    Beschreibung: The Radiat experiment was one of 17 investigations which used the ESA Biorack on IML-1 (International Microgravity Laboratory) and it had two objectives. The first objective was to isolate and characterize mutations induced by cosmic rays; the second was to assess the fidelity of development in 0-gravity over two consecutive generations. Two strategies were used to isolate mutations in a set of essential genes or a specific gene and to correlate the genetic events with the passage of charged particles. The results were isolation of 60 lethal mutations whose phenotypes are related to the local pattern of energy deposition. 12 mutations in the unc-22 gene include large deletions as characterized by DNA hybridization studies. Development of nematodes proceeded through two consecutive generations with no obvious defects. Cytoplasmic determinants in embryos, nuclear location and symmetry of cellular anatomy were normal as were Mendelian segregation and recombination of genetic markers.

    Beschreibung: An experiment using isolated skeletal tissues under microgravity, is reported. Fetal mouse long bones (metatarsals) were cultured for 4 days in the Biorack facility of Spacelab during the IML-1 (International Microgravity Laboratory) mission of the Space Shuttle. Overall growth was not affected, however glucose consumption was significantly reduced under microgravity. Mineralization of the diaphysis was also strongly reduced under microgravity as compared to the on-board 1 g group. In contrast, mineral resorption by osteoclasts was signficantly increased. These results indicate that these fetal mouse long bones are a sensitive and useful model to further study the cellular mechanisms involved in the changed mineral metabolism of skeletal tissues under microgravity.

    Beschreibung: The free-flyer biostack experiment is part of a radiobiological space research program that includes experiments in space as well as in accelerators on Earth. The program has been specially designed to increase knowledge concerning the importance, effectiveness, and hazards of the structured components of cosmic radiation to man and to any biological specimen in space. Up to now, our understanding of the ways in which HZE particles might affect biological matter is based on a few spaceflight experiments from the last Apollo missions and on the limited data available from heavy-ion irradiation from accelerators. In the near future, accelerators capable of accelerating particles up to higher atomic numbers and higher energies will promote increased activity in ground-based studies on biological effects of HZE particles. Comparison of data from such irradiation experiments on Earth with those from an actual spaceflight experiment will show any potential influence of the inevitably attendant spaceflight factors (e.g., weightlessness) on the radiobiological events.

    Beschreibung: The need for 24-h operations creates nonstandard and altered work schedules that can lead to cumulative sleep loss and circadian disruption. These factors can lead to fatigue and sleepiness and affect performance and productivity on the job. The approach, research, and results of the NASA Ames Fatigue Countermeasures Program are described to illustrate one attempt to address these issues in the aviation environment. The scientific and operational relevance of these factors is discussed, and provocative issues for future research are presented.

    Beschreibung: The key to understanding the mechanisms of many important biological processes such as photosynthesis and respiration is a better understanding of the electron transfer processes which take place between metal atoms (and other groups) fixed within large protein molecules. Research is currently focused on the rate of electron transfer and the factors that influence it, such as protein composition and the distance between metal atoms. Current models explain the swift transfer of electrons over considerable distances by postulating bridge-mediated tunneling, or physical tunneling pathways, made up of interacting bonds in the medium around and between donor and acceptor sites. The program PATHWAYS is designed to predict the route along which electrons travel in the transfer processes. The basic strategy of PATHWAYS is to begin by recording each possible path element on a connectivity list, including in each entry which two atoms are connected and what contribution the connection would make to the overall rate if it were included in a pathway. The list begins with the bonded molecular structure (including the backbone sequence and side chain connectivity), and then adds probable hydrogen bond links and through-space contacts. Once this list is completed, the program runs a tree search from the donor to the acceptor site to find the dominant pathways. The speed and efficiency of the computer search offers an improvement over manual techniques. PATHWAYS is written in FORTRAN 77 for execution on DEC VAX series computers running VMS. The program inputs data from four data sets and one structure file. The software was written to input BIOGRAF (old format) structure files based on x-ray crystal structures and outputs ASCII files listing the best pathways and BIOGRAF vector files containing the paths. Relatively minor changes could be made in the input format statements for compatibility with other graphics software. The executable and source code are included with the distribution. The main memory requirement for execution is 2.6 Mb. This program is available in DEC VAX BACKUP format on a 9-track 1600 BPI magnetic tape (standard distribution) or on a TK50 tape cartridge. PATHWAYS was developed in 1988. PATHWAYS is a copyrighted work with all copyright vested in NASA. DEC, VAX, VMS, and TK50 are trademarks of Digital Equipment Corporation.     Beschreibung: The role of the tail in the altered thermoregulation of rats during acute exposure to hypergravity was investigated, using groups of rats of two ages: 55 days (young) and 138 days (old). Rectal and foot temperature changes were measured in intact and tailless rats subjected to 1 h centrifugation of 2.8 G, with preceding (1 h) and following (1-3 h) 1 G periods. At 22 C, the loss of body heat from the tail per se does not measurably contribute to the hypothermia induced by hypergravity. However, the heat loss from the feet was greater in the tailless rats than in the intact rats from the young group of animals, although there was no significant difference between the tailless and intact rats in the old animal group. It is concluded that the inhibition of heat production is a significant factor in the hypothermia of centrifuged tailless rats, as it has been previously shown to be in the intact animals.

    Beschreibung: The reduction of protein content in skeletal muscle undergoing disuse-induced atrophy is correlated with accelerated rates of protein degradation and reduced rates of protein synthesis (Goldspink, 1977). It is not known in what manner myofibers are partially disassembled during disuse atrophy to fibers of smaller diameter; nor is it known which proteases are responsible for this morphological change in contractile protein mass. Dayton and colleagues (1975) have suggested that the Ca(2+)-activated protease (CaP) may initiate myofibril degradation. The discovery of a form of CaP that is activatable by nano-molar concentrations of Ca(2+) indicates that CaP activity may be regulated by physiological concentrations of Ca(2+) (Mellgren, 1980). The enhancement of proteolysis by the Ca(2+) ionophore A23187, reported by Etlinger (1979), is consistent with a significant role for CaP in protein degradation. It was of interest, therefore, to measure the levels of CaP activity and the CaP inhibitor in extracts obtained from skeletal muscles of rat and chicken limbs undergoing disuse atrophy or stretch hypertrophy, respectively.

    Beschreibung: The tail suspension model (head-down tilt) simulates hypogravity in terms of musculoskeletal loss in the rat. However, little is known of tissue fluid shifts and body weight distribution in this model. Tissue fluid pressures were measured by wick catheters in 12 Munich-Wistar rats before, during, and after 48 hrs of tail suspension (about 30 deg head-down tilt). Subcutaneous tissue fluid pressure in the neck increased from -2.2 + or - 0.4 (normal horizontal position) to +4.0 + or - 1.5 cm H2O during tail suspension, indicating a cephalic fluid shift and significant edema during head-down tilt. In a separate study, six rats were suspended at 30-70 deg, and forelimb load and tail tension were measured by a balance and force transducer, respectively. Approximately 50 percent of body weight (BW) was loaded on forelimbs at a head-down tilt angle of 30 deg and forelimb load declined linearly to 10 percent BW at 70 deg. Furthermore, tail tension increased from 50 percent BW at 30 deg to 85 percent BW at 70 deg. These results indicate that less than normal loads are applied to forelimbs of rats suspended at angles of less than 30 deg and that the tail bears an increasing proportion of the rat's body weight at head-down tilt angles of less than 30 deg.

    Beschreibung: Antibody production by two different cell lines was measured as the media were supplemented with varied amounts of glucose and fetal bovine serum. Both cell lines elaborated antidinitrophenyl hapten antibodies. Two basic media were used: RPMI 1640 and Dulbecco's modified Eagle's medium. The production of antibodies was followed from 0 to 180 h and was assayed by radioimmunoassay.

    Beschreibung: The immune response in rats exposed to simulated hypergravity (2.1 G and 3.1 G) by chronic centrifugation was assessed. Rats were immunized with sheep red blood cells (SRBC), either on the day of initial exposure to hypergravity (hyper-G), or after being centrifuged for 28 d and remaining on the centrifuge thereafter. Pair-fed and ad libitum fed noncentrifuged controls were used. Although there were some alterations in leukocyte counts, hyper-G did not systematically affect the primary or secondary anti-SRBC response, hematocrits, or the sizes of the liver, spleen, kidneys, thymus, or adrenal glands. The immune system is thus remarkably homeostatic under hypergravity conditions which do affect other physiologic parameters.

    Beschreibung: Cancer is the most important risk of radiation exposure. There is a definite lack of suitable test systems, human epidemiological data are only available for certain radiation types, especially not for charged particles. We use the Xiphophorus model which is genetically well characterized. As a prelude to experiments with heavy ions we report here on results obtained with x-rays to establish the necessary baseline for future studies. Apart from this direct aim we hope to obtain also a better insight in the genetical determination of cancer formation.

    Beschreibung: Since more than 20 years ago, when the National Academy of Sciences and the National Research Council of the U.S.A. released their report on 'HZE particle effects in manned spaced flight', it has been emphasized how difficult - if not even impossible - it is to assess their radiobiological impact on man from conventional studies where biological test organisms are stochastically exposed to 'large' fluences of heavy ions. An alternative, competing approach had been realized in the BIOSTACK experiments, where the effects of single cosmic as well as accelerator - heavy ions on individual biological test organisms could be investigated. Although presented from the beginning as the preferable approach for terrestrial investigations with accelerator heavy ions too ('The BIOSTACK as an approach to high LET radiation research'), only recently this insight is gaining more widespread recognition. In space flight experiments, additional constraints imposed by the infrastructure of the vehicle or satellite further impede such investigations. Restrictions concern the physical detector systems needed for the registration of the cosmic heavy ions' trajectories as well as the biological systems eligible as test organisms. Such optimized procedures and techniques were developed for the investigations on chromosome aberrations induced by cosmic heavy ions in cells of the stem meristem of lettuce seeds (Lactuca sativa) and for the investigation of the radiobiological response of Wolffia arriza, which is the smallest flowering (water) plant. The biological effects were studied by the coworkers of the Russian Institute of Biomedical Problems (IBMP) which in cooperation with the European Space Agency ESA organized the exposure in the Biosatellites of the Cosmos series. Since biological investigations and physical measurements of particle tracks had to be performed in laboratories widely separated, the preferred fixed contact between biological test objects and the particle detectors until the geometrical correlation between tracks and organisms has been established could not be maintained. This gave rise to half a dozen of coordinate systems for different measurements which finally had to be related to a single stack reference system.

    Beschreibung: The spatial distribution of energy deposition is an important determinant in the formation of biologically significant lesions. It has been widely realized that Linear Energy Transfer (LET) being an average quantity is not sufficient to describe the situation at a submicroscopic scale. To remedy this to some extent 'energy-cut-off' values are sometimes used but since they are related to secondary electron energy and only indirectly to their range they are also not adequate although they may be easily calculated. 'Range-restricted LET' appears to be better but its determination is usually quite involved. Xapsos (1992) suggested a semi-empirical approximation based on a modified Bethe-formula which contains a number of assumption which are difficult to verify. A simpler and easier way is to use existing beam-models which describe energy deposition around an ion's path. They all agree that the energy density (i. e., energy deposited per unit mass) decreases with the inverse square of the distance from the track center. This simple dependence can be used to determine the fraction of total LET which is deposited in a cylinder of a given radius. As an example our own beam model. Energy density depends on distance x (measured in m) from the track center according to the presented formula.

