    Notes: Abstract Point defect agglomerates in dislocation-free silicon crystals, usually called “swirls”, have been investigated by means of high-voltage electron microscopy. It was found that a single swirl defect consists of a dislocation loop or a cluster of dislocation loops. By contrast experiments it could be shown that these loops are formed by agglomeration of self-interstitial atoms. Generally the loops have a/2〈110〉 Burgers vectors, but in specimens with high concentrations of carbon (∼1017 cm−3) and oxygen (∼1016 cm−3) also dislocation loops including a stacking fault were observed. In crystals grown at growth rates higher thanv=4 mm/min no swirls are observed; lower growth rates do not markedly affect the size and shape of the dislocation loops. With decreasing impurity content (particulary of oxygen and carbon) the swirl density decreases, whereas the dislocation loop clusters become larger and more complex. A model is presented which describes the formation of swirls in terms of agglomeration of silicon self-interstitials and impurity atoms.

    Notes: Summary The pharmacokinetics of the anti-epileptic drug di-n-propylacetate (DepakineR) have been studied in 7 patients, in whom plasma concentrations were determined during and following subchronic treatment. Elimination of the drug appeared to follow a monophasic exponential course; biological half lives were 8 to 15 hours. The data supported the assumption that an open one-compartment model can be used to describe the kinetics of dipropylacetate in man. The drug appeared to have a relatively restricted distribution: calculated relative distribution volumes ranged from 0.15 to 0.40 1/kg. There were large interindividual differences in clearance rate. The therapeutic range was considered to be between 50 and 100 mg/1 plasma. Plasma levels of phenobarbital were markedly raised during treatment with dipropylacetate for an unknown reason. Determination of the plasma concentrations of drugs at accurately fixed times appears to be a reliable method for pharmacotherapeutic monitoring of epileptic patients.

    Notes: Summary An oral dose of 5 mg of14C-isosorbide dinitrate was rapidly absorbed, biotransformed and excreted by human subjects. Peak whole blood concentrations of radioactivity were reached after 1.5 to 2 hours and declined relatively slowly. The radioactivity in whole blood mainly represented metabolites, isosorbide mononitrates. The peak concentrations found were 4.5, 11.7 and 34.3 ng/ml of isosorbide dinitrate, isosorbide 2-mononitrate and isosorbide 5-mononitrate, respectively, in the blood of one subject and 5.9, 15 and 61.3 ng/ml, respectively, in the blood of another subject. However, concentrations of the metabolites declined relatively slowly during 6 h after the oral dose. Up to 99% of an oral dose of isosorbide dinitrate was excreted during 5 days, mainly in the urine of the first day (ca. 78%). The results showed that isosorbide mononitrates were available to contribute to the pharmacological action.

    Notes: Summary In 11 healthy untrained volunteers the increase in plasma dopamine-β-hydroxylase (DBH) activity during graded physical exercise has been examined as a true measure of increased activity of the sympathetic nervous system. The correlation between DBH activity, catecholamine concentration (CA) in plasma and heart rate was studied. When work on an electrically braked bicycle ergometer was gradually increased from 12.5 to 100, 200 and 300 watts there was a linear increase in DBH activity and heart rate; the increase in CA concentrations followed an exponential function. The peak values for DBH activity and CA concentration in plasma after the 300 watt work load (as percentages of the resting levels) were 130±3% and 820±71%, respectively; the adrenaline concentration in plasma increased only to 150±19% (p〉0.05). There were significant correlations between heart rate and work load, DBH and work load and log CA and work load. The data imply direct correlations between heart rate and DBH, heart rate and log CA and DBH and log CA. The exponential increase in noradrenaline concentration in plasma might be due either to a greater net “overflow” from sympathetic nerve endings, and/or to increased secretion by the adrenal medulla. In the latter case, the release of noradrenaline would not be accompanied by secretion either of adrenaline or DBH. After work ceased there were sharp falls in heart rate and CA concentration, which indicate an immediate drop in sympathetic activity. DBH activity in plasma returned to normal very slowly; it reached half maximum values after 20 – 22 min. It is concluded that increased sympathetic activity in man can be estimated in vivo as changes in DBH and/or CA concentration in plasma. In contrast, a rapid decrease in sympathetic activity is directly reflected only by a rapid fall in the plasma concentrations of CA.

    Notes: Summary The diminished sympathomimetic pressor activity of monohydroxylated phenylalkylamines after oral administration has been attributed to incomplete enteric absorption. Therefore, urinary excretion of the unchanged drug and its metabolites has been compared after intravenous and oral administration of3H-m-octopamine to eight patients. Identical amounts of3H-activity (80% of the dose) were excreted after the two routes of dosing, so enteric absorption has been assumed to be complete. Significant differences were found in the fraction of free urinarym-octopamine, which amounted to 10.5% of the dose after infusion and 0.58% after oral administration. The only metabolic pathways form-octopamine are deamination and conjugation. Following oral administration the percentage of conjugates was considerably higher than after intravenous infusion. This metabolic pattern appears typical of all phenylalkylamines with a hydroxyl group in themeta position. Ring hydroxylation to catecholamines was not observed. The enzymes mainly responsible for conjugation after oral administration are located in the gut wall. The resulting “first pass effect”, i.e. metabolism prior to the access to the central compartment, can account for the diminished pharmacodynamic effect after dosing by this route.

    Notes: Summary After oral administration of oxyfedrine to healthy volunteers, norephedrine was identified in the urine by thin layer chromatography and gas liquid chromatography and mass spectrography. 30 hours after single oral doses of 8, 16 or 24 mg of oxyfedrine, about 4, 8 and 9 mg, respectively, of norephedrine were found in the urine, i.e. on a molar base 75–100% of the dose was excreted as norephedrine. The peak of excretion occurred within 2–4 hours after administration of the drug. No accumulation of oxyfedrine and/or its metabolite was observed after administration of 16 mg of oxyfedrine t.i.d. for three days. It could not be decided whether oxyfedrine was metabolized to norephedrine by liver enzymes, as in rats, or was spontaneously degraded to norephedrine, e.g. in duodenal fluid before absorption. 30–150 min after oral oxyfedrine (24 mg) norephedrine was demonstrable in duodenal fluid. Thus, in addition to the directβ-sympathomimetic effects of oxyfedrine, it may also have indirect sympathomimetic effects because of the noradrenaline-releasing properties of its metabolite norephedrine.

    Notes: Summary It is apparent from studying recent articles on pharmacokinetics that a number of misunder-standings exist, both about the design of experiments and the analysis of results. The purpose of this paper is to outline many of the common pitfalls associated with the design of experiments and also the limitations upon the analysis of results. The paper describes mathematical, laboratory and clinical aspects which must be examined in designing a protocol for pharmacokinetic experiments. Simulated data is presented to demonstrate the dangers of using standard computer programs for parameter estimation. Even when convergence is obtained the answers may be dependent on the method employed. A mathematical model is of little use unless a reasonable amount of good, accurate data is obtained.

    Notes: Summary Clonazepam (CNP) and its principal metabolite in plasma, 7-amino-CNP (ACNP), have been investigated in a prospective study of 27 newly diagnosed epileptics and correlated with specified side-effects. At a daily dose of 6 mg, the average plasma levels of both substances were about 50ng/ml, and individual values ranged from 30 to about 80ng/ml. There was a linear correlation between changes in dose and the resulting plasma levels, which indicates first order elimination kinetics. Side-effects were frequent, but neither their severity nor their occurrence could be related to plasma levels or to the rate of increase in plasma concentration of the drug. Three out of five patients who developed serious dysphoria had significantly high CNP levels. The concentration of ACNP was considerably increased in four patients who subsequently suffered from withdrawal symptoms. Drug interaction with diphenylhydantoin, i.e. decreased CNP level, was observed in all five patients who received both compounds. In general it is not yet possible to define an upper limit for the plasma levels of CNP and ACNP at which toxicity occurs. In patients treated with conventional doses of CNP, measurement of plasma concentration is not required, except in special circumstances, because of the lack of correlation between plasma level and side-effects.

    Notes: Summary After administration of a single loading dose (80 mg p.o.) of fluorophenindione, the prothrombin level decreased to 37 % in 24 h, and the effect lasted for 48 h. Accordingly, fluorophenindione can be classified as an anticoagulant with an “intermediate” effect. Its elimination half-life was 31 h, which is longer than that of phenindione, because of the greater stability of the fluorinated derivate.

The data were evaluated according to a two compartment open model. The calculated best-fitting curves were in good agreement with the experimental data, better than could be expected from a simpler model. This justifies the assumption that the kinetics of nortriptyline in man may be described by this model with an appropriate input function. The data permitted estimation of all the parameters of the model. The meaning of the parameters is discussed, particularly in relation to individual variation.

    Notes: Summary A quantitative thin layer chromatographic (TLC) method has been developed for determination of the antiarrhythmic quaternary ammonium compound N, N-bis (phenylcarbamoylmethyl) dimethylammonium chloride (QX-572) in biological materials. Prior to chromatography QX-572 was transferred into chloroform as perchlorate by ion pair extraction. Tritium-labelled QX-572 was used as the internal standard and a TLC scanning spectrophotometer equipped with a linear detector system afforded the required accuracy, specificity and simplicity. The method was used to determine QX-572 in plasma from 11 patients with various cardiac diseases who received QX-572 8 mg/kg body wt. as an intravenous infusion over 30 min. There was a rapid initial decay of the plasma levels from 11.0±1.1 µg/ml (mean ± SE) at the end of infusion to 3.5±0.5 µg/ml after 30 min. 240 min after commencement of the infusion the plasma level was 0.7±0.1 µg/ml. In these patients 22±2% (mean±SE) of the total administered dose of QX-572 was excreted unchanged in urine during the 24 hours following infusion of the drug. A second group of 28 patients with acute myocardial infarction also received QX-572 8 mg/kg body wt. Their plasma levels did not differ significantly from those found in the first group of patients. There was a poor correlation between the amount of QX-572 administered and plasma level at the end of the infusion. The study has provided some preliminary data about the pharmacokinetics of QX-572, but before a detailed analysis can be done data from longer periods of observation is required. The present results suggest that in future QX-572 can be administered in a standardized dosage, what would be advantageous in practice.

