. Later in inflorescence development, GA1 and GA4 can be detected at the shoot apex and are florally active if applied. The 2-oxidase restricting accessibility to the apex has probably declined at this time so there is a second florigenic, LD-regulated GA action. A growing body of molecular evidence supporting these actions of GA may provide a future basis for manipulating flowering of grasses and cereals.

    Notes: Abstract In this review we focus on photosynthetic behavior of overwintering evergreens with an emphasis on both the acclimative responses of photosynthesis to cold and the winter behavior of photosynthesis in conifers. Photosynthetic acclimation is discussed in terms of the requirement for a balance between the energy absorbed through largely temperature-insensitive photochemical processes and the energy used for temperature-sensitive biochemical processes and growth. Cold acclimation transforms the xanthophyll-mediated nonphotochemical antenna quenching of absorbed light from a short-term dynamic response to a long-term sustained quenching for the whole winter period. This acclimative response helps protect the evergreen foliage from photooxidative damage during the winter when photosynthesis is restricted or prevented by low temperatures. Although the molecular mechanisms behind the sustained winter excitation quenching are largely unknown, it does involve major alterations in the organization and composition of the photosystem II antenna. In addition, photosystem I may play an important role in overwintering evergreens not only by quenching absorbed light photochemically via its support of cyclic electron transport at low temperatures, but also by nonphotochemical quenching of absorbed light irrespective of temperature. The possible role of photosystem II reaction centers in nonphotochemical quenching of absorbed energy in overwintering evergreens is also discussed. Processes like chlororespiration and cyclic electron transport may also be important for maintaining the functional integrity of the photosynthetic apparatus of overwintering evergreens both during periods of thawing in winter and during recovery from winter stress in spring. We suggest that the photosynthetic acclimation responses of overwintering evergreens represent specific evolutionary adaptations for plant species that invest in the long-term maintenance of leaf structure in cold climatic zones as exemplified by the boreal forests of the Northern Hemisphere.

    Notes: Abstract Potassium (K+) plays a number of important roles in plant growth and development. Over the past few years, molecular approaches associated with electrophysiological analyses have greatly advanced our understanding of K+ transport in plants. A large number of genes encoding K+ transport systems have been identified, revealing a high level of complexity. Characterization of some transport systems is providing exciting information at the molecular level on functions such as root K+ uptake and secretion into the xylem sap, K+ transport in guard cells, or K+ influx into growing pollen tubes. In this review, we take stock of this recent molecular information. The main families of plant K+ transport systems (Shaker and KCO channels, KUP/HAK/KT and HKT transporters) are described, along with molecular data on how these systems are regulated. Finally, we discuss a few physiological questions on which molecular studies have shed new light.

    Notes: Abstract Plant systems utilize a diverse array of cytochrome P450 monooxygenases (P450s) in their biosynthetic and detoxicative pathways. Those P450s in biosynthetic pathways play critical roles in the synthesis of lignins, UV protectants, pigments, defense compounds, fatty acids, hormones, and signaling molecules. Those in catabolic pathways participate in the breakdown of endogenous compounds and toxic compounds encountered in the environment. Because of their roles in this wide diversity of metabolic processes, plant P450 proteins and transcripts can serve as downstream reporters for many different biochemical pathways responding to chemical, developmental, and environmental cues. This review focuses initially on defining P450 biochemistries, nomenclature systems, and the relationships between genes in the extended P450 superfamily that exists in all plant species. Subsequently, it focuses on outlining the many approaches being used to assign function to individual P450 proteins and gene loci. The examples of assigned P450 activities that are spread throughout this review highlight the importance of understanding and utilizing P450 sequences as markers for linking biochemical pathway responses to physiological processes.

    Notes: Abstract Cryptochromes are photosensory receptors mediating light regulation of growth and development in plants. Since the isolation of the Arabidopsis CRY1 gene in 1993, cryptochromes have been found in every multicellular eukaryote examined. Most plant cryptochromes have a chromophore-binding domain that shares similar structure with DNA photolyase, and a carboxyl terminal extension that contains a DQXVP-acidic-STAES (DAS) domain conserved from moss, to fern, to angiosperm. In Arabidopsis, cryptochromes are nuclear proteins that mediate light control of stem elongation, leaf expansion, photoperiodic flowering, and the circadian clock. Cryptochromes may act by interacting with proteins such as phytochromes, COP1, and clock proteins, or/and chromatin and DNA. Recent studies suggest that cryptochromes undergo a blue light-dependent phosphorylation that affects the conformation, intermolecular interactions, physiological activities, and protein abundance of the photoreceptors.

    Notes: Since the completion of the human genome sequencing project, the discovery and characterization of human genetic variation is a principal focus for future research. Comparative studies across ethnically diverse human populations and across human and nonhuman primate species is important for reconstructing human evolutionary history and for understanding the genetic basis of human disease. In this review, we summarize data on patterns of human genetic diversity and the evolutionary forces (mutation, genetic drift, migration, and selection) that have shaped these patterns of variation across both human populations and the genome. African population samples typically have higher levels of genetic diversity, a complex population substructure, and low levels of linkage disequilibrium (LD) relative to non-African populations. We discuss these differences and their implications for mapping disease genes and for understanding how population and genomic diversity have been important in the evolution, differentiation, and adaptation of humans.

    Notes: Given the unique biological requirements of sound transduction and the selective advantage conferred upon a species capable of sensitive sound detection, it is not surprising that up to 1% of the approximately 30,000 or more human genes are necessary for hearing. There are hundreds of monogenic disorders for which hearing loss is one manifestation of a syndrome or the only disorder and therefore is nonsyndromic. Herein we review the supporting evidence for identifying over 30 genes for dominantly and recessively inherited, nonsyndromic, sensorineural deafness. The state of knowledge concerning their biological roles is discussed in the context of the controversies within an evolving understanding of the intricate molecular machinery of the inner ear.

    Notes: Human narcolepsy is a genetically complex disorder. Family studies indicate a 20-40 times increased risk of narcolepsy in first-degree relatives and twin studies suggest that nongenetic factors also play a role. The tight association between narcolepsy-cataplexy and the HLA allele DQB1*0602 suggests that narcolepsy has an autoimmune etiology. In recent years, extensive genetic studies in animals, using positional cloning in dogs and gene knockouts in mice, have identified abnormalities in hypothalamic hypocretin (orexin) neurotransmission as key to narcolepsy pathophysiology. Though most patients with narcolepsy-cataplexy are hypocretin deficient, mutations or polymorphisms in hypocretin-related genes are extremely rare. It is anticipated that susceptibility genes that are independent of HLA and impinge on the hypocretin neurotransmitter system are isolated in human narcolepsy.

    Notes: Geneticists are interested in finding genes associated with disease. Because of widespread health disparities, race is a variable that is often said to be relevant in this context. The idea is that members of a preconceived "race" share common ancestry that may include genetic risk factors. Human variation has been shaped by the long-term processes of population history, and population samples that reflect that history carry statistical information about shared genetic variation or "ancestry." But race is an elusive concept and a term difficult even to define rigorously. Unfortunately, these problems are neither new nor related to recent genetic knowledge. Race is also one of the most politically charged subjects in American life because its associated sociocultural component has notoriously led to categorical treatment that has been misleading and politically misused. There are ways in which the concept of race (whether or not the term is used) can be a legitimate tool in the search for disease-associated genes. But in that context race reflects deeply confounded cultural as well as biological factors, and a careful distinction must be made between race as a statistical risk factor and causal genetic variables.
