    Notes: Small-angle neutron scattering (SANS) measurements were carried out on two series of ion-exchanged charged polystyrene latex (average particle diameters: 340 and 380 Å) dispersions in deuterated water [88%(v/v) D2O] with particle volume fractions between 0.0075 and 0.074. Absolute scattering intensity profiles obtained were fitted to a model based on generalized one-component macroion theory to evaluate average particle diameter with the polydispersity and effective surface charge. The particle size parameters computed agreed well with electron microscopy data. However, effective surface charges obtained were only 6–7% of that determined by conductometric titration.

    Notes: Small-angle X-ray scattering (SAXS) and small-angle neutron scattering (SANS) techniques have been used to confirm and determine the size and shape of cylindrical micelles formed by a comb-shaped copolymer, poly(1-octadecene-co-maleic anhydride), abbreviated as PODMA, in aqueous solution. Upon full neutralization by the addition of alkaline hydroxide, PODMA becomes a polyelectrolyte carrying two negative charge per repeating unit. SANS measurement gives the dimension of the cylinder with a radius of 20 Å, and a length of 110 Å in D2O solutions with Cs as counterion. SAXS measurements were made for a series of 1% PODMA solutions fully neutralized by CsOH. Linear charge density at the micellar surface was varied by adding an ionic surfactant, sodium dodecyl sulfate (SDS). The dimensionless linear charge density parameter, ξ ≡ lB/b, where lB is the Bjerrum length (lB = 7.15 Å for water solvent at 295 K) and b is the linear spacing between unit charges on the micellar surface, varies from 32.96 to 67.21. Since the electron densities of the micellar core and the aqueous solvent are nearly equal, the SAXS intensity distribution function is a Bessel transform of the Cs counterion distribution around the cylindrical micelles. Comparison is made between the results obtained from SAXS intensity distribution analyses and the prediction of Manning's condensation theory.

    Notes: Abstract Neuromuscular preparations from third instar larvae of Drosophila are not well-maintained in commonly used physiological solutions: vacuoles form in the muscle fibers, and membrane potential declines. These problems may result from the Na∶K ratio and total divalent cation content of these physiological solutions being quite different from those of haemolymph. Accordingly haemolymph-like solutions, based upon ion measurements of major cations, were developed and tested. Haemolymph-like solutions maintained the membrane potential at a relatively constant level, and prolonged the physiological life of the preparations. Synaptic transmission was well-maintained in haemolymph-like solutions, but the excitatory synaptic potentials had a slower time course and summated more effectively with repetitive stimulation, than in standard Drosophila solutions. Voltage-clamp experiments suggest that these effects are linked to more pronounced activation of muscle fiber membrane conductances in standard solutions, rather than to differences in passive muscle membrane properties or changes in postsynaptic receptor channel kinetics. Calcium dependence of transmitter release was steep in both standard and haemolymph-like solutions, but higher external calcium concentrations were required for a given level of release in haemolymph-like solutions. Thus, haemolymph-like solutions allow for prolonged, stable recording of synaptic transmission.

    Notes: Abstract Bang-sensitive mutants of Drosophila melano gaster (bas 1, bssMW1, eas2, tko25t) display seizure followed by paralysis when subjected to mechanical shock. However, no physiological or biochemical defect has been found to be common to all of these mutants. In order to observe the effects of bang-sensitive mutations upon an identified neuron, and to study the nature of mechanically induced paralysis, we examined the response of a mechanosensory neuron in these mutants. In each single mutant and the double mutant bas 1 bssMW1, the frequency of action potentials in response to a bristle displacement was reduced. This is the first demonstration of a physiological defect common to several of the bang-sensitive mutations. Adaptation of spike frequency, cumulative adaptation to repeated stimulation (fatigue) and the time course of recovery from adaptation were also examined. Recovery from adaptation to a conditioning stimulus was examined in two mutants (bas 1 and bss MW1), and initial recovery from adaptation was greater in both mutants. Quantification of receptor potentials was complicated by variability inherent in extracellular recording conditions, but examination of the waveform and range of amplitudes did not indicate clear mutant defects. Therefore the differences observed in the spike response may be due to an alteration of the transfer from receptor potentials to action potential production. DNA sequence analysis of tko and eas has indicated that they encode apparently unrelated biochemical products. Our results suggest that these biochemical lesions lead to a common physiological defect in mechanoreceptors. Although this defect does not provide a straightforward explanation for bang sensitivity, the altered cellular process may lead to bang sensitivity through its action in different parts of the nervous system.

    Notes: In sections the protoplasm of the spinning gland of Platyphylax designatus Walker appears to be a syncytium with large, branching nuclei, which contain both nucleoli and chromatin granules embedded in a delicate linin reticulum.In the normal gland the nucleus is sharply marked off by a nuclear membrane from the granular, homogeneous cytoplasm. The Golgi apparatus appears as rings, loops, or comma-like structures, evenly distributed and without orientation to the nucleus or lumen of the gland.The nucleoli first increase both in number and size within the nucleus, then migrate out into the cytoplasm, where they undergo further growth, and finally are dissolved and passed out into the lumen of the gland as liquid secretion.Small vacuoles appear near the periphery of the gland in early periods of activity.In glands active for forty-eight hours all stages of the secretory process may be seen.In glands active for longer periods only vacuoles are present which we interpret as the remains of the secretory inclusions. There is a progressive decrease in the nuclear content of the nucleus.Throughout the activity of the gland the Golgi apparatus changes little from the normal condition, both in form and distribution. As activity progresses, the bodies become smaller and slightly more dispersed. No relation of Golgi apparatus to the secretory phenomenon is apparent.