    Beschreibung: A material balance model for ethylene was developed and applied retrospectively to data obtained in the Biomass Production Chamber of CELSS in order to calculate true plant production rates of ethylene. Four crops were analyzed: wheat, lettuce, soybean, and potato. The model represents an effort to account for each and every source and sink for ethylene in the system. The major source of ethylene is the plant biomass and the major sink is leakage to the surroundings. The result, expressed in the units of ppd/day, were converted to nl of ethylene per gram of plant dry mass per hour and compare favorably with recent glasshouse to belljar experiments.

    Beschreibung: Electrophoresis has been used for the separation of particles, ions, and molecules for a number of years. The technology for separation and detection of the results has many applications in the life sciences. One of the major goals of the scientific community is to separate DNA molecules and intact chromosomes based upon their different lengths or number of base pairs. This may be achieved by using some of the commercially available and widely used methods, but these processes require a considerable amount of time. The challenge is to achieve separation of intact chromosomes in a short time, preferably in a matter of minutes.

    Beschreibung: Heavy ions (HZE-particles) constitute an important part of radiation in space. Although their number is small the high amount of energy transferred by individual particles may cause severe biological effects. Their investigation requires special techniques which were tested by experiments performed at the UNILAC at the GSI (Darmstadt). Diploid yeast was used which is a suitable eucaryotic test system because of its resistance to extreme conditions like dryness and vacuum. Cells were placed on nuclear track detector foils and exposed to ions of different atomic number and energy. To assess the action of one single ion on an individual cell, track parameters and the respective colony forming abilities (CFA) were determined with the help of computer aided image analysis. There is mounting evidence that not only the amount of energy deposited along the particle path, commonly given by the LET, is of importance but also the spatial problem of energy deposition at a submicroscopical scale. It is virtually impossible to investigate track structure effects in detail with whole cell populations and (globally applied) high particle fluences. It is, therefore, necessary to detect the action of simple ions in individual cells. The results show that the biological action depends on atomic number and specific energy of the impinging ions, which can be compared with model calculations of recent track structure models.

    Beschreibung: Our investigations of heavy ion-induced mutations in mammalian cells, which had been begun a few years ago, were systematically continued. For the first time, it was possible to cover a large LET range with a few kinds of ions. To do this, both UNILAC and SIS were used to yield comparable data for a large energy range. This is a necessary condition for a comprehensive description of the influence of such ion parameters as energy and LET. In these experiments, the induced resistance against the poison 6-thioguanin (6-TG), which is linked to the HPRT locus on the genome, is being used as mutation system. In addition to the mutation-induction cross-section measurements, the molecular changes of the DNA are being investigated by means of Multiplex PCR ('Polymerase Chain Reaction') gene amplification. From these experiments we expect further elucidation of the mutation-inducing mechanisms composing the biological action of heavy-ion radiation.

    Beschreibung: From a compilation of experimental data on the mutagenic effects of heavy ions in bacteria, main conclusions have been drawn as follows: (1) The mutagenic efficacy of heavy ions in bacteria depends on physical and biological variables. Physical variables are the radiation dose, energy and charge of the ion; the biological variables are the bacterial strain, the repair genotype of bacteria, and the endpoint investigated (type of mutation, induction of enzymes related to mutagenesis); (2) The responses on dose or fluence are mainly linear or linear quadratic. The quadratic component, if found for low LET radiation, is gradually reduced with increasing LET; (3) At low values of Z and LET the cross section of mutation induction sigma m (as well as SOS response, sigma sos. and lambda phage induction, sigma lambda versus LET curves can be quite consistently described by a common function which increases up to approximately 100 keV/mu m. For higher LET values, the sigma(m) versus LET curves show the so-called 'hooks' observed also for other endpoints; (4) For light ions (Z is less than or equal to 4), the cross sections mostly decrease with increasing ion energy, which is probably related to the decrease of the specific energy departed by the ion inside the sensitive volume (cell). For ions in the range of Z = 10, sigma(m) is nearly independent on the ion energy. For heavier ions (Z is greater than or equal to 16), sigma(m) increases with the energy up to a maximum or saturation around 10 MeV/u. The increment becomes steeper with increasing atomic number of the ion. It correlates with the increasing track radius of the heavy ion; (5) The mutagenic efficiency per lethal event changes slightly with ion energy, if Z is small indicating a rough correlation between cellular lethality and mutation induction, only. For ions of higher Z this relation increases with energy, indicating a change in the 'mode' of radiation action from 'killing-prone' to 'mutation-prone'; and (6) Repair genotype substantially influences the radiation induced mutagenesis. Different mechanisms of mutation induction and/or different types of biologically significant lesions in wild type cells compared to repair deficient strains are a likely explanation.

    Beschreibung: Investigation of radiation induced damage in mutant strains of mammalian cells which show a defect in the rejoining of DNA double strand breaks provides an unique opportunity to examine the role of double strand breaks and the mechanisms of double strand break rejoining in the production of chromosome aberrations. This is particularly important, because there is increasing evidence that the DNA double strand break is the major lesion responsible for the formation of chromosome aberrations. To address this issue, we studied the induction of chromosome aberrations in xrs-5 cells, an x-ray sensitive strain of a Chinese hamster ovary cell line, which shows a defect in the rejoining of double strand breaks and their wild-type parent CHO-cells. Because radiosensitivity depends strongly on cellular age, the experiments were performed with synchronous cells.

    Beschreibung: Besides inactivation one of the major interests in our experiments is to study the primary damage in the DNA double strand breaks (DSB) after heavy ion irradiation. These damages lead not only to cell death but also under repair activities to mutations. In further experiments we have investigated the inactivation with two different strains of Deinococcus radiodurans (R1, Rec 30) and the induction of DSB as well as the rejoining of DSB in stationary cells of E. coli (strain B/r) irradiated with radiations of different quality. In the latter case irradiations were done so that the cell survival was roughly at the same level. We measured the DSB using the pulse field gelelectrophoresis which allows to separate between intact (circular) and damaged (linear) DNA. The irradiated cells were transferred to NB medium and incubated for different times to allow rejoining.

    Beschreibung: DNA double strand breaks (DSB's) are the critical lesions involved in cellular effects of ionizing radiation. Therefore, the evaluation of DSB induction in mammalian cells after heavy ion irradiation is an essential task for the assessment of high-LET radiation risk in space. Of particular interest has been the question of how the biological efficiency for the cellular inactivation endpoint relates to the initial lesions (DSBs) at varying LETs. For cell killing, an increased Relative Biological Efficiency (RBE) has been determined for highLET radiation around 100-200 keV/mu m. At higher LET, the RBE's decrease again to values below one for the very heavy particles. At GSI, DSB-induction was measured in CHO-K1 cells following irradiation with accelerated particles covering a wide LET range. The electrophoretic elution of fragmented DNA out of agarose plugs in a constant electrical field was applied for the detection of DSB's. The fraction of DNA retained was determined considering the relative intensities of ethidium bromide fluorescence in the well and in the gel lane. Dose-effect curves were established, from which the RBE for DSB induction was calculated at a fraction of 0.7 of DNA retained In summary, these rejoining studies are in line with an enhanced severity of the DNA DSB's at higher LET's, resulting in a decreased repairability of the induced lesions. However, no information concerning the fidelity of strand breaks rejoining is provided in these studies. To assess correct rejoining of DNA fragments an experimental system involving individual DNA hybridization bands has been set up. In preliminary experiments Sal I generated DNA fragments of 0.9 Mbp were irradiated with xrays and incubated for repair However, restitution of the original signals was not observed, probably due to the high radiation dose necessary for breakage of a fragment of this size. A banding pattern with NotI hybridization signals in a higher MW range (3Mbp) has been obtained by varying the electrophoretic conditions and correct rejoining studies will be further developed in this system.

    Beschreibung: Molecular changes at the DNA are assumed to be the main cause for radiation effects in a number of organisms. During the course of the last decades techniques have been developed for measuring DNA double-strand breaks (dsb), generally assumed to be the most critical DNA lesions. The outcome of all those different approaches portrays a collection of data useful for a theoretical description of radiation action mechanisms. However, in the case of heavy ion induced DNA dsb the picture is not quite clear yet and further projects and strategies have to be developed. The biological systems studied in our group are yeast and mammalian cells. While in the case of yeast cells technical and methodical reasons highlight these organisms mammalian cells reach greater importance when dsb repair studies are performed. In both types of organisms the technique of pulsed-field gel electrophoresis (PFGE) is applied, although with different modifications and evaluation procedures mainly due to the different genome sizes.

    Beschreibung: Radiation effects caused by single cosmic ray particles have been studied for many years in radiobiological experiments for different biological objects and biological end-points. Additionally, single event effects in microelectronic devices have gained large interest. There are two fundamental mechanisms by which a single particle can cause radiation effects. On the one hand, a cosmic ray ion with high linear energy transfer can deposit a high dose along its path. On the other hand, in a nuclear collision, a high dose can be deposited by short range particles emitted from the target nucleus. In low earth orbits a large contribution to target fragmentation events originates from trapped protons which are encountered in the South Atlantic Anomaly. These protons have energies up to a few hundred MeV. We study the fragmentation of C, O and Si nuclei - the target nuclei of biological material and microelectronic devices - in nuclear collisions. Our aim is to measure production cross sections, energy spectra, emission directions and charge correlations of the emitted fragments. The present knowledge concerning these data is rather poor. M. Alurralde et al. have calculated cross sections and average energies of fragments produced from Si using the cascade-evaporation model. D.M. Ngo et al. have used the semiempirical cross section formula of Silberberg and Tsao to calculate fragment yields and the statistical model of Goldhaber to describe the reaction kinematics. Cross sections used in these models have uncertainties within a factor of two. Our data will help to test and improve existing models especially for energies below 300 MeV/nucleon. Charge correlations of fragments emitted in the same interaction are of particular importance, since high doses can be deposited if more than one heavy fragment with a short range is produced.

    Beschreibung: The membrane-bound NADH oxidase of Paracoccus halodenitrificans was inhibited by dicoumarol, 2-n-heptyl-4-hydroxyquinoline-N-oxide (HQNO), and exposure to ultraviolet light (at 366 nm). When the membranes were extracted with n-pentane, NADH oxidase activity was lost. Partial restoration was achieved by adding the ubiquinone fraction extracted from the membranes. Succinate oxidation was not inhibited by dicoumarol or HQNO but was affected by ultraviolet irradiation or n-pentane extraction. However, the addition of the ubiquinone fraction to the n-pentane-extracted membranes did not restore enzyme activity. These observations suggested the reducing equivalents from succinate entered the respiratory chain on the oxygen side of the HQNO-sensitive site and probably did not proceed through a quinone.

