    Notizen: To investigate the mechanism of trophoblast adhesion to fibronectin, we cultured blastocysts in serum-free medium on proteolytic fibronectin fragments containing its major functional domains, and localized fibronectin-binding integrins in outgrowing trophoblast cells by immunofluorescent staining. Outgrowth comparable to that obtained with intact fibronectin was observed using a 120 kD chymotryptic fragment containing the central cell-binding domain (FN-120) and the Arg-Gly-Asp (RGD) recognition sequence. A 40 kD COOH-terminal chymotryptic fragment of fibronectin containing both a heparin-binding region and an alternate (non-RGD) cell-binding site was inactive in supporting trophoblast adhesion. Three synthetic peptides derived from the heparin-binding domain, including the CS1 alternate cell-binding site, were also unable to promote trophoblast cell adhesion. A 75 kD recombinant protein, ProNectin F, containing 13 copies of the cell recognition epitope of fibronectin, Val-Thr-Gly-Arg-Gly-Asp-Ser-Pro-Ala-Ser, vigorously supported blastocyst outgrowth. Blastocyst outgrowth was not significantly different when surfaces were precoated with cellular fibronectin, which contains an alternatively spliced type III repeat and is the form actually encountered in vivo. Several putative fibronectin receptors were localized in trophoblast outgrowths by immunofluorescent labeling. Antibodies reactive with integrin subunits α3, α5, αllb, αv, β1 and β3, but not α4, all bound to trophoblast cells. Antibodies raised against either the β1 or β3 integrin subunits significantly inhibited fibronectin-mediated outgrowth. These findings demonstrate the key role of the central cell-binding domain of fibronectin in trophoblast adhesion, and suggest four RGD-binding integrins, α3β1, α5β1, αllbβ3, and αvβ3, that could mediate trophoblast adhesion in vitro and may play an important role during implantation. © 1995 Wiley-Liss, Inc.

    Notizen: Amphiregulin (Ar) and Cripto-1 (Cr-1) are growth promoting peptides that share amino acid sequence homology with epidermal growth factor (EGF). The present study examined Ar and Cr-1 mRNA and protein expression during various stages of C57BL/6 mouse mammary morphogenesis. Reverse transciption-polymerase chain reaction (RT-PCR) was used to detect transcripts for Ar and Cr-1 at all stages of mammary development. Immunocytochemical (ICC) localization demonstrated that in virgin 4-week to mature 12-week-old mouse fourth inguinal mammary gland, Ar and Cr-1 are expressed in the stromal cells, luminal epithelial cells, and myoepithelial cells of the branching ducts. Ar, and to lesser extent Cr-1, were also found in the epithelial cap cells and in the luminal epithelial cells of the advancing terminal end bud (TEB) from virgin 4-week and 6-week-old mice. Western blot analysis demonstrated that both Ar (28 and 26 kDa) and Cr-1 (90, 67, 56, and 21 kDa) proteins are expressed in virgin, 13.5 day midpregnant and in the 14 day lactating mammary gland. In addition, Ar and Cr-1 are associated with developing alveolar structures as determined by ICC. These results imply that together with EGF and transforming growth factor alpha (TGFα), Ar and Cr-1 may play salient roles as modifiers in the morphogenesis and differentiation of the mammary gland. © 1995 Wiley-Liss, Inc.

    Notizen: Intermediate subviral particles (ISVP) derived from reovirus represent a simple model system for the switch-on of transcriptase function. In such particles the endogenous transcriptase is present in a switched-off form, one step removed from the switched-on state. Switch-on of transcriptase function is an active process in this system and can be triggered by K+ ions. A variety of agents which affect gene expression in cells were tested for an effect on switch-on in ISVP. Marked effects on switch-on in ISVP were observed with a diverse group of test agents, including DMSO and other solvents, BUdR, TdR, caffeine, theophylline, and temperature. The correlation in response between ISVP and cells suggests that the ISVP system may be useful as a model for studying the biochemical mechanisms underlying the perturbative effects of such agents on gene expression in cells.

    Notizen: The morphological and ultrastructural changes that occur during preparation of porcine, bovine, and murine spermatozoa for flow cytometric quantification of the relative DNA content of the X- and Y-chromosome-bearing sperm populations were examined. Ejaculated spermatozoa from the boar and bull were washed using a series of dimethyl sulfoxide (DMSO) solutions prior to fixation, whereas the epididymal mouse spermatozoa were washed only in phosphate-buffered saline (PBS). Spermatozoa from all three species were then fixed in ethanol and processed for fluorochrome staining by a treatment regimen consisting of sulfhydryl reduction and proteolysis. The processed sperm nuclei were stained for DNA with the fluorochrome, 4′-6-diamidino-2-phenylindole (DAPI) before quantification by flow cytometry. Scanning and transmission electron micrographs of sperm heads taken at various steps of the preparation and staining procedures show 1) that the rigorous washing procedure disrupted the plasma and outer acrosomal membranes, 2) that ethanol fixation resulted in removal of the outer membranes and disintegration of the nuclear envelope, and 3) that thiol and proteolysis treatment removed the remaining cellular organelles including the tail and rapidly induced partial decondensation of the tightly packed chromatin. Sequential micrographs showed that the nuclear matrix of all three species increased in thickness about twofold during the preparation and staining. Consequently, the harsh procedures currently used for quantitative staining of DNA for high-resolution flow cytometric analyses destroy most cellular organelles and thereby prevent simultaneous characterization of DNA content and other sperm cell constituents.

