    Notes: Biological mineralization is a cell-mediated process which is believed to be triggered by a “nucleating agent.” Various matrix structures, such as matrix vesicles, collagen fibrils and macromolecules, have been claimed to be the source of this substance, since these components have been found by transmission electron microscopy (TEM) of thin sections to be associated with early mineral crystals. Systematic image analysis of the relationships revealed in electron micrographs between specific matrix components and early mineral deposits has shown that unequivocal image interpretation is not possible. This is due principally to the problems posed by overprojection and truncation phenomena, since the structures being analyzed lie within the same dimensional range as thin section thickness. Various examples are illustrated and discussed. The site at which mineral crystals are initially laid down thus cannot be identified with any matrix structure using thin section TEM. Possible technical approaches to resolve this problem of image analysis are discussed.

    Notes: A computer graphics system for reconstruction from serial section micrographs was applied to intracellular details of a yeast target cell (Saccharomyces cerevisiae cell) induced by the α factor mating pheromone and was also applied to a periodontal structure of a dog tooth moved orthodontically. In the former, intracellular organelles and a distribution of vesicles could be clearly observed through the cell membrane using the transparent display method in which the smoothing of the reconstructed outer cell membrane surface by computer processing was applied to the transparent display. In the latter case, by cutting through a reconstructed dog tooth and its periodontal tissues, labiolingual and mesiodistal cut surfaces of the tooth and of adjacent alveolar bone could be observed with fine details (232 sections were used).

    Notes: In order to determine the best conditions to carry out quantitative ultrastructural studies in plant specimens, five different fixation techniques, including some of the most reported electron microscopy fixatives (glutaraldehyde-paraformaldehyde, osmium tetroxide, potassium permanganate), were assayed in onion root meristems to check their ability to induce morphometric changes in Golgi apparatus ultrastructure. Although the parameters evaluated showed in all cases the same tendency, values obtained after permanganate fixation were always higher than those found after aldehyde techniques (especially aldehyde-osmium). Aldehyde followed by osmium fixation appears as the most indicated fixation method when accurate quantitative ultrastructural studies are to be developed.

    Notes: Optimal imaging of complex structures requires proper alignment relative to the optic axis of the electron microscope. This is especially important for high-voltage and intermediatevoltage microscopes, which form an in-focus image throughout the entire thickness of the object. As a result, structures at different specimen heights form overlapping and confused images that severely curtail the usefulness of these instruments.The work described here provides a generalized, flexible method for optimizing specimen orientation and eliminating or limiting image overlap by means of a commonly used double-tilt stage. Analysis of the motion about the two axes provides accurate tilting for any azimuthal direction whether or not it corresponds to a mechanical axis of the stage. An object can be positioned to minimize image overlap, to record stereopairs for any parallax axis, and to record three-dimensional data sets by the conical collection geometry.Images of muscle paracrystals are shown after tilting about an axis perpendicular to a symmetry direction. The tilted image displays higher-order symmetry, which is altered by changes of one degree. Precision double-tilting for optimizing stereopairs is shown for a desmosome recorded using different parallax axes and pretilts. A tomographic conical data-collection scheme is demonstrated by imaging a microtubule axoneme for a specific cone half-angle and arbitrary azimuthal angles.

    Notes: The relative sensitivity of an analytical electron microscope and energy-dispersive x-ray detector to x-rays of various elements is investigated through an extensive kASi factor study. Elemental standards, primarily National Bureau of Standards multielement research glasses, were dry-ground into submicrometer-sized particles and analyzed at 200 kV accelerating potential. The effect of self-absorption of x-rays by the particle has been corrected for, allowing the experimental kASi factors from this study to approximate those that could be obtained from “infinitely thin” specimens.Whenever possible, elemental k-factors were determined by the analysis of many (up to a maximum of nine) different standard materials. Experimental kASi factors were calculated for a wide range of Kα, Lα, and Mα x-ray lines. For comparison, theoretical kASi factors, employing a variety of ionization cross sections, were computed. Good agreement is obtained between several of the theoretical k-factor models and the experimental results. Mass volatilization of Na and K from the small glass particles during analysis is discussed, as are observations that the grinding and/or dispersing of standard materials in a liquid (such as ethanol) may promote leaching of certain elements from the particle matrix.

    Notes: Experimental conditions for obtaining the optimum signal-to-background (S/B) ratio in reflection electron energy-loss spectroscopy (REELS) are investigated. It is shown that the S/ B ratio can be improved by lowering the incident energy of the electrons. The spectra taken from the GaAs (660) specular reflection spot under the surface resonance condition is demonstrated to have the best S/B ratio and lowest surface plasma excitation, which is capable of providing structural information on the top few atomic layers.

