Notes: A computer graphics system for reconstruction from serial section micrographs was applied to intracellular details of a yeast target cell (Saccharomyces cerevisiae cell) induced by the α factor mating pheromone and was also applied to a periodontal structure of a dog tooth moved orthodontically. In the former, intracellular organelles and a distribution of vesicles could be clearly observed through the cell membrane using the transparent display method in which the smoothing of the reconstructed outer cell membrane surface by computer processing was applied to the transparent display. In the latter case, by cutting through a reconstructed dog tooth and its periodontal tissues, labiolingual and mesiodistal cut surfaces of the tooth and of adjacent alveolar bone could be observed with fine details (232 sections were used).

Notes: Recently, the reliability of field-emission electron guns has increased. In addition, the cost of computer systems for on-line processing has dropped. Hence, we should now consider the use of scanning transmission electron microscopy (STEM) for routine work, especially, in the field of biology where one may expect to utilize digital image processing techniques. An STEM has been constructed, without disturbing the original functions, by converting a commercial scanning electron microscope equipped with a fieldemission gun. The STEM is generally operated at accelerating voltage 30 kV, focal length 7.5 mm, and beam current 1-2 × 10-10 A. Several improvements have been incorporated for removing the effects of vibration, contamination, and stray magnetic fields. Also, an adjustable detector aperture was utilized. The modified instrument was connected to an on-line digital image processing system for utilizing the information obtained from STEM images. The advantages of the modified system were studied from various viewpoints.

Notes: A linear relationship in each of the torsion angle pairs, α-β, β-∊, ∊-ζ, and α-γ, has been found by applying a statistical method based on the concept of circular variates to backbone torsion angle data of helical in yeast tTNAPhe. A series of helical dimer models generated with these relationships have been found to be stereochemically acceptable, and the models also indicate that the backbone unit in the RNA helix is geometrically capable of an oscillatory motion with the distance of about 3.4 Å between adjacent bases. The motion of the backbone unit is analogous to that of a helical spring. The adjacent bases, because of being attached to the backbone, oscillate in a manner similar to the oscillatory dimer model proposed by Davis and Tinoco [Davis, R. C. & Tinoco, I., Jr. (1968) Biopolymers 6, 223-242]. Here, the oscillation of the backbone unit in the RNA helix is discussed in terms of two geometrical quantities: the torsion (τ) and curvature (κ) of the helix. On these lines, a stereochemical model of RNA strand separation is proposed.

Notes: The Hückel-London theory has been applied to the calculation of the “ring currents” octopole hypersusceptibilities of a series of conjugated compounds. Our calculations have been performed using fourth-order Rayleigh-Schrödinger perturbation technique. This is believed to be the first calculation of this kind. It is found that the values of “ring currents” hypersusceptibilities of fulvene and azulene are larger, in absolute magnitude, than the values corresponding, respectively, to benzene and naphthalene. It is found that, in most cases, the “ring currents” hypersusceptibilities are diamagnetic (negative values) for aromatic molecules and paramagnetic (positive values) for pseudoaromatic or antiaromatic compounds.

Notes: We have shown that integrals Irs and Jrs which occur in coupled Hartree-Fock perturbation, on a basis of gauge invariant atomic orbitals with the London approximation and neglect differential overlap, can be reduced, by appropriate transformations, to the overlap integral type. The computational program of Srs, Irs, and Jrs integrals is elaborated for Slater-type atomic orbitals. The process proposed presents a double advantage: it is extended over the entire Periodical Table and does not use the analytical formulas of Mulliken.

Notes: 1,3-Dioxa-2-stannolanes 1 are readlily activated by Lewis bases such as Bu3P and Et3N to give cycloadducts on reaction with carbon disulfide or phenyl isothiocyanate under mild conditions. In particular, bases play a characteristic role in the reaction with carbon disulfide to produce 1,3-dioxolane-2-thiones 2 in good yields, while spiro compounds 3 (1:2 adducts) are predominantly obtained in the absence of bases.

Description / Table of Contents: Die Verdünnungswärme und die Volumenänderung des Systems Polystyrol/Äthylacetat wurden gemessen.
Notes: The heats of dilution and the volume change for the system polystyrene/ethyl acetate were measured. It was shown that the values of the interaction heat parameter as well as those of the excess volume change their positive sign to a negative one at about the same molar mass of the polymer. This is explained in terms of drastic changes of the conformation of the polymer molecules as soon as a certain molar mass is attained.

Notes: The heats of mixing of polyriboadenylic acid (Poly A) and polyribouridylic acid (Poly U) in various proportions by changing the mole ratio n = [Poly U]/[Poly A] in 0,001, 0,1, and 0,5 molar NaCl solutions containing 0,1 molar tris-HCl buffer solution were measured. Constant values for the heats of mixing of the (Poly A+n Poly U) system are found for n ≤ 1 and n ≥ 2. From the results of UV and CD spectra, it may be considered that the mixing of Poly A and n (Poly U) leads to the formation of Poly(A+U) for n ≤ 1, and to that of Poly(A+2U) for n ≥ 2, even at [NaCl] = 0,001 mol/l.The enthalpies of formation of Poly(A+U) and that of Poly(A+2U) depend on the concentration of NaCl. This dependence may be attributed to the differences in conformation of Poly(A+U) in NaCl solution.

Notes: The heat of solution and the osmotic pressure of bovin hemoglobin in aqueous solution have been measured at 298 K using a twin microcalorimeter and an osmometer. The heat of association, ΔH, from calorimetric measurement and the association equilibrium constant, K, from osmometric measurement have been estimated, respectively. Also the thermodynamic quantities of the hemoglobin molecule which associates from 2αβ to the original α2β2 form are calculated. In all systems of the hemoglobin solutions, the value of dissociation equilibrium constant, K', calculated as the reciprocal of K, and that of ΔH seem to be of comparable order to those determined by the other investigators.

Notes: The interaction between DNA and dyes such as acridine orange (1), proflavine (2), and 3,8-diamino-5-ethyl-6-phenylphenanthridinium bromide (3) were studied by means of the heat of mixing and UV spectra. From the results obtained, the thermodynamic quantities of DNA-dye complexes were estimated. It is concluded that the intercalation process forms a more stable binding than the stacking process does. Further, in each process, ΔG of DNA-2 and DNA-3 systems is nearly equal to that of the DNA-1 system, but their  -  ΔH and  -  ΔS values are higher than those of the DNA-1 system. The stability of binding in DNA-2 and DNA-3 systems may be governed largely by ΔH, while in the DNA-1 system ΔS may contribute largely to the stability of binding.
