    Relationship between the number and function of human ribosomal genes (1988)

    Notes: Summary The relative number of ribosomal RNA genes of the acrocentric chromosomes in one individual was measured by counting grains after in situ hybridization of 3H-labeled human 18S rDNA to fixed metaphase chromosomes. The relative amount of ribosomal RNA gene activity of each of the same chromosomes was estimated by determining the frequency with which the chromosome's nucleolus organizer region (NOR) was silver stained, the size of the silver-stained region, and how often the chromosome was found in satellite association. Results were similar in phytohemagglutinin-stimulated T-lymphocytes, Epstein-Barr virus transformed lymphoblasts, and fibroblasts. One chromosome 21 had few gene copies and low activity. One chromosome 22 had many gene copies but low activity. Both chromosomes 14 had few gene copies but high activity. The level of expression that can be achieved by rRNA gene clusters can, therefore, be determined by factors other than the number of gene copies.

    Notes: We report the first observation of the self-electro-optic effect in InGaAs/InP multiple quantum wells, grown by organometallic vapor phase epitaxy. Clear bistability and switching are observed over a range of 40 nm around 1.61 μm with 20–30 V bias. We demonstrate the operation of a modulation convertor, which converts a modulation from a carrier at 1.6 μm onto a carrier at 0.85 μm and vice versa.

    Notes: Summary A cloned alphoid sequence, p82H, hybridizes in situ to the centromere of every human chromosome. After washing under stringent conditions, no more than 8% of the grains are located on any specific chromosome. p82H thus differs from other centromeric sequences which are reported to be chromosome specific, because it detects sequences that are conserved among the chromosomes. Two experimental approaches show that the p82H sequences are closely associated with the centromere. First, p82H remains with the relocated centromeres in an inv(19) and an inv(6) chromosome. Second, p82H hybridizes at the centromere but not to the centromeric heterochromatin of chromosomes 1, 9 and 16 that have elongated 1qh, 9qh and 16qh regions produced by short growth in 5-azacytidine. The only noncentromeric site of hybridization is at the distal end of the 9qh region.

    Notes: Abstract A survey of the terrestrial tardigrades inhabiting growths of algae, lichens and mosses in the Vestfold Hills, Antarctica, was carried out at 11 and 35 sites during the austral summers of 1980 and 1982, respectively. In all, 24 species of plants were collected from which four genera and four species of Tardigrada were recovered. A key to the tardigrades of the area is presented. The distribution and associational patterns of the tardigrades are discussed in the context of other studies of antarctic Tardigrada.

    Notes: Abstract The macroinvertebrate community composition was compared in two Alaskan streams (USA) for numeric and species constancy during the ice-free period from 1981 to 1983. Imnavait Creek is a first order arctic stream (60° 39′ N, 149° 21′ W) draining upland tundra in the foothills of the Brooks Range. Caribou-Poker Creek is a 4th order subarctic stream (65° 08′ N, 147° 28′ W) draining the taiga forest north of Fairbanks, Alaska. The aquatic insect larvae and other macroinvertebrates were sampled with drift nets and Hess bottom samplers for four periods, each 1 week long in the ice free season of three years. We found 112 species in the arctic stream and 138 species in the subarctic stream in a chironomid-dominated community. In any sample period the communities contained 51–60 species in the arctic and 49–92 species in the subarctic. Between the four sample periods on average 39% and 50% of the species were present in two sequential samples in the arctic and subarctic stream, respectively. New immigrants, never before found in the system, averaged 37% and 31% of the community, respectively. These systems are exposed to several intermediate disturbances: prolonged and variable freeze-up, extreme variation in discharge, wide diel and seasonal changes in temperature, and erosion by frazil and anchor ice. The dipterans that compose the most numerous and variable taxa must have variable diapause, ability to grow in cold waters, and good dispersal powers, even migrating across drainages in the arctic. Much of the seasonal dominance pattern appears therefore to be stochastic.

    Notes: Magnetic field systems were added to existing electric field exposure apparatuses for exposing cell suspensions in vitro and small animals in vivo. Two horizontally oriented, rectangular coils, stacked one directly above the other, have opposite electric currents. This configuration minimizes leakage fields and allows sham- and field-exposure systems to be placed in the same room or incubator. For the in vitro system, copper plates formed the loop-pair, with up to 900 A supplied by a 180:1 transformer. Electric fields were supplied via electrodes at the ends of cell-culture tubes, eight of which can be accommodated by each exposure system. Two complete systems are situated in an incubator to allow simultaneous sham and field exposure up to 1 mT. For the in vivo system, four pairs of 0.8 × 2.7-m coils made of copper bus bar are employed. This arrangement is energized from the power grid via a 30:1 transformer; horizontal magnetic flux densities up to 1 mT can be generated. Pairs of electrode plates spaced 30.5 cm apart provide electric field exposure of up to 130 kV/m. Four systems with a capacity of 48 rats each are located in one room. For both the in vitro and in vivo systems, magnetic exposure fields are uniform to within ± 2.5%, and sham levels are at least 2,500-fold lower than exposure levels. Potential confounding factors, such as heating and vibration, were examined and found to be minimal.

    Notes: This study describes a differential frequency of spontaneous fusion between metastatic and nonmetastatic subpopulations derived from a single mouse mammary tumor. Subpopulations 66, 66cl4 (a variant of 66 which is resistant to both thioguanine and ouabain), 410.4, and 44FTO (a thioguanine-resistant, ouabain-resistant derivative of 410.4) spontaneously metastasize from subcutaneous and mammary fatpad sites. Subpopulations 168, 168FARO (a diaminopurine-resistant, ouabain-resistant derivative of 168), 67, 68H, and 410 do not. The ability of these subpopulation lines to fuse spontaneously in vitro was determined after coculturing a drug-resistant line with a wild-type line in nonselective media. After 16-20 h of coculture, cells were plated in the appropriate media to select for fusion products - either HAT (hypoxanthine, aminopterin, thymidine) plus ouabain or AA (alanosine, adenine) plus ouabain - to determine the number of colony-forming cells (fusion products) present per 104 cells plated. When both subpopulations of the pair in the fusion mixture were metastatic, a significantly greater number of fusion products was recovered than if one or both of the subpopulations in the fusion mixture was nonmetastatic, with one exception: line 410 readily fused with both 66cl4 and 44FTO. Subline 410 was highly metastatic when originally isolated but lost its metastatic competence after a brief time in tissue culture.
