    Notes: Abstract Hypoxanthine-guanine-phosphoribosyltransferase (HGPRT; EC 2.4.2.8) was determined as an enzyme following an ordered bireaction in the presence of substrate inhibition due to hypoxanthine. This kind of inhibition has not been postulated for HGPRT so far. The mechanism and the kinetic constants of the reaction of HGPRT from Saccharomyces cerevisiae were investigated by initial velocity studies involving a non-linear regression analysis. In addition, two kinds of experimental designs were compared: the variation of hypoxanthine concentrations over wide ranges at different of fixed levels of 5-phosphoribosyl-1-pyrophosphate, and the use of five appropriate sets of experimental conditions each characterized by different hypoxanthine and 5-phosphoribosyl-1-pyrophosphate concentrations. Both experimental designs were consistent with an ordered bi bi mechanism including a dead-end-complex between the enzyme and hypoxanthine.

    Notes: Cemented layers predominantly consisting of gels/poorly crystalline mineral phases havebeen formed as a consequence of mineral weathering in sulfidic tailings near Freiberg, Saxony,Germany. These layers function as natural attenuation barrier for toxic compounds and reduceoxidation and erosion processes of tailings surfaces. Quantitative molecular biological andcultivation methods were applied to investigate the role of microorganisms for mineral weatheringand cemented layer formation. High resolution depth profiles of numbers of microorganismsshowed maximal cell numbers in the oxidation zone where cemented layers had been formed.Highest total cell numbers of 〉109 cells g-1 dry weight (dw) were detected by SybrGreen directcounting. Using quantitative real-time PCR (Q-PCR) between 107 and 109 Bacteria g-1 dw and up to108 Archaea g-1 dw were determined. As well high numbers of cultivable and living Bacteria couldbe detected by MPN (most probable number) for Fe(II)- and S-oxidizers and CARD-FISH(catalyzed reporter deposition - fluorescence in situ hybridization). Overall, the high numbers ofmicroorganisms determined with various quantification techniques argue for a significant role ofmicroorganisms in cemented layer formation due to microbial mineral weathering. It ishypothesized that EPS (extracellular polymeric substances) mediate the formation of secondarymineral phases

    Notes: The Gaussian-2 (G2) theoretical procedure, based on ab initio molecular orbital theory, is used to calculate the energies of CH2OH, CH3O, and their cations. The resulting G2 adiabatic ionization potential for CH3O of 10.78 eV supports the new value of 10.726±0.008 eV reported by Ruscic and Berkowitz [J. Chem. Phys. 95, xxxx (1991)] from a photoionization study. A previous photoelectron result of 7.37±0.03 eV is probably an incorrect assignment. The G2 ionization potential for CH2OH is 7.45 eV, consistent with the experimental value of 7.55 eV. The calculated O–H and C–H bond dissociation energies of CH3OH are 105.0 and 96.2 kcal/mol, respectively. The results suggest that the D0(H–CH2OH) from kinetics measurements may be too low.

    Notes: Abstract The vitamin requirements of a strain of Candida shehatae for the fermentation of d-xylose was determined using a statistical procedure with a 23 factorial design. Biotin as well as thiamine exerted a dramatic stimulatory effect on the rate of ethanol production, coupled with a significant improvement in the ethanol yield. The greatest enhancement of the fermentation was found in the presence of both these vitamins. Pyridoxine exerted only a minor effect, but was essential for complete substrate utilization in the absence of either biotin or thiamine. Only biotin caused a significant increase in the growth rate.

    Notes: Abstract The cellular long-chain fatty acids of 32 strains representing 10 species of the genus Kluyveromyces were extracted by saponification and analyzed as methyl esters by gas chromatography. The Kluyveromyces strains were characterized by the presence of palmitate, palmitoleate, oleate, and linoleic acid as the major fatty acids. These strains were divided into 3 groups on the basis of fatty acid content. The first group was characterized by a high percentage of linolenic acid, the second group of a lower percentage and the third group by the absence of linolenic acid.

    Notes: Much of the evaluative research on groupware in organizations sofar has been preoccupied with the role of groupware as a new interactionmedium to replace or extend face-to-face communication in groups. It hasfocused on gains and losses from a group interaction point of view,typically disregarding the impact of other functions such as groupaccess, and contribution to information concerning the organization.Attempts to bridge this gap with an action research into the effects ofthe introduction of an asynchronous groupware system to support businessprocess improvement groups in a service company based in Brazil. Theresearch suggests that improvements in business redesign efficiency andeffectiveness can be attained not only from asynchronous groupwaresupport to group communication, but also from public sharing ofhistorical information about previous business process improvementefforts, and by providing a repository of information about businessprocesses which could be candidates for improvement. Concludes with theproposal of an explanatory model, describing the relationship betweenthe introduction of technology, its integration with a business processimprovement meta-process, and its effects on the efficiency andeffectiveness of that meta-process.

    Notes: Describes an action research study of a computer-mediated business process redesign (BPR) group in a New Zealand university. The BPR group used an integrated BPR framework, which comprises a group process methodology, called MetaProi, and an asynchronous groupware tool. BPR group members were from two different departments and successfully redesigned two course-related processes. Reveals some possible effects of computer mediation on BPR groups, namely: lower demand for leadership skills, much lower overall running cost, and much lower degree of interaction. No impact on the group effectiveness was observed. Also indicates that computer mediation lowers barriers to, and, in turn, fosters more interdepartmental communication. This favours the occurrence of new BPR groups involving different departments. On the other hand, indicates that those groups lead to more threats to management, which can lead to lack of support from managers for future BPR groups. Finally, suggests that strategic BPR groups, as opposed to those dealing with local operational issues, can better benefit from computer mediation when this is combined with face-to-face and other types of oral interaction.

Business process data were collected in the context of action research projects where the researcher was involved in organizational development activities. Suggests that business processes tend to cut across different departments owing to a contemporary phenomenon - the specialization of knowledge. This leads to the specialization of work with the multiplication of functions and departments in organizations. Also suggests that 70 per cent or more of the product flow through business processes in organizations is made up of information.