    Notes: Summary The importance of individualizing the dosage of potent drugs in order to maximize their therapeutic effectiveness and safety is generally accepted. Whenever possible the dosage of a drug should be “titrated” directly in each patient against the intensity of its therapeutic or toxic actions. Unfortunately, for many drugs convenient clinical yardsticks of the intensity of their pharmacologic effects are lacking. Determination of the serum concentration of such compounds can help to guide adjustment of dosage during their therapeutic use. By measuring the serum level of drugs one bypasses the largest source of individual differences in doseeffect relationships — the pharmacokinetic variation between subjects. However, the relationship between the serum concentration of a drug and the intensity of its pharmacodynamic action is influenced by many other factors, which must always be considered in interpretation of serum levels. Therapeutic decisions should never be based solely on the serum concentration of a compound, nor can such measurements ever substitute for careful medical observation and judgement.

The average gastrointestinal uptake estimated by comparison of the urinary recovery of label and the areas under the plasma curves after the two routes of administration was 65%. The half life of radioactivity in the plasma after oral35S-furosemide was 90 ± 17 min (estimated on the slope between 2 and 6 h); the corresponding figure after35S-furosemide i.v. was 47–53 min (slope 0.5–4 h). There was probably a slower phase after 4–6 h. Fractionation of labelled material in urine from two subjects demonstrated that approximately two thirds of the label recovered at 24 h had the same chromatographic properties as furosemide. A major part of the metabolite(s) was probably furosemide glucuronide. There was no evidence that 4-chloro-5-sulfamoylanthranilic acid was formed in man. The total urinary recovery of label (5–7 d) after oral and intravenous administration was 55.1±3.2 (mean±SD) and 82–84%, respectively. After35S-furosemide i.v., 6–9% of the label was recovered in faeces, and it could not be accounted for solely by biliary excretion of furosemide.

    Notes: Summary Twenty male volunteers received oral doses (2100, 1050, and 525 mg) of a pivampicillin-probenecid salt in a 1 to 1 molar ratio (MK-356) at 12 hour intervals. After each dose peak serum concentrations of probenecid were observed 2 hours later than peak concentrations of ampicillin. Following the first dose of MK-356 the apparent elimination rate of ampicillin was dose-dependent and did not follow first order kinetics, as it showed a longer apparent half life after a higher dose. An equal dose of MK-356 administered 12 hours later caused an increase in the peak serum ampicillin level greater than expected from the concentration of ampicillin after the preceding dose. In twelve male volunteers who received at random 525 mg of MK-356 or 350 mg of pivampicillin, each three times daily for 4 days, the areas under the ampicillin concentration curve were the same after the first or last dose of either drug. When 2100 or 1050 mg of MK-356 was taken as an initial dose, 30 to 40 per cent of the ampicillin was recovered from urine in the ensuing 12 hours. The results indicate that when at least 400 mg probenecid was coadministered twice daily with 700 mg pivampicillin (MK-356), the peak serum concentrations of ampicillin were increased and its elimination rate slowed following successive doses.

    Notes: Summary The absorption, biotransformation and elimination of sulfinpyrazone, 1,2-diphenyl-3,5-dioxo-4-(2′-phenylsufinylethyl)-pyrazolidine, have been studied by administration of single 200 mg oral doses of a14C-labelled preparation to two male volunteers. Absorption from the gastro-intestinal tract was rapid and complete and the plasma concentration of unchanged drug reached maximum values of 22.67 and 13.04 µg/ml, respectively, after 1 – 2 hours. The elimination half-life in the two subjects, calculated from the decline between 3 and 8 hours, was 2.7 and 2.2 hours. The integrated concentration of unchanged sulfinpyrazone in plasma, estimated from the area under the concentration curves (AUC), was almost as high as that of total14C-substances, so the proportion of metabolized drug in plasma was low. In no case did the AUC of the three specifically determined metabolites, i.e. the sulphone G 31 442, the “para-hydroxy”-compound G 32 642 and the “4-hydroxy”-compound GP 52 097, exceed 4% of the sulfinpyrazone value. More than 95% of whole blood radioactivity was confined to plasma. The oral dose was rapidly and completely excreted, since within 4 days more than 95% was recovered, 85% from urine and 10% from faeces. A large proportion of the dose was excreted as unchanged drug in the two volunteers: 51 and 54% of total urinary radioactivity was present as sulfinpyrazone; 8.2 and 8.8% was present as “para-hydroxy”-metabolite, 2.7 and 3.0% as sulphone-metabolite, and 0.6 and 0.8% as “4-hydroxy”-metabolite. About 30% of urinary radioactivity consisted of highly polar metabolites. Spectroscopy of them showed that they were the C-β-glucuronides of sulfinpyrazone (28%) and the corresponding sulfone (2%). In these metabolites the C(4) of the pyrazolidine ring was directly attached to glucuronic acid, and thus they represent a new type of biosynthetic conjugate.

    Notes: Summary Eight healthy volunteers were given single i.v. and oral doses of clonazepam (2 mg). The disposition curves after i.v. administration showed a biexponential decline and the data were applied to a two-compartment open model. The volume of distribution ((Vd)β) ranged between 1.5 and 4.4 l/kg and the plasma half-life (t1/2) between 19 and 60 hours. Absorption after oral administration was fast, with peak plasma concentrations within 4 hours in all subjects. Five of the subjects received repeated oral doses of clonazepam 0.5 mg bid for 15 days. The plasma level during steady state (estimated as Cmin within the dose interval) could be predicted from the constants A, B, α and β obtained in the single dose study with a coefficient of variation of 6%. The plasma half-lives after cessation of the subchronic dosing were of the same magnitude as after single doses.

    Notes: Summary The metabolic fate of orally administered (−)-[14C]-ephedrine has been studied in 3 human subjects and the urinary excretion of metabolites determined quantitatively by solvent extraction, paper chromatography and reverse isotope dilution procedures. Following an oral dose of the drug (0.35 mg/kg, 1.6 µCi), 97% of the dose was excreted in the urine within 48 h, 88% in the first 24 h. Unchanged drug was the major urinary excretory product (53–74%), with N-demethylation occurring to a variable extent (8–20%) although there was little interindividual variation in urine pH. Oxidative deamination was also variable (4–13%); the main identified products of this were benzoic acid (free and conjugated) and 1,2-dihydroxy-1-phenylpropane (free and conjugated). No phenolic metabolites could be detected, and thus it would appear that these compounds cannot be implicated in the acquisition of tolerance to ephedrine which can occur on repeated dosage.

    Notes: Summary Pharmacokinetic and metabolic studies with3H-etilefrine were performed to assess the importance of a first-pass effect on the pharmacodynamic action of this sympathomimetic amine. Identical amounts of3H-activity, ca. 80% of the dose, were excreted in the urine after intravenous or oral administration, which indicates complete enteral absorption of the drug. Comparison of the areas under the plasma curves of unchanged etilefrine after both routes of administration resulted in a bioavailability factor of 0.55, which can be explained by an extensive first-pass effect. The time curve of plasma levels of etilefrine was compatible with an open 2-compartment model characterized by a rather large volume of distribution (Vd, β) of 160 1, and a predominant half life of 2 hours. The pharmacodynamic action corresponded to the amount of drug in the central compartment. The major pathway of metabolism of etilefrine was conjugation to form the phenolic sulphate, and a very minor proportion of the drug was excreted as the corresponding hydroxymandelic acid. This metabolic pattern seems to confirm our hypothesis that phenylalkylamines with the hydroxyl group in the m-position of the benzene ring are predominantly conjugated in contrast to p-hydroxylated compounds which are mainly deaminated.

    Notes: Summary A method has been developed for the quantitative determination of heptabarbital [5-(1-cyclohepten-1-yl)-5-ethylbarbituric acid] in human plasma after administration of single therapeutic doses of the drug. It involves a single extraction step followed by gas chromatography with alkali flame ionization detection, and the results were linear in the concentration range 0.125 – 5.0 µg/ml plasma. The pharmacokinetics and relative bioavailability of heptabarbital and heptabarbital sodium were studied in a crossover design in 7 healthy volunteers after oral administration of 20 tablets containing 200 mg heptabarbital and hard gelatine capsules containing an equivalent amount of its sodium salt. Heptabarbital concentrations in plasma were determined at regular intervals. The absorption of heptabarbital from the tablets was quite slow and peak level times varied from 1.5 to 4 h. The sodium salt was absorbed more rapidly and peak concentrations occurred between 1/3 and 2 h. In all cases the elimination of heptabarbital could be described by a single first-order process with an average half-life of 7.6 h (range 6.1 – 11.2 h). The half-life of the drug in each individual was about the same in the two trials. The relative bioavailability in each volunteer was estimated by comparing the areas under the plasma concentration curves. The sodium salt had an average bioavailability of 83% relative to the free acid. In some volunteers urinary excretion of unchanged heptabarbital was measured; cumulative excretion amounted to 0.16 – 0.30% of the administered dose. Four volunteers received one tablet each night for eight or ten days, but no accumulation was found. In three volunteers the half-life of the drug prior to and after these experiments did not change, whereas in the other volunteer the half-life decreased from 7.1 to 4.6 h. The possibility of enzyme induction should be considered when heptabarbital is taken regularly. It was concluded that heptabarbital was a suitable drug for the treatment of insomnia, since its half-life was rather short. Heptabarbital sodium may be used for induction of sleep, whereas Medomin® tablets, i.e. heptabarbital free acid, may be prescribed when the maintenance of sleep is the primary reason for treatment with a hypnotic drug.

The plasma concentrations of the two drugs were determined during and after infusion. The average elimination half-life of hexobarbital had decreased from 325 to 122 min and of tolbutamide from 418 to 183 min following rifampicin treatment. It was calculated that the metabolic clearance of hexobarbital had increased about three-fold and that of tolbutamide more than two-fold. Significant changes in the distribution kinetics of the two drugs were not observed. The results suggest that rifampicin is capable of inducing drug metabolism in man, which leads to an increased rate of elimination of drugs that undergo biotransformation in the liver.

