    Description: The project of Age, growth, feeding regime and fecundity of Kutum (Rutilus frisii Kutum) survey started in south of the Caspian Sea from 2006 to 2007. The aim of project was related to determination of length maturity (Lm50%), identification of feeding animals, and investigation some of biological aspects uncluded in sex ratio, growth and age structure in southern of the Caspian Sea. Total 2706 specimens of Kutum were surveyed. The age structure of Kutum has nine age groups that included in 1, 2, 3, 4, 5, 6, 7, 8, and 9 years. The result showed that maximum frequency was about 39.3 % and were belong to 4 years and minimum frequency which was about o.1 % were belong to 9 years. The maximum and minimum fork length and weight were 58 and 21cm with mean (±SD) 38.4 (± 6.4) cm; 2450 and 104g with mean (±SD) 784.2 (± 383.30) g , respectively. There are significant different between fork length and weight in male and female of Kutum at difference months (ANOVA, P〈0.05). The sex ratio of males to female was 0.65: 1 (males: females). The least and high condition factor was 1.5 in January and 2.17 in October, respectively. Length maturity (Lm50%) was 39.07cm. Minimum and maximum absolute fecundity was varied from 15713 to 130737 eggs with mean (±SD) 69961.7 (± 3836.4). Significant different was between absolute fecundity with fork length and weight (p〈0.05). Gonadosomatic index (GSI) mean for females were 2.5±0.2 (autumn) and 17± 0.2 (spring); 1.2± 0.1 (autumn) and 4.1± 0.2 (spring) for males, respectively wich coincided to spawning period. The highest feeding intensity (Gastrosomatic index) for females was 313.9 ± 65.9 in October and 119.4 ±59.2 in March; 350.7 ±103.7 in October and 144.1±85.04 in March for males, respectively. There are significant different between GSI with male anfemale in difference months (ANOVA, p〈0.05). The Bertalanffy growth equation for all populations of Kutum was L (t) = 63.92[1-e ^0.19 (t (-1.00) ]. Main prey mature of Kutum was Cerastoderma lamarki; Gastropoda and Balanus were subordinate, and Crab, Hypanis, Fish (Neogobous sp.) Algae, egg, and scale were random preys. Planktone groups comprise to Exuviaella, Nitzchia, Osillatoria, Synedra, Nematoda, Navicolla, Diatoma, Rholcosphenia were preys juvenile of Kutum. Based on ISI (Important species index), Cerastoderma lamarki and Balanus were dominant species in feeding of Kutum. As a result, growth, fecundity and length maturity of Kutum declined which may due to loss natural breeding area, overfishing (overlapping between fishing and spawning period) and artificial breeding that commenced since 1982. Hopefully, this information can be used in fisheries management.