    Beschreibung: The present study was motivated by an interest in seeing whether hair cell types in the bullfrog utriculus might differ in their voltage responses to hair bundle displacement. Particular interest was in assessing the contributions of two factors to the responses of utricular hair cells. First, interest in examining the effect of hair bundle morphology on the sensitivity of hair cells to natural stimulation was motivated by the observation that vestibular hair cells, unlike many auditory hair cells, are not free-standing but rather linked to an accessory cupular or otolithic membrane via the tip of their kinocilium. Interest also laid in examining the contribution, if any, of adaptation to the response properties of utricular hair cells. Hair cells in auditory and vibratory inner ear endorgans adapt to maintained displacements of their hair bundles, sharply limiting their low frequency sensitivity. This adaptation is mediated by a shift in the displacement-response curve (DRC) of the hair cell along the displacement axis. Observations suggest that the adaptation process occurs within the hair bundle and precedes mechanoelectric transduction. Recent observations of time-dependent changes in hair bundle stiffness are consistent with this conclusion. Adaptation would be expected to be most useful in inner ear endorgans in which hair cells are subject to large static displacements that could potentially saturate their instantaneous response and compromise their sensitivity to high frequency stimulation. The adaptation process also permits hair cells to maintain their sensory hair bundle in the most sensitive portion of their DRC. In vestibular otolith organs in which static sensitivity is desirable, any adaptation process in the hair cells may be undesirable. The rate and extent of the decline of the voltage responses was measured of utricular hair cells to step and sinusoidal hair bundle displacements. Then for similar resting potentials and response amplitudes, the voltage responses of individual hair cells were compared to both hair bundle displacement and intracellular current.

    Beschreibung: Hair cells in the bullfrog sacculus are specifically adapted to sense small-amplitude, high-frequency linear accelerations. These hair cells display many properties that are undesirable or inappropriate for hair cells that must provide static gravity sensitivity. This study resulted in part due to an interest in seeing how the transduction mechanisms of hair cells in a gravity-sensing otolith endorgan would differ from those in the bullfrog sacculus. The bullfrog utriculus is an appropriate model for these studies, because its structure is representative of higher vertebrates in general and its function as a sensor of static gravity and dynamic linear acceleration is well known. Hair cells in the bullfrog utriculus, classifiable as Type 2 by cell body and synapse morphology, differ markedly in hair bundle morphology from those in the bullfrog sacculus. Moreover, the hair bundle morphologies of utricular hair cells, unlike those in the sacculus, differ in different membrane regions.

    Beschreibung: Vestibular nerve afferents innervating the bullfrog utriculus differ in their response dynamics and sensitivity to natural stimulation. They also supply hair cells that differ markedly in hair bundle morphology. To examine the peripheral innervation patterns of individual utricular afferents more closely, afferent fibers were labeled by the extracellular injection of horseradish peroxidase (HRP) into the vestibular nerve after sectioning the vestibular nerve medial to Scarpa's ganglion to allow the degeneration of sympathetic and efferent fibers. The peripheral arborizations of individual afferents were then correlated with the diameters of their parent axons, the regions of the macula they innervate, and the number and type of hair cells they supply. The utriculus is divided by the striola, a narrow zone of distinctive morphology, into media and lateral parts. Utiricular afferents were classified as striolar or extrastriolar according to the epithelial entrance of their parent axons and the location of their terminal fields. In general, striolar afferents had thicker parent axons, fewer subepithelial bifurcations, larger terminal fields, and more synaptic endings than afferents in extrstriolar regions. Afferents in a juxtastriolar zone, immediately adjacent to the medial striola, had innervation patterns transitional between those in the striola and more peripheral parts of the medial extrastriola. moast afferents innervated only a single macular zone. The terminal fields of striolar afferents, with the notable exception of a few afferents with thin parent axons, were generally confined to one side of the striola. Hair cells in the bullfrog utriculus have perviously been classified into four types based on hair bundle morphology. Afferents in the extrastriolar and juxtastriolar zones largely or exclusively innervated Type B hair cells, the predominant hair cell type in the utricular macula. Striolar afferents supplied a mixture of four hair cell types, but largely contacted Type B and Type C hair cells, particularly on the outer rows of the medial striola. Afferents supplying more central striolar regions innervated fewer Type B and larger numbers of Type E and Type F hair cells. Striolar afferents with thin parent axons largely supplied Type E hair cells with bulbed kniocilia in the innermost striolar rows.

    Beschreibung: Surface glycoconjugates of hair cells and supporting cells in the vestibular endorgans of the bullfrog were identified using biotinylated lectins with different carbohydrate specificities. Lectin binding in hair cells was consistent with the presence of glucose and mannose (CON A), galactose (RCA-I), N-acetylgalactosamine (VVA), but not fucose (UEA-I) residues. Hair cells in the bullfrog sacculus, unlike those in the utriculus and semicircular canals, did not stain for N-acetylglucosamine (WGA) or N-acetylgalactosamine (VVA). By contrast, WGA and, to a lesser extent, VVA, differentially stained utricular and semicircular canal hair cells, labeling hair cells located in peripheral, but not central, regions. In mammals, WGA uniformly labeled Type 1 hair cells while labeling, as in the bullfrog, Type 2 hair cells only in peripheral regions. These regional variations were retained after enzymatic digestion. We conclude that vestibular hair cells differ in their surface glycoconjugates and that differences in lectin binding patterns can be used to identify hair cell types and to infer the epithelial origin of isolated vestibular hair cells.

    Beschreibung: Adult bullfrogs were given single intraotic injections of the aminoglycoside antibiotic gentamicin sulfate and sacrificed at postinjection times ranging from 0.5 to 9 days. The saccular and utricular maculae of normal and injected animals were examined in wholemount and cross-section. Intraotic 200 (mu) M gentamicin concentrations resulted in the uniform destruction of the hair bundles and, at later times, the cell bodies of saccular hair cells. In the utriculus, striolar hair cells were selectively damaged while extrastriolar hair cells were relatively unaffected. Regenerating hair cells, identified in sectioned material by their small cell bodies and short, well-formed hair bundles, were seen in the saccular and utricular maculae as early as 24-48 h postinjection. Immature versions of mature hair cell types in both otolith organs were recognized by the presence of absence of a bulbed kinocilia and the relative lengths of their kinocilia and longest sterocilia. Utricular hair cell types with kinocilia longer than their longest stereocilia were observed at earlier times than hair cell types with shorter kinocilia. In the same sacculus, the hair bundles of gentamicin-treated animals, even at 9 days postinjection, were significantly smaller than those of normal animals. The hair bundles of utricular hair cells, on the other hand, reached full maturity within the same time period.

    Beschreibung: Among the many problems of growing plants in completely controlled environments, such as those anticipated for the space station and the CELSS program, is the need to provide light that is both adequate for photosynthesis and of proper quality for normal growth and development. NASA scientists and engineers have recently become interested in the possibility of utilizing densely packed, solid state, light emitting diodes (LED's) as a source for this light. Unlike more conventional incandescent or electrical discharge lamps, these sources are highly monochromatic and lack energy in spectral regions thought to be important for normal plant development. In addition, a recent observation by NASA scientist has suggested that infra-red LED's, that are routinely used as photographic safelights for plants grown in darkness, may interact with the ability of plants to detect gravity. In order to establish how plants respond to light from these LED light sources we carried out a series of experiments with known pigment mutants of the model mustard plant, Arabidopsis thaliana, growing in either a gravity field or on a clinostat to simulate a micro-gravity environment. Results indicate that only red light from the 665 nm LED's disrupts the ability of normal wildtype seedlings to detect a gravity stimulus. There was no consistent effect found for the far-red (735 nm) LED's or either of the infrared (880 nm or 935 nm) LED sources but both showed some effect in one or more of the genotypes tested. Of these five members of the phytochrome multigene family in Arabidopsis, only the phytochrome B pigment mutant (hy3) lacked the ability to detect gravity under all conditions. There was no effect of either micro-gravity (clinostat) or the infra-red LED's on the light induced inhibition of hypocotyl elongation. Measurements of the pigment phytochrome in oats also showed no photoconversion by 15 min irradiations with the infra-red LED's. We conclude that phytochrome B is required for the perception of gravity and that only red light is able to disrupt this perception. The infra-red LED's also do not appear to interact with gravity perception in Arabidopsis, but caution should be exercised if infra-red LED's are to be used as photographic safelights for these types of experiments.

    Beschreibung: Light scattering experiments performed at Mississippi State University using MSFC ovostatin preparations indicated that at low ovostatin concentrations, below 0.2 mg/ml, the protein was dissociating from a tetramer into dimers. Since the proposed mechanism of action involved the tetrameric form of the protein, we hypothesized that perhaps under the conditions of our assays at various O/T ratios the ovostatin was becoming dissociated into an inactive dimer. To examine this possibility we assayed the ovostatin activity as a function of ovostatin concentration and of temperature of the assay. Data are presented that show the results of these assays at 23 C, 30 C, 37 C and 42 C respectively. The data are highly suggestive that there is a decrease in ovostatin activity as the concentration of the protein falls below 0.06 mg/ml. This may not be of any physiological importance, however, since the concentration of ovostatin in the egg is about 0.5 mg/ml. Curiously, the dissociation of the tetramer into dimers does not show a significant temperature dependence as would be expected for an equilibrium reaction. Whether this is in fact the case, or whether the differences are so small as to not be discerned from the current data remains to be seen. Another aspect to consider is that in the egg the primary role of the ovostatin may or may not be as a protease inhibitor. Although the inhibition of collagenase by ovostatin may be an important aspect of embryogenesis, it is also possible that it functions as a binding protein for some substance. In this regard, all ovostatin preparations from MSFC have shown an approximately 88,000 MW protein associated with the ovostatin. The identity of this protein is not currently known and may be the subject of future studies.

    Beschreibung: Attempts to use artificially lit cabinets to grow plants identical to those growing in sunlight have provided compelling evidence of the importance of light quality for plant growth. Changing the balance of red (R) to far-red (FR) radiation, but with a fixed photosynthetic input can shift the phytochrome photoequilibrium in a plant and generate large differences in plant growth. With FR enrichment the plants elongate, and may produce more leaf area and dry matter. Similar morphogenic responses are also obtained when light quality is altered only briefly (15-30 min) at the end-of-the-day. Conversely, for plants grown in natural conditions the response of plant form to selective spectral filtering has again shown that red and far-red wavebands are important as found by Kasperbauer and coworkers. Also, where photosynthetic photon flux densities (PPFD) of sunlight have been held constant, the removal of far-red alone alters plant growth. With FR depletion plants grown in sunlight are small, more branched and darker green. Here we examine the implications for plant growth and flowering when the far-red composition of incident radiation in plant growth chambers is manipulated.

    Beschreibung: An important motivation for studying photomorphogenesis is to understand the relationships among plant photophysiology in canopies, canopy productivity, and agronomic yield. This understanding is essential to optimize lighting systems used for plant farming in controlled environments (CE) and for the design of genetically engineered crop strains with altered photoresponses. This article provides an overview of some basic principles of plant photomorphogenesis in canopies and discusses their implications for (1) scaling up information on plant photophysiology from individual plants in CE to whole canopies in the field, and (2) designing lighting conditions to increase plant productivity in CE used for agronomic purposes (e.g. space farming in CE Life Support Systems). We concentrate on the visible (lambda between 400 and 700 nm) and far-infrared (FR; lambda greater than 700 nm) spectral regions, since the ultraviolet (UV; 280 to 400 nm) is covered by other authors in this volume.

