    Notizen: Abstract  N2O emissions were measured from three contrasting onion (Allium cepa L.) production systems over an 8.5-month period. One system was established on soil where a clover sward had 3 months earlier been ploughed in (ploughed clover site). This production system followed conventional production management practices. The other two systems were established on soil where a mixed herb ley had 3 months earlier been either ploughed or rotovated. These last two production systems followed the guidelines of the International Federation of Organic Agriculture Movements (IFOAM). Cumulative N2O emissions were significantly greater from the ploughed clover site compared to the ploughed ley site (3.8 and 1.6 kg N2O-N ha–1, respectively), while cumulative N2O emissions from the ploughed ley and rotovated ley sites were not significantly different from each other. Emissions from all sites were dominated by episodes of high N2O flux activity following seedbed preparation and drilling, when soil water suction (SWS) was shown to be the rate-controlling variable. The decline in the N2O fluxes after these peak emissions followed clear exponential relationships of the form F=Ae– kt (r≥0.91), where F is the daily flux and A is the y-intercept. First-order decay constants (k) during these periods of declining N2O fluxes (corresponding to half-lives of 2.6–3.0 days) were not significantly different in magnitude from the first-order rate constants that characterised the increasing SWS. Gross differences in cumulative emissions between the clover and ley sites were attributed to the influence of differing soil pHs at the two sites on the N2O:(N2O+N2) ratio in the denitrification products. It also appeared that fertiliser applications to the clover site had both direct and indirect effects on N2O emissions by: (1) enhancing N2O emissions via potential nitrification, (2) increasing the NO3 – supply for enhanced N2O emissions via denitrification, and (3) influencing the N2O:(N2O+N2) ratio by lowering soil pH and increasing NO3 – concentrations. Onion crop yields were greater at the clover site, mainly due to the higher density of planting made possible under a conventional production philosophy. Expressing the yield on the basis of net N2O emissions, 23 t onions kg–1 N2O-N was obtained from the ploughed clover, which was double that obtained for the two systems based on the ley site. However, when the N2O emissions from the cultivation of the soils prior to the sowing of the onions was included, all three systems produced a similar yield per kilogram of N2O-N emitted, averaging 10 t kg–1.

    Notizen: Abstract  Nitrous oxide (N2O) emissions via the nitrification (I nit) and denitrification (I den) pathways were successfully measured with in-field incubation of soil cores in preserving jars at 0 Pa and 5–10 Pa acetylene. From the incubations, fractions of nitrification – N2O over total N2O (I nit / I tot) – and denitrification – N2O over total N2O (I den / I tot) – were obtained. Actual field emissions of N2O via nitrification (F nit) and denitrification (F den) were calculated by multiplying the fractions from the incubation technique with the daily N2O emission (F day) determined with a direct soil cover method. The approach presented here was successful for a whole range of soil moisture conditions in intensive grassland. F nit and F den followed the trends of soil ammonium and soil nitrate.

    Notizen: Abstract  Nitrous oxide (N2O) emissions and methane (CH4) consumption were quantified following cultivation of two contrasting 4-year-old pastures. A clover sward was ploughed (to 150–200 mm depth) while a mixed herb ley sward was either ploughed (to 150–200 mm depth) or rotovated (to 50 mm depth). Cumulative N2O emissions were significantly greater following ploughing of the clover sward, with 4.01 kg N2O-N ha–1 being emitted in a 48-day period. Emissions following ploughing and rotovating of the ley sward were much less and were not statistically different from each other, with 0.26 and 0.17 kg N2O-N ha–1 being measured, respectively, over a 55-day period. The large difference in cumulative N2O between the clover and ley sites is presumably due to the initially higher soil NO3 – content, greater water filled pore space and lower soil pH at the clover site. Results from a denitrification enzyme assay conducted on soils from both sites showed a strong negative relationship (r=–0.82) between soil pH and the N2O:(N2O+N2) ratio. It is suggested that further research is required to determine if control of soil pH may provide a relatively cheap mitigation option for N2O emissions from these soils. There were no significant differences in CH4 oxidation rates due to sward type or form of cultivation.

    Notizen: Abstract We have used DNA amplification fingerprinting (DAF) to study the genetic variation of bermudagrass (Cynodon) species and cultivars of interspecific crosses that exhibit leaf-blade textural characteristics ranging from coarse to fine. Arbitrary octamer primers produced complex and reproducible amplification profiles with high levels of polymorphic DNA. Phylogenetic analysis using parsimony (PAUP) and unweighted pair group cluster analysis using arithmetic means (UPGMA) grouped 13 bermudagrass cultivars into several clusters, including one containing the African-type bermudagrasses (C. transvaalensis) and another containing the common-type bermudagrasses (C. dactylon). The latter group included C. magennissii (‘Sunturf’) and a interspecific C. transvaalensisxC. dactylon cross (‘Midiron’), 2 cultivars that exhibited leaf textural characteristics closer to the common-types. All other C. transvaalensisxC. dactylon crosses grouped between the African and common types. An extended screen of 81 octamer primers was needed to separate cultivar ‘Tifway’ from the irradiation-induced mutant ‘Tifway II’. The use of either template endonuclease digestion prior to amplification or arbitrary mini-hairpin primers increased detection of polymorphic DNA and simplified the task of distinguishing these closely related cultivars. Alternatively, the use of capillary electrophoresis (CE) resolved fingerprints adequately and detected products with high sensitivity, thereby promising to increase throughput and the detection of polymorphic DNA. When used to fingerprint samples from commercial sources, DAF identified bermudagrass plant material on the basis of unique reference profiles generated with selected primers. DAF represents an excellent technique for bermudagrass cultivar verification, seed certification, varietal protection, and for the identification of mistakes in plantings, mislabeled plant materials, and contamination or substitutions of sod fields.

