    Notes: A new method for the visualization of proteolytic activity in cell culture supernatant from Pseudomonas lundensis after sodium dodecyl sulfate (SDS) - gel electrophoresis is described. Following conventional electrophoresis, the gel is washed in a methanol-containing buffer to facilitate partial removal of SDS. After incubation with 0.5% casein the gel is stained for protein with Coomassie Brilliant Blue R-250. Bands with proteolytic activity appear as clear areas in the gel against a blue-stained background. Molecular weight standards electrophoresed in the same gel stain more intensely than the background and allow determination of the molecular weights of the proteolytic components. The sensitivity of post-electrophoretic reactivation in SDS-gels was determined using trypsin as standard. A slight modification of the technique allowed detection of proteolytic activity in nondenaturing and in isoelectric focusing gels.

    Notes: Luminol-dependent chemiluminescence of normal human monocytes activated by zymosan is demonstrated to be inhibited by tetrahydrobiopterin in a concentration-dependent manner. The reduced pterins tetrahydrobiopterin, dihydrobiopterin, and dihydroneopterin are all shown to be readily oxidized by the hydroxyl radical.The susceptibility of reduced pterins to free radical attack may explain the inhibition of chemiluminescence observed and an additional role of reduced pterins as free radical scavengers in tissues is considered.

    Notes: The vegetation eaten by animals on large areas of several continents is deficient in phosphate and deleterious effects on physiology, particularly reproduction, ensue. Records on bone chewing behaviour by both pastoral andwild game animals extend over two centuries. In laboratory investigation of this apt behaviour it has been shown that the appetite for bones is innate and specific and cued predominantly by olfactory stimuli. It is suppressed by rapidly increasing the plasma phosphate concentration to normal but not influenced by increasing the phosphate concentration in cerebrospinal fluid. The central organization of this genetically programmed behaviour appears to differ from systems subserving thirst and sodium appetite.

    Notes: Elastic wave radiation from an equivalent cylindrical cavity due to an underground explosion is considered in detail. Both static and steady state dynamic finite element models are used to investigate the surface displacements due to the interaction of such wave radiation with both the free surface topography and stope formation. For a blast 430 m below the surface the models predict a negligible displacement effect due to the irregular topography but a significant effect due to the stope. A comparison between the models and a site experiment verified the cylindrical wave nature of the problem. Due to experimental uncertainties however it was difficult to clearly detect the surface displacement effect due to the large underground open stope.

    Notes: The dynamic response due to a spherical source of radius a embedded in an elastic and viscoelastic full-space is investigated at a distance R from the source. Previous solutions to the elastic case are extended to incorporate realistic source pressure functions. The elastic solution is then cast in a scale independent form in order to generalize the application. The results show that the near-field of the spherical source may be defined by R/a 〈 5. For this region the particle velocity and displacement decrease as R-2, and the risetime decreases as R-1. However. in the far-field region (R/a 〉 5) the particle velocity and displacement decrease as R-1, and the risetime is independent of R. A non-constant Q model is developed to model viscoelastic attenuation and a complete analytical solution for wave propagation is obtained by cascading the separate mechanisms of geometric attenuation and viscoelastic attenuation. A comparison of our analytical model with the results of dynamic finite element modelling shows excellent agreement. This suggests that the method of cascading the separate transfer functions is a valid approach for wave propagation in viscoelastic media.

    Notes: High-performance liquid chromatography coupled with graphite furnace atomic absorption spectroscopy (HPLC—GF AA) gives element-specific detection of environmental samples containing trace amounts of organotin or organolead species. The direct GF AA of organotin and organolead species is subject to errors arising primarily from loss of analyte prior to atomization, probably through the formation of refractory carbides and of compounds or complexes that are volatile at low temperatures. Examples abound in the literature of signal suppression in the GF AA of organometallic species in environmental samples, and several furnace tube modifications have been developed to overcome this suppression. Here, the analyte and a modifier are co-pipetted into a conventional furnace tube, from either a solution of analyte or an HPLC effluent. Oxides of transition metals (e.g. chromium, manganese, or tungsten) are shown to enhance both tin and lead signals, whereas chlorides do not, suggesting the low-temperature formation of relatively involatile metal oxides or volatile metal chlorides, respectively. In the absence of modifier, GF AA signal intensities decrease consecutively for equal quantities of mono-, di-, tri- and tetra-butyltin species, but are nearly equal for the first three in the presence of complexing dichromate (Cr2O72-). The lesser signal increase for tetrabutyltin indicates a dissimilar low-temperature complexation chemistry for the fully ligated neutral organometal to that for the ligated ions. similar results are demonstrated in post-column addition of a matrix modifier to effluent containing either organotin or organolead species.

    Notes: The reactivity of six monoclonal antibodies with fragments of fibronectin produced with trypsin, chymotrypsin, and Staphylococcus aureus V8 protease is described. All these antibodies reacted with fragments derived from the C-terminal one-third of fibronectin. This region probably contains sites for the binding of fibronectin to cells, and to heparin and may also contain active sites for the reattachment, spreading, and alignment of transformed cells. Analysis of the reactivities of different sets of proteolytic fragments with the antibodies and with other ligands (eg. heparin) allows one to determine overlaps between the fragments and to locate the positions of the different binding sites for antibodies and ligands. One of the antibodies has allowed us to identify a site of structural difference between cellular and plasma fibronectins from hamsters. The site recognized by this antibody is located near to, but not at, the C-terminal end and docs not involve carbohydrate groups. Because of its internal location in fibronectin, this difference suggests that there are probably different genes for cellular and plasma fibronectin. These monoclonal antibodies should be useful for further probing the functions present in the C-terminal regions of fibronectin and for determining their locations.

    Notes: An assay for the quantitative analysis of six biologically important prostanoids based on combined gas chromatography negative ion chemical ionization mass spectrometry has been developed. Prostanoids were extracted from biological fluids by liquid chromatography on Sep-Pak cartridges and converted to pentafluorobenzyl ester derivatives. Samples were injected on capillary columan by the splitless technique and injections were made in a high boiling hydrocarbon solvent (n-dodecane) in order to minimize chromatographic run times. Quantification was carried out using selected ion monitoring of the appropriate [M-pentafluorobenzyl]- anion. The assay has been used for profiling cyclooxygenase metabolites of arachidonic acid in guinea pig lung perfusate after induction of anaphylaxis and platelet rich plasma after collagen-stimulated aggregation.

    Notes: The surface areas of dyed and undyed secondary cellulose acetate fibers have been determined using low-temperature nitrogen gas adsorption by the method of Brunauer, Emmett, and Teller. The results confirmed earlier work using p-nitrophenol, in which it was shown that there is a marked change of surface area of dyed fiber in the region corresponding to a theoretical monolayer adsorption of the dye.

    Notes: The effect of several variables on the adsorption rate of four dyestuffs onto chitin was studied. A model is proposed enabling the film mass transfer coefficients to be determined. The coefficients were independent of initial dye concentration, chitin mass, chitin particle size, and temperature; a slight dependence with agitation was obtained. The film mass transfer coefficients at 400 rpm were 2.8×10-3, 2.9×10-3, 3.9×10-3, and 0.9×10-3 cm/s for Acid Blue 25, Acid Blue 158, Mordant Yellow 5, and Direct Red 85, respectively.