    Notes: Summary A clinical-pharmacological study was performed to determine the effect of acetylsalicylic acid upon the serum concentration of indomethacin. 14 rheumatic patients were given indomethacin orally (25 mg × 4 for 4 days) and concurrently acetylsalicylic acid 3.7 g orally (0.9 g × 3 and 1.0 g × 1 daily), and 21 rheumatic patients were given indomethacin rectally in the morning (100 mg × 1) and concurrently acetylsalicylic acid 3.7 g orally (0.9 g × 3 and 1.0 g × 1 daily). On comparison with treatment with oral or rectal indomethacin alone, it was found that peak serum concentrations of indomethacin were significantly reduced (1% level), the times of the peaks were not shifted, and the areas beneath the serum concentration curves of indomethacin were smaller, but significantly so only if compared with rectal administration. In 12 rheumatic patients given indomethacin by rectum in the evening (100 mg × 1) and concurrently acetylsalicylic acid 3.7 g (0.9 g × 3 and 1.0 g × 1 daily), the serum level of indomethacin on the following morning (after 11 h) did not differ from that found after rectal treatment. A statistically but not biologically significant difference was observed between the mean serum half-lives of indomethacin given orally and rectally. For unknown reasons, concurrent doses of acetylsalicylic acid and indomethacin made the mean serum half-life of indomethacin longer than after its oral administration, but shorter than when the same dose of indomethacin was given rectally. There was no difference between serum levels of salicylate after oral administration of acetylsalicylic acid alone or after a concurrent oral or rectal dose of indomethacin. The results have been related to those reported previously, with respect to the interaction between indomethacin and acetylsalicylic acid, the serum levels of indomethacin after oral and rectal dosing, and the serum half-life of indomethacin based upon a one- or two-compartment model. The clinical relevance of the study is discussed.

The data after iv administration were analyzed according to a two compartment open model with half lives of 8.8±1.2 min for the α phase and 2.4±0.2 h for the β phase (predominant half life), respectively. Protein binding was 〉98%. The apparent volume of distribution was small at 0.34±0.03 l/kg. Of the intravenous dose 7.4±1.2% was excreted in urine as a conjugated metabolite, and 0.65±0.1% was excreted unchanged. After oral administration no measurable plasma concentrations could be detected, nor was any quercetin found in urine, either unchanged or in a metabolized form. These results exclude absorption of more than 1% of unchanged drug. Recovery in faeces after the oral dose was 53±5%, which suggests extensive degradation by microorganisms in the gut. The data obtained show that oral administration of flavonoids may be of questionable value.

    Notes: Summary Whole night EEG and polygraphic recordings were made in ten young, healthy, male volunteers after dixyrazine (12.5 mg, 25 mg, 50 mg), methaqualone (250 mg) and Isonox® (methaqualone 250 mg + etodroxizine 50 mg). A total of 156 recording nights (36 adaptation nights were not included in the analyses) were scored for different sleep stages according to accepted criteria. The smallest dose of dixyrazine (12.5 mg) had no significant effect upon sleep pattern: the larger doses (25 mg and 50 mg) caused significant decreases in REM-sleep during the first nights of administration. The decrease disappeared during the following two nights of treatment. No withdrawal effects were seen. Methaqualone also caused moderate depression of REM-sleep during the first night of treatment, and this effect, too, disappeared during prolonged administration. Isonox® (methaqualone + etodroxizine) had a somewhat stronger surpressive effect upon REM-sleep than methaqualone alone.

    Notes: Summary Carbamazepine (2.7–3 mg/kg) was administered orally as an alcoholic solution (50% v/v) to eight healthy volunteers. Two of the subjects were also given 50 mg and 100 mg of carbamazepine in alcoholic solution and 200 mg as a tablet. Plasma concentrations, which were analysed by mass fragmentography, reached a maximum 1 – 7 hours after dosing, and then declined monoexponentially with half-lives ranging from 24 to 46 hours. The half-lives were independent of dose. The apparent distribution volume ranged from 0.79 to 1.40 l/kg. It was found that 72% of carbamazepine was bound to plasma proteins with little interindividual variation, and this was not influenced by the presence of diphenylhydantoin or phenobarbital in therapeutic concentrations. The pharmacokinetic parameters calculated from single oral doses were used to predict the steady-state plasma concentration expected after treatment with multiple doses of 200 mg three times daily. The predicted steady-state concentration was 2 – 3 times higher than that reported in patients undergoing chronic treatment with carbamazepine at this dose level, i.e. the pharmacokinetics of carbamazepine apparently change during multiple dosing.

    Notes: Summary The haemodynamic effects of disopyramide have been studied in 11 patients with manifest or imminent heart failure. Disopyramide, 2 mg per kg body weight, was given intravenously during right and left heart catheterisation. The cardiac index decreased by an average of 28% (p〈0.01); mean maximal left and right ventricular end-diastolic pressures were increased by 5.0±0.9 mm Hg (〈0.01) and 5.0±0.6 mm Hg (p〈0.05), respectively; and left ventricular systolic pressure fell slightly but significantly (p〈0.05). No significant change in right ventricular systolic pressure was seen. Pulmonary wedge pressure rose on average by 2.7 mm Hg (p〈0.05). No significant change in heart rate was observed in 5 patients with sinus rhythm. In 6 patients with atrial fibrillation, there was a significant (p〈0.01) increase in heart rate; the average increase in heart rate for the entire group was 19,6 heats per minute. The maximum effect on all the parameters occurred 7–11 minutes after the injection, and it gradually subsided during the following 10 minutes. It was concluded that disopyramide had a potentially serious myocardial depressant effect.

The log. plasma concentration-time curve of chlormethiazole after cessation of the infusion was found to be curvilinear and was fitted therefore, by a bi-exponential equation computed by non-linear least squares regression analysis. Half-lives for the inital α-phase (0.54±0.05 h) and the terminal β-phase (4.05 ±0.60 h) were calculated together with other pharmacokinetic parameters of the two compartment open model. An explanation for the discrepancy between the presently reported plasma half-lives and those appearing in the literature has been presented. The pharmacokinetic treatment of the plasma concentration-time data obtained following intravenous infusion also enabled the prediction that the maximal systemic availability of an orally administered dose of chlormethiazole would be of the order of 15%.

    Notes: Summary Various brands of digoxin tablets, and even different batches of one brand, may differ greatly in bioavailability. Digoxin-Sandoz® tablets have been compared with Lanoxin® manufactured between 1969 and 1972 and after May 1972. Comparisons were also made between and within batches of Digoxin-Sandoz tablets. Three separate cross-over studies were conducted involving a total of 20 volunteers. Digoxin-Sandoz tablets were shown to have a constant bioavailability and to produce plasma concentrations very similar to “new” Lanoxin. Storage for 2 years of one batch of Digoxin-Sandoz did not alter the bioavailability. Particle size was shown to influence bioavailability. Care should be exercised when plasma data alone are interpreted as an index of bioavailability. Measures of bioavailability based on plasma data obtained up to 6 h after administration differed from those based on cumulative urinary excretion data (in this study by a factor of about 2), which can lead to the belief that a difference in bioavailability is much greater than is actually the case. Data from cumulative urinary excretion, collected over a sufficiently long period of time, are likely to be the most reliable method for determining the bioavailability of a substance such as digoxin.

After rapid i.v. injection of 250 mg phenacetin, the log plasma concentrations versus time curves were evaluated according to the rules of a two-compartment open model. The elimination half-life (t 1/2) β varied from 37 to 74 minutes. The volume of distribution (Vd) β ranged from 1.0 to 2.1 1 per kg body weight. The total clearance of the drug was high and approximated the average value of hepatic blood flow in normal adults. In agreement with this finding, the bioavailability of a small oral dose of phenacetin (0.25 g) was almost nil, as the bulk of the drug was cleared during its first pass through the liver. With large oral doses (1.0 g) the first-pass effect decreased and availability increased. The results are discussed and related to current general views of the liver-first-pass phenomenon.

At rest, the central pressures increased slightly, which was taken as a sign of a moderate negative inotropic effect, but there was no change in pre-ejection period or maximal dp/dt of the aortic pressure. The heart rate increased and there was a small decrease in systemic arterial pressure, probably due to a fall of systemic vascular resistance. The PQ time was prolonged. During exercise, with its positive inotropic stimulation, the moderate negative inotropic effect of verapamil disappeared, whereas the increase in heart rate and decrease in aortic pressures persisted. Some variables that reflected the oxygen demand of the heart decreased. The slight negative inotropic effect does not appear to be a particular contraindication to the use of verapamil, but it should be employed cautiously in conditions with a compensatory rise in systemic vascular resistance, or if atrioventricular conduction is impaired.

    Notes: Summary Carbamazepine (Tegretol®) was administered orally to four patients as a single dose, and one week later three times daily for 15–21 days. The plasma half-lives of the drug were shorter in all patients after multiple doses (20.9±5.0 hours) than after the initial single dose (35.6±15.3 hours). During multiple doses the plasma concentrations of the metabolite carbamazepine-10,11-epoxide followed those of the parent drug. The steady-state plasma concentrations expected during multiple doses were calculated from the pharmacokinetic parameters obtained in the single dose studies. The calculated levels were higher (17.2±7.2 µg/ml) than the observed maximal concentrations (8.4±1.6 µg/ml on day 4), which were obtained 3–4 days after starting the multiple doses. The levels tended to decrease further during the experimental period. The results suggest that carbamazepine induces its own metabolism in man.

In two subjects a second peak was observed between 6 and 12 hours. The plasma half life of total radioactivity was 51 hours. 20% and 56% respectively of the dose were eliminated in urine and faeces during the following 7 days. 55% of the total radioactivity in plasma, 80% in urine and 20% in faeces consisted of CHCl3-insoluble compounds. 50 – 60% of the latter in plasma and urine could be hydrolysed by β-glucuronidase. More than 90% of the split products were identified as conjugates of methylproscillaridin. TLC-separation of the CHCl3-soluble fractions of plasma and urine yielded two unidentified metabolites, P2 and P3, as the main compounds, besides methylproscillaridin, proscillaridin and scillarenin. In faeces more than 90% of the non-polar fraction was identified as methylproscillaridin. Shortly after administration of3H-methylproscillaridin, the radioactivity in plasma consisted mainly of CHCl3-in-soluble conjugates and of the metabolite P2.

