    Notes: Abstract The in vitro effect of rubratoxin B on the electron transport system of rat liver mitochondria was investigated. This mycotoxin depressed oxygen consumption in ADP-lacking mitochondria and in ADP-coupled mitochondria, using succinate or β-hydroxybutyrate as substrats. Rubratoxin B is neither an oxidative-phosphorylation inhibitor nor uncoupling agent. Its effect is compared with aflatoxin B1.

    Notes: Summary This is a study of the fine structure of cells of the 20-day fetal rat calvarium. Special attention is given to identifying and characterizing preosteoclasts. These cells are relatively common and located largely, but not exclusively, at the endocranial bone surface. The preosteoclasts are characterized by abundant mitochondria, an incomplete perinuclear Golgi apparatus, and variable-shaped dense granules. The dense granules are unique in appearance in that they contain an internal dense matrix surrounded by a clear halo. Most granules are circular in shape but some are elongate or tubular in form. Granules with identical appearance are observed in osteoclasts. The preosteoclasts are mononucleate, or occasionally binucleate. It is suggested that because preosteoclasts are morphologically distinctive and relatively abundant, it should be feasible to separate these cells from a heterogeneous cell isolate.

    Notes: Summary A method for quantitative studies of the formation rate of bone has been developed. After vital staining with calcein, the fluorescence of a bone section was measured with a microphotometer controlled by a mini computer. After staining the bone structure with alizarin red S in a second step, the section was measured in transmitted light. The two data sets were combined and the shortest distances between the bone surface and the fluorescence lines were computed. With this information the distance distribution and the bone area between the label and the surface could be calculated in two different ways: with the single labeling and the continuous labeling techniques. The advantages and disadvantages of the two methods are discussed and compared with those of other techniques.

    Notes: Summary The purpose of this work was to analyze the proximal tibial metaphysis of the 170 g rat in a quantitative histologic fashion which would allow some relation to tissue age to be established. Stained 3 µm thick tissue sections were analyzed with the aid of a Merz grid on an eyepiece reticule and a light microscope. Tissue mass and cell distribution were studied in all areas. The rate of change in tissue mass during aging of the metaphysis was calculated. Two regions of the metaphysis were identified. One, corresponding to the primary spongiosa, less than 4.45 days of age, is a region of high turnover of hard tissue and high numbers of osteoblasts and osteoprogenitor cells. The other, corresponding to the secondary spongiosa, is a region of relatively low net tissue turnover and low numbers of osteoblasts and osteoprogenitor cells. Osteoclasts were found relatively more uniformly distributed through the metaphysis than were osteoblasts and osteoprogenitor cells. The rate of bone formation in the primary spongiosa is 50 times that found in the Haversian bone of the rib of 5-year-old humans and about 500 times that found at the cortical-endosteal surface of ribs of 5-year-old humans. It is argued that both cell distribution and tissue distribution in the metaphysis support the concept that osteoblasts and osteoclasts, rather than osteocytes, are responsible for the maturation of the metaphysis. The inhomogeneous distribution of both cells and tissue in the metaphysis has definite meaning for the interpretation of findings concerning the incorporation of radionuclides into the skeleton.

    Notes: Summary The demands of growth are known to exacerbate the effect of phosphorus deprivation (PD). We examined whether changes associated with PD could be prevented in young rats in which growth and growth hormone (GH) were eliminated by hypophysectomy (HPX) and whether PD in normal intact rats (INT) was associated with increased secretion of GH. INT or thyroxine- and ACTH-replaced HPX rats were fed one of the three diets: 0.31% P (NP); 0.027% P (LP), and 0.31% P, pair-fed with LP-mates (NP-PF). The results indicate that HPX did not qualitatively alter several physiologic responses to PD: (a) serum and urinary phosphorus (P) decreased and urinary calcium (Ca) increased; (b) net intestinal Ca retention fell and duodenal sac uptake of45Ca rose; and (c) external P balance was restored and duodenal sac uptake of32P-phosphate increased. Only the hypercalcemia seen in INT, LP rats was prevented by HPX. In INT rats serum immunoassayable GH levels, measured in single samples, were not different between different dietary groups while pituitary bioassayable GH was reduced in both LP and NP-PF rats when compared to the NP rats. Thus, except for hypercalcemia, the physiologic responses associated with PD are not prevented by the elimination of growth and GH, and the development of these responses in INT rats was not associated with a consistent or specific alteration in GH secretion.

    Notes: Summary Eighty-eight adult male Sprague-Dawley rats were given a diet with either (a) 0.5% Ca and 0.6% P or (b) 0.01% Ca and 0.6% P. Osteopenia was created by adding prednisolone to the diet. The prophylactic effect of oral 1,25(OH)2D3 on the osteopenia was studied. It was found that prednisolone osteopenia in the rat was associated with defective Ca absorption. By giving an oral dose of 1,25(OH)2D3, it was possible to maintain normal Ca absorption during prednisolone treatment and to prevent the bone loss. No significant hypercalcemia or any kidney calcifications were seen. These results are in contrast to earlier findings, in which subcutaneous administration of 1,25(OH)2D3 failed to prevent prednisolone osteopenia because of its tendency to increase bone resorption.

    Description / Table of Contents: Zusammenfassung In Fortsetzung einer früheren Arbeit wurde die Eminentia mediana von männlichen und weiblichen Ratten, sowie die trächtiger und durstender Tiere auf die Fragestellung hin untersucht, ob 1. die Eminentia mediana der erste Ort ist, wo die beiden Neurohormone Oxytocin und Vasopressin freigesetzt werden, und 2. ob sich die Axone in der externen Schicht der Eminentia mediana aufgrund der mittleren Durchmesser ihrer Granula in verschiedene Populationen differenzieren lassen. Bei männlichen und weiblichen Kontrolltieren haben die Oxytocin-bzw. Vasopressin-granula in der Eminentia mediana bereits ihre endgültige Größe erreicht; ihr mittlerer Durchmesser ändert sich nicht mehr wesentlich bis zum Hypophysenhinterlappen. Allerdings gibt der unterschiedliche prozentuale Anteil beider Populationen in der Eminentia mediana, in Zusammenhang mit den früher bestimmten Werten für die jeweiligen Kerne und die Neurohypophyse, Hinweise auf gewisse Unterschiede im turnover beider Hormone bei männlichen und weiblichen Tieren. Bei graviden und durstenden Ratten findet sich eine scheinbare Abnahme der Granula Durchmesser in den oxytocin-bzw. vasopressinführenden Axonen. In Verbindung mit sehr charakteristischen morphologischen Befunden, besonders bei langdurstenden Tieren, sprechen die morphometrischen Ergebnisse dafür, daß in der Eminentia mediana bereits Neurohormon freigesetzt wird, wenn eine schnelle Deckung des peripheren Bedarfes notwendig ist. Die Axone mit Granula, die kleiner sind als 115 nm, lassen sich in der internen Schicht der Eminentia mediana aufgrund der mittleren Durchmesser ihrer Granula in vier Populationen aufteilen. Ihre mittleren Granula-Durchmesser liegen bei etwa 84 nm, 96 nm, 103 nm und 110 nm. Dieselben vier Populationen finden sich sowohl in der Zona palisadica als auch in der perikapillären Schicht der Eminentia mediana. Im Unterschied zur Zona interna findet sich aber in diesen beiden Schichten noch eine fünfte Population von Axonen mit einem mittleren Granula-Durchmesser von etwa 70–75 nm. Der mengenmäßige Anteil dieser fünf Populationen aber ist unterschiedlich in der Zona palisadica und in der perikapillären Schicht einerseits und in den verschiedenen Tiergruppen andererseits. Einige morphologische Befunde an den Tanycyten werden im Zusammenhang mit der neueren Literatur diskutiert.
    Notes: Summary In connection with previous studies (Krisch, 1974), a morphologic analysis of the median eminence of male, female, pregnant, and thirsting rats has been made, in order to evaluate: 1. whether the median eminence is an additional area where oxytocin and vasopressin may be released from neurosecretory axons in the zona interna; 2. whether it is possible to distinguish different populations of axons according to the mean diameters of their granules in the external zone of the median eminence. In the zona interna of the male and the female control animals, the oxytocin-and vasopressin-containing granules have already reached their final diameters, i.e. their size remains constant until they reach and are stored in the neurohypophysis. However, the relative proportions of these populations in the fibre layer of the median eminence indicate certain differences between male and female animals, concerning the turnover of oxytocin and vasopressin. In pregnant, as well as in thirsting animals, there is an apparent decrease of the mean granule diameters in oxytocin and vasopressin-containing axons. In particular in the latter group, ballooned axons in the fibre layer of the median eminence, containing disintegrating granules, tubular structures filled with electron dense material, and fine-flocculent material in the axoplasm, together with the morphometric results, support our concept that in case of highly increased peripheral demand the contents of the mature, large granules are released into the axoplasm of the fibre layer in the median eminence. In addition, axons containing granules smaller than 115 nm may be classified into four populations with mean diameters of 84 nm, 96 nm, 103 nm, and 110 nm. The same populations are found in the palisade zone of the median eminence as well as in the pericapillary zone, but in contrast to the zona interna there is a fifth population of axons containing very small granules with a mean diameter of about 70–75 nm. The relative proportions of these five populations differ between the zona palisadica and the pericapillary zone on the one hand and between the experimental groups on the other hand. Some morphological findings concerning the tanycytes are discussed with respect to previously published data.