    Description: Study on Viral Nervouse necrosis (isolation, characterisation and pathogenesis) in Golden grey mullet in the Caspian Sea and study of pathogenecity and possibility of transmission to the other fish species (Sturgeon fishes, Rutilus frisii kutum and reared Rainbow trout and Carp) Viral Nervous Necrosis (VNN) is a worldwide disease affecting several species of cultured marine fish. For the past two decades, betanodavirus infections that cause Viral Nervous Necrosis (VNN) have emerged as major constraints on the culture and sea ranching of marine fish in almost all parts of the world. More than forty species mainly of marine origin have been so far affected and this number is likely to rise in future following the introduction of new species and the increase of aquaculture trade. Unknown acute mortality occurred in wild golden grey mullet Lisa auratus and Liza saliens in Iranian waters of Caspian Sea in recent years. In order to isolation and confirmation of causative agents of golden grey mullet mortality in the Caspian Sea, a complementary research investigation project was designed in 2005 and approved immediately in Iranian Fisheries Research Organization (IFRO). Many diagnostic aspects such as Virology (Cell culture and Elctereone Microscopy), Hemathology, Bacteriology, Histopathology, Molecular biology (NestedRT-PCR), Heavy metals measuerment and Serology (IFAT and IHC) were employed in mentioned multidiciplinary project. About 322 moribund fish samples which revealed skin darkening, erratic swimming behavior such as spiral and belly-up at rest and high distention of swimming bladder. Suspected samples were collected from coastal capture sites in iranian north proviences in 2006 till 2009. Targets tissue such as brain and eye were removed in strile condition and then kept in -80oC frezzer for cell culture and Nested-RT-PCR. Other tissue samples from liver, kidney, intestine, stomach, gill, skin and muscle, gall bladder and gonads were taken and fixed in 10% buffer formalin and same parts fixed in glutaraldehyde 3% for histopathology, IHC and EM respectively. Cytopathic effect (CPE) was observed in those cell cultures just six days after inoculation with the dilutions of the tested 312 homogenate supernatants. CPE in monolayers of cells cultured (SSN-1 cell line) was characterised by thin or rounded, refractile, granular cells with vacuoles. Nine samples were positive in virology assay. Nested- RT-PCR was done on suspected tissue samples and supernatant of CPE positive samples and 21 tissue samples and all CPE positive samples were positive. IFAT was selected as a confirmatory method for identifying viral strains replicating on cell cultures and carried out with rabbit anti-betanodavirus serum on suspected tissue samples and some smears of CPE positive samples. Some bright points approved betanodavirus antigen and confirmed cell culture and Nested-RT-PCR findings. In fixed tissue samples widespread and massive vacuolation were observed in brain, spinal cord, retina and optical nerve. In order to confirmation of diagnostic findings , IHC was done with monoclonal antibody antibetanodavirus and some red-brown points were observed. Theses findings revealed expected viral antigens and confirmed previous results. Moreover, virus particles with 25-30 nm in diameter were visualized in infected brain and retina using positive staining in TEM. Also pathogenicity test was employed to confirm the obtained results. So Guppy fish Poecilia reticulata and sturgeon fry were used instead of the experimental host due to ease of handling and susceptibility. After 15 days post infection, guppy bathed in VNN-infected tissue culture with 104 TCID50 showed clinical signs similar to naturally infected Golden grey mullet, and the mortality rate reached up to 100% in 75 dpi. When target organs were examined by cell culture isolation, serology, and histopathology, all revealed the presence of virus in the Guppy. Suspected supernatant injected to sturgeon fry through intravitreous injection and widespread vacuolation were observed in brain and spinal cord buy IHC and Real time PCR were negative. In conclusion, with attntion to obtained results in this investigation such as ecological factors, clinical signs, histopathological, virological and bacteriological results, molecular analysis, (IHC, IFAT, PCR), TEM demonstration, serological and hematological findings, it could be confirmed that VNNV was the main causative agent for disease outbreak in Golden grey mullet in Southern coastline of Caspian Sea.

    Description: This study was conducted to determine catch and fishing effort trends, length and age composition, growth and mortality parameters and estimate biomass and acceptable biological catch of Kutum in the Iranian coastal waters of Caspian Sea from 2013 to 2015. The catch of Kutum was 6112.8 and 5599.2 tons and CPUE were 181.1 and 183.5 kg/haul in 2013-2014 and 2014-2015, respectively. The mean (± SD) of fork length were observed 39.4±6.1 and 40.7±5.6 cm and for total weight were 805.1±378.0 and 891.5±384.6 gram in 2013-2014 and 2014-2015, respectively. The most predominated of length class was obtained 40-42 cm and frequency of immature individuals were observed 37.8 and 44.4 percent during two sampling period, respectively. The range of age was from 2 to 11 year and fish with 4 to 6 years comprised up to 84 percent. In length – weight relationship a, b and r2 were calculated 0.011, 3.02 and 0.96 respectively. K value, L∞ and t0 were calculated 0.19 (/year), 61.3 cm and -0.99 respectively. Mortality parameters such as Z, M and F were calculated 0.82, 0.36 and 0.46 respectively and Exploitation rate were calculated 0.56. The biomass of kutum estimated 41000 tons and the acceptable biological catch calculated 8250 to 9750 tons. The biomass of Kutum decreased in recently years in spite of produce and release of huge fingerlings. This recruitment (produce and release fingerlings) must be continued for stability of stocks and fishing stress must be decrease by anyway.