    Notes: Abstract Effects of diazepam (Valium) on photosynthesis, chlorophyll/photosynthesis ratios, respiration, uptake of rubidium ions, and ultrastructure of Scenedesmus obliquus synchronized by a light-dark regimen of $$14:\overline {10}$$ hrs were determined. 80 and 160 μM diazepam, added to the nutrient medium at the start of the light-dark change (i.e., start of the cell cycle) gradually reduced rates of photosynthesis below the initial rates from the beginning of the experiment. Contents of chlorophyll, however, remained nearly unaffected. Consequently, the diazepam-treated cells had a higher chlorophyll/photosynthesis ratio—also with regard to respiration in order to calculate the gross photosynthesis. The occurrence of photorespiration cannot be assumed. The net influx or rubidium was slightly reduced by 100 μM diazepam 0.5 and 2.0 hrs after the start of the cell cycle and was strongly inhibited after 5 to 14 hrs. 80 and 160 μM diazepam caused separation of thylakoids, formation of giant mitochondria and enlargement of vacuoles. The results are discussed and it is finally suggested that diazepam acts on different membrane systems. Furthermore an ATP deficiency cannot be excluded.

    Notes: Abstract Spores of the thermophilic, acidophilic, Bacillus acidocaldarius were covered by a thick outer coat and a laminated inner coat (5.5 nm periodicity). Small membranous vesicles were present in the spore core and they disappeared as germination proceeded. After depolymerization of the cortex, and a 30% increase in spore diameter, a localized gap appeared in the laminated inner coat only. This inner coat gap was narrow and could be the whole length of the spore. The germ cell appeared to grow, or to be pushed towards the inner coat gap, at which stage the outer coat disappeared in the same localized area. As the vegetative cell grew out the spore coat fell away, with loose cortical material still attached to it. The young germ cell developed a large spherical electron dense inclusion body in the cytoplasm, at the same time as the ribosomal and nuclear areas became distinct.

    Notes: Abstract A cell observation chamber was designed to perform continuous photomicroscopic observations of hyphal anastomosis and the origin of intra-hyphal hyphae in Trichophyton terrestre and T. rubrum. These data were correlated with ultrastructural features of intra-hyphal hyphae. Hyphal fusions occurred commonly in either species of Trichophyton when incubated alone. In T. terrestre, empty hyphal segments adjoined by live units were invaded at the septa from both directions by new hyphal ingrowth. Continuous observations revealed that the intra-hyphal hyphae subsequently anastomosed via a lateral fusion peg. Similar intra-hyphal hyphae were shown in T. rubrum. Electron microscopic studies revealed ascomycetous septa in both conventional hyphae and intra-hyphal hyphae. For the latter, the cytoplasm and wall of the inner hypha were bounded by cytoplasmic organelles and another cell wall of the outer hypha.

    Notes: Abstract During the process of degradation of the cell wall of the yeast form of Pullularia pullulans by the lytic system of Micromonospora chalcea samples were withdrawn at different times and observed under phase contrast and electron microscope. The progressive lysis of the walls reveals a fibrillar component inside the apparently amorphous wall. Freeze etched preparations of cells during the formation and regeneration of protoplasts show that the cellular membrane is split and this method allows the smooth external face of the membrane and other internal face covered by particles to be seen. The fact that the smooth face of the membrane is only visible during the preparation or the regeneration of protoplasts and very rarely when intact cells are fractured, suggests a strong adherence between cell wall and this external layer of the membrane. During the regeneration which takes place as in most of the yeasts and moulds, a special study of the extension of the cell wall is made and a possible mechanism for this extension of the regenerated cell wall is proposed.

    Notes: Abstract Tritium labeled uredospores of Uromyces phaseoli were produced be feeding the host, Phaseolus vulgaris, with 3H-orotic acid. These spores were allowed to germinate on and to penetrate into a bean leaf. 24 hrs after inoculation, the bean rust had formed the first haustorium. All fungal structures, including the fungus walls, were heavily labeled. No label could be detected in the cells that had come into contact with the hyphae. In the infected host cell, the haustorium was labeled heavily, but the sheath around the haustorium and the host cell remained free of label. These results indicate that no detectable amounts of label leach from the bean rust into the host at this stage of infection although it is known that the rust takes up many metabolites. Since the sheath remains free of label and all fungal structures are evenly labeled, it is concluded that the sheath is formed by the host.

    Notes: Abstract Cr was oxidized in 1 aim O2 at 980, 1090, and 1200°C. ElectropolishedCr and some orientations of etched Cr oxidize rapidly and develop compressive stress in the growing Cr2O3; other orientations oxidize slowly, apparently free of stress. SEM examination of fracture sections shows that the thick oxide is polycrystalline whereas the thin oxide on etched Cr is monocrystalline. It is deduced that the monocrystalline oxide grows by lattice diffusion of cations outward, and the polycrystalline layer by the two-way transport of cation diffusion outward and anion diffusion inward along oxide grain boundaries. The consequent formation of oxide within the body of the polycrystalline layer generates compressive stress and leads to wrinkling by plastic deformation. The activation energy for oxidation of Cr by cation lattice transport is 58 kcal/mole. Polycrystalline Cr2O3 forms on Fe-26Cr alloy, whether electropolished or etched; oxidation is accordingly rapid and accompanied by compressive stress.

    Notes: Abstract A Zr-0.5 Y alloy was found to oxidize about 6 times faster than pure zirconium over the temperature range of 400 to 565°C. The activation energies were nearly identical (∼32 kcal/mole). The activation energies correspond to grain boundary diffusion of oxygen through the scale. The higher oxidation rate of the alloy was attributed to a higher anion vacancy concentration and the assumption that diffusion sites in the lattice and boundaries were in local equilibrium. Measurements on yttria-doped zirconia showed that ionic conductivity was increased markedly by yttrium and extended over a wide range of oxygen pressure. The defect structure of the doped oxide was changed to one of oxygen vacancies, even at the high end of the oxygen pressure range, 10−8 to 0.2 atm, over which pure ziconia contains oxygen interstitials. The doped oxide was found to be extrinsic over the entire range of oxygen pressure and, although ionic conductivity predominated, electronic conductivity was still appreciable. The electronic conductivity, however, was still sufficiently high so that electron transport was not rate-controlling in the predominantly ionic-conducting scale.

    Notes: Abstract The oxidation of ilmenite (FeTiO3) in air and dry oxygen was investigated over the temperature interval 600 to 970°C. Dense platelets of ilmenite crystals as well as powder samples of ilmenite were oxidized. The weight data were recorded employing a thermobalance. The oxidation kinetics of ilmenite platelets were parabolic except for the initial stages during which logarithmic kinetics were observed. For powder samples the logarithmic rate law was followed primarily. The logarithmic rate law was attributed to free penetration of oxygen through cracks and short-circuit paths. The activation energies associated with the logarithmic rate law were nearly one-half of those obtained from parabolic oxidation. The growth morphology of the products of oxidation of ilmenite was observed with a scanning electron microscope. The effect of growth morphology on the kinetics is discussed, and a probable reaction mechanism is suggested for the oxidation of ilmenite.

    Notes: Abstract A new fast method is proposed for the determination of parabolic rate constants of metal oxidation as a function of pressure and temperature. The method consists of determining rate constants by oxidation of a single metal sample in a continuous manner, periodically changing the oxidant pressure or temperature. This method eliminates a number of errors inherent in the classical method which involves the use of a new metal specimen in each experiment and it further shortens the time of evaluating the functions k p '' = f (p, T). The method is particularly suitable for the determination of rate constants of slow processes. To verify the proposed method measurements of the kinetics of oxidation of cobalt and nickel at different oxygen pressures over the temperature range 1000–1300° C were carried out.

    Notes: Summary The integumental melanophores of Latimeria chalumnae were studied by light and electron microscopy. The epidermal melanophore located in the mid-epidermis consists of a round perikaryon with long slender dendrites extending into epidermal cells and intercellular spaces. The dermal melanophores occur in the loose dermal matrix underlying a relatively thick layer of collagen fibers. The dermal melanophores are usually flattened and their dendrites lie parallel to the collagen layer. Both epidermal and dermal melanophores contain oval, electron-opaque melanosomes, large mitochondria, agranular vacuoles of endoplasmic reticulum and microtubules. Microfilaments and RNP particles are less conspicuous. While the peripheral cytoplasm of both dermal and epidermal melanophores is filled with a large number of melanosomes, the perinuclear cytoplasm of many dermal melanophores is occupied by premelanosomes in various stages of differentiation, and that of the epidermal melanophore contains numerous large vacuoles. Despite the scarcity of epidermal melanophores, the epidermal melanin unit is present in the form of melanosome complexes. In addition, the melanophores of Latimeria possess the basic characteristics common to other vertebrates, but they more closely resemble those of lungfish and other aquatic vertebrates.

    Notes: Summary Sinus and venous walls of normal human spleens were studied with enzyme histochemical and electron microscopic methods. Particular attention was paid to the connections between sinuses and veins. Histochemically the sinus lining cells revealed a distinct naphthol-AS-acetate-esterase activity but no reaction for alkaline phosphatase. Venous endothelial cells were positive for the latter but negative for the former enzyme. In the sinusvenous junctional area there were no endothelial cells with reactivity for both enzymes. Electron microscopically both the sinus lining cells and the venous endothelial cells could be clearly characterized and therefore easily distinguished from one another on morphological grounds. There were no clear ultrastructural indications of transitional forms between sinus lining cells and venous endothelial cells in the sinus-venous area. According to these findings, sinus lining cells represent a specialized endothelium, but one with practically no morpholgical similarities to the venous endothelium.