    Notes: C-peptide and insulin antigenic sites in rat pancreatic β cells were both labeled by the protein A and immunogold techniques. Gold bead density in the secretory granules was correlated with granule size distribution. Our results demonstrate that the distribution patterns of antigenic sites for C-peptide (which also represents proinsulin sites) and insulin are similar when granule size distribution is correlated to the density of antigenic sites. It was found that the ratio of C-peptide to insulin in granules is independent of granule size. Examination of frequency histograms by a Kolmogorov-Smirnov analysis excluded the possibility that the antigenic sites of C-peptide and insulin are distributed in the same manner. Mean gold bead density for insulin antigen (97 ± 24) was slightly higher than C-peptide density (64 ± 18). The efficiency of labeling was found to be very low; less than 0.1% of the theoretically estimated antigenic sites were labeled.

    Notes: We designed and built a freeze-drying device that ensures the protection of the specimens against contaminants during mounting on the cold stage of the freeze-fracture machine, transferring into the vacuum chamber and deep etching. The device consists of a copper cap that covers the specimen and a thermal connection that ensures thermal transfer between the microtome arm and the copper cap. This device was used to study the ultrastructural features of the erythrocyte membrane skeleton and the immunocytchemical localization of spectrin in an “in situ” approach, by freeze drying and platinum rotary shadowing. Human erythrocytes adhered to polylysine-coated coverslips and were broken by a stream of buffer that mimics the intracellular ionic environment (“inside buffer”). The samples were prefixed in periodate-lysine-paraformaaldehyde fixative, labeled with antispectrin 5-nm gold particles, fixed in glutaraldehyde, mordanted in tannic acid, postfixed in OsO4, repeatedly washed in water, rinsed quickly in 30% ethanol, freeze-dried, and rotary-shadowed. Electron microscopic examination of the replicas revealed the skeletal network on the inner surface of the erythrocyte membrane. Immunocytochemical labeling proved that spectrin represents a fibrillar component of the network. Our data confirm the speculative model of the molecular organization of the erythrocyte skeleton, based on studies on in vitro association of proteic constituents. Both the technique and the device developed by us may lead to a deeper understanding of the spatial organization of the cytoskeletal network of more complex cell types.

    Notes: The use of osmium-potassium ferrocyanide as the secondary fixative greatly improved chondrocyte preservation and stabilized the cartilage matrix proteoglycan. The proteoglycan was similar in appearance to that seen following fixation in the presence of cationic dyes. Extensive glycogen preservation was noted in these cells, occupying the area prior to and during the formation of the secondary center of ossification. The volume and organization of the glycogen within the cell cytoplasm were greater than that following buffered osmium fixation, and the cellular vacuoles within were greatly reduced.The cells forming the secondary center prior to the onset of mineralization were of greatest interest, because other studies compared them with the primary growth plate and described them as showing signs of hypertrophy as early as 5 days postnatally, as is found in the primary growth plate. Our observations indicate that glycogen is present in these cells, and cellular enlargement was not present. The cells do not resemble the hypertrophic chondrocytes of the primary growth plate, as far as cytoplasmic content is concerned, and we suggest that they may contribute to the development of the secondary center in a different fashion.

    Notes: A magnetic-field-superimposed field emission gun with low aberrations and equipped with a zirconiated tungsten emitter has been developed for applications where very stable high probe currents are required. It has been tested on a conventional electron microscope at 10 kV and on an electron beam testing system at 1 kV. Probe current i = 250 nA in a probe size d = 0.4 μm is obtained at 10 kV; at 1 kV the resolution is 0.1 μm with i = 5 nA, and 0.4 μm with i = 30 nA. For these probe currents, the spatial broadening effect due to electron-electron interactions in the beam is the preponderant factor limiting the probe size.

    Notes: The properties of oxides may be drastically influenced by their microstructures. Thus the knowledge of nonstoichiometric mechanisms is an essential tool for the conception of new inorganic compounds by solid-state chemist. Through three chosen examples of complex oxides with perovskite-related structure, these points are illustrated. The new families of phosphate tungsten bronzes, the antiferromagnetic AMn1 - x FexO3 - y, and the copper oxides LCuO3 - x (La = Ba, Y, La) were studied by high-resolution electron microscopy (HREM) in order to analyze the mechanisms of nonstoichiometry, the defects of which can modify their physical properties (magnetism or electronic conductivity) and the factors that govern their stabilization.

    Notes: The applications of electron diffraction and diffraction contrast electron microscopy with which to study the structure and dynamics of organic thin films are discussed. The techniques of making thin film deposits on substrates and of forming free-standing thin films over holes on the substrate are described. Selected area electron diffraction and diffraction contrast imaging techniques for thin film studies are elaborated, and examples are given. Methods to reduce radiation damage and environmental protection of the thin film specimen are outlined.The interpretation of electron diffraction and imaging data is given for three cases: (1) The effects of film tilting and molecular tilting (with respect to the film plane) are examined. (2) The detection of phase transition is illustrated. (3) The use of labels to mark film domains is shown together with the measurement of dynamic movement.

