    Notes: Sunflower lines RHA-274, HA-61 and RHA-325 were studied for their resistance to race 330 of downy mildew (Plasmopara halstedii). The same inbred line, with normal (HA-89) or sterile cytoplasm (cmsHA-89) was used in all the crosses as susceptible parent, and, in each cross, only one genotype of the resistant parent was studied. The resistant-to-susceptible ratios obtained in the BC1 and F2 progenies from the crosses of the lines RHA-274 and HA-61 to cmsHA-89 and HA-89, respectively, suggested that, in each resistant line, two dominant genes are responsible for resistance to this downy mildew race. One of the genes (A) is epistatic to the other (B), and the recessive allele b in homozygosity is also epistatic to aa, with plants carrying aabb genotypes being resistant. Resistance to race 330 seemed to be controlled by two complementary genes in the sunflower inbred line RHA-325, the dominant allele of one of them being present in cmsHA-89. In the genotypes HA-89 or cmsHA-89, the existence of genes that modify the expected segregations following the crosses with resistant parents is proposed. It is concluded that, although major genes have been described as responsible for monogenic resistance to downy mildew, other types of regulation of this character, such as complementarity and epistatic relationships, do occur.

    Notes: Abstract A method for long-term plant regeneration of Phaseolus coccineus L, is described. Shoot-tips and cotyledonary nodes cultured on a Murashige and Skoog medium supplemented with N6-benzylaminopurine, 10 μM, and α-naphthaleneacetic acid, 1μM, formed multiple bud-shoots. These shoots were transferred to medium containing BAP 1 μM, NAA 0.1 μM, and gibberellic acid 3 μM to promote shoot growth and further shoot multiplication. Rooting was achieved in medium with 11 μM indole-3-acetic acid. Rooted plants grew to maturity and were fertile. Cultures have maintained their ability to regenerate plants for more than two years. A sample of 30 regenerated plants (R0) was tested for chromosome number, all of them being diploid; seven isozymatic systems were electrophpretically analyzed in 82 R0 regenerated plants. No differences were observed in their electrophoretic patterns in comparison with those shown by seedlings. Histological studies revealed the origin of buds from calluses via organogenesis.

    Notes: Abstract The effect of benzylaminopurine (BAP) on the formation of roots from lentil shoots regenerated on media containing BAP was studied. Seedling shoot tips, first nodes and bractlets, and immature seeds cultured on the initiation media containing 2.25 or 0.225 mg/l of BAP regenerated multiple bud shoots. The regenerated shoots formed roots in percentages ranging from 4.6 to 39.9% on a rooting medium (R medium) containing 2 mg/l of indoleacetic acid. Rooting success on R medium depended upon the cytokinin used in the initiation media, its concentration, and the time elapsed during shoot formation on these media prior to transplanting regenerated shoots to R medium. In vivo study of root growth of lentil seedlings demonstrated the strong inhibitory effect of BAP on root growth reflected in a drastic reduction of the mitotic index of the root meristem.

    Notes: Abstract In vitro culture of immature embryo and young leaf tissues was carried out with five cultivars of barley, Hordeum vulgare. Two cultivars (Albacete and Porthos) responded poorly from both types of explants, while the three others (Dissa, Golden Promise and Ingrid) produced a high frequency of embryogenic callus from these explants (25–60%). For ‘Dissa’ and ‘Ingrid’, young leaf explants were slightly better than immature embryo explants for embryogenic callus induction, while immature embryo cultures of ‘Golden Promise’ responded better than young leaf explants. Thus, there appears to be a significant genotype × explant interaction in the initiation of embryogenic callus in barley. Some phenotypic variants were detected among the regenerated plants of ‘Golden Promise’ and ‘Ingrid’, most originating by epigenetic changes. Only in one case was the variant phenotype heritable, probably due to a mutation in the chloroplast DNA. Mitotic alteractions were not detected. Consequently, somaclonal variation did not appear to be a very frequent event in plants regenerated from 1- to 6- month-old cultures of barley.

    Notes: Summary A study of the chromosome numbers of calluses obtained fromin vitro cultured stem nodes of barley over a period of time was carried out. The cells in division in the calluses were found only in growth centres (g.c.) which were independent of one another and easily isolated. At the beginning of the observations, the cell dividing population showed the diploid chromosome number. Tetraploid dividing cells appeared in the 5-month-old calluses and their frequency increased gradually. After 36 months of culture, the frequency of diploid and tetraploid cells was fixed at approximately 50%. The histological observation showed that in the 2-month-old cultures, the calluses were being formed from the epidermis as well as from the stem cortical parenchyma. The medullar parenchyma of the stem was not involved in the callus formation until 5 months of culture. Thus, it was possible to correlate the increase of the tetraploid dividing cell population with the formation of the calluses from the medullar parenchyma.

    Notes: Summary The effect of colchicine upon the embryos of barley (Hordeum vulgare L.) culturedin vitro has been studied. Different concentrations of the drug as well as different times of culture in its presence have been tested, in order to ascertain the optimal conditions for the induction of non diploid cells. Tetraploid cells were always infrequent after the treatments, reaching the higher values during the first days of culture; aneuploid cells being frequent in some cases.

    Notes: Summary Over a period of time a comparative study of the evolution of the different number of nuclei as well as the different chromosome number in the cells of severalin vitro tissue cultures of barley (Hordeum vulgare L.) was carried out. The cultures were obtained by culturing embryos in a control culture medium or in a media supplemented with 0.5 g/1 or 1 g/1 of caffeine, and thereafter transferred to the control medium. The drug effect was noticeable by the increase of polynucleated cells during the first days in culture, but the nuclei tended to fuse quickly, becoming polyploid mononucleated cells. Thus, a similar percentage of polynucleated cells is reached after a few days in both control and treated cultures. From the study of the chromosome number of the cells in the different cultures, it can be deduced that the predominance of the diploid class during the time observed depends upon the frequency of diploid cells at the time of starting the culture. When this frequency was higher than 40% the diploid cells constituted the class most frequent at the end of our observations, while the tetraploid cells were the predominant ones if the diploid initial frequency was lower.