    Notes: Summary The fine structure of epidermal mucous cells of two species of salmonid fish has been described. Mucous cells are, next to filament-containing cells, the most commonly encountered cells in fish epidermis. The development of the cells as they progress to the periphery has been characterised. They are initially difficult to distinguish from filament-containing cells: later, they can be recognised by the presence of much smooth-surfaced E.R. The mucigenesis and the subsequent secretion of mucus has been observed and it is essentially comparable to that which occurs in the mucous cells of the mammalian intestine. The mucous layer of the epidermal surface seems to mainly comprise of the products of these mucous cells and the “cuticle” seen in other species has not yet been observed in the salmonid species investigated here.

    Notes: Summary Smooth muscle cells of the mouse vas deferens fixed with 5% glutaraldehyde contained three types of filaments, namely, thin (50–80 Å) filaments, intermediate (100 Å) filaments and thick (120–180 Å) filaments. However, in 2 out of 16 experiments, under identical conditions, the cells did not contain thick filaments. With OsO4 fixation, thin filaments were not prominent, the most obvious being thick (120–250 Å) and intermediate (100 Å) filaments. After soaking in a modified Ringer solution under no applied tension for one hour, thick filaments (120–180 Å) appeared prominently in smooth muscle cells of the mouse vas deferens and thin filaments were in ordered bundles. By 4 hours, thick filaments had increased in size and density, with thin filaments distributed randomly around them. After 8 hours in Ringer, thin filaments were diffuse and difficult to discern, while thick filaments were large (up to 300 Å) and electron-dense. Intermediate (100 Å) filaments were present in association with dark bodies. Physiological experiments indicated that the intracellular components responsible for the development of a mechanical response were still functional at this time. The presence of “thick filaments” is also reported in degenerating smooth muscle cells of the guinea-pig vas deferens in tissue culture.

    Notes: Summary Triad density relative to sarcomeres, size of T-system tubules, sarcomere length, muscle fiber diameter in native and fixed states, and size of myofibrils were measured in four striated muscle fiber types in Atlantic hagfishes (Myxine glutinosa, L.) of different sizes. Triads occur at A/I — junctions in all fiber types. The density of triads relative to sarcomeres is higher in “white” than in “red” muscle fibers. The T-tubules show no sign of branching. The area of the T-system tubules is 3–4 times the surface area in 80 μm “white” muscle fibers and 1–2 times that in 60 μm “red” fibers. The size of myofibrils is similar in “white”, “intermediate”, and “red” fibers of m. parietalis, and constant through a large span of animal size. In “white” fibers, increase in diameter up to 90 μm is accompanied by an increase in the number of myofibrils, not by an increase in the individual size of the myofibrils. Above 90 μm, “white” fibers grow by increasing the amount of intermyofibrillar space. This is reflected by an extensive shrinkage of the thicker “white” fibers during the preparative procedure for electron microscopy, a shrinkage that is limited only by complete packing of the myofibrils. “Red” fibers shrink much less.

    Notes: Summary This study describes distinctive cells with ultrastructural and histochemical features of APUD-type endocrine cells within the tracheal epithelium of human fetuses, newborns and children as well as different animal species. These cells referred to as Kultschitzky cells (K cells) were found to be argyrophilic, but not argentaffin, and are considered analogous to the same type of cells in lung and gastro-intestinal tract. Fluorescence histochemistry demonstrated the presence of intracellular amine within tracheal K cells, but only after in-vitro or in-vivo administration of amine precursor (L-DOPA). Ultrastructurally, these cells are characterized by the presence of numerous cytoplasmic granules (dense core vesicles) which show species related morphologic variations. Two different types of K cells were found in trachea of lamb and armadillo, each type possessing morphologically different dense core vesicles. In human and rabbit tracheas, only one type of K cell was identified. K cells in the trachea are distributed as single cells between other epithelial cells; neuroepithelial bodies such as those found in bronchial mucosa were not identified. Well differentiated K cells were found in tracheas of early human fetuses and throughout gestation, infancy, and childhood. Preservation of K cells in human autopsy material and widespread occurrence of these cells in various laboratory animals will permit further studies into the nature and function of tracheobronchial endocrine cells.

    Notes: Summary Nerve cells of the superior cervical ganglion of young rats (20 g body weight) were investigated electron microscopically 6 h, 24 h, 48 h and 5 days after subcutaneous injection of nerve growth factor (10 μ/g body weight every 24 h). By means of a planimetric method with high accuracy significant changes of the Nissl substance and the Golgi apparatus could be demonstrated as early as 6 h after injection. Within the Nissl bodies both the density of bound ribosomes on the cisternae of the rough endoplasmic reticulum and the density of free ribosomes and polysomes decreased in a given field. This finding reflects a rearrangement of the Nissl substance and a spreading over larger areas of the cytoplasm, indicating an activation of the ribosomal system. The Golgi apparatus, in particular its outer part, increases in volume with time of nerve growth factor treatment. On the other hand, the total cell volume does not show significant changes before 48 h of nerve growth factor treatment. At this time an increase in the cytoplasmic volume can be seen, whereas the nuclear volume remains unchanged. The possibility of correlations of the present findings with data from biochemical studies done under similar experimental conditions is discussed.

    Notes: Summary The ultrastructure of neuromuscular connections on developing dorsolongitudinal flight muscles was studied in the moth Antheraea polyphemus, Undifferentiated membrane contacts between axon terminals and muscle-fiber anlagen are present in the diapause pupa. They persist during the period of nerve outgrowth, which probably provides a pathway of contact guidance. By the 4th day of adult development some of these contact areas have differentiated into structures similar to neuromuscular junctions although differentiation of muscle structure does not start earlier than the eighth day. Dense-cored vesicles are abundant in many axon terminals at the beginning of development. They later decrease in number quite rapidly. The significance of the above-mentioned early junctions, their possible mode of action and the role of the dense-cored vesicles are discussed. It is proposed that they exercise a stimulating (trophic) influence on the growth of the undifferentiated muscular tissue. The imaginai neuromuscular junctions are formed during the second half of adult development. Clusters of vesicles and electron-dense depositions along the inner face of the axolemma seem to initiate junction formation. Glial processes then grow between axoand sarcolemma and divide the large contact area into several small segments. Mutual invaginations and protrusions of the sarcolemma and the glial cell membrane subsequently form an extensive “rete synapticum.” Six days before eclosion the glial and sarcoplasmic parts of the rete synapticum are similar in size. Up to eclosion, all glial processes shrink and increase in electron density. Most of the observations are discussed also in relation to findings in vertebrates.

This cellular part is surrounded by a thick electron dense capsule which is secreted by the bud itself. The capsule is probably impervious and protective to its contents. Blood vessels join the buds to the zooids of the colony. They form the probable route of transfer of yolk from the zooids to the dormant bud. The capsule of the dormant bud has some structural features in common with the black stolon of the adult zooids. The black stolon is probably formed in a manner similar to that which made the fusellar fabric of the periderm of fossil graptolities.

    Notes: Summary The ultrastructure of normal and glycerol treated fibers of the closer muscle of the ghost crab, Ocypode cursor, was studied. The muscle is composed of presumably phasic (short sarcomeres) and tonic (long sarcomeres) fibers, the latter greatly predominating. Horseradish peroxidase (HRP) was used as an extracellular tracer to delineate the tubular system (TS), and to determine to what extent this system becomes detached from the extracellular space as a result of glycerol treatment. Sarcolemmal clefts invade deeply into the muscle at Z-lines and I-bands; tubules invaginate into the muscle from the clefts and from the surface sarcolemma at the Z-lines, A-I overlaps and A-bands. A tubules are in frequent diadic or tetradic contact with the sarcoplasmic reticulum (SR), whereas Z tubules appear to be randomly associated with SR, terminal cisterns (TC) and Z-line fibrils. When HRP was administered to normal muscle, black reaction product was found adjacent to the outer surface of the sarcolemma, within the clefts and within profiles of the TS throughout the tissue. In glycerol treated muscle peripheral vacuolation frequently occurred; black reaction product penetrated only as far as the vacuoles and into dilated Z-line tubules, but was virtually absent from the rest of the TS. This lack of continuity between the extracellular space and the A tubules indicated disruption or constriction of the A tubules as a result of glycerol treatment, although Z tubule contact with the extracellular space appeared unimpaired. These findings provide ultrastructural correlates of the electrophysiological changes produced by glycerol treatment of the closer muscle of the ghost crab (Papir, 1973), namely, interference with excitation-contraction (e-c) coupling. The random association of the Z tubules with SR and TC, and their resistance to disruption by glycerol treatment, tend to endorse the claims that the Z tubules in crustacean muscle are not directly involved in e-c coupling (Brandt et al., 1965; Peachey, 1967; Selverston, 1967).

Différenciation d'un système de microvillosités externes, avec un niveau dense aux électrons, très contrasté par rapport aux extrémités d'où semblent partir de fins filaments englobant une population assez dense de Bactéries. 2. Cette plage de microvillosités recouvre un relief assez particulier de la procuticule, évoquant par ses cratères un paysage lunaire. 3. Présence de canaux cytoplasmiques ramifiés et terminés par une expansion arrondie, sous l'épicuticule. 4. Existence d'un système de vésicules reliant la base des microvillosités aux extrémités renflées des canaux cytoplasmiques. L'association des Bactéries est régulière et une grande proportion de celles-ci est en cours de cytolyse. La système de microvillosités et de canaux ne nous parait pas étranger à la présence de cette population bactérienne.
    Notes: Summary Two species of Porcellidium have been studied by scanning and transmission electron microscopy: P. viride from Banyuls-sur-mer, Mediterranian Sea and P. fimbriatum from Roscoff, English Channel. Both species live on green Algae of the genus Ulva. We confirmed previous descriptions of the cuticular ultrastructure in the main groups of Copepods. Four new characteristics however were shown to occur in the backcuticle of Porcellidium: 1. The presence of a system of highly differentiated external microvilli showing electron dense basal portions and less electron dense tips, from which thin filaments project towards a large population of Bacteria associated with the Copepod. 2. The surface of the cuticle resembles a lunar landscape with craters. The sheet of microvilli closely follows the contour of the cuticle. 3. The presence of branched cytoplasmic canals with swollen extremities (beneath the epicuticle) extending from the epidermal cells. 4. Systems of vesicles lying between the bases of the microvilli and the expansions (ampullae) at the tip of the cytoplasmic canals. The association of Bacteria with the cuticle is constant and many of these are apparently undergoing cytolysis. The system of microvilli and of cytoplasmic canals are apparently related to the presence of the bacterial microflora.