    Beschreibung: The widespread application of electric (often called artificial) light in greenhouses, growing rooms, and plant growth chambers would presuppose that the role of phytochrome would be considered in the selection and use of such lighting systems. Unfortunately this is not usually the case. Part of the problem is that many students, and indeed an unfortunate number of senior scientists, seem to regard phytochrome as a laboratory phenomenon without much application in the real world. They simply have not grasped the concept that phytochrome is functioning through all stages of plant development, wherever plants are grown. It is certainly true, as Meijer (1971) stated, that one cannot compare experimental results obtained under very strict laboratory conditions with plant irradiation in glasshouses and in growth rooms. When Karl Norris developed the first practical portable spectroradiometer about 1962, some of the first measurements were to determine the red/far-red ratios under tree canopies. These measurements showed clearly the predominance of far-red in the understory and suggested that far-red was contributing to the elongation exhibited by many species growing in the shade, and possibly was a factor in the induction of light requirements in seeds. Subsequently we used Catalpa leaves as far-red filters to make light-insensitive lettuce seed light requiring. Much more detailed work has since been done on phytochrome effects in the natural environment, and it is encouraging to note that efforts are bring made to apply phytochrome research to horticulture.

    Beschreibung: We have shown that the photosynthetic period is important in regulating carbon partitioning. Even when the same amount of carbon is fixed over a 24h period considerably more is translocated out of the leaf under the longer photosynthetic period. This is extremely important when parts of the plant other than the leaves are to be sold. It is also important to notice the amount of carbon respired in the short photosynthetic period. The light period effect on carbohydrate fixation, dark respiration, and translocation is shown in this report.

    Beschreibung: Light is undoubtedly the most important environmental variable for plant growth and development; plants not only use radiant energy in photosynthesis, they also respond to the quantity, quality, direction and timing of incident radiation through photomorphogenic response that can have huge effects on the rate of growth and the pattern of development. It is surprising, therefore, that the manufacturers and suppliers of controlled environment facilities have been singularly uninventive in the design of the lighting assemblies they provide. The consumer has one choice only - a lighting assembly that provides irradiance levels usually only a fraction of sunlight, and a control system that is limited to regulating the timing of the on-off switch. The reasons for these limitations are partly technological, but in the main they result from ignorance on the part of both the consumer and the manufacturer. A specific and powerful example of this ignorance relates to the importance of the so-called far-red wavelengths (FR = 700-800 nm). Because the human eye can hardly detect wavelengths above 700 nm, and photosynthesis also cuts off at about 700 nm, the majority of plant and crop physiologists are still almost completely unaware that FR radiation can have massive effects on growth rate and development. In consequence, most growth cabinets have light sources based on fluorescent tubes, and provide very little FR apart from that emitted by a token number of small incandescent bulbs. Larger growth facilities often use broader spectrum light sources, but growth facilities that provide the capability to vary the FR incident upon the plants are about as abundant as seals in the Sahara. This article sets the background of the significance of FR radiation in the natural environment and its importance for plant growth and development in the hope that it might inform intelligently those concerned with improving the design of plant growth facilities.

    Beschreibung: The absorption spectrum of a leaf is often thought to contain some clues to the photosynthetic action spectrum of chlorophyll. Of course, absorption of photons is needed for photosynthesis, but the reverse, photosynthesis when there is absorption, is not necessarily true. As a check on the existence of absorption limits we measured spectra for a few different leaves. Two techniques for measuring absorption have been used, viz. the separate determination of the diffuse reflectance and the diffuse transmittance with the leaf at a port of an integrating sphere and the direct determination of the non-absorbed fraction with the leaf in the sphere. In a cross-check both methods yielded the same results for the absorption spectrum. The spectrum of a Fuchsia leaf, covering the short-wave region from 350 to 2500 nm, shows a high absorption in UV, blue and red, the well known dip in the green and a steep fall-off at 700 nm. Absorption drops to virtually zero in the near infrared, with subsequent absorptions, corresponding to the water absorption bands. In more detailed spectra, taken at 5 nm intervals with a 5 nm bandwidth, differences in chlorophyll content show in the different depths of the dip around 550 nm and in a small shift of the absorption edge at 700 nm. Spectra for Geranium (Pelargonium zonale) and Hibiscus (with a higher chlorophyll content) show that the upper limit for photosynthesis can not be much above 700 nm. No evidence, however, is to be seen of a lower limit for photosynthesis and, in fact, some experiments down to 300 nm still did not show a decrease of the absorption although it is well recognized that no photosynthesis results with 300 nm wavelengths.

    Beschreibung: The main conclusions of many investigations about general requirements of plants for spectral composition of PAR (photosynthetically active radiation) are based on phylogenetic aspects of plant growth. We think that these aspects are not the main criteria in choosing the spectral composition required for growing plants in controlled conditions. Our approach to this problem is based on plant and crop reaction under long duration growth with specific spectra and intensity. Only in this way can we determine correctly the role of light characteristics for developing crops.

    Beschreibung: Electroluminescence in solid materials is defined as the generation of light by the passage of an electric current through a body of solid material under an applied electric field. A specific type of electroluminescence, first noted in 1923, involves the generation of photons when electrons are passed through a p-n junction of certain solid materials (junction of a n-type semiconductor, an electron donor, and a p-type semiconductor, an electron acceptor). The development of this light emitting semiconductor technology dates back less than 30 years. During this period of time, the LED has evolved from a rare and expensive light generating device to one of the most widely used electronic components. A number of LED characteristics are of considerable importance in selecting a light source for plant lighting in a controlled environment facility. Of particular importance is the characteristic that light is generated by an LED at a rate far greater than the corresponding thermal radiation predicted by the bulk temperature of the device as defined by Plank's radiation law. This is in sharp contrast to other light sources, such as an incandescent or high intensity discharge lamp. A plant lighting system for controlled environments must provide plants with an adequate flux of photosynthetically active radiation, plus providing photons in the spectral regions that are involved in the photomorphogenic and phototropic responses that result in normal plant growth and development. Use of light sources that emit photons over a broad spectral range generally meet these two lighting requirements. Since the LED's emit over specific spectral regions, they must be carefully selected so that the levels of photsynthetically active and photomorphogenic and phototropic radiation meet these plant requirements.

    Beschreibung: This essay will consider both physical and photobiological aspects of controlled environment lighting in the spectral region beginning in the blue and taken to the normal limit of the solar spectrum in the ultraviolet. The primary emphasis is directed to questions of plant response to sunlight. Measurement and computations used in radiation dosimetry in this part of the spectrum are also briefly treated. Because of interest in the ozone depletion problem, there has been some activity in plant UV-B research and there are several recent reviews available. Some aspects of growth chamber lighting as it relates to UV-B research were covered earlier. Apart from work related to the blue/UV-A receptor, less attention has been given to UV-A responses.

    Beschreibung: The effects of daylength and spectral quality on assimilate partitioning and leaf carbohydrate content should be considered when conducting controlled environment experiments or comparing results between studies obtained under different lighting conditions. Changes in partitioning may indicate alterations to photoregulatory processes within the source leaf rather than disruptions in sink strength. Moreover, it may be possible to use photoregulatory responses of assimilate partitioning to probe mechanisms of growth and development involving translocation of carbon or adaptation to environmental factors such as elevated CO2. It may also be possible to steer assimilate partitioning for the benefit of controlled environment agriculture using energy-efficient manipulations such as daylength extensions with dim irradiances, end-of-day alterations in light quality, or shifting plants between different spectral qualities as a part of phasic control of growth and development. Note that high starch levels measured on a one-time basis provide little information, since it is the proportion of photosynthate stored as starch that is meaningful. Large differences in starch content can result from small changes in partitioning integrated over several days. Rate information is required.

    Beschreibung: Both plants and animals have one general commonality in their perception of light. They both are sensitive primarily to the 400 to 700 nm wavelength portion of the electromagnetic spectrum. This is referred to as the visible spectrum for animals and as the photosynthetically active radiation (PAR) spectrum for plants. Within this portion of the spectrum, animals perceive colors. Relatively recently it has been learned that within this same spectral range plants also demonstrate varying responses at different wavelengths, somewhat analogous to the definition of various colors at specific wavelengths. Although invisible to the human eye, portions of the electromagnetic spectrum on either side of the visible range are relatively inactive photosynthetically but have been found to influence important biological functions. These portions include the ultraviolet (UV approximately equal to 280-400 nm) and the far-red (FR approximately equal to 700-800 nm). The basic photoreceptor of plants for photosynthesis is chlorophyll. It serves to capture radiant energy which combined with carbon dioxide and water produces oxygen and assimulated carbon, used for the synthesis of cell wall polysaccarides, proteins, membrane lipids and other cellular constituents. The energy and carbon building blocks of photosynthesis sustain growth of plants. On the other hand, however, there are other photoreceptors, or pigments, that function as signal transducers to provide information that controls many physiological and morphological responses of how a plant grows. Known photomorphogenic receptors include phytochrome (the red/far-red sensor in the narrow bands of 655-665 nm and 725-735 nm ranges, respectively) and 'cryptochrome' (the hypothetical UV-B sensor in the 280-320 nm range). Since the USDA team of W. L. Butler, S. B. Hendricks, H. A. Borthwick, H. A. Siegleman and K. Norris in Beltsville, MD detected by spectroscopy, extracted and identified phytochrome as a protein in the 1950's, many other investigators have found evidence of its control functions in plants. Considerably less, however, is known about the yet non-isolated cryptochrome. The information-transferring roles of photoreceptors in plants at specific spectral ranges quite naturally stimulated plant scientists and engineers to consider physically manipulating light to achieve desired physiological and morphological characteristics. One way to manipulate light is to filter it through materials that selectively transmit portions of the sun's spectrum in and near the PAR range.

    Beschreibung: At the Odessa Phytotron we have installed over 50 climatic chambers and cabinets made by various companies of the United States, Canada, Germany and U.S.S.R. They employ different light sources including Sylvania fluorescent lamps of various types, fluorescent lamps produced in the former Soviet Union with a special luminophore, ordinary tungsten lamps, xenon, mercury, mercury-iodide, sodium, etc. Our objective in lighting is that the intensity distribution over the wave lengths should be maximal in the photosynthetically active part of the spectrum and minimal in the IR part to avoid plant sterilization. Phytotrons are extremely energy consuming entities, and the large part of their energy consumption falls into the lighting category in our electric bills. Therefore, we are in a constant search of the processes to reduce energy. However, the main way to increase effectiveness would be the development of new types of light sources, which would come close to the threshold of 150 to 200 lumens per watt.