    Notes: Cytoskeletons underneath the postsynaptic membrane of neuromuscular junctions were studied by using a quick-freeze deep-etched method and immunoelectron microscopy of ultrathin frozen sections. In a quick-freeze deep-etched replica of fresh, unfixed muscles, 8.9 ± 1.5-nm particles were present on the true postsynaptic membrane surface. Underneath this receptor-rich postsynaptic membrane, networks of fine filaments were observed. These cytoskeletal networks were more clearly observed in extracted samples. In these samples, diameters of the filaments which formed networks were measured. In the platinum replica, three kinds of filament were recognized - 12 nm, 9 nm, and 7 nm in diameter. The 12-nm filament seemed to correspond to the intermediate filament. The other two filaments formed meshworks between intermediate filaments and plasma membrane. In ultrathin frozen sections vinculin label was localized just beneath the plasma membrane. Thirty-six percent of the label was within 18 nm from the cytoplasmic side of the plasma membrane and 50% was within 30 nm. Taking the size of the vinculin molecule into account, it was concluded that vinculin is localized just beneath the plasma membrane and might play some role in anchoring filaments which formed meshworks underneath the plasma membrane.

    Notes: A scanning electron microscope of ultra-high-vacuum (UHV-SEM) with a field emission gun (FEG) is operated at the primary electron energies of from 100 eV to 3 keV. The instrument can form the images that contain information on surface chemical composition, chemical bonding state (electronic structure), and surface crystal structure in a microscopic resolution of several hundred angstroms (Å) using the techniques of scanning Auger electron microscope, scanning electron energy loss microscope, and scanning low-energy electron diffraction (LEED) microscope. A scanning tunneling microscope (STM) also has been combined with the SEM in order to obtain the atomic resolution for the solid surface.The instrumentation and examples of their applications are presented both for scanning LEED microscopy and STM.

    Notes: Four kinds of works on the detection of displacement of atoms in crystals are shown. The irregular small displacement of atoms has been detected, with an accuracy of about 0.1 Å around dislocations and stacking faults in Au crystals as shown by their electron microscope images. The displacement of the atomic images is recorded by aberration-free focus (AFF). Even when the periodic displacement of atoms in SiC and TaS2 crystals is around 0.1% of the lattice constant, this displacement has been revealed as the weak-contrast anomaly in the images. Using in situ observations by a TV system attached to an electron microscope, the rapid movements of atoms that have taken place within 1/30 sec have been recorded. Using the technique of successive subtraction of the images by TV system and image sigma, only the images of moving atoms in Au crystal have been recorded each 1/10 sec.

Neuropeptides may function as hormones secreted from chromaffin cells or as neurotransmitters/neuromodulators released from nerve terminals. Thus, to understand the role of neuropeptides, it is important to explore their localization in different species.     Notes: Understanding the relationship between the molecular structure and the macroscopic properties of polymer solutions and gels, oil-water-surfactant emulsions, lyotropic and thermotropic liquid crystals, colloidal dispersions, detergents, and other such “microstructured fluids” is essential to the optimal use of these commercially important materials. Modern rapid-freezing methods followed by freeze-fracture replication techniques are ideally suited to allow the direct visualization of the three-dimensional structure of the particles or units that make up the dispersion, while simultaneously revealing their orientation and distribution with molecular resolution. This paper reviews the necessary experimental conditions required to successfully exploit the freeze-fracture technique as it applies to microstructured fluid systems. The benefits and limitations of structural studies by freeze-fracture techniques as opposed to the more commonly used light, X-ray, and neutron-scattering methods are discussed. Freeze-fracture replicas can also be imaged by scanning tunneling microscopy to reveal directly three-dimensional fracture contours with improved resolution.

    Notes: In this paper, we present a technique for analysis of composition gradients, using an analytical electron microscope, within the primary phase of a two-phase alloy for the case where the second-phase particle size is similar to the size of the irradiated volume. If the composition difference between the two phases is large, the detected compositional fluctuations associated with varying phase fractions may mask any underlying composition gradient of the primary phase. The analysis technique was used to determine grain boundary chromium concentration gradients in a nickel-base superalloy, alloy X-750. The technique may also be of use in other alloy systems.

HRTEM images were submitted to a filtering in order to improve their interpretation. One type of dislocation - (010)[001]/2 - appears to be dissociated, with (010)[001]/2 planar defect, whereas the other one - (010)[110]/2 - is perfect.     Notes: With the aid of epitaxial orientation techniques originally designed for linear polymer crystallization, it is found that a large assortment of linear chain molecules can be prepared for electron diffraction study in a projection onto the molecular axes. This not only facilitates a study of ordered monodisperse molecular crystal structures but also of the disordered state as well, including thermotropic phase transitions and the structure of binary solids. Representative studies of monodisperse and polydisperse phase behavior based on electron diffraction and differential scanning calorimetry measurements are reviewed for n-paraffins, glycerolipids, and cholesteryl esters. The importance of observing the microcrystalline state is readily apparent from these studies - not only because the symmetry of individual small crystals can be determined, but also because local structural variations not detectable in bulk measurements are readily observed.