    Description: This study aimed to investigate the toxic effects of atrazine herbicide on the fry of Caspian Kutum (Rutilus frisii kutum, Kamensky, 1901). First the 96-h LC50 of the fry were exposed to atrazine at the concentration of 24.95 ppm was determined. Then the toxicity of this herbicide on Caspian kutum fry exposed to the concentration of 12.47ppm (1/2 LC50), for four days was measured and compared with a control group. Comparison of the length, weight and condition factor showed no significant differences between atrazine exposed and control group. The concentration of Na+, K+, Ca2+, Mg2+ and Cl- in the whole body of fry in control and atrazine exposure groups were as the following order: Ca2+〉K+〉 Na +〉 Cl- 〉Mg2+ and Ca2+〉Na+〉K+〉Mg2+〉Cl-, respectively. Results showed that the concentration of all these ions were higher in atrazine exposure group than control group, except for Cl-, and the only significant differences was found in Na+ concentration. Major histopathological effects of atrazine on the gills were hyperplasia and thickening of the filaments, separation of the pavement cells of the lamellae epithelium from the pillar cells and swelling of the epithelial cells. Results of the present study showed that atrazine could affect the ion composition of the body, and caused major damages in gill epithelium even at sublethal concentration and acute exposure, but had no effects on the growth parameters.

    Description: The histopathological effects of Linear Alkylbenzene Sulfonate on the gill, liver and kidney tissues of the Caspian kutum, Rutilus frisii kutum were studied. The fish were exposed to three sublethal concentrations of anionic detergent, Linear Alkylbenzene Sulfonate (LAS) for short term intervals (192 h). Gill, liver and kidney samples were collected after 192 h of exposure to LAS and lesions were analyzed by light microscopy. The histological changes to gills were edema, fusion of lamellae and lamellar aneurism. Some alterations like reduction of the interstitial haematopoietic tissue, tubular shrinkage, degeneration in the epithelial cells of renal tubule and necrosis were observed in the kidney. In the liver tissue, hepatocyte degeneration, congestion and dilation of sinusoid and vacuolar degeneration were seen. It seems that sublethal concentration of LAS may affect sever changes to gill, kidney and liver of R. frisii kutum specimens that leads to malfunction of these organs which cause damage to health of the fish.

    Description: Overfishing, destniction of spawning grounds together with intensive artificial reproduction and release of Rutilus fish/ kutum fingerlings have caused remarkable fluctuations in the stocks of the fish in the Caspian Sea over the past decades. The mean catch size of the fish during periods 1937-1947, 1967-1977 and 1987-1997 were 3110, 990 and 8505 tons respectively. Also the minimum catch was recorded as 121 tons in 1964 and the maximum catch was recorded as 11175 tons in 1994. The catch per unit effort (CPUE) of the species shows high variations such that it has been 17 kg/ton in 1971, 216 kg/ton in 1989 and 112 kg/ton in 1999. The decline in CPUE in 1999 was due to increased fishing efforts of 3.8 orders of magnitude (in comparison to the year 1989) as a result of fishing cooperatives increasing in number during that period. The calculated stock biomass of the fish was 1300 tons in 1971 fluctuating between 18489 to 25400 tons in the period 1990-2000. The mean biomass of the fish in the recent past 10 years was 22750 tons which shows 17-fold increase in comparison to the year 1971. The accomplished catch in the recent 10 years has been 35 to 46 % of the annual stock. The exploitation rate being more than MSY and decreased mean weight of the released fingerlings plus lowered return rate might have caused the decrease in the stock size of the fish during 1998-1999.