    Beschreibung: Xenon lamps are available as low and high power lamps with relatively high efficiency and a relatively long lifetime up to several thousand hours. Different construction types of short-arc and long-arc lamps permit a good adaptation to various applications in projection and illumination techniques without substantial changes of the spectral quality. Hence, the xenon lamp was the best choice for professional technical purposes where high power at simultaneously good spectral quality of the light was required. However, technical development does not stand still. Between the luminous efficacy of xenon lamps of 25-50 lm/W and the theoretical limit for 'white light' of 250 lm/W is still much room for improvement. The present development mainly favors other lamp types, like metal halide lamps and fluorescent lamps for commercial lighting purposes. The enclosed sections deal with some of the properties of xenon lamps relevant to plant illumination; particularly the spectral aspects, the temporal characteristics of the emission, and finally the economy of xenon lamps will be addressed. Due to radiation exceeding the natural global radiation in both the ultraviolet (UV) and the infrared (IR) regions, filter techniques have to be included into the discussion referring to the requirements of plant illumination. Most of the presented results were obtained by investigations in the GSF phytotron or in the closed Phytocell chambers of the University of Erlangen. As our experiences are restricted to area plant illumination rather than spot lights our discussion will concentrate on low pressure long-arc xenon lamps which are commonly used for such plant illuminations. As the spectral properties of short-arc lamps do not differ much from those of long-arc lamps most of our conclusions will be valid for high pressure xenon lamps too. These lamps often serve as light sources for small sun simulators and for monochromators which are used for action spectroscopy of plant responses.

    Beschreibung: Artificial lighting is very important for experimental purposes, as well as for the practical use of plants when not enough sunlight is available. To grow green higher plants in their normal forms under artificial lighting constructing efficient and economically reasonable lighting systems is not an easy task. One possible approach would be to simulate sunlight in intensity and the radiation spectrum, but its high construction and running costs are not likely to allow its use in practice. Sunlight may be excessive in irradiance in some or all portions of the spectrum. Reducing irradiance and removing unnecessary wavebands might lead to an economically feasible light source. However, removing or reducing a particular waveband from sunlight for testing is not easy. Another approach might be to find the wavebands required for respective aspects of plant growth and to combine them in a proper ratio and intensity. The latter approach seems more practical and economical, and the aim of this Workshop lies in advancing this approach. I summarize our present knowledge on the waveband requirements of higher plants for the regions of blue, UV-A and UV-B.

    Beschreibung: All life on earth depends on light. A variety of photoreceptors capture the light for a wide range of reactions. Photosynthetic organisms absorb the light necessary for energy transformation and charge separation facilitating photosynthesis. In addition to the bulk pigments there is a great diversity of photoreceptors present in minute concentrations that control development, metabolism and orientation of plants and microorganisms. Based on its spectral absorbance, the well-studied phytochrome system acts in the RL (red light) region as well as in the UV-A/BL (blue light) region where the above mentioned reactions are mediated by a variety of photoreceptors whose natures are largely unknown. Phyllogenetically the UV-A/BL photoreceptors seem to be more ancient pigments that eventually were replaced by the phytochrome system. However, there are many reports that suggest a coaction between the UV-A/BL receptors and the phytochrome system. In several cases the UV-A/BL activation is the prerequisite for the phytochrome reaction. Historically it was the German botanist Julius Sachs who first discovered in 1864 that phototropism in plants was due to BL reactions. It took over 70 years until Bunning (1937) and Galston and Baker (1949) rediscovered the BL response. Since then, an ever-increasing attention has been paid to this effect. In this contribution, the general aspect of UV-A/BL responses and especially the responsiveness of algae will be covered.

    Beschreibung: This paper presents data that suggests some criteria for evaluating growth chamber and greenhouse lighting. A review of the general lighting requirements for photosynthesis reveals that four aspects of light are important: irradiance, quality, timing, and duration. Effective lighting should produce plants that perform according to the goals of the project. For example, for physiological studies the plants probably should exhibit morphology and physiology similar to that found in field-grown plants. For other projects the criteria will obviously be set according to the reason for raising the plants.

    Beschreibung: An increase in the demand for and production of vegetables in the winter, mainly in northern and Siberian regions, inevitably leads to mass building of structures for growing plants under completely artificial conditions. An industrial lighting technology is required whose main parameters (spectrum, irradiance, photoperiod) should be assigned carefully and should uniquely determine, along with other important characteristics of the artificial climate, the productivity of the plant-production facility. The most widespread crops grown in our country under indoor conditions are cucumber and tomato plants, which account for more than 98% of the area in greenhouses. These plants are good prospects for growing completely under intense artificial lighting conditions (photocultures). Optimization of the main parameters of optical radiation when growing these plants is the most important task of achieving their profitable production. At present, considerable experience has been gained in studying the dependence of productivity of cucumber and tomato communities on irradiation conditions. Fundamental studies of the Agrophysical Research Institute of the Russian Academy of Sciences, Timiryazev Institute of Plant Physiology of the Russian Academy of Sciences, Timiryazev Agricultural Academy, and other institutes create a good basis for a detailed study of the given problem. Commercial sources of radiation substantially differing in spectral characteristics in the region of photosynthetically active radiation (PAR) were used in the studies.

    Beschreibung: The Generic Bioprocessing Apparatus (BPA) payload was developed by BioServe to support the commercial flight development needs of our specialized consortia comprised of business, academic, and government entities. The consortia have commitments to explore commercial opportunities in bioprocessing, biomedical models, and closed agricultural systems. In addition, some members of BioServe have interests in the development and/or qualification of enabling flight hardware used in life sciences space flight testing. Some business and academic entities have interests in more than one of these consortia. To aid in payload development, flight, and analysis, each consortium member contributes resources ranging from proprietary expertise and materials, to hardware and cash. Professionals from business, academia, and government often interact with each other via graduate research assistants who do much of the 'hands-on' payload preparation and subsequent data analyses. The GBA supported research, testing, and development activities for each different BioServe consortium. It produced an environment in which professionals from diverse backgrounds came together with a single focus. And, it provided a truly novel learning environment for a youthful new cadre of space professionals committed to the exploration of commercial opportunities presented by space. Since the GBA supported a large number of different experiments, this paper briefly describes the payload characteristics and the essential operations of the payload. A summary of the experiments is presented. Finally, a few of the experiments are described in detail highlighting some novel effects of space flight on life science systems. Portions of the reported work have or will appear in appropriate archival journals as cited in the bibliography. In several instances, data collected from USML-1 have been supplemented with related data collected on more recent STS missions.

    Beschreibung: The mission of the Life Science Division at the NASA Ames Research Center is to investigate the effects of gravity on living systems in the spectrum from cells to humans. The range of these investigations is from microgravity, as experienced in space, to Earth's gravity, and hypergravity. Exposure to microgravity causes many physiological changes in humans and other mammals including a headward shift of body fluids, atrophy of muscles - especially the large muscles of the legs - and changes in bone and mineral metabolism. The high cost and limited opportunity for research experiments in space create a need to perform ground based simulation experiments on Earth. Models that simulate microgravity are used to help identify and quantify these changes, to investigate the mechanisms causing these changes and, in some cases, to develop countermeasures.

    Beschreibung: The Life Science Division at NASA's Ames Research Center has a suite of specialized facilities that enable scientists to study the effects of gravity on living systems. This paper describes some of these facilities and their use in research. Seven centrifuges, each with its own unique abilities, allow testing of a variety of parameters on test subjects ranging from single cells through hardware to humans. The Vestibular Research Facility allows the study of both centrifugation and linear acceleration on animals and humans. The Biocomputation Center uses computers for 3D reconstruction of physiological systems, and interactive research tools for virtual reality modeling. Psycophysiological, cardiovascular, exercise physiology, and biomechanical studies are conducted in the 12 bed Human Research Facility and samples are analyzed in the certified Central Clinical Laboratory and other laboratories at Ames. Human bedrest, water immersion and lower body negative pressure equipment are also available to study physiological changes associated with weightlessness. These and other weightlessness models are used in specialized laboratories for the study of basic physiological mechanisms, metabolism and cell biology. Visual-motor performance, perception, and adaptation are studied using ground-based models as well as short term weightlessness experiments (parabolic flights). The unique combination of Life Science research facilities, laboratories, and equipment at Ames Research Center are described in detail in relation to their research contributions.

    Beschreibung: One of the most difficult obstacles to establishing a causal connection between mass extinctions and large body impacts is the existence of what appear to be many more KT survivor species than previously suspected. Though interpretations of 'Cretaceous' faunal elements in lowermost Danian sediments differ, this enigmatic fauna has not been recovered from every biozone-complete boundary section, including the El Kef stratotype. In terms of their potential for providing constraints on scenarios seeking to account for the KT extinction event, the significance of such observations cannot be overstated. Owing to the consistency with which these observations have been made over the last several years, the possibility of widespread trans-KT biotic survivorship can no longer be dismissed. Rather, the survivorship hypothesis must be tested alongside its alternative (the reworking hypothesis) to determine which explains the available data in the most complete yet parsimonious manner. Moreover, valid tests for survivorship cannot be based on negative evidence or on the assumption that only a small cohort of species could have survived the KT boundary event. Several authors have recently proposed various criteria that might be used to test alternative interpretations for this aspect lowermost Danian biotic record.

    Beschreibung: The energy transducing ATPase from Halobacterium saccharovorum was studied in order to define the origin of energy transducing systems. The ATPase required high salt concentration (4M NaCl) for activity; activity was rapidly lost when NaCl was below 1 Molar. At low salt concentration, the membrane bound ATPase activity could be stabilized in presence of spermine. However, following solubilization spermine was ineffective. Furthermore, F1 ATPase activity was stabilized by ammonium sulfate even when the NaCl concentration was less than 1 Molar. These studies suggest that stabilization by hydrophobic interactions preceded ionic ones in the evolution of the energy transducing ATPases.

    Beschreibung: The peripheral (tail) circulatory responses of six male albino rats were measured at ambient temperatures between 5 and 40 C, using impedance plethysmography. Each animal was anesthetized, instrumented, and placed in a thermal environmental chamber to reach equilibrium. Tail blood flow, respiration rate, heart rate, core temperature, and tail skin temperature were then monitored at each ambient temperature. The mean tail blood flow was significantly higher (p less than 0.05) at 5 C than at 10 C. The mean blood flow increased significantly (p less than 0.01) at each of the temperatures above 10 C. Tail skin temperature and internal body (core) temperature increased significantly with increasing ambient temperature.

    Beschreibung: Intracerebroventricular administration of adrenocorticotropin (ACTH 1-24) and alpha-melanotropin (alpha-MSH), peptides which occur naturally in brain induced dose-related hypothermia in guinea-pigs at room temperature (21 C) and also produced greater hypothermia at low (10 C) ambient temperature. However, when the experiments were repeated in a warm (30 C) environment, no effect on body temperature was observed. These results indicate that the peptides did not reduce the central set-point of temperature control. The hypothermia induced by ACTH and alpha-MSH was not mediated via histamine H1- or H2-receptors and serotonin since the H1-receptor antagonist, mepyramine, the H2-receptor antagonist, cimetidine, and the serotonin antagonist, methysergide, had no antagonistic effects. The peptides were antipyretic since they reduced pyrogen-induced-fever and hyperthermia due to prostaglandin E2, norepinephrine and dibutyryl cAMP, at a dose which did not affect normal body temperature. The powerful central effects of these peptides on normal body temperature, fever and hyperthermia, together with their presence of the brain regions important to temperature control, suggest that they participate in thermoregulation.