    Notes: Summary In the hypothalamus of the roach (Leuciscus rutilus) green and yellow fluorescent cells were found in the nucleus recessus lateralis (NRL) and the nucleus recessus posterioris (NRP) and green fluorescent cells in the nucleus recessus preopticus (NRPO). The green fluorescence indicates the presence of noradrenaline or dopamine and the yellow one the presence of 5-hydroxytryptamine. The cells of the NRL and NRP contained electron dense granules averaging 70 nm in diameter. The NRL is divided into two parts. These and the NRP are connected by large fluorescent tracts. The NRL and NRP send axons towards the nucleus lateralis tuberis (NLT) and the NRPO sends axons towards the nucleus preopticus (NPO). It could not be established whether the aminergic nuclei described are the origin of the fluorescent fibers in the hypophysis of the roach.

    Notes: Summary Spermatogenesis in the Budgerigar can be arrested by reducing the birds' photoperiod to 8 hours of daylight or less. When this occurs, Sertoli cell cytoplasm shows a great increase in the size and number of residual bodies, while the smooth endoplasmic reticulum is reduced. If the bird is kept at 8 hours of daylight for some weeks large lipid droplets are seen in Sertoli cytoplasm, and degenerated spermatids are apparently phagocytosed. The interstitium shows fewer active Leydig cells, a paucity of lipids and occasional ovoid mitochondria. The basal lamellae of the tubule which are thick and convoluted before and during spermatogenesis become thinner and straighter. It is thought that these morphological changes reflect changes in metabolic activity.

    Notes: Summary Neural lobes of rats subjected to dehydration by drinking 2% saline for four days were examined electron microscopically and compared to untreated controls. The ultrastructure of the blood vessels and the tissues surrounding them were examined and it was found that, although few exocytotic figures could be seen in either group of animals, a significantly larger (P〈0.01) number of small vesicles were found in nerve endings adjacent to the perivascular space in the saline treated group when compared to nerve endings not closely associated with blood vessels. No differences were found in the control group of animals, which supports the suggestion that the vesicles could arise from a membrane recapture process.

Nerve profiles, identified as cholinergic, were found in close apposition with all four cell types. No examples of a close association between processes of chromaffin cells and sympathetic neurons were found.

    Notes: Summary Examination of the squirrel monkey (Saimiri sciureus) area postrema (AP) revealed this circumventricular organ to be primarily composed of two types of glial cells and a single type of neuronal element. No pattern of neuronal arrangement could be discerned, however, this cell type was frequently observed in close relation to the perivascular spaces. The neuronal elements, although slightly larger than the glial cells, were characteristically less electron dense. The neurons routinely displayed an infolded nuclear membrane, a single nucleolus and the normal complement of subcellular organelles. Synaptic terminals were numerous, and both axo-somatic and axo-dendritic varieties were observed with the latter being more numerous. Both clear-cored and dense-cored vesicles could be observed in the same ending. Unmyelinated neuronal processes were the predominant type within the interior of the AP, although myelinated processes were also regularly present. Non-neuronal elements within the AP resembled CNS astrocytes and were as numerous as the neuronal elements. This cell type appeared to envelope completely the vasculature and separated the parenchyma from the perivascular spaces. The ventricular surface of the AP was covered by modified ependyma which lacked kinocilia but frequently demonstrated microvillar projections. Opposed ependymal cell membranes showed interdigitations, and zonula adherens-type cell junctions connected the ependymal cells near the ventricular lumen. Two types of bulbous projections were observed in the ventricular lumen close to the ependymal surface. The most characteristic feature of the AP, however, was its vascularity. Perivascular spaces surrounding fenestrated capillaries contained fibroblasts and collagen. The vascular endothelium routinely demonstrated pinocytotic activity, and the basal lamina was prominent.

    Notes: Summary The failure of denervated muscle to undergo effective regeneration, despite reported increases in the number of muscle satellite cells, warranted an investigation of the viability and myoblastic capacity of these cells present in denervated muscle. Four types of satellite cells present in muscle denervated for three weeks are described, based on their ultrastructure and relationship to their principal fiber. The increased number of ribosomes, including helically arranged polysomes; the number of Golgi complexes; the presence of microtubules; the branching subsarcolemmal tubular system; and the appearance of regularly arranged 96 Å microfilaments with diffuse electron dense areas are structural features of satellite cells that are similar to those of developing myoblasts in growing and regenerating muscle. The electron microscopic observations suggest that “activated” satellite cells do have myoblastic potential. Possible explanations for the ultimate failure of denervated muscle to regenerate include: 1) the inability of the muscle to produce satellite cells rapidly enough to keep pace with muscle degeneration; 2) a cytotoxic effect produced by the degenerating muscle fiber on the satellite cell; and 3) the inability of satellite cells to form stable, mature multinucleated fibers in the absence of the trophic effect of the nerve.

    Notes: Summary Compensatory hypertrophy was induced in the rat soleus muscle by sectioning the tendon of the ipsilateral gastrocnemius and plantaris muscle. Seven days after tenotomy of synergistic muscles, when soleus hypertrophy attains about 40%, the number of satellite cells (expressed as percentage of all muscle nuclei found in the same cross-sections) as revealed by electron microscopy, was increased from 5.8±0.06% in the normal soleus muscle to 16.6±1.26%. After four days' denervation of the soleus muscle the percentage of satellite cells was increased to 7.2±0.62%. In experiments where hypertrophy of the soleus muscle was combined with denervation three days after tenotomy of synergists, and examined after another four days (during which time it loses, as has previously been shown, over 40% of its predenervation weight), the number of satellite cells was greatly increased to 29.9±3.42%. This increase is apparently due to two independent processes which take place during the first postoperative period: a) mitotic division of satellite cells during the early stages of compensatory hypertrophy and b) pinching off of muscle nuclei from rapidly atrophying muscle fibres due to subsequent denervation. Activation of satellite cells was mainly manifested by expansion of smooth and especially of rough endoplasmic reticulum, a rich Golgi complex, high pinocytotic activity, increased number of ribosomes and by nuclear changes. Concomitantly with the increased number of satellite cells, proliferation of fibroblasts, macrophages and mast cells could be observed.

    Notes: Summary Two methods are described for measuring the mitochondrion-vesicle association seen by electron-microscopy in thin sections of the guinea-pig taenia coli. Both methods are based on comparisons of the observed distributions with predicted random distributions. It was found in control muscles that mitochondria were consistently nearer to vesicles than corresponding random points. 1 mM ouabain treatment reduced the mitochondrion-vesicle association for mitochondria which were closer to the membrane surface than 130 nm. Quantitative investigation of the freeze-etch structure of the membrane fracture faces is also reported, confirming the observation that membrane particles are more numerous in vesiculated membrane regions of smooth muscle.

    Notes: Summary The ultrastructure and acetylcholinesterase activity of the intrinsic innervation of the sphincter of Oddi of eight adult dogs was studied by electron microscopy. A rich distribution of unmyelinated axons embedded individually or as groups within Schwann cell cytoplasm (“innervation fasciculée”), is to be observed. A few myelinated fibres were also observed. Many of the axons are acetylcholinesterase-positive. Three main types of nerve terminals are distinguished according to their vesicle populations. Individual nerve cells or small groups of nerve cells were scattered between the smooth muscle bundles and in the lamina glandularis mucosae. The cytoplasm of some neurons contains many electron dense spherical bodies resembling “myeloid bodies”, and many lysosomes. Nerve terminals synapse onto both neuronal perikarya and their dendrites. Within the nerve fascicles, close appositions between the terminals occur frequently probably representing the most peripheral inter-neuronal integrative link in the neural regulation of the function of the sphincter of Oddi. — The gap between nerve terminals and smooth muscle cells usually measures several thousands of Å. Closer appositions are seldom seen, and no synaptic complexes can be observed.

    Notes: Summary The ultrastructure of the rectal papillae of the parasitoid hymenopteran, Nasonia vitripennis (Walk), is described. These organs in this insect consist of four distinct cell types arranged as a closed, hollow cone. The majority of the cells are present in the raised cone, and are characterised by large numbers of mitochondria arranged in a membranous labyrinth. A series of cells form a collar around the base of the cone. Junction cells have been identified which are present at the point of insertion of the cone into the rectal epithelium. The base of the cone consists of cells with elaborately folded plasma membranes facing both the central cavity of the cone, and the haemolymph. The structure of this rectal papilla is compared with those found in other insects.

    Notes: Summary Ultrastructure of osmiophilic bodies identified as lipofuscin granules occurring at extraneuronal sites in the brain tissue of both young and old monkeys was studied. The present work revealed that lipofuscin granules were detected normally in the neuroglia cells, phagocytic cells and pericytes surrounding the blood capillaries, as well as in the capillary endothelium. However, their presence in these sites was more marked in young animals. The findings presented in this report are strongly suggestive of the normal removal of lipofuscin from the nerve cells to the capillary endothelium, and suggest further that the phagocytic cells as well as the glia cells participate in this removal mechanism. Being a more active process during youth, few lipofuscin granules are present in neurones from young animals. Failure of the removal mechanism due to diminished activity of the participating cells with ageing, is probably the cause of lipofuscin accumulation in senescent neurones.

    Notes: Summary Barbiturates were examined for in vitro effects on ultrastructure of the frog sciatic system and polymerization of microtubules (MT) in a brain supernatant. Exposure for 5–17 h to 2.0 mM barbiturates caused a considerable loss of MT in ganglionic cell bodies and sciatic axons. This was mostly followed by a proliferation of 10 nm filaments. Under similar conditions treatment with 1 mM NaCN or 0.1 mM 2,4-DNP did not change the number or ultrastructure of MT and filaments. Eight barbiturates, varying in binding ratios to serum albumin and partition coefficients, were tested for effects on polymerization of MT using viscometry. Inhibitory effects were found which correlated with their reported ability to bind to albumin and brain fractions. Dimethylsulphoxide and ethanol were used as solvents for some of the barbiturates. These solvents at 1% had stabilizing effects on MT. The present results are discussed in relation to previous findings of inhibition of rapid axonal transport in vitro in the frog sciatic system by barbiturates.