    Notes: Summary The effect of pilocarpine and food uptake on the rate of incorporation of [3H]-leucine in vivo was measured by means of quantitative radioautography in three exocrine cells of the rat: the acinar and the granular duct cells of the submandibular and the acinar cells of the parotid gland. The three cell types react differently. The submandibular acinar cells showed a decrease in incorporation rate after pilocarpine administration but not after feeding. The incorporation rate of the granular duct cells of the submandibular gland remains constant after both stimulations. The acinar cells of the parotid gland show an increase in incorporation rate of [3H]-leucine in response to both. The contrast between the submandibular and the parotid gland could also be demonstrated radiobiochemically, the results reflecting the incorporation rates of the acinar cells of both glands, giving no information on the contribution of other cell types. The decrease in incorporation rate of the submandibular gland acinar cells is accompanied by a shift of polyribosomes towards monomers.

    Notes: Summary After stimulation of the protein secretion by pilocarpine or feeding the rate of incorporation of [3H]-leucine increases in the acinar cells of the parotid gland of the rat while the secretory cells of the submandibular gland show a moderate decrease (Kuijper et al., 1975b). Since the rate of labelled amino acid incorporation depends on the specific radioactivity of the amino acid used, which is not easy to determine in vivo, experiments in vitro were performed to get an idea of the influence of this factor on the measured changes in [3H]-leucine incorporation. In vitro both cell types showed a more pronounced but essentially identical reaction as in vivo. Since in these experiments the specific radioactivity of the extracellular leucine is the same whether fragments of stimulated or unstimulated glands incorporate the radioactive amino acid, the increase of incorporation in the parotid and the decrease in the submandibular cells cannot be ascribed to differences in specific radioactivity of leucine, unless the intracellular leucine pool should show great differences between secreting and non-secreting cells. However, in vitro the submandibular gland cells under both conditions appear to use the extracellular leucine for their protein synthesis (or a small compartmentalized pool in rapid exchange with the extracellular pool). In the parotid cells the whole intracellular pool showed such a rapid exchange with the extracellular one that for practical reasons one may say that these cells, too, rely on the extracellular specific radioactivity of leucine in their protein synthesis. We conclude that the rat parotid gland cells show a rapid and substantial increase of protein synthesis after stimulation of their enzyme secretion, while the submandibular gland cells do not.

    Notes: Summary In untreated, fasting animals the cells of the serous demilunes of the sublingual gland incorporate [3H]-leucine at a higher rate than any other of the 5 main cell types of the 3 major salivary glands. The acinar cells of the submandibular and the mucous cells of the sublingual gland show intermediate values, while the cells of the granular ducts of the submandibular and the acini of the parotid gland have a low rate of incorporation. In fasting animals extrusion of newly synthesized protein starts early in the cells of the serous demilunes. It starts between 4 and 7 hrs after [3H]-leucine injection in the acinar cells of the submandibular gland, while the other cell types did not lose substantial amounts of labelled (glyco)protein within 7 hrs. The secretion of protein is stimulated by the cholinergic drug pilocarpine in all but one of the 5 types of salivary gland cells studied. The acinar cells of the submandibular gland react strongly, the granular duct cells less strongly. Still less are the reactions of the acinar cells of the parotid and of the mucous cells of the sublingual gland. The cells of the serous demilunes of the latter appear to be insensible to pilocarpine. The effect of food uptake on secretion does not differ from pilocarpine stimulation, with one exception: the acinar cells of the parotid gland react more strongly on food uptake than on cholinergic stimulation.

    Notes: Summary Ganglion cells and paraganglionic (PG) cells in the developing rat superior cervical ganglion were studied following postnatal treatment with p-chlorophenylalanine (pCPA) for 5 to 8 days. Litter mates, injected with saline solution, served as controls. Ganglion cells of control animals were differentiated ultrastructurally according to L. Eränkö (1972 a) into late sympathicoblasts and young sympathetic nerve cells. In both maturation stages treatment with pCPA caused marked swelling of mitochondria, concomitant with minor changes of other cell organelles. Parallel to the ultrastructural alterations, fluorescence microscopy and cytophotometry revealed a slight diminution of diffuse fluorescence intensity in sympathetic neurons as the expression of a mainly extragranular amine depletion. In distinction from ganglion blocking agents the alterations are regarded as a general toxic effect of pCPA upon maturing sympathetic neurons, which secondarily influences catecholamine storage sites. Following treatment with pCPA, in PG-cells an alteration of mitochondria was scarcely to recognize. Specific granules were distinctly decreased in number, in some cases to an almost complete degree. Concordant to ultrastructural observations a marked diminution of fluorescence intensity was demonstrable in SIF-cells. In addition in these elements the fluorescence spectrum shifted towards the green field. Fluorescence cytophotometric evaluations confirmed the optical impression. Provided, that PG-cells, demonstrated with electron microscopy, are identical with SIF-cells in fluorescence microscopy, the results are discussed on the basis of a specific decrease of primary catecholamines due to an enzyme inhibition involved in catecholamine synthesis.

    Notes: Summary The central nervous system and the pituitary of adrenalectomized male rats injected with 3H-dexamethasone were examined by radioautography. At 1 hr after the injection, radioactivity concentration was high in the medial basal hypothalamus, the pituitary and the pineal gland. In the hypothalamus, radioactive material was found to be selectively concentrated in neurons in the ventral part of nucleus arcuatus and in the infundibular region. In the anterior pituitary, a large proportion of cells showed silver grains both in the cytoplasm and over the cell nuclei. However, in a small number of cells, the radioactive material was associated with the cell nuclei. Less radioactivity was present in the intermediate and posterior lobes. The pineal gland contained more silver grains than did other regions of the brain. The results obtained in the present study suggest essentially an action of dexamethasone in the medial basal hypothalamus and at the level of the pituitary.

    Notes: Summary Earlier statements to the contrary, the present study demonstrates the presence of a cell surface coat (glycocalyx) on the luminal plasma membrane of the super ficial transitional epithelial cells lining the urinary bladder of male Buffalo rats. This coat was demonstrated with ruthenium red, an electron dense stain, which revealed a surface layer, 60–80 Å thick, separated from the outer leaflet of the plasma membrane by an electron lucent layer, approximately 30 Å thick. The structure of the glycocalyx was not affected by 12 weeks of treatment with dibutylnitrosamine, a known bladder carcinogen.

    Notes: Summary To further evaluate the role of autonomic ganglia in the regulation of pelvic visceral activity, the neural elements in the major pelvic ganglion of the male rat have been studied with histochemical and electron microscopic techniques. The principal findings are that the ganglion is composed of cholinergic and adrenergic ganglion cells as well as small intensely fluorescent (SIF) cells. Polarity in the ganglion is indicated by clustering of small ganglion cells which stain intensely for acetylcholinesterase (AChE) along the pelvic nerve while larger cells, with weak to moderate AChE activity, collect near small branches of the hypogastric nerve. Some cholinergic ganglion cells are enclosed by a plexus of adrenergic terminals. SIF cells appear to be in contact with both cholinergic and adrenergic cells, although many of the fluorescent beads around adrenergic neurons may be short dendrites of ganglion cells, rather than processes of SIF cells. Two types of SIF cells may be distinguished on the basis of size and morphology of their granulated vesicles. Afferent synapses of the cholinergic type were common on SIF cells of the large granule and small granule type. Portions of SIF cells with large granules occur within the capsule of ganglion cells. Contacts seen here were interpreted as efferent synapses from SIF cells to the dendrites of ganglion cells.