    Description: Bony fish stocks are being legally utilized by 134 fishing cooperatives in the Iranian side of the Caspian Sea mainly through beach seine. This fishing method has been invariantly used over the past. Fifty years with actually no alteration or technical modifications in the fishing nets despite the substantial changes in the quality & quantity of bony fish resources. Based on the data obtained from the stock assessment project for bony fish, the amount of premature or young fish (substandard size) caught during the past decade has been far greater than those of authorized fish size and even the figures far certain species suggest heavy over fishing to such an extent that in the case of frisii kutum, there has been a reduction by 50% in the overall kutum landings.Thus in order to modify the technical design of the fishing with a view to alleviate the catch of non-standard fish and whereby prevent over fishing by beach seine, it seems imperative therefore to launch a biometric study on bony fish stocks particularly the two intended fish species namely Rutilus frisii kutum and mullet which would involve identification of their standard weight and length using pertinent mathematical equations as well as determining proper mesh size of the code-end and other parts of the nets & consequently the H. R, d/a coefficients. The operations related to project were carried out in the "Inland Aquaculture Research Institute" net mating plant and beach seine site in Anzali during 2005-7 which aimed at designing and constructing standardized beach seines and comparing the fishing performance & yields with those of ordinary beach seine nets under equal conditions with a ratio of 3:1. The results concerning the quality and quantity of fish caught during a whole fishing cycle were recorded in the related data forms. The statistical analysis consisting the comparison of the quality of fishing performance in both the experimental and control beach seine nets was made using T. student and excel, spss 12 software packages. The results indicated that the reduction in the amount of non-standard fish caught in standardized beach seine net (the experimental net) was meaningfully different from that of the ordinary (control) net showing the superior fishing performance of the former. The catch level of non-standard R.frisii kutum decreased from 67.1% in the control beach seine net down to 7.1% in the standardized beach seine net and these figures for mullet were 29.98% and 1.1% respectively. Based on the results obtained and considering the apparent superiority of the experimental net in catching greater number of mature fish and less number of premature or non-standard sized fish, the use of this modified beach seine net can be proposed to Iranian fisheries organization as a viable alternative to the traditional beach seine nets.

    Description: The toxicity of the pesticides, Malathion and Diazinon, and the herbicides, Machete and Saturn, on mortality of fingerling of Rutilus frisii kutum were examined. Lc10 values for Diazinon, Malathion, Machete and Saturn were 0.14 - 0.24, 0.10 - 0.35, 0.14 - 0.55, and 1.45 - 2.50 mg/L respectively. The result showed that Diazinon, Malathion and Machete have a highly toxic effect on fingerling of R. frisii kutum and the herbicide, Saturn, in comparison with other poisons was less toxic.

    Description: The study was done in Nutrition and Live Food Station was located in Bandar Anzali Ghaziyan. Juveniles weighted average 0.5, 1, 2.5, 5, 10, 15 and 20 g were randomly selected in three water conditions with a salinity of 11 ppt (Caspian sea water), water 7 ppt and freshwater (with three replicates per group) were included. At intervals of 0, 3, 6, 12, 24, 72, 168, 240 hours, blood samples were heparinized capillary tubes by caudal juveniles and ion concentrations Mg, Ca,Cl using the spectrophotometer and sodium and potassium ions with Flaym photometry (flame photometric), the osmotic pressure of blood plasma by osmometer and cortisol levels were measured by RIA method. To study the microstructure of gill and kidney tissue for each treatment , tissue samples by classical histological methods and stained with hematoxylin - eosin slides were prepared. The frequency and location of the enzyme Na +, K + - ATPase and chloride cells with immunohistochemical localization was performed. Studies micrometric gill chloride cells and renal glomerular networks by software Image tool (version 2.0) was performed. Measurement of enzyme Na +, K +-ATPase, by Zaugg (1982) method was conducted. Data analyzed by one-way ANOVA (Oneway ANOVA) with Tukey's test was performed. Overall, the results of measuring ions and osmotic pressure on the tenth day of treatment, the osmotic potential juveniles 2.5, 5, 10, 20 gr in Caspian sea water and all groups except the 0.5 in water of 7 ppt confirmed. But in case of unfavorable conditions for the release in estuaries river and river, fish with weight 1 to 3 release directly to beach (where the salinity is 7 grams per liter) and fishes with weight from 10 to 20 gr to sea. Although suitable river conditions necessary condition for release of juveniles in riverine areas to adaptation juveniles occur gradually.