    Beschreibung: Smith (1981) has presented three alternative models of the origin of eukaryotes. In the present investigation, alternative theories are discussed along with the status of serial endosymbiotic theory. It is pointed out that the usefulness of the serial endosymbiotic theory is attested by the great flourishing of productive scientific studies it has generated. Attention is given to the origin of mitochondria and plastids from free-living bacteria, the polyphyly of mitochondria, the origin of the nuclear membrane, the origin of undulipodia (cilia and other 9 + 2 organelles), the sequence of acquisition of organelles, large ribosomes, genome organization, animals and plants, and systematics and axonomy of the microorganisms.

    Beschreibung: In a recent study of the effects of hypergravity (HG) on the reproductive system of the rat, it was found that the estrous cycle is perturbed in a pseudopregnancy-like pattern upon first exposure of animals to HG. This can be prevented by previous exposure to HG or by administration of bromergo cryptin. Adapted rats can mate and deliver in HG. However, little is known of the condition of the individual pregnant rat, fetuses, and newborn pups in this environment. There are also questions regarding the effects of HG on the number of fetuses, the mortality rate in different HG levels, and the effects on production and secretion of PRL and TSH. The present investigation is concerned with such questions, taking into account possible approaches to avoid or minimize the lethal effects of HG. The obtained results suggest that peripartum plasma PRL levels and thyroid hormone levels are critical for the survival of the litter.

    Beschreibung: Paracoccus halodenitrificans produced a membrane-associated nitrite reductase. Spectrophotometric analysis showed it to be associated with a cd-cytochrome and located on the inner side of the cytoplasmic membrane. When supplied with nitrite, membrane preparations produced nitrous oxide and nitric oxide in different ratios depending on the electron donor employed. The nitrite reductase was maximally active at relatively low concentrations of sodium chloride and remained attached to the membranes at 100 mM sodium chloride.

    Beschreibung: Steady-state cultures of Paracoccus halodenitrificans were grown anaerobically prior to establishing steady states at different concentrations of oxygen. In the absence of oxygen, nitrate-limited cultures produced dinitrogen, and as the oxygen supply increased, these cultures produced nitrous oxide, then nitrite. These changes reflected two phenomena: the inactivation of nitrous oxide reductase by oxygen and the diversion of electrons from nitrite to oxygen.

    Beschreibung: A diversification of the genetic code based on the number of codons available for the proteinous amino acids is established. Three groups of amino acids during evolution of the code are distinguished. On the basis of their chemical complexity and a small codon number those amino acids emerging later in a translation process are derived. Both criteria indicate that His, Phe, Tyr, Cys and either Lys or Asn were introduced in the second stage, whereas the number of codons alone gives evidence that Trp and Met were introduced in the third stage. The amino acids of stage one use purines rich codons, thus purines have been retained in their third codon position. All the amino acids introduced in the second stage, in contrast, use pyrimidines in this codon position. A low abundance of pyrimidines during early translation is derived. This assumption is supported by experiments on non enzymatic replication and interactions of DNA hairpin loops with a complementary strand. A back extrapolation concludes a high purine content of the first nucleic acids which gradually decreased during their evolution. Amino acids independently available form prebiotic synthesis were thus correlated to purine rich codons. Conclusions on prebiotic replication are discussed also in the light of recent codon usage data.

    Beschreibung: We have demonstrated that etiolated soybean seedlings grown under the altered gravity conditions of clinorotation (1 rpm) and centrifugation (5xg) exhibit changes in starch metabolism. Cotyledon starch concentration was lower (-28%) in clinorotated plants and higher (+24%) in centrifuged plants than in vertical control plants. The activity of ADP-glucose pyrophosphorylase in the cotyledons was affected in a similar way, i.e. lower (-37%) in the clinorotated plants and higher (+22%) in the centrifuged plants. Other starch metabolic enzyme activities, starch synthase, starch phosphorylase and total hydrolase were not affected by the altered gravity treatments. We conclude that the observed changes in starch concentrations were primarily due to gravity-mediated differences in ADP-glucose pyrophosphorylase activity.

    Beschreibung: Understanding the physical chemical processes which affect the structure and composition of astrophysical ices is of central importance to our understanding of the early solar system and early earth. Laboratory experiments on amorphous ice analogues of cometary ices have shown that amorphous ices exhibit a rich and complex morphology. Computer simulation results for ice deposition with the solutes Ne, CH4, and CO are presented. The relative fraction of adsorbed solutes which are subsequently ejected from the growing ice has implications on the composition of comets relative to the composition of the primitive environment in which they formed.

    Beschreibung: Cometary missions are of enormous fundamental importance for many different space science disciplines, including exobiology. Comets are presumed relics of the earliest, most primitive material in the solar nebula and are related to the planetesimals. They undoubtedly provided a general enrichment of volatiles to the inner solar system (contributing to atmospheres and oceans) and may have been key to the origin of life. A Discovery class, comet rendezvous mission, the Cometary Coma Chemical Composition (C4) Mission, was selected for further study by NASA earlier this year. The C4 Mission is a highly focused and usefully-limited subset of the Cometary Rendezvous Asteroid Flyby (CRAF) Mission, concentrating exclusively on measurements which will lead to an understanding of the chemical composition and make-up of the cometary nucleus. The scientific goals of the Cometary Coma Chemical Composition (C4) Mission are to rendezvous with a short-period comet and (1) to determine the elemental, chemical, and isotopic composition of the nucleus and (2) to characterize the chemical and isotopic nature of its atmosphere. Further, it is a goal to obtain preliminary data on the development of the coma (dust and gas composition) as a function of time and orbital position.

    Beschreibung: Amino acids in the Murchison carbonaceous chondrite are anomalously enriched in deuterium. Synthesis in Strecker reactions from D-enriched interstellar precursors during low temperature aqueous alteration of the parent body has been proposed by Cronin et al. (1988) to account for the isotopic observations. To test this hypothesis, we have measured the retention of deuterium in the glycine, alanine, and alpha-amino isobutyric acid produced, respectively, by reactions of formaldehyde-D2, acetaldehyde-D4, and acetone-D6 with HCN and NH3 in water.

    Beschreibung: Theories for the origin of life require the availability of reduced nitrogen. In the non-reducing atmosphere suggested by geochemical evidence, production in the atmosphere and survival of NH3 against photochemical destruction are problematic. Electric discharges and impact shocks would produce NO rather than HCN or NH3. Conversion of NO to nitrous and nitric acid (by way of HNO) and precipitation in acid rain would provide a source of fixed nitrogen to the early ocean. One solution to the NH3 problem may have been the reduction of nitrite/nitrate in the ocean with aqueous ferrous iron, Fe(2+): 6Fe(+2) + 7 H2O + NO2(-) yields 3Fe2O3 + 11 H(+) + NH3. We have measured the kinetics of this reaction as a function of temperature, pH, and concentrations of salts, Fe(+2), and NO2(-). Cations (Na(+), Mg(2+), K(+)) and anions (Cl(-), Br(-), SO4(2-)) increase the rate by factors of 4 to 8. Although a competing pathway yields N2, the efficiency of the conversion of nitrite to ammonia ranges from 25% to 85%. Nitrate reduction was not consistently reproducible; however, when it was observed, its rate was slower by at least 8X than that of nitrite reduction. If the prebiotic atmosphere contained 0.2 to 10 atmospheres CO2 as suggested by Walker (1985), the Fe(+2) concentration and the rate would have been limited by siderite (FeCO3) solubility.

    Beschreibung: The growth and development of more than 500 Zea mays seeds flown on Long Duration Exposure Facility (LDEF) were studied. Somatic mutations, including white-yellow stripes on leaves, dwarfing, change of leaf sheath color or seedling color were observed in plants developed from these seeds. When the frequency of white-yellow formation was used as the endpoint and compared with data from ground based studies, the dose to which maize seeds might be exposed during the flight was estimated to be equivalent to 635 cGy of gamma rays. Seeds from one particular holder gave a high mutation frequency and a wide mutation spectrum. White-yellow stripes on leaves were also found in some of the inbred progenies from plants displayed somatic mutation. Electron microscopy studies showed that the damage of chloroplast development in the white-yellow stripe on leaves was similar between seeds flown on LDEF and that irradiated by accelerated heavy ions on ground.

    Beschreibung: The biological effects of high Linear Energy Transfer (LET) charged particles are a subject of great concern with regard to the prediction of radiation risk in space. In this report, mutagenic effects of high LET charged particles are quantitatively measured using primary cultures of human skin fibroblasts, and the spectrum of induced mutations are analyzed. The LET of the charged particles ranged from 25 KeV/micrometer to 975 KeV/micrometer with particle energy (on the cells) between 94-603 MeV/u. The X-chromosome linked hypoxanthine guanine phosphoribosyl transferase (hprt) locus was used as the target gene. Exposure to these high LET charged particles resulted in exponential survival curves; whereas, mutation induction was fitted by a linear model. The Relative Biological Effect (RBE) for cell-killing ranged from 3.73 to 1.25, while that for mutant induction ranged from 5.74 to 0.48. Maximum RBE values were obtained at the LET of 150 keV/micrometer. The inactivation cross-section (alpha i) and the action cross-section for mutant induction (alpha m) ranged from 2.2 to 92.0 sq micrometer and 0.09 to 5.56 x 10(exp -3) sq micrometer respectively. The maximum values were obtained by Fe-56 with an LET of 200 keV/micrometer. The mutagenicity (alpha m/alpha i) ranged from 2.05 to 7.99 x 10(exp -5) with the maximum value at 150 keV/micrometer. Furthermore, molecular analysis of mutants induced by charged particles indicates that higher LET beams are more likely to cause larger deletions in the hprt locus.

    Beschreibung: Regenerative Life Support Systems (RLSS) will be required to regenerate air, water, and wastes, and to produce food for human consumption during long-duration missions to the Moon and Mars. It may be possible to supplement some of the materials needed for a lunar RLSS from resources on the Moon. Natural materials at the lunar surface may be used for a variety of lunar RLSS needs, including (1) soils or solid-support substrates for plant growth, (2) sources for extraction of essential, plant-growth nutrients, (3) substrates for microbial populations in the degradation of wastes, (4) sources of O2 and H2, which may be used to manufacture water, (5) feed stock materials for the synthesis of useful minerals (e.g., molecular sieves), and (6) shielding materials surrounding the outpost structure to protect humans, plants, and microorganisms from harmful radiation. Use of indigenous lunar regolith as a terrestrial-like soil for plant growth could offer a solid support substrate, buffering capacity, nutrient source/storage/retention capabilities, and should be relatively easy to maintain. The lunar regolith could, with a suitable microbial population, play a role in waste renovation; much like terrestrial waste application directly on soils. Issues associated with potentially toxic elements, pH, nutrient availability, air and fluid movement parameters, and cation exchange capacity of lunar regolith need to be addressed before lunar materials can be used effectively as soils for plant growth.