    Notes: Summary Immuno-enzyme histochemical investigations on the hypothalamus of the normal rat showed (1) that the Suprachiasmatic nuclei produce vasopressin; (2) that it is highly probable that these nuclei do not produce oxytocin.     Notes: Summary The uptake of L-dopa into the cells of the adenohypophysis of the rat was studied during the postnatal development and at adult age using the formaldehyde-induced fluorescence method (FIF). The cells taking up L-dopa were classified by Alcian blue-PAS-Orange G staining. The correlation between the cells taking up L-dopa and those containing tryptophyl-peptide was estimated during the postnatal period and in adult rats. The cells containing tryptophyl-peptide were demonstrated using fluorescence induced by treatment with combined formaldehyde and acetyl chloride vapour. The following observations were made: 1) Great majority of the cells taking up L-dopa did not contain tryptophyl-peptide. Thus the accumulation of L-dopa into the cells of pars distalis is not due to accumulation of L-dopa into the cells by the same transport mechanism as the amino acids for tryptophyl-peptide. 2) Of the cells taking up L-dopa in the adult rats 96% were chromophobes, 2.0% acidophilic cells (somatotrophs and cells producing prolactin), 0.9% R-mucoid cells (corticotrophs), and 1.2% S1- and S2-mucoid cells (gonadotrophs and thyrotrophs). At 10 and 25 days' age the relative numbers of the cells taking up L-dopa were about the same. 3) Pretreatment with nialamide caused only a slight increase in the number of the cells taking up L-dopa. The decrease in the number of the cells uptaking L-dopa of the pars distalis, which takes place after 5 weeks' age is thus not caused by the increased MAO-activity. 4) Strongly chromophilic cells did not take up L-dopa. At the light of our results it seems evident that L-dopa is taken up by the chromophobic cells when these differentiate into chromophilic cells. The accumulation of L-dopa may be a sign of an active transport of amino acids into the cells. The accumulation of L-dopa into the chromophobic stellate and follicular cells may reflect their metabolic activity. These cells probably have an important role in the production of the hormones of the pars distalis.

Neurophysin-containing perikarya appeared to be surrounded by catecholamine-containing varicosities. This phenomenon was seen to varying degrees within the supraoptic and paraventricular nuclei. The juxtaposition of varicosities and peptidergic neurons suggests an afferent fiber-target neuron relationship that might favor a functional interaction between monoamines and neuropeptides.

    Notes: Summary To identify and describe ovarian follicles committed to undergo follicular degeneration (atresia), immature rats were primed with pregnant mare serum gonadotropin (PMSG). After PMSG treatment, preovulatory follicles develop but subsequently degenerate. Prior to the appearance of pyknotic nuclei (Stage I of atresia), degenerative changes were observed in focal areas of the granulosa cell layer. These changes include “blebbing” of the cytoplasm and alterations in the shape of the granulosa cells. The appearance of these degenerative changes coincides with a decrease in ovarian concentrations of estradiol and testosterone. Since estrogens and androgens maintain the follicle, the decline in estradiol and testosterone could be responsible for the further degenerative alterations that lead to complete deterioration of the preovulatory follicle. In Stage I atretic follicles, lysosome-derived autophagic vacuoles develop and macrophages invade both the thecal and granulosa cell layers. The combined actions of the autophagic vacuoles and macrophages could destroy both the granulosa-cell and thecal layers and thereby transform the preovulatory follicle into an ovarian cyst.

One week after operation the number of macrophages was considerably reduced and continued to decrease during the subsequent time periods studied. A 6 weeks most macrophages had disappeared. Simultaneously the immunological activity diminished and had completely disappeared 8 weeks after operation. Three weeks after operation the endothelial cells of the postcapillary high endothelial venules had flattened, and the number of immigrating lymphocytes was greatly reduced. Subsequently the lymph nodes became depleted of both macrophages and lymphocytes, leaving only the reticuloendothelial framework.

    Notes: Summary By removing connective tissue components with enzymatic digestion followed by HCl-hydrolysis, myoepithelial cells (MECs) of the terminal portion in a variety of exocrine glands of the rat were examined with the scanning electron microscope. The profile of MECs varied considerably from gland to gland; MECs in the lactating mammary gland have a few long cytoplasmic processes in close contact with those of adjacent cells forming a continuous network around the terminal portion. Those of the exorbital lacrimal gland are stellate with many thin radiating processes with tapered ends that terminate freely. MECs in the sublingual gland are characterized by a number of broad and extensive cellular processes. MECs in the submandibular gland are similar in appearance to those of the exorbital lacrimal gland, but with more extensive cellular processes that form a more or less continuous network with those of the adjacent cells. No MECs were observed on the terminal portion of the parotid gland where the cells appear to be lodged on the intercalated duct. The relative surface area covered by MECs per terminal portion was also found to vary significantly, being 24% in the lactating mammary, 17% in the exorbital lacrimal, 48% in the sublingual, and 25% in the submandibular glands. The findings are discussed in relation to the physical properties of secretions in different glands.

    Notes: Summary The vasopressin system of the rat was examined in the course of the first 12 h of rehydration after prolonged thirst at light and electron microscopic levels and by use of the peroxidase anti-peroxidase (PAP) method. Light microscopically, the median eminence was the only part of the system that not only displayed distinct differences between animals of different rehydration times but also showed a characteristic pattern of immunohistochemical reactivity in its rostro-caudal distribution. Ultrastructurally, in the perikarya a maximal labeling of the rough endoplasmic reticulum was observed after 2 h of rehydration, whereas an extensive labeling of the enlarged Golgi zones was attained after 4 h of resupplying water. A labeling of the intercellular clefts in the basal glial labyrinth of the supraoptic nucleus (and to a lesser degree in the subependymal neuropil adjacent to the paraventricular nucleus) was increased 30 min after the onset of drinking, as compared with water-deprived animals; it decreased slightly after 12 h of rehydration. The filling of the swollen fibers by increasing amounts of labeled axoplasmic reticulum, evident in the nuclear areas already after 30 min of water supply, begins in the median eminence after 2 h of rehydration and is fully developed after 4 and 8 h. Corresponding results hold true for the neural lobe but are somewhat delayed in comparison to the findings in the median eminence. The discussion considers (i) synthesis and transport of nongranular vasopressin within the axoplasmic reticulum, and (ii) release not only from the neural lobe but also from the nuclear areas and from the fibers of the median eminence.

Afferent axo-dendritic, axo-somatic and putative axo-axonic synapses with immunostained vasopressinergic neurons can be identified. The presynaptic profiles do not contain immunoreactive material.     Notes: Summary The maturation of the capillaries of the primary portal plexus in rats during the perinatal period has been studied lightand electronmicroscopically. The number of capillaries covering the median eminence and of those invading the nervous tissue (capillary loops) increases significantly with age. Capillary loops were observed as early as the 18th fetal day. The mitotic divisions of the endothelial cells within the preexisting capillaries seem to be the main reason for the vascular growth. Immature capillaries with a characteristic narrow lumen are surrounded by a fuzzy basal lamina; their wall is formed by a generally expanded endothelium with rather sparse organelles and inclusions, and by minute flattened areas. The maturation of the capillary results in a progressive spread of flattened endothelium followed by an enlargment of the capillary lumen. Moreover, a rising concentration of organelles and inclusions, relatively numerous luminal microvilli, and a dense and uniform basal lamina become noticeable as capillary differentiation proceeds. These data are thought to reflect the progressive increase in the metabolic activity of the endothelium as well as the establishment of capillary patency during the perinatal period of rats.

    Notes: Summary Dissociated cell suspensions were prepared from the substantia nigra of 15–17 day-old rat embryos and grafted via an intraparenchymal injection into the depth of the neostriatum of adult recipient rats. The survival and fibre outgrowth of the dopamine-containing neurones in the implants were studied by fluorescence histochemistry, and the functional capacity of the grafts was monitored by repeated testing of the amphetamine-induced turning behaviour of the implanted rats. Before transplantation the target neostriatum of the recipient rats was denervated of its normal dopaminergic innervation by an injection of 6-hydroxydopamine into the ipsilateral nigrostriatal dopamine pathway. The completeness of the denervation was ascertained by measurement of the intensity of the amphetamine-induced turning response. After injection of the dissociated cells large numbers of dopamine-containing neurones were found in clusters at the site of injection as well as scattered in the apparently intact neostriatal tissue up to a distance of about 0.5 mm from the site of injection. Extensive dopamine-containing fibre networks had developed around the implant. These newly formed fibres, which were most abundant around the cell clusters at the injection site, extended in a loose network into large areas of the initially denervated caudate-putamen. In all animals with surviving dopamine neurones the amphetamine-induced turning response was reduced, and in the most extensively reinnervated cases even reversed, within 3–5 weeks after transplantation. This strongly suggests that the implanted dopamine neurones are capable of restoring dopaminergic neurotransmission in the denervated neostriatum, probably via reinnervation of the denervated neostriatal tissue. The use of dissociated brain tissue preparations thus permits reliable intraparenchymal grafting of neurones to plausibly any desired site within the central nervous system, and should open entirely new possibilities for investigation of neuronal growth dynamics and functional reconstruction of damaged brain circuits, perhaps even in brains of larger mammals.