    Description: The Kutum, Rutilus frisii kutum, is one of the most important bony fishes in Iranian coastal of Caspian Sea. Its harvest range is between 9000-10000 tons in a year, nearly 60% of the income of Bony fish fishery produced by kutum fishery. The stock of this species reduced drastically in 1982 and the catch slumped to the less than 250 tons in a year. Kutum spawning grounds deterioration, illegal catch, lack of restocking program were the main cause of the decline. This Spices in nature comprised by two distinct form, autumn and spring form. It is worth to be mentioned, by the effect of Caspian Sea Bony fishes Research Center s experts in 1983, artificial spawning and releasing the fries to the sea were commenced and the catch steadily improved. But all activities concerning restocking of kutum concentrated in spring form, as at present about 260 million its fries are released into sea for restocking by Iranian Fisheries Organization, but for above reasons and lack of restocking program, the populations of autumn form gravely shrinked and neared to be extinct. Therefore, to enhance the biodiversity and boost fishers livelihood of kutum in Caspian Sea this project implemented by cooperation of Iranian Fisheries Organization (IFRO) and Caspian Environment Program (CEP) in Aquaculture Institute (Inland Waters). In this project, brooders caught from Anzali lagoon and maintained in two different condition, include of floating cages in Anzali lagoon and earthen ponds in Sefidrud Fisheries Research Station. The results showed that there weren t significant differences between two maintenance statuses in maturation period and other reproductive characteristics of brooders. The ratio of male to female was 1 to 1.4. Minimum and maximum weight measured 1450 to 3100 g (with average of 1850 g) in female and 670 to 1900 g (with average of 1165 g) in male, respectively. The first natural spawning of brooders occurred in the end of January in temperature of 8 till 10 °C in concrete ponds. Also, some of maintained brooders in earthen ponds spawned in February. The average number of absolute, function and relative fecundity determined 88565 16809, 73805 14008 and 48670 12056, respectively. For artificial spawning, male and female brooders injected by pituitary gland with dose of 2-3 and 4-5 mg/kg body weight, respectively. Approximately, 10 and 8 present of female were over-ripe and immature in March (artificial spawning time), respectively. More than 59 % of injected female brooders induced to spawning in first stage after 10-12 hours and 13 % of them in twice stage and 7-8 hours after first stage. And also, 27.6% of females didn’t positive response to injection. Dry method used for eggs fecundity and incubation period lasted 7- 10 days in 14-16 °C. In totally, eggs fertilization were more than 95% and the average of eggs fertilization percent in throughout of period measured more than 92.7 6 %. Eyed eggs appearance occurred 3 days after fecundity and its mean was 92.7 15.1%. Larvae after yolk sac absorption feed with dry milk for 4-5 days and then introduced into fertilized earthen ponds (500 m2 and equipped to aerators) in intensive condition and fed with micro pellet food for 3-4 month. In finally, more than 1.8 millions fries of 1-2 g and some more than 5 g produced and released into Anzali lagoon to its restocking for first time. It is expected that continuing of restocking process of autumn form kutum by Iranian Fisheries Organization eventuate to population increasing of this form in Caspian Sea in future.