    Beschreibung: Genetic and developmental effects of heavy ions in maize and rice were investigated. Heavy particles with various charges and energies were accelerated at the BEVALAC. The frequency of occurrence of white-yellow stripes on leaves of plants developed from irradiated maize seeds increased linearly with dose, and high Linear Energy Transfer (LET) heavy charged particles, e.g., neon, argon, and iron, were 2-12 times as effective as gamma rays in inducing this type of mutation. The effectiveness of high-LET heavy ion in (1) inhibiting rice seedling growth, (2) reducing plant fertility, (3) inducing chromosome aberration and micronuclei in root tip cells and pollen mother cells of the first generation plants developed from exposed seeds, and (4) inducing mutation in the second generation, were greater than that of low-LET gamma rays. All effects observed were dose-dependent; however, there appeared to be an optimal range of doses for inducing certain types of mutation, for example, for argon ions (400 MeV/u) at 90-100 Gy, several valuable mutant lines with favorable characters, such as semidwarf, early maturity and high yield ability, were obtained. Experimental results suggest that the potential application of heavy ions in crop improvement is promising. Restriction-fragment-length-polymorphism (RFLP) analysis of two semidwarf mutants induced by argon particles revealed that large DNA alterations might be involved in these mutants.

    Beschreibung: We have flown two new charged particle detectors in five recent Shuttle flights. In this paper we report on the dose rate, equivalent dose rate, and radiation quality factor for trapped protons and cosmic radiation separately. A comparison of the integral Linear Energy Transfer (LET) spectra with recent transport code calculations show significant disagreement. Using the calculated dose rate from the omni-directional AP8MAX model with IGRF reference magnetic field epoch 1970, and observed dose rate as a function of (averaged over all geographic latitude) and longitude, we have determined the westward drift of the South Atlantic anomaly. We have also studied the east-west effect, and observed a 'second' radiation belt. A comparison of the galactic cosmic radiation lineal energy transfer spectra with model calculations shows disagreement comparable to those of the trapped protons.

    Beschreibung: This overview covers the basic microbial ecology of recirculating hydroponic solutions. Examples from NASA and Soviet Controlled Ecological Life Support Systems (CELSS) tests and the commercial hydroponic industry will be used. The sources of microorganisms in nutrient solutions include air, water, seeds, plant containers and plumbing, biological vectors, and personnel. Microbial fates include growth, death, and emigration. Important microbial habitats within nutrient delivery systems are root surfaces, hardware surfaces (biofilms), and solution suspension. Numbers of bacteria on root surfaces usually exceed those from the other habitats by several orders of magnitude. Gram negative bacteria dominate the microflora with fungal counts usually much lower. Trends typically show a decrease in counts with increasing time unless stressed plants increase root exudates. Important microbial activities include carbon mineralization and nitrogen transformations. Important detrimental interactions include competition with plants, and human and plant pathogenesis.

    Beschreibung: The structure and functions of the earliest ancestors of contemporary cells are focal points in studies of the origin of life. Probably the first cell-like structures were vesicles - closed, spheroidal structures with aqueous medium trapped inside. The membranous walls of vesicles were most likely bilayers composed of simple amphiphilic material available on early earth. The membrane studied was composed of glycerol 1-monooleate (GMO). Glycerol forms the polar head group and the oily tail contains 18 carbon atoms. All head groups have been found to be located in two narrow regions at the interfaces with water. The membrane interior, formed by the hydrophobic tails, is quite fluid with chain disorder increasing towards the center of the bilayer. These results are in agreement with x-ray and neutron scattering data from related bilayers. The width of the membrane is not constant, but fluctuates in time and space. Occasional thinning defects in the membrane, observed during the course of the simulations, may have a significant influence on rates of passive transport of small molecules across membranes. It has been found that water penetrates the head group region but not the oily interior of the membrane. Water molecules near the interface are oriented by dipoles of the head groups. The resulting electrostatic potential across the interface, determined in our simulations, has been found to be markedly larger than across the water-oil interface. This quantity has been implicated as the source of selectivity, with respect to the sign of the charge, as an ion approaches the interface and during transport of hydrophobic ions across membranes.

    Beschreibung: In connection with a model of a primitive translation based entirely on nucleic acids the structural requirements and interactions of small nucleic acids were studied. As shown previously hairpins bind to a complementary single-strand by base pairs of their loop nucleotides if the loop contains at least five nucleotides (Baumann et al 1987). Here, the question is approached to which degree a single-strand, the assembler-strand, is occupied with hairpin molecules. A gapless occupation would allow the close proximity necessary for the assumed peptide bond formation by hairpins bearing an amino acid at their 3'-end. Oligo(dC)(sub n), n = 12, 15, 18, and the investigated hairpins form complexes which are detectable under nondenaturing electrophoretic conditions. The size range is estimated to be 3-4 hairpin molecules to one assembler molecule when oligo(dC)(sub n), n = 15, 18, is used. Due to the limitation of the method, dependence of the migration velocity on the shape which is unique and thus limits the use of marker nucleic acids, and due to the uncertainty if the ends of the assembler form base pairs, a gapless occupation of the assembler is conceivable but not proven.

    Beschreibung: Thermodynamic and kinetic information available on the synthesis of prebiotic monomers and polymers will be examined in order to illuminate the prebiotic plausibility of polymer syntheses based on (a) phosphoryl transfer that yields phosphodiester polymers, (b) acyl transfer that gives polyamides, and (c) electron transfer that produces polydisulfide or poly(thio)ester polymers. New experimental results on the oxidative polymerization of 2,3-dimercaptopropanol by ferric ions on the surface of ferric hydroxide oxide will be discussed as a chemical model of polymerization by electron transfer. This redox polymerization that yields polymers with a polydisulfide backbone was found to give oligomers up to the 15-mer from 1 mM of 2,3-dimercaptopropanol after one day at 25 C. High pressure liquid chromatography (HPLC) analysis of the oligomers was carried out on an Alltech OH-100 column eluted with acetonitrile-water.

    Beschreibung: Aquatic microcrustaceans of the genus Daphnia are known to orient to light during the day. At night, in the absence of visual cues, daphnids were suspected of maintaining equilibrium by monitoring the direction of gravity through their swimming antennae. Recent investigations using simulated, weightlessness conditions coupled with absence of illumination revealed hair like structures or setae on the basal, articulating socket of the antennae that, when surgically removed, resulted in disorientation. Given the simulated weightlessness or neutrally buoyant condition that eliminated sinking of the normally negatively buoyant Daphnia, it was proposed that the antennal socket setae function as rheoceptors stimulated by the upward rush of water currents during gravity induced, sinking phase of daphnid swimming movements. This rheoceptively mediated, gravity perception hypothesis is further supported by morphological investigations. Scanning electron micrographs indicate that antennal socket setae are anatomically similar to proprioceptors used by higher crustaceans to monitor gravitational direction.

    Beschreibung: Amyloplast DNA (apDNA), when subjected to digestion with restriction endonucleases, yields patterns nearly identical to that of DNA from mature pea chloroplasts (ctDNA). Southern transfers of apDNA and ctDNA, probed with the large subunit (LS) gene of ribulose-1,5-bisphosphate carboxylase (Rubisco), shows hybridization to the expected restriction fragments for both apDNA and ctDNA. However, Northern transfers of total RNA from chloroplasts and amyloplasts, probed again with the LS gene of Rubisco, shows that no detectable LS meggage is found in amyloplasts although LS expression in mature chloroplasts is high. Likewise, two dimensional polyacrylamide gel electrophoresis of etiolated gravisensitive pea tissue shows that both large and small subunits of Rubisco are conspicuously absent; however, in greening tissue these two constitute the major soluble proteins. These findings suggest that although the informational content of these two organelle types is equivalent, gene expression is quite different and is presumably under nuclear control.

    Beschreibung: Changes in contical bone formation found in rats on the model and wheather they are due to hind limb unloadng or involve a stress response was studied. Variables known to effect bone histomorphometry and physiology in that model are defined. The variables studied were: (1) changes in bone formation with age in two commonly used rat strains; (2) effects of cold stress on bone formation and apposition rates relative to changes in the same parameter in rats on the model. Body mass, growth rates, and tibial radographic and histomorphometric measurements were compared in Sprague-Dawley (S/D) and Fischer 344 (F) rats aged 6 to 68 weeks. Histomorphometric measurements of rat tibia from the juvenile to the adult period were studied, and tibial growth rates relative to both age and weight were compared. The physiological effects of unweighting the hind-limbs with those of cold stress in rats studied after 1 or 3 weeks of exposure. Adrenal weights, thymus weights, and corticosteroid levels were used as indictors of environmental stress.

    Beschreibung: Differences in radiation quality from the six most common electric lamps have little effect on photosynthetic rate. Radiation quality primarily alters growth because of changes in branching or internode elongation, which change radiation absorption. Growth and yield in wheat appear to be insensitive to radiation quality. Growth and yield in soybeans can be slightly increased under high pressure sodium (HPS) lamps compared to metal halide lamps, in spite of greatly reduced chlorophyll concentrations under HPS lamps. Daily integrated photosynthetic photon flux (mol m(exp -2)d(exp -1)) most directly determines leaf anatomy and growth. Photosynthetic photon flux (PPF) levels of 800 (mu)mol m(exp -2)s(exp -1) are adequate to simulate field daily-integrated PPF levels for both short and long day plants, but plant canopies can benefit from much higher PPF levels.

    Beschreibung: In vitro characteristics of cardiac cells cultured in simulated microgravity are reported. Tissue culture methods performed at unit gravity constrain cells to propagate, differentiate, and interact in a two dimensional (2D) plane. Neonatal rat cardiac cells in 2D culture organize predominantly as bundles of cardiomyocytes with the intervening areas filled by non-myocyte cell types. Such cardiac cell cultures respond predictably to the addition of exogenous compounds, and in many ways they represent an excellent in vitro model system. The gravity-induced 2D organization of the cells, however, does not accurately reflect the distribution of cells in the intact tissue. We have begun characterizations of a three-dimensional (3D) culturing system designed to mimic microgravity. The NASA designed High-Aspect-Ratio-Vessel (HARV) bioreactors provide a low shear environment which allows cells to be cultured in static suspension. HARV-3D cultures were prepared on microcarrier beads and compared to control-2D cultures using a combination of microscopic and biochemical techniques. Both systems were uniformly inoculated and medium exchanged at standard intervals. Cells in control cultures adhered to the polystyrene surface of the tissue culture dishes and exhibited typical 2D organization. Cells in cultured in HARV's adhered to microcarrier beads, the beads aggregated into defined clusters containing 8 to 15 beads per cluster, and the clusters exhibited distinct 3D layers: myocytes and fibroblasts appeared attached to the surfaces of beads and were overlaid by an outer cell type. In addition, cultures prepared in HARV's using alternative support matrices also displayed morphological formations not seen in control cultures. Generally, the cells prepared in HARV and control cultures were similar, however, the dramatic alterations in 3D organization recommend the HARV as an ideal vessel for the generation of tissue-like organizations of cardiac cells in simulated microgravity.

