    Notizen: The subject of this article is the self-consistent-field (SCF) treatment of low-lying molecular vibrations in molecules subject to solvent effects and light atom migration. The analyses use a Cartesian Gaussian basis and Gaussian functional expansions of potential energy operators. The objective of the work was to establish approximate and practical methods of analysis of vibrational degrees of freedom in molecules that build on and compare well with the highly accurate treatments of vibrations in small molecular systems of the past decade. An application to a system in which hydrogen bonding contributes the major anharmonic effect illustrates the method. © 1995 John Wiley & Sons, Inc.

    Notizen: Two different approaches were used to explain the distribution of water coordinates in the crystallographically invisible part of the unit cell. Monte Carlo calculations were done starting from different initial water structures. Extended Monte Carlo calculations using equal initial structures were also used to obtain two further water structures. The differences between the Monte Carlo water structures were used to calculate the mean-square displacements of the water molecules. Monte Carlo calculations starting from different structures lead to a mean-square displacement of 0.58 Å2, whereas extended Monte Carlo moves using the same initial structure only show a mean-square displacement of 0.17 Å2. The mean-square displacement of 0.58 Å2 can be used to explain the experimental data. © 1996 John Wiley & Sons, Inc.

    Beschreibung: Spatially resolved fluorescence resonance energy transfer (FRET) measured by fluorescence lifetime imaging microscopy (FLIM), provides a method for tracing the catalytic activity of fluorescently tagged proteins inside live cell cultures and enables determination of the functional state of proteins in fixed cells and tissues. Here, a dynamic marker of protein kinase Calpha (PKCalpha) activation is identified and exploited. Activation of PKCalpha is detected through the binding of fluorescently tagged phosphorylation site-specific antibodies; the consequent FRET is measured through the donor fluorophore on PKCalpha by FLIM.     Beschreibung: Reactive and potentially toxic cofactors such as copper ions are imported into eukaryotic cells and incorporated into target proteins by unknown mechanisms. Atx1, a prototypical copper chaperone protein from yeast, has now been shown to act as a soluble cytoplasmic copper(I) receptor that can adopt either a two- or three-coordinate metal center in the active site. Atx1 also associated directly with the Atx1-like cytosolic domains of Ccc2, a vesicular protein defined in genetic studies as a member of the copper-trafficking pathway.     Beschreibung: A candidate gene for the chromosome 1 Alzheimer's disease (AD) locus was identified (STM2). The predicted amino acid sequence for STM2 is homologous to that of the recently cloned chromosome 14 AD gene (S182). A point mutation in STM2, resulting in the substitution of an isoleucine for an asparagine (N141l), was identified in affected people from Volga German AD kindreds. This N141l mutation occurs at an amino acid residue that is conserved in human S182 and in the mouse S182 homolog.
