Abstract: This book focuses on the fundamentals and applications of messenger RNA (mRNA)-based therapeutics and discusses the strengths and key challenges of this emerging class of drugs. In the past 30 years, extensive research and technological development in many areas have contributed to the emergence of in vitro transcribed mRNA as a therapeutic that has now reached clinical testing. Formulations that protect the mRNA from nucleases and accelerate its cellular uptake, combined with improvements to the mRNA molecules themselves, have been critical advancements for mRNAs to become viable therapeutics. Though once regarded as a serious impediment, the transient nature of mRNA technology is now considered a major advantage in making mRNA therapies safe and, ultimately, a potential game changer in the field of medicine. This new book in the RNA Technologies series provides a state-of-the-art overview on the emerging field of mRNA therapeutics covering essential strategies for formulation, delivery, and application. It also reviews the promising role in cancer immunotherapy, respiratory diseases, and chronic HBV infection and discusses RNA vaccines in light of the current COVID-19 pandemic. mRNA-based approaches have great potential to revolutionize molecular biology, cell biology, biomedical research, and medicine. Thus, this handbook is an essential resource for researchers in academia and industry contributing to the development of this new area of therapeutics.

Abstract: Despite the critical importance of the cerebellum in brain functions, the scientific community still lacks effective treatments for most cerebellar ataxias. This book provides a link between the pathogenesis and therapies of cerebellar ataxias while also providing a comprehensive assessment of the preclinical and clinical trials dedicated to cerebellar ataxias over the past 20 years of progress. This is the first book fully dedicated to the trials and therapies of these disorders. It is a truly authoritative and comprehensive reference and comes at a time of major advances in genetic tools and neuroimaging assessments. The coverage begins by laying a foundation of the basic science of the cerebellum and ataxias, proceeds to discuss biomarkers and the tools of trials, offers guidelines on conducting trials, and then explores the full range of therapeutics and their trials, including gene therapy and cell transplantation. The authors are top experts on cerebellar research and the contributing authors have all made seminal contributions in the field.

Abstract: This edited volume provides insights about the molecular virology, pathoenesis of infection and host responses to varicella-zoster virus, which is a medically important human herpesvirus. The chapters not only discuss genetics and molecular evolution of the virus, but also new structure-based knowledge on the functions of viral glycoproteins. Further, the authors illustrate how mechanisms from the virus are used to take over and remodel host cells during pathogeneis. These mechanisms also enable viral modulation of host cells and evasion of the host immune response. Part of these evasion and survival strategies, is the reaction to triggered host cell responses like autophagy, the stress response, modulation of apoptosis and other cell death pathways. This hands-on knowledge is crucial for students and clinicians in Virology, Immunology, and Infectious Diseases. Abstract: This book reviews a novel and exciting field of cellular and molecular biology called epitranscriptomics, which focuses on changes in an organism’s cells resulting from the posttranscriptional modification of cellular RNA. RNA-binding proteins (RBPs) play a crucial role in these posttranscriptional modifications and also support several cellular processes necessary for maintaining RNA homeostasis. Exploring the mechanisms underlying RNA modifications and RBP function is an emerging area of biomedical research, taking the study of gene regulation a step beyond epigenetics. This book reveals that the RNA molecule is not just an information-carrying molecule with some secondary structures. Accordingly, how RNA is modified, regulated, packaged, and controlled is an important aspect. Leading experts address questions such as where the over 170 distinct posttranscriptional RNA modifications are located on the genome, what percentage of mRNAs and noncoding RNAs these modifications include, and how an RNA modification impacts a person’s biology. In closing, the book reviews the role of RNA modifications and RBPs in a variety of diseases and their pathogenesis. Addressing some of the most exciting challenges in epitranscriptomics, this book provides a valuable and engaging resource for researchers in academia and industry studying the phenomena of RNA modification.

Description / Table of Contents: Ancillary Benefits and Development Co-effects -- Conceptual and Theoretical Approaches for the Analysis of Ancillary Benefits -- Ancillary Benefits in Different Sectors and in Adaptation to Climate Change -- Climate Actions in Urban Areas and their Ancillary Benefits. .
Abstract: This volume presents new developments in the research on ancillary benefits. Twenty years after the influential OECD report on ancillary benefits, the authors discuss theoretical innovations and offer new empirical findings on various ancillary effects in different world regions. Covering topics such as ancillary health effects associated with reduced air pollution, the influence of ancillary benefits on international cooperation on climate protection, co-effects of carbon capture and storage, ancillary effects of adaptation to climate change, multi-criteria decision analysis covering multiple effects of climate protection actions, and the analysis of primary and ancillary effects within an impure public goods framework, it provides starting points for further research on integrated climate policies seeking to address a range of policy objectives simultaneously.
