    Notes: A preparative electrolysis apparatus for isoelectric focusing of large amounts of proteins is described. It contains 46 separation compartments, and its total volume is about 7.6 liters. The compartments are closed, and internal cooling and stirring are effected by slow rotation of the whole apparatus in a tank filled with cold water. The apparatus can be run with an electric load of up to 5 kW, and an isoelectric focusing takes 2-3 days. It has been shown that crude pepsin can be purified in this apparatus. Preparations of hemoglobin and bovine whey have also been run in the apparatus. Fourteen grams of whey protein has been completely separated into its main components, serum albumin (pI 4.60), α-lactalbumin (pI 5.01), and β-lactoglobulin (pI 5.13-5.23), whereas the latter component was partially separated into its subcomponents A and B.

    Notes: A pH gradient providing maximal separation between pH 4.7 and 4.9 was generated by systematic variation of carrier constituent mixtures, and/or selection of anolyte/catholyte. In the optimized pH gradient, separation on gel electrofocusing between proteins with pI's between 4.7 and 4.9 was shown to be superior to that achieved on relatively steep pH gradients. This was the case in the separation of either human growth hormone isohormones D and E, or of bovine serum albumin and ovalbumin.

    Notes: In isoelectric focusing on polyacrylamide gel, pH gradients exhibited a systematic shift along the pH-axis in proportion to the pH of the anolyte. Thus, more acidic anolytes produced more acidic pH gradients, and more basic ones gave rise to more basic pH gradients. In all cases the pH's of the anolyte and catholyte were kept outside the pI-range of the carrier ampholytes used. Increasing the pH of the anolyte stabilized pH gradients, while the pH of the catholyte, within the range tested, had no systematic effect on pH gradient stabilization.From a practical viewpoint, modification of pH gradients by the anolyte provides increased flexibility in pH gradient design for electrofocusing. From a theoretical viewpoint, the selective anolyte effect on pH gradients in isoelectric focusing on polyacrylamide gels supports the hypothesis previously advanced by Murel et al., which relates pH gradient decay in electrofocusing to progressive protonation from the anodic pH gradient terminus.

    Notes: The photometry of two-dimensional electrophoretograms is described in some detail. This method has a high sensitivity of 5·10-4 O.D., because it takes advantage of the noise characteristics of photometric signals. Digital image analysis is used to locate and evaluate the individual spots on the electrophoretogram. The presented procedure of segmentation detects the faintest protein spots visible by eye and separates even confluent spots reliably and automatically. The combination of scanning photometry and image analysis is a sensitive, versatile, and accurate method for the evaluation of slab gels.

    Notes: Polyacrylamide gel isoelectric focusing was carried out using 250 μm thick gels backed with polyester films at voltage gradients up to 460 V/cm. Peltier cooling was employed with berrylium oxide heat transfer plates to achieve adequate cooling. Visualization of low protein concentrations was obtained by the use of a highly sensitive silver staining technique. Results indicated that over separating distances of 5.4 cm 150 to 200 bands should be resolved and that considerable differences between two commercial carrier ampholytes exist. Pharmalyte carrier ampholytes from Pharmacia have better continuity and conductance over the range of pH 3-10 but require much higher field strengths to achieve maximal separation than do LKB Ampholines over a similar pH range.

    Notes: A simple and versatile technique of horizontal slab gel electrofocusing is described which is based on a discontinuity of the gel volume along the pH axis achieved by overlaying ampholyte containing gel strips, of a defined volume and shape, at various locations on a base gel during the prerun of isoelectric focusing. In the electric field the overlaid gel strip delivers ampholytes of a distinct pI interval into the strip-free part of the base gel and removes from the latter some ampholytes present in the system. Flattening of the pH gradient in the strip-free part of the gel is the consequence. The technique is especially useful for thin gels and can be performed with any commercially available horizontal slab apparatus.

    Notes: We have demonstrated the possibility of performing fingerprints in the single isoelectric focusing dimension, able to resolve at least 50 peptide zones, down to a minimum critical length of 12 amino acids. Isoelectric focusing can be coupled to SDS-gel gradient electrophoresis, provided the fragments have a minimum molecular mass of 3500 daltons. We have applied this system to the analysis of digests of casein micelles. Upon digestion with extracts of lactobacilli, α and β caseins are broken preferentially into two major fragments, a pI 7.1 chain of 10 000 daltons, and a pI 6.4 chain of 14 000 daltons. Upon digestion with an extract of L. jugurti, only two fragments remain undigested, a pI ∼ 3 chain of 5500 daltons (probably the 43 to 90 amino acid fragment of α casein) and a pI ∼ 9 chain of 4200 daltons (probably the 1 to 36 amino acid sequence of casein).

    Notes: Ultrathin-layer isoelectric focusing in 50-100 μm polyacrylamide gels covalently bound to glass plates or polyester films, pretreated with methacryloxypropyl-trimethoxy-silane, is described. The gels adhere firmly to the silanized supports during all steps of operation and visualization. Ultrathin gels are prepared by the new and simple “flap technique”. Artificial mixtures of marker proteins and crude fungal enzymes were focused on 5-25 cm separation distances. In 11 cm gels containing pH 4-9 “Servalyt T” carrier ampholytes, 2-3 zones per mm can be resolved, with a zone width of 70-150 μm and differences in pI values of 0.023-0.033 pH. The gel volume per focused sample is 15-120 μl and up to 100 samples can be analyzed on 12.5 × 26 cm gels. Polyacrylamide gels of intermediate and high porosity, e. g. 5 % T, 3 % C and 3 % T, 20 % C, are compared. Small marker proteins attain equilibrium in both gels after focusing for 1300-2000 V x h at a final field strength of 100 V/cm, whereas thyroglobulin (690 000) and ferritin (465 000), applied at the anode and cathode, coalesce only in high-porosity gels. pH Gradients were determined, with the aid of a glass electrode, directly on the gel surface for wide range carrier ampholytes “Servalyt T”, “Ampholine”, and “Pharmalyte” after focusing for 2000-5000 V × h. Identical focusing patterns are found for some marker proteins and crude fungal enzymes in different carrier ampholytes. The effect of purification of commercial carrier ampholytes by ultrafiltration and charcoal treatment, and of “forced aging” at 80°C, on isoelectric focusing of marker proteins was studied. The molecular size distribution of different carrier ampholytes was assessed by thin-layer gel chromatography on Bio-Gel P-10. Protein staining with Serva Blue G, Serva Blue R and Serva Violet 49 is described. The sensitivity of protein detection is 50-100 ng/mm2. Fixation, staining and destaining can be completed within 10-15 min in 50 μm gels. Ultrathin-layer isoelectric focusing on silanized glass plates or polyester films recommends itself as an excellent analytical technique for research and routine applications. The method combines high resolution, speed, versatility and reagent economy with simplicity of operation.
