    Notizen: A theoretically contracted agonist conformation of potent phenoxypropanolamine derivatives on the β1-adrenoceptor has been analyzed in detail. The main effect of the enthalpic contraction of some 6.0-7.0 kcal/mol arises from the movement of the nitrogen atom toward the aromatic ring by 0.7-0.8 Å, requiring some 3.0-3.5 kcal/mol. A second effect arising from the contraction can be a dihedral rotation of some 30° around the O—CH2 bond of the planar anisole moiety. This rotation is correlated with an effect arising from “in-plane” deformation of the anisole moiety where opening of the relevant bond angle releases steric constraints for this rotation. Ortho-substituents assist this rotation indirectly through hyperconjugation with the lone pair of the OCH2 group, electron-attracting substituents opening this bond angle and lowering the energy required to reach a given bond-angle deformation. The adjacent ring meta-substituent can be similarly affected, the strength of the total effect being also of the order of 3.0-3.5 kcal/mol. The net effect gives rise to a further contraction of the nitrogen atom and the betahydroxyl group toward the aromatic ring, the beta-hydroxyl group showing a contraction of up to 0.4-0.5 Å along the main axis of the conformer. The deformed conformation is consistent with the predicted conformer of a fixed-ring benzdioxepine molecule that possesses the highest degree of partial agonism within the set of phenoxypropanolamine agents. It is concluded that ortho-substituents in phenoxypropanolamine derivatives can retain steric freedom in both agonist and antagonist action provided that the substituent can accommodate the required deformation, both agonist and antagonist conformer forms lying within one unbound conformation. The agonist conformer is consistent with the proposed model for the ligand-activated transmembrane proton transfer in the β1-adrenoceptor where a contraction along the main axis of the ligand conformer (with some attendant distortion in the position of the β-hydroxyl moiety) is required to activate proton transfer. © 1994 John Wiley & Sons, Inc.

    Notizen: Molecular dynamics simulations are used to study hypervelocity impacts of an ultrathin flyer plate with a semi-infinite two-dimensional model diatomic molecular solid. We show that these hypervelocity impacts can produce a dissociative phase transition from a molecular to a close-packed solid in the target material. We also show that hypervelocity impacts of ultrathin plates can produce extensive chemical reactions leading to a detonation accompanied by a phase transition in an energetic version of the model. © 1994 John Wiley & Sons, Inc.

    Notizen: Formulas are derived for the density matrices belonging to an n-particle wave function built on the basis of single-center explicitly correlated Gaussian basis functions. An explicit formula for the first-order density matrix, P(r1, r′1), is obtained for computing the probability distribution P(r1, r1). Other formulas are derived for matrix elements of the first-order density operator P on a basis of single-particle Gaussian orbitals so that natural orbitals (NOs) can be expressed in such a basis. The method is illustrated for the case of the ground state of the helium atom using the 16-term (geminal) wave function by Singer and Longstaff (E = -2.90233 au) and a set of even-tempered Gaussian orbitals. The resulting natural orbitals compare favorably with natural orbitals from Cl expansions. The method is also applied to our 20 term (trimal) wave function for the ground state of dipositronium (E = -0.51560 au). Analysis is made in this case for pair correlation functions of both the electron-electron and the positron-electron pairs; results include the radial distributions of these pairs and their relative angular momentum. © 1996 John Wiley & Sons, Inc.

    Notizen: The pairwise intercorrelation between 12 topological indices is studied in benzenoid hydrocarbons with up to six rings. The functions used in the intercorrelations were linear, quadratic, cubic, power low, and logarithmic relationships. The last two relationships failed to intercorrelate the selected topological indices. The results obtained suggested that there are basically three subclasses of independent topological indices in the benzenoids studied. When contrasted with the previous findings in alkanes, our results suggest that the cyclicity leads to the stronger intercorrelation of topological indices than the branching of a molecule. © 1992 John Wiley & Sons, Inc.