    Notizen: The effects of the male antifertility agent, α-chlorohydrin, six of its derivatives, and glycidol were studied on the metabolism of washed ram spermatozoa in vitro with fructose as substrate. The α-chlorohydrin derivatives were the amino, the phosphorylated, and four glycol-bridge (ketal) compounds. All compounds except glycidol, in a concentration between 0.1 and 100 mM, reduced the aerobic glycolsis and/or oxidation of fructose. However, there was not a high correlation between the ability of these compounds to inhibit the metabolism of ram spermatozoa in vitro and their antifertility activity when administered to male rats. Other factors are clearly involved in their antifertility activity, eg, the concentration of the compounds in the epididymis and their conversion of either more or less spermicidal compounds in the body.

    Beschreibung: Space-based and airborne coherent Doppler lidars designed for measuring global tropospheric wind profiles in cloud-free air rely on backscatter, beta from aerosols acting as passive wind tracers. Aerosol beta distribution in the vertical can vary over as much as 5-6 orders of magnitude. Thus, the design of a wave length-specific, space-borne or airborne lidar must account for the magnitude of 8 in the region or features of interest. The SPAce Readiness Coherent Lidar Experiment under development by the National Aeronautics and Space Administration (NASA) and scheduled for launch on the Space Shuttle in 2001, will demonstrate wind measurements from space using a solid-state 2 micrometer coherent Doppler lidar. Consequently, there is a critical need to understand variability of aerosol beta at 2.1 micrometers, to evaluate signal detection under varying aerosol loading conditions. Although few direct measurements of beta at 2.1 micrometers exist, extensive datasets, including climatologies in widely-separated locations, do exist for other wavelengths based on CO2 and Nd:YAG lidars. Datasets also exist for the associated microphysical and chemical properties. An example of a multi-parametric dataset is that of the NASA GLObal Backscatter Experiment (GLOBE) in 1990 in which aerosol chemistry and size distributions were measured concurrently with multi-wavelength lidar backscatter observations. More recently, continuous-wave (CW) lidar backscatter measurements at mid-infrared wavelengths have been made during the Multicenter Airborne Coherent Atmospheric Wind Sensor (MACAWS) experiment in 1995. Using Lorenz-Mie theory, these datasets have been used to develop a method to convert lidar backscatter to the 2.1 micrometer wavelength. This paper presents comparison of modeled backscatter at wavelengths for which backscatter measurements exist including converted beta (sub 2.1).

    Beschreibung: Aerosol backscatter coefficient data were examined from two nights near Japan and Hawaii undertaken during NASA's Global Backscatter Experiment (GLOBE) in May-June 1990. During each of these two nights the aircraft traversed different altitudes within a region of the atmosphere defined by the same set of latitude and longitude coordinates. This provided an ideal opportunity to allow flight level focused continuous wave (CW) lidar backscatter measured at 9.11-micron wavelength and modeled aerosol backscatter from two aerosol optical counters to be compared with pulsed lidar aerosol backscatter data at 1.06- and 9.25-micron wavelengths. The best agreement between all sensors was found in the altitude region below 7 km, where backscatter values were moderately high at all three wavelengths. Above this altitude the pulsed lidar backscatter data at 1.06- and 9.25-micron wavelengths were higher than the flight level data obtained from the CW lidar or derived from the optical counters, suggesting sample volume effects were responsible for this. Aerosol microphysics analysis of data near Japan revealed a strong sea-salt aerosol plume extending upward from the marine boundary layer. On the basis of sample volume differences, it was found that large particles were of different composition compared with the small particles for low backscatter conditions.

    Beschreibung: Water vapor plays an important role in the energetics of the boundary layer processes which in turn play a key role in regulating regional and global climate. It plays a primary role in Earth's hydrological cycle, in radiation balance as a direct absorber of infrared radiation, and in atmospheric circulation as a latent heat energy source, as well as in determining cloud development and atmospheric stability. Water vapor concentration, expressed as a mass mixing ratio (g kg(exp -l)), is conserved in all meteorological processes except condensation and evaporation. This property makes it an ideal choice for studying many of the atmosphere's dynamic features. Raman scattering measurements from lidar also allow retrieval of water vapor mixing ratio profiles at high temporal and vertical resolution. Raman lidars sense water vapor to altitudes not achievable with towers and surface systems, sample the atmosphere at much higher temporal resolution than radiosondes or satellites, and do not require strong vertical gradients or turbulent fluctuations in temperature that is required by acoustic sounders and radars. Analysis of highly-resolved water vapor profiles are used here to characterize two important mesoscale flows: thunderstorm outflows and a cold front passage. The data were obtained at the Atmospheric Radiation Measurement Site (CART) by the groundbased Department of Energy/Sandia National Laboratories lidar (CART Raman lidar or CARL) and Goddard Space Flight Center Scanning Raman Lidar (SRL). A detailed discussion of the SRL and CARL performance during the IOPs is given by others in this meeting.

    Beschreibung: Ion acceleration and flux increase associated with substorm energetic particle injections are investigated on the basis of geosynchronous observations and test proton orbits in the dynamic fields of a three dimensional magnetohydrodynamic simulation of neutral line formation and dipolarization in the magnetotail. The energetic particle flux changes obtained from the test particle orbits agree well with observations that demonstrate rapid ion flux increases at energies of above 20 keV. The injection region inferred from the test particles has a sharp earthward boundary and a sharp ragged tailward boundary. The earthward portion of the enhanced ion flux can be traced to the enhanced cross-tail electric field associated with the near-earth x-type neutral line. Due to the rapid earthward motion of accelerated ions away from the neutral line, this boundary is displaced earthward to where the energetic ions become more adiabatic in the stronger dipolar field.