    Notes: Lactoferrin, a DNA-binding protein, was complexed to colloidal gold and applied on ultrathin sections of resin-embedded plant tissues and bacterial cultures. Optimal results were obtained when lactoferrin was tagged to colloidal gold particles at pH 9.2. Postfixation with osmium tetroxide and embedding with Epon did not prevent the accessibility of the protein towards its corresponding binding sites. In plant nuclei, labeling was observed over the dense chromatin and to a lesser extent over the dispersed chromatin. Nucleolar labeling was preferentially located over the dense fibrillar component. Gold particles were also found to be associated with chloroplasts and mitochondria. In prokaryotic cells, a dispersed labeling was noted over the cytoplasm and, in some cases, the aggregation of few gold particles suggested the presence of packed DNA fibrils. Various control experiments confirmed the specificity of the labeling pattern obtained. Lactoferrin-gold complex appears to be a valuable probe for the intracellular demonstration of DNA molecules in double-fixed and Epon-embedded tissues.

    Notes: A technique for performing light, scanning, and transverse transmission electron microscopy on cultured cells grown within a single tissue culture flask is described. Permanent light microscopy slides are obtained by removing selected portions of the plastic tissue culture vessel and mounting them on glass slides with an aqueous mounting solution. The images obtained from these slides are superior to viewing through the bottom of the flask with an inverted stage microscope. For scanning electron microscopy, selected areas are also cut from the remainder of the vessel and prepared for viewing. The final portion of the culture container is transferred and attached to a new tissue culture vessel and prepared for transmission electron microscopy using alcohol instead of acetone and propylene oxide during dehydration, infiltration, and embedding.

    Notes: As a microscale tool for observing GaAs-Alx Ga1-xAs heterostructures, backscattered electron (BE) images in the scanning electron microscope (SEM) were compared with conventional secondary electron (SE) images. BE images were found to be more sensitive to compositional differences between GaAs and AlxGa1-xAs and less sensitive to surface roughness. BE images have a spatial resolution of 10 nm or better. This method enables the nondestructive observation of ultrafine lateral periodic structures, such as quantum-well-wire (QWW) structures, fabricated by compositional disordering technology using focused Ga ion-beam (Ga-FIB) implantation into GaAs-AlxGa1-xAs material.

    Notes: A new method was developed which is suitable for the preparation of mammalian sperm for scanning electron microscopy under either laboratory or field conditions. Samples of ejaculates from humans, two ferret species, and epididymal sperm from the African elephant were diluted in Millonig phosphate buffer and then fixed in glutaraldehyde solution. A small sample of the fixed sperm suspension was diluted in the same buffer, withdrawn with a syringe, and injected very slowly onto either a cellulose acetate or a polycarbonate membrane filter. This step was essential to concentrate the dilute sperm samples. During the various dilution steps most of the granular prostatic secretions were lost. However, a protein-like sheath, which remained attached to most sperm, obscured the surface features and had to be removed for SEM studies. It was removed by prolonged fixation/etching in 1% osmium tetroxide. Membrane filters containing sperm on their surfaces then were dehydrated, dried by the critical point drying method, and sputter coated with gold. Polycarbonate filters were superior to cellulose acetate filters in producing a flat and homogeneous background.

    Notes: The term “etching,” in electron microscopy, refers to the removal of specimen surface layers and includes chemical, electrolytic, and ion-beam methods. The ion-beam etching process is used to remove layers of a target material by bombarding it with ionized gas molecules. Recently, the method has been applied to the field of biological specimens; however, the practical procedures for such organic materials have not been developed. In the present study, we used an apparatus in which a beam of argon ions is collimated and focused by electrostatic lenses onto an appropriate target. We demonstrated the optimum conditions to observe biological specimens that were treated with osmium tetroxide and tannic acid. The specimens were examined uncoated at low accelerating voltage using a field emission scanning electron microscope.According to our experiments, when a biological specimen was observed under high-resolution conditions at over 50,000x magnification, the optimum condition of ion-beam etching consisted of an accelerating voltage of E = 1 keV and an ion-beam dose of It = 360 ∼ 400 μA. min, depending on parts of the specimens. In order to decrease overetching, we had to choose factors such as E = 1 ∼ 2 keV and It = 500 μA. min.

    Notes: Number, length, and diameters of dendritic spines of the granule cell in the dorsal leaf of the rat dentate gyrus were measured by using high-voltage electron microscope stereo images of 5-m̈m-thick Golgi preparations with the aid of a three-dimensional image analyzer system.Spine densities of 2.02 ± 0.28, 2.28 ± 0.33, and 3.36± 0.35 per 1 μm at distal, middle, and proximal portions of the dendrite were obtained. These values were about 1.6-fold of the previous light microscopical report.Mean three-dimensional spine length were 1.244 ± 0.506 μm, 1.262 ± 0.563 μm, and 1.254 ± 0.584 μm at distal, middle, and proximal portions, respectively, which were about 1.4 times longer than those measured in two dimensions.By using measured morphometrical parameters of spines such as lengths, diameters, and population densities, total spine surface areas of 2.401 μm2, 2.806 μm2, and 4.180 μm2 per 1 μm of the dendrite at distal, middle, and proximal portions, respectively, were obtained. The total surface area of dendrite was about doubled by the addition of the spines at each dendritic portion.The advantageous features and the problems of the present method are discussed.

