    Description / Table of Contents: Abstract The application of differtent fractionation procedures has shown that ferritin components separated by gel electrophoresis in polyacrylamide differ in respect to solubility and to behaviour in chromatography and gel filtration on Sephadex G-200. The ferritin has been identified with ferrocyanid or—after exchange with59Fe—by its radiation. On extraction of a ferritin precipitate with solutions of ammonium sulphate of decreasing molarity α-ferritin was distributed over the whole extraction range. The slower β-, γ- und δ-ferritins appeared in increasing amounts in solutions of decreasing molarities. Differences were observed in the extraction of ferritins of different origin (horse, beef, human). Chromatography of ferritin on DEAE cellulose gave several peaks. In paper electrophoresis, the mobility of the chromatographically different ferritins was the same. In gel electrophoresis, however, differences in the distribution of the ferritin components of these peaks were found. A partial separation of the different ferritin components was also obtained in gel filtration. The leading part of the only peak contained the β-, γ- and δ-components, while α-ferritin was found in the trailing part. This result may be ascribed to differences in the molecular sizes of the components.
    Notes: Zusammenfassung Die Anwendung verschiedener Fraktionierverfahren zeigt, daß die bei der Gelelektrophorese in Polyacrylamid gefundenen Ferritin-Komponenten sich auch bezüglich der Löslichkeit, der chromatographischen Eigenschaften und des Verhaltens bei der Gelfiltration auf Sephadex G-200 unterscheiden. Das Ferritin wurde mit Ferrocyanid oder — nach Austausch mit59Fe — durch die Aktivität identifiziert. Bei der Extraktion eines Niederschlages von Ferritin mit Ammoniumsulfatlösungen fallender Molarität war das α-Ferritin über den ganzen Extraktionsbereich verteilt. Die β-, γ- und δ-Ferritine wurden in zunehmender Menge mit Lösungen fallender Molarität extrahiert. Es wurden Unterschiede in der Extraktion von Ferritin verschiedenen Ursprungs (Pferd, Rind, Mensch) festgestellt. Bei der Chromatographie von Ferritin auf DEAE-Cellulose wurden mehrere Peaks erhalten. Bei der Papierelektrophorese wanderte das Ferritin verschiedener chromatographischer Peaks mit identischer Beweglichkeit, während sich bei der Gelelektrophorese deutliche Unterschiede in der Verteilung der Ferritin-Komponenten dieser Peaks zeigten. Eine partielle Trennung der Ferritin-Komponenten konnte auch bei der Gelfiltration erreicht werden. Der aufsteigende Teil des einzigen Peaks enthielt die elektrophoretisch langsameren β-, γ- und δ-Ferritine, im absteigenden Teil des Peaks war das α-Ferritin lokalisiert. Dieses Ergebnis weist auf Unterschiede in der Molekülgröße der Komponenten hin.

    Notes: Zusammenfassung Unter Einwirkung von Hitze (90°C) auf die Meerrettich-Peroxidase wurden die basischen Isoenzyme bevorzugt inaktiviert. Eine quantitative Verlagerung der Isoenzymverteilung mit einer Umwandlung zu Isoenzymen mit niedrigeren isoelektrischen Punkten wurde beobachtet. Es wurden nur geringe Mengen neuer enzymatisch aktiver Komponenten mit Hilfe der dünnschicht-isoelektrischen Fokussierung gefunden. Mittels der Dünnschicht-Gelfiltration wurde eine Aggregatbildung gefunden, wobei die Aggregate ein sehr hohes Molekulargewicht (〉 500 000) aufwiesen und nur Spuren von Aktivität aufzeigten.

    Notes: Summary Horseradish peroxidase has beenγ-irradiated in dilute aqueous solution under argon, air, and nitrous oxide. The results indicate that the OH⋅ radicals are of primary importance for the radiation inactivation of peroxidase. By thin-layer isoelectric focussing a shift in the distribution of enzyme activity towards lower isoelectric points was detected in the order argon 〈 air 〈 N2O. Thin-layer gel chromatography revealed enzymatically active aggregates, the formation of which could be attributed mainly to the OH⋅ radicals.

    Description / Table of Contents: Abstract The water soluble proteins of the unicellular freshwater alga Chlorella pyrenoidosa were obtained after homogenization of the cells. The proteins were then fractionated either by DEAE chromatography, followed by electrophoresis in polyacrylamide gel, or by gel filtration in Sephadex G-200. The proteins were found to consist of many components differing in electric charge and molecular size. Some of these components may be characterized by combined chromatography and electrophoresis. Appreciable amounts of nucleic acid have been found in chromatography only in the fraction eluted last. In contrast, in gel filtration nucleic acid is distributed over several fractions.
    Notes: Zusammenfassung Die wasserlöslichen Proteine aus der einzelligen süßwasseralge Chlorella pyrenoidosa wurden nach Homogenisierung der Zellen erhalten. Die Proteine wurden dann einerseits durch Chromatographie auf DEAE-Zellulose, gefolgt von Elektrophorese in Polyacrylamid-Gel, und anderseits durch Gel-Filtration in Sephadex G-200 fraktioniert. Man findet, daß die Proteine nach elektrischer Ladung und Molekulargewicht aus vielen Komponenten bestehen. Einige dieser Komponenten lassen sich jedoch durch die Methode der kombinierten Chromatographie und Elektrophorese charakterisieren. Wesentliche Gehalte an Nukleinsäure wurden bei der Chromatographie nur in der letzten — am schwersten eluierbaren — Fraktion gefunden. Dagegen ist die Nukleinsäure bei der Gel-Filtration über mehrere Fraktionen verteilt.

    Notes: A washed carrot substrate, prepared with high yields and easy handling properties, was found to be a suitable substrate for studying cellulolytic and pectinolytic degradation processes. A cellulase from Trichoderma reesei, and Rohament P, a macerating enzyme from Aspergillus alleaceǔs in endopolygalacturonase, degraded the washed carrot substrate to an extent of 60%. With the combined action of both enzymes, degradation was more than 80%. Simultaneous action of both enzymes was more efficient than their sequential use. The effect of temperature, pH, incubation time, enzyme concentration, and substrate concentration on the degradation by the single enzymes and their mixture were studied. Gas chromatographic sugar analysis revealed that Rohament P liberated glucose, arabinose, and galactose in the low-molecular-weight fraction obtained by ultrafiltration, in addition to high amounts of galacturonic acid. These carbohydrates were also found in the high-molecular-weight fraction (retentate) together with rhamnose and mannose. Cellulase BC released mainly glucose, although galacturonic acid, arabinose, xylose, and mannose were also detected both in the ultrafiltrate and retentate. Morphologically, during Rohament P degradation of the washed carrot substrate, damaged tissues and disintegrated cells were seen, whereas on cellulase BC action mainly disintegrated cell walls were observed.