    Beschreibung: MIP-101 is a poorly differentiated human colon carcinoma cell line established from ascites that produces minimal amounts of carcinoembryonic antigen (CEA), a 180 kDa glycoprotein tumor marker, and nonspecific cross-reacting antigen (NCA), a related protein that has 50 and 90 kDa isoforms, in vitro in monolayer culture. MIP-101 produces CEA when implanted into the peritoneum of nude mice but not when implanted into subcutaneous tissue. We tested whether MIP-101 cells may be induced to express CEA when cultured on microcarrier beads in three-dimensional cultures, either in static cultures as non-adherent aggregates or under dynamic conditions in a NASA-designed low shear stress bioreactor. MIP- 101 cells proliferated well under all three conditions and increased CEA and NCA production 3 - 4 fold when grown in three-dimensional cultures compared to MIP-101 cells growing logarithmically in monolayers. These results suggest that three-dimensional growth in vitro simulates tumor function in vivo and that three-dimensional growth by itself may enhance production of molecules that are associated with the metastatic process.

    Beschreibung: Fusion Systems Corporation has developed a mercury-free, low infrared, efficient microwave lamp using a benign sulfur based fill optimized for visible light. Our literature search and discussions with researchers directed us to enhance the bulbs red output. We have demonstrated a photosynthetic efficacy of over 2 micro-moles per microwave joule which corresponds to over 1.3 micro-moles per joule at the power main. Recent work has shown we can make additional increases in overall system efficiency. During the next two years, we expect to demonstrate a system capable of producing more than 1.5 micro-moles/joule measured at the power main with significantly less IR than alternative lamp systems. We determined optimal plant growth light requirements via a literature search and researcher input. We surveyed candidate lamp fill materials to be used in combination with sulfur and explored several methods of increasing photosynthetic efficacy.

    Beschreibung: Soluble protein expression in Arabidopsis thaliana L. (Heynh.) leaf and stem tissue was examined after chronic clinorotation. Seeds of Arabidopsis were germinated and plants grown to maturity on horizontal or vertical slow-rotating clinostats (1 rpm) or in stationary vertical control units. Total soluble proteins and in vivo-labeled soluble proteins isolated from these plants were analyzed by two-dimensional sodium doedocyl sulfate polyacrylamide gel electrophoresis (SDS PAGE) and subsequent fluorography. Visual and computer analysis of the resulting protein patterns showed no significant differences in either total protein expression or in active protein synthesis between horizontal clinorotation and vertical controls in the Arabidopsis leaf and stem tissue. These results show chronic clinorotation does not cause gross changes in protein expression in Arabidopsis.

    Beschreibung: The Porous Tube Plant Nutrient Delivery System (PTPNDS), a hydrophilic, microporous ceramic tube hydroponic system designed for microgravity, will be tested in a middeck locker of the Space Shuttle. The flight experiment will focus on hardware operation and assess its ability to support seed germination and early seedling growth in microgravity. The water controlling system of the PTPNDS hardware has been successfully tested during the parabolic flight of the KC-135. One challenge to the development of the spaceflight experiment was to devise a method of holding seeds to the cylindrical porous tube. The seed holder must provide water and air to the seed, absorb water from the porous tube, withstand sterilization, provide a clear path for shoots and roots to emerge, and be composed of flight qualified materials. In preparation for the flight experiment, a wheat seed-holder has been designed that utilizes a cellulose acetate plug to facilitate imbibition and to hold the wheat seeds in contact with the porous tube in the correct orientation during the vibration of launch and the microgravity environment of orbit. Germination and growth studies with wheat at a range of temperatures showed that optimal moisture was 78% (by weight) in the cellulose acetate seed holders. These and other design considerations are discussed.

    Beschreibung: The objective of this feasibility study was to subject pregnant rats of the same age, strain, and size that will be utilized in a shuttle flight experiment to all flight conditions except the unique microgravity of space flight and determine the feasibility of the proposed experimental design to meet the experimental objectives. The study utilized facilities at NASA, Ames Research Center, Moffett Field, CA to subject the rats to the gravitational stresses of a simulated shuttle launch and simulated shuttle landing. One hundred pregnant rats were received on gestation day (G) 2 (day 1 = day of vaginal sperm) and on G7, eighty rats were laparotomized to determine the condition of pregnancy and allow assignment to test groups. The five test groups (N=10 each group) were as follows: Group 1, Nominal Flight; Group 2, Laparotomy Control; Group 3, Hysterectomy Control; Group 4, Vivarium Control; Group 5, Delayed Recovery. On G9, animals in groups 1,2,3, and 5 were subjected to a shuttle launch simulation. On G18, groups 1,2, and 3 were subjected to a shuttle landing simulation and on this same day groups 1 and 2 were subjected to unilateral hysterectomy to obtain fetuses and placentas for evaluation. Fetal crown-rump length and fetal weight of the Nominal Flight group was significantly less than the Laparotomy Control group, but placentas were similar. On G20, group 5 was subjected to a shuttle landing simulation and on this day this group received a unilateral hysterectomy and fetuses and placentas were weighed. Animals in all groups were allowed to go to term and all animals delivered between 06:00 hours G22 and 18:00 hours G23. After delivery, a blood sample was taken from each experimental dam, and they were euthanized and the thymus and adrenal glands weighed. The thymus weight from all experimental group dams was decreased relative to the Vivarium Control group but adrenal glands and hormone values in dam plasma was similar in all groups. Pups from experimental groups were tattooed for identification, the anogenital distance of male pups was measured, and all pups placed with foster dams and litter sizes were standardized to 10. There was no difference in anogenital distances between male pups from different test groups. Pups delivered from Delayed Recovery animals were smaller than pups delivered from Nominal Flight animals. On neonatal day 7, all pups were euthanized and pup adrenal glands and thymus weighed. There was no difference in weights of thymus and adrenal glands in pups euthanized at neonatal day 7. Collectively, these data confirm the feasibility of the experimental design to meet objectives of the studies proposed for shuttle flight.

    Beschreibung: Radish plants (Raphanus sativus L. cvs. Cherry Belle, Giant White Globe, and Early Scarlet Globe) were grown in four different CO2 enriched environments, 0.04, 0.10, 0.50, and 1.00 kPa (400, 1000, 5000, 10000 ppm). Cultivar responses to CO2 treatments varied, where cv. Cherry Belle showed no significant response to CO2 enrichment, cv. Giant White Globe was moderately affected and Early Scarlet Globe was strongly affected. Enrichment at 0.10 kPa led to greater root dry matter (DM) than 1.00 kPa for cv. Giant White Glove, whereas 0.10 kPa produced greater storage root, shoot, and root DM than 1.00 kPa for cv. Early Scarlet Globe. The data suggest that 1.00 kPa CO2 may be detrimental to the growth of certain radish cultivars. Root:shoot ratios tended to increase with increasing CO2 concentration. Water use efficiency (g biomass/kg H2O) increased with increasing CO2 enrichment, up to 0.5 kPa but then declined at the 1.00 kPa treatment. The total nitric acid used to maintain nutrient solution pH was lowest at the 1.00 kPa treatment as well, suggesting a decreased demand of nutrients by the plants at the highest CO2 level.

    Beschreibung: Body temperatures and thermoregulatory responses were measured at rest and during submaximal exercise under normal ambient conditions in 11 aerobically-conditioned men (age = 29.2 +/- 6.2 yr, VO2(max) = 3.73 +/- 0.46 min(sup -1), relative body fat = 12.3 +/- 3.7 percent, mean +/- SD) with (CT) and without (NCT) the ingestion of 10 mg of caffeine per kg of body weight. Oxygen uptake (VO2), heart rate (HR), and rectal (T(sub re)) and mean skin (T-bar(sub sk)) temperatures were recorded for 100 minutes starting one minute after ingestion of caffeine or a placebo. Data were collected throughout 30 minutes of rest (sitting) and the following 70 minutes of sitting leg ergometer exercise using the same constant load (1,088 +/- 153 kgm/min) in both NCT and CT. The load resulted in a mean relative exercise intensity equal to approximately 68 percent of VO2(sub max). Skin heat conductance (H(sub sk)) and sweat rate were calculated. Two-way analysis of covariance revealed no significant (P greater than 0.05) differences between NCT and CT in VO2, HR, T(sub re), T-bar(sub sk), or H(sub sk). A dependent t-test indicated no significant difference between NCT and CT in sweat rate. Thus, a high level of caffeine ingestion has no detrimental effects on body temperatures and thermoregulatory responses during moderately heavy exercise in normal ambient conditions.

    Beschreibung: The hypothesized fluid shifts and resultant responses that occur during spaceflight are simulated by six-degree head down tilt (HDT) and water immersion (WI). The purpose of this study was to compare exercise thermoregulation before and after physiologic mechanisms reduce plasma volume (PV) in response to 24-hr HDT (HDT24). A secondary study utilized WI to reproduce the PV reduction of HDT24. Seven males were studied in two conditions: during 70 minutes of supine cycling ergometry at 58 percent of peak oxygen consumption following 1-hr HDT (HDT1) and HDT24; and up to 6 hr WI at 34.5 C. Plasma volume was reduced by 10.4 percent in HDT24 when compared to HDT1. Pre-exercise rectal temperature, T(sub re), was an average 0.22 C higher after HDT24. Rectal temperature increased during exercise with no interaction between time and treatment. The reduced PV and elevated pre-exercise T(sub re) had offsetting effects on thermoregulatory mechanisms, suggesting no alteration in the response at a given T(sub re). Plasma volume was reduced by 4.3 +/- 2.3 percent and 1.1 +/- 1.8 percent following HDT24 and WI, respectively, compared to upright chair rest. Although the reductions in PV were not significantly different, great intra-individual variability was evident. The ability to reproduce PV changes consistently with HDT and WI is limited by this variability.

    Beschreibung: During a centrifuge experiment, any interruption that requires stopping the centrifuge may influence the results. Centrifuges often must be stopped for animal maintenance (food, water and waste removal), especially in cases of timed feedings. To eliminate the need for stopping the centrifuge while still providing timed feeding, an automatic paste diet feeder was developed. The feeder is based on a constant volume concept and can deliver a predetermined amount of paste diet at specified time intervals. This unit was supported by water delivery and waste collection systems. The entire system performed reliably and maintained the animals well for a continuous centrifugation experiment of 14 days.

    Beschreibung: In shoots of many plants, of which tomato (Lycopersicon esculentum Mill.) is an example, ethylene production is substantially increased during gravitropism. As a first step toward elucidating the role of ethylene in gravitropism, detailed time courses of ethylene production in isolated hypocotyl segments and whole plants were measured for gravistimulated and upright tomato seedlings. In the first experiment, seedlings were set upright or laid horizontal and then, at 15 min intervals, sets of hypocotyls were excised and sealed into gas tight vials. A steady long term rise in ethylene production begins after 15 min gravistimulation. It is possible that this increase is a consequence of the accumulation of indoleacetic acid (IAA) in the lower tissue of the hypocotyle. In a second kind of experiment, whole seedlings were enclosed in sealed chambers and air samples were withdrawn at 5 min intervals. Stimulated seedlings produced more ethylene than controls during the first 5 min interval, but not appreciably more during the second. This suggests the possibility that the ethylene production induced during the first 5 min occurs immediately rather than after a lag, and thus much too soon to be controlled by redistribution of IAA.