    Notes: HIV particles were usually seen on the surface of established lymphoid cells derived from AIDS patients or on CEM cells infected with HIV, and sometimes in cytoplasmic vacuoles. The virus particles were formed by a budding process from the plasma membrane of an infected cell. The budding particles were of a doughnut form. Various profiles of virus particles were seen extracellularly: type 1 had a bar-shaped, electron-dense core, type 2 had a central and type 3 an eccentric electron-dense round core, type 4 was doughnut-shaped, and type 5 had a layered core. However, projection patterns of the AIDS virus model suggested that type 1, 2 and 3 particles are similar. Therefore, the AIDS virus may be one of three main types: with or without a dense core, and with a layered core. It is thought that a particle with a layered core and a doughnut-type particle may be immature viruses.

    Notes: Single crystalline composite films of iron and MgO are prepared by a simultaneous vacuum deposition technique. The structures of the composite films, especially of the iron crystallites embedded, are studied by high-resolution electron microscopy and nanometer-area electron diffraction. The α-iron (b.c.c.) crystallites of 1 nm in size are epitaxially embedded in single crystalline MgO films, the orientation being (011)[100]Fe/(001)[100]Mgo and (001)[110]Fe//(001)[100]Mgo. A heat treatment of the as-grown films at 500-1,000°C brings about a phase transformation of the crystallites from α-iron to γ-iron (f.c.c.), followed by a grain growth of α-iron and finally the growth of the spinel, MgFe2O4. The γ-iron crystallites transformed are circular plates and have strains at the periphery to accommodate the surrounding MgO-matrix. The magnetic property of the composite films is also reported.

    Notes: The structure of amorphous Se films, in the topological random system, has been studied by the computer-simulation, electron diffraction (ED), and high-resolution transmission electron microscopy (HRTEM). As an example of HRTEM of the cellular random system, our recent investigation on Ba-ferrites is reviewed beforehand. In HRTEM images of spin-glass BaTi2-xSnxFe4O11 (x 〈 0.6), magnetic clusters or clusters of FeO6-octahedra surrounded by TiO6-octahedra have been found. The structure of BaSn2Fe4O11 (x = 2) has been determined. The ordering of Sn (Ti) and Fe ions increases with increasing x, which interprets the change from the spin-glass state to nonspin-glass state at x = 0.6. It is shown that a cluster of polarized lattice ions is detectable in images of polar-glass BaxK2-xFexTi6-xO13 (x ≥ 1.2). The structure models of amorphous Se films forming on a substrate have been constructed on a computer, and their radial distribution function (RDF) and HRTEM images have been calculated. Experimental RDF and HRTEM images have been obtained from vacuum-deposited amorphous Se films and are compared with the calculated ones. It is concluded that in the as-deposited films most of Se molecules may be composed of Se atoms as small as three and that by the electron-beam irradiation the molecules link to form spiral chains in amorphous state and then arrange to have the hexagonal crystal structure.

    Notes: The field of neural transplantation has moved rapidly forward in the last decade. Initially, fetal cells were used as implants to investigate their potential to ameliorate deficits in animal models of Parkinson's disease. However, because of the moral and legal problems associated with the use of fetal tissues in humans, alternative sources of donor tissue were sought which possessed the structural and functional characteristics needed to improve motor function in Parkinsonian patients. To date, one of the most promising tissues being investigated is the adrenal medulla, whose chromaffin cells possess an inherent plasticity of form and function. Transplanted chromaffin cells currently are being studied by a variety of approaches, including electron microscopy, in mouse, rat, and primate models of Parkinson's disease. An overview of the role of the chromaffin cell in this exciting and clinically important arena is briefly reviewed, with an emphasis on the fine structure of implanted chromaffin cells.

    Notes: Evidence is presented for morphological proof of exocytosis in the rat adrenal medulla in situ. Techniques were modified to allow perfusion of the intact adrenal gland with secretagogues (or electrical stimulation) followed by tannic acid. Unstimulated specimens demonstrated exocytotic (omega-shaped) profiles filled with flocculent material. This flocculation was also seen in the intercellular space. Stimulation of the adrenal medulla also resulted in the appearance of exocytotic profiles and an accumulation of the flocculent mass. This was often most evident in the subendothelial space. This is the first demonstration of exocytosis in the rat adrenal medulla by electron microscopy. The techniques used in this study will be useful for studying the pathway of secretory products of the adrenal chromaffin cell before they enter the vascular system.

    Notes: Mitotic activity often has been reported in embryonic and fetal sympathetic neuroblasts, principal sympathoblasts, and primitive sympathetic cells in various species at different stages of development. Postnatal adrenal medullary cells also are known to undergo mitosis, but such dividing capabilities rarely have been observed in the true postnatal extraadrenal chromaffin system. Although few in number, this work nevertheless has clearly identified such cells in varying stages of the mitotic cycle in the young dog, Syrian hamster, mouse, rabbit, and rat. The dividing cells were noted in paraaortic chromaffin organs, paraganglia, and within the inferior mesenteric ganglion as well. They displayed the morphological character usually associated with their adrenal medullary catecholaminergic counterparts, including numerous dense-cored vesicles known to be the harbingers of catecholamines and various peptides. Nerve endings were not noticed upon the mitotic cells. The phenomenon of dividing extraadrenal chromaffin cells augments existing data and perhaps suggests that these cells are more endocrine than neural in type and subservient to the adrenal medulla in its classic endocrine function.