This procedure obviates the use of aldehyde fixation and cryoprotection (glycerol). The present work demonstrates its applicability to the freeze-etching of mammalian brain tissue. Freeze-fracture replicas from rat cerebellar cortex and subfornical organ prepared by this method are compared to conventionally processed material using aldehyde fixation, glycerination and freezing with Freon. The formation of large ice crystals is prevented in tissue blocks up to 0.5 mm thick; deep etching is markedly enhanced. Cytoplasmic microstructures such as mitochondrial cristae, microtubules and microfilaments, are readily observable against a finely granulated cytosol matrix. An additional advantage is the combined application with freeze-substitution.

    Notes: Summary It has previously been reported that the primitive primate Tupaia belangeri develops a renal failure when exposed to psychosocial stress. In order to learn if this high susceptibility to stress of the Tupaia kidney can be correlated with morphological and functional parameters of the Juxtaglomerular apparatus (JGA) and the renin-angiotensin system, comparative experiments were performed on Tupaia and rat. Our results reveal an outstandingly high potency of the JGA and the renin-angiotensin system in Tupaia as evident from the following findings: The Tupaia JGA contains a great number of epithelioid cells abounding in renin granules (electron microscopy). The renin content of the Tupaia kidney is considerably higher than in the rat (radio-immunoassay). The sympathetic innervation of the kidney and especially of the JGA is abundant in Tupaia (fluorescence and electron microscopy). Catecholamine contents of the kidney and other organs are significantly higher in Tupaia than in rats (spectrophotofluorometry). Our results support the previously developed concept of a potent intrarenal neuroendocrine interaction at the JGA level favouring, under certain conditions of social stress, the development of acute renal failure in Tupaia belangeri.

    Notes: Summary Transection of Purkinje cell axons in adult male rats made 1.5 mm or further from the cell body does not lead to the death of the neuron and results in compensatory structural alterations of the surviving axonal portions of the nerve cell. Near to, and at the emergence of recurrent collaterals of Purkinje cell axons, huge varicosities filled with filaments, granular material, lysosomes and mitochondria develop. Terminals of recurrent axon collaterals also exhibit different degrees of structural changes. Most striking of the morphological alterations is the regular presence of nematosomes in the hypertrophic axonal branches, especially in synaptic terminals. Since nematosomes were shown to contain RNA in other types of neurons, their presence in recurrent collaterals may indicate an enhanced synthetic activity in Purkinje axonal processes and endings after axotomy.

    Notes: Summary The process of myelination in the central nervous system (CNS) of the rat (optic nerve) was studied with the freeze-fracturing technique and ultrathin sectioning to obtain information on the developmental mechanisms of interlamellar tight junctions. Using a tilting cartridge for analysis of thin sections, it could be demonstrated that during the initial phase of wrapping a tight junction formation develops between the joining tips of the oligodendrocytic process. In tannic acid-stained samples these junctions appear as typical quintuple-layered membrane fusions, while in potassium permanganate-stained material membrane thickenings between the apposing glial tips are prevalent. The latter configuration represents the characteristic feature of the so-called radial component of central myelin. Using the freeze-fracturing technique, a biphasic mode of the myelinic tight junction assembly was detected. It is suggested that tight junctions represent a prerequisite of the myelination process.

These features were present in vessels larger than 5 μm in diameter and more than 100 μm away from the uterine epithelium, both in the mesometrial and antimesometrial wall of the endometrium. Vessels from interimplantation sites displayed neither fenestrations nor interruptions. The endothelial cells of the implantation sites displayed morphological signs of metabolic activation. These consisted of increased numbers of polyribosomes, well developed Golgi complexes and prominent nucleoli. The fenestrations and gaps in the vessel wall were interpreted as constituting the morphological basis for the increase in vascular permeability and the consequent edema which characterize the Pontamine Blue Reaction.

    Notes: Summary Conspicuous differences in the distributional pattern of nerve fibers containing corticotropin-releasing factor (CRF) or posterior lobe hormones, respectively, were shown in the median eminence of the adult male rat by means of immunoperoxidase histochemistry, with the use of anti-CRF, anti-oxytocin, and anti-vasopressin sera. In the rostral and central divisions of the median eminence, a high concentration of CRF-immunoreactive nerve fibers was found in the median portion of the external layer; these fibers terminated on the capillary loops of the hypophysial portal system. In the caudal division of the median eminence, the CRF-immunoreactive nerve fibers were located in the median to paramedian portions of the external layer. Numerous oxytocin- and vasopressin-immunoreactive nerve fibers were observed evenly distributed throughout the internal layer of the median eminence. In the external layer, a small number of the oxytocin- and vasopressin-containing nerve fibers was found around the capillary loops, particularly in the median to paramedian portions. The distributional patterns of the CRF and the posterior lobe hormones in the hypothalamo-hypophysial system and their functional interrelationship are discussed.

    Notes: Summary Apart from the known hypothalamic controls, which have been well documented, a myriad of compounds both endogenous and exogenous have proved effective in influencing secretion of prolactin (PRL). Recent studies have shown that somatostatin (SRIF), when injected intra-atrially as a bolus, is able to inhibit PRL secretion in vivo. However, the inhibitory effect of SRIF occurs only in adenohypophyses previously primed with estradiol. This research was undertaken to examine the ultrastructural effects of that inhibition using male Sprague-Dawley rats primed for three weeks with subcutaneous implants of estradiol. Within 2 min of injection of SRIF (1 mg/kg), the adenohypophyses were removed and processed for electron microscopy. We found dramatic changes in the estradiol-primed mammotrophs, including 1) an apparent rearrangement of rough endoplasmic reticulum (RER) into concentric cisternae, and 2) the appearance of intracellular bodies closely associated with granules. These changes were not observed in non-estradiolprimed male rats injected with SRIF which is consistent with the fact that in the normal male rats, SRIF failed to inhibit PRL secretion. These findings suggest that SRIF causes reorganization of cellular organelles so that PRL granules are sequestered thereby inhibiting secretion of PRL.

    Notes: Summary The localization of T-and B lymphocytes and interdigitating cells (IDC) was investigated during the regeneration process of splenic implants. For this purpose a two-step immunoperoxidase technique was used to visualize T-cell antigen, immunoglobulins and Ia-antigen on cryostat sections. The specific localization of the repopulating lymphocytes occurred simultaneously with the development of non-lymphoid elements characteristic for the different compartments of the white pulp, i.e., the periarteriolar lymphocyte sheaths (PALS) and follicles. The marginal zone (MZ) developed after the PALS and primary follicles, but before germinal center reactions were found. During ontogeny, however, the development of a broad MZ precedes the formation of follicles. This difference in sequence of events is discussed.

    Notes: Summary The enzymatic activity of bone matrix vesicles from parathyroidectomized rats was determined and compared to the activity of vesicles from sham operated and normal animals. The vesicles were isolated from the alveolar bone by collagenase digestion and differential centrifugation and further purified on a discontinuous sucrose density gradient. The amount of extractable protein and the activity of alkaline phosphatase, acid phosphatase, and ATPase in the vesicle fractions thus obtained did not differ significantly from the values characteristic of preparations from control rats. It may therefore be suggested that parathyroid hormone depletion and the associated hypocalcemia have no significant effect on the occurrence and phosphatase activity of bone matrix vesicles.

    Notes: Summary The hypothesis that melatonin (aMT) is implicated in the regulation of the secretory process involved in the synthesis and release of protein/peptide “hormones” of the pineal organ has been tested in the present study by the use of an in vitro system in two mammalian species. In these species the secretory process studied is characterized by either large (hamster) or very small (rat) numbers of granular vesicles. In both species, aMT clearly participates in the regulation of the process of protein/peptide secretion. However, the effect of aMT varies with the presence of noradrenaline (NA) in the medium and is not identical in both species. Melatonin, in the absence of NA, induces the formation of granular vesicles by the Golgi apparatus in pinealocytes of the rat but not in those of the hamster, while in the presence of NA, aMT provokes a decrease in the number of these vesicles in both species. The present experiments show (i) that the pineal is one of the target organs for aMT, and (ii) that aMT is implicated in the control of protein/ peptide secretion in the pineal organ.