    Notizen: An alignment of the α-helices of the β1-adrenoceptor on the bacteriorhodopsin structure has produced a hypothesis that the receptor can act as a proton pump during excitation (through helices III, IV, V, VI, and VII) while at the same time, through a correlated gating mechanism, develop an intermittent ion channel (via helices I, II, III, and VII). Proton transfer is achieved through a proton shuttle consisting of Tyr377-Arg156-Tyr157 in which Arg156 is unprotonated in the resting state of the receptor. Activation of Tyr377 produces the initial transfer of a proton to Arg156, while the hydrogen-bond reorganization involved in this activation realigns two aspargine residues 369 and 373 that move toward the interface of helices III and VII. The β-hydroxyl of the natural ligand via Ser145 together with the para-hydroxyl group of the natural ligand activate the proton transfer, displacing Trp183 and Asn373 from the phenolic oxygen atom of Tyr377 by stronger hydrogen-bond proton donor interactions. An estimate of the enthalpic changes produced by the full agonist, isoprenaline, at the CHARMM modeled hydrogen-bond distances of 2.1-2.2 Å where significant proton transfer is not yet expected to take place, gives a minimum value of 5.0-7.0 kcal for the hydrogen-bond reorganization based on 3-21G calculations and scaled values based on 6-31G** hydrogen-bond pairs. The enthalpic change approaches the 6-7 kcal observed in the polar bonding differences between agonist and antagonist components of the partial agonist, prenalterol, on the cardiac β1-adrenoceptor and in that predicted from agonists and antagonists using a comparative ligand-receptor binding model for turkey erythrocyte β-adrenoceptor data. In the CHARMM-based model, the loss of this activating energy brings the energetics of interaction close to alternative interactions of the natural ligand catechol moieties at higher levels within the membrane. The unusually high entropy of antagonist binding of phenoxypropanolamine β1-adrenoceptor agent is qualitatively accounted for. In line with experimental prediction, it is concluded that agonist-ligand conformer-receptor interaction is a dominantly enthalpic process and major α-helical movement is not expected in agonist action. © 1992 John Wiley & Sons, Inc.

This fact allows an approximate separation of the hyperradius, leading to many-dimensional hydrogenlike radial solutions. Kindred applications to dimensional scaling are also discussed, where d = DN, with D the spatial dimension. For the large D regime, illustrative analytic formulas are obtained giving the energy and effective nuclear charge for the lowest few S states of the helium atom.

    Notizen: We define the algebrant, a mathematical generalization of the determinant, the immanant, the permanent, and the Schur functions. Algebrants are classified as multilinear matrix functions or multicomponent symmetrized tensors. In applications, such as N-electron quantum mechanics, where extensive computation is required, it is vital to reduce computational effort, e.g., the well-known N-factorial problem. We derive certain mathematical properties that can be incorporated in efficient computing algorithms for algebrants. Foremost is our “elimination theorem,” which allows (in important special cases) zeros to be introduced into an algebrant in close analogy with Gaussian elimination for determinants. Savings accruing from such elimination can be substantial. We show examples from Matsen's spin-free quantum chemistry where elimination effectively removes the N-factorial problem that has hitherto stifled possible applications.

    Notizen: This work proposes a very detailed ab initio study of the hypothesis for a one-step serine protease acylation process with one water molecule acting as the main catalyst reactant. For the 11 increasing complexity models considered, the minimum and transition state conformations for the reaction are determined by full geometry optimizations at the ab initio self-consistent field (SCF) levels within several basis sets, from MINI-1 to 6-31G, and, for the smallest complexes, at the post-SCF MP2 level within the 6-31G** basis set. The related thermodynamical quantities are calculated for all the conformations. The influence of the oxyanion hole stabilizer and of the dyad His57-Asp102 is quantified and a very good agreement is obtained with point mutagenesis. The activation barrier is found in the range 15-18 kcal/mol. © 1996 John Wiley & Sons, Inc.

    Notizen: Several configurations of the C2H4 · HF molecular complex were studied using the 4-31G basis set. The most stable configuration was found to be the π hydrogen bonded complex. For this structure the interaction energy was computed employing a double-zeta basis plus polarization functions. The changes in electronic properties of the components C2H4 and HF due to complex formation were discussed.