    Notes: Current investigations on the immunohistochemical occurrence and co-occurrence of biogenic polypeptides in the mammalian carotid body were reviewed and extended by our own recent findings. The family of chromogranins and related peptides in glomus cells appears to have a widespread interspecies distribution, whereas other peptides investigated occur in a species-specific pattern. Immunoreactivity to antisera against opioids, which derive from the proenkephalin sequence, appears to be present in glomus cells of the rabbit, cat, dog, and a shrew. Conversely, glomus cells of pig and guinea pig predominantly are immunoreactive to cleavage products of prodynorphin, which co-occur in some cells with substance P and met-enkephalin-arg-phe, respectively. In the rat and Callithrix jacchus, opioid immunoreactivity is present in nerve fibres but not in glomus cells. Immunoreactivity to other peptides, such as neurotensin, cholecystokinin, neuropeptide Y, and galanin, is found only in one or two particular species. Neurotensin immunolabelling occurs in beagle dog glomus cells, which are known to lack substance P. Cholecystokinin immunoreactivity is present in glomus cells of dog and Callithrix, and co-exists with chromogranin A, neuropeptide Y, and substance P. Substance P appears to exist in both carotid body glomus cells and nerve fibres. Substance P immunoreactivity is present in glomus cells of all species investigated, except dog. Coexistence of substance P and calcitonin gene-related peptide (CGRP) is demonstrated in nerve fibres of the guinea pig carotid body, which originate in the petrosal and jugular ganglia. Other peptides visualized immunohistochemically in mammalian carotid body nerve fibres are vasoactive intestinal peptide and neuropeptide Y. The functional significance of the various peptides present in the carotid body is discussed.

    Notes: The methods by which Convergent Beam Electron Diffraction (CBED) is used to solve practical problems in materials science are reviewed. Particular emphasis is placed on those applications where a consensus has developed on the potential difficulties, but these problems have never been explicitly treated in the literature. Point-group determination using CBED is a standard technique, but there are several different experimental methods which are outlined here; the different approaches and the information obtained are compared. These point-symmetry determinations are shown to rely heavily on elimating the large number of potential artefacts: the possible sources of symmetry-breaking are summarised, and their salient features are identified. This should give a prescription for demonstrating whether symmetry-breaking arises through crystal structure, defects, strain, or specimen morphology. In space-group determinations the practical approach is now fairly standard, but the article draws attention to some anomalies. Strain and lattice parameter determinations are also standard, but a consensus is beginnning to emerge that the interpretive difficulties involved in allowing for thin film relaxation are extremely severe: a few of these difficulties are outlined.     Notes: This paper explains how the convergent beam electron diffraction (CBED) and large angle convergent beam (LACBED) techniques can be used to study crystal defects, bicrystals, and multilayers. It is shown how the LACBED technique in particular can be used to derive the magnitude and sign of the Burgers vectors of dislocations and displacements at stacking faults. For bicrystals and multilayers examined in plan-view, LACBED gives the rocking curve for a chosen reflection. This enables layer strains to be measured to ∼ 0.1% and composition profiles derived for both periodic and aperiodic structures. It is shown that a simple kinematic approach can be used to interpret the essential results in most cases.

    Notes: The use of whole, intact plant tissue for freeze-fracture electron microscopy provides important information that cannot be obtained from the use of isolated biological membranes or of artifical (phospholipid) membrane preparations. This is not to imply that these exminations of such preparations are not useful, since it would be difficult to interpret our observations of intact cells and tissues without the analysis of these model systems. Analysis of intact tissue and cells reveals the relative densities of membrane proteins of the different membranes within a cell; the three-dimensional organization of various organelles, especially the endoplasmic reticulum; changes in intramembranous particle (IMP) distribution due to stress or injury; and, in conjunction with the use of filipin, membrane sterol content and relative distribution.It is our intention that this survey of freeze-fracture images of intact plant tissues will illustrate the uniqueness of the information gained from an analysis of whole plant tissues compared to isolated membrane fractions.

    Notes: Model membrane systems are used extensively in all aspects of membrane research, and freeze-fracture is the preeminent procedure for directly visualizing local structure in these lipid dispersions. Here we describe in detail the formation of liposomes and how freeze-fracture is routinely employed as a complementary technique to biophysical and biochemical procedures in the characterization of multilamellar vesicles (most commonly known as liposomes) and unilamellar vesicles. Many preparative procedures exist for the formation of multi- and unilamellar vesicles. Examples of each system are given and their properties as well as freeze-fracture morphology are discussed. The detection of lipid-phase transitions is considered, in particular, with emphasis on the application of freeze-fracture to the study of lipid polymorphism. We briefly discuss the fracturing of apolar lipids which do not adopt bilayer structures but which can be stabilized into microemulsions by a phospholipid monolayer. Finally, a critical assessment is made of filipin as a morphological marker for cholesterol domains in the plane of the bilayer.