    Notes: Summary Pituitary folliculo-stellate cells and associated cells were studied immunohistochemically throughout the life of male and female rats. The marginal layer cells of the pars distalis and intermedia as well as the folliculo-stellate cells were immunostained with anti-S 100 protein serum. The immunostained folliculo-stellate cells in one section were identified as themselves by their ultrastructural properties on the adjacent section. Corticotrophs, thyrotrophs and prolactin cells, characterized by the stellate shape, were not immunostained with anti-S 100 protein serum. Reactivity for S-100 protein appeared on postnatal Day 6 in the marginal layer cells of the pars intermedia, and appeared on Day 10 in those cells of the pars distalis. No immunostained folliculostellate cells appeared before Day 6, but a few of them appeared on Day 10. Thereafter they increased in number, having more intense immunostaining with advancing age. In castrated rats, the immunostained folliculo-stellate cells appeared quite numerously, with branched cytoplasmic processes surrounding the gonadotrophs. In the thyroidectomized rats, however, folliculo-stellate cells lacked ramified cytoplasmic processes, and their topographic affinity for thyrotrophs was negligible.

    Notes: Summary Extensive secretion by exocytosis was demonstrated in the glomus (type I) cells of the adult rat after perfusion of carotid bodies with a potassium-rich (high K) glutaraldehyde fixative. Similar secretory profiles were very rare with a glutaraldehyde fixative containing a low concentration of potassium (low K). The increase in the incidence of exocytotic profiles in glomus cells with the high K fixative was highly significant, whereas no statistical difference could be observed in the incidence of coated pits with the different fixatives. Exocytotic profiles were characterized by the following features: (1) they predominated in non-synaptic regions, but were occasionally observed near synapses between two glomus cells; they were not observed near synapses between glomus cells and nerve terminals; (2) extruded electron-dense material associated with coating of the cell membrane was frequent; (3) different stages of dissolution of the extruded granule material was evident. The possible role of exocytosis as a mode of secretion in the glomus cells and the characteristics of the new high K-glutaraldehyde fixative are discussed.

    Notes: Summary A specific rabbit anti-CRF serum and the immunoperoxidase technique were used to show that CRF-containing neurons are mainly distributed in the paraventricular and supraoptic nuclei of the rat hypothalamus. In addition, immunoreactive neurons are scattered in other hypothalamic regions. These neurons are 20–30 μm in diameter. From the present and previous investigations it may be concluded that the hypothalamic magnocellular nuclei, i.e., paraventricular and supraoptic, and other hypothalamic accessory nuclei, are the producing sites not only for vasopressin and oxytocin, but also for corticotropin-releasing factor.

    Notes: Summary The present report describes the genesis, development and topographical distribution of ectopic cells of the external granular layer in the subarachnoid space covering the rat cerebellum. Following one intracisternal injection to newborn rats of 100 μg 6-hydroxydopamine (6-OHDA), the meningeal cells degenerate and are removed by phagocytosis within 24 h post injection (p.i.), leaving the cerebellar cortex without a pia-arachnoid cover. Defects appear in the basal lamina investing the cerebellar cortex 3 to 5 days p.i., and both external granule cells and ‘sprouts’ from Bergmann-glia endfeet grow into the subarachnoid space. The latter form large, flat glial lamellae and cover extensive areas of the denuded cerebellar surface, although they do not form a glial scar over the exposed neuropil of the cerebellar cortex. The numbers of ectopic external granule cells increase within the subarachnoid space both by proliferation and a continuous efflux of cells from the cerebellar cortex. They migrate, aggregate, and ultimately develop into granule, stellate and basket cells, the morphology of which is indistinguishable from their counterparts in situ; they make specific afferent and efferent connections, both among themselves and with the underlying cerebellar cortex and brainstem. The distribution of ectopic external granule cells and their derivatives is restricted to the anterior vermal fissures and the vermal-hemispheric junctions. The present results indicate that external granule cells and their derivatives are capable of both differentiating normally and surviving in the subarachnoid space if they become associated with glial cells and establish synaptic connections.

    Notes: Summary As luminal fluid moves from the seminiferous tubule and enters the rete testis, its protein concentration declines from approximately 6 mg/ml to 1 mg/ml. It was therefore suggested that protein is either 1) utilized by the spermatozoa, 2) transported across the epithelium of the terminal segment of the seminiferous tubule, the tubuli recti or rete testis, or 3) absorbed and degraded by the epithelium. Horseradish peroxidase (HRP), a protein marker, was microperfused into single seminiferous tubules or perfused directly into the rete. After fixation, the HRP was localized histochemically and the tissue observed under the light- and electron microscope. HRP was taken up via pinocytotic vesicles into the cytoplasm of the Sertoli cells and germ cells but did not permeate extracellularly beyond the tight junctions. Similar results were obtained in the cells lining the terminal segment and the tubuli recti. The rete epithelium showed uptake of HRP into coated and noncoated vesicles, while some cells additionally revealed diffuse cytoplasmic distribution of HRP. The terminal segment, tubuli recti, and rete testis may be important routes by which proteins may leave the testicular fluid either to be degraded or to enter the blood.

    Notes: Summary When the resorption zones of the distal femoral and proximal tibial epiphyseal plates of rats are examined with the electron microscope, numerous macrophages can be seen surrounding degenerated chondrocytes. Macrophages are also found in the neighbourhood of the invading sinusoids and in close proximity to multinucleate osteoclasts. Cell fusion, however, could not be observed. Repeated transfusions of female leukocytes to isohistogeneic growing male rats caused an increase in the number of nuclei containing a Barr body in the osteoclasts of the males. The result suggests that leukocytes — of which only the monocytes come in question — are the source of multinucleate osteoclasts in endochondral ossification.

    Notes: Summary The ontogeny of the somatostatin-containing neuron system was investigated by light-microscopic immunohistochemistry. During development, immunoreactive somatostatin-containing neurons arise from three discrete regions of the neuroepithelium of the third ventricle and show a chronological difference. The neurons are first evident within the third ventricle floor on day 12.5 of gestation; they move thereafter to the arcuate nucleus. The second generation occurs in the dorsal region of the arcuate nucleus during days 17.5–19.5; these neurons migrate sequentially into the arcuate-ventromedial nuclear region. The third generation is recognized in the neuroepithelial cell layer of the rostral hypothalamus on day 17.5 of gestation; these cells move to the periventricular area. This latter generation is most prominent during days 3–6 after birth, and some of the cells are seen sporadically even up to day 20. The first two generations give rise to the somatostatin neuron system in the arcuate-ventromedial nuclear region, while the latter gives rise to that in the rostral periventricular region in the adult rat hypothalamus.

    Notes: Summary This paper reports a stereological study of the cellular aspects of development of the adrenal cortex of male and female rats in the course of maturation. Rats of the Wistar strain were studied at weekly intervals from day 21 to day 77 of postnatal life. From day 42 homogenates of adrenals from female rats produced more corticosterone than homogenates from males of corresponding age. In the strain of rats studied sex differences in adrenal weight appeared at about day 49. From this day onward relative, and subsequently absolute adrenal weights were higher in females than in males. In the course of maturation the total volume of all adrenocortical zones gradually increased. From day 70 the zona glomerulosa and from day 49 the zona fasciculata were larger in female than in male rats, while no distinct sex difference was observed in the volume of the zona reticularis. The appearance of sex-related differences in the adrenal cortex depends mainly on changes in the zona fasciculata, the cells of which markedly increased in volume in female rats by comparison with males. The total number of all adrenocortical cells increased from ca 13 million on day 21 to ca 25 million on day 77 and at all intervals studied adrenal glands of male and female rats contained a similar number of parenchymal cells.

    Notes: Summary The architecture of the pancreas was revealed by retrograde injection of the pancreatic ductal system of normal rats with a silicone rubber compound, and subsequent study of the preparation by light microscopy and scanning electron microscopy. The injected material became associated with both ducts and “acinar” areas. Examination of these specimens suggests that the arrangement of the exocrine pancreas is that of a complexly curving and branching system of tubules which anastomose and end blindly. This architecture, which is not that of a true acinar gland, provides a rational basis for the understanding of the simple dedifferentiative changes that accompany pancreatic carcinogenesis, and which have been generally interpreted as representing ductular proliferation.

    Notes: Summary The present ultrastructural results indicate that, in the rat, the vasopressin-synthesizing perikarya of the supraoptic nucleus (NSO) attain a certain degree of maturity earlier than those of the paraventricular nucleus (NPV). In the neonate rat, the stainability of the nuclear areas is very weak; in the perikarya of the NSO a few labeled granules can be found, whereas the perikarya of the NPV often display only a labeled Golgi area, the cytoplasm being devoid of granules. At the end of the first (NSO) and the second (NPV) postnatal weeks, the filling of the neurosecretory granules with vasopressin is inhomogeneous with irregular spots of reaction product distributed on the granules. This feature is less obvious during the following week and has nearly disappeared after the third and fourth postnatal weeks. Already in the neonate two types of vasopressin-positive fibers are observed in the median eminence, characterized by the different diameters of their granules and by their typical location in the internal and the external pericapillary contact zone. Especially in one and two week-old animals, in the internal zone of the median eminence and, to a lesser degree in the neural lobe, the immunocytochemical reaction product is deposited on an axonal tubular network. Judging from the presence of very few vasopressin-negative fibers in the neural lobe of the neonate, the development of the oxytocin system appears to be delayed. A characteristic relationship between pituicytes and the neurosecretory fibers can be observed during the first two postnatal weeks. After the third postnatal week the immunocytochemical features of the vasopressin system correspond approximately to that in adult rats.