    Notizen: Poly[(methylphenylsilanediyl)methylenecyclopropylenemethylene] (3a) was prepared by cyclopropanation of the C—C double bonds of poly[(methylphenylsilanediyl)-(Z)-butenylene] (2a) by a Simmons-Smith reaction. Alternatively, 3a was prepared by a Birch reduction of the dichlorocarbene adduct poly[(methylphenylsilanediyl)methylene(2,2-dichloro-1,3-cyclopropanediyl)-methylene] (4a). Both 3a and 4a show the characteristic absorptions in the 1H, 13C, and 29Si NMR, IR and UV spectra. Their molecular weight distributions, determined by GPC, are rather broad and their thermal stabilities (by TG) are relatively high.

    Notizen: Diesel fuels from different parts of the UK have been analyzed for polycyclic aromatic hydrocarbons (PAH), nitrogen-containing PAH, and sulfur-containing PAH using capillary column GC with simultaneous parallel triple detection. The concentrations of polyaromatic compounds (PAC) were high and showed considerable variability amongst the fuels. The PAH are mainly naphthalene, fluorene, and phenanthrene and their alkylated homologs; the PANH are mainly carbazole and its methyl derivatives; the PASH are mainly dibenzothiophene and its methyl derivatives.

    Notizen: The interaction of κ-carrageenan with locust bean gum and dextran has been studied by rheology, differential scanning calorimetry (DSC), and electron spin resonance spectroscopy (ESR). Rheological measurements show that the carrageenan gel characteristics are greatly enhanced in the presence of locust bean gum but not in the presence of dextran. Carrageenan/locust bean gum mixtures show two peaks in the dsc cooling curves. The higher temperature peak corresponds to the temperature of gelation and its intensity increases at the expense of the lower temperature peak as the proportion of locust bean gum in the mixture increases. Furthermore, the DSC heating curves show enhanced broadening when locust bean gum is present, indicating increased aggregation. These results are taken as evidence of carrageenan/locust bean gum association. The gelation process has also been followed by ESR using spin-labeled carrageenan. On cooling carrageenan solutions, an immobile component appears in the ESR spectra signifying a loss of segmental mobility consistent with chain stiffening due to the coil → helix conformational transition and helix aggregation. For carrageenan/locust bean gum mixtures, carrageenan ordering occurs at temperatures corresponding to the higher temperature DSC setting peak and the temperature of gelation. Similar studies using spin-labeled locust bean gum show that its mobility remains virtually unaffected during the gelation process. It is evident, therefore, that carrageenan and locust bean gum interact only weakly. It is proposed that at low carrageenan concentrations the gel network consists of carrageenan helices cross-linked by locust bean gum chains. At high carrageenan concentrations the network is enhanced by the additional self-aggregation of the “excess” carrageenan molecules. For carrageenan/dextran mixtures, only one peak is observed in the dsc cooling curves. The onset of gelation shifts to higher temperatures only at very high (20%) dextran concentrations and this is attributed to volume exclusion effects. Furthermore, there is no enhanced broadening of the peaks in the DSC heating curves as for the carrageenan/locust bean gum systems. It is therefore concluded that carrageenan/dextran association does not occur. The difference in behavior between locust bean gum and dextran is attributed to the greater flexibility of the dextran chains. © 1996 John Wiley & Sons, Inc.

    Notizen: The solution properties of κ-carrageenan and κ-carrageenan/locust bean gum mixtures have been studied by small deformation oscillation measurements and differential scanning calorimetry (DSC) in the presence of sodium chloride and sodium iodide. Both salts induced the κ-carrageenan to undergo a coil-helix conformational change as noted by an increase in the storage and loss moduli (G′, G′) and by an exothermic peak in the DSC cooling curves. The enthalpy ΔHc-h and temperature of the conformational transition Tc-h were higher in Nal compared to NaCl and Tc-h increased with increasing the concentration of both electrolytes. Gelation was not observed for carrageenan or carrageenan/locust bean gum mixtures in the presence of up to 200 mM Nal. Although carrageenan alone did not gel in the presence of 100 mM NaCl, a weak gel was obtained for a mixture containing 0.9%/0.1% carrageenan/locust bean gum. Furthermore, the mixture showed hysteresis in both the rheological and DSC cooling and heating curves. A strong gel was produced for carrageenan alone in the presence of 200 mM NaCl and the gel strength increased on adding a small proportion of locust bean gum (0.9%/0.1%). © 1997 John Wiley & Sons, Inc.     Notizen: The phase behavior of ternary (protein + dextran + solvent) and quaternary (protein 1 + protein 2 + dextran + solvent) aqueous solutions have been monitored at various ionic strengths and pHs by turbidity measurements. For the ternary systems, phase separation was favored when the solvent conditions promoted protein self-association. Bovine serum albumin (BSA) and γ-globulin were believed to self-associate in the presence of dextran at their respective isoelectric points through the electrostatic interactions between positive and negative charged patches at their surface. The fact that phase separation for BSA became less evident on increasing the ionic strength supported this concept. In the case of lysozyme alone, self-association was promoted by the addition of electrolyte as was phase separation in the lysozyme + dextran ternary system. For this biopolymer self-association is presumably as a consequence of van der Waals and/or hydrophobic forces. The influence of dextran has been discussed in terms of polymer depletion theory where an additional attractive force is created due to the exclusion of the dextran molecules from the immediate vicinity of the protein surface. For the quaternary systems, by subtle choice of solvent conditions, BSA and globulin could be exclusively partitioned into separate liquid layers through a segregative mechanism, while BSA and lysozyme could be separated into a single highly concentrated layer through an associative mechanism. © 1998 John Wiley & Sons, Inc.