    Notes: An image can be represented digitally as a matrix of numbers. When those numbers are linearly related to a property of the object, such as mass per unit area, a simple integration of an image area leads to a total of that property, such as the mass of a particle that is represented in a selected area.Following techniques pioneered by Bahr and Zeitler, we illustrate the use of photographic densitometry of films exposed in an electron microscope to measure electron scattering. The transmission of an electron micrograph will be linear with respect to mass thickness for a particular value of background brightfield density, hence allowing determination of the mass of microscopic particles. We show here a digital computer method for conveniently establishing the linear condition by quantitative image processing using micrographs of polystyrene spheres. The method also serves to produce calibration curves for cases where the transfer from transmission to mass thickness is not linear. We also illustrate how an inexpensive computer is used to display and integrate regions of micrographs to determine particle mass.

    Notes: Silver enhancement of immunogold-labeled cells was carried out to increase the applicability of colloidal gold probes for visualization in the backscatter electron imaging (BEI) mode of a scanning electron microscope. Optimum conditions were established for single particle discrimination and differential counting of labeling density at low magnifications. Red blood cells double-labeled with 15 + 40 nm and 5 + 20 nm gold probes were silver-enhanced for 6 min and 20 min, respectively, at which times both pairs of labels increased to about 25 + 50 nm. The gold probes still appeared spherical after enhancement and were easily discriminated. Cells were also single-labeled with the above probes and enhanced accordingly. The present method enables visualization of individual particles of any probe size, labeling one, or simultaneously two, antigenic sites on cell surfaces. The silver enhancement procedure thereby allows cells to be labeled with small probes with increased labeling efficiency.

    Notes: A new chemical, Peldri II, is evaluated as a compound for drying soft biological tissues for scanning electron microscopy. Peldri II, a fluorocarbon, is a solid at room temperature and is a liquid above 25°C. Cells or tissues are embedded in Peldri II by immersing them in the liquid form and allowing it to solidify. Once solidified, Peldri II will sublime with or without vacuum to dry tissues, probably without introducing surface tension. Several types of cells and tissues have been examined to compare preservation with Peldri II and critical point drying techniques. No differences were detected between the two techniques when normal surface structures were examined. Peldri II appears to be a significant improvement over hexamethyldisilazane as a drying agent for scanning electron microscopy. It is also very convenient for drying large numbers of samples.

    Notes: Cells grown on type I hydrated collagen gels require special techniques for sample preparation and processing in order to optimize the removal of all reagents from the collagen matrix and prevent artifactual shrinkage. This method includes cutting out a small block of the collagen gel, postfixation, and transfer to a scintillation vial for further processing. These additional steps ensure that all sides of the block will come in contact with solutions and reduces the possibility of reagent trapping within the collagen matrix. Additionally, in our study the collagen gel and endothelial cells that form a monolayer on the surface are oriented to allow the microtomist greater assurance of cutting true cross sections, thus saving time and increasing reproducibility. The dehydration sequence is also modified, with an increase in the times and additional steps, especially in the higher concentrations of dehydrant.

    Notes: A two-dimensional detector system, designed for the observation and recording of microdiffraction patterns formed in an HB 5 scanning transmission electron microscope (STEM) is described and discussed. Possibilities are described and demonstrated for the simultaneous or successive recording of microdiffraction patterns from regions of diameter 3 å or more, bright- or dark-field STEM images, EELS spectra, secondary electron images, and in-line holograms. Applications of the system have been made to studies of catalyst particles, reflection-mode imaging of bulk surfaces, and image reconstruction from microdiffraction patterns obtained from each point of a STEM image.

    Notes: We studied the penetration of fluorescein isothiocyanate (FITC) T-40 dextran into the paracellular spaces in the tracheal mucosa and its appearance in the blood plasma of guinea pigs exposed to cigarette smoke or (controls) breathing room air. Under general anesthesia, the dextran solution was instilled onto the tracheal surface via a tracheotomy tube, arterial blood was sampled serially for 40 min, and then the trachea was fixed by perfusion or immersion. Examination of the tracheal mucosa with light microscopy, with and without epifluorescence, and transmission electron microscopy, revealed dextran in the paracellular spaces in mucosa from experimental animals but not controls. In all control animals, the levels of the dextran in plasma was below the sensitivity of the assay. By contrast, dextran levels in the plasma from the experimental animals fell within the sensitivity range of the assay and increased in blood at a rate of 0.00125 ± 0.00023 (SE) expressed as a percentage of the instilled dose/min. We conclude that FITC T-40 dextran provides a reliable, fairly simple, fast method for assessing major, but not subtle, changes in paracellular permeability of the tracheal mucosa.

    Notes: Loss of 14C-labeled compounds from bean leaf tissue was monitored during all the stages of routine specimen preparation. No significant differences in extraction were associated with the use of acetone, ethanol, or dioxane as dehydration fluids. Fixation at low temperature increased the loss of label. Prolonged fixation in glutaraldehyde increased the loss, but fixation in osmium solutions for periods as long as 4 hr had no influence on extraction. Buffer rinses and dehydration fluids caused appreciable amounts of label to be extracted. The use of propylene oxide as transition fluid resulted in low extraction. Some embedding media caused the loss of small amounts of labeled compounds, but one of the media tested (LR-white) extracted significant amounts of label.

