Notes: Abstract A method for long-term plant regeneration of Phaseolus coccineus L, is described. Shoot-tips and cotyledonary nodes cultured on a Murashige and Skoog medium supplemented with N6-benzylaminopurine, 10 μM, and α-naphthaleneacetic acid, 1μM, formed multiple bud-shoots. These shoots were transferred to medium containing BAP 1 μM, NAA 0.1 μM, and gibberellic acid 3 μM to promote shoot growth and further shoot multiplication. Rooting was achieved in medium with 11 μM indole-3-acetic acid. Rooted plants grew to maturity and were fertile. Cultures have maintained their ability to regenerate plants for more than two years. A sample of 30 regenerated plants (R0) was tested for chromosome number, all of them being diploid; seven isozymatic systems were electrophpretically analyzed in 82 R0 regenerated plants. No differences were observed in their electrophoretic patterns in comparison with those shown by seedlings. Histological studies revealed the origin of buds from calluses via organogenesis.

Notes: Abstract The effect of an allelopathic compound, diacetyl-piquerol on the H+ -ATPase activity of the microsomal fraction from the radicles of a common weedIpomoea purpurea was studied. The diacetyl-piquerol inhibited the germination and radicle growth fromI. purpurea; the radicle growth was increasingly inhibited (10% to 100%) as piquerol concentrations were raised (10 μM to 1000 μM). The H+-ATPase activity was inhibited (48%) by 500 μM diacetyl-piquerol, and this inhibition was higher in plasma membrane ATPase (67.2%) than in tonoplast membrane ATPase (31.4%). Additional studies of the precise physiological mechanisms of interference caused by allelopathic compounds are needed.

Notes: Abstract A resin glycoside mixture isolated fromIpomoea tricolor inhibited radicle growth ofEchinochloa crusgalli. The effect of the resin was tested on the activity of the plasma membrane H+-ATPase fromE. crusgalli. For this purpose, plasma membrane vesicles were purified by the method of aqueous two-phase partitioning. The resin glycoside inhibited by 30% the activity of the plasma membrane ATPase. The same result was obtained with the purified main component of the resin. This indicates that the plasma membrane ATPase can be one of the cellular targets of the resin. Hence it is possible that the mechanism of action of the resin involves an inhibition of the plasma membrane ATPase.

Notes: Summary The effect of colchicine upon the embryos of barley (Hordeum vulgare L.) culturedin vitro has been studied. Different concentrations of the drug as well as different times of culture in its presence have been tested, in order to ascertain the optimal conditions for the induction of non diploid cells. Tetraploid cells were always infrequent after the treatments, reaching the higher values during the first days of culture; aneuploid cells being frequent in some cases.