    Notes: Summary The ultrastructure of human umbilical cord vein endothelium in situ, after isolation by collagenase treatment, and in primary culture is described. The cultured cells formed a monolayer with typical “butt” and interdigitated junctions with specialized areas, and contained Weibel-Palade bodies, rod-shaped tubular organelles considered specific of endothelial cells. These morphological features were not present in cultures of human skin fibroblasts and fibroblast-like cells derived from umbilical cords. It is thus concluded that endothelial cells retain their characteristic fine structure in primary culture. Simple ultrastructural studies can thus be used to identify endothelial cells in culture.

    Notes: Summary The secondary X zone induced by castration in the adrenal cortex of adult male mice was examined by electron microscopy and radioautography with 3H-thymidine. 10–15 days after castration a thin layer of small eosinophilic cells is formed in the inner-most cortex. Such eosinophilic cells contain irregulary shaped nuclei and spherical or ellipsoidal mitochondria with tubulolamellar cristae, 20–25 days after castration a prominent zone of small eosinophilic cells was clearly identified as the secondary X zone. The typical secondary X zone cells were characterized by the formation of peculiar mitochondrial complexes and whorled sER. The X zone cells with their characteristic organelles incorporated 3H-thymidine. The ultrastructure and formation of the secondary X zone were very similar to those of the primary X zone which appears during normal postnatal development. We demonstrate here the capacity of reticularis cells of adult male mice to transform into typical X zone cells following castration.

    Notes: Summary The spermatheca of the female mealworm beetle is an inflorescence of branching cuticular ducts which is connected to the bursa copulatrix via a cuticular neck surrounded by a muscular coat. The infolded bursal cuticle consists of a distinct outer epicuticle, inner epicuticle, procuticle, and a subcuticular zone; the latter is rich in mucopolysaccharides. The cuticle of the neck lacks a distinct procuticle. The cuticle of the spermatheca itself is mostly inner epicuticle with two thin underlying lamellae of procuticle. The cells of the bursa are loosely coupled to the procuticle, whereas cuticular projections bind the epithelia of the “neck” and the spermatheca proper to the underlying epithelia. The apical plasma membranes of the spermathecal epithelium are sinuous and much infolded; we believe that this epithelium controls the micro-environment within the cuticular ducts.

    Notes: Summary The corpora allata of the three last larval instars were studied in newly molted animals, at the beginning, middle, and end of the feeding period, and during the molt period. They were found to consist of uniform gland cells, whose ultrastructure changes in the course of the instars. In gland cells considered to be resting, the outer and inner nuclear membranes run in parallel without forming a dilated perinuclear space. Mitochondria are small, polymorphic, with an electron-dense matrix. The smooth endoplasmic reticulum (SER) appears as stacks of parallel cisternae near the nuclear envelope and in the rest of the cytoplasm, and as accumulations of twisted profiles. Occasionally, the SER takes the form of paracrystalline bodies. There are few small smooth-surfaced vesicles in the cytoplasm. In cells considered as active, a dilated perinuclear space occurs. The peripheral ends of profiles forming the SER are swollen, and numerous vesicles and vacuoles bud off from them to fill the cytoplasm. Mitochondria are large, with a more transparent matrix. The plasma membrane of gland cells located just beneath the connective tissue sheath forms numerous small invaginations. The corpora allata consist of resting cells during the molt periods. At the beginning of each instar, few active gland cells appear. In the middle of the second to last and the third to last instars, the bulk of the gland cells is active. At the end of these instars, there are both active and inactive cells. In the middle of the last instar, the gland cells are inactive or subactive, and at its end, all gland cells are completely inactive.

    Notes: Summary Electron microscopic studies of the carotid body of the domestic fowl (Gallus gallus domesticus) have shown Type I and Type II cells combined with axons into compact groups. The many Type I cells in the depths of the organ had a body, containing the nucleus, and an elongated, flared process. Some of the Type I cells in the superficial regions tended to be spindle-shaped. Type I cells were characterised by membrane-bound, dense-cored vesicles about 120 nm in diameter. Type II cells invested the Type I cells and had axons embedded in them as in Schwann cells. The fine structure of the carotid body in the domestic fowl resembles that of the Lovebird (Uroloncha domestica) and of various amphibia and mammals. The possibility is discussed that the Type I cells may have a chemoreceptor or a general secretory function, or even both pathway for functions together. The main role of the Type II cells seems to be to provide a of these axons leading to or from Type I cells.

    Notes: Summary The germinal or surface epithelium covering rabbit Graafian follicles contains occasional small, dark, lysosome-like bodies. After an ovulatory dose of human chorionic gonadotropin (HCG) such bodies gradually increase in size and number. At 8 hr after HCG there is a maximal accumulation in the apical follicle cells; then the dense bodies decrease and just prior to ovulation, 9.5 hr after HCG, only few of them remain in the attenuated surface epithelium. Most of the growing membrane-surrounded bodies probably represent lysosomes, since electron microscopy combined with cytochemistry revealed that many of them contain the lysosomal “marker” enzyme, acid phosphatase. The role of sex steroids and prostaglandins regarding lysosomal growth and labilization is discussed. The close temporal relation between disappearance of the apical surface epithelial lysosomes and disintegration of the underlying tunica albuginea gives further support to our working hypothesis that at least part of the “ovulatory enzymes” emanate from the surface epithelium.     Notes: Summary In the moth Antheraea polyphemus the innervation of the anlage of the dorsolongitudinal flight muscle (dlm) was transected at the onset of adult development. The subsequent breakdown of the isolated motor stumps during early adult development was studied at the ultrastructural level. First reactions are seen on the second day of development when axonal mitochondria shrink. Later, elongated vesicles similar in structure to channels of smooth ER, appear in large numbers in the axoplasm. Their nature as well as the functional aspects of early axonal changes are discussed. From the 7th day onward two types of axonal breakdown become prominent. The first is characterized by swelling axon profiles, distorted vesicles and strongly shrunken mitochondria, while shrinking axon profiles containing tightly packed mitochondria and unaltered vesicles are typical of the second. Both types presumably take place independently of each other in different axon terminals. Axons and the contents of at least the first type are finally removed by transformation into lamellar bodies. Glial processes obviously behave independently of degenerating terminals; they loose any contact with them and never act as phagocytes for axon remnants. During the whole period of breakdown undifferentiated contacts between nerve fibers and muscle anlagen are present but synaptic structures as in normal developing dlm have never been observed. This fact, in comparison with earlier studies, suggests a lack of trophic nervous activity on the muscle anlagen tissue. A short time after removal of the isolated stumps new nerve tracts appear between dlm-fibers (which are, of course, strongly retarded in development). They are presumably sensory wing nerves which lack a guide structure to the central target, due to axotomy. Neuromuscular contacts or even junctions formed by axons of these nerves have occasionally been detected on the dlm. Their nature is discussed. Wallerian axon degeneration is compared to the normal, metamorphic breakdown of the innervation of the larval dlm-precursor. In contrast to the former, glial processes here remain in contact with the terminals. Glia and axons first swell. Then most glial processes are transformed into lamellar bodies whereas neuntes shrink and become electron-dense. Axonal organelles remain intact for a long period.

    Notes: Summary Regularly, four different types of cellular contacts are found in synovial folds and villi of the cubital joint of the cat (interdigitations, desmosomes, intermediate junctions, gap junctions). The same types of contact-with the exception of intermediate junctions — occur sporadically also in synovial fat folds of the knee joint of the rabbit. In both species, hemidesmosomes and discontinuous basement membranes are seen in the synovial lining layer. Cellular contacts predominate between A-cells and cells of the intermediate type, hemidesmosomes and incomplete basement membranes predominate in intermediate cells and B-cells. The latter are rare in A-cells. The importance of such contacts for mechanical, metabolic and electrical interactions of cellular elements in the synovial membrane is discussed. No unanimous concept as to their function can be advanced at present.

    Notes: Summary The pineal organ of the migratory antarctic penguin, Pygoscelis papua, has a lobular structure. Clusters formed by different types of parenchymal cells are separated by connective tissue septa containing blood vessels. The predominant cell type displays a well-developed Golgi complex, free ribosomes, clear and granular vesicles (secretory granules), and lysosomes. Other cell types found in the gland are supporting and ependymal-like cells. The former contain dense bodies and filament bundles, the latter possess abundant cilia and clusters of ribosomes. Typical photoreceptor elements are lacking. Blood vessels are located within a perivascular space bordered by basal laminae. This perivascular space extends between the basal protrusions of the parenchymal cells. The presence of pinocytotic vesicles, secretory granules and cytoplasmic processes in the vicinity of these spaces suggests active sites of transport and exchange of substances. Intercellular canaliculi-like spaces are surrounded by parenchymal cells rich in microvilli. These canaliculi are continuous with the cavities (invaginations) of secretory and other parenchymal cells.

    Notes: Summary The effect of ovariectomy on the adrenal gland was studied in the Mongolian gerbil. Castration stimulated cells in the zona fasciculata as well as those in the region between the zona fasciculata and zona reticularis. No alterations occurred in the zona reticularis. The width of the intermediate region was enlarged, the cells were hypertrophic and contained an increased number of concentric whorls of rough endoplasmic reticulum. Smooth endoplasmic reticulum was hypertrophie in cells of the zona fasciculata and intermediate region. Many mitochondria in cells of the intermediate region were larger than those in controls. Stimulation of cellular ultrastructure is directly correlated with a significant elevation in plasma cortisol in ovariectomized gerbils.

    Notes: Summary Proximal to an experimental stenosis of the small intestine of rats and guinea-pigs a remarkable hypertrophy of the muscle coat develops 3–5 weeks after the operation. There is no increase in the length of the intestine but an overall increase in volume of the muscularis externa up to 10 times. This increase is accounted for by an increase in size and in number (by mitosis) of smooth muscle cells of both the longitudinal and circular layers. Bundles of newly-formed smooth cells appear in the serosa and are circularly arranged. In the hypertrophic smooth muscle cells of the circular layer the ratio of surface to volume is 0.80 (0.80 μm2 of cell surface for every μm3 of cell volume) as against 1.4 in the control muscle. The hypertrophic muscle cells have a highly developed sarcoplasmic reticulum and show a large number of nexuses. The density of innervation (number of axons per given number of smooth muscle cells) is smaller than in controls. Few collagen fibrils are visible in the extracellular space.