    Notes: Conventional fixation for thin-section electron microscopy is insufficient to preserve bacterial capsular material, a highly hydrated structure which collapses during dehydration and embedding. Boyles et al. (1984, 1985) have recently introduced a modified aldehyde fixation process in which the addition of a primary amine was found to improve the preservation of actin filaments, the basement membrane material and cell surface glycocalyx of mouse tissues. To the best of our knowledge, the application of diamines to the analysis of bacterial ultrastructure has not been reported in the literature. In the present study, excellent preservation of bacterial capsular material was seen when a diamine (lysine) was used together with glutaraldehyde in conventional embedding procedures.

    Notes: Using an instrument equipped with two electron guns, an electron analyzer, and a Si(Li) diode detector, we developed microanalytical techniques based on inner-shell electron excitations by incident electrons and X-rays, that is, electron energy-loss spectroscopy (EELS) in the reflection mode; electron probe microanalysis (EPMA) and X-ray appearance potential spectroscopy (XAPS); electron-induced Auger electron spectroscopy (e-AES); X-ray photoelectron spectroscopy (XPS), X-ray absorption spectroscopy (XAS); X-ray induced AES (XAES), X-ray fluorescence analysis (XRF), and scanning X-ray radiography (SXR).     Notes: The first stages of crystallization have been investigated for a glass in the system SiO2-Al2O3-Li2O with small amounts of titanium and zirconium as nucleating elements. The studies were performed by electron diffraction and electron microscopy in high-resolution mode. It has been found that the two phases forming the final glass ceramic crystallize successively. The first one gives small crystallites (〈5 nm) randomly dispersed. The lattice plane images in several orientations are in good agreement with TiZrO4 crystal lattice. At 780°C the average crystal size increases with increasing time of the heat treatments from 2.5 nm at 0.5 h to 4.5 nm at 2 h and then remains nearly constant. The crystallization of the second phase, a silicate solid solution with β-quartz structure, occurs after complete crystallization of TiZrO4.

    Notes: The current framework for the quantitative analysis of transmission electron diffraction data from organic microcrystals is discussed. The appropriate formulation depends upon the crystal perfection: solution-grown material tends to be nearly perfect with regular elastic bends whilst the epitaxial samples are usually considerably less perfect with highly irregular bends. It is possible to obtain Bragg intensity data which are near-kinematical and thus amenable to ab initio structure analysis. Thereafter, the further refinement of the model proceeds with a consideration of dynamical and crystal bending effects. If both of these are small it is a good approximation to analyse them independently rather than use the correct description of dynamical scattering from a deformed foil. The analysis of three-dimensional data sets from tilt series is considered. Although the technique has been successfully demonstrated in several instances there are still details of the analysis that need further clarification.

    Notes: For three-dimensional electron microscopical structure research the specimen must be imaged in a tilted position. Specimen tilt is also often needed to achieve an optimal molecular packing orientation. The tilt with respect to the optical axis causes a defocus gradient alongside the imaged area and thus entails the following complications: (1) The phase-contrast transfer function fades for strong defocus; (2) the Fourier coefficients are split; and (3) the signal-to-noise ratio cannot be enhanced by simple averaging. An image procedure with small-spot scanning and simultaneous defocus compensation is proposed which helps to reduce these problems.

    Notes: High-voltage electron microscopy has shown itself advantageous for the study of natural science, including biology, but especially for materials science. The most important advantage for materials science is for in situ experiments about the detailed processes of the phenomena that occur in bulk materials. The present paper is mainly concerned with several types of in situ experiments that have been carried out in the Research Center for Ultra-High Voltage Electron Microscopy, Osaka University. The following subjects have been studied: (a) fundamental problems, such as the conditions necessary for in situ experiments, functional features of specimen treatment devices, and the effects of electron irradiation; (b) the dislocation behavior of crystals under various conditions; (c) high-temperature behavior of refractory materials, mainly ceramic composites; (d) new applications of electron irradiation effects, such as amorphization of crystalline materials and electron-irradiation-induced foreign-atom implantation; (e) environment-matter interaction, mainly chemical amorphization of alloys; and (f) future trends of the in situ experiment, such as combinations with Auger valency electron spectroscopy and high-resolution electron microscopy.

    Notes: Recent studies of high-resolution electron microscopy on the high-Tc superconductors of Y-Ba-Cu-O and Bi-Ca-Sr-Cu-O are presented. The observed images of crystals thinner than 3 nm, taken under conditions that approached the Scherzer defocus condition, directly show the arrangements of cations and oxygen-vacant positions. The results reveal structural characteristics of the atomic scale; this offers important insights into the origin of the high-Tc superconductivity. The usefulness of high-resolution electron microscopy for studying complicated crystal structures is demonstrated for the high-Tc oxides.
