    Notes: Proteins have been immobilized in porous support particles held in a fixed-bed reactor through which protein solution is continuously circulated. Changing the recirculation flow rate alters the observed immobilization kinetics and the maximum enzyme loading which can be achieved for glucose oxidase and glucoamylase on carbodiimide-treated activated carbon and for glucoamylase immobilized on CNBr-Sepharose 4B. Direct microscopic examination of FITC-labelled protein in sectioned Sepharose particles and indirect activity-loading studies with activated carbon-enzyme conjugates all indicate that immobilized enzyme is increasingly localized near the outer surface of the support particles at larger recirculation flow rates. Restricted diffusion of enzymes may be implicated in this phenomenon. These contacting effects may be significant considerations in the scaleup of processes for protein impregnation in porous supports, since apparent activity and stability of the final preparation depend on internal protein distribution.

    Notes: A carrier-supported mycelial growth of Penicillium chrysogenum was applied to penicillin fermentation system using celite as a support material. Hyphal growth through the pore matrices of the material showed strong anchorages and provided highly stable biofilm growth. With bioparticles developed in such a manner, both cell growth and penicillin production were observed to increase significantly compared to the conventional dispersed filamentous cultures. Maximum values of specific penicillin production rate were found to be constant regardless of the growth form. A three-phase fluidized-bed fermentor was designed and tested for penicillin production using the bioparticles. Two modes of operation, semicontinuous and repeated fed batch, of the fermentor were tried. It was noted that the overgrowth of free mycelia and the development of fluffy loose bioparticles caused poor mixing and made the fermentor operation quite difficult. Control of the bioparticle size and the extension of production phase were therefore considered important to maintain the reactor productivity at a desired level. From the results of repeated fed-batch operation it was found that the control of bioparticle size could be successfully achieved by phosphate-limiting culture condition. Penicillin production under this condition was also observed to be maintained at a high level (about 80% of the maximum) for at least 1 month.

    Notes: Raman spectra of polycrystalline NH4VO3 and ND4VO3 were recorded at various temperatures. Splittings were observed for most fundamentals and the external mode region contains at least eleven bands. The spectra are interpreted in terms of a group theoretical analysis based on the molecular symmetry and the known crystal structure.Wavenumber vs temperature curves for several internal and external modes show minor anomalies near 150 K, which might be due to an order-disorder phase transition. The bands assigned to ammonium ion librations are observed at room temperature, which implies that the onset of ‘free’ rotation of the NH4+ ion (on the vibrational time scale) occurs at a much higher temperature in NH4VO3 than it does in other ammonium salts.

    Notes: Raman spectra of polycrystalline normal and three 15N isotopic ammonium nitrates (15NH4NO3, NH4 15NO3 and 15NH4 15NO3) were recorded at various temperatures between 10 and 300 K. Isotopic shifts of both internal and external modes of the ammonium ion relative to isotopic normal NH4NO3 were observed. Various components of the hetero-ionic coupled internal mode (ν3 of the nitrate ion with ν4′ of the ammonium ion) were assigned with more certainty on the basis of the 15N isotopic shifts. Detailed studies of the temperature dependence of one internal mode (ν1) of the NO3 - ion and several external modes were made. The frequency and half band width versus temperature plots for both the internal mode and the external modes of 15NH4NO3 indicate only one crystal structure (phase V) between 10 and 257 K. Comparison of the frequency shifts between various isotopic compounds in the external mode region permitted a more precise assignment of the lattice modes associated with the NH4+ and NO3- ions to be made.

    Notes: Infrared and Raman spectra of polycrystalline films of CF3 Br have been recorded at 77 K. All six fundamentals of the molecule have been observed in both spectra. Observed splittings of fundamentals have been attributed to Fermi resonance (ν1), bramine isotope splitting (ν3) and crystal splitting (ν4, ν5 and ν6). Some bands due to 13CF3 Br have also been identified. The crystal splittings can be explained if it is assumed that the structure is monuclinic with factor group C2h and two molecules per unit cell on sites of Cs symmetry.

    Notes: The nitrile stretching region of binary solutions of phenol in propionitrile has been studied by means of Raman spectroscopy. The number of independently varying spectral components in this region was determined by Factor Analysis to be two. The results of Factor Analysis were supported by the observation of a pseudo-isosbestic point. The band envelope was fitted with Lorentzian - Gaussian product functions using a non-linear least squares procedure. The results indicate the formation of a 1:1 complex. Equilibrium constants calculated for the complex formation reaction showed a strong concentration dependence.

    Notes: 15NH4ReO4 was synthesized and the Raman spectrum recorded at various temperatures between 10 and 300 K. 15N isotopic shifts of both internal and external modes of the ammonium ion relative to 14NH4ReO4 were observed. The temperature dependence of the external modes was studied. The librational lattice mode associated with the ammonium ion disappeared above about 180 K. At this temperature anomalies were observed in the temperature dependence of the perrhenate ion lattice modes.