    Notes: Summary Scanning electron microscopy of the lining of the pituitary cleft was carried out in normal, lactating, castrated, adrenalectomized, and cyproterone-treated adult rats. Four cell types could be differentiated in the posterior wall in control and experimental animals: (1) cells with a smooth surface, (2) cells with microvilli located at the cellular borders, (3) ciliated cells, and (4) cells with evenly distributed microvilli. The anterior wall showed mainly cells with few microvilli located at their margins, and clusters of ciliated cells. In normal, and more frequently in experimental animals, the anterior wall showed shriveled cells, and variously sized cavities. Colloid appeared either as a network of finely granular material or as compact bodies adhering to the epithelial surface. These observations suggest that a compact component of the colloid is derived at least in part from degraded cells.

Ces cellules réagissent également quand un anticorps est utilisé contre la fraction épiphysaire UMO5R, fraction qui est douée de propriétés antigonadotropiques in vivo. Il a également été montré que le nombre des cellules immunoréactives était plus important dans la pinéale du jeune rat que dans celle de l'adulte. La comparaison des résultats obtenus avec différents anticorps et l'étude des propriétés de ces anticorps aprés absorption sur différents peptides ou sur différentes fractions épiphysaires, a permis de conclure que les réactions obtenues dans la pinéale du rat n'étaient que la conséquence d'une réaction croisée de ces anticorps avec une/des substance(s) inconnue(s) synthétisée(s) par la pinéale elle-même.     Notes: Summary Using antibodies against AVT, α-MSH, LHRH and somatostatin, immunoreactive cells were detected in the rat pineal gland. All of these antibodies stain the same cells, which also react immunocytochemically when an antibody against the UMO5R sheep pineal fraction, a fraction that presents antigonadotropic properties in vivo, is used. Relatively more immunoreactive cells are present in the pineals of young rats than in the pineals of adult animals. Comparison of the results obtained with different potent antibodies against each of the peptides, and a study of the staining properties of the antibodies in the pineal after solid phase adsorption to different peptides or to different sheep pineal fractions, led to the proposal that the immunoreactivity found in the rat pineal is not due to the presence of AVT, α-MSH, LHRH or somatostatin, but to a cross-reaction of each of these antibodies with (an) unidentified compound(s). This compound is synthetized in the pineal gland, as was demonstrated using cultured pineals. The UMO5R and the Prot. 4 fractions of the sheep pineal seem to be chemically related to this unknown compound, the possible endocrine nature of which is discussed.

    Notes: Summary The pineal gland of the rat receives a rich nervous supply originating from the superior cervical ganglia. These fibers contain serotonin in addition to their neurotransmitter, noradrenaline. Cytochemical studies at the ultrastructural level have shown that both amines are present in the cores of the granular vesicles that are characteristic of these nerves. It is presently shown that the bilateral electrical stimulation of the preganglionic fibers innervating the ganglia markedly reduces the number of small sites reacting cytochemically for both noradrenaline and serotonin, these sites corresponding to the cores of small granular vesicles, while the larger reactive sites (cores of large vesicles) remain unaltered. The vesicles are retained in nerve terminals after stimulation, as observed in conventionally processed tissues, although with altered sizes and shapes. Apart from these cytochemical and structural changes, nerve stimulation also reduces the endogenous noradrenaline content of the pineal gland. Thus, both noradrenaline and serotonin are released from their storage sites in pineal sympathetic nerves after electrical stimulation in vivo. This suggests the possibility that several substances with presumed transmitter or modulatory functions might be simultaneously released by nerve impulses from a given nerve terminal.

    Notes: Summary The B-cells of the endocrine pancreas constitute an adequate model for in vitro study of the aging process in highly differentiated cells. In the present study, collagenase-isolated islets of Langerhans from young and senescent rats were cultured up to 28 days. The response of the B-cells to the stimulatory conditions of the culture medium involved the nucleus, ribosomes, endoplasmic reticulum, Golgi apparatus, and secretory granules. Correlated data from light microscopy, electron microscopy, and insulin radioimmunoassay show that the differentiation and function of senescent B-cells are maintained in culture, as it has been proven for the B-cells of younger animals. On the other hand, signs of cytological deficiency not directly concerned with the specific function of B-cells were observed: abnormal mitochondria and lysosomes are more numerous in the senescent B-cells. The proliferative capacity of the B-cells of aged rats is reduced.

    Notes: Summary The dynamics of horseradish peroxidase (HRP) transport in primary sensory neurons were studied in rats by demonstration of the reaction product in spinal nerves, spinal ganglia, dorsal roots and in the spinal cord at different survival times after application of the enzyme to the transected sciatic nerve and to the spinal cord. Using tetramethylbenzidine as the chromogen according to Mesulam (1978), transganglionic transport of HRP was shown in both the disto-proximal direction after peripheral application, and proximo-distal direction after central application. Significant differences in staining intensity between the central and peripheral processes of primary sensory neurons were found after all survival times used in this study. After peripheral application the number of labeled axons and the staining intensity were higher in spinal nerves than in dorsal roots; an inverse situation occurred after central application. These differences as well as the time sequences in staining of different parts of primary sensory neurons suggest that HRP applied to a peripheral nerve and to the spinal cord, respectively, enters the perikarya of spinal ganglion cells in any case before continuing its movement in a cellulifugal direction. Lysosomal degradation of the major portion of the applied HRP is supposed. However, in the post-perikaryal portion of a considerable number of neurons HRP-transport still occurs to a varying extent, thus resulting in labeling of nerve endings. In some neurons a post-perikaryal transport could not be detected light microscopically. The transport rates differ: the calculated transport rate of disto-proximal, cellulipetal movement in the fastest transporting neurons was 7.5 mm/h, that of the disto-proximal cellulifugal movement 2.5 to 3 mm/h.

On the day of birth, numerous villi of various sizes and shapes were present on the caecal mucosa. After the 5th day, the villi decreased very rapidly in length and in number. A strong constriction was observed at the basal region of the caecal villi. During postnatal days 5 ∼ 9 the villi probably separated and disappeared from the caecal mucosa. Electron microscopy revealed that all ependymal cells including the tanycytes disappear or degenerate in rats subjected to electric cauterization of the ependymal layer lining the third ventricle. However, the granular axons in the palisade layer of the median eminence remain intact. In rats subjected to electric lesion, no significant change was found in either the serum-LH level or in the weight of the adenohypophysis, testes, adrenal and thyroid glands. It is concluded that the tanycytes do not participate in the hypothalamic regulation of hypophysial function.

    Notes: Summary With the aid of light- and electron- microscopic immunocytochemistry, somatostatin- and luliberin (LRF)-positive fibers can be demonstrated in the rat subfornical organ (SFO). Each of the neurohormones has a specific location: LRF in the lateral parts of the organ, and somatostatin in the center of the posterior zone. Common to both neurohormone-containing fibers is the pattern in which they reach the organ as well as the fact that their terminals are located in the perivascular spaces of fenestrated vessels, i.e., within the limited neurohemal regions of the organ. Since injection of India ink of different colors demonstrates that the capillary bed of the SFO is connected with the central capillaries of the choroid plexus, the question arises as to whether the neurohormones released in the area of the SFO influence the choroid plexus.

    Notes: Summary The results presented here reveal that in adult Sprague-Dawley and Wistar rats the pineal gland represents a complex rather than a single organ. Regularly one can distinguish (i) pineal tissue in the intercommissural region as a deep pineal, (ii) a superficial pineal, which represents the major part of the pineal complex, and (iii) nearly always a parenchymal stalk of variable length. The volume of the deep pineal with the adjacent parenchymal stalk exhibits great interindividual variation. It amounts to 127±39×105 μm3 (mean ±standard deviation). The histological appearance of the deep and superficial pineal tissue is fairly similar. The intrinsic cells of the deep and superficial pineal differ in nuances only. Karyometry reveals that the nuclear volumes of the intrinsic cells of the deep pineal are very variable ranging from 90–450 μm3, with a mean value of 207 μm3. The changes over a period of 24 h reach statistical significance.