    Notes: Summary The effects of chronic administration of dexamethasone (for up to 15 consecutive days) on both the morphology and DNA-synthesis of the mitochondria of the rat adrenal zona fasciculata were investigated by stereologic and autoradiographic techniques. Up to the 3rd day of continuous dexamethasone treatment, the average volume of mitochondria did not change, whereas the number of mitochondria per cell was significantly decreased. From the 3rd to the 15th day of hormonal administration both the volume and number of mitochondria were found to decrease in proportion to the duration of treatment. Autoradiography showed that after the 3rd day of dexamethasone administration there is virtually no incorporation of 3H-thymidine into the mitochondrial compartment. These findings are discussed in the light of evidence indicating that dexamethasone blocks ACTH-release by inhibiting the hypothalamo-hypophyseal axis. The results confirm the view that ACTH controls the maintenance of growth and proliferation of rat adrenocortical mitochondria.

The principal changes in the growth-hormone cells of anterior pituitary glands of partially hepatectomized rats were: (1) increased numbers of secretory granules in exocytosis, (2) increased numbers of microtubules, and (3) enlargement of endoplasmic reticulum and occurrence of dilated cisternae of the endoplasmic reticula. Many growth-hormone cells contained a reduced number of secretory granules. Exocytosis of growth-hormone granules was more frequently observed in animals sacrificed at 32 hours after the operation than in those killed at 80 or 104 hours after surgery. The above results in which appearance of numerous microtubules and active secretory granule extrusion in the growth-hormone cells were observed after hepatectomy indicate that ultrastructure of growth-hormone cells and growth hormone secretion were markedly stimulated by the operation.

    Notes: Summary The occurrence of structural connections between myocardial granules and tubular elements of the sarcoplasmic reticulum in the cardiac ventricle of the Atlantic hagfish is described. The core substance of the myocardial granules is shown to be uranophilic and in this respect similar to the granular cores within the lumen of the sarcoplasmic reticulum. After application of CaCl2-containing fixatives, the ultrastructure of the core substance of these organelles resembles that described for calcium-containing structures in vertebrate glial cells. Incidences of uranophilic core substance in the extracellular space suggest a secretory function of the myocardial granules. Possible implications of the sarcoplasmic reticulum and the myocardial granules in storage, intracellular transport, and secretion of bound calcium are discussed.

The diameter distributions of thick myofilaments from fibre cross sections in the relaxed, phasic contracted, tonic contracted, and in the catch states show that a characteristic filament spectrum is formed in the catch state and its preceding active state. The significant structural differences are discussed in relation to earlier hypotheses related to the catch state.

    Notes: Summary A transmission electron microscopic study of demineralized, methaphyseal bone of the young guinea pig is presented. Special attention is paid to the lysosomal system of the different cell types. Visualization of acid phosphatase and aryl sulfatase activity was used to identify tissue components as belonging thereto. The distribution of alkaline phosphatase activity, a plasma membrane marker, was also examined. Osteoblasts were distinguished by a marked development of the granular endoplasmic reticulum and the Golgi complex. Perivascular cells type A, morphologically resembled the osteoblasts, and are believed to represent an early stage in the specialization of the latter. A few lysosomes were normally found in the osteoblasts; they were less common in the type A cells. In contrast to their regular occurrence in guinea pig epiphyseal cartilage, dense bodies of lysosomal nature (“type I vesicles”) were only rarely seen in the bone matrix. Structures analogous to the type II vesicles in cartilage were, however, normally present. Their membrane showed activity of alkaline phosphatase. Possible functions of lysosomes and matrix vesicles in osteogenesis are discussed. Perivascular cells type B and chondroclasts both contained a prominent Golgi complex and large numbers of free ribosomes, mitochondria and lysosomes. In the type B cells, inclusion material of varying appearance often occurred in the lysosomes and in endocytic vesicles. The chondroclasts sometimes presented a ruffled border, with associated vacuoles and lysosomes in the subjacent cytoplasm. It is suggested that both cell types participate in the resorption of the epiphyseal cartilage. Chondroclasts presumably arise by fusion of type B cells and/or monocytic precursors from the peripheral blood.     Notes: Summary Guinea pig metaphyseal bone was exposed to horse spleen ferritin in vitro and to colloidal thorium dioxide in vivo. The cellular uptake and intracellular accumulation of these marker particles were studied ultrastructurally. In vitro, the ferritin molecules were found to spread evenly throughout the tissue. After 1–2 hours ferritin was mainly found in plasma membrane invaginations and in endocytic vesicles of varying size. At 4–6 hours a successive accumulation of the marker in secondary lysosomes could be observed. In addition to ferritin, the lysosomes and the large endocytic vesicles often contained other inclusions. In vivo, the pattern of intracellular accumulation of the marker particles was identical to that in vitro. Moreover, the presence within the cells of similar amounts of thorium dioxide after 1 and 4 days suggested that these indigestible molecules are stored intracellularly for a considerable time. In accordance therewith there were no definite signs of extrusion of labeled bodies or secretion of the exogenous marker by exocytosis. Ferritin and thorium dioxide were taken up by all cell types in the metaphysis. Both in vitro and in vivo perivascular cells type B ingested large amounts of marker particles, whereas chondroclasts, endothelial cells, perivascular cells type A and osteohlasts showed a more restricted endocytizing ability. On the basis of these observations, the functional significance of different cell types in the resorption of the epiphyseal cartilage and the formation of bone is discussed.     Notes: Summary The six types of neurosecretory cell in the optic lobe of Carcinus maenas described by light microscopy are recognised by electron microscopy. They are categorised according to size, distribution of organelles and type of neurosecretory product. The neuro secretory material, produced as granules by the Golgi bodies, migrates to the cell periphery eventually reaching the sinus gland via the neurosecretory cell axon extension. No change in size occurs in the granules but the density does alter. Each cell type has its own characteristic type of neurosecretory granule based on size and electron density. Multivesicular and lytic bodies in cell types 1, 2, 3,4 and 6 suggest a cycle for degrading neurosecretory material. Such a cycle is not so evident in cell type 5. Peripheral release of neurosecretory material is suggested for cell type 6 although the fate of the material is unknown.

    Notes: Summary During blastoderm formation, the Drosophila embryo produces a large area of new membrane to accommodate the simultaneous demands of approximately four thousand newly cleaved cells. The embryo was examined with the electron microscope at various stages during cleavage in order to investigate the high membrane forming capacity of these cells. Embryos were subjected to the histochemical procedure for the demonstration of thiamine pyrophosphatase. The enzyme was present in the cisternae of the endoplasmic reticulum, Golgi bodies and the nuclear envelope. No activity could be demonstrated on the furrow surface or at the furrow tip despite closely adjacent reactive cisternae. It is concluded that the endoplasmic reticulum and Golgi bodies are not major contributors to the new surface. Lamellar bodies were frequently observed in the cytoplasm of all stages examined. The bodies showed a lamellar periodicity of approximately 3.5 nm, and were ultrastructurally similar in appearance after a variety of fixation procedures. The distribution of these bodies was markedly related to the stage of blastoderm formation. Before the commencement of cleavage, lamellar bodies were very prominent within a region 4–5 μm. below the cell surface. As cleavage progressed, the bodies became sparse or absent from this region but were apparent at the base of the blastoderm cells or in the sub-blastoderm region, where they were not previously present. Lamellar bodies with leaflets closely associated with, or in apparent continuity with, the cleavage furrow membrane were frequently observed. In these regions the lamellar periodicity was the same as the thickness of the membrane laminae. It is suggested that these bodies play a role in the synthesis of new membrane in the furrow. Intercellular contact specializations between the developing membranes of the furrow were restricted to incipient desmosones and point contacts where the intercellular gap was reduced to 3 nm or less.

    Notes: Summary Electron-microscope autoradiographs have been prepared from the neural lobes of the pituitary glands of rats which had received intracisternal injections of [35S] cysteine at various times before gland removal. The rate of appearance and disappearance of autoradiographically demonstrable radioactivity in the neural lobe closely paralleled that previously determined, biochemically, for radioactive hormones and neurophysins. Radioactivity was appreciably associated with the undilated parts of neurosecretory axons only during the first few hours after injection of the label. The axonal dilations were subdivided into those in which small vesicles could be seen (“endings”) and those in which no small vesicles could be seen (“swellings”). Radioactivity appeared first in “endings” and then in progressively larger and larger profiles of “swellings”. It appeared that newly arrived granules were found close to the limiting membrane of the nerve swelling and that as time progressed they moved deeper and deeper into the swelling. On the basis of the results, suggestions were made for an anatomical explanation of the readily-releasable pool of hormone which has been demonstrated pharmacologically.

    Notes: Summary The fat globules of the cream fractions of human and rat milk were stained with ruthenium red. Under the electron microscope, discrete granules and an amorphous coat of lesser density are seen at the surface of the milk fat globules. Since ruthenium red binds anionic groups selectively, it is probable that the granules containe the greatest concentration of these groups. The cream fraction of rat milk contains hexoses, hexosamines, methylpentoses and sialic acid. Methylpentoses and hexosamines are significantly enriched in the cream fraction. It is concluded that the finding of a surface coat in milk fat globules is in keeping with the Bargmann-Knoop model and suggests a distinct mechanism for carrying certain complex carbohydrates in milk. The role of the negative charges at the outer surface of the membrane coat in maintaining fat globules in suspension and in binding certain cations such as calcium is suggested.

    Notes: Summary The structure of the tapetum of the sheep's eye has been examined by transmission electron microscopy. It consists of regularly arranged layers of collagen fibrils, estimated from the hydroxyproline content to make up about 60% of the dry weight of the tissue. There are several hundred of these layers in the whole depth, a majority being orientated in the same direction. The collagen is associated with strands of microfibrillar material as well as with cells of both fibrocyte and smooth muscle types.
