    Notizen: The role of the cytoskeleton and cell attachments in the alignment of baby hamster kidney fibroblasts to ridge and groove substratum topography was investigated using confocal scanning microscopy. This was carried out with normal cells and cells treated with the cytoskeleton modifiers cytochalasin D, colcemid, and taxol. Actin was localised with fluorescent phalloidin. Tubulin, Vinculin, and intracellular adhesion molecule-1 were visualised by indirect immunofluoresence. The spreading, elongation, and orientation of the cells after 24 h of culture in these conditions were measured on grooves of 5, 10, and 25 μm width and 0.5, 1, 2, and 5 μm depth. We have also observed events over the first 30 min of cell attachment. Five minutes after cell attachment, F-actin condensations were seen close to the intersection of groove wall and ridge top, that is, at a topographic discontinuity. The condensations were often at right angles to the groove edge and showed a periodicity of 0.6 μm. Vinculin arrangement at the early stages of cell spreading was similar to that of actin. Organisation of the microtubule system followed later, becoming obvious at about 30 min after cell plating. The Curtis and Clark theory (that cell react to topography primarily at lines of discontinuity in the substratum by actin nucleation) is supported by these results. The use of cytoskeletal poisons did not entirely abolish cell reaction to grooves. Colocemid increased cell spreading and reduced cell orientation and elongation. Cytochalasin D reduced cell spreading, orientation, and elongation. Taxol reduced cell elongation but did not affect cell spreading and orientation. We conclude that the aggregation of actin along groove/ridge boundaries is a primary driving event in determining fibroblast orientation on microgrooved substrata.

    Notizen: A test mixture containing 28 compounds plus their stable isotopically labeled analogs was analyzed on a daily basis for one month during a series of routine fused silica capillary column GC/MS analyses in order to establish the precision with which retention times and responses were produced. These long-term precision data were evaluated to determine how to best predict retention times and how to best reproduce quantification. Results clearly indicate that relative retention times should be calculated using the reference compound eluted most closely to the target compound and that quantification should be based on relative response using a chemically “similar” compound.

    Notizen: Operational characteristics have been determined for fused silica capillary column (FSCC) GC/MS as applied to “extractable” priority polutants. Chromatographic data show excellent relative retention time (RRT) intralaboratory precision and interlaboratory accuracy when multiple internal standards are empolyed. Potential chromatographic problems, such as column overload and “double peaking”, are addressed. Response factor relative standard deviations (RSD) at 50 ng for most of the extractable priority pollutants over the long term indicated precise determination (i.e. RSD generally ≤ 10%). Linearity was demonstrated over two orders of magnitude for FSCC GC/MS analysis of compounds with relatively low and high RF (response factor) values. Potential quantitative problems, such as saturation, are discussed. For certain aromatic priority pollutants interlaboratory RF agreement was observed. This was noted as perhaps the most important property of FSCC GC/MS analysis when the multiple internal standard approach is utilized. Determinations of extractable priority pollutants are directly compared for paced column GC/MS and FSCC GC/MS analysis of separate and composited extracts. For six extracts analyzed in triplicate, the latter configuration was shown to produce more consistent results. In view of the superior analysis logistics of composite extract FSCC GC/MS analysis, this approach was established as the preferred method for the analysis of priority pollutants classified as extractable.

    Notizen: A method is described for the determination of 22 tetrachlorodibenzo-p-dioxin isomers at the low part-per-trillion (ppt) level. High resolution, narrow bore, open tubular columns (OTCs) with 100 μm i.d. can achieve better separations than presently used 320 or 250 μm i.d. columns in about half the total time. Relative retention times and response factors for all 22 TCDD isomers are presented for the electron capture detection and for the molecular ion mass of individual isomers under electron impact ionization with selected ion monitoring (SIM).

    Notizen: The influence of two distinct types of water leachable polymeric additives upon the transport properties across polymer latex films is considered. Hydroxypropyl methylcellulose, added as an aqueous solution, and a methyl methacrylate-methacrylic acid copolymer, added as latex particles, are both shown to be more effective than lower molecular weight rapidly water soluble additives, typified by sucrose, in enhancing solute transport and creating pathways for convective diffusion.

    Notizen: An investigation of the effect of leachable sucrose on the permeability of polymer latex films showed that the additive can be used to vary the solute permeability in a systematic manner, as a result of increased free volume resulting from additive leaching. However, the extent of the permeability enhancement is dependent on both the glass transition temperature of the polymer and its ability to sustain porosity and prevent pore reduction by wet sintering and/or film drying, and also the compatibility of the sucrose additive with the polymer. The loading of additive dictates how readily it is leached and, hence, the presence, or not, of continuous porosity through the full thickness of film. Although continuous porosity can be conferred on the film by the sucrose leaching, the resulting porosity still greatly inhibits the permeation of electrolyte, and the film fails well before the level of porosity approaches that required for unimpeded transport.

    Notizen: A range of permeation techniques were employed to investigate the transport mechanisms across polymer films cast from lattices containing either no or only low levels of water soluble leachates such as residual surfactant from polymerisation or added plasticiser essential to film formation. Hydrophilic pathways can provide a mechanism of transport, in addition to partition into and solution-diffusion in the polymer phase. Loss of porosity by low Tg polymers on drying highlights the value of solute probes in studying film morphology.

    Notizen: A gas chromatographic mass spectrometric assay using selected ion monitoring for meperidine and normeperidine in blood and plasma is described. Both unlabeled and [2H5]phenyl ring labeled drugs may be determined simultaneously using [2H10] analogs as internal standards. Calibration curves are linear over the range 5-100 ng ml-1, the results reproducible (coefficient of variation 〈5%), the sensitivity limit is about 5 ng using a 1 ml sample and no interference is present in blank blood or plasma, or by isotope impurity. The method is shown to be applicable to measuring the concentration of meperidine and its metabolite, normeperidine, in blood, following simultaneous administration by the intravenous and oral routes of 25 mg each of unlabeled and [2H5] labeled drug, respectively.

    Notizen: Segmented copolyureas (PUr) and copoly(urethane-urea)s (PUU) comprising 50% by weight of polyurea hard segments (HS) and polyether soft segment (SS) with different functionalities, have been formed by reaction injection moulding (RIM). The HS were formed from 4,4′-diphenylmethane diisocyanate reacted with mixed isomers of 3,5-diethyltoluene diamine. The nominal functionality of the SS prepolymers used (either amino- or hydroxyl-functionalised polyoxypropylenes with a constant molar mass per functional group of ∼ 2000 g mol-1) was systematically increased from 2 to 4. RIM materials were characterised using dynamic mechanical thermal analysis, differential scanning calorimetry, tensile stress-strain and fracture mechanics studies. Generally, the PUr exhibited far superior thermal-mechanical properties than equivalent PUU materials but inferior fracture resistance, owing to morphological variations resulting from differences in copolymerisation behaviour. For both systems, tensile behaviour was shown to be dominated by the degree of phase separation, whereas fracture properties showed a degree of dependence on SS functionality.
