    Description: Marmosets represent an attractive and widely used animal species in biomedical research, and the routine monitoring of female reproductive cycles is often mandatory in the fields of reproductive biology and stem cell research. Today, the established method for the reliable detection of ovulation is the determination of progesterone concentrations from blood samples. This method is based on relatively frequent handling and blood collections; therefore, less invasive alternatives would help to reduce stress on the animals. Here, we investigated whether the core body temperature of marmosets would show a correlation with cycle-dependent hormonal fluctuations, as has been described for humans and other primate species. In particular, the objective was to investigate whether the telemetric recording of core body temperature could replace progesterone measurements as a reliable, less invasive method for the detection of ovulation in these animals. Here we show that the core body temperature parameters in female marmosets were characterized by frequent variations, but they were not related to particular days or phases during the reproductive cycle. Therefore, the recording of core body temperature in our controlled standard experimental setting is not an appropriate method to monitor the reproductive cycle in female marmosets, and cannot replace serum progesterone measurement as a state-of-the-art method.

    Description: Although non-human primates (NHPs) represent only a minor fraction of all animals used in biomedical research, there is a continuous effort to further reduce, refine and replace research with NHPs in accordance with the principles of the three R's. Most of the NHP model species are genetically highly diverse, and significant variation occurs among populations of different geographic origins, particularly in macaques. Since such differences can considerably affect the outcome of biomedical experiments, genotyping represents a promising tool to refine research approaches and to reduce the number of NHPs in biomedical research. Accordingly, the European Primate Network (EUPRIM-Net) developed an anonymous online survey to evaluate possibilities and potential hindrances for the application of genotyping in NHP research. On the one hand, our results point to the importance of genetic variation in NHPs and the need to consider the genetic background for future research approaches. On the other hand, our survey identified several hindrances and limiting factors for the application of genotyping and its incorporation in research, primate husbandry and breeding. We provide some fundamental recommendations on how to meet these challenges and how genotyping can be efficiently used to refine NHP research and to reduce the number of NHPs in biomedical research in the long term.

    Description: Dispersal and grouping patterns form the foundations of social interactions in group-living mammals and are the outcomes of a complex interplay between inbreeding avoidance, kin cooperation and competition, predation pressure and food resource distribution. In species where both sexes disperse, the potential for kin-biased associations would seem limited. In one such species, the western lowland gorilla (WLG), short-term data suggest that female kin associations may be present due to directed local dispersal decisions, but monitoring of groups over longer timescales is needed to better elucidate this pattern. Using autosomal genotyping of 419 faecal samples representing 85 unhabituated gorillas collected non-invasively over 5 years in a 132 km2 section of Loango National Park, Gabon, we investigated the dynamics of WLG group composition, social structure and patterns of dispersal. By revealing two group dissolutions, one group formation and the movement of 13 gorillas between groups, this study demonstrates the utility of genetic analysis as a way to track individuals, groups and population dynamics on a larger scale than when monitoring the behaviour of a limited number of habituated groups or through one-time genetic sampling. Furthermore, we find that females are found in groups containing their female kin more often than expected by chance, suggesting that dispersal may not impede female kin associations in WLGs.

    Description: We observed a putative case of empathy among wild black-fronted titi monkeys (Callicebus nigrifrons) from two different groups (D and R). In over 10 years of behavioural observations of five habituated groups of this species, only low levels of inter-group tolerance have been observed. However, on one day, we encountered the adult male from group D limping (poor hind limb motor coordination) as he travelled alone along the ground. Interestingly, we observed that members of group R did not express any agonistic behaviour towards this neighbouring male and apparently allowed this disabled individual to follow them in the forest for over 5 h. They stayed low in the forest (〈 2 m above the ground) and 〈 10 m horizontally from the individual, and remained in visual contact with him. At the end of the day, this male from group D slept in the sleeping site of group R and was groomed by the adult female of group R. Such tolerance between members of different groups has never been previously observed in this species. Furthermore, group R exposed themselves to increased predation risk by staying close to the ground for protracted periods. The behaviour of group R could be interpreted by as a putative case of empathic responding in this species.