    Notes: Summary With the aid of electron microscopic immunocytochemistry following the application of antisera against somatostatin and luliberin (LRF), a labeling of the intercellular clefts in different areas of the brain was observed. This labeling is especially conspicuous near the basal pole of the cuboidal ependymal cells, but is also generally present in all regions containing neurohormone-producing perikarya or their processes (for example, the preoptic area, the basal ganglia and the cortex). Furthermore, in all these regions displaying labeled intercellular clefts, glialike cells and sparsely ciliated ependymal cells are found, the secondary lysosomes of which exhibit an immunoreactivity resembling that observed in the intercellular clefts. As sources of the immunoreactive material the following possibilities are discussed: (i) perikarya producing somatostatin or LRF, situated in the wall of the third ventricle and sending fibers between the cuboidal ependymal cells, (ii) hypothalamic and extrahypothalamic projections of both peptidergic systems, and (iii) in the case of somatostatin, immunoreactive perikarya in the cortex.

    Notes: Summary Evans blue was injected into either the medial and lateral gastrocnemius or the soleus muscle of adult albino rats to mark retrogradely the corresponding motoneurones in the spinal cord. The labelled motoneurones were identified by the red fluorescence of their perikarya in the ventral horn of segments L4–L6. In addition, a monosynaptic reflex action potential was recorded only in the ventral roots L4–L6 after stimulation of the nerves to the medial and lateral gastrocnemius and the soleus muscle. Excitability and reflex latencies of labelled and unlabelled motoneurones of segments L4–L6 as well as the conduction velocities of their axons were measured. Apart from a small but statistically nonsignificant increase in excitability, no functional differences were found between labelled and control neurones. Thus, retrograde labelling of motoneurones with Evans blue prior to performing electrophysiological experiments has the advantage that the marked motoneurones can be identified under the fluorescence microscope without the need of additional staining or fixation.

    Notes: Summary In female Wistar rats the influence of adrenalectomy and NaCl administration on the amount of “classical” neurosecretory material (cNSM) in the supraoptico-hypophysial system and on vasopressin-like substance-containing granules (vlG) in the outer layer of the median eminence has been studied. In conjunction with appropriate sodium replacement, adrenalectomy induces an increase in the amount of vlG but does not alter the amount of cNSM. Administration of hypertonic saline diminishes cNSM but has no or only little influence on the amount of vlG. From the findings it is concluded that cNSM and vlG, in spite of their identical histochemical and immunohistochemical properties, have different functions. The functional significance of the vlG is discussed.

    Notes: Summary At the light microscopic level, following immunostaining with a single antiserum against luliberin (LRF), two types of hormone-producing perikarya in the preoptic area are demonstrated. The two cell types differ in their morphological features: a bipolar, smooth-contoured cell type can be differentiated from an irregularly contoured unipolar type. Intermediate forms between both cell types occurring in the same area are not observed. Electron microscopically, both cell types contain labeled granules of similar size and immunoreactivity. It is dicussed whether the uneven surface of the one cell type is due to areas of synaptic contacts, and whether both cell types are integrated in different neuronal and functional circuits. Moreover, at the ultrastructural level, from the irregularly contoured LRF-producing perikarya a further positively stained cell type, probably a glial cell, can be differentiated. The specific labeling of the latter is caused by its content of immunoreactive lysosomal bodies. Differentiation between the labeled glial cells and the irregularly contoured LRF-producing perikarya is not possible at the light microscopic level.

Another antiserum raised against somatostatin (antiserum “BS 102”) stains only the axons but not the perikarya of the hypothalamic system; the cortical somatostatin system does not react with this antiserum. The electron microscopic findings do not allow decision whether the above-mentioned hypothalamic and cortical neurons possess a common prohormonal form of somatostatin, immunoreactive only with antiserum “F”. They show, however, that the granules in both neuronal systems differ considerably; in the cortical neurons they measure approximately 65 nm in diameter, in the hypothalamic neurons 90–120 nm in diameter. Thus, both somatostatin systems are different and independent from one another.

    Notes: Summary Immunocytochemical and autoradiographic localization of thyrotropin-releasing hormone (TRH)-and 3H-TRH-binding sites was studied in the arcuate nucleus-median eminence region of the rat. TRH-like immunoreactivity was found in dense granular vesicles (90–140 nm in diameter) in TRH-like immunoreactive nerve fibers and terminals. In the median eminence, the immunoreactive terminals were observed to be in direct contact with the perivascular basal lamina of the portal vessel and to form synaptoid contacts with tanycytes. In the arcuate nucleus, the immunoreactive terminals were often found to form axosomatic and axo-axonic, and/or axo-dendritic synapses. The uptake of tritiated TRH into the nerve fibers and terminals of the median eminence was also observed by autoradiography and the distribution and localization of silver grains in them were analyzed quantitatively by circle analysis. Thirty minutes after intraventricular infusion of 3H-TRH, radioactive labeling occurred in type-2 and 3-nerve fibers and terminals containing dense granular vesicles in the median eminence. It is therefore suggested that the neurons labeled after 3H-TRH infusion possess certain functions as physiological recognition sites or receptors for TRH.

    Notes: Summary Freeze-fracture images of the parenchymal cells in the parathyroid gland of rats were observed after vitamin D2 plus calcium chloride-suppression and EGTA-activation of secretion. In cells of the suppressed glands, large bulges protruded from the Golgi cisternae, and large granules with a stalk, which are identified as storage granules, suggest that, during maturation, some storage granules may be connected by long tubules with the Golgi cisternae and supplied with secretory products from the Golgi cisternae via these tubules. In the activated glands, presumptive exocytotic and endocytotic specializations of intramembranous particles of the parenchymal cell plasma membrane were frequently observed. In addition, elevations and complementary shallow depressions of various shape and extent were occasionally encountered in the intercellular space. From their morphological characteristics it was concluded that these originated from secretory granule cores, which are discharged from the parenchymal cells into the intercellular space by exocytosis, and it was suggested that discharged granule cores may retain their spherical shape until they fuse to form a flat conglomerate.

In the olfactory epithelium proper, the cilia of receptor cells and microvilli of supporting cells possess a cell coat measuring approximately 10 nm in thickness. In the vomeronasal neuroepithelium, the apical cell coat is thicker than in the olfactory epithelium proper. On microvilli of vomeronasal receptor cells the cell coat varies in thickness from 15 to 20 nm, and on microvilli of supporting cells it measures approximately 75 nm. The functional implications of these findings are discussed.

    Notes: Summary The function of the microtubules that are present in the villus enterocytes of the mammalian small intestine is virtually unknown. In order to advance our knowledge about enterocyte microtubules, a quantitative ultrastructural comparison was carried out on enterocytes from rats injected intraperitoneally with colchicine (0.5 mg/100 g body weight) in saline and from rats injected with saline alone. Our morphometric and stereologic study demonstrates that colchicine treatment results in 1) an absolute decrease in microtubules, 2) a reduction in microvilli, essentially in length, 3) an increased thickness of the terminal web, 4) an increase in total lysosomal volume, apparently by an increased number of smaller lysosomes, and 5) a decrease in the number of Golgi lamellae. These results along with those from other studies suggest to us that enterocyte microtubules are involved in the biogenesis of microvillus plasma membrane. Our morphometric data from the saline-treated rats essentially agree with comparable data from other studies. However, comparison with comparable data from hamster enterocytes demonstrates species differences.

    Notes: Summary Pseudointranuclear inclusions resulting from invaginations of the nuclear envelope, characteristic of myopathic muscles, were found in the soleplate nuclei of normal soleus muscles of rats. Such inclusions had annulate lamellae (AL), nemaline or rod bodies, lamellar structures, a concentric system of membranes, as well as tightly packed sarcoplasmic organelles. An accumulative origin of such included structures was suggested. The AL were sometimes connected directly to the outer membrane enclosing the inclusions, dilated into cisterns and positioned perpendicularly against the nuclear surface. AL, some of which were welldeveloped, were observed in the same loci as above during the course of denervation atrophy.

    Notes: Summary Cells emitting orange-red autofluorescence have been found in the pars intermedia of aging rats. The fluorescence maximum of the emission is localized in an area of the spectrum where the most intense band maxima of porphyrins are located. The fluorescence fades when the excitation wavelength is about 400 nm, which is specific (Soret band) for the absorption spectra of porphyrins. The fluorescence is emitted by coarse inclusions in the cytoplasm of a few cells. These inclusions are also stainable with paraldehyde-fuchsin and exibit a high endogenous peroxidase activity. The inclusions observed have morphologic features similar to those of porphyrin-containing astrocytes from the periventricular area of the hypothalamus. The inclusion-bearing cells of the pars intermedia also contain debris that might be due to the phagocytic activity of these cells.

