    Notizen: Ammonia/ammonium thiocyanate (NH3/NH4SCN) is an excellent swelling agent and solvent for cellulose, even at a high degree of polymerization. Because polymorphic conversion in cellulose has been a long-standing, perplexing, troublesome problem, we have undertaken to study that mechanism. Solid state CP/MAS 13C-NMR and X-ray analysis proved to be very useful analytical techniques for the task. It appears that during temperature cycling, specific cellulosic inter- and intramolecular hydrogen-bonds are broken as polymorphic conversion proceeds sequentially from the polymorph I to III, and finally at total solvation to amorphous. This proceeds correspondingly via transformation of the polymorph conformations of CH2OH from trans-gauche, “tg,” to gauche-trans, “gt,” to gauche-gauche, “gg.” © 1994 John Wiley & Sons, Inc.

    Notizen: In a previous article,1 we reported on the interaction of cellulose with NH3/NH4SCN by using solid state CP / MAS 13C-NMR, wide-angle X-ray and other techniques. It appears that during an imposed temperature cycling sequence, specific cellulosic inter- and intramolecular hydrogen bonds are broken as polymorphic conversion and, ultimately, dissolution occurs. Cellulose is converted from the polymorph I to II to III and, finally, to amorphous. We speculate that these changes proceed via transformation of the polymorph conformations of CH2OH from trans-gauche, “tg,” to gauche-trans, “gt,” to gauche-gauche, “gg.” Remarkably, the temperature cycling effectively and rapidly effects these changes, seemingly, by invoking at two different temperatures (22°C and -78°C), a subtle but powerful temperature-related interplay of enthalpic and entropic forces. At the higher of the cycling temperature limits, entropy and van der Waals forces dominate, causing NH3 to partition in favor of the liquid phase. At the lower of the temperature cycling limits, enthalpy dominates and the situation is reversed favoring NH3 partition toward the cellulose, resulting in breakage of inter- and intracellulosic H-bonds by the interpenetrating ammonia to form new H-bonds between cellulose and ammonia, leading ultimately to conformational changes and, ultimately, even dissolution. © 1994 John Wiley & Sons, Inc.

    Notizen: The collisional activation (xenon) decomposition of actinomycin V, dactinomycin, 7-nitrodactinomycin, 7-aminodactinomycin, and a mixture of dactinomycin, actinomycin C1 and actinomycin C2 was studied using a triple-quadrupole mass spectrometer with fast atom bombardment (xenon) ionization. Fragmentation pathways of the structurally significant ions were rationalized by assuming an initial McLafferty rearrangement at the ester linkage between the methylvaline and the threonine of both rings to form linear pentapeptides, followed by fragmentation in either pentapeptide yielding typical sequence ions and sequence cleavages from both rings.

    Notizen: The acrosome of the stallion spermatozoon was visualized by indirect immunofluorescence with monoclonal antibody (18.6) which recognized an integral acrosomal membrane component. Localization was confirmed by electron microscopy using peroxidase labelled antibody. In fresh semen samples (n = 19), 73.9 ± 9.1% of the spermatozoa from five fertile stallions displayed a uniform bright fluorescence over their acrosome region. In two semen samples from an infertile stallion only 28% and 35% of spermatozoa showed the same pattern of fluorescence. Spermatozoa from fertile stallions incubated for up to 12 hours in TALP medium maintained motility and exhibited a significant progressive loss of acrosomes as detected by immunofluorescence. Alternatively, a similar loss of acrosomes could be induced with calcium ionophore A23187 over a 90 minute incubation. Ultrastructural observations and incubation with zona-free hamster eggs indicated that only with ionophore treatment was immunofluorescent acrosome loss correlated with a physiological acrosome reaction, while prolonged sperm incubation led to degenerative membrane changes. It was concluded that, if carefully validated, immunofluorescent localization of the acrosome of stallion sperm with monoclonal antibody could be used to monitor the acrosome reaction. Furthermore, definitive acrosome visualization would be valuable in assessing semen quality.

    Notizen: A comprehensive 13C-NMR method for the analysis of composition in the most common commercial polyethylene copolymers has been established. The method covers ethene copolymers with propene, butene-1, hexene-1, octene-1, and 4-methyl pentene-1 in the composition range of 1-10 mol %. The chemical shift assignments and T1 values of the resonances of the copolymers are presented. Results of precision studies and interlaboratory analyses showed that the molar composition could be determined with a relative precision at 2δ of about 6%. This method is being proposed to ASTM as Method X70-8605-2.

    Notizen: The mechanical properties of a propylene-ethylene copolymer resin depend strongly on the mold temperature employed during injection molding. Average spherulite size correlates well with most physical properties; good impact properties are related to small average size, and high heat distortion temperatures to large average size. Tensile specimens have three distinct regions when viewed through crossed polars. The outer region (Region 1) does not contain spherulites, but Regions 2 and 3 do. The largest spherulites are found in Region 3, the center of the specimen. Region 1 has a molecular weight higher than that of the other two regions.

    Notizen: The amount of residual n-pentane in expandable polystyrene bead foam (PSBF) has an effect on the strength and dimensional stability of the foam. Data were needed to determine how fast the residual n-pentane could be removed to enhance the properties of PSBF. An experiment was designed to determine the diffusivity of n-pentane through PSBF at various temperatures. Mathematical models based on the appropriate diffusion differential equations were then used to predict n-pentane concentrations in in cylinders of PSBF. The diffusivity of n-pentane through PSBF was found to have a magnitude of 1×10-8 to 1×10-6 cm2/sec and to vary with temperature, foam density, n-pentane concentration, and foam structure.
