    Notes: One of the requirements for enhanced productivity by the animal culture systems used in biotechnology is the direct assessment of the metabolic rate by on-line biosensors. Based on the fact that cell growth is associated with an enthalpy change, it is shown that the specific heat flow rate is stoichiometrically related to the net specific rates of substrates, products, and indeed to specific growth rate, and therefore a direct reflection of metabolic rate. Heat flow rate measured by conduction calorimetry has a technical advantage over estimates for many material flows which require assays at a minimum of two discrete times to give the rate. In order to make heat flow rate specific to the amount of the living cellular system, it would be advantageous to divide it by viable biomass. This requirement has been fulfilled by combining a continuous flow microcalorimeter ex situ with a dielectric spectroscope in situ, the latter measuring the viable cell mass volume fraction. The quality of the resulting biosensor for specific heat flow rate was illustrated using batch cultures of Chinese hamster ovary cells (CHO 320) producing recombinant human interferon-γ (IFN-γ) during growth in a stirred tank bioreactor under fully aerobic conditions. The measuring scatter of the probe was decreased significantly by applying the moving average technique to the two participant signals.It was demonstrated that the total metabolic rate of the cells, as indicated by the specific heat flow rate sensor, decreased with increasing time in batch culture, coincident with the decline in the two major substrates, glucose and glutamine, and the accumulation of the by-products, ammonia and lactate. Furthermore, the specific heat flow rate was an earlier indicator of substrate depletion than the flow rate alone. The calorimetric-respirometric ratio showed the intensive participation of anaerobic processes during growth and the related IFN-γ production. Specific heat flow rate was monotonically related to specific cell growth rate and associated with specific IFN-γ production. Specific heat flow rate is potentially a valid control variable for the growth of genetically engineered cell lines producing target proteins. © 1998 John Wiley & Sons, Inc.     Notes: The effect of the growth phase of Spodoptera frugiperda (Sf9) cells on the production of recombinant proteins (β-galactosidase and glucocerebrosidase) was investigated. Cells infected with the recombinant Autographa californica nuclear polyhedrosis virus at the late exponential and stationary phases yielded low quantities of expressed protein. Highest enzyme yields were obtained using Sf9 cells from the early exponential phase (0.9 mg β-galactosidase/106 cells and 1.7 μg glucocerebrosidase/106 cells). Infection of resuspension of cells collected from various phases of growth in fresh medium resulted in 75% restoration of maximal expression levels. This finding suggested either nutrient limitation or waste product accumulation as the cause of the decrease in productivity at the latter phases of growth. Further experiments revealed that the highest productivity levels could be obtained with cultures of Sf9 cells grown in a fermentor to a cell concentration of 4 × 106 mL-1. The medium needed to be replaced prior to infection with the recombinant virus and supplemented with a mixture of glucose, L-glutamine, and yeastolate ultrafiltrate. © 1993 John Wiley & Sons, Inc.

    Notes: Tissue-engineered implants require appropriate biomaterials to serve the required physical function of the tissue being repaired or replaced while facilitating remodeling of the implant. We report on the development of implantable fabrics manufactured from continuous collagen threads. The collagen threads are formed by extrusion of native, acid-extracted bovine colagen into a buffered solution of polyethylene glycol, followed by rinsing and air drying. The high manufacturing rate of such threads permits the production of colagen fabrics of various configurations. The fiber diameter can be controlled, and threads with dry diameters as low as 25 μm have been produced. Braids and bundles of collagen threads implanted as a replacement of the anterior cruciate ligament in a dog model were completely remodeled into host tissue by 12 weeks. Knitted collagen fabrics implanted in a rat abdominal repair model prevented herniation, and connective tissue ingrowth was observed within the fabric by 12 weeks.

Die Strukturen wurden anhand von Modellsubstanzen mittels Massenspektroskopie identifiziert. Die ursprünglichen Resole wurden auf gleiche Weise analysiert.
    Notes: The characterisation of two resols which had been etherified with methanol under acidic conditions is described for the first time. The resins were converted into trimethylsilyl derivatives and the individual components were separated by means of capillary gas chromatography. Structural assignments were made using mass spectrometry measurements and with the aid of model compounds. The precursor resols have also been analysed by the same technique.

In the reaction of CpMn(CO)2CNPPh2 with CpMn(CO)2 THF or of K[CpMn(CO)2CN] with CpMn(CO)2PPh2Cl a complex with an isocyanophosphane bridge, {CpMn(CO)2}2(CNPPh2) (4), is obtained. The IR, NMR, and mass spectra of the compounds 3a-d and 4 are discussed.

    Notes: Uptake of orthophosphate (Pi) by osteoblast-like cells is known to be stimulated by parathyroid hormone (PTH), but effects on intracellular [Pi] have not been investigated. Here we show in rat osteoblast-like cells (UMR 106-06) that PTH (10-11 to 10-7 M) increases both 32Pi uptake and cellular [Pi] by up to 50 per cent. 1,25 Dihydroxyvitamin D3 (1,25D) (10-12 to 10-6 M) and salmon calcitonin (CT) (10-12 to 10-6 g ml-1) also increased cellular [Pi] (by up to 60 per cent), but the percentage increases in total cellular 32Pi uptake were smaller. The effects of 1,25D were transient (observable at 80 min and 6 h but not 24 h), and were also observed with 24,25 dihydroxy- and 25 hydroxyvitamin D3. Transient degradation of organic phosphorus pools to Pi might contribute to this increased [Pi]. These pools remain to be identified but were not shown to be phospholipids. Foetal bovine serum also affected cellular [Pi]. Care is therefore needed in distinguishing direct hormonal effects on cellular [Pi] from indirect effects arising from changes in the rate of cell growth.

    Notes: Osteoblast-like cells possess Na-dependent transporters which accumulate orthophosphate (Pi) from the extracellular medium. This may be important in bone formation. Here we describe parallel measurements of Pi uptake and cellular [Pi] in such cells from the rat (UMR 106-01 and UMR 106-06) and human (OB), and in non-osteoblastic human fibroblasts (Detroit 532 (DET)). In UMR 106-01, cellular [Pi] was weakly dependent on extracellular [Pi] and higher than expected from passive transport alone. [32Pi]-uptake was inhibited by Na deprivation, but paradoxically increased on K deprivation. With Na, 87 per cent of cellular 32P was found in organic phosphorus pools after only 5 min. Na deprivation also decreased cellular [Pi], in both UMR 106-01 and DET, but the decrease was smaller than that in [32Pi]-uptake. Ouabain decreased [32Pi]-uptake and cellular [Pi] in DET, but not in UMR 106-01. Regulation of cellular [Pi] is therefore at least partly dependent on Na/Pi co-transport, but this does not seem to be an exclusive property of osteoblasts.