    Notes: Summary The intervascular segments of the leptomeninges of the rat were studied by the use of horseradish peroxidase (HRP) in short-term experiments. HRP was injected (i) intravenously, (ii) into the lateral ventricle, (iii) into the cortex, and (iv) into the meninges. The composition of the meninges covering the median eminence (ME) was analyzed in comparison to the results obtained with the parietal cortex. The meninges covering the cortex show the following pattern of layers and compartments: The intercellular compartment comprises the intercellular clefts of the neuropil, the subpial space, and the intercellular clefts of the leptomeninges. The pial space establishes a second compartment. The third compartment is the arachnoid space. The intercellular clefts of the dura form the fourth compartment. At the border of the ME, the neurothelium and the outer arachnoid layer are rolled up to form a tissue frame around a hollow pit that is covered by a diaphragm consisting of meningeal cells; the latter separate the hemal milieu of the ME from that of the dura. The hemal and the cerebrospinal fluid (CSF) milieus may communicate to a limited extent only within the subpial space adjacent to the ME. The CSF-containing compartments of the pial and arachnoid spaces terminate at the brain-facing insertion of the tissue frame. According to the present results, an anatomical basis for a short-loop feedback from and to the neurohemal region of the ME via the CSF does not exist.

    Notes: Summary Immunohistochemical studies with the use of the peroxidase-antiperoxidase (PAP) method revealed that “amoeboid microglial cells”, in the brains of neonatal rats and “brain macrophages” in lesioned brains of adult rats react positively to an antiserum raised against macrophages. In brains of neonatal rats, “amoeboid microglial cells” stained by means of the PAP-method were observed in the corpus callosum, internal capsule, dorso-lateral region of the thalamus, subventricular zone of the lateral ventricle, and the subependymal layer of the ventricular system. These cellular elements were not detected in brains of rats aged 21 days or older. Resting microglial cells displaying a typical ramified structure were not specifically stained. Cells reacting positively to the macrophage antiserum appeared (i) in the cerebral cortex of adult rats following placement of a stab wound, or (ii) in the hippocampal formation after kainic acid-induced lesions; in the damaged areas immunoreactive cells exhibited the typical features of “brain macrophages”. “Brain macrophages” and “amoeboid microglial cells” are considered to belong to the class of exudate macrophages derived from blood monocytes. Thus, elements of hematogenous origin do exist in the intact brain parenchyma of neonatal rats and in lesioned brains of adult rats. The relationship between brain macrophages and resting microglial cells is discussed.

    Notes: Summary The influence of adrenalectomy and administration of hypertonic saline on the amount of vasopressin, oxytocin, and neurophysin contained in the median eminence and the neural lobe of rats was studied by means of the following methods: (i) morphometric and microphotometric analyses of aldehyde fuchsin-stained histological sections of the neurohypophysis; (ii) immunohistochemical demonstration of vasopressin, oxytocin, and neurophysin in the neurohypophysis, and (iii) radioimmunological measurement of vasopressin and oxytocin in extracts of the median eminence and the neural lobe. Adrenalectomy increases the amount of vasopressin and neurophysin in the external layer of the median eminence but does not change the content of oxytocin. It has no influence on the amount of vasopressin, oxytocin, and neurophysin demonstrable in the inner layer of the median eminence and in the neural lobe two weeks after the operation. Hypertonic saline markedly diminishes the vasopressin, oxytocin, and neurophysin content of the inner layer of the median eminence and the neural lobe but reduces only slightly, if at all, the amount of vasopressin and neurophysin in the outer layer of the median eminence. The findings support the concept that osmotic stress reduces only the vasopressin and oxytocin content of the hypothalamus-neural lobe system and has no or only little influence on the vasopressin content of the outer layer of the median eminence.

    Notes: Summary The ultrastructure of macrophages and reticular cells of regional lymph nodes of the rat after administration of large doses of cortisone acetate, estrone, progesterone, and cholesterol in aqueous suspensions was investigated. A large number of vacuoles, most of which were surrounded by unit membrane, and lipid droplets not surrounded by unit membrane were observed in the cytoplasm of both macrophages and reticular cells. They were not seen in these cells of control animals and in experimental animals that had received smaller doses of these steroid hormones. After cholesterol injection, many lipid droplets were observed in the cytoplasm of macrophages. These observations suggest that steroids injected in suspension accumulate in macrophages and reticular cells of the regional lymph nodes. Electron-dense material was often present in vacuoles of macrophages but not in those of reticular cells.

    Notes: Summary The ability of ova collected from mated middle-aged rats (9–10 months of age) to undergo preimplantation development in vitro was compared to that of younger controls (3–4 months of age). The majority of ova from young rats underwent at least one cleavage division for every 24 h of the 96 h culture period. Seventy-two percent reached the morula/blastocyst stage at the termination of the culture. Aged ova had a reduced capacity to undergo preimplantation development in vitro with the number of fragmented and/or degenerated ova increasing after each day in culture. At the end of the culture period, 81.6% of the ova had degenerated. It is concluded that middle-aged rats ovulate defective ova and that these defective ova account, in part, for the preimplantation losses that characterize middle-aged rats.

    Notes: Summary Previous studies have shown that LHRH or LHRH-like substances are present in the pineal gland. In order to investigate whether exogenous LHRH may affect the pineal gland, in the present study the effects of a single dose of LHRH (1 μg, i.p.) on pineal “synaptic” ribbons and spherules as well as serum melatonin levels were examined in diestrous Wistar rats. One hour after the injection both ribbons and spherules exhibited a statistically significant decrease in number. Serum melatonin levels were not affected. It is concluded that humoral feedback mechanisms may exist between the hypothalamus and the pineal gland.

    Notes: Summary Immunoreactive neuropeptide Y (NPY) was demonstrated in neuronal elements in the gut and pancreas of the rat. Immunoreactive endocrine cells could not be detected. The occurrence of NPY containing nerve-cell bodies in the submucosal and myenteric ganglia indicates an intrinsic origin of the NPY fibers. However, an additional extrinsic supply of NPY fibers is suggested by the finding that abdominal sympathectomy caused the disappearance of some NPY fibers, notably those around blood vessels. The distribution of NPY fibers in all layers of the gut wall suggests multiple functions of NPY, including a role in the regulation of intramural neuronal activities, smooth muscle tone, and local blood flow.

    Notes: Summary An electron microscopic study has been carried out in order to examine the permeability of the blood-brain barrier in the median eminence of perinatal rats. After several minutes, intravascularly injected electron-dense tracers (lanthanum nitrate; horseradish peroxidase, 40000 MW MW: molecular weight , ferritin, 500000 MW) pass the capillary wall, the perivascular space, and become incorporated into neurosecretory axons and basal processes of tanycytes both in fetuses and young rats. In the case of immature capillaries, the materials diffuse freely through the endothelial cells, and to a lesser extent are transferred via occasional plasmalemmal vesicles and fenestrae. As the maturation of capillaries proceeds, their permeability via plasmalemmal vesicles and fenestrae increases considerably due to a gradual rise of the number of these structures. The plasmalemmata of the differentiated endothelial cells become impermeable to all of the tracers. Only ionic lanthanum appears to penetrate through transendothelial channels and intercellular junctions between adjacent endothelial cells.

    Notes: Summary Satellite cells were studied in the undamaged part of the rat soleus muscle rendered partially necrotic by a subcutaneous injection of notexin, the myotoxic toxin purified from the venom of the Australian snake Notechis scutatus scutatus. Nuclei of satellite cells were twice as numerous as in the controls, with a decreasing gradient in the number of satellite cells with distance from the necrotic area. The cells were in an activated state with an increased cytoplasmic volume and prominent organelles. Occasionally, mitosis of some satellite cells was observed. Between the satellite cell and the muscle fibre, an unusually wide space was frequently seen, within which a “new” basal lamina was often visible. It is suggested that the role of the satellite cells of undamaged muscle fibres in the regeneration of necrotic muscle is worthy of more detailed investigation.

    Notes: Summary The axonal transport of neurosecretory material was studied in neurosecretory axons of the supraoptico-posthypophyseal system after in-situ transection of the median eminence. Two hours, 8 h, and 18 h after the lesion, both vasopressin and oxytocin antibodies revealed progressive accumulations of immunoreactive material not only in the proximal but also in the distal stumps of the transected axons. The electronmicroscopic examination of these axonal portions revealed that such intense immunopositive labelings could be correlated, in both stumps, to a conspicuous accumulation of neurosecretory granules. It is concluded that, under normal physiological conditions, a significant amount of axoplasmic neurosecretory material is transported in retrograde direction and that such a retrograde transport mainly involves neurosecretory granules.
